Quantification of cow milk in adulterated goat milk by HPLC-MS/MS using N-acetylglucosamine as a reliable biomarker of cow milk

Jing-Yan Li,Qian Ge,Bin Liu,Zhen-Peng Gao,Chun-Feng Guo

This study aimed to develop a non-protein-based method for quantify cow milk in adulterated goat milk. The raw cow milk and raw goat milk samples were mixed separately with the same volume of cold acetone and centrifuged to remove their proteins. The supernatants were evaporated, and the residues were derivatized by silylation followed by GC-MS analysis. N-acetylglucosamine was identified as a thermostable non-protein biomarker of cow milk. Its concentration in the raw cow milk (146.7 mg/L) was 32 times higher than that in the raw goat milk. On this basis, a HPLC-MS/MS method using positive-ion electrospray ionization in parallel reaction monitoring mode was developed for quantifying cow milk in adulterated goat milk by determining peak area of the biomarker. The method showed good linearity over cow milk concentrations of 1–100% and could detect as low as 0.3% cow milk in adulterated goat milk. Besides, the method was characterized by a simple sample pretreatment process, rapid detection, high accuracy, and high precision. With this method, a sample could be analyzed in 10 min. The present study demonstrated the feasibility of quantifying cow milk in adulterated goat milk using N-acetylglucosamine as a biomarker of cow milk.

A “wave-riding” biocatalysis: all-enzyme system toward genuine-green and flexible manufacture of machine-washable dyed wool fabrics

Xinrui Zhang,Jun Wang,Man Zhou,Yuanyuan Yu,Ping Wang,Qiang Wang

The well-known phenomenon of felt shrinkage that occurs during home laundry of wool textiles has long bothered consumers. Currently, to address this issue, factories have no choice but to employ the chlorination-Hercosett process, which predominates in the production of machine-washable wool fabrics. However, this method often results in color alternations in dyed fabrics and generates absorbable organic halides (AOX), thereby significantly exacerbating environmental challenges. Enzymatic processing has been consistently recognized as a promising alternative, while most necessitate the assistance of chemical reagents. To accomplish fully green manufacturing of machine-washable wool fabrics, this study has innovatively developed a compound-protease system based on a "wave-riding" enzymatic mechanism, reaching localized surface modification of wool fibers. The results demonstrated that dyed wool fabrics treated exclusively with proteases exhibited excellent dimensional stability (3.90% ± 0.12%) and controllable strength loss (11.39% ± 0.61%). Additionally, the wearability of the fabrics, which was quantified through assessing factors such as air and moisture permeability, the hand feeling, and the wettability, was improved. The original color properties of dyed wool fabrics were also preserved; specifically, there was no adverse effect on color fastness and minimal color difference (∆ECMC < 1.00). This approach further fills the technical gap in the field of genuinely green anti-felting solutions for dyed wool fabrics, offering a new pathway toward sustainable textile manufacturing.

Assembly of Chitosan/Caragana Fibers to Construct an Underwater Superelastic 2D Layer-Supported 3D Architecture for Rapid Congo Red Removal

Ning Luo,Hanwen Ge,Xiangyu Liu,Qingdong He,Wenbo Wang,Wenyuan Ma,Fang Guo

Developing environmentally friendly bulk materials capable of easily and thoroughly removing trace amounts of dye pollutants from water to rapidly obtain clean water has always been a goal pursued by researchers. Herein, a green material with a 3D architecture and with strong underwater rebounding and fatigue resistance ability was prepared by means of the assembly of biopolymer chitosan (CS) and natural caraganate fibers (CKFs) under freezing conditions. The CKFs can randomly and uniformly distribute in the lamellar structure formed during the freezing process of CS and CKFs, playing a role similar to that of “steel bars” in concrete, thus providing longitudinal support for the 3D-architecture material. The 2D layers formed by CS and CKFs as the main basic units can provide the material with a higher strength. The 3D-architecture material can bear the compressive force of a weight underwater for multiple cycles, meeting the requirements for water purification. The underwater compression test shows that the 3D-architecture material can quickly rebound to its original shape after removing the stress. This 3D-architecture material can be used to purify dye-containing water. When its dosage is 3 g/L, the material can remove 99.65% of the Congo Red (CR) in a 50 mg/L dye solution. The adsorption performance of the 3D architecture adsorbent for CR removal in actual water samples (i.e., tap water, seawater) is superior than that of commercial activated carbon. Due to its porous block characteristics, this material can be used for the continuous and efficient treatment of wastewater containing trace amounts of CR dye to obtain pure clean water, meaning that it has great potential for the effective purification of dye wastewater.

Effect of Antifreeze Glycopeptides on the Quality and Microstructure of Frozen Lamb Meatballs

Rong Dong,Shengkun Yan,Guoqiang Wang,Pei Wang

This study explored the protective effects of antifreeze glycopeptide and alginate on the quality of −18 °C frozen lamb meatballs across various storage periods. Methods: Measurements of volatile salt nitrogen (TVB-N), thiobarbituric acid (TBARS), water retention, water distribution, microstructure, and metabolite changes were taken in the lamb meatballs. Results: The results showed that the addition of antifreeze glycopeptides (AFGs) significantly preserved the quality characteristics of lamb meatballs. In particular, the 0.30% antifreeze glycopeptide demonstrated the strongest protective effect on water retention and metabolites during freezing. The ice crystal area within the microstructure of lamb meatballs with added antifreeze glycopeptides was markedly reduced compared to the others after 14 days of freezing (p< 0.05). Additionally, AFGs lessened the lipid oxidation reaction and prolonged the oxidation time of lamb after 28 days of freezing. Conclusion: In summary, AFGs beneficially affected the quality of frozen lamb meatballs and are a potential, safe, and efficient cryoprotectant.

Eco-friendly shrink-resist finishing of wool through synergistic effect of disulfide bond reducing agent and papain

Kun Li,Zirong Li,Jiaqi Zhang,Jun Wang,Yuanyuan Yu,Man Zhou,Qiang Wang,Ping Wang

The environmental benefits of utilizing protease as a biocatalyst for wool shrink-resist finishing have been widely recognized. However, the efficacy of individual protease treatment is unsatisfactory due to its incapability towards the outermost cuticle layer of wool fibers that contains hydrophobic fatty acids. In order to weaken the structural integrity of the highly cross-linked scales and promote the enzymatic anti-felting, sodium sulfite and tris (2-carboxyethyl) phosphine hydrochloride (TCEP) were employed in combination with papain , respectively, aiming at obtaining a low shrinkage without unacceptable fiber damages. Based on the synergistic effect of papain and TCEP, the edges of wool scales were slightly destroyed by the reduction of disulfide bonds, accompanied by enzymatic hydrolysis of the keratin component. Through the controlled reduction and hydrolysis of wool scales, satisfactory anti-felting result was achieved without causing severe damage to the fiber interiors. In the presence of 0.25 g/L TCEP and 25 U/mL papain, the area shrinkage of wool fabric decreased to approximately 6 %, with a low strength loss of less than 8 %. Meanwhile, the dyeing behavior of the wool fabric under low-temperature conditions was dramatically improved, leading to decreased energy consumption during production. The present work provides an alternative for eco-friendly finishing of wool fabrics, which can be applied commercially.

Synthesis of acid dyes containing polyetheramine moieties and their low-temperature dyeing properties on wool fiber

Haiying Wang,Guowei Wang,Chunlin Zheng,Tijian Zhou,Jie Sun

To realize the low-temperature dyeing of wool fibers, the use of auxiliaries and wool modification are common methods. Low-temperature auxiliaries may cause water-pollution problems, and wool modification is a costly and uncontrollable process. In this study, new acid dyes, named D1–D3, containing polyetheramine groups were synthesized and applied to wool fiber by conventional and low-temperature exhaust dyeing procedures. The results indicate that the new acid dyes could interact with wool-fiber-like auxiliaries and render a high exhaustion rate to the wool fiber at a dyeing temperature of 80 °C. In comparison with Acid Blue 25, the D1–D3 dyes showed an increased dyeing rate, especially under a dyeing temperature of 80 °C. Despite the bigger relative molecular masses of D1–D3, the exhaustion rates of D1–D3 were still higher, and the times of half-dyeing were shorter than that of Acid Blue 25. The color differences between the wool fabrics dyed with the four dyes at 80 and 98 °C, respectively, were compared. We found that the color differences of D1–D3 between 80 and 98 °C were smaller than that of Acid Blue 25. The interactions between the dyes and wool fiber were analyzed and manifested by the measurement of the ζ potential of the dyes and wool fiber. The leveling and transfer properties of the D1–D3 dyes were also investigated, and the color differences of the wool fabrics dyed with Acid Blue 25, D1, and D2 were very low at all measured pH values and temperatures. The fastnesses of D1–D3 on wool fabric were almost the same as that of Acid Blue 25. © 2017 Wiley Periodicals, Inc. J. Appl. Polym. Sci. 2018 , 135 , 45793.

A Sponge-Like Double-Layer Wound Dressing with Chitosan and Decellularized Bovine Amniotic Membrane for Promoting Diabetic Wound Healing

Yang Yang,Yanyan Zhang,Yishu Yan,Qian Ji,Yutong Dai,Suyuan Jin,Yanxian Liu,Jinghua Chen,Liping Teng

The diabetic wounds do not heal easily in part because they are susceptible to infection due to environmental influences. Wound dressing is crucial to wound healing, as it can basically protect the wound from external damages and provide a suitable microenvironment for tissue regeneration. In this study, a double-layer membrane that consists of chitosan sponge and decellularized bovine amniotic membrane (dBAM) has been developed by freeze-casting method. The results showed that the porous structure of the sponge layer improved the performances of blood coagulation and swelling. The dense dBAM can optimize the mechanical property of wound dressing. In vitro studies revealed that the bilayer membrane had favorable biocompatible, especially for human foreskin fibroblast cells (HFF-1) cell adhesion and proliferation. Moreover, the full-thickness skin defects of diabetic model mice that treated with bilayer membrane showed over 80% closure in 8 days. Our findings imply that the double-layer dressing has great potentials to be used in diabetic patients. Keywords:chitosan;decellularized bovine amniotic membrane;diabetic wound healing;double-layer dressing

Ultrafine, self-crimp, and electret nano-wool for low-resistance and high-efficiency protective filter media against PM0.3

Yuyao Li,Leitao Cao,Xia Yin,Yang Si,Jianyong Yu,Bin Ding

Frequent outbreaks of emerging infectious diseases (EIDs) make personal protective filter media in high demand. Electrospun nanofibrous materials are proved to be very effective in resisting virus-containing fine particles owing to their small fiber diameters; however, hindered by the intrinsic close-packing character of fine fibers, electrospun filters suffer from a relatively high air resistance, thereby poor breathing comfort. Here, we report a biomimetic and one-step strategy to create ultrafine and curly wool-like nanofibers, named nano-wool, which exhibit fluffy assembly architecture and powerful electret effect. By achieving the online self-crimp and in-situ charging of nanofibers, the curly electret nano-wool shows a small diameter of ~0.6 μm (two orders of magnitude lower than natural wool: ~20 μm) and an ultrahigh porosity of 98.7% simultaneously, together with an ultrahigh surface potential of 13260 V (one order of magnitude higher than previous filters). The structural advantages and powerful electret effect enable nano-wool to show excellent filtration efficacy (>99.995% for PM 0.3 ) and low air resistance (55 Pa). Additionally, nano-wool can be easily scaled up, not only holding great industrial prospect in personal protective respirators, but also paving the way for developing next-generation wool in a cost-efficient and multifunctional form.

Dissolution and regeneration of wool keratin in the deep eutectic solvent of choline chloride-urea

Zhe Jiang,Jiugang Yuan,Ping Wang,Xuerong Fan,Jin Xu,Qiang Wang,Lianbing Zhang

Wool keratin is a kind of renewable protein which has great potential in environment-friendly materials. In this paper, the dissolution of wool fiber and the regeneration of wool keratin in the deep eutectic solvent (DES) of choline chloride-urea was detailed studied. The results showed that 35.1 mg wool completely dissolved in 1 g DES at 130 °C in 5 h. The polarizing microscope, scanning electron microscope (SEM) images along with the amino acid analysis suggested that the DES mainly dissolved the wool cortex layer. Compared with the raw wool, the regenerated wool keratin contained much more fragments of low molecular weight polypeptide chains from SDS-PAGE results. And the percentage of α-helix dramatically decreased, while the content of β-sheet and disordered structure increased. All the results indicated that the DES can be used as a kind of green solvent for the regeneration of wool keratin without losing the long peptide chains. The method also offered a possibility of large-scale for wasted wool reuse.

Tracing antibacterial compounds from Acalypha australis Linn. by spectrum-effect relationships and semi-preparative HPLC

Sui Xiao,Li-Fang Zhang,Xu Zhang,Su-Mei Li,Fei-Qun Xue

Several Acalypha australis Linn. species are used in traditional medicine in Southeast Asia. In this work, the ultra-performance liquid chromatography/mass spectrometry fingerprints and the antibacterial activities of A. australis Linn. were investigated. An in-depth discussion on the reliability of identifying and obtaining potentially active compounds by spectrum-effect relationship and semi-preparative high performance liquid chromatography was conducted. The result shows that gallic acid and a compound with molecular weight of 634.1 in the fingerprints were the main antibacterial compounds. Compared to the crude extract of A. australis Linn., both compounds increase the antibacterial efficacy 10 to 20 times. Compounds with molecular weights of 154.0, 292.0, and 485.1 in the fingerprints were the auxiliary antibacterial compounds. Through the entire isolation procedure, we obtained these antibacterial compounds with purities of 92.53, 87.98, 90.73, 89.36, and 88.14%, respectively. This work provides a general model of the combination of ultra-performance liquid chromatography/mass spectrometry fingerprinting and antibacterial test to study the spectrum-effect relationships of A. australis Linn. This model can be used to discover further the active compounds of this herb.

Effect of nisin and potassium sorbate additions on lipids and nutritional quality of Tan sheep meat

Wei Jia,Xixuan Wu,Ruiting Li,Shuxing Liu,Lin Shi

Nisin and potassium sorbate as preservatives are used in a broad range of meat. A lipidomic evaluation was performed on Tan sheep meat treated by two types of preservatives. The addition of potassium sorbate resulted in higher lipid losses compared with nisin treatment. Furthermore, 106 significant lipids of 12 lipid classes (PC, PS, LPS, LPC, PE, PI, LPE, TG, Cer, DG, SM, Sph) with variable importance in projection scores greater than 1.0 were detected and qualified to distinguish different preservatives added meat using UHPLC-Q-Orbitrap MS/MS. LOD and LOQ were 0.12–0.32 μg kg −1 and 0.35–0.89 μg kg −1 , indicating high sensitivity and excellent analytical characteristics in the study. Nisin was confirmed to be the better preservative for prolonging the shelf life of Tan sheep meat while reducing the loss of nutrients. These results could provide a strong cornerstone for future research on preservatives in meat products.

Characterization and Discrimination of Key Aroma Compounds in Pre- and Postrigor Roasted Mutton by GC-O-MS, GC E-Nose and Aroma Recombination Experiments

Huan Liu,Teng Hui,Fei Fang,Qianli Ma,Shaobo Li,Dequan Zhang,Zhenyu Wang

The key aroma compounds in the pre- and postrigor roasted mutton were studied in this study. The results showed that 33 and 30 odorants were detected in the pre- and postrigor roasted mutton, respectively. Eight aroma compounds, including 3-methylbutanal, pentanal, hexanal, heptanal, octanal, nonanal, 1-octen-3-ol, and 2-pentylfuran, were confirmed as key odorants by aroma recombination and omission experiments. The aroma profiles of pre- and postrigor roasted mutton both presented great fatty, roasty, meaty, grassy, and sweet odors. Particularly, the concentrations of hexanal, heptanal, octanal, nonanal, 1-octen-3-ol, and 2-pentylfuran in postrigor roasted mutton were significantly higher (p< 0.05) than those in the prerigor roasted mutton. The postrigorback strapwas more suitable for roasting than the prerigorback strap. The pre- and postrigor roasted mutton could be obviously discriminated based on the aroma compounds by orthogonal partial least squares discrimination analysis (OPLS-DA) and principal component analysis (PCA). Hexanal and 1-octen-3-ol might potential markers for the discrimination of the pre- and postrigor roasted mutton. Keywords:roasted mutton;pre- and postrigor;key aroma compounds;marker;recombination and omission experiments
