Preparation, characterization, release and antioxidant activity of curcumin-loaded amorphous calcium phosphate nanoparticles

Xia Guo,Wenfeng Li,Heping Wang,Yan-Ying Fan,Huifang Wang,Xianghua Gao,Baolong Niu,Xuechen Gong

In the study, amorphous calcium phosphate (ACP) nanoparticles were prepared by the coprecipitation method to optimize release profile of curcumin (Cur) and avoid burst releases, which were used to overcome the weakness of Cur, such as poor chemical stability and bioavailability. To find the best preparation condition, the influence of reaction concentration, temperature, time and pH on crystal phase of the samples was investigated by XRD and FTIR. The results showed that amorphous calcium phosphate (ACP) was obtained, when pH value and the concentration of PO 4 3− were 8 and 0.024 mM, prepared at 30 °C for 10 min. In vitro drug release assay, ACP nanoparticles showed high loading capacity of Cur and favorable pH-responsive drug release properties. Furthermore, the Cur-load ACP nanoparticles showed an excellent ability to scavenge free radical and damage A549 cells, resulting in a high antioxidant properties and low cell viability. Therefore, the as-prepared nanoparticles have promising applications in the food and biomedical fields.

Hydrazine hydrate and organosolv synergetic pretreatment of corn stover to enhance enzymatic saccharification and co-production of high-quality antioxidant lignin

Lingxia Zhong,Xiao Zhang,Chenglun Tang,Yanjun Chen,Tao Shen,Chenjie Zhu,Hanjie Ying

Hydrazine hydrate and organosolv synergetic pretreatment of corn stover was developed. With the aid of alkaline and reductive hydrazine hydrate, a high delignification rate (77.94%) and total sugar yield of 90.27% (96.60% glucose, 78.82% xylose) based on the pulp could be obtained using hydrazine hydrate (10 mmol/g corn stover) and aqueous ethanol (60 vt%) as the solvent system. Analysis techniques such as FT-IR, SEM, XRD, and NMR were employed to characterize the structure and property changes of stock before and after pretreatment to evaluate the effect of hydrazine hydrate. A mechanism is proposed for the presented system.

Preparation, characterization and antioxidant activity of polysaccharide from spent Lentinus edodes substrate

Hongji Zhu,Li Tian,Lei Zhang,Jingxiu Bi,Qianqian Song,Hui Yang,Jianjun Qiao

This study explored the potential of spent Lentinus edodes substrate, a by-product of mushroom industries causing environmental pollution, serving as materials to produce antioxidant polysaccharide. The extraction process of spent Lentinus edodes substrate polysaccharide (SLSP) was optimized and the effects of drying methods on chemical composition, morphological property and antioxidant activity were investigated. Results showed that freeze-dried SLSP (SLSP-F) exhibited the best quality in terms of the polysaccharide yield (13.00%) and antioxidant activity. The EC 50 values of SLSP-F on DPPH, ABTS and superoxide anion radicals was 0.051   mg/mL, 0.379   mg/mL, 0.719   mg/mL, respectively, which was significantly lower than that of freeze-dried Lentinus edodes polysaccharide (LP-F). After purification by Sephadex G-150, the purified SLSP-F (PSP) has a molecular weight of 16.77   kDa. Compared with LP-F, PSP has more reducing sugars and uronic acids in chemical composition and higher contents of xylose, glucose and galactose in monosaccharide composition. FT-IR and NMR spectroscopy analysis revealed that PSP has both α and β glycosidic bonds and massive acetyl groups, which is different from LP-F mainly composed of 1, 3 linked α-D-Man p residue with some acetyl groups. The findings provided a reliable approach for the development of antioxidant polysaccharide from spent Lentinus edodes substrate.

Quantification of Neoagaro-Oligosaccharide Production through Enzymatic Hydrolysis and Its Anti-Oxidant Activities

Shu-Ying Xu,Jie Kan,Zhong Hu,Yang Liu,Hong Du,Guang-Chang Pang,Kit-Leong Cheong

Neoagaro-oligosaccharides (NAOS) have health benefits that are related to their amount and degree of polymerization (DP). However, the current methods that are used to quantify enzymatically released NAOS are un-specific and time-consuming. Agar has been extracted fromGelidium amansiiand has been degraded by AgaXa (a recombinant β-agarase). Polysaccharide analysis using carbohydrate gel electrophoresis (PACE) has been adapted in order to quantify NAOS. In addition, the anti-oxidant activity of the degraded samples has been assessed. We have found that the PACE method provided sensitive, precise, and accurate quantification for each of the six NAOS samples. PACE has revealed that the DP of the enzymatic products from the AgaXa digestion were mainly neoagaro-octaose and neoagaro-decaose. The degraded samples exhibited increased radical-scavenging activity towards 2,2-diphenyl-1-picrylhydrazyl and 2,2-azino-bis(3-ethylbenzothiazoline sulfonic acid) radicals. While the anti-oxidant activity may have been from NAOS activity and contributions from neoagaro-octaose and neoagaro-decaose. The adapted PACE method that has been presented here is promising for large sample analysis during quality control and for characterizing novel β-agarase degradation mechanisms. Keywords:Gelidium amansiiagar;neoagaro-oligosaccharides;β-agarase;anti-oxidant

Structural characterization, in vitro and in vivo antioxidant activities of a heteropolysaccharide from the fruiting bodies of Morchella esculenta

Zheng-Nan Cai,Wan Li,Shomaila Mehmood,Wen-Juan Pan,Ya Wang,Fan-Ju Meng,Xiao-Fang Wang,Yong-Ming Lu,Yan Chen

This study aimed to investigate the structural features, in vitro and in vivo antioxidant activities of a heteropolysaccharide from the fruiting bodies of Morchella esculenta (FMP-1). FMP-1 had an average molecular weight of 4.7 × 10 3  Da and consisted of mannose, glucose and galactose. By methylation and NMR analysis, the backbone of FMP-1 was deduced to be made up of 1,4-linked Glc p and 1,6-linked Gal p . Hydroxyl, DPPH and superoxide radicals could be efficiently scavenged by FMP-1, with IC 50 values of 74.26, 119.32 and 161.49 μg/mL, respectively. Furthermore, FMP-1 could significantly protect zebrafish embryos against AAPH-induced oxidative damage. Decrease in malformations and mortalities was observed along with the reduction of ROS production, NO production and cell death. The protective effects were by decreasing MDA content and increasing SOD, CAT and GSH-Px levels. The current work provided a good suggestion of the potential utilization of FMP-1 as an attractive natural antioxidant.

Fermentation and complex enzyme hydrolysis for improving the total soluble phenolic contents, flavonoid aglycones contents and bio-activities of guava leaves tea

Lu Wang,You Luo,Yanan Wu,Yan Liu,Zhenqiang Wu

There are both soluble and insoluble-bound forms of phenolics in tea-leaf products. In order to increase total soluble phenolics contents, guava leaves tea (GLT) was first fermented with Monascus anka and Saccharomyces cerevisiae , and then hydrolyzed with complex enzymes . The changes in phenolics profiles, antioxidant activities and inhibitory effect on α -glucosidase in processed GLT were investigated. Compared with the un-fermented GLT, fermentation and complex enzymatic processing (FE) significantly increased the total phenolics, total flavonoids, quercetin and kaempferol contents by 2.1, 2.0, 13.0 and 6.8 times, respectively. After the FE, a major proportion of phenolics existed in the soluble form. Quercetin was released in the highest amount among different phenolics. In addition, soluble phenolic extracts from GLT following FE exhibited a highest antioxidant activity and inhibitory effect on α -glucosidase. The paper suggested an improved method for processing GLT into high-value products rich in phenolics and flavonoids aglycones with enhanced health benefits.

Edible coating based on whey protein isolate nanofibrils for antioxidation and inhibition of product browning

Zhibiao Feng,Guangxin Wu,Chunhong Liu,Dongmei Li,Bin Jiang,Xiaosong Zhang

Whey protein isolates (WPI) have been studied for film formation because of their favorable functional properties and industrial surplus; however, little is known regarding whey protein nanofibrils (WPNFs) as raw material for edible films . In the present study, WPNF-based edible coatings plasticized with glycerol (Gly) and trehalose (Tre) were studied. WPI (5%) can self-assemble into nanofibrils at 80 °C for 10 h under 220 rpm constant magnetic stirring. Compared with that of WPI, the hydrophobic and antioxidant activities of WPNFs were significantly increased. The microstructures and functions of WPI/Gly, WPNF/Gly, and WPNF/Gly/Tre films were characterized by scanning electron microscopy and atomic force microscopy , and by analyzing their contact angles, film thickness, transparency, moisture content, and solubility in water. Results showed that the formation of WPNFs increased surface smoothness, homogeneity, continuity, hydrophobicity, and transparency, and decreased the moisture content and water solubility of the films. To evaluate the performance of the edible coating, coated fresh cut apples were analyzed during storage. We found that the coatings containing WPNF (5%, w/v), Gly (4%, w/v) and Tre (3%, w/v) afforded the best protective action toward retarding the total phenolic content, browning, and product weight loss. Overall, the low cost, high biocompatibility, and consumer acceptance in taste tests support the ready application of these optimized WPNF films for commercial purposes.

The similarity and variability of the iridoid glycoside profile and antioxidant capacity of aerial and underground parts of Lamiophlomis rotata according to UPLC-TOF-MS and multivariate analyses

Dan Zhang,Yun-ling Gao,Sheng Jiang,Yiwen Chen,Yi Zhang,Zheng Pan

Lamiophlomis rotata (L. rotata) is a Tibetan medicinal herb used for centuries that contains iridoid glycosides (IGs), which are pharmacologically active ingredients and can be used for quality control. The IG profiles of the underground and aerial parts of the plant were determined by UPLC-TOF-MS to evaluate the similarity and variability of the different herbal parts listed in the Chinese Pharmacopoeia. Twenty-six IGs were detected in the total ion current (TIC) profile of L. rotata, and twenty-two of these were identified by comparing the retention times and mass spectra of the compounds to those of authentic standards. Among these compounds, five IGs with the same molecular formula (C17H26O11) were identified for the first time by mass spectrometry based on their different hydroxyl group-substituted positions. The aerial part has a similar chemical profile to that of the roots. The difference between the two parts was determined by multivariate statistical analysis of the UPLC-TOF-MS data of 24 specimens. Sesamoside was explored as the most characteristic marker to distinguish the two parts of L. rotata. To further estimate the distinction between the two parts, the content of total IGs and the antioxidant capacity were investigated in samples from different locations. The aerial parts showed a high content of total IGs and high antioxidant capacity, although not higher than those of the roots. The results also suggest the dosage should be increased when the aerial parts are used as crude medicinal materials instead of the underground parts.

Effects of metal ions in tea polysaccharides on their in vitro antioxidant activity and hypoglycemic activity

Minghao Fan,Xun Sun,Yilin Qian,Ying Xu,Dongfeng Wang,Yanping Cao

Total tea polysaccharides (TTPS) were extracted from two kinds of pruning leaves of tea plant and fractionated into neutral tea polysaccharides (TPSI) and acidic tea polysaccharides (TPSII) by anion exchange resin D315. Some physicochemical properties, including structure, monosaccharide composition, and molecular weight distribution, as well as the 4 in vitro antioxidant activities and inhibitory effects on α-glucosidase of above polysaccharides before and after removing metal ions were investigated. By comparing TTPS and TPSII, we found that they exhibited different antioxidant activities and inhibitory actions against α-glucosidase after their metal ions were removed. However, the in vitro antioxidant activities and inhibitory effects on α-glucosidase of TPSI were substantially improved. The study can be a certain reference for tea and soil selection. At the same time, we suggested that pruning leaves of tea plant could be treated as a potential resource for the development of polysaccharide antioxidants and hypoglycemic products.

Seasonal variations of phenolic profiles and antioxidant activity of walnut (Juglans sigillata Dode) green husks

Binbin Shi,Wene Zhang,Xue Li,Xuejun Pan

The phenolics content and antioxidant activity of walnut (Juglans sigillata Dode) green husks were analysed during different stages of development. Contents of total phenolics and total flavonoids decreased from April 25th to June 7th, then increased during June and July, and decreased again at the beginning of August. Thirteen types of phenolic compounds were identified using HPLC of which juglone had the highest content. Because of seasonal variations, the trends of different phenolic compounds were different. Contents of both total phenolics and total flavonoids were strongly correlated with antioxidant activity; however, compared with totals, less significant correlations were observed between individual phenolic compounds and antioxidant activities.

Investigation of inhibition of lipid oxidation by L-carnosine using an oxidized-myoglobin-mediated washed fish muscle system

Shulan Xiao,Hong Zhuang,Guanghong Zhou,Jianhao Zhang

The objective of the present study was to understand mechanisms of inhibitory effect of l -carnosine on lipid oxidation using an oxidized myoglobin (metMb)-mediated washed cod muscle system. Experiments were conducted to compare l -carnosine with commercial antioxidants, tert-Butylhydroquinone (TBHQ) and α-tocopherol, and metal chelating reagent EDTA for inhibition of lipid oxidation, to compare l -carnosine with TBHQ for scavenging free radicals and to evaluate the effect of l -carnosine on structure of pro-oxidant metMb with spectral methods. Results showed that the inhibitory effect of l -carnosine on lipid oxidation was comparable with TBHQ and better than α-tocopherol. However, 1,1-diphenyl-2-picrylhydrazyl assay showed that TBHQ was much more effective in scavenging free radicals than l -carnosine. Comparing with 1.5% EDTA, 1.5% l -carnosine was much more effective against lipid oxidation. Incubation of l -carnosine with metMb resulted in spectral changes of metMb in wavelengths between 370 nm and 450 nm, Soret band shifted from 409 nm to 420 nm, and reduced molar ellepticity values. These results indicate that the high inhibitory effects of l -carnosine on meat lipid oxidation may result from not only its scavenging and chelating functions but also its effect on pro-oxidant metMb structure.

Quantification of polyphenol composition and multiple statistical analyses of biological activity in Portuguese red wines

Bai Shuang,Cui Can,Liu Jing,Li Pei,Li Qing,Bi Kaishun

The objective of the research was to evaluate potential biological activities of Portuguese red wines with different grape types on the level of composition characterization. An ultrafast liquid chromatography coupled with triple quadrupole mass spectrometry method was validated and used for qualitative analysis of commercial red wine samples through simultaneously determining 31 major active components. Meanwhile, the antioxidant activities of red wines were measured by DPPH (1,1-diphenyl-2-picrylhydrazyl), ABTS [2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid)] and CUPRAC (cupric ion reducing antioxidant capacity) assay. The relations between polyphenols and antioxidant values were further considered by multivariate statistical analysis. Results of correlation analysis and stepwise multiple regression analysis showed that antioxidant activities were highly correlated to polyphenolic profiles, with significant contributions of procyanidins and phenolic acids. Consequently, the assessment method we established appeared to be an effective tool for appraising the nutritional value of polyphenols in wines, and the research strategy could be applied to the evaluation of other beverages.
