Waterborne Benzo[a]pyrene (B[a]P) pollution is a global threat to aquatic organisms. The exposure to waterborne B[a]P can disrupt the normal locomotor behavior of zebrafish ( Danio rerio ), however, how it affect the locomotor behavior of adult zebrafish remains unclear. Herein, B[a]P at two concentrations (0.8 μg/L and 2.0 μg/L) were selected to investigate the molecular mechanisms of the affected locomotor behavior of zebrafish by B[a]P based on transcriptome profiling. Adverse effects of B[a]P exposure affecting locomotor behavior in zebrafish were studied by RNA sequencing, and the locomotion phenotype was acquired. The gene enrichment results showed that the differentially highly expressed genes ( atp2a1, cdh2, aurka, fxyd1, clstn1, apoc1, mt-co1, tnnt3b, and fads2 ) of zebrafish are mainly enriched in adrenergic signaling in cardiomyocytes (dre04261) and locomotory behavior (GO:0007626). The movement trajectory plots showed an increase in the locomotor distance and velocity of zebrafish in the 0.8 μg/L group and the opposite in the 2.0 μg/L group. The results showed that B[a]P affects the variety of genes in zebrafish, including motor nerves, muscles, and energy supply, and ultimately leads to altered locomotor behavior.

N-(1,3-dimethylbutyl)- N ′-phenyl- p -phenylenediamine (6PPD) is a type of p -phenylenediamine (PPD), which is widely used in the manufacture of rubber tires owing to its excellent antiozonant properties. In this study, the developmental cardiotoxicity of 6PPD was evaluated in zebrafish larvae, and the LC50 was approximately 737 μg/L for the larvae at 96 h post fertilization (hpf). In the 6PPD treatment of 100 μg/L, the accumulation concentrations of 6PPD were up to 2658 ng/g in zebrafish larvae, and 6PPD induced significant oxidative stress and cell apoptosis in the early developmental stages of zebrafish. Transcriptome analysis showed that 6PPD exposure could potentially cause cardiotoxicity in larval zebrafish by affecting the transcription of the genes related to the calcium signal pathway and cardiac muscle contraction. The genes related to calcium signaling pathway ( slc8a2b , cacna1ab , cacna1da , and pln ) were verified by qRT-PCR, which were significantly downregulated in larval zebrafish after exposing to 100 μg/L of 6PPD. Simultaneously, the mRNA levels of the genes related to cardiac functions ( myl7 , sox9 , bmp10 , and myh71 ) also respond accordingly. H&E staining and heart morphology investigation indicated that cardiac malformation occurred in zebrafish larvae exposed to 100 μg/L of 6PPD. Furthermore, the phenotypic observation of transgenic Tg (myl7: EGFP) zebrafish also confirmed that 100 μg/L of 6PPD exposure could change the distance of atria and ventricles of the heart and inhibit some key genes ( cacnb3a , ATP2a1l , ryr1b ) related to cardiac function in larval zebrafish. These results revealed the toxic effects of 6PPD on the cardiac system of zebrafish larvae.

Trichlorocarban (TCC) is ubiquitously detected in environmental matrices, while there is a paucity of information regarding its systemic toxicity. In the present study, we observed that TCC exposure led to high embryo mortality, delayed hatching and yolk absorption, as well as increased malformations, such as closure of swim sac and yolk sac edema. Meanwhile, TCC affected the formation and branch of subintestinal veins (SIVs), intersegmental vessels and posterior cardinal veins. Especially, the SIVs were shrunk, and their branches were reduced or even broken along with reduced coverage area. TCC-induced oxidative stress and excessive apoptosis resulted from abnormal expression of the anti/pro-apoptotic genes. Significant reduction in the number and aggregation function of immune cells proved that TCC had prominent immunotoxicity to zebrafish. TCC-targeted TLR4 signaling pathway was demonstrated by abnormal expression of the marker genes ( tlr4 , MyD88 and nf-κb ) and release of the downstream inflammatory factors (TNF-α, IL-6, etc.). Inhibition of TLR4/MyD88/NF-κB pathway by an inhibitor (CA-4948) rescued the decreasing trend of the immune cells induced by TCC. Molecular docking results demonstrated that TCC could stably bind to TLR4 receptor to form hydrogen bonds and hydrophobic interactions with amino acids. Overall, these findings reveal the underlying molecular mechanisms on TCC-induced developmental and immune toxicity to zebrafish.

Accumulated evidence has demonstrated that microplastics and oxytetracycline (OTC) affect organisms, but few studies have investigated their combined effects on aquatic organisms. In this study, adult zebrafish ( Danio rerio ) were exposed to single and binary-combined contamination of micro-, nano-sized polystyrene plastics and OTC for 30 days, and the intestinal histopathology, gut microbiota and antibiotic resistance genes (ARGs) of zebrafish were measured. The results showed that the intestinal epithelial damage increase with the decrease of plastic sizes. Nano-sized plastics, OTC and their combined exposure caused intestinal epithelial damage, and co-exposure with micro-sized plastics reduced the intestinal damage caused by single OTC exposure. The gut microbial communities were affected by the combined exposure to microplastics and OTC. Compared with the blank control, the relative abundance of Fusobacteria increased 12.7 % and 21.1 % in OTC combined with 45–85 μm micro-plastics (MOTC) and 40–54 nm nano-plastics (NOTC), respectively, and that of Bacteroidetes increased 26.2 % and 18.6 % in the MOTC and NOTC treatments, respectively. The effects of MOTC and NOTC on the biodiversity of the zebrafish gut microbiome were different; MOTC increased the biodiversity by 11.3 % compared with the blank control, whereas NOTC decreased the biodiversity by 8.8 % compared with the blank control. Furthermore, the abundance of ARGs in 40–54 nm nano-plastics, MOTC and NOTC treatments was increased 96.9 %, 96.6 % and 68.8 % compared with the control group, respectively. Additionally, significant differences were observed in ARGs characteristics between the micro- and nano-plastics treated groups whether combined with OTC or not. These results are essential to further understand the combined ecotoxicological effects of micro- or nano-plastics and antibiotics on aquatic organisms.

Triclosan (TCS), as a broad-spectrum bactericide, is extensively used in the fine chemical and textile industries. It is recognized as a new type of environmental endocrine disruptor with frequent detection and environmental pollution. However, the toxicity mechanism regarding neurodevelopment and neurobehavior remains unclear. This study is intended to explore the underlying toxic mechanism of TCS based on gut-brain axis. TCS-chronic exposure affected the development of zebrafish, induced feminization, obesity physical signs and abnormal organ index and caused neurobehavioral abnormalities by inhibiting both neurotransmitter acetylcholinesterase and dopamine activity, promoting brain neuron apoptosis and accelerating diencephalic lesions. Meanwhile, TCS-chronic exposure led to gut microbiota dysbiosis and decreased diversity, such as increased pathogenic bacteria and decreased probiotics in adult zebrafish gut, which caused many pathological damages, including partial shedding and ablation of intestinal villi, inflammatory infiltration, thinning of intestinal wall, and increased goblet cell in villus. Based on the communication between intestinal peripheral nerves and CNS, the above histopathological injuries and disorders were well underpinned and illustrated by the changes of biomarkers and the expression of related marker genes in the gut-brain axis. Additionally, short-chain fatty acids (SCFA), as the regulators of intestinal sympathetic nerve activation, are also secreting products of intestinal microflora and play a crucial role in regulating the balance of intestinal flora and protecting intestinal homeostasis. SCFA in low doses can effectively alleviate and rescue the toxic effects under TCS exposure, which evidenced that TCS exerted systemic toxic effects on the gut-brain axis by influencing the composition and diversity of gut flora in zebrafish, and fully demonstrated the interaction effect between intestine and brain. Hence, these findings contribute to the understanding, prevention, and diagnosis of endocrine disrupting diseases caused by environmental pollutants from the perspective of the gut-brain axis, and strengthening the early warning, management and control of TCS pollution.

The broad spectrum applications of CoFe 2 O 4 NPs have attracted much interest in medicine, environment and industry, resulting in exceedingly higher exposures to humans and environmental systems in succeeding days. Their health effects and potential biological impacts need to be determined for risk assessment. Zebrafish ( Danio rerio ) embryos were exposed to environmentally relevant doses of nano-CoFe 2 O 4 (mean diameter of 40 nm) with a concentration range of 10–500 μM for 96 h. Acute toxic end points were evaluated by survival rate, malformation, hatching delay, heart dysfunction and tail flexure of larvae. Dose and time dependent developmental toxicity with severe cardiac edema, down regulation of metabolism, hatching delay and tail/spinal cord flexure and apoptosis was observed. The biochemical changes were evaluated by ROS, Catalase (CAT), Lipid peroxidation (LPO), Acid phophatase (AP) and Glutatione s- transferase (GST). An Agglomeration of NPs and dissolution of ions induces severe mechanical damage to membranes and oxidative stress. Severe apoptosis of cells in the head, heart and tail region with inhibition of catalase confirms ROS induced acute toxicity with increasing concentration. Increased activity of GST and AP at lower concentrations of CoFe 2 O 4 NPs demonstrates the severe oxidative stress. Circular dichroism (CD) spectra indicated the weak interactions of NPs with BSA and slight changes in α-helix structure. In addition, CoFe 2 O 4 NPs at lower concentrations do not show any considerable interference with assay components and analytical instruments. The results are possible elucidation of pathways of toxicity induced by these particles, as well as contributing in defining the protocols for risk assessment of these nanoparticles.

p-Chloro-m-xylenol (PCMX) is a widely used broad-spectrum antimicrobial agent. Its extensive production, utilization and discharge have led to frequent detection in aquatic environments and biological samples, posing potential ecological risks. However, its toxic effects on the visual system and visual-related behaviors of aquatic organisms remain poorly understood. Here, zebrafish larvae were exposed to 0.4, 1.2, 2 and 2.8 mg/L PCMX to investigate impacts on ocular morphology, retinal development, visual-mediated behaviors and circadian rhythms. The results demonstrated that PCMX at ≥0.4 mg/L induced ocular malformations. Specifically, PCMX suppressed the expression of key genes regulating photoreceptor proliferation and differentiation and disrupted thyroid hormone signaling, inhibiting photoreceptor development and causing retinal pathology. At concentrations of 2 and 2.8 mg/L, these impairments further impaired image-forming (IF) vision and visuomotor responses to light-dark transitions. Concurrently, PCMX disrupted the rhythmic expression of melanopsin family genes in the non-image-forming (NIF) visual system. Furthermore, at higher concentrations (≥1.2 mg/L), PCMX disturbed the circadian rhythmic expression of core clock genes, ultimately induced abnormal sleep/wake behaviors featuring daytime hyperactivity and insomnia-like sleep fragmentation at night. Melatonin (MT) intervention effectively alleviated the sleep/wake behavioral deficits. In conclusion, PCMX damages IF vision by inhibiting photoreceptor development, leading to visual-mediated behavioral abnormalities, while also disrupting the NIF visual system through altered rhythmic expression of melanopsin genes. PCMX-induced circadian and sleep/wake behavioral disorders can be ameliorated by exogenous MT. This study provides novel molecular targets for PCMX ecological risk assessment and support MT as a potential intervention strategy for pollutant-induced toxicities.

Environmentally relevant DMP exposure induced embryotoxicity in zebrafish. • Early-life exposure to DMP caused ovarian damage in adult zebrafish, resulting in fewer mature oocytes. • DMP exposure impaired maternal reproductive capacity and triggered embryotoxicity in the offspring. • DMP disrupted sex hormone balance and altered the HPGL axis. • DMP inhibited oocyte development by affecting key pathways for cell adhesion and steroidogenesis. Dimethyl phthalate (DMP) is a ubiquitous environmental contaminant due to its persistence and inefficient removal during wastewater treatment. However, the long-term consequences of early-life DMP exposure for female reproductive health remain insufficiently understood. In this investigation, zebrafish embryos were subjected to environmentally relevant concentrations (10, 100, 1000 μg/L) of DMP from 2 h post-fertilization (hpf) to 28 days post-fertilization (dpf), and maintained in clean water until adulthood (150 dpf). The results showed that early-life DMP exposure significantly impaired oocyte maturation and fecundity in adult females. Offspring derived from DMP-exposed females mated with unexposed males exhibited reduced survival and increased malformation rates. DMP exposure also induced endocrine disruption, altering hormone levels at 45 dpf (E2 and T) and in adults (E2, T, FSH, LH, and AMH). This was associated with dysregulated expression of key genes ( gnrh2 , gnrh3 , fshβ , lhβ , cyp11a1 , and cyp19a ) along the HPGL axis. Additionally, molecular docking analysis further demonstrated relatively strong binding of DMP to the aromatase isoforms CYP19A and CYP19B, suggesting interference with estrogen–androgen conversion. Transcriptomic analysis of ovarian tissue identified significant disruption of pathways related to extracellular matrix–receptor interaction, focal adhesion, and steroid hormone biosynthesis. These findings reveal the mechanisms through which early-life DMP exposure causes lasting reproductive toxicity in adult female zebrafish, highlighting its potential ecological and health risks. Nanoplastics (NPs) are ubiquitous in the natural aquatic environment, thus there is a major need to understand their potential health risk toward aquatic organisms. Proteins are widely spread in the water environment, they may interact with NPs and significantly impact the ecotoxicology of NPs in vivo . In this study, positively charged lysozyme (LYZ) and negatively charged ovalbumin (OVA) were selected as model proteins, and their effects on the bioaccumulation kinetics of polystyrene nanoplastics (PS-NPs) in zebrafish were investigated quantitatively using a biokinetic model. The adsorption of OVA decreased the uptake rates of PS-NPs, thereby reducing the bioconcentration factors (BCF) of PS-NPs in zebrafish. However, in the presence of LYZ, the uptake rates of PS-NPs increased while the depuration rates decreased significantly, as a consequence, the BCF of PS-NPs in zebrafish were elevated. Dynamic light scattering, transmission electron microscopy, and confocal laser scanning microscopy analysis demonstrated that the adsorption of OVA significantly increased the negative charge and hydrodynamic size of PS-NPs, inhibited the interaction between the nanoparticles and negatively charged cell membrane, thus leading to the alleviated bioaccumulation. In contrast, LYZ changed the surface charge of PS-NPs to positive, enhanced the electrostatic attraction and interaction between PS-NPs and cell membrane, thereby increasing the bioavailability of PS-NPs. Our results highlight the impacts of proteins on the biokinetics of NPs, providing novel insights into the ecological risk of nanoplastics.

Levonorgestrel (LNG) is a prevalent contraceptive progestin frequently detected in aquatic systems. Although the endocrine disruptive effect of levonorgestrel on parental fish has been documented, its intergenerational effects at environmentally relevant concentrations remain unclear. In this study, zebrafish embryos were exposed to LNG at nominal concentrations of 0.0, 5.0, 50.0, or 500.0 ng L -1 from 2 hours post fertilization (hpf) to 144 days post fertilization (dpf). Chronic exposure to LNG caused sex ratio skew toward males in zebrafish. These F0 males were subsequently crossed with solvent control females to establish the F1 lineages. Integrated transcriptomic and neurochemical analyses revealed a thyro-catecholaminergic-GABAergic (TCGA) axis dysregulation, characterized by up-regulation of tpo , dio2 , DBH and downregulation of gadA/B . This multi-level dysregulation converged on neurotransmitter imbalance: elevated dopamine (DA) and reduced γ-aminobutyric acid (GABA), resulting in elevated triiodothyronine (T3) and reduced thyroxine (T4). These interdependent hormonal shifts were paternally transmitted to F1 offspring, manifesting as heritable hyperlocomotion. Elevated DNA methyltransferase (DNMT) activity and reduced Ten-eleven translocation (TET) activity in the F0 testes and 120 hpf F1 larvae suggested altered DNA methylation maintenance capacity. In addition, paternal exposure led to reduced fecundity, survival, and motor deficits in F1 offspring. Our data indicate that LNG at concentrations from 5 ng L -1 upward disrupts the integrated TCGA axis, with DNMT/TET alterations suggesting potential paternal epigenetic mechanisms, supporting a framework for progestin-induced intergenerational toxicity.

Triclocarban (TCC) is widely used in personal care products and medical disinfectants, and it was frequently detected in the environment and in humans. In this study, no abnormalities were observed in zebrafish (F0) during exposure to 1, 10, 100 μg/L of TCC (0-4 dpf). However, body length and weight were significantly suppressed at adulthood. Meanwhile, the number of spermatocytes and late-stage oocytes was reduced, and egg production was also decreased. Developmental defect phenotypes were observed in unexposed F1 generation during early life-stage. In addition, the developmental defect phenotypes were still observed in F2 and F3 generation. This indicated that TCC exhibited transgenerational developmental toxicity. RNA-seq results revealed that slc25a4 was significantly up-regulated and mitochondrial autophagy relative pathway was activated in F1-F3 generations. Treatment with Carboxyatractyloside (inhibitor of slc25a4 ) suppressed the expression of slc25a4 while attenuating the developmental defect phenotypes of F1-F3 generations. In addition, both hdac5 and hdac7 were down-regulated in F1-F3 generations. ChIP-PCR results indicated that H3K27ac levels were significantly increased in the promoter region of slc25a4 in F1-F3 generation. These results suggest that the up-regulation of slc25a4 induced by TCC is the key to the occurrence of the developmental defect phenotypes, and that H3K27ac of slc25a4 may mediate the transgenerational transmission of developmental defect phenotypes. This study provides new perspectives for evaluating the health safety and environmental risks of environmentally relevant doses of TCC.

Extensive use of triazophos for the chemical control of pests in agriculture or aquaculture might strongly disturb the aquatic environment due to residue accumulation through various routes like surface run-off, spray-drift and effluent from factories, which have potential negative effects to non-target aquatic organisms . Previous studies have documented the antioxidative effects of triazophos to mammals, however, the oxidative toxicity of triazophos to fish has not been adequately studied to date. Thus, an acute exposure (96 h) to triazophos at different concentrations of 0.06, 0.3 and 1.5 mg/L (corresponding to 1/50th, 1/10th and 1/2th of 96 h-LC 50 , respectively), was conducted to investigate the triazophos-induced oxidative stress in adult zebrafish ( Danio rerio ). The results showed that the time- and dose-dependent induction of oxidative stress except for the ROS level in liver after 24 and 48 h exposure, as indicated by increased reactive oxygen species (ROS), malondialdehyde (MDA) level and a compromised antioxidant defense system, including increased superoxide dismutase (SOD) activity, catalase (CAT) activity, glutathione (GSH) content as well as the increased at first and decreased afterwards genes expression ( Sod1 , Sod2 , Cat and Gpx ) in brain. Simultaneously, ROS and MDA showed an increased trend, SOD activity, CAT activity and GSH content showed a trend of increasing at 24 h and decreasing at 48 h, and then increasing at 96 h and Sod1 , Sod2 , Cat and Gpx gene showed decreasing at first and then increasing in liver tissue. The present study concluded that the damage of the antioxidant system by triazophos induced oxidative stress in the brain and liver of zebrafish with concomitant lipid peroxidation , which is an important mechanism underlying the triazophos-induced acute toxicity .
