Background: The effects of heat acclimation (HA) on the hypothalamus after exertional heatstroke (EHS) and the specific mechanism have not been fully elucidated, and this study aimed to address these questions. Methods: In the present study, rats were randomly assigned to the control, EHS, HA, or HA + EHS groups (n = 9). Hematoxylin and eosin (H&E) staining was used to examine pathology. Tandem mass tag (TMT)-based proteomic analysis was utilized to explore the impact of HA on the protein expression profile of the hypothalamus after EHS. Bioinformatics analysis was used to predict the functions of the differentially expressed proteins. The differential proteins were validated by western blotting. An enzyme-linked immunosorbent assay was used to measure the expression levels of inflammatory cytokines in the serum. Results: The H&E staining (n = 5) results revealed that there were less structural changes in hypothalamus in the HA + EHS group compared with the EHS group. Proteomic analysis (n = 4) revealed that proinflammatory proteins such as argininosuccinate synthetase (ASS1), high mobility group protein B2 (HMGB2) and vimentin were evidently downregulated in the HA + EHS group. The levels of interleukin (IL)-1β, IL-1, and IL-8 were decreased in the serum samples (n = 3) from HA + EHS rats. Conclusions: HA may alleviate hypothalamic damage caused by heat attack by inhibiting inflammatory activities, and ASS1, HMGB2 and vimentin could be candidate factors involved in the exact mechanism.

Objective Liver fibrosis is a reversible pathological process, and its prevention and treatment hold great significance for patients with chronic liver disease. This study combined 16S rRNA analysis of gut microbiota and serum metabolomics to explore the mechanism of curcumol’s effect on liver fibrosis in mice. The results clarified the relationship between the gut microbiota and metabolites in the process of liver fibrosis. Materials and methods In this study, we randomly divided mice into a control group, a model group, and a curcumol treatment group to analyze the pathological changes in the liver tissue as well as the activities of the toll-like receptor 4 (TLR4)/nuclear factory kappa B (NF-κB) signaling pathway and inflammatory factors, such as tumor necrosis factor (TNF), interleukin 6 (IL-6), and IL-8. The gut microbiota were analyzed by 16 S rRNA sequencing, and serum metabolites were examined by liquid chromatography-mass spectrometry (LC-MS) metabolomic analysis. Results Molecular biological testing found that curcumol could significantly improve the pathological changes of the liver tissue and inhibit the occurrence of liver inflammation. Intestinal flora testing found that curcumol could significantly change the abundances of Veillonellaceae, Prerotella_oulorum, and Alistipes_finegoldii. Metabolomics analysis found that curcumol’s antihepatic fibrosis effect may be related to its regulation of arachidonic acid metabolism. Correlation analysis suggested that curcumol regulated the abundances of Bacteroidota and Bacteroides and participated in the metabolism of Prostaglandin B2. Conclusions When liver fibrosis occurs, the intestinal flora and metabolic network are altered. The effect of curcumol on liver fibrosis may be related to its regulation of intestinal flora and the resulting interference with metabolic pathways, thereby reducing liver inflammation.

Aflatoxins are common food contaminants that pose serious public health risks. This study presents a straightforward, eco-friendly hydrothermal method for synthesizing carbon quantum dots (CQDs) with high selectivity for aflatoxins detection. The CQDs with a 48% quantum fluorescence yield show a strong linear fluorescence response to toxin concentration, achieving a detection limit of 0.16–0.29 ng/mL. Furthermore, the total aflatoxins in citri reticulatae pericarpium (CRP) and coicis seed (CS) were quantified, using a relative correction factor based on the AFB1 standard curve. The method, combined with immunoaffinity column pretreatment, effectively eliminated interferences and enabled accurate quantification, while reducing assay costs. The relative standard deviation (RSD) was below 8%, and spiked recoveries ranged from 99.02% to 106.5%, matching with those obtained using chromatographic methods. This approach provides a rapid, sensitive, and reliable solution for aflatoxins screening in food, enhancing detection efficiency and accuracy, for food safety and applications.

Objective:
Protein kinase B2 (AKT2) is implicated in cardiomyocyte survival during various stress conditions. However, the role of AKT2 in heart function, cardiac hypertrophy and blood pressure (BP) control during hypertension is not fully understood. Therefore, we sought
to determine whether the deletion of AKT2 protects cardiac function during angiotensin II/high-salt-diet (AngII/HSD) treatment and find out the signaling pathway. Methods:
Male C57BL/6J (wild type), AKT2 knockout and interleukin (IL)-6 knockout
mice were fed a 4% NaCl diet for 5 weeks. In the last week, mice were split in two groups and infused subcutaneously with either vehicle or AngII (1.5 μg/h per mouse) for 1 week. Then, BP and cardiac function were assessed. Immunohistology of IL-6 and monocyte chemoattractant protein
1 was performed to detect inflammation in the heart. Masson's trichrome staining was performed to evaluate cardiac fibrosis. Heart tissue homogenates and neonatal mice cardiomyocytes were collected to analyze oxidative stress. Results:
Compared
with wild-type mice, AKT2 knockout mice maintained BP and showed better left ventricle ejection fraction, lower level of fibrosis, reduced oxidative stress, reduced IL-6 expression and less macrophage infiltration, when treated with AngII/HSD. IL-6 knockout mice treated with AngII/HSD also
showed alleviated left ventricular function, fibrosis, oxidative stress and macrophage infiltration compared with wild type. Conclusion:
AKT2 deficiency prevents the development of AngII/HSD-induced hypertension, cardiac dysfunction
and myocardial injury including oxidative stress, fibrosis and inflammation by suppressing IL-6 expression. These data reveal an important role of the AKT2-IL-6 pathway in mediating AngII/HSD-induced hypertension and cardiomyopathy.

Purpose: To investigate the impact of grape seed proanthocyanidin B2 (GSPB2) pretreatment on hypoxia-reoxygenation model of HK-2 cells in vitro . Methods: The experiment was divided into five groups: control group (control), GSPB2 group (GSPB2), hypoxia-reoxygenation group (HR), GSPB2 + HR group (GSPB2+HR), and GSPB2 + brusatol (BRU) + HR group (GSPB2 + BRU + HR). Flow cytometry was used to detect apoptosis of cells. Transmission electron microscopy was employed to observe ultrastructural changes of cells. Mitochondrial membrane potential (MMP) was measured. Cellular immunofluorescence was used to assess intracellular Ca ²+ concentration. Immunofluorescence staining and Western blotting were conducted to detect expression levels of Nrf2, HO-1, glucose-regulated protein 78 (GRP78), C/EBP homologous protein (CHOP), and cleaved-caspase3. Results: Compared to HR group, GSPB2 + HR group showed significantly increased cell viability, and reduced mitochondrial damage in the cytoplasm. MMP in GSPB2 + HR group was significantly restored, and intracellular Ca ²+ concentration was significantly decreased. The expression of Nrf2 and HO-1 proteins was significantly upregulated, while the expression of GRP78, CHOP, and cleaved-caspase3 proteins was markedly downregulated. Conclusion: GSPB2 pretreatment can alleviate oxidative stress damage, mitochondrial injury, and endoplasmic reticulum stress induced by hypoxia-reoxygenation in HK-2 cells in vitro . This effect may be related to the ability of GSPB2 pretreatment to activate the endogenous antioxidant system, particularly through the activation of the Nrf2/HO-1 signaling pathway.

Gillespite-type SrCu 0.95 B 0.05 (B 2+ :Cu,Co,Mn,Ni,Mg,Zn)Si 4 O 10 (SCBS) ceramics were prepared by high-temperature solid-state method. Their microwave dielectric properties and crystal structure evolution were studied by P–V-L theory, bond valence model, and X-ray photoelectron spectroscopy. Varying B 2+ ion changed ionic polarizability and bond ionicity, causing dielectric constant ε r of SCBS to change. The change in quality factor Q × f was explained by lattice energy, oxidation state of Cu, and internal strain/fluctuation. The temperature coefficient of resonator frequency τ f was regulated by B–O bond length and valence. The optimal microwave dielectric properties were exhibited by SrCu 0.95 Mn 0.05 Si 4 O 10 after sintering at 1050 °C: ε r ∼5.8 ± 0.03, Q × f ∼65,589 ± 2611 GHz (@15.8 GHz), and τ f ∼ -50 ± 4 ppm/°C. These materials show great potential application prospects for 5G/6G wireless communication.

Low-temperature sintering and phase/microstructure tailoring of multi-element ultra-high temperature ceramics remain challenging, restricting the improvement of mechanical properties and the development of ultra-high temperature ceramic materials. In this work, a novel low-temperature reactive sintering technology was used to tailor the content of multiple borides and carbides in (Hf, Ta)B 2 -(Hf, Ta)C-SiC ceramics. It was found that the introduction of borides promoted the densification process of multi-phase ceramics. With the increase of borides content, the grain size of multi-phase ceramics gradually increased. The introduction of moderate amount borides helped to form a uniformly refined microstructure. When the molar ratio of borides to carbides was 2:1, the flexural strength and fracture toughness of multi-phase ceramics were the highest, which were 487.57 ± 38.3 MPa and 5.04 ± 0.09 MPa·m 1/2 , respectively. The main strengthening and toughening mechanisms included pullout of (Hf, Ta)B 2 grains and crack deflection.

It is crucial to address the challenges for low-temperature densification and interfacial reactions during the fabrication of multi-element ultra-high temperature ceramic matrix composites. In this paper, C sf /(Hf, Ta)B 2 -(Hf, Ta)C-SiC composites were prepared via reactive hot-press sintering at low temperatures. The intrinsic connection between the fiber content, microstructure, and mechanical behavior of the composites was elucidated. The results indicated that the ultrasonic dispersion combining with fast hot-press sintering effectively promoted the uniform dispersion of C sf in the matrix and achieved densification at 1700 °C. The composites exhibited an undamaged fiber-matrix interface without observable interfacial phase, demonstrating that low-temperature preparation effectively prevented fiber degradation from interfacial reactions. Compared to the unreinforced ceramics, the composites showed decreased flexural strength and fracture toughness, yet possessed higher work of fracture and larger critical crack size. This indicated that fiber addition improved the damage tolerance and structural reliability by inhibiting the catastrophic brittle fracture behavior inherent to monolithic ceramics. The composite with 20 vol% C sf offered the best balance of strength, toughness and non-catastrophic failure behavior. The flexural strength and fracture toughness were 260.6 MPa and 4.82 MPa m 1/2 , respectively. Download: Download high-res image (531KB) Download: Download full-size image

This is the first report on a polysaccharide (RCJ2-Ib) isolated from Rosa Chinensis Flos. RCJ2-Ib was obtained through the extraction with water, precipitation with ethanol, separation with DEAE-52 column and purification with DEAE-Sepharose Fast Flow column and Sephadex G100 column. GC, FT-IR and NMR analyses revealed that RCJ2-Ib (3.3 kDa) was a 1,4-linked polymannuronic acid containing substantial β - D -anomers units. The anticoagulant effect of RCJ2-Ib evaluated by using rabbit ear venous blood and an acute blood stasis rat model showed that RCJ2-Ib had obvious anticoagulant activity in regulating endogenous and exogenous coagulation pathways and reducing serum thromboxane B2 and endothelin-1. In addition, RCJ2-Ib could also increase the number of Lactobacillus and Escherichia coli . As a result, RCJ2-Ib has the potential to inhibit thrombosis and maintain the intestinal environment.

The properties of molybdenum diboride are intricately tied to its crystal structure, which includes two distinct phases, α-MoB 2 and β-MoB 2 . The α-MoB 2 exhibits superior electrical and catalytic properties, akin to those of graphite due to the similar HCP structure. Nevertheless, the transformation between α-MoB 2 and β-MoB 2 is complicated and the formation of α-MoB 2 remains challenge due to higher Gibbs free energy. The MoB 2 powder synthesized through boro/carbothermal reduction in this study suggests the β phase, transforming into α-MoB 2 completely via a high-entropy strategy. It founds that the introduction of high-entropy diborides (HEB) effectively inhibits the grain growth of samples, resulting in a significant enhancement in their hardness. The potential for the formation of a single-phase between MoB 2 and HEB is associated with Pauling electronegativity difference, δ χP . Furthermore, the formation of a single-phase solid solution was found to contribute to the improvement in the fracture toughness of the samples. This work presents a novel strategy for stabilizing α-MoB 2 , offering valuable insights into understanding phase-transition behavior in MoB 2 and the solid solubility in multi-component ceramics.

Diabetes is a chronic disease, and long term hyperglycemia and oxidative stress in diabetes patients will lead to non-enzymatic glycosylation of proteins and eventually produce advanced glycation end products (AGEs). The non-enzymatic glycosylation of proteins and the formation and accumulation of AGEs in blood vessels and tissues are closely related to the complications of diabetes. Here, the anti-glycation mechanisms of four natural flavonoids, baicalein (BA), baicalin (BE), catechin (CT), and proanthocyanidinB2 (PB2), are studied using a bovine serum albumin (BSA) glycosylation model. The results show that fructose may induce non-enzymatic glycosylation of BSA with a significant increase in glycation stage products, increase aggregation and decrease hydrophobicity. In the presence of reactive oxygen species H 2 O 2 , it exacerbates non-enzymatic glycosylation of BSA and causes more severe damage to its function and structure. BA, BE, CT, and PB2 may inhibit non-enzymatic glycosylation of BSA, exert anti-glycosylation stage products, anti-aggregation, inhibit hydrophobicity reduction, and protect BSA structure. BA/BE/CT/PB2 forms a complex with BSA to statically quench the endogenous fluorescence of BSA. Molecular docking and molecular dynamics simulation are used to further confirm that the main binding forces between BSA and flavonoids are hydrophobic, but there are also hydrogen bonding and electrostatic interactions.

Background The root of Salvia miltiorrhiza f. alba (RSMA) (Lamiaceae) is used for the treatment of patients with thromboangiitis obliterans (TAO) in traditional Chinese medicine . Previously, a mixture of phenolic acids extracted from RSMA has shown significant protective effects on TAO rats. Purpose This study investigates the inhibitory effects of salvianolic acid B on TAO induced by sodium laurate injection in rats to explore the effective constituents of RSMA in TAO treatment. Methods TAO rats were developed using injected sodium laurate. After treatment with ligustrazine hydrochloride (15 mg/kg) and various doses of salvianolic acid B (10, 20, 40 mg/kg) by tail intravenous injection , levels of thromboxane B 2 (TXB 2 ), 6-keto-prostaglandin F 1α (6-keto-PGF 1α ) and endothelin-1 (ET-1) in plasma were determined using enzyme-linked immunosorbent assay. The right femoral arteries were studied by hematoxylin and eosin staining and immunohistochemical analysis to determine pathological changes and overexpression of tumor necrosis factor (TNF)–α and inducible nitric oxide synthase (iNOS) in the femoral artery walls of TAO rats. Results Salvianolic acid B significantly decreased the expressions of TXB 2 and ET-1 and increased the expression of 6-keto-PGF 1α in plasma, and significantly inhibited the overexpression of TNF-α and iNOS in the femoral artery walls of TAO rats at medium and high doses. Conclusion Salvianolic acid B has a protective effect on TAO rats. The mechanism may involve inhibition of thrombosis and TAO-associated inflammatory responses, which may explain the success of RSMA treatment of TAO in humans in traditional Chinese medical practice. Hence, it may be a potential drug for TAO treatment in conventional medicine.
