Near-infrared fluorescent probes for non-invasive, real-time, and dual-color in vivo NIR-II imaging

Huixian Jia,Yujie Liu,Wei Tang,Chenghui Liu,Xinrui Duan

Multiplexed fluorescence imaging is mainly applied to cellular imaging. However, the application of an analogous technique to mammals remains challenging due to the presence of substantial scattering and autofluorescence in biological tissues in the visible (350–700 nm) and near-infrared I region (NIR-I, 700–900 nm). Herein, we report multiplexing excitation and single-channel detection in the near-infrared II region (NIR-II) with two fluorescent probes for dual-color imaging in vivo. The first one, FNIR-1090 was constructed on Pr-1099 derivative by attaching with 1-Ethylpiperazine through the SRN1 displacement reaction. The second one, IR-765 was synthesized by a one-step reaction between the Vilsmeier–Haack reagent with 2-(3,5,5-trimethylcyclohex-2-enylidene) malononitrile (TEMO). It is noteworthy that the absorption spectrum of FNIR-1090 is almost completely separated from that of IR-765, which facilitates dual-color in vivo imaging. The two dyes can be used for high-contrast dual-channel vivo imaging of different tissues. All these prominent performances show the great promise of FNIR-1090 and IR-765 in non-invasive, real-time, and dual-color NIR-II imaging (>900 nm) in vivo.

Bioheterojunction-Engineered System: A New Mechanism of BP/MoS2 Photothermal-Photocatalytic Dual-Power Synergistic Sterilization under NIR

Shuyan Zhang,Yueqi Pan,Yihan Chen,Zhiwei Yang,Dongcai He,Nan Feng,Weijie Wang,Yunfeng Li,Danna Chen,Xianchun Chen,Jing Zhang

Graphical BP/MoS 2 heterojunction is prepared by solvothermal method using NMP, BP nanosheets and (NH 4 ) 2 MoS 4 . Under NIR irradiation, BP/MoS 2 can be heated up to 51.8 °C within 5 minutes by photothermal conversion. In addition, the existence of ⋅O 2 − and 1 O 2 in BP/MoS 2 -NIR system is verified to sterilize efficiently. With a loading content of 10 wt % MoS 2 , bacterial cells, especially drug-resistant bacteria (10 6 cfu ⋅ mL −1 ) completely can lose the viability within 10 min of NIR irradiation. In this research, a high-efficiency nano-fungicide with synergistic effect was obtained by combining polycrystalline MoS 2 with layered nano-BP to form heterojunction, which had a new photothermal-photocatalytic antibacterial mechanism to Gram-positive bacteria, Gram-negative and drug-resistance bacteria under NIR. BP/MoS 2 whose lethality of bacteria could reach nearly 100 % at 400 μg/L, exhibited the highest photothermal lysis efficiency against E. coli, B. pumilus and A. baumannii. It was found that the heterojunction could be heated up to 51.8 °C within 5 minutes by photothermal conversion, and it also had an excellent performance of photothermal cycle. High temperature and stability of the material endowed it with an outstanding and long-lasting bactericidal effect. In addition, a series of in vitro free radical experiments was designed, the existence of ⋅O 2 − and 1 O 2 in BP/MoS 2 -NIR system was verified. And the Way of ROS production and the mechanism of action were discussed by EPR test. Subsequently, two mechanisms of synergistic antibacterial action were summarized, that is, abundant ROS and high temperature could destroy cell wall damage and lysis of bacteria effectively. This study provides a theoretical basis for the synergistic antibacterial action of NIR photothermal and photocatalysis of BP system.

Wan-Ni Wang,Chen-Xi Huang,Chen-Yang Zhang,Meng-Li Zhao,Jun Zhang,Hua-Jian Chen,Zheng-Bao Zha,Tingting Zhao,Hai-Sheng Qian

Photocatalysts driven by near infrared light (NIR) are of great importance for making full use of solar energy and wide applications in biomedicine, biosensor, photocatalysis, and etc. In this work, we demonstrate a facile template assisted hydrothermal approach to fabricate upconversion nanoparticles (UCNPs) and Zn x Cd 1-x S yolk-shell nanoparticles, although the yolk-shell nanoparticles were made from hexagonal phase of β-NaYF 4 (NaYF 4 :Yb/Tm@NaYF 4 :Yb/Er) and II–VI semiconductor materials with a large lattice mismatch of more than 10%. The growth mechanism of the formation of yolk-shell nanostructures has been extensive investigated and the shape evolution with the time has been demonstrated. Steady and dynamic fluorescence spectra show that the as-prepared yolk-shell nanoparticles of UCNPs/Zn x Cd 1-x S can be used as efficient nanotransducer for NIR light owing to enhanced energy transfer efficiency. Interesting, the as-prepared yolk-shell nanoparticles exhibit excellent photocatalytic performance on degradation towards RhB dyes, excellent biocompatible properties and high production ability for hydroxyl radical ( OH) and single oxygen ( 1 O 2 ), which might be employed to wide applications in solar cell, hydrogen production, and photodynamic therapy related nano-biotechnology.

pH/NIR-responsive and self-healing coatings with bacteria killing, osteogenesis, and angiogenesis performances on magnesium alloy

Yanbin Zhao,Peng He,Junyan Yao,Mei Li,Bin Wang,Linyuan Han,Zhihai Huang,Chao Guo,Jing Bai,Feng Xue,Yu Cong,Weihua Cai,Paul K. Chu,Chenglin Chu

Although biodegradable polymer coatings can impede corrosion of magnesium (Mg)-based orthopedic implants , they are prone to excessive degradation and accidental scratching in practice. Bone implant-related infection and limited osteointegration are other factors that adversely impact clinical application of Mg-based biomedical implants. Herein, a self-healing polymeric coating is constructed on the Mg alloy together with incorporation of a stimuli-responsive drug delivery nanoplatform by a spin-spray layer-by-layer (SSLbL) assembly technique. The nanocontainers are based on simvastatin (SIM)-encapsulated hollow mesoporous silica nanoparticles (S@HMSs) modified with polydopamine (PDA) and polycaprolactone diacrylate (PCL-DA) bilayer . Owing to the dynamic reversible reactions, the hybrid coating shows a fast, stable, and cyclical water-enabled self-healing capacity. The antibacterial assay indicates good bacteria-killing properties under near infrared (NIR) irradiation due to synergistic effects of hyperthermia, reactive oxygens species (ROS), and SIM leaching. In vitro results demonstrate that NIR laser irradiation promotes the cytocompatibility, osteogenesis, and angiogenesis . The coating facilitates alkaline phosphatase activity and expedites extracellular matrix mineralization as well as expression of osteogenesis-related genes. This study reveals a useful strategy to develop multifunctional coatings on bioabsorbable Mg alloys for orthopedic implants .

NIR-dependent photothermal-photodynamic synergistic antibacterial mechanism for titanium carbide nanosheets intercalated and delaminated by tetramethylammonium hydroxide

Jun Wang,Jinnan Xuan,Yisi Liu,Zhong Li,Yuqiang Han,Zhen Wang

The infectious disease epidemics caused by pathogens are a serious and growing worldwide public health problem. More seriously, the multiple resistant bacteria extensively spread in hospitals or communities due to antibiotic abuse. In this paper, we fabricate two-dimensional Ti 3 C 2 nanosheets with excellent biocompatibility and photothermal-photodynamic synergistic antibacterial activity against E. coli and S. aureus based on the strategy of tetramethylammonium hydroxide (TMAOH)-driven intercalation and delamination. Compared with the traditional Ti 3 C 2 nanosheets exfoliated by HF or situ HF (HCl + LiF), the photothermal-photodynamic Ti 3 C 2 nanosheets show higher synergistic antibacterial efficiency. In addition, the antibacterial mechanism demonstrates that biofilm disruption and leakage of bacterial contents contribute to reactive oxygen species reaction (ROS) and photothermal antibacterial activity irradiated by NIR after most Ti 3 C 2 nanosheets adhering to target bacteria. In conclusion, the Ti 3 C 2 nanosheets have great potential as an effective antibacterial material without causing drug resistance, relying on intercalating and delaminating by TMAOH.

NIR-II Responsive Nanohybrids Incorporating Thermosensitive Hydrogel as Sprayable Dressing for Multidrug-Resistant-Bacteria Infected Wound Management

Weilun Pan,Bodeng Wu,Chengtao Nie,Tingting Luo,Zhenli Song,Jie Lv,Yong Tan,Chunchen Liu,Mingzhen Zhong,Tong Liao,Zhenxun Wang,Guanghui Yi,Limin Zhang,Xiaoliu Liu,Bo Li,Jinxiang Chen,Lei Zheng

Developing an effective dressing against bacterial infection and synchronously addressing wound complications, such as bleeding, long-term inflammation, and reinfection, are highly desirable in clinical practice. In this work, a second near-infrared (NIR-II) responsive nanohybrid consisting of imipenem encapsulated liposome with gold-shell and lipopolysaccharide (LPS)-targeting aptamer, namely ILGA, is constructed for bacteria elimination. Benefiting from the delicate structure, ILGA exhibits strong affinity and a reliable photothermal/antibiotic therapeutic effect toward multidrug-resistant Pseudomonas aeruginosa (MDR-PA). Furthermore, by incorporating ILGA with a thermosensitive hydrogel poly(lactic-co-glycolic acid)–polyethylene glycol–poly(lactic-co-glycolic acid) (PLGA-PEG-PLGA), a sprayable dressing ILGA@Gel was prepared, which enables a quick on-demand gelation (10 s) for wound hemostasis and offers excellent photothermal/antibiotic efficacy to sterilize the infected wound. Additionally, ILGA@Gel provides satisfactory wound-healing environments by reeducating wound-associated macrophages for inflammation alleviation and forming a gel layer to block exogenous bacterial reinfection. This biomimetic hydrogel reveals excellent bacteria eradication and wound recovery effectiveness, demonstrating its promising potential for managing complicated infected wounds.

NIR-AIEgens nanospheres featuring high-fidelity dynamic lipid droplet targeting, expediting ferroptosis to annihilating tumor in hypoxia

Ke Xue,Yongfei Zhao,Saidong Sun,Yuanhang Li,Jiankang Liang,Zhengjian Qi

Severe ferroptosis is a non-apoptotic iron-dependent cell death pathway, and extreme hypoxia and anomalous iron metabolism in tumors critically suppress the efficacy of ferroptosis therapy. Overcoming tumor hypoxia while facilitating ferroptosis is an extremely challenging task. Herein, the photosensitizer named BOTTQ with specific targeting of lipid droplet (LD) was synthesized by increasing the tail chain length of triphenylamine. The addition of alkoxy chains optimized the molecular structure and fostered inter-system crossing and type I reactive oxygen species (ROS) generation. Besides, BOTTQ yielded large amounts of type I ROS under light excitation oxidize efficiently polyunsaturated fatty acids in lipid monolayer of LD and contribute to the formation of ferroptosis substrate. Meanwhile, the generation of type I ROS accelerated the Fenton reaction, thus effectively boosting the occurrences of ferroptosis. Furthermore, the successful encapsulation of BOTTQ into DSPE-PEG-MAL material not only elevates the stable type of nanoparticle colloids, but the MAL moiety can deplete the functional thiols of glutathione in a mild environment, which leads to glutathione failure and facilitates ferroptosis. Thus tumor-specific ferroptosis-inducing strategy offers a universally applicable therapy with the capacity to enhance type I ROS production and effectively contain tumorigenesis.

Microenvironment responsive nanocomposite hydrogel with NIR photothermal therapy, vascularization and anti-inflammation for diabetic infected wound healing

Shuangli Zhu,Bangjiao Zhao,Maocai Li,Hao Wang,Jiayi Zhu,Qingtao Li,Huichang Gao,Qi Feng,Xiaodong Cao

Bacterial infection, excessive inflammation and damaging blood vessels network are the major factors to delay the healing of diabetic ulcer. At present, most of wound repair materials are passive and can't response to the wound microenvironment, resulting in a low utilization of bioactive substances and hence a poor therapeutic effect. Therefore, it's essential to design an intelligent wound dressing responsive to the wound microenvironment to achieve the release of drugs on-demand on the basis of multifunctionality. In this work, metformin-laden CuPDA NPs composite hydrogel ( [email protected] CuPDA NPs/HG) was fabricated by dynamic phenylborate bonding of gelatin modified by dopamine (Gel-DA), Cu-loaded polydopamine nanoparticles (CuPDA NPs) with hyaluronic acid modified by phenyl boronate acid (HA-PBA), which possessed good injectability, self-healing, adhesive and DPPH scavenging performance. The slow release of metformin was achieved by the interaction with CuPDA NPs, boric groups (B–N coordination) and the constraint of hydrogel network. Metformin had a pH and glucose responsive release behavior to treat different wound microenvironment intelligently. Moreover, CuPDA NPs endowed the hydrogel excellent photothermal responsiveness to kill bacteria of >95% within 10 min and also the slow release of Cu 2+ to protect wound from infection for a long time. [email protected] CuPDA NPs/HG also recruited cells to a certain direction and promoted vascularization by releasing Cu 2+ . More importantly, [email protected] NPs/HG effectively decreased the inflammation by eliminating ROS and inhibiting the activation of NF-κB pathway. Animal experiments demonstrated that [email protected] NPs/HG significantly promoted wound healing of diabetic SD rats by killing bacteria, inhibiting inflammation, improving angiogenesis and accelerating the deposition of ECM and collagen. Therefore, [email protected] NPs/HG had a great application potential for diabetic wound healing.

Dual enzyme-like Co–FeSe2 nanoflowers with GSH degradation capability for NIR II-enhanced catalytic tumor therapy

Jingge Zhang,Enna Ha,Danyang Li,Shuqing He,Luyang Wang,Shaolong Kuang,Junqing Hu

Nanozymes mediated catalytic therapy can produce toxic reactive oxygen species (ROS) and destroy the metabolic balance of tumor cells, providing a new direction for cancer treatment. However, the catalytic efficiency of a single nanozyme is limited by the complexity of the tumor microenvironment (hypoxia, GSH overexpression, etc.). In order to overcome these problems, we designed flower-like Co-doped FeSe2 (Co–FeSe2) nanozymes by a simple wet chemistry method. Co–FeSe2 nanozymes not only exhibit high POD and OXD-mimicking activities for facile kinetics, but also effectively consume over-expressed glutathione (GSH), inhibiting the consumption of generated ROS and destroying the metabolic balance of the tumor microenvironment. These catalytic reactions trigger cell death through apoptosis and ferroptosis dual pathways. More importantly, under the NIR II laser irradiation, the catalytic activities of Co–FeSe2 nanozymes are boosted, confirming the photothermal and catalytic synergistic tumor therapy. This study takes advantage of self-cascading engineering that offers new ideas for designing efficient redox nanozymes and promoting their clinical translation.

NIR-light-responsive chemo-photothermal hydrogel system with controlled DOX release and photothermal effect for cancer therapy

Jun Yang,Zhen Sun,Qingqing Dou,Shuhan Hui,Peng Zhang,Rui Liu,Di Wang,Shan Jiang

Here a multiplex near-infrared (NIR) light-responsive hydrogel platform (SA/ [email protected] ) has been constructed for controlled synergetic chemotherapy and photothermal cancer therapy. Ag nanoparticles doped SnS 2 nanoflowers (SA), which can convert NIR light into thermal energy with efficiency of 30.0%, were embedded in low-melting-temperature agarose hydrogel as a photothermal agent. Moreover, anticancer drug doxorubicin (DOX) is loaded into the hydrogel for tumor treatment. The heat generated from NIR irradiation of SA promotes melting of hydrogel, thus the incorporated DOX can be released into the surrounding environment. By adjusting the irradiation power and time, the drug will be released with a control. In vitro study shows that the SA/ [email protected] system is highly effective to kill cancer cells (∼ 93%) at a relatively low -laser irradiation (1.2 w cm −2 , 10 min) treatment. In the mice tumor model, SA/ [email protected] with laser irradiation is capable to suppress or even eliminate the tumor completely from the mice within two weeks. The excellent anti-cancer efficiency benefits from the combined results of photothermal effect from NIR-irradiated SA and chemotherapy from DOX controlled release by heat. Therefore, this chemo-photothermal therapeutic system exhibits a great potential for cancer treatment.

A biomimetic nanoplatform for precise reprogramming of tumor-associated macrophages and NIR-II mediated antitumor immune activation

Yang Du,Xiaohui Qian,Fenghao Lin,Bingqiang Gao,Weili Wang,Huang Yang,Weilin Wang,Yuan Ding

The therapeutic effects of photothermal therapy (PTT) are dependent on the photothermal conversion efficiency of photothermal agents (PTAs) in tumors and the subsequent activation of the antitumor immune system. However, the insufficient tumor accumulation of current PTAs and the inevitable recruitment of tumor-associated macrophages (TAMs) could further compromise the antitumor activities of PTT. To address these issues, a biomimetic photothermal nanoplatform [email protected] is developed for the targeted remodeling of TAMs, which promotes the antitumor immunity of PTT. Au nanorods with second near-infrared (NIR-II) absorptions are fabricated to serve as PTAs to induce immunogenic cell death in tumor cells. The ferric hydroxide shell coated on Au nanorods can release iron ions to repolarize M2-like TAMs into the tumoricidal M1 phenotype via P38 and STAT1-mediated signaling pathways . Moreover, the surface decoration of platelet membranes endows biomimetic nanoplatform with enhanced tumor targeting ability for precise tumor ablation and TAM regulation. Consequently, [email protected] under NIR-II laser irradiation exhibits significantly higher inhibitory effects in a poor immunogenic 4T1 tumor-bearing mouse model with a 50% complete remission rate compared to conventional PTT (0%). By simultaneously reversing the immunosuppressive tumor microenvironment, this biomimetic nanoplatform offers a promising strategy for enhancing the antitumor efficacy of PTT. Statement of significance The therapeutic effects of current photothermal therapy (PTT) are hindered by the insufficient tumor accumulation of conventional photothermal agents and the recruitment of immunosuppressive tumor-associated macrophages (TAMs) after PTT. Herein, we report a biomimetic iron-based second near-infrared (NIR-II) photothermal nanoplatform ( [email protected] ) for targeted TAMs reprogramming and NIR-II mediated anti-tumor immunity. [email protected] can actively target the tumor site with the help of surface-decorated platelet membranes. Meanwhile, iron ions would be released from [email protected] in acidic lysosomes to reprogram TAMs into tumoricidal M1-like macrophages, which promotes the antitumor responses elicited by NIR-II PTT, thereby contributing to remarkable tumor inhibitory effects, with 50% higher complete remission rate than that of conventional PTT.

Surface Engineering of Metal–Organic Framework as pH-/NIR-Responsive Nanocarrier for Imaging-Guided Chemo-Photothermal Therapy

Haibin Guo,Yanqing Xia,Ke Feng,Xiaowei Qu,Cuilian Zhang,Feng Wan

Background Metal–organic frameworks (MOFs) have attracted intensive research interest in the biomedical field because of their unique properties. However, in order to realize the high loading capacity and therapeutic efficacy, it is still urgent to develop a multifunctional MOFs-based nanoplatform.Materials and Methods Herein, a pH/near-infrared (NIR) dual-responsive drug delivery system based on zeolitic imidazolate framework-8 (ZIF-8) is constructed for synergistic chemo-photothermal therapy and dual-modal magnetic resonance (MR)/photoacoustic (PA) imaging. The doxorubicin hydrochloride (DOX) is embedded into ZIF-8 through one-pot method, and the resultant ZIF-8/DOX is then successively modified with polydopamine, Mn ions and poly(ethylene glycol). The obtained ZIF-8/DMPP is systematically characterized, and both its in vitro and in vivo biological effects are evaluated in detail. Results The ZIF-8/DMPP possesses a high drug-loading content of 18.9% and displays appropriate size and morphology. The pH-dependent degradation and drug release behavior of prepared ZIF-8/DMPP are confirmed. Importantly, the results demonstrate that the photothermal effect of ZIF-8/DMPP under NIR laser irradiation can significantly accelerate its drug releasing rate, further improving the intracellular drug concentrations. Thereafter, the augmented chemotherapeutic efficiency by photothermal effect against cancer cells is verified both in vitro and in vivo. Besides, the favorable MR and PA imaging capacity of ZIF-8/DMPP is also evidenced on the tumor model. Conclusion Taken together, the surface engineering of ZIF-8-based nanocarrier in this work offers a promising strategy for the multifunctional MOFs-based drug delivery system.
