Herein, a sandwich electrochemical sensing strategy for aflatoxin b 1 (AFB 1 ) detection based on hybrid-nanoengineering was presented. First, Au nanoparticle was doped into zeolitic imidazolate framework-8 (ZIF-8) to form Au@ZIF-8 by in-situ growth method, followed by multi-walled carbon nanotubes (MWCNTs) addition to synthesize MWCNTs/Au@ZIF-8 via self-assembly. The structural “confinement effect” of ZIF-8 afforded a microenvironment for Au nanoparticles and SMCNTs in a certain spatial region, giving MWCNTs/Au@ZIF-8 excellent electrochemical property as the substrate material. In addition, Au-Pt bimetallic nanoparticle, which exhibited excellent stability and catalytic activity was loaded on the hollow cerium oxide (CeO 2 ) to form AuPt@CeO 2 nanoparticle through one-step aqueous phase reduction. Owning to its high surface-to-volume ratio, satisfied electron transfer efficiency and biocompatibility, massive toluidine blue (TB) and AFB 1 antibody (Ab) could be modified on the AuPt@CeO 2 to form AuPt@CeO 2 -Ab-TB, which acted as signal tag for the ultrasensitive assay of AFB 1 . The proposed electrochemical sensing system exhibited wide detection range (2 × 10 -5 − 20 ng/mL) and low detection limit (2.13 fg/mL), which has been successfully applied to AFB 1 determination in four real samples. The hybrid nanoengineering presented in this work is an active attempt to prepare high-performance substrate material and signal tag, which provides a new insight for the development of highly sensitive and specific electrochemical sensing systems.

Visual and on-site monitoring of aflatoxin B1 (AFB1) with high sensitivity and selectivity is essential for ensuring food safety. We herein developed a colorimetric biosensor for AFB1 monitoring based on target-mediated DNA self-replicating circuits. The aptamer-AFB1 recognition liberated the trigger DNA (T), which initiates DNA assemblies involving hairpin probes H1, H2 and H3. Through a series of DNA toehold hybridizations and strand displacements, a ternary hybrid H1-H2-H3 will be produced. At one arm of the H1-H2-H3 hybrid is a newly regenerated T and at other two arms are the Mg 2 + -DNAzymes. The Mg 2+ -DNAzyme can cleave the substrate chain to liberate another blocked T sequence and a G-quadruplex sequence. All three types of T probes can be recycled to form the DNA self-replicating circuits than can continuously generate H1-H2-H3 products. The G-quadruplex DNAzyme catalyzes the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to produce a colorimetric signal that is observable directly by the naked eye. This enzyme-free DNA circuit-based biosensor exhibits high sensitivity, with a visual detection limit reaching 0.1 pM. This biosensor is robust and can be employed to detect AFB1 in corn, peanut, and rice samples with good reliability. Using AFB1, deoxynivalenol (DON), and fumonisin B1 (FB1) as three logic inputs, we further built cascaded INHIBIT-AND and AND-OR logic gates, which possess great potential for intelligent sensing of various mycotoxins.

Effective truncation and mutation can obtain highly active nucleic acid aptamers for the target, especially avoiding non-specific binding, uncertainty to the tertiary structure and low binding affinity of the lengthy aptamer. In this work, a new aptamer, T-AF28 (28 nt) with better affinity to AFB1 was obtained by truncating 22 bases and mutating 3 bases of the reported AFB1 aptamer (50 nt), and the thermodynamic information, structural changes and binding domains of aptamer T-AF28 in the presence of AFB1 or AFM1 were further investigated by isothermal titration calorimetry, circular dichroism, and molecular dynamics. On this basis, Using optimized aptamers as recognition elements, a novel label-free and enzyme-free fluorescent aptasensor for AFB1 and AFM1 was developed by synergistic amplification effect of hybridization chain reaction (HCR) coupled molybdenum disulfide nanosheets and copper nanoparticles in situ synthesized (CuNPs) on an extra-long double-stranded template generated by HCR as the fluorescent probe. Under the optimal experimental conditions, sensitive and selective fluorescent detection of AFB1 and AFM1 was achieved with wide linear detection ranges of 0.030 ng/mL-46.8 ng/mL and 0.030 ng/mL-32.8 ng/mL, respectively. And this aptasensor can be applied to accurate detection of AFB1 and AFM1 in grains, edible oil, tea, and milk with the recovery in the range of 85.20 %-117.0 %. This work not only provides a theoretical basis for the design of AFB1 and AFM1 aptasensors and expands the application of AFB1 aptasensors in the field of food safety, but also breaks a new path for the development of HCR-MoS 2 Ns CuNPs aptasensors.

Background DNA tweezers, classified as DNA nanomachines, have gained prominence as multifunctional biosensors due to their advantages, including a straightforward structure, response mechanism, and high programmability. While the DNA tweezers demonstrate simultaneous, rapid, and stable responses to different targets, their detection sensitivity requires enhancement. Some small molecules , such as mycotoxins , often require more sensitive detection due to their extremely high toxicity. Therefore, more effective signal amplification strategies are needed to further enhance the sensitivity of DNA tweezers in biosensing. Results We designed programmable DNA tweezers that detect small-molecule mycotoxins and miRNAs through simple sequence substitution. While the DNA tweezers demonstrate simultaneous, rapid, and stable responses to different targets, their detection sensitivity requires enhancement. We introduced the Strand Displacement Amplification (SDA) technique to address this limitation, proposing a strategy of novel programmable DNA tweezers-SDA ultrasensitive signal amplification fluorescence sensing . We specifically investigate the effectiveness of this approach concerning signal amplification for two critical mycotoxins: aflatoxin B1 (AFB1) and zearalenone (ZEN). Results indicate that the detection ranges of AFB1 and ZEN via this strategy were 1–10,000 pg mL −1 and 10–100,000 pg mL −1 , respectively, with corresponding detection limits of 0.933 pg mL −1 and 1.07 pg mL −1 . Compared with the DNA tweezers direct detection method for mycotoxins, the newly constructed programmable DNA tweezers-SDA fluorescence sensing strategy achieved a remarkable 10 4 -fold increase in the detection sensitivity for AFB1 and ZEN. Significance The constructed programmable DNA tweezers-SDA ultrasensitive signal-amplified fluorescence sensing strategy exhibits excellent detection performance for mycotoxins. The superb versatility of this strategy allows the developed method to be easily used for detecting other analytes by simply replacing the aptamer and cDNA, which has incredible potential in various fields such as food safety screening, clinical diagnostics, and environmental analysis .

An aptamer affinity based microextraction column is developed to be directly in-line coupled to capillary electrophoresis-mass spectrometry (CE-MS) for analyzing mycotoxins in food samples. Single-stranded DNA aptamers for selective recognition of aflatoxin B1 (AFB1) and ochratoxin A (OTA) targets are co-immobilized via covalent bonds on the surface of the inlet end of a capillary, which is pre-modified with three-dimensional porous layer and gold nanoparticles to enhance the specific surface area and loading capacity. The outlet of the capillary is designed as a porous tip to serve as the spray source for injection to the mass spectrometry. All the necessary processes for pretreatment and analysis of a sample are accomplished in one injection, including aptamer affinity-based microextraction, CE separation and MS detection of analytes. AFB1 and OTA are simultaneously determined in a wide linear range with sample consumption of only 1 μL and the limit-of-detection as low as 1 pg/mL. The microextraction column exhibits excellent repeatability and stability, which can be used over 45 runs within a month with CE separation efficiency and only MS intensity slightly decreased. Mycotoxins in three kinds of cereal based infant foods are accurately analyzed using the proposed method. The study provides a robust and universal approach that would have potential applications in a variety of analytical fields based on selective molecular recognition coupling to CE-MS analysis.

Simple, rapid, convenient, and economical surface enhanced Raman scattering (SERS) substrate is developed for on-site evaluation of Aflatoxin B 1 (AFB 1 ) in food matrix using handheld Raman Spectrometer. Self-assembly of gold nanobipyramids (Au NBPs) into the nanoholes of anodic aluminum oxide (AAO) template/pattern using ‘drop-dry’ approach provides a reliable pathway for the rapid fabrication of highly active and uniform SERS substrate. It shows enhanced and reproducible SERS signals towards the probe molecule, 4-aminothiophenol (4-ATP) with a relative standard deviation (RSD) of less than 10% and an average enhancement factor (EF) of 1 × 10 8 . For practical application, the proposed method is demonstrated for the detection of aflatoxin B 1 (AFB 1 ) in peanut extracts. The results show that the AFB 1 in peanut extracts can be identified within 1 min, with a limit of detection of 0.5 μg/L. Compared with conventional ELISA based AFB 1 analysis, our method is much more efficient (1 min versus >30 min).

The potential homogeneous assay employing immunomagnetic beads (IMB) has been receiving attention as a screening tool in food-safety control; the method is simple, efficient, and does not require long incubation times or complex separation steps. In this study, a homogeneous immunoassay has been successfully developed and applied in the determination of aflatoxin B1 (AFB1) contamination in agricultural products by coupling IMB and the biotin-streptavidin (BSA) (BSA-IMB) system. Under optimal conditions, the limit of detection (LOD, IC10), half-maximal inhibition concentration (IC50) and detection range (IC20–IC80) of BSA-IMB are 0.00579, 0.573 and 0.0183–17.9 ng mL−1, respectively, for AFB1. The detection of AFB1 by BSA-IMB can be achieved in 40 min (ELISA needs at least 180 min). The cross-reactivities of BSA-IMB with its analogues are negligible (<3.82%); these results indicate high selectivity. The spiked recoveries are in the range from 89.6 to 118.2% with relative standard deviations (RSDs) of 3.4 to 13.2% for AFB1 in agricultural product samples. Furthermore, the results of BSA-IMB for authentic samples show reliability and high correlation of 0.9928 with an HPLC-fluorescence detector. The proposed BSA-IMB system is demonstrated to be a satisfactory tool for homogeneous, efficient, sensitive, and alternative detection of AFB1 in a wide detection range for agricultural product samples.

High-toxicity secondary metabolites called aflatoxin are naturally
produced by the fungus Aspergillus. In a warm, humid
climate, Aspergillus growth can be considerably
accelerated. The most dangerous chemical among all aflatoxins is
aflatoxin B1 (AFB1), which has the potential to cause cancer and
several other health risks. As a result, food forensicists now
urgently need a method that is more precise, quick, and practical for
aflatoxin testing. The current study focuses on the development of a
highly sensitive, specific, label-free, and rapid detection method for
AFB1 using a novel humanoid-shaped fiber optic WaveFlex biosensor
(refers to a plasmon wave-based fiber biosensor). The fiber probe has
been functionalized with nanomaterials (gold nanoparticles, graphene
oxide and multiwalled carbon nanotubes) and anti-AFB1 antibodies to
enhance the sensitivity and specificity of the developed sensor. The
findings demonstrate that the developed sensor exhibits a remarkable
low detection limit of 34.5 nM and exceptional specificity
towards AFB1. Furthermore, the sensor demonstrated exceptional
characteristics such as high stability, selectivity, reproducibility,
and reusability. These essential factors highlight the significant
potential of the proposed WaveFlex biosensor for the accurate
detection of AFB1 in diverse agricultural and food samples.© 2023 Optica Publishing Group under the terms

The multimodal lateral flow immunoassay (LFIA) has provided accurate and reliable results for fast and immediate detection. Nonetheless, multimodal LFIA remains challenging to develop biosensors with high sensitivity and tolerance to matrix interference in agro-food. In this study, we developed a self-assembled multivalent fluorescence-nanobody (Nb26-EGFP-H6) with 16.5-fold and 30-fold higher affinity and sensitivity than a monovalent nanobody (Nb26). Based on the Nb26-EGFP-H6, we synthesized enhanced immune-probes Zn-CN@Nb26-EGFP-H6 by pyrolyzing and oxidizing an imidazolating zeolite framework-8 (ZIF-8) to obtain photothermal metal–carbon nanomaterials (Zn-CN) for immobilizing Nb26-EGFP-H6. The rough and porous structure of Zn-CN with a large surface area facilitates the enrichment and immobilization of antibodies. A trimodal lateral flow immunoassay (tLFIA) with colorimetric, fluorescent, and photothermal triple signal outputs was constructed for the detection of aflatoxin B1 (AFB1) in maize. Attractively, the Zn-CN-based tLFIA’s multiplex guarantees accurate and sensitive detection of AFB1, with triple signal detection limits of 0.0012 ng/mL (colorimetric signals), 0.0094 ng/mL (fluorescent signals), and 0.252 ng/mL (photothermal signals). The sensitivity of the trimode immunosensor was 628-fold and 42-fold higher than that of the original Nb26-based ELISA (IC50) and the unimodal LFIA (LOD). This work provides an idea for constructing a sensitive, tolerant matrix and efficient and accurate analytical platform for rapidly detecting AFB1 in food.

An electrochemical aptamer sensor for the sensitive detection of aflatoxin B1(AFB1) in corn samples was developed using nanocomposites loaded with copper nanoparticles (CuNPs) on zeolitic imidazolate framework-8 (ZIF-8), which were modified on a glassy carbon electrode (GCE). The CuNPs@ZIF-8 nanocomposites served as modifying materials for electrodes, exhibiting a high specific surface area and excellent compatibility with aptamers, thereby enhancing the electron transfer rate and increasing the aptamer loading and immobilization efficiency. The electrochemical properties before and after modification were investigated and validated using cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) techniques, while the sensor’s performance was analyzed through quantitative detection via differential pulse voltammetry (DPV). Furthermore, various conditions, including the volume of ZIF-8 dispersion, electrodeposition time of copper nanoparticles, aptamer concentration, and AFB1incubation time, were optimized. The results indicated that the sensor exhibited a wide linear range (10.0 to 1.0 × 106pg/mL), with a lower limit of detection (LOD) of 1.13 pg/mL under optimized conditions, outperforming other reported aptamer sensors. The spiked recoveries in corn samples ranged from 96.663% to 105.72%. In conclusion, this sensor presents a novel solution for low-cost and high-sensitivity detection of AFB1.

A sensitive electrochemical biosensor was designed for determination of aflatoxin B1 (AFB1) using a copper-based metal-organic framework (Cu-MOF), which has strong electrochemical activity and exonuclease III (Exo III)-assisted recycling for dual signal amplification. Hairpin DNA (S1) was immobilized on the electrode. The AFB1 was recognized by aptamer DNA (S2) and complementary DNA (S3) was released. The S3 hybridized with the hairpin S1 to form the Exo III hydrolyzed double-stranded DNA, leaving a partial sequence of hairpin DNA (S1′) on the electrode and releasing S3 for the next cycle of the opening and digestion of hairpin S1. The amplified S1′ then was able to combine with more signal probes. Cu-MOF bond gold nanoparticles (AuNPs) by -NH2 were immobilized to capture DNA (S4) to obtain Cu-MOF/AuNPs/S4. This signal probe Cu-MOF/AuNPs/S4 was able to hybridize with the electrode and generate an amplified electrochemical signal. Under the optimized conditions, this electrochemical biosensor for AFB1 exhibited a low detection limit of 6.7 × 10−7 ng/mL at a signal-to-noise equal to 3 and a wide linear range from 10−6 to 1 ng/mL. The biosensor was also used to analyze AFB1-spiked beer sample with recovery values between 96% and 103%. This method has the potential to become a valuable technology for detecting various toxins by the selection of the appropriate aptamer DNA.

In the present study, for the first time, immunoaffinity columns under the term MIL (Al) −53 deep eutectic solvents @MIPs (MIL(Al)-53-DES@MIPs) were synthesized based on two technologies, metal–organic framework (MOF) and molecular imprinted polymers (MIPs). The MOFs and MIPs with DES used in synthesizing immunoaffinity columns, was subsequently used to extract and preconcentrate aflatoxins AFB1, AFB2, AFG1, and AFG2. One factor at a time (OFAT) method was used to optimize the parameters affecting on extraction efficiency of the target compounds. Under optimized conditions, the limits of detection of the four aflatoxin ranged from 0.023 to 0.033 µg kg −1 (S/N = 5), and the calibration graphs were linear in the concentration range from 0.1 to 400 μg kg −1 , the inter-day and intraday relative standard deviations (RSD) for various aflatoxins at three different concentration level (n = 5) using a single column were 1.3–4.4 %, and the column to column RSD % (n = 5) was 4.3–6.7 % at 5 μg L -1 of each analyte. The method was successfully applied to quantify aflatoxin in cereals samples of Khuzestan province of Iran, with the recoveries between 95.3 and 98.5 %.
