Arsenic Bioaccessibility in Rice and Its Application to Derive Health-Based Limits in China

Di Zhao

Arsenic (As) contamination in rice is a global public health concern, particularly in Asian countries where rice is the staple food. Current health-based limits for As in rice are typically derived from total As concentrations, resulting in overly stringent values. This study aimed to determine As bioaccessibility in rice, estimate dietary intakes of inorganic As (iAs) at different consumption rates, evaluate the cancer and non-cancer risks associated with iAs exposure through rice consumption, and assess the feasibility of deriving more accurate health-based limits for As in rice after incorporating rice As bioaccessibility. Bioaccessibility of As ranged from 60.7% to 104.5% in rice samples. Estimated iAs intake varied from 0.04 to 1.40 μg/kg BW/day at rice consumption rates of 100–300 g/day. Incorporating rice As bioaccessibility resulted in lower iAs intake estimates of 0.03–1.18 μg/kg BW/day. The non-cancer and cancer risks associated with iAs exposure are concerning for populations with higher rice consumption rates and elevated rice iAs concentrations. Health-based limits for iAs in rice for different regions across China are discussed after incorporating rice As bioaccessibility. This study contributes to the development of regional or national safety limits for As in rice, based on As bioaccessibility in rice.

Andrias davidianus bone peptides alleviates hyperuricemia-induced kidney damage in vitro and in vivo

Wei Li,Haihong Chen,Hongliang Chen,Ziyin Li,Wei Hu,Qinxuan Zhou,Bing Xu,Yi Wang,XinHui Xing

Hyperuricemia (HUA) is a vital risk factor for chronic kidney diseases (CKD) and development of functional foods capable of protecting CKD is of importance. This paper aimed to explore the amelioration effects and mechanism of Andrias davidianus bone peptides (ADBP) on HUA-induced kidney damage. In the present study, we generated the standard ADBP which contained high hydrophobic amino acid and low molecular peptide contents. In vitro results found that ADBP protected uric acid (UA)-induced HK-2 cells from damage by modulating urate transporters and antioxidant defense. In vivo results indicated that ADBP effectively ameliorated renal injury in HUA-induced CKD mice, evidenced by a remarkable decrease in serum UA, creatinine and blood urea nitrogen, improving kidney UA excretion, antioxidant defense and histological kidney deterioration. Metabolomic analysis highlighted 14 metabolites that could be selected as potential biomarkers and attributed to the amelioration effects of ADBP on CKD mice kidney dysfunction. Intriguingly, ADBP restored the gut microbiome homeostasis in CKD mice, especially with respect to the elevated helpful microbial abundance, and the decreased harmful bacterial abundance. This study demonstrated that ADBP displayed great nephroprotective effects, and has great promise as a food or functional food ingredient for the prevention and treatment of HUA-induced CKD.

Antidepressant advantage of Chaihushugan san in female mice: A novel signaling mechanism in hippocampus

Chao Lu,Lingang Zhao,Liyuan Tian,Chenguang Lin,Lei Wu

Ethnopharmacological relevancy Chaihushugan san (CSS), a classic formula for soothing the liver and relieving depression, has been identified to produce rapid antidepressant-like effects in female mice. However, the gender predominance and underlying mechanisms of CSS's antidepressant remain unclear. Aim of the study In this study, we focused on unraveling the gender predominance of CSS in antidepressant and the specific neuronal mechanisms that mediate this predominance. Methods and materials Tail suspension test (TST), forced swimming test (FST) and sucrose preference test (SPT) were used to evaluate depressive phenotypes or antidepressant-like effects of CSS in female and male chronic unpredictable mild stress (CUMS) mice model. RNA-sequencing was used to screen specific target for CSS antidepressant gender dominance. RT-PCR and elisa were used to detect the expressions of specific molecule, hormones, and inflammatory factors in the hippocampus. hippocampal viral overactivation and pharmacological blockade were used to detect the correlation between CSS antidepressant gender dominance and related targets. Results In the present study, both female and male mice displayed depressive phenotypes including significant increasing immobility time in TST and reducing sucrose preference ratio in SPT after exposing CUMS for 3 weeks. However, acute administration of CSS (2, 4 g/kg) improved the depressive phenotypes only in female mice or not male mice at 2 h later. Moreover, the expressions of TC2N were increased only in female mice after exposing CUMS for 3 weeks, which were also reversed by CSS after a single administration 2 h later, but no alterations in male mice. The hippocampal expressions of estrogen receptor β (Erβ), pro-inflammatory factors (IL-1β and TNF-α) and anti-inflammatory factors (IL-10, TGF-β and IL-1Rα) were all abnormal in female CUMS mice model, which were all normalized by CSS. Furthermore, overactivation of hippocampal TC2N by AAV-TC2N +/+ blocked the antidepressant-like effects of CSS and the up-regulation of hippocampal Erβ in female mice. However, inhibition of Erβ blunted the antidepressant-like effects of CSS and CSS's suppression of pro-inflammatory factors (IL-1β and TNF-α), which had no any effect on hippocampal TC2N and anti-inflammatory factors (IL-10 and TGF-β). Conclusions The study revealed that CSS had antidepressant superiority in female mice depending on inhibiting hippocampal TC2N and then activating Erβ, further inhibiting the release of pro-inflammatory factors to produce antidepressant effects, which provided a basis for the guidance of CSS in clinical application, new ideas and targets for the development of drugs for depression with gender differences.

The matrix effect derived from proteins in fluorescence detection for ciprofloxacin residues in eggs

Kai Wang,Hui Li,Wenya Liu,Pan Liu,Chuang Jiang,Wanqi Jiang,Yaqing Xiao,Yingnan Liu

Based on the previously developed Al 3+ /CuNCs probe, the matrix effect (ME) derived from protein was investigated in the fluorescence determination of fluoroquinolones (FQs) residues in eggs, and a sample pretreatment method was established to rapidly reduce the ME. Molecular docking results indicated that the three main egg proteins and FQs were bound by hydrogen bond, van der Waals force, and alkyl groups, leading to the loss of FQs response signal, and then produced ME. After trichloroacetic acid treatment, the mean particle size of egg matrix decreased, the protein secondary structure changed, and the crystal structure was destroyed. The corresponding ME of the supernatant obtained by centrifugation was only 0.7 %. The standard addition experiment confirmed that the proposed pretreatment method improved the accuracy and reliability of the fluorescence probe. This work is helpful to promote the practical application of fluorescence analysis in the rapid monitoring of food safety hazard factors.

The role of exosomal lncRNAs in acetaminophen-induced induced liver injury in SD rats

Zixuan Yang,Lei Shi,Minhui Zheng,Minbo Hou,Mengdi Zhou,Naying Su,Hui Lang,Liyuan Zhao,Mengyun Gu,Naping Tang,Yan Chang

Background Drug-induced liver injury (DILI) is a leading cause of drug development failures during clinical trials and post-market introduction. Current biomarkers, such as ALT and AST, lack the necessary specificity and sensitivity needed for accurate detection. Exosomes, which protect LncRNAs from RNase degradation, could provide reliable and easily accessible options for biomarkers. Materials and methods RNA-sequencing was used to identify differentially expressed LncRNAs (DE-LncRNAs), followed by isolation of LncRNAs from plasma exosomes in this study. Exosome characterization was conducted by transmission electron microscopy (TEM), nanoparticle tracking analysis (NTA), and Western blot (WB). Bioinformatics analysis included functional enrichment and co-expression network analysis. Five rat models were established, and quantitative real-time PCR was used to verify the specificity and sensitivity of two candidate exosomal LncRNAs. Results The APAP-induced hepatocellular injury model was successfully established for RNA-sequencing, leading to the identification of several differentially expressed exosomal LncRNAs. Eight upregulated exosomal DE-LncRNAs were selected for validation. Among them, NONRATT018001.2 (p < 0.05) and MSTRG.73954.4 (p < 0.05) exhibited a more than 2-fold increase in expression levels. In hepatocellular injury and intrahepatic cholestasis models, both NONRATT018001.2 and MSTRG.73954.4 showed earlier increases compared to serum biomarkers ALT and AST. However, no histological changes were observed until the final time point. In the fatty liver model, NONRATT018001.2 and MSTRG.73954.4 increased earlier than ALT and AST at 21 days. By the 7th day, minor steatosis was evident in liver tissue, while the expression levels of the two candidate exosomal LncRNAs exceeded 2 and 4 times, respectively. In the hepatic fibrosis model, NONRATT018001.2 and MSTRG.73954.4 showed increases at every time point. By the 49th day, hepatocellular necrosis and fibrosis were observed in the liver tissue, with NONRATT018001.2 showing an increase of more than 8 times. The specificity of the identified exosomal DE-LncRNAs was verified using a myocardial injury model and they showed no significant differences between the case and control groups. Conclusion NONRATT018001.2 and MSTRG.73954.4 hold potential as biomarkers for distinguishing different types of organ injury induced by drugs, particularly enabling early prediction of liver injury. Further experiments, such as siRNA interference or gene knockout, are warranted to explore the underlying mechanisms of these LncRNAs.

Passivating defects in SnO2 electron transport layer through SnF2 incorporation in perovskite solar cells

Fengxuan Chen,Xin Hu,Longhao Jisi,Liping Su,Huiyao Zhao,Yanbei Wei,Rui Zhou,Yangdi Chen,Jun Qu,Yonglian Xiong,Mao Liang,Wenfeng Zhang

Metal halide perovskite solar cells have received a lot of attention from researchers because of their excellent performance, and their efficiency has increased rapidly over the past decade and is now comparable to that of conventional crystalline silicon solar cells. Perovskite solar cells (PSCs) are currently most commonly constructed with SnO 2 as an electron transport layer (ETL). Nevertheless, its existence of bulk phase defects and surface defects pose an impediment to developing a higher level as the most favorable electron transport layer. This paper suggests an efficient technique to decrease the defects of SnO 2 through incorporating a metal halide SnF 2 in planar (n-i-p type) perovskite solar cells. Divalent Sn in SnF 2 is easily oxidized to tetravalent Sn, leading to more conversion of divalent Sn to tetravalent Sn, which inhibits the formation of oxygen vacancies from the source. F replaces defective sites, passivates oxygen vacancy defects, and reduces the content of hydroxyl oxygen on the surface. Doping SnF 2 enhances the conduction band value and conductivity of SnO 2 , making the energy levels of the ETL and perovskite interfaces more compatible. This eliminates energy barriers in charge transport. Consequently, the fill factor of planar PSCs that were prepared through SnO 2 :SnF 2 ETL exhibited an increase from 80.01% to 82.12%. Furthermore, the efficiency of PCE underwent an improvement from 21.43% to 22.65%.

Sweet taste receptors play roles in artificial sweetener-induced enhanced urine output in mice

Cai Shuangfeng,Xie Ningning,Zheng Ling,Li Quan,Zhang Siyu,Huang Qinghua,Luo Wei,Wu Mei,Wang Yidan,Du Yilun,Deng Shao-ping,Cai Lei

Sweet taste receptors found in oral and extra oral tissues play important roles in the regulation of many physiological functions. Studies have shown that urine volume increases during the lifetime exposure to artificial sweeteners. However, the detailed molecular mechanism and the general effects of different artificial sweeteners exposure on urine volume remain unclear. In this study, we investigated the relationship between urinary excretion and the sweet taste receptor expression in mice after three artificial sweeteners exposure in a higher or lower concentration via animal behavioral studies, western blotting, and real-time quantitative PCR experiment in rodent model. Our results showed that high dose of acesulfame potassium and saccharin can significantly enhance the urine output and there was a positive correlation between K + and urination volume. The acesulfame potassium administration assay of T1R3 knockout mice showed that artificial sweeteners may affect the urine output directly through the sweet taste signaling pathway. The expression of T1R3 encoding gene can be up-regulated specifically in bladder but not in kidney or other organs we tested. Through our study, the sweet taste receptors, distributing in many tissues as bladder, were indicated to function in the enhanced urine output. Different effects of long-term exposure to the three artificial sweeteners were shown and acesulfame potassium increased urine output even at a very low concentration.

Study of the liquid resistance effect of water-in-oil emulsions in porous media

Lei-Lei Jia,Li-Guo Zhong,Shi-Hao Li,Yu-Hao Liu,Chang-Hao Hu,Guo-Dong Wang,Yu-Ning Gong,Ce Shang,Xiao-Cheng Zhang,Yao-Tu Han,Jin Li

During heavy oil recovery in the Bohai Oilfield, substantial emulsification of oil and water occurred, primarily forming water-in-oil emulsions. This phenomenon could alter fluid dynamics within the subsurface porous media, potentially impacting well production performance. To elucidate the properties of water-in-oil emulsions and their associated liquid resistance effects, this study conducted a series of rheological tests, microscopic examinations, and injection experiments. The results show that the droplet size and distribution of water-in-oil emulsions were primarily influenced by shear rate and water content, which in turn modified emulsion viscosity. The stability of water-in-oil emulsions was reduced when they flowed through porous media. The increase in emulsion viscosity and the liquid resistance effect collectively enhanced the seepage resistance of water-in-oil emulsions flowing through porous media. Notably, when the emulsion droplet size exceeded the pore throat size, over 90% of the total seepage resistance was attributable to the liquid resistance effect. Conversely, when the majority of the emulsion droplets were smaller than the pore throat, the viscosity accounted for more than 60% of the seepage resistance. Water-in-oil emulsions flowed through cores with permeabilities ranging from 50 to 100 mD, exhibiting threshold pressure gradients between 35 and 43 MPa/m. At a core permeability of 300 mD, the threshold pressure gradient was significantly reduced to 1 MPa/m. The presence of a water-in-oil emulsion in the reservoir could result in a production pressure differential falling below the threshold pressure, thereby reducing reservoir productivity.

Rapid and In-Field Sensing of Hydrogen Peroxide in Plant by Hydrogel Microneedle Patch

Xinyue Wu,Yuxiang Pan,Xunjia Li,Yuzhou Shao,Bo Peng,Chao Zhang,Chi Zhang,Shiyun Yao,Jianfeng Ping,Yibin Ying

The rapidly changing climate is exacerbating the environmental stress that negatively impacts crop health and yield. Timely sensing of plant response to stress is beneficial to timely adjust planting conditions, promoting the healthy growth of plants, and improving plant productivity. Hydrogen peroxide (H 2 O 2 ) is an important molecule of signal transduction in plants. However, the common methods for detecting H 2 O 2 in plants are associated with certain drawbacks, such as long extraction time, cumbersome steps, dependence on large instruments, and difficulty in realizing in-field sensing. Therefore, it is urgent to establish more efficient detection methods to realize the rapid detection of H 2 O 2 content in plants. In this research, poly (methyl vinyl ether- alt -maleic acid) (PMVE/MA) hydrogel microneedle (MN) patch for rapid extraction of leaf sap are prepared, and the extraction mechanism of PEG-crosslinked PMVE/MA hydrogel MN patch is studied. A method of rapid detection of H 2 O 2 content in plants based on MN patch with optical detection technology is constructed. The hydrogel MN patch can be used for timely H 2 O 2 analysis. This application enables new opportunities in plant engineering, and can be extended to the safety and health monitoring of other plants and animals.

Superior strength-ductility combination in TiC/TC4 composites via In situ construction Ti2Cu nanoparticles

Xiaoqi Mao,Longlong Dong,Yiya Zhang,Shuanglin Qin,Guodong Sun,Lianwen Wang,Yusheng Zhang

Synergistic improvement of strength and ductility is challenging in the engineering applications of titanium matrix composites (TMCs). Herein, we fabricated the TC4 composites reinforced with grain boundary micron-TiC and intragranular nano-Ti 2 Cu using spark plasma sintering (SPS) and hot rolling (HR) to achieve a good balance between strength and ductility. An interfacial reaction between Ti and C forms the grain boundary TiC, and the vast majority of intragranular nano-Ti 2 Cu precipitates to accommodate the severe lattice distortion caused by the solid solution of Cu in β-Ti phase. Benefiting from Ti 2 Cu nanoprecipitates and their ability to effectively suppress dislocation motion , precipitation strengthening, and dislocation strengthening are the main factors, in addition to grain refinement strengthening due to deformation processing , that contribute to the realization of the superior yield strength (YS) of 1360 MPa and ultimate tensile strength (UTS) of 1504 MPa in composites than TC4 matrix (YS = 986 MPa, UTS = 1160 MPa). Meanwhile, the interfacial strain-delocalization effect of TiC/Ti 2 Cu with Ti matrix, the shear-lag effect of TiC, and the sustained strain-hardening effect provided by TiC/Ti 2 Cu empower the composites with an outstanding ductility (fracture elongation of 8.4 %). This study highlights the untapped potential for improving the mechanical properties of Ti matrix composites.

Optimal porosity in zeolitic imidazole framework for cost-effective furfural removal in biomass hydrolysates

Kanghong Wang,Chaozhong Xu,Wei Xiong,Shanshan Tong,Jia Ouyang,Xiaoli Gu

Efficient removal of fermentation inhibitors, particularly furan aldehydes like furfural is crucial for enhancing microbial fermentation efficiency in lignocellulosic biomass processing. In this study, zeolitic imidazolate framework-8 with different molar ratios were synthesized by regulating the Zn 2 + /2-methylimidazole molar ratio (from 1:4–1:15), and their structural properties and adsorption performance were systematically evaluated. The results showed that the pore structure was crucial to the adsorption performance, and the pore structure was optimal when the Zn 2+ /2-methylimidazole ratio was 1:8. The optimized zeolitic imidazole framework-8 showed rapid, selective and efficient adsorption of furfural under different operating conditions, thereby preventing the formation of inhibitors from affecting fermentation while minimizing sugar loss. This study provides a generalizable design principle for the development of adsorbents in biomass conversion. This work provides a scalable solution for biomass hydrolysate detoxification, with potential cost advantages due to the material’s reusability and high adsorption capacity compared to conventional resin-based methods.

Region-Specific Metabolic Alterations in the Tryptophan Pathway in Cisplatin-Induced Acute Kidney Injury

Yue Feng,Ying Zhang,Jinxin Li,Junzhe Yu,Yuan Tian,Zunjian Zhang,Fengguo Xu,Pei Zhang

Cisplatin (DDP) is widely utilized in the clinical treatment of malignant tumors, but its effectiveness is significantly compromised by the adverse effects of acute kidney injury (AKI). Renal tubular cells are primarily responsible for DDP-induced AKI (DDP-AKI); however, the responses of heterogeneous renal tubular cells to DDP exposure have not been thoroughly explored. In this study, we employed a targeted metabolomics approach to investigate the metabolic responses of renal tubular cells in DDP-AKI rats. Tubular cells were isolated from the renal cortex and outer medulla, and a chemical derivatization-based liquid chromatography–tandem mass spectrometry (LC–MS/MS) metabolomics method was applied. Our findings revealed distinct metabolic profiles in tubular cells from the renal cortex and outer medulla, with outer medullary cells exhibiting greater sensitivity to DDP exposure. Further analyses identified the tryptophan pathway as a critical factor contributing to these regional differences. Additional functional investigations showed that intermediate metabolites of the tryptophan pathway alleviated DDP cytotoxicity in both cortical and outer medullary tubular cells primarily through modulation of the Bcl2/Bax and Caspase-3 pathway. This study enhances our understanding of the metabolic characteristics of tubular cells across heterogeneous renal regions in DDP-AKI and facilitates further exploration of the underlying mechanisms of DDP-induced nephrotoxicity.
