An acoustic squeezer for assessment of multiparameter cell mechanical properties

Wei Zhou,Yingyin Li,Yifan Liu,Hao Quan,Pengqi Li,Fei Li,Lili Niu,Hairong Zheng,Long Meng

Cells’ ability to sense and respond to mechanical stimuli is fundamental to various biological processes and serves as a crucial biomarker of their physiological and pathological states. Traditional methods for assessing cell mechanical properties, such as atomic force microscopy and micropipette aspiration, are hindered by complex procedures and the risk of cellular damage due to direct contact. Here we introduce a novel non-contact acoustic squeezer that leverages focused interdigital transducers to induce cell deformation through a robust standing surface acoustic wave (SSAW) field. This approach enables the multiparametric quantification of multiple mechanical properties, including elasticity (Young’s modulus, stiffness) and viscosity, without requiring labeling or physical contact, providing a comprehensive understanding of the cell mechanical properties. Our acoustic squeezer is capable of generating a maximum squeezing force of 25.70 pN, inducing a deformability of 1.27 ± 0.017. Combined with thin-shell deformation model, the quantized Young’s modulus of normal red blood cells (RBCs) is approximately 919.04 ± 55.64 Pa. Furthermore, our method demonstrates that cells treated with the anti-cancer drug (doxorubicin) exhibited reduced deformability, increased Young’s modulus and viscosity. Our acoustic squeezer offers a standardized, non-invasive, and highly sensitive approach for characterizing cell mechanical properties, with significant promise for clinical applications in disease diagnosis and drug development.

Integrated plasma and red blood cell membrane lipidomics analysis unveils novel biomarker panel for Alzheimer’s disease

He Dandan,Bao Yaran,Yuan Siyi,Wang Yingji,Cai Jingbo,Xu Fangfang,Chang Tiantian,Zhang Pei,Yue Menglu,Pan Xiaodong,Hao Haiping,Zheng Qiuling

Background Discovering new biomarkers for Alzheimer’s disease (AD) are significant for early diagnosis and monitoring disease’s progression. Blood-based lipid biomarkers, particularly from red cell membranes, offer a non-invasive alternative, providing insights into AD progression and potential therapeutic targets. Methods This study innovatively incorporates red cell membrane (RCM) lipids, which reflect chronic physiological alterations and are linked to AD pathogenesis. A comprehensive lipid detection platform was employed to analyze lipid profiles from 156 individuals, including normal subjects, those with amnestic mild cognitive impairment, and AD patients. Differential lipids were identified and validated to be AD-associated by correlation analysis and big data analysis. Multi-dimensional criteria were applied to select potential lipid biomarkers. Results PLS-DA revealed distinct lipidomic profiles between groups, with RCM samples showing superior separation. 138 differentially expressed lipids were identified and enriched in AD-related pathways, many uniquely associated with the RCM. Six lipids were selected as a potential biomarker panel for AD based on multi-dimensional criteria. Conclusion sRCM shows broader lipidomic relevance and stronger biomarker potential compared to plasma. Integrating RCM lipids with plasma analysis enhances AD diagnostics and highlights the potential of RCM as an underexplored biomarker resource.

Red Blood Cell-Hitchhiking Delivery of Simvastatin to Relieve Acute Respiratory Distress Syndrome

Mengjuan Sun,Jun Wei,Yanhui Su,Yangjingwan He,Liang Ge,Yan Shen,Bohui Xu,Yanlong Bi,Chunli Zheng

Purpose The purpose of this study is to address the high mortality and poor prognosis associated with Acute Respiratory Distress Syndrome (ARDS), conditions characterized by acute and progressive respiratory failure. The primary goal was to prolong drug circulation time, increase drug accumulation in the lungs, and minimize drug-related side effects. Methods Simvastatin (SIM) was used as the model drug in this study. Employing a red blood cell surface-loaded nanoparticle drug delivery technique, pH-responsive cationic nanoparticles loaded with SIM were non-covalently adsorbed onto the surface of red blood cells (RBC), creating a novel drug delivery system (RBC@SIM-PEI-PPNPs).Results The RBC@SIM-PEI-PPNPs delivery system effectively extended the drug’s circulation time, providing an extended therapeutic window. Additionally, this method substantially improved the targeted accumulation of SIM in lung tissues, thereby enhancing the drug’s efficacy in treating ARDS and impeding its progression to ARDS. Crucially, the system showed a reduced risk of adverse drug reactions. Conclusion RBC@SIM-PEI-PPNPs demonstrates promise in ARDS and ARDS treatment. This innovative approach successfully overcomes the limitations associated with SIM’s poor solubility and low bioavailability, resulting in improved therapeutic outcomes and fewer drug-related side effects. This research holds significant clinical implications and highlights its potential for broader application in drug delivery and lung disease treatment.

“Plug and Play” Functionalized Erythrocyte Nanoplatform for Target Atherosclerosis Management

Yuan Zhong,Xian Qin,Yi Wang,Kai Qu,Li Luo,Kun Zhang,Boyan Liu,Essam Abdo Mohammed Saad Obaid,Wei Wu,Guixue Wang

For atherosclerosis (AS) management, a therapeutic drug intervention is the most widely used strategy. However, there are some problems such as low location specificity, high intake, and side effects. Nanomedicine can prolong the half-life of drug solubilization, reduce toxic and side effects, and improve the distribution of drug objects. Herein, to overcome the challenges, an erythrocyte-based “plug and play” nanoplatform was developed by incorporating the vascular cell adhesion molecule-1 (VCAM-1) targeting and the acid stimulus responsibility. After the function moieties conjugated with DSPE-PEG, the targeting peptide and the acid-sensitive prodrug were conveniently integrated into red blood cells’ surface for enhancing target AS drug delivery and controlling local drug release. As a proof of principle, a plug and play nanoplatform with targeted drug delivery and acid-control drug release is demonstrated, achieving a marked therapeutic effect for AS.

Unloading of cryoprotectants from cryoprotectant-loaded cells on a microfluidic platform

Kang Yufeng,Zou Lili,Qiu Bensheng,Liang Xin,Sun Sijie,Gao Dayong,Ding Weiping

In this paper, a multistep dilution-filtration microdevice (MDFD) is developed for unloading cryoprotectants from cryoprotectant-loaded cells. The MDFD contained a diluent producing region, a dilution-filtration execution region, and a filtrate collection region. It was made of two patterned PMMA stamps with four pieces of sandwiched PVDF membranes. Firstly, the performances of the mixers that were used in the diluent producing region and the dilution-filtration execution region were assessed using fluorescence experiments. Then, the effect of the MDFD structure on the loss of cells was investigated by applying the MDFD to unload glycerin from glycerin-loaded porcine red blood cells. Finally, the effects of the cell density, glycerin concentration, and membrane pore size on the clearance efficiency of glycerin ( C G ), the survival rate of cells ( S C ) and the recovery rate of cells ( R C ) have been studied. Under the designed conditions, C G achieved ~80% and S C reached ~90%. However, R C was only ~40%, mainly resulting from the cells detained on the membrane surface and squeezed through the membrane pores into the filtrate. Increasing the membrane pore size caused high C G and S C , but low R C . For a low glycerin concentration, C G , S C , and R C were all high. For a high cell density, C G was high, but both S C and R C were low. This work is of significance to develop a microfluidic chip for unloading cryoprotectants from a small amount of cryopreserved cell samples.

Using Mesoporous Silica-Based Dual Biomimetic Nano-Erythrocytes for an Improved Antitumor Effect

Ziyue Xi,Yingying Jiang,Zitong Ma,Qun Li,Xinran Xi,Chuanyong Fan,Shuang Zhu,Junjie Zhang,Lu Xu

The nano-delivery system with a dual biomimetic effect can penetrate deeper in tumor microenvironments (TMEs) and release sufficient antitumor drugs, which has attracted much attention. In this study, we synthesized erythrocyte-like mesoporous silica nanoparticles (EMSNs) as the core loaded with doxorubicin (DOX) and coated them with calcium phosphate (CaP) and erythrocyte membrane (EM) to obtain DOX/EsPMs. The transmission electron microscopy (TEM), fluorescent co-localization and protein bands of SDS-PAGE were used to confirm the complete fabrication of EsPMs. The EsPMs with erythrocyte-like shape exhibited superior penetration ability in in vitro diffusion and tumor-sphere penetration experiments. Intracellular Ca2+and ROS detection experiments showed that the CaP membranes of EsPMs with pH-sensitivity could provide Ca2+continuously to induce reactive oxide species’ (ROS) generation in the TME. The EM as a perfect “camouflaged clothing” which could confuse macrophagocytes into prolonging blood circulation. Hemolysis and non-specific protein adsorption tests proved the desirable biocompatibility of EsPMs. An in vivo pharmacodynamics evaluation showed that the DOX/EsPMs group had a satisfactory tumor-inhibition effect. These advantages of the nano-erythrocytes suggest that by modifying the existing materials to construct a nano-delivery system, nanoparticles will achieve a biomimetic effect from both their structure and function with a facilitated and sufficient drug release profile, which is of great significance for antitumor therapy.

Enhancing Chemotherapy Efficacy via an Autologous Erythrocyte-Anchoring Strategy with a Closed-System Drug-Transfer Device

Lingzi Feng,Xiangqian Wang,Ziyi Gao,Yuqing Tong,Xiaopeng Yuan,Ting Wu,Donglin Xia,Yong Hu

Chemotherapeutic drugs often fail to localize efficiently to tumors when administered intravenously, causing off-target effects. This study proposes an autologous erythrocyte (ER)-anchoring strategy to improve chemotherapy efficacy and reduce side effects. Utilizing a modified hemodialysis instrument, a closed-system drug-transfer device was developed for autologous ER procurement and immunogenicity mitigation. Doxorubicin (DOX) and indocyanine green (ICG) were encapsulated in autologous ERs and then modified with DSPE-PEG-FA. The final product, DOX-ICG@ER-D, was reintroduced into circulation to enhance chemotherapy. These obtained DOX-ICG@ER-D showed good stability, minimal cardiotoxicity, and extended circulation time. Compared to free DOX, DOX-ICG@ER-D had a higher accumulation of DOX in hepatocellular carcinoma and the release of DOX could be controlled by laser irradiation. Tumor-bearing rats treated by these DOX-ICG@ER-D demonstrated improved antitumor efficacy and reduced cardiotoxicity. Thus, this autologous ER-anchoring strategy offers a promising alternative to intravenous chemotherapy in the clinic.

A novel collaborative signal amplification biosensor based on sandwich-type composite film for ultrasensitive detection of rhEPO

Zhang Weiqian,Zhang Yichen,Xiong Shisheng

This research established a highly sensitive and low-cost electrochemical biosensor for the detection of recombinant human erythropoietin. A sandwich-like gold nanoparticles/Chitosan/gold nanoparticles composite film was prepared on a glassy carbon electrode. The gold nanoparticles/Chitosan/gold nanoparticles film, comprising gold nanoparticles with a large specific surface area, high conductivity, and excellent biocompatibility, was paired with Chitosan, which was naturally high in content, biodegradable, and biocompatible. Immobilization of the erythropoietin receptor on gold nanoparticles/Chitosan/gold nanoparticles films allowed recombinant human erythropoietin to recognize erythropoietin receptor, thereby altering the number of pores. The electrochemical results demonstrated that the gold nanoparticles/Chitosan/gold nanoparticle-modified electrochemical biosensor exhibited a broad detection range (5 × 10 –13 to 5 × 10 –9  g L −1 ) and a low detection limit (5.16 × 10 –14  g L −1 ). The overall preparation process was straightforward, and the detection process was rapid, with an incubation period of only 20 min. Furthermore, the biosensor exhibited high specificity, repeatability, and stability. In conclusion, the electrochemical biosensor was a simple and rapid method for the specific and accurate detection of trace recombinant human erythropoietin, which was suitable for clinical monitoring and the identification of doping. Effective Cell and Particle Sorting and Separation in Screen-Printed Continuous-Flow Microfluidic Devices with 3D Sidewall Electrodes

Xiaoguang Lin,Jie Yao,Hua Dong,Xiaodong Cao

In recent years, microfluidic dielectrophoresis (DEP) devices, as one of the most promising tools for cell and particle sorting and separation, are facing the bottleneck in the development of practical products due to the high-cost yet low-yield device manufacturing via traditional microelectromechanical systems (MEMS) and the challenge of maintaining the cell viability during DEP treatment. In this paper, we demonstrate a facile, low-cost, and high-throughput method of constructing continuous-flow microfluidic DEP devices via screen-printing technology. The new device configuration and operation strategy not only facilitate cell and particle sorting and separation using 3D electrodes as sidewalls of microchannel but also improve cell viability by reducing the exposure time of cells to high electrical-field gradients. Furthermore, we propose and validate a semiempirical formula with which to simplify the complicated calculation and plotting of DEP spectra. As a consequence, the optimal DEP parameters and crossover frequencies can be obtained directly using our devices instead of typical electrorotation method. To evaluate the performance of a screen-printed continuous flow microfluidic DEP device, a suspension containing polystyrene (PS) microspheres and erythrocytes is used as the biosample. Our results show that a high sorting efficiency (ca. 93%) with a high cell viability (hemolysis ratio of <4.8%) can be achieved, indicating the excellent performance and promising application of such devices for cell and particle sorting and separation.

Ultradense Erythrocyte Bionic Layer Used to Capture Circulating Tumor Cells and Plasma-Assisted High-Purity Release

Taoye Zhang,Wei Peng,Wanli Jiang,Kefan Gao,Wei Liu

The isolation and detection of rare circulating tumor cells (CTCs) from patient peripheral blood can help early diagnosis of cancer and evaluation of therapeutic outcomes. At present, most of the available strategies for enriching CTCs face serious problems with purity due to the nonspecific interactions between the capture medium and leukocytes. Inspired by the immune evasion ability of homologous red blood cells (RBCs), we modified the tumor-targeting molecule folic acid (FA) on the surface of RBCs by hydrophobic interactions. Under the treatment of polybrene, the charges on the surface of RBCs are neutralized, which reduces the mutual repulsion force. Furthermore, RBCs treated with polyethylene also have excellent deformability, thereby enabling engineered RBCs to form a dense bionic layer on the adhesive glass slide, which can greatly inhibit the nonspecific adhesion of leukocytes. The bionic layer can achieve high-purity enrichment of tumor cells in phosphate-buffered saline (PBS), and we can achieve high-activity release in plasma. The cell count showed over 80% capture efficiency and over 70% release rate, and the purity of CTCs obtained in the artificial blood sample after release was higher than 90%. The RBC bionic surface coating is notably cost-effective and highly applicable for CTC isolation in clinic practice, and thus provides new prospects for designing cell–material interfaces for advanced cell-based biomedical studies in the future.

Inner filter with carbon quantum dots: A selective sensing platform for detection of hematin in human red cells

Qian Qian Zhang,Bin Bin Chen,Hong Yan Zou,Yuan Fang Li,Cheng Zhi Huang

Hematin plays a crucial role in various physiological functions, and the determination of hematin in complex biological matrixes is a significant but difficult issue. Considering the unique photophysical/photochemical properties of carbon quantum dots (CQDs) prepared with p-aminobenzoic acid (PABA) and ethanol, a new strategy for the design of fluorescent probes for hematin has been achieved. The proposed sensor array is fabricated based on the inner filter effect (IFE) between hematin and CQDs with phenomenon of selective fluorescence quenching of CQDs which results from the strong absorption of the excitation and emission spectrum of CQDs by hematin. The fluorescence quenching of CQDs is closely related to the amount of hematin and there is a good linear relationship over the range of 0.5–10   μM with a detection limit of 0.25   μM. What's more, the fluorescence assay has been successfully applied for hematin sensing in healthy human red cells showing this sensing assay has a great potential prospect for detection of hematin in the complex matrixes .

Full encapsulation of oncolytic virus using hybrid erythroctye-liposome membranes for augmented anti-refractory tumor effectiveness

Hanwei Huang,Mengchi Sun,Mingyang Liu,Siwei Pan,Pengfei Liu,Zhenguo Cheng,Jia Li,Huimian Xu,Funan Liu,Zhiqing Pang

Intravenous delivery of oncolytic virus (OVs) is promising in cancer treatment. Unfortunately, fast clearance of OVs and the severe cytokine release syndrome impede its wide application. It has been shown that nanoparticles coated with cell membranes display less toxicity and slower clearance. However, different from conventional nanoparticles, the characteristic spike-like structure and abundant antigens on the surface make it difficult for intravenously delivered OVs to take advantage of cell membrane coating to shield their surface antigens. To overcome this challenge, we, for the first time, used erythrocyte-lipid hybrid membrane vesicle (erythroliposome) to fully encapsulate OVs for their intravenous delivery. We found that adding artificial membranes to cell membranes reduced the fluidity of the membranes, leading to an extraordinary shielding effect on OV antigens. Consequently, circulation of OVs was significantly prolonged and their oncolytic efficacy to metastatic and refractory tumors was markedly enhanced.
