A cancer-specific activatable theranostic nanodrug for enhanced therapeutic efficacy via amplification of oxidative stress

Xie-an Yu,Mi Lu,Yingping Luo,Yiting Hu,Ying Zhang,Zhiming Xu,Shuaishuai Gong,Yunhao Wu,Xiao-Nan Ma,Bo-Yang Yu,Jiangwei Tian

Rationale: Despite considerable advances, the reactive oxygen species (ROS)-mediated cancer treatment suffers from the problems of up-regulation of adaptive antioxidants in cancer cells as well as side effects to normal cells. Therefore, development of a new generation of cancer-specific nanomedicine capable of amplifying oxidative stress would be of great interest for accurate and effective cancer treatment. Methods: Herein, transferrin (Tf)-decorated, dihydroartemisinin (DHA), L -buthionine-sulfoximine (BSO), and CellROX-loaded liposomal nanoparticles (Tf-DBC NPs) were developed for precise cancer theranositcs. Tf-DBC NPs could specifically recognize cancer cells via Tf-Tf receptor binding and be uptaken into the lysosomes of cancer cells, where Tf-DBC NPs were activated to release Fe(II), DHA, and BSO. ROS was generated by DHA in the presence of Fe(II), and GSH was depleted by BSO to disrupt the redox balance in cancer cells. Furthermore, CellROX, as a fluorescent probe for imaging of intracellular oxidative stress, was used to monitor the therapeutic efficacy. Results: The integration of Tf, DHA, and BSO into the acidic pH-responsive liposomes selectively and effectively killed cancer cells and prevented the oxidative injury to normal cells. The high oxidative state was visualized at the tumor site and the amplification of oxidative stress enabled tumor eradication by Tf-DBC NPs, demonstrating the successful implementation of this novel strategy in vivo . Conclusion: Our study provides a new paradigm for the design of ROS-mediated therapeutics and offers a promising perspective for precise cancer treatment.

Alleviation effect of lutein Pickering emulsion formed by casein-dextran conjugates through Maillard reaction against blue light retinal degeneration

Kangjing Liu,Yu Li,Jiaxuan Li,Xiaoting Yu,Xu Zhong,Wentao Su,Mingqian Tan

With the increasing prevalence of electronic devices, awareness of the risks linked to blue light exposure has significantly heightened. Lutein, a powerful antioxidant, safeguards eye tissue by filtering blue light, while supplementation with docosahexaenoic acid (DHA) enhances retinal function. Adequate intake of these nutrients can help reduce the potential damage from prolonged blue light exposure. The protective effects of lutein and algal oil stabilized with Pickering emulsion were investigated using casein-dextran (CD) conjugates via Maillard reaction. Microstructural analysis revealed a three-dimensional network structure surrounding oil droplets formed by CD conjugates. With the increase of the oil phase ratio from 55 % to 80 %, the average size of Pickering emulsion droplets decreased. Pickering emulsion demonstrated higher viscoelasticity, excellent recovery, thixotropy, and good thermal stability as the oil phase ratio increased. The retention of lutein in CD-75 % Pickering emulsions showed significant improvement under various conditions. Simulated gastrointestinal digestion demonstrated that CD-75 % Pickering emulsions effectively enhanced the lutein bioaccessibility from 19.97 % to 48.99 %. In vivo experiments showed that lutein-loaded Pickering emulsion could effectively relieve blue light-induced retinal degeneration in mice. These findings suggested that Pickering emulsion can serve as a delivery system to protect lutein, offering a nutritional intervention to mitigate blue light-induced retinal degeneration.

Boosting middle hydroxyl adsorption on lignin-derived carbon loaded MnO–Ni for glycerol conversion to dihydroxyacetone

Haichao Zhou,Jianglin Liu,Dalang Chen,Yanlin Qin,Tiejun Wang,Xueqing Qiu,Xuliang Lin

The electrochemical conversion of glycerol into dihydroxyacetone (DHA) under mild conditions efficiently addresses glycerol surplus, enhancing its economic value. However, DHA production efficiency remains constrained by steric hindrance from secondary hydroxyl groups and sluggish glycerol electro-oxidation reaction (GOR) kinetics in weakly alkaline conditions. Herein, lignin-derived carbon-supported MnO–Ni heterojunction catalyst (MnO–Ni/C) is synthesized via self-assembly precipitation and in situ pyrolysis. The optimized MnO–Ni/C catalyst requires a low potential of 1.58 V to achieve 10 mA cm −2 , 270 mV lower than the oxygen evolution reaction (OER). It attains a 53.37% DHA selectivity with a high production rate of 864.75 mmol m −2 h −1 at 1.8 V. MnO–Ni/C heterostructures modulate the local electronic structure of Ni sites, optimizing glycerol adsorption configuration and enhancing selective adsorption of secondary hydroxyl groups and oxygen active species near the electrode surface. These synergistic effects significantly improve GOR kinetics and DHA production efficiency.

Gastric Acid-Responsive ROS Nanogenerators for Effective Treatment of Helicobacter pylori Infection without Disrupting Homeostasis of Intestinal Flora

Jiayin Yu,Zhihao Guo,Jiachang Yan,Changxin Bu,Chang Peng,Cuie Li,Rui Mao,Jian Zhang,Zhi Wang,Shi Chen,Meicun Yao,Zhiyong Xie,Chuan Yang,Yi Yan Yang,Peiyan Yuan,Xin Ding

Helicobacter pylori ( H. pylori ) has infected more than half of the world's population, and is the major cause of gastric cancer. The efficacy of standard antibiotic-based triple therapy is declining due to drug resistance development. Herein, a pH-responsive reactive oxygen species (ROS) nanogenerator (Fe-HMME@DHA@MPN) composed of acid-responsive metal polyphenol network (MPN) shell and mesoporous metal-organic nanostructure core [Fe-HMME (hematoporphyrin monomethyl ether, sonosensitizer)] loaded with dihydroartemisinin (DHA) is reported. These nanoparticles generate more ROS singlet oxygen than sonosensitizer HMME under ultrasonication, and this sonodynamic process is fueled by oxygen generated through Fenton/Fenton-like reactions of the degraded product in gastric acid Fe (II) and hydrogen peroxide (H 2 O 2 ) in the infection microenvironment. The encapsulated DHA, as a hydroperoxide source, is found to enhance the peroxidase-like activity of the Fe-HMME@DHA@MPN to generate ROS hydroxyl radical, beneficial for the microenvironment without sufficient H 2 O 2 . In vitro experiments demonstrate that the ROS nanogenerators are capable of killing multidrug-resistant H. pylori and removing biofilm, and ROS nanogenerators show high therapeutic efficacy in a H. pylori infection mouse model. Unlike the triple therapy, the nanogenerators display negligible side effects toward the normal gut microbiota. Taken together, these self-enhanced ROS nanogenerators have a great potential for treatment of H. pylori infection.

Different tea polyphenol treatments on lipid oxidation of scallop (Argopecten irradians) adductor muscle during hot air drying

Hongkai Xie,Chenlinrui Yue,Fengjiao Fan

This study investigated the effects of different tea polyphenol (TP) treatments (control, unmarinated, and marinated) on scallop adductor muscle lipid oxidation during hot air drying. Results showed that the marinated sample had the lowest peroxide value, thiobarbituric acid reactive substances, polyunsaturated fatty acids loss, free radical level, and lipoxygenase activity during drying, indicating that the marination process maximized the antioxidant effects of TP. These results were achieved by the distributing of TP on lipid oxidation site, effectively intercepting free radical chain reaction and inhibiting lipoxygenase activity. The significant loss of total phenolic content in the marinated sample confirmed this result. The control, unmarinated, and marinated sample lost 26.52%, 9.99%, and 6.46% of eicosapentaenoic acid and docosahexaenoic acid during drying, respectively, indicating that the marination more efficiently maintained its high nutritive value in terms of lipid. This study provided a more effective way of using TP to control lipid oxidation in seafood during drying. Novelty impact statement Marinated with TP improved its antioxidant effect during scallop drying. More than 26% of EPA + DHA lost in scallop adductor muscle, while less than 7% of EPA + DHA lost in marinated scallop adductor muscle during drying. Marinated with TP maintained the higher nutritive value of scallop during drying.

Hepatic macrophage targeted siRNA lipid nanoparticles treat non-alcoholic steatohepatitis

Jing-E Zhou,Lei Sun,Li Liu,Yujie Jia,Yuqiao Han,Jiaqi Shao,Jing Wang,Yiting Wang,Lei Yu,Zhiqiang Yan

HMGB1 is an inflammatory factor produced by macrophages after liver injury, which plays a key role in promoting NASH progression and further developing into liver fibrosis and cirrhosis. In this study, a mannose-modified HMGB1-siRNA loaded stable nucleic acid lipid particle delivery system (mLNP-siHMGB1) was constructed to target liver macrophages with mannose receptor mediation, thereby silencing HMGB1 protein expression and treating NASH. We also examined the effect of co-administration with docosahexaenoic acid (DHA), a kind of unsaturated fatty acid , on NASH. The results showed that mLNP-siHMGB1 could target macrophages through mannose receptors, effectively silence HMGB1 gene, reduce the release of HMGB1 protein in the liver, regulate liver macrophages to be an anti-inflammatory M2 phenotype, effectively reduce hepatic lobular inflammation and bullous steatosis in the liver, and restore the liver function of NASH model mice to a normal level. After 8 weeks of combined treatment with mLNP-siHMGB1 and DHA, the liver function of NASH model mice recovered rapidly and the hepatic steatosis returned to normal level. In view of inflammation, a key factor in the progression of NASH, we provided an actively targeted siRNA delivery system in this study, and clarified the important role of the delivery system in phenotypic regulation of liver macrophages in NASH. In addition, we also demonstrated the effectiveness of DHA co-administration in NASH treatment. This study provided a useful idea and scientific basis for the development of therapeutic strategies for NASH in the future.

A newly identified polyunsaturated macamide alleviates dextran sulfate sodium-induced colitis in mice

Rui Zha,Enhui Ge,Lirong Guo,Qing Gao,Qiqi Lin,Wan Zhou,Xiaobao Jin,Weiquan Xie,Hui Yin,Tao Liu

Macamides are a class of bioactive amide alkaloids found only in maca ( Lepidium meyenii ). Recent studies have shown that macamide-rich extracts possess various biological activities , such as antioxidative, immune-enhancing, and reproductive health-improving activities. In the present study, N-benzyl docosahexaenamide (NB-DHA), a newly identified macamide with the highest degree of unsaturation among all identified macamides, was identified from the maca extract. Microalgae oil, a docosahexaenoic acid-rich substance, was used as the starting material for the synthesis of NB-DHA. The effects of NB-DHA in colitis-induced mice were evaluated. NB-DHA significantly alleviated weight loss, shortening of colon length, and occult blood occurrence in mice with dextran sulfate sodium-induced colitis . Histological analysis revealed that following the administration of NB-DHA in mice with colitis, the infiltration of inflammatory cells and levels of proinflammatory factors, such as tumor necrosis factor-α, interleukin (IL)-1β, IL-6, and myeloperoxidase , decreased, whereas the level of the anti-inflammatory factor IL-10 increased. Furthermore, the decreased expression of intestinal tight junction proteins caused by colitis was upregulated by the administration of NB-DHA. These results indicate that NB-DHA could be developed as a therapeutic agent for ulcerative colitis .

Small-Sized MOF-Constructed Multifunctional Diagnosis and Therapy Platform for Tumor

Chao Wang,Xiaodan Jia,Wenyao Zhen,Mengchao Zhang,Xiue Jiang

In this article, a type of small-sized metal organic framework (MOF), MIL-101(Fe), as an intelligent delivery system was fabricated to load chemotherapy drug dihydroartemisinin (DHA) and photosensitizer methylene blue (MB). In addition, the Fe ions releasedfrom the MOFs in the tumor environment not only enhanced the curative effect of DHA but also catalyzed H2O2 to release O2, which further improved the photodynamic therapeutic effect of the nanocomposites. The nanocomposites can serve as a T2 magnetic resonance imaging contrast agent at the same time. The polylactic acid (PLA) and polyethylene glycol (PEG) were used to modify the surface of MOFs-MB-DHA to acquire the excellent controllable release of drugs and good biocompatibility to decrease the side effects for normal cells. All the results show remarkably increase of the therapeutic efficiency by synergistic chemo-photodynamic therapy. Thus, a smart multifunctional drug delivery system for diagnosis and therapy based on MOFs-MB-DHA@PLA@PEG was constructed for not only real-time imaging but also chemotherapy and photodynamic synergetic therapy to kill the tumor selectively, showing great potential for conquering the existing barrier in chemo-photodynamic synergetic therapy.

Cancer Nutritional-Immunotherapy with NIR-II Laser Controlled ATP Release Based on Material Repurposing Strategy

Jingsong Lu,Yupei Wang,Xiaohan Gao,Ying Li,Neema Ufurahi Pambe,Abdul Fahad,Zeping Jin,zhijun He,Zhenhu Guo,Wensheng Xie,Shi Wang,Xiaodan Sun,Xiumei Wang,Jing Yu,Shenglei Che,Guifeng Zhang,Yen Wei,Lingyun Zhao

Enlightened by the great success of drug repurposing strategy in pharmaceutic industries, in the current study, material repurposing was proposed where the performance of carbonyl iron powder (CIP), a nutritional intervention agent of iron supplement approved by the US FDA for iron deficiency anemia in clinical was explored in anti-cancer treatment. Except for the abnormal iron metabolic characteristics of tumors that serving as potential targets for CIP-based cancer therapy under repurposing paradigm, the efficacy of CIP as catalyst in Fenton reaction, activator for dihydroartemisinin (DHA) and thus increase the chemo-sensitivity of tumors, as well as the potent agent for NIR-II photothermal therapy (PTT) was fully evaluated by an injectable alginate hydrogel form. The CIP-ALG gel could cause rapid temperature rise in the tumor site under NIR-II laser irradiation, leading to complete ablation in the primary tumor. Further on, such photothermal-ablation would lead to ATP release significantly and in the bilateral tumor model, both primary tumor ablation and inhibition of secondary tumor were observed simultaneously under synergistic tumor treatment of nutritional-photothermal therapy (NT/PTT). Material repurposing could thus be confirmed by our pioneering trial and the CIP-ALG meditated NT/PTT/immunotherapy would provide new choice of strategy for safe and efficient tumor therapy.

Mitochondrial folate metabolism-mediated α-linolenic acid exhaustion masks liver fibrosis resolution

Yanjie Gao,Bingfeng Zheng,Shuaiqi Xu,Zhibo Zhao,Wanyue Liu,Tingyu Wang,Manman Yuan,Xueqing Sun,Yang Tan,Qiang Xu,Xingxin Wu

Sustainable TGF-β1 signaling drives organ fibrogenesis. However, the cellular adaptation to maintain TGF-β1 signaling remains unclear. In this study, we revealed that dietary folate restriction promoted the resolution of liver fibrosis in mice with nonalcoholic steatohepatitis (NASH). In activated hepatic stellate cells (HSCs), folate shifted toward mitochondrial metabolism to sustain TGF-β1 signaling. Mechanistically, nontargeted metabolomics screening identified that α-linolenic acid (ALA) is exhausted by mitochondrial folate metabolism in activated HSCs. Knocking down serine hydroxymethyltransferase 2 (SHMT2) increases the bioconversion of ALA to docosahexaenoic acid (DHA) which inhibits TGF-β1 signaling. Finally, blocking mitochondrial folate metabolism promoted liver fibrosis resolution in NASH mice. In conclusion, mitochondrial folate metabolism/ALA exhaustion/TGF-βR1 reproduction is a feedforward signaling to sustain profibrotic TGF-β1 signaling and targeting mitochondrial folate metabolism is a promising strategy to enforce liver fibrosis resolution.

Engineered Biomimetic Nanovesicles Based on Neutrophils for Hierarchical Targeting Therapy of Acute Respiratory Distress Syndrome

Xiaonan Li,Qi Qiao,Xiong Liu,Qian Hu,Yulin Yu,Xianya Qin,Tianyi Tian,Yinmei Tian,Xiangjun Ou,Boning Niu,Conglian Yang,Li Kong,Zhiping Zhang

Acute Respiratory Distress Syndrome (ARDS) is a clinically severe respiratory disease that causes severe medical and economic burden. To improve therapeutic efficacy, effectively targeting delivery to the inflamed lungs and inflamed cells remains an ongoing challenge. Herein, we designed engineered biomimetic nanovesicles (DHA@ANeu-DDAB) by fusion of lung-targeting functional lipid, neutrophil membrane containing activated β2 integrins, and the therapeutic lipid, docosahexaenoic acid (DHA). By the advantage of lung targeting lipid and β2 integrin targeting adhesion, DHA@ANeu-DDAB can first target lung tissue and further target inflammatory vascular endothelial cells, to achieve “tissue first, cell second” hierarchical delivery. In addition, the β2 integrins in DHA@ANeu-DDAB could bind to the intercellular cell adhesion molecule-1/2 (ICAM-1/2) ligand on the endothelium in the inflamed blood vessels, thus inhibiting neutrophils’ infiltration in the blood circulation. DHA administration to inflamed lungs could effectively regulate macrophage phenotype and promote its anti-inflammatory activity via enhanced biosynthesis of specialized pro-resolving mediators. In the lipopolysaccharide-induced ARDS mouse model, DHA@ANeu-DDAB afforded a comprehensive and efficient inhibition of lung inflammation and promoted acute lung damage repair. Through mimicking physiological processes, these engineered biomimetic vesicles as a delivery system possess good potential in targeting therapy for ARDS.

Protection effect of lutein-loaded Pickering emulsion prepared via ultrasound-assisted Maillard reaction conjugates on dry age-related macular degeneration

Kangjing Liu,Yu Li,Xu Zhong,Yitong Hou,Siyuan Fei,Entao Chen,Mingqian Tan

Age-related macular degeneration (AMD) is a prominent cause of vision loss among the elderly, and the treatment options for dry AMD (dAMD) are severely limited. Lutein has a favorable effect on the treatment of dAMD. Algae oil, rich in docosahexaenoic acid (DHA), is considered an effective intervention for eye diseases. In this study, casein-mannose conjugates were prepared to form algal oil-in-water Pickering emulsions by ultrasound-assisted Maillard reaction. As the ultrasound time increased from 0 to 25 min, the droplet size decreased to 648.2 ± 21.18 nm, which substantially improved the stability of the Pickering emulsions. The retention of lutein in the Pickering emulsions under ultrasonic treatment for 20 min was significantly improved under different conditions. The simulated gastrointestinal digestion revealed that ultrasound-assisted Pickering emulsions are an effective method for improving the bioaccessibility of lutein (19.76%–53.34%). In vivo studies elucidated that the lutein-loaded Pickering emulsions could effectively alleviate retinal thinning induced by sodium iodate (NaIO3) in mice with dAMD. Mechanistically, lutein-loaded Pickering emulsions significantly reduced oxidative stress by decreasing the MDA level, increasing the SOD production, and reducing the retinal ROS production. These findings explored the protective effects of lutein-loaded Pickering emulsions on dAMD and offered promising prospects for the nutritional intervention of dAMD.
