Inflammation and fibrosis caused by inserting cochlear implants into the inner ear are of great concern in clinical treatment. To address these issues, we studied new cochlear implant electrode analogs, polydimethylsiloxane (PDMS) filaments loaded with anti-inflammation/fibrosis dexamethasone (Dex) and coated with hyaluronic acid (HA) as a surface modifier. The PDMS filaments were prepared by incorporating Dex into PDMS that contained different amounts of poloxamer 188 (P188) as a drug release enhancer. HA coatings were performed to prevent protein adsorption by treating the surfaces of the PDMS filaments with O 2 plasma, 3-aminopropyltriethoxysilane and HA in sequence. The prepared PDMS filaments were investigated by attenuated total reflection Fourier transform infrared spectroscopy , contact angle measurement and atomic force microscopy . Their protein adsorption and drug release behaviors were examined as well. L929 mouse fibroblast cells were used to evaluate the anti-cell adhesion/proliferation effects of the PDMS filaments. The results showed that the prepared PDMS filaments had the expected structure, good surface properties and excellent anti-fibroblast cell proliferation effects. The HA coatings provided filament surfaces that were more hydrophilic, smoother and more protein-repelling. Dex release was significantly facilitated by adding P188 and slightly slowed down by the HA coatings. The PDMS filaments that contained 5% Dex and 5% P188 and that were coated with HA showed a significant decrease in the fibroblast cell number by approximately 51%, with obviously reduced cell adhesion on the surfaces.

The transcription factor NF-kB plays a pivotal role in the pathogenesis of rheumatoid arthritis. Here we attempt to slow arthritis progression by co-delivering the glucocorticoid dexamethasone (Dex) and small-interfering RNA targeting NF-kB p65 using our previously developed polymeric hybrid micelle system. These micelles contain two similar amphiphilic copolymers : polycaprolactone-polyethylenimine (PCL-PEI) and polycaprolactone-polyethyleneglycol (PCL-PEG). The hybrid micelles loaded with Dex and siRNA effectively inhibited NF-kB signaling in murine macrophages more efficiently than micelles containing either Dex or siRNA on their own. In addition, the co-delivery system was able to switch macrophages from the M1 to M2 state. Injecting hybrid micelles containing Dex and siRNA into mice with collagen-induced arthritis led the therapeutic agents to accumulate in inflamed joints and reduce inflammation, without damaging renal or liver function. Thus, blocking NF-kB activation in inflammatory tissue using micelle-based co-delivery may provide a new approach for treating inflammatory disease.

Selective exhaustion of over-expressed reactive oxygen species (ROS) is of great significance in the therapy of osteoarthritis (OA) because of the inhibiting effect on oxidative stress and inflammation. Herein, a ROS-scavenging and drug-release platform was prepared via encapsulating dexamethasone acetate (DA)-loaded ROS erasable poly(ethylene glycol)- b -polythioketal- b -poly(ethylene glycol) (PEG-PTK-PEG) micelles (PDM) into an injectable hydrogel. The hydrogel ( [email protected] ) was constructed by Schiff base reaction between hydrazide-grafted hyaluronic acid (HA-ADH) and aldehyde-modified dextran (Dex-ALH), achieving a self-healing property for viscosupplementation. The PDM imparted enhanced antioxidant capability to the hydrogel, which, in turn, endowed the PDM with prolonged retention and sustained DA release. The intraarticularly administered multifunctional injectable hydrogel potently diminished inflammation via depleting ROS and suppressing inflammatory cytokines, as well as downregulating pro-inflammatory M1 macrophages ratio in a rat OA model. The developed therapeutic system significantly alleviated OA symptoms, embodying the excellent capability of preventing cartilage extracellular matrix degeneration with negligible toxicity in vivo .

The presence of dexamethasone (DEX) residues and the illegal addition of DEX to food have garnered widespread public concern over recent decades. Based on the bicycled hairpin assembly (BHA) mechanism and the catalytic oxidase effect of urchin-shaped Au–Ag alloy loaded with Pt nanoparticles (Au@Ag@Pt), an accurate and ultrasensitive surface-enhanced Raman scattering (SERS)/colorimetric dual-mode method for DEX detection is pioneered. When DEX is introduced into the sensor system, the BHA cycle begins. The Fe3O4 nanosphere loaded with Au nanoparticles (Fe3O4@Au) surface facilitates the anchoring of a large amount of Au@Ag@Pt, resulting in synergistic signal amplification and facile magnetic separation from the complex system. Finally, the collected product effectively catalyzes the conversion of 3,3′,5,5′-tetramethylbenzidine (TMB) to the oxidized product, producing a strong SERS response, an ultraviolet–visible absorption signal, and color deepening. Our dual-mode platform achieves ultrasensitive detection of DEX, achieving a detection limit (LOD) of 6.61 × 10–17 M within the broad range of 10–16 to 10–9 M for SERS and a LOD of 8.81 × 10–13 and 6.98 × 10–13 M within the range of 10–12–10–5 M for the colorimetric method. Additionally, this platform shows high selectivity and satisfactory recovery rates for DEX in milk and pork samples, with recoveries of 92.3%–106.5% (RSDs <8.35%) for SERS and 95.2%–107.6% (RSDs <8.81%) for colorimetric analysis. This study demonstrates that by combining the BHA amplification process with multifunctional Au@Ag@Pt and Fe3O4@Au substrates, the DEX content in actual food samples can be accurately determined using a dual-mode approach that combines the high sensitivity of SERS with the simplicity of colorimetric analysis.

A novel covalently dexamethasone-conjugated drug release system based on cross-linked vegetable oil (DEX-CVs) was designed and prepared by simple curing reaction of modified vegetable oil and dexamethasone phosphomonoester (DEX-P) without any toxic catalyst and initiator. UV and HPLC analysis confirmed that the dexamethasone (DEX) drug could be released linearly with high purity by adjusting the component ratios. The analysis of the model reaction indicated that DEX-P in the optimized DEX-CV (A4) was mainly covalently conjugated in the cross-linked vegetable oil network by cyclic phosphotriester bond, and the selective hydrolysis induced the pure release of dexamethasone drug. Finally, the low cytotoxicity and successful osteogenic differentiation effect of A4 on human bone marrow mesenchymal stem cells (hBMSCs) was corroborated by the detection of MTT analysis, ALP activity, staining experiments and some gene expressions, which provided the promising applications in drug release areas and bone tissue engineering.

Shape memory polymers (SMP) with better biodegradability and better stability have great potential applications in biomedical fields, such as the drug carriers or the tissue engineering scaffolds. In this study, poly(ε-caprolactone)(PCL)-based polyurethane(PU) microfibers were fabricated with the containing of hydroxyapatite(HA) to enhance the biodegradability and to exhibit excellent shape memory performance. The composition, the morphology, the thermal stability, and the mechanical properties of the microfibers were characterized and detected using Fourier transform infrared spectroscopy (FTIR), 1 H nuclear magnetic resonance ( 1 HNMR), scanning electron microscopy (SEM), differential scanning calorimetry (DSC). and dynamic mechanical analysis (DMA), etc. And dexamethasone was selected as drug model to investigate the delivery and release behaviors of the carrier of the microfibers. It was revealed that HA enhanced the degradation rate of the shape memory polyurethane (SMPU) fibers, and the fibers could guarantee a sustained long time drug release. The detection on the shape memory performance found that, with the different addition amounts of HA, the composite microfibers of (SMPU) and HA exhibited the different shape memory transition temperature ( T trans ) values. And with the addition of 3 wt% of HA, the excellent shape recovery ratios of R r (> 97%) and the shortest recovery time of ~ 6 s could be obtained. With further increase of the amounts of HA, the recovery force and the recovery time were reduced and prolonged, respectively. The obtained results proved that the biodegradable SMPU/HA composite microfibers have more valuable application prospects in biomedical fields.

The brain‑derived neurotrophic factor (BDNF) and c‑Jun NH 2‑terminal kinase (JNK) signaling pathways are therapeutic targets to prevent degeneration in the central nervous system. Dexamethasone (DXMS), a glucocorticoid, protects against vestibular brain injury, however, the molecular mechanisms have yet to be fully elucidated. To investigate whether the BDNF and JNK signaling pathways are involved in the protective effects of DXMS in rats with vestibular dysfunction, a rat model of severe vestibular deficits was established by middle ear injection of arsanilic acid (AA; 100 mg/ml; 0.05 ml). After 3 days, rat symptoms and behavior scores with vestibular disorders were detected. In brain tissues, histopathological alterations, cell apoptosis, expression levels and patterns of BDNF signaling pathway‑associated BDNF, tyrosine receptor kinase B (TrKB) and K+/Cl‑ cotransporter isoform 2 (KCC2), and the expression of apoptosis‑related cleaved‑caspase 3 and the JNK signaling pathway were detected. It was identified that DXMS relieved AA‑induced vestibular dysfunction, leading to improvement in rat behavior scores to normal levels, minimizing brain damage at the histopatholojnnkngical level, reducing cell apoptosis, enhancing the expression of BDNF, TrKB and KCC2, and downregulating cleaved‑caspase 3 and phosphorylated‑JNK1/2 in brain tissues. Together, these findings indicated the protective effect of DXMS on AA‑induced rat vestibular dysfunction, and that activating BDNF and inhibiting JNK singling pathways were the underlying mechanisms. In addition, with additional treatment of mifepristone (RU486), a specific glucocorticoid agonist, all the events elicited by DXMS mentioned above in the AA‑treated rat rats were reversed. In conclusion, DXMS was identified as a therapeutic agent targeting the BDNF and JNK singling pathways for AA‑induced rat vestibular dysfunction.

The implantation of neural electrodes usually induces acute and chronic inflammation, which can result in the formation of glial scars encapsulating the implanted electrodes and the loss of neurons near the active electrode sites. Local presentation of anti-inflammatory drugs or neural protective factors has been evidenced as an effective strategy to modulate inflammatory responses and promote electrode–neuron integration. Here, a novel delivery system for the simultaneous presentation of both anti-inflammatory drugs (dexamethasone, Dex) and nerve-growth-promoting factors (nerve growth factor, NGF) from the electrode sites was developed via layer-structured carbon nanotubes and conductive polymers. The modified electrodes exhibited higher charge storage capacitance and lower electrochemical impedance compared to unmodified electrodes and electrodes coated with polypyrrole/Dex. Dex released from the functional coating under controlled electrochemical stimulation was able to inhibit the lipopolysaccharide-induced secretion or mRNA transcription of inflammatory cytokines, including nitric oxide, TNF-α, and IL-6 in RAW264.7 cells, and control the activation of cultured astrocytes. In addition, the functional coatings did not show a toxic effect on PC12 cells and primary neural cells but exhibited promoted activities on the adhesion, growth, and neurite extension of neural cells.

This research aimed to better exert the efficacy of dexamethasone (DEX) and fabricate an intercellular adhesion molecule A (ILDMA) monoclonal antibody (mAb)-modified nanostructured lipid carrier (NSLC). The anionic DEX NSLC was fabricated by the aqueous solvent diffusion methodology
using DEX as the model drug and in combination with various types of lipids. Using N,N’-succinimidyl carbonate as the link, anti-ILDMA mAb-modified anion DEX NSLC (ILDM/DEX/NSLC) and anti-IgG mAb-modified cation DEX NSLC (IgG/DEX/NSLC) were prepared. The total lipid content was controlled
unchanged, 3% mass ratio of glyceryl monostearate (MS) in the original prescription was replaced with 3% mass ratio of octadecylamine (ODA), and ILDM/DEX/ODA-NSLC and IgG/DEX/ODA-NSLC were fabricated in the same way. The four NSLCs prepared in the experiment were round in shape and uniform
in size. The nanoparticles with a size of approximately 230 nm were similar, and the zeta potentials were (−29.8±21.5) mV, (−27.9±1.6) mV, (36.8±0.8) mV, and (33.7±2.9) mV, respectively. In vitro drug release demonstrated a cumulative release
rate of more than 55% of DEX NSLC. The inhibitory rate of DEX NSLC on the activated human vascular endothelial cell line (EAhy926 cell) was dose dependent, and ILDM/DEX/NSLC could transport DEX to activated endothelial cells more efficiently, thus enhancing the intervention ability on diseased
endothelium. For the establishment of a sepsis-induced acute lung injury (ALI) mouse model, ILDM/DEX/NSLC was highly distributed in the lung of the model, and its infiltration effect on inflammatory cells was superior to that of other drugs (P < 0.05). Meanwhile, ILDM/DEX/NSLC could
more markedly repair the pathological features in the mouse model than other drugs did (P < 0.05). The nanodrug inhibited the protein level of TLR9 in mouse lung tissue to the maximum extent (P < 0.05), thereby enhancing the survival rate of the mice.

Whey protein isolate (WPI) is a promising carrier for bioactive compounds, yet its application for muscle health requires further investigation. This study constructed a WPI-Quercetin (Que) complex using the pH-shift method and evaluated its binding mechanism, structural evolution, and mitigating effects on dexamethasone (DEX)-induced muscle damage. The complex achieved optimal stability at a 1:40 Que: WPI mass ratio, characterized by a sulfhydryl (SH) content of 5.94 μmol/g, a particle size of 427 nm, a Polydispersity Index (PDI) of 0.28, and a zeta potential of −32.1 mV. Multispectral analysis and molecular docking suggested that Que primarily embeds into the hydrophobic cavity of WPI through hydrophobic interactions and hydrogen bonds, inducing protein secondary structure rearrangement. In vitro experiments using C2C12 myotube models demonstrated that the WPI-Que complex significantly enhanced myosin heavy chain (MHC) expression, increased myotube diameter, and improved the fusion index, both in the presence and absence of DEX stimulation. Furthermore, RT-qPCR and Western blot analyses indicated that the WPI-Que complex upregulates myogenic regulators more effectively than WPI or Que alone. These findings indicate that the WPI-Que complex possesses both structural stability and biological activity, offering a theoretical foundation for developing novel functional food ingredients to counteract muscle decline.

Dexamethasone, a potent synthetic glucocorticoid, is widely used for its anti-inflammatory and immunosuppressive properties. However, its misuse as a performance-enhancing drug in sports has raised concerns among regulatory authorities. To address this issue, an electrochemical sensor based on Fe 3 O 4 /polyaniline (PANI)-Cu(II) microspheres and hematite nanoparticles was developed for the sensitive and selective detection of dexamethasone. The sensor exhibited excellent electrocatalytic activity towards the oxidation of dexamethasone due to the synergistic effect of the high conductivity and large surface area of the Fe 3 O 4 /PANI composite microspheres, the electrocatalytic properties of the loaded copper(II) ions, and the enhanced electron transfer of the hematite nanoparticles. Under optimized conditions, the sensor showed a wide linear range of 0.05–50 μM, with a detection limit of 0.015 μM. The sensor also demonstrated good selectivity, reproducibility (RSD = 3.2 %, n = 5), and long-term stability (92 % retention after 4 weeks). Furthermore, the sensor was successfully applied for the determination of dexamethasone in spiked human serum and urine samples, with recoveries ranging from 97.5 % to 103.5 % and RSDs less than 3.5 %.

Osteoarthritis (OA) is a degenerative joint disease characterized by cartilage degradation, chronic inflammation, and subchondral bone remodeling. Conventional intra-articular therapies provide limited relief and fail to address its multifactorial pathogenesis. Here, we present an injectable hydrogel microsphere platform that integrates antioxidative, anti-inflammatory, and regenerative functions for localized OA management. Uniform (∼125 µm) chondroitin sulfate methacrylate (ChSMA)-based microspheres are fabricated via microfluidic photocross-linking. Manganese dioxide nanoparticles provided catalytic reactive oxygen species (ROS) scavenging, while dexamethasone sodium phosphate enabled sustained release, reducing TNF-α and IL-6 levels by ∼30%. Bone marrow mesenchymal stem cells (BMSCs) are co-delivered to promote cartilage repair. In vitro, the microspheres reduce intracellular ROS, induce M2 macrophage polarization, and suppress inflammatory cytokines by 60–70%, with IL-10 levels increased by ∼90%. 3D co-culture supports chondrocyte/BMSC viability and matrix production. In vivo, intra-articular injection in a rat OA model markedly reduces cartilage erosion, decreases osteophyte volume by 80%, and improves subchondral bone microarchitecture. Histological staining confirms matrix restoration and structural preservation, with Osteoarthritis Research Society International (OARSI) scores reduced by 88%. Collectively, this injectable hydrogel microsphere system offers a minimally invasive and integrated strategy, simultaneously delivering antioxidative, anti-inflammatory, and regenerative effects for comprehensive OA management.
