CD-MOF Based pH/ROS Sensitive 5-ASA Delivery System for Ulcerative Colitis Therapy

Yanxue Si,Ruitao Cha,Pai Zhang,Fengshan Zhou,Wenbing Yuan,Xingyu Jiang

5-Aminosalicylic acid (5-ASA) is the first-line drug for ulcerative colitis (UC), which encounters therapeutic constraints due to the poor water solubility and absorption by the upper gastrointestinal. Here, a cyclodextrin-based metal-organic framework (CD-MOF) is used as a carrier for 5-ASA (ASA@CM) to enhance its solubility. To improve the targeted release 5-ASA, ASA@CM is incorporated into sodium alginate/ L -arginine complex (SA-LA) to obtain the desired microspheres (ASA@CM/SL) by electrospray. ASA@CM/SL safeguards 5-ASA against the harsh gastrointestinal environment and provides favorable delivery to the colon. The anti-inflammatory and anti-oxidant activity is studied by measuring inflammatory factors and reactive oxygen species (ROS). The therapeutic effect of ASA@CM/SL on UC mice induced with dextran sulfate sodium is assessed. The biosafety by evaluating cytotoxicity and organ toxicity is studied. ASA@CM/SL shows excellent therapeutic effects for UC, as evidenced by its alleviation of inflammatory response and restoration of disrupted intestinal barriers. ASA@CM/SL features low cytotoxicity and low organ toxicity. This study provides a potential strategy for improving 5-ASA solubility and delivery efficiency, which may also be applied to broadly tackle various small hydrophobic molecules to treat UC and other intestinal diseases.

Reduced glycoprotein hormone β-5 links male aging and testosterone decline to increased adiposity

Gengmiao Xiao,Aijun Qian,Zhuo Gao,Tingting Dai,Hui Liang,Shuai Wang,Mulan Deng,Yunjing Yan,Xindan Zhang,Xuedi Zhang,Yunping Mu,Jiqiu Wang,Aibo Gao,Huijie Zhang,Fanghong Li,Allan Zijian Zhao

Aging commonly causes decline of testosterone or estrogen, leading to overaccumulation of fatness in males or females, respectively. Although such phenomenon can be readily explained by estrogen’s direct action on adipocytes in females, accumulative evidence does not support the direct action of testosterone in adipocyte lipid metabolism, suggesting that there is a missing intermediary link. Herein, we propose that glycoprotein hormone β5 (GPHB5) is the intermediary linkage between testosterone and the regulation of adiposity. In clinical samples, blood levels of GPHB5 were correlated negatively with men’s ages, and positively with circulating testosterone. Testosterone directly stimulated the expression of GPHB5 in cultured cells, pharmacological blockade of androgen receptor (AR) functions abrogated such effect. Knockout of AR led to not only development of obesity but also reduction of GPHB5 expression. Genetic ablation of GPHB5 in the males, but not in the females, lowered the browning of white adipose tissue, diminished energy expenditure and caused severe obesity. Importantly, elevated blood testosterone didn’t exert its catabolic actions in GPHB5 null mice, and yet, recombinant GPHB5 protein was able to stimulate energy expenditure and reduce adiposity. Taken together, these results provided the strong proof that GPHB5 is the “missing” intermediary hormone linking testosterone (and aging) and its well-known catabolic effect on adipose tissue.

Phosphorus-assisted reconstruction creates NiOOH/CoOOH interfaces for efficient biomass-derived 5-hydroxymethylfurfural electrooxidation

Yuwei Xiong,Chen Deng,Wei Ji,Yifei Sun,Richen Lin

Although cobalt (Co) and phosphorus (P) are widely employed to enhance the electrooxidation performance of Ni-based electrocatalysts, their synergistic roles in regulating metal-site activation and interfacial reconstruction during 5-hydroxymethylfurfural (HMF) oxidation remain unclear. Herein, by comparatively studying the electrochemical behavior and structural evolution of Ni-P/NF and Ni 0.8 Co 0.2 -P/NF during the HMF oxidation reaction (HMFOR), we identify a Co-P synergistic reconstruction mechanism that governs the dynamic formation of active interfaces. Ni 0.8 Co 0.2 -P/NF delivers a low onset potential of 1.33 V (vs. RHE) in 50 mM HMF-containing alkaline electrolyte and achieves a high current density of 900 mA cm −2 at 1.60 V (vs. RHE), with a Faradaic efficiency of 97 % toward FDCA. Mechanistic studies reveal that Co modulates the initial electronic structure of Ni sites, while P stabilizes the evolution of Co species into CoOOH under reaction conditions. The in situ generated CoOOH dynamically couples with neighboring Ni sites, thereby promoting the low-potential reconstruction of Ni 2+ into active NiOOH. In situ Raman spectroscopy further confirms the formation of a stable NiOOH/CoOOH composite active interface during HMFOR, which accounts for the reduced onset potential and accelerated reaction kinetics. This work highlights the importance of coupling static electronic modulation with dynamic electrochemical reconstruction, providing mechanistic insights for the rational design of multicomponent electrocatalysts for biomass-derived 5-hydroxymethylfurfural electrooxidation.

A novel non-enzymatic sensing platform for determination of 5′-guanosine monophosphate in meat

Qiqi Fei,Nana Zhang,Chong Sun,Panpan Zhang,Xiaodi Yang,Yunhui Hua,Li Li

Graphitic carbon nitride (g-C 3 N 4 ) doped carboxylated MWCNTs nanocomposite was synthesized using a simple method. The composite films containing 45 wt%, 50 wt%, 56 wt%, 67 wt% fraction of the carboxylated MWCNTs doped into g-C 3 N 4 were fabricated and characterized. An electrochemical non-enzymatic sensor for determination of 5′-guanosine monophosphate (GMP) based on the nanocomposite was developed. The results indicate that the g-C 3 N 4 -carboxylated MWCNTs nanocomposite has highly electrocatalytic activity, good conductivity and biocompatibility, which plays an essential role in the determination of GMP . Under the optimum conditions, the linear fitting equation was I (µA) = −0.0022c (μg·mL −1 ) + 0.3560 (R 2  = 0.9982). The linear range was from 0.5 to 100 μg·mL −1 and the detection limit (LOD, S/N = 3) was 0.109 μg·mL −1 . This non-enzymatic sensor can offer a better alternative to other methods for the analysis of GMP because of cheap cost, low detection limit and good anti-jamming capability in meat quality evaluation.

Hydrothermally Synthesized HZSM-5/SAPO-34 Composite Zeolite Catalyst for Ethanol Conversion to Propylene

Duan Chao,Zhang Xin,Zhou Rui,Hua Yuan,Chen Jie,Zhang Li

HZSM-5/SAPO-34 zeolite composites were prepared by hydrothermal synthesis (ZS-HS) and mechanical mixture (ZS-MM), respectively. ZS-HS showed higher propylene yield and better catalytic stability in ethanol conversion than ZS-MM as well as HZSM-5 and SAPO-34. In case of ZS-HS, SAPO-34 at least partially overlapped on HZSM-5, which resulted in the interfacial interaction of HZSM-5 and SAPO-34. Such interaction significantly modified the texture and the acidity of ZS-HS, which in turn affected its catalytic reactivity. ZS-HS showed the higher apparent formation rate of propylene, due to its moderate concentration and strength distribution of acid sites. Propylene, ethylene and butylenes might produce through the parallel reaction pathways and the common intermediate on ZS-HS. Role of Al pairs on effective N2O decomposition over the ZSM-5 zeolite catalyst

Bin Kang,Ming Li,Zhaoying Di,Xiaonan Guo,Ying Wei,Jingbo Jia,Runduo Zhang

Series of ZSM-5 zeolites with diverse contents and locations of Al pairs were synthesized, and Co 2+ ions were subsequently anchored at the Al pairs of ZSM-5 zeolites by wet ion-exchange with H protons. The Co-ZSM-5 sample derived from the mixtures of AlCl 3 and TEOS of Si/Al=30.3 showed the best de N 2 O performance. Analysis on Co (Ⅱ) ion exchange capacity demonstrated the content of Al pairs followed the sequence: AlCl 3 (0.84) > Al(NO 3 ) 3 (0.73) >  aluminum isopropoxide (0.65) and TEOS (0.84) >  silicic acid (0.57). More Al pairs coordinated with protons or cobalt ions were essentially associated with better performances for N 2 O catalytic dissociation. Moreover, Si source had a significant effect on the location of Al pairs determined by UV–vis–DRS of bare Co (Ⅱ), Al pairs of ZSM-5 zeolite synthesized from TEOS were enriched in the sinusoidal channel (γ site), whereas those synthesized from silicic acid were concentrated in the channel intersections (6-MRs β site). The theoretical calculation revealed that the O 2 desorption at γ-site Co 2+ of ZSM-5 zeolite required to overcome less energy barrier (23.8 kcal/mol) than that at β-site one.

Catalytic pyrolysis of oleic acid to BTX over Ni-modified nano-ZSM-5 zeolite

Guo Hanxu,Fu Sen,Yuan Hong

Hierarchical zeolites can provide multidimensional spatial networks, thereby holding significant potential as catalysts for BTX production from biomass pyrolysis. In this study, nano-ZSM-5 zeolite (Z X-X ) with a hierarchical pore structure was synthesized using glycine as a growth inhibitor. Due to the exceptional ability of Ni to cleave C–C and C–H bonds, Ni modification was performed on the Z 30-0.2 catalyst. The effects of glycine and Ni loading on the morphology and pore structure of the zeolites were systematically investigated using scanning electron microscopy (SEM), transmission electron microscopy (TEM), and N 2 adsorption–desorption. The results demonstrated that the particle size of 10 wt% Ni-Z 30-0.2 zeolite ranged from 200 to 300 nm, with a BET surface area of 314 m 2 /g and abundant mesopores. The NH 3 -TPD results showed that the acid amount of 10 wt% Ni-Z 30-0.2 was 0.72 mmol/g. The prepared zeolite was employed in a fixed-bed reactor to catalyze the pyrolysis of oleic acid for the production of benzene, toluene, and xylene (BTX). The introduction of metal Ni significantly enhanced the BTX yield. At 500 ℃, the average BTX yield achieved with the 10 wt% Ni-Z 30-0.2 catalyst was 19.4 wt%, which was higher than that of the zeolite without Ni loading.

Ammonia-Steam Treatment of ZSM-5 for the efficient conversion of ethylenediamine to triethylenediamine

Leqing He,Changyue Shao,Ping Liu,Yingxia Li,Jiqin Zhu,Chongpin Huang

Conventional ZSM-5 zeolites face significant challenges in achieving high product selectivity and avoiding rapid catalyst deactivation due to coke deposition during the condensation of ethylenediamine (EDA) to triethylenediamine (TEDA). Based on the characteristic of ammonia generation during the synthesis of TEDA from EDA, this study treats ZSM-5 with high-temperature ammonia-steam to investigate its effect on the catalytic synthesis of TEDA. The results show that ZSM-5 sample treated with 5   wt% ammonia-steam at 600 ℃ exhibited higher crystallinity and more mesoporous structure. A large amount of framework aluminum  in the catalyst was removed, resulting in significant decrease of acid strength and a high L/B ratio. The modified catalyst achieved complete EDA conversion during the reaction, with 64% TEDA selectivity—a 17% improvement over that of unmodified catalyst. It also exhibited a significant reduction in coke deposition from 6% to 2.3% and a 15-days continuous fixed-bed reaction test confirmed the high stability of the modified catalyst without noticeable deactivation. Compared to acid-base treated and metal ion treated ZSM-5, ammonia-steam treated ZSM-5 demonstrates superior selectivity and stability, and it has been verified that the TEDA yield is higher than that of TS-1. This study provides a novel strategy for designing efficient catalysts for amine cyclization reactions.

Efficient Production of 5-Methoxymethyl-2-furfural from Fructose Catalyzed by Amide-Modified Resin

Chenfeng Li,Jiahao Ju,Beizhan Li,Jilei Xu,Xin Su,Zuoyi Xiao,Qingda An,Jiahui Huang

5-Methoxymethyl-2-furfural (MMF) serves as a crucial biobased platform molecule that can be transformed into various high-value chemicals and biobased polyester monomers. However, the current production of MMF still faces several challenges, such as low yield and prolonged reaction time. In this study, we prepared a series of amide-modified strongly acidic resin catalysts and discovered that they have a higher efficiency in converting fructose to prepare MMF in 1-Butyl-3-methylimidazolium chloride ([BMIM]Cl) and methanol. Among the synthesized catalysts, DB757-NMP demonstrated superior performance, achieving an MMF yield of approximately 61.5% under the optimized conditions, with a combined yield of HMF and MMF reaching about 66.6%. The catalyst formation mechanism was analyzed using FTIR, and NMR, confirming the transformation of proton between NMP and the sulfonic acid groups of the resin, which collectively promoted the conversion of fructose to MMF. In addition, we investigated main reasons for catalyst deactivation and successfully restored catalytic activity through regeneration. The regenerated catalyst could be reused for three times with only a slight decrease in MMF yield. The results suggested that DB757-NMP is a more sufficient and recyclable catalyst for the production of MMF from fructose. This work presented a simple and environmentally benign approach for the synthesis of MMF.

Ternary ZnCdS quantum dots enable efficient photocatalytic oxidation of 5-hydroxymethylfurfural to 2,5-diformylfuran

Shuo Yan,Qing-Xu Fan,Fen-Lian Wang,Yu-Ji Gao,Ling-Bao Xing

Solid-solution Zn 0.2 Cd 0.8 S QDs balance redox ability, band alignment, and stability. • The QDs affords 95.4% HMF conversion and 70.1% DFF yield under visible light. • Synergisticeffects of h + , •O 2 − , and 1 O 2 drive highly selective HMF oxidation to DFF. The selective photocatalytic oxidation of biomass-derived 5-hydroxymethylfurfural (HMF) into high-value platform chemicals represents a promising pathway toward achieving carbon neutrality and sustainable development. To overcome the limited visible-light response and severe photocorrosion of monometallic sulfide quantum dots (QDs), solid-solution ZnCdS QDs were fabricated, enabling efficient photocatalytic oxidation of HMF to high-value 2,5-Diformylfuran (DFF). Systematic characterizations reveal that the ternary ZnCdS QDs achieve an ideal balance between robust redox capability and favorable band alignment, alongside remarkable structural stability and sustainable recyclability. Under optimal conditions, the Zn 0.2 Cd 0.8 S QDs achieved 95.4 ± 0.3% HMF conversion and 70.1 ± 0.9% DFF yield. The reaction mechanism was elucidated via radical trapping experiments and electron paramagnetic resonance (EPR) measurements, demonstrating that reactive species, including h + , •O 2 − , and 1 O 2 synergistically promoted the high-value conversion of HMF to DFF. Protective Effect of Pediococcus pentosaceus Li05 on Constipation via TGR5/TPH1/5-HT Activation

Hui Chen,Fengjiao Wang,Shengjie Li,Longxian Lv,Hao Xu,Jiafeng Xia,Lvwan Xu,Ding Shi,Lanjuan Li

Pediococcus pentosaceus Li05, a strain of lactic acid bacteria isolated from the faeces of healthy volunteers, exhibited potential protective effects against various diseases. This study performed third-generation sequencing and detailed characterisation of its genome. The Li05 chromosome harboured conserved genes associated with acid resistance ( atp ), bile salt resistance ( bsh ), oxidative stress resistance ( hsl , dltA , and et al.), and adhesion ( nrd , gap , and et al.), whereas the plasmid did not contain antibiotic resistance or virulence genes. Following intervention with Li05 in loperamide-induced constipated mice, constipation symptoms improved. Meanwhile, alterations in gut microbiota, increased BSH activity in faeces, and modifications to the faecal bile acid profile were observed. Additionally, expression levels of TGR5 and TPH1 in the colon of the mice increased, leading to elevated 5-HT levels. When the TGR5 gene was knocked out or the TPH1 inhibitor LX1606 was administered to suppress 5-HT synthesis in constipated mice, the beneficial effects of Li05 on gastrointestinal motility and mucus secretion were reversed. Culturing intestinal organoids demonstrated that increased bile acids such as DCA, Iso-LCA, and EALCA could enhance 5-HT levels through the TGR5/TPH1 axis. Therefore, we concluded that Li05 regulated bile acid metabolism, subsequently increasing 5-HT levels through the TGR5/TPH1 axis, thus alleviating constipation. Graphical The mechanism of Li05 in alleviating constipation. Li05 intervention regulated gut microbiota and bile acid metabolism, increasing 5-HT levels through the TGR5/TPH1 activation to ameliorate constipation.

Boosting 5-hydroxymethylfurfural electrooxidation in neutral electrolytes via TEMPO-enhanced dehydrogenation and OH adsorption

Hongfang Wang,Leitao Xu,Jingcheng Wu,Peng Zhou,Shasha Tao,Yuxuan Lu,Xianwen Wu,Shuangyin Wang,Yuqin Zou

5-Hydroxymethylfurfural (HMF) electrooxidation in neutral conditions is a promising strategy to suppress the formation of humins and corrosive effects on electrochemical devices. However, scarce studies have been reported in neutral media due to the deficiency in electrophilic oxygen (eg OH – ) required for the activation of HMF. Herein, 2,2,6,6-tetramethyl-piperidine-1-oxyl (TEMPO)/Co 3 O 4 was utilized to co-catalyze HMF in neutral media, successfully achieving 2,5-furandicarboxylic acid (FDCA) with yield > 99%. It was found that TEMPO could promote HMF dehydrogenation to 2,5-diformylfuran (DFF) and simultaneously activated water via hydrogen-bonding interactions. As a result, the formation of OH* in neutral electrolytes was favored, which was absorbed by electrogenerated active Co species to facilitate subsequent conversion of formyl-group-involved intermediates to FDCA. This work provides a current understanding of the catalytic mechanism for HMFOR in neutral media and guides the design of highly efficient electrocatalysts for biomass upgrading.
