COMT1 overexpression resulting in increased melatonin biosynthesis contributes to the alleviation of carbendazim phytotoxicity and residues in tomato plants

Yanyan Yan,Shasha Sun,Ning Zhao,Wanying Yang,Qinghua Shi,Biao Gong

Melatonin (Mel) serves as an important signalling molecule in various aspects of stress tolerance in plants. However, the function of Mel in pesticide metabolism remains unknown. Here, selecting the widely used fungicide carbendazim (MBC) as the model, we found that exogenous Mel had the ability to alleviate pesticide phytotoxicity and residues in tomato as well as in some other vegetables. Additionally, overexpression of the Mel biosynthetic gene caffeic acid O-methyltransferase 1 ( COMT1 ) significantly enhanced the capacity of the tomato to reduce MBC phytotoxicity and residue. This outcome was mainly because of the Mel-induced antioxidant capability, as well as the key detoxification process. Indeed, levels of reactive oxygen species (ROS) and lipid peroxides significantly decreased after applying exogenous Mel or overexpressing COMT1 , which resulted from direct ROS scavenging, and increased Mel levels significantly enhanced antioxidant enzymatic activity. More importantly, Mel activated the ascorbate-glutathione cycle to participate in glutathione S-transferase-mediated pesticide detoxification. A grafting experiment showed that rootstocks from COMT1 transgenic plants increased the Mel accumulation of wild-type scions, resulting in MBC metabolism in the scions. To our knowledge, this is the first report providing evidence of Mel-induced pesticide metabolism, which provides a novel approach for minimizing pesticide residues in crops by exploiting plant self-detoxification mechanisms.

A highly sensitive detection of carbendazim pesticide in food based on the upconversion-MnO2 luminescent resonance energy transfer biosensor

Qin Ouyang,Li Wang,Waqas Ahmad,Yawen Rong,Huanhuan Li,Yuqian Hu,Quansheng Chen

Carbendazim (CBZ) pesticide residues in food products have become a growing concern in recent years. Herein, a sensitive biosensor for detecting CBZ was developed based on luminescent resonance energy transfer (LRET) from aptamer labeled upconversion nanoparticles (UCNPs, donor) to manganese dioxide (MnO 2 , acceptor) nanosheets. The strong overlap between the absorption spectrum of MnO 2 and the UCNPs fluorescence emission allowed the luminescence quenching. With the addition of CBZ, it tended to bind with specific aptamers, which culminated in the UCNPs-aptamer dropping off MnO 2 nanosheets and restoring the fluorescence. A linear calibration plot between logarithmic CBZ concentration and fluorescence intensity was acquired in the range of 0.1–5000 ng·mL −1 , with a limit of detection 0.05 ng·mL −1 , indicating that the UCNPs- MnO 2 aptasensor is a rapid, sensitive and specific quantitative detection platform for CBZ. Furthermore, the precision and accuracy of the developed LRET biosensor was validated by HPLC method with no significant differences.

Sub-chronic carbendazim exposure induces hepatic glycolipid metabolism disorder accompanied by gut microbiota dysbiosis in adult zebrafish (Daino rerio)

Zhiwei Bao,Yao Zhao,Anyi Wu,Ze Lou,Huahui Lu,Qianxuan Yu,Zhengwei Fu,Yuanxiang Jin

Carbendazim (CBZ) as a broad spectrum fungicide is widely used in the whole world to contorl plant diseases. With the application of CBZ in the agriculture, it has been detected in vegetables and fruits. Nowadays, it even has been detected in the watercourse and indoor dust. However, the toxic effects of CBZ on aquatic organisms have received limited attention. In this study, male adult zebrafish were exposed at 0, 30 and 100 μg/L CBZ for 21 days to assess its effects on hepatic glycolipid metabolism. After exposure, the body weight and length decreased, but the condition factor increased significantly. Some hepatic biochemical parameters including the levels of glucose, pyruvate, low density lipoprotein (LDL) and triglyceride (TG) decreased significantly in the liver of zebrafish after exposure with CBZ. Two transaminases alanine transaminase (ALT) and aspartate transaminase (AST) also increased significantly, indicating that subchronic CBZ exposure influenced the liver function. Moreover, the relative mRNA levels of some key genes related to the glycolysis and lipid metabolism in the liver also changed significantly. Furthermore, the transcriptome analysis showed that the carbon metabolism, lipid metabolism and detoxification metabolism were also affected in the liver of CBZ exposed zebrafish. Interestingly, we also found the amounts of the Firmicutes , Bacteroidetes , Actinobacteria , α-Proteobacteria , γ-Proteobacteria and Verrucomicrobia at phylum level significantly decreased in the gut. Sequencing V3-V4 region of 16S rRNA also demonstrated gut microbiota composition changed significantly according to weighted UniFrac distance analysis. Consequently, subchronic CBZ exposure induced hepatic metabolic disorder accompanied by gut microbiota dysbiosis in adult male zebrafish.

Facile Method to Fabricate Cactus-like Ag NPs/CuO/Cu2O Nanocomposites for Recyclable SERS Detection of Trace Carbendazim Residues

Ya Lu,Zhuanfang Bi,Guangyi Shang

Detection of trace carbendazim in agricultural products or environment is challenging but important for human health and environmental protection. Developing a sensitive and recyclable surface-enhanced Raman scattering (SERS) substrate is one of the effective methods to detect such pesticide residues. In this work, we developed a facile method to fabricate a three-dimensional cactus-like Ag nanoparticles/CuO/Cu2O nanocomposite (CACN) based on a galvanic replacement reaction between Tollens’ reagent and Cu foam. This design combined the Raman enhancement effect and photocatalytic activity, providing a multifunctional and reusable substrate for highly sensitive SERS detection. It was found that the enhancement and photocatalysis of the CACN substrate were closely related to the concentration of Tollens’ reagent, and the 10-CACN substrate (prepared at 10 mM) demonstrated excellent sensitivity, reproducibility, and recyclability. The substrate was successfully applied for the detection of carbendazim solution with a detection limit of 1.5 × 10–10 M and could be reused up to 7 times. More importantly, the substrate can be employed for the detection of trace carbendazim in real apple peels with a detection limit of ∼8.5 × 10–4 ppm, which is far below that of 2.0 × 10–1 ppm suggested by China, United Kingdom, and European Union. These results suggest that the facile fabrication method and the excellent substrate would facilitate the SERS technology closer to practical application in the detection of pesticide residues.

Insights into enhanced toxic effects by the binary mixture of carbendazim and procymidone on hepatic lipid metabolism in mice

Zhiwei Bao,Dou Wang,Yao Zhao,Ting Luo,Guilin Yang,Yuanxiang Jin

Carbendazim (CBZ) and procymidone (PRO) are two widely used fungicides in agriculture. However, there are still gaps in knowledge regarding about the potential hazards of joint exposure to CBZ and PRO in animals. Here, 6-week-old ICR mice were exposed to CBZ, PRO and CBZ + PRO for 30 days, and metabolomics were performed to discover the mechanism by which the mixture enhanced the effects on lipid metabolism. Co-exposure to CBZ + PRO elevated the body weights, relative liver weights and relative epididymis fat weights, but not in the single exposure groups. Molecular docking analysis suggested that CBZ and PRO combined with peroxisome proliferator-activated receptor (PPARγ) at the same amino acid site as the agonist rosiglitazone. The RT-qPCR and WB results demonstrated that the levels of PPARγ were higher in the co-exposure group than in the single exposure groups. In addition, hundreds of differential metabolites were discovered by metabolomics and enriched in different pathways, such as pentose phosphate pathway and purine metabolism. A unique effect, a decrease in glucose-6-phosphate (G6P) that promoted more NADPH production, was observed in the CBZ + PRO group. These results demonstrated that exposure to CBZ + PRO caused more serious lipid metabolism disorder in the liver than exposure to a single fungicide, which could provide some new insight for the toxic effects after fungicides joint exposure.

Simultaneous determination of imidacloprid and carbendazim in water samples by ion chromatography with fluorescence detector and post-column photochemical reactor

Qamar Subhani,Zhongping Huang,Zuoyi Zhu,Yan Zhu

A new analytical method has been developed and validated for the simultaneous determination of pesticides from different classes using ion chromatography-online photochemical derivatisation-fluorescence detector (IC- hv -FD). Fluorimetric detection was performed at λ ex / λ em =332   nm/367   nm for imidacloprid and then detector was set at λ ex / λ em =247   nm/470   nm for carbendazim . The two pesticides imidacloprid and carbendazim were successfully separated isocratically on an IonPac ® AS11 (250   mm×4   mm i.d; 13   µm particle size, Dionex) anion-exchange column using 40   mM KOH with 10% (v/v) acetonitrile and pumped at a flow rate of 1.0   mL   min −1 . Under the optimized conditions, the limit of detection (LOD, S/N=3) of imidacloprid and carbendazim were 7.8   µg   L −1 and 67   µg   L −1 , respectively. The experimental results showed that there was good linearity with a correlation coefficient ( r )≥0.9966 over the range of 0.05–10   mg   L −1 for imidacloprid and 0.2–15   mg   L −1 for carbendazim. Good reproducibility with a relative standard deviation (RSD, n =7) less than 4.5%. Finally, the proposed method was applied with satisfactory results to the analysis of these pesticides in ground water, lake water and river water without any pre-treatment of samples. The average spiked recoveries were in the range of 90–104%.

Cation distribution and defect engineering in spinel oxides MCo2O4 (M = Zn and Cu) toward efficient electrocatalytic interfaces for carbendazim sensing

Nguyen Ngoc Huyen,Ngo Xuan Dinh,Pham Thi Tuyet Nhung,Ong Van Hoang,Tran Van Tan,Nguyen Duc Toan,De Pham-Cong,Pham Duc Thang,Anh-Tuan Le

Tunable cation distribution in spinel MCo2O4 enhances surface redox activity. • Microwave-assisted hydrothermal yields ZCO nanosheets and CCO nanorods with defects. • Oxygen vacancies and mixed cations boost interfacial charge transfer kinetics. • CCO/SPE shows high sensitivity and low LOD for Carbendazim detection. • Strong Carbendazim adsorption on spinel surfaces confirmed by experiment and theory. Spinel cobaltites MCo 2 O 4 (M is a divalent metal ion) nanomaterials have emerged as promising electrocatalysts for electrochemical sensing owing to their tunable cation distribution and redox-active metal centers. In this work, ZnCo 2 O 4 (ZCO) nanosheets and CuCo 2 O 4 (CCO) nanorods were prepared using a simple microwave-assisted hydrothermal method and subsequently employed to modify screen-printed electrodes (SPEs) for carbendazim (CBZ) detection. Structural analysis revealed distinct cation arrangements within the spinel lattice, particularly CCO exhibited mixed Cu/Co occupancy and abundant oxygen vacancies. Both modified electrodes significantly enhanced the electrochemical oxidation of CBZ compared with bare SPE, primarily due to the catalytic activity of the Co 3+ /Co 2+ redox couple. Notably, the CCO/SPE demonstrated superior sensing performance, arising from the synergistic effects of Cu 2+ /Cu + redox activity, high defect density, and one-dimensional morphology that promoted charge transfer and mass diffusion. The CCO/SPE sensor delivered promising analytical performance, featuring two linear ranges (0.1–10 and 10–500 µM), a detection limit of 0.07 µM, and a sensitivity of 5.97 µA µM −1 cm −2 . Combined experimental and theoretical analyses suggest strong CBZ adsorption on the MCo 2 O 4 spinel surface, highlighting spinel oxides as efficient electrocatalysts for CBZ pesticide sensing. Dual-Emission Carbon Dots Hybrid for Ratiometric Fluorescence and Visual On-Site Detection of Carbendazim Residues in Fruits and Vegetables

Yong Han,Xiaoyu Luo,Chengqian Li,Yanxin Ma,Chenxi Jiang,Fei Shang,Zhonghong Li,Hui Wang

A ratiometric fluorescence sensor based on dual-emissive carbon dots (d-CDs) and a smartphone-assisted colorimetric platform was developed for the sensitive visual detection of carbendazim (MBC) in complex food matrices. The d-CDs were obtained by hybridizing blue-emissive CDs (B-CDs) and red-emissive CDs (R-CDs), showing emission peaks at 430 and 610 nm under 365 nm excitation. In the presence of MBC, the 430 nm emission was quenched by π–π stacking and photoinduced electron transfer (PET), while the 610 nm emission increased due to PET inhibition through amino-MBC coordination. The fluorescence intensity ratio (F 430 /F 610 ) showed a linear response to MBC from 0 to 2.5 μM, with a detection limit of 0.0209 μM, recoveries of 93.1%–108.7%, and relative standard deviations below 3.5% in real food samples. The smartphone-assisted method exhibited a distinct color change from dark blue to magenta under a portable UV lamp as the MBC concentration increased for the d-CDs, allowing for instrument-free visual semiquantitative analysis with a detection limit of 0.0700 μM. This work contributes to a reliable, portable, and economical method for quantitative and semiquantitative detection of MBC residues in fruits and vegetables, supporting real-time food safety monitoring.

Between intestinal inflammation and renal disulfidptosis caused by carbendazim exposure in common carp: Insights into the potential gut-kidney axis

Yan Wang,Lingyu Zhu,Sijin Zhou,Xinchi Shang,Yuanhang Zhao,Yuhao Liu

Carbendazim induces renal disulfidptosis in common carp. • Carbendazim triggers intestinal inflammation and gut microbiota dysbiosis. • Inflammatory cytokines are elevated in the intestine, serum, and kidney, may act as mediators of gut-kidney cross-talk. • IL-1β may drive disulfidptosis under carbendazim stress. Carbendazim is a widely used fungicide and a pervasive water contaminant detected in aquaculture systems worldwide, yet its chronic toxicity mechanisms in farmed fish remain poorly understood. Here, we exposed common carp ( Cyprinus carpio ), a globally significant aquacultural species, to environmentally relevant concentrations of carbendazim (10 and 20 μg/L) for 30 days. We found that carbendazim dose-dependently reduced weight gain (WG), weight gain ratio (WGR), and specific growth rate (SGR). Histology revealed both intestine and kidney damage. Intestinal transcriptomics showed significant up-regulation of genes in toll-like receptor, cytosolic DNA-sensing, RIG-I-like receptor, and NOD-like receptor pathways, meaning that carbendazim induced inflammation in intestines. Metagenomics revealed expansion of pro-inflammatory bacteria ( Aeromonas , Plesiomonas , Acinetobacter ) and enrichment of microbial immune-evasion genes, suggesting that the inflammatory response may be amplified by gut microbiota dysbiosis. In the kidney, carbendazim induced inflammation and altered disulfidptosis markers: SLC7A11 , GYS1 , abi2a , TLN1 , NDUFA11 , and NDUFB10 . The inflammatory cytokines IL-1β, TNF-α, and IL-6, which are elevated in the intestine, serum, and kidney, may serve as mediators of gut-kidney communication. Furthermore, in vitro epithelioma papulosum cyprini (EPC) cell experiments revealed that carbendazim increased both IL-1β and disulfidptosis markers. siRNA knockdown of IL-1β significantly attenuated the changes in all the disulfidptosis-related factors, demonstrating a potential mediator role of this cytokine. While our correlative in vivo data and mechanistic in vitro findings suggest cytokine-mediated cross-talk, we acknowledge that definitive proof of the renal IL-1β originating specifically from the gut awaits lineage-tracing or organ-specific neutralization studies. This is the first study to establish a link between pesticide exposure, gut-kidney axis perturbation, and disulfidptosis in an aquatic vertebrate, providing testable biomarkers and mechanistic hypotheses for future causality validation.

Signal switching electrochemical and fluorescence dual-mode sensing platform for carbendazim determination based on “two-in-one” magneto-fluorescent Cdots

Yuzhen Jia,Yuanmeng Ke,Zhixiang Liu,Huaixia Yang,Mingsan Miao,Liang Guo

An innovative method for carbendazim (CBZ) detection was developed, consisting of an electrochemical-fluorescence dual-mode biosensor based on magneto-fluorescent composite M-CDs. M-CDs, as the fluorescent probe of this sensor, could combine the electrical signal-ferrocene to achieve the “signal switching” by specifically recognizing CBZ through aptamers, of which magnetic property was used to quickly separate from complex substrates without interference. The dual-mode sensor based on M-CDs demonstrated excellent linear responses in both electrochemical and fluorescence assays. It achieved detection ranges of 10 fg/mL − 300 ng/mL and 60 fg/mL – 100 ng/mL with detection limits (LODs) of 1.4 fg/mL and 2.3 fg/mL. The sensor exhibited exceptional detection performance, stability and anti-interference. In addition, the results of the sensor in actual samples were consistent with those of enzyme-linked immunosorbent assay (ELISA), which further demonstrated that the sensor could accurately trace detecting CBZ in real samples and had a certain application prospect.

Health risks of chlorothalonil, carbendazim, prochloraz, their binary and ternary mixtures on embryonic and larval zebrafish based on metabolomics analysis

Guiling Yang,Yanhua Wang,Jian Li,Dou Wang,Zhiwei Bao,Qiang Wang,Yuanxiang Jin

Chlorothalonil (CTL), carbendazim (CBZ), and prochloraz (PCZ) are fungicides widely used in many countries. The use of these fungicides raises concerns because they are often applied together or used in the same agricultural area. However, the toxicity of these fungicides or mixtures, especially to aquatic organisms, has received limited attention. Here, embryonic and larval zebrafish were exposed to indicated concentrations of CTL, CBZ, and PCZ and their binary (CTL+CBZ, CTL+PCZ and CBZ+PCZ) and ternary (CTL+CBZ+PCZ) mixtures for 24 h. Based on metabolomics analysis, we observed that hundreds of metabolites were altered, and glycolysis metabolism and amino acid metabolism were the two most affected pathways. Interestingly, a total of 9 and 26 metabolites changed significantly in embryos and larvae treated with all fungicides, respectively. Among these altered metabolites, 2-aminoadipic acid (2-AAA) levels increased significantly in all groups, indicating that 2-AAA potentially represents a useful biomarker for evaluating the toxicity of fungicides. Furthermore, the joint effects of CTL+PCZ on embryos and larvae, especially on amino acid metabolism, were weaker than those in other groups, but combined treatment did not influence individual fungicidal activity. Data acquired from metabolomics provided important insight for understanding the mechanism by which fungicides or their mixtures affect zebrafish.

Colorimetric determination of carbendazim based on the specific recognition of aptamer and the poly-diallyldimethylammonium chloride aggregation of gold nanoparticles

Song Wang,Lantian Su,Lumei Wang,Dongwei Zhang,Guoqing Shen,Yun Ma

This paper proposes the idea of establishing carbendazim (CBZ) colorimetric determination in spiked water samples by specific aptamers of unlabeled carbendazim (CBZ), gold nanoparticles (AuNPs) and cationic polymer poly-diallyldimethylammonium chloride (PDDA). In the absence of CBZ, the CBZ aptamer will react with the cationic polymer PDDA by electrostatic interaction to form a complex structure. Therefore, the gold nanoparticles will remain dispersed due to the lack of PDDA. However, when CBZ is added into the sensory system, the CBZ-specific aptamer can selectively capture CBZ to form a stable complex structure. Due to the consumption of the aptamer, PDDA is unable to interact with the aptamer and begins to induce aggregation of AuNPs, thereby causing the color of the solution to change from red to blue. Colorimetric determination of CBZ based on the specific recognition of aptamer and the PDDA-induced aggregation of AuNPs has a detection limit of 2.2 nM, a linear range (R = 0.9960) from 2.2 to 500 nM. The method has good sensitivity and specificity, and the average recovery of CBZ is 94.9–104.8% in the application of actual water samples. This colorimetric method is simple, time-saving and low requirements for equipment, therefore, it holds great potential for CBZ detection in the environmental water samples.
