Catalytic medicine, using enzymes or nanozymes, is an emerging method for cancer treatment. However, its applicability is limited by the low catalytic activity in the tumor microenvironment (TME). In this work, a versatile and synthesis-friendly nanozyme, CeO 2 Mn 1.08 O x nanoclusters, is prepared. This novel Ce─Mn heterojunction is formed by oxidation of CeO 2 nanoparticles through H 2 SO 4 /KMnO 4 . CeO 2 Mn 1.08 O x exhibits high multi-enzymatic catalytic activities and acts as a catalase (CAT), peroxidase (POD), and oxidase (OXD) mimics under acidic conditions. It can regulate the TME by relieving hypoxia and consuming endogenous glutathione (GSH). Glucose oxidase (GOx) is then incorporated into CeO 2 Mn 1.08 O x and linked with poly(ethylene glycol) (PEG) to obtain the cascade enzyme system (Ce─Mn)-PEI/GOx-PEG. CeO 2 Mn 1.08 O x exhibits CAT-like properties, which sensitize GOx-based starvation therapy, and POD- and OXD-like properties, which generate highly cytotoxic reactive oxygen species (ROS) in cancer cells. The glucose catabolic product, H 2 O 2 , is also used to generate O 2 and ROS. In addition, the heterojunction structure provides CeO 2 Mn 1.08 O x with near-infrared (NIR) photothermal capability, making it suitable for photothermal therapy (PTT). Density functional theory (DFT) calculations provide possible reasons for the high catalytic activity and photothermal capability of CeO 2 Mn 1.08 O x . When combining mild PTT with catalytic therapy, the cascade enzyme system (Ce─Mn)-PEI/GOx-PEG can efficiently ablate tumors.

Abstract The present article introduced an natural enzyme-covered/amino-modified Pd–Pt bimetallic-doped zeolitic imidazolate framework (NAPPZ) for ultrasensitive and specific detection of glucose. The dodecahedral nanomaterial zeolitic imidazolate framework (ZIF-8)-loaded Pd–Pt bimetallic nanoparticles endowed the composite with peroxidase-like activity. The modification with glucose oxidase (GOx) facilitated the rapid access of H 2 O 2 produced through glucose oxidation to the Pd–Pt nanoparticles vicinity reducing diffusion. GOx specifically catalyzes the transformation of glucose into H 2 O 2 , which then H 2 O 2 rapidly migrates to the Pd–Pt nanoparticles, catalyzing the oxidation of colorless o-phenylenediamine into the orange–yellow product 2,3-diaminophenazine. Based on the aforementioned cascade reaction, the NAPPZ and NAPPZ based on ChOx were utilized for detecting glucose in human urine samples and cholesterol in milk, respectively. The NAPPZ strategy presented a broad detection range (20–1100 μmol L −1 ) and a low detection limit (15.9 μmol L −1 ) for glucose, and the NAPPZ based on ChOx strategy approach offered a broad detection range (10–500 μmol L −1 ) and low detection limit (6.4 μmol L −1 ) for cholesterol. Therefore, this novel method holds significant potential in the areas of clinical diagnostics and food safety. Graphical abstract

Starvation therapy, utilizing glucose oxidase (GOx) to deplete glucose and disrupt tumor energy, has shown promise in cancer treatment. However, this approach alone is insufficient to block ATP generation in tumors. Here, we introduce a self-‘cycling, boosting and propelling’ strategy, overcoming the ‘Achilles' heel’ of traditional starvation therapy by regulating the energy supply with CO. The high concentration of H 2 O 2 in the tumor microenvironment reacts with MnO 2 to generate Mn 2+ and O 2 . Mn 2+ catalyzes a Fenton-like reaction to produce hydroxyl radicals ( OH) for chemodynamic therapy and T1-weighted magnetic resonance imaging (MRI), while O 2 further enhances GOx-mediated glucose consumption, producing more H 2 O 2 . This process not only recycles H 2 O 2 but accelerates CO release, disrupting ATP synthesis and inhibiting tumor energy supply. Additionally, the gas released enhances nanoplatform motility, improving tumor penetration and overcoming the limited permeability of conventional nanoparticles. In vitro and in vivo studies demonstrated superior antitumor and antimetastatic efficacy, coupled with excellent biocompatibility. This strategy offers a promising solution to traditional starvation therapy's challenges, with strong potential for clinical translation and transformative cancer treatment.

Periodontitis, a chronic inflammatory condition associated with bacterial infection, is the leading cause of tooth loss in adults. Pathogenic bacteria that persist in deep periodontal pockets with hyperglycemic levels following traditional manual debridement can contribute to a persistent local inflammatory microenvironment, posing a significant challenge and highlighting the critical need for improved therapeutic approaches. In addition, the anaerobic environment during these treatments is beneficial to the colonization of anaerobes and may amplify inflammatory responses. Herein, we designed a glucose oxidase (GOx)-coating MXene-SnS 2 bio-heterojunctions (MS@G bio-HJs) for multi-modal and effective treatment of anti-bacteria and anti-inflammatory. Upon the hyperglycemic nidus in periodontal pockets, GOx catalyzes the conversion of glucose into hydrogen peroxide (H 2 O 2 ). Subsequently, the breakdown of H 2 O 2 into hydroxyl radicals through Sn 2+ /Sn 4+ -involved Fenton-like reactions enables in situ liberation of oxygen (O 2 ), contributing to relieve local hypoxia. The proposed bio-HJs presents a satisfactory bactericidal capability with anti-bacterial rates of more than 99 % upon near-infrared irradiation, in virtue of multi-modal therapies including photodynamic, photothermal and chemodynamic treatments. Furthermore, in vivo assays utilizing infectious rat models demonstrated that the proposed bio-HJs exhibits high biocompatibility and capacity to alleviate hypoxia and reduce pathogenic virulence factors, potentially contributing to periodontal tissue regeneration. These findings suggest that the versatile bio-HJs could be a promising therapeutic candidate for treating infectious periodontal diseases.

Insufficient accumulation of reactive oxygen species (ROS) due to tumor hypoxia significantly contributes to increased radiation resistance and the failure of radiotherapy (RT). Therefore, developing methods to alleviate hypoxia and boost ROS levels represents a promising strategy for enhanced radiosensitivity. This study introduced a self-cascade catalytic Pt@Au nanozymes as a radiosensitizer, using glucose oxidase (GOx)-, catalase (CAT)-, and peroxidase (POD)-like activities to improve hypoxia and increase ROS accumulation, thereby affecting glucose metabolism and enhancing the effects of RT. Pt@Au nanozymes exhibit GOx-like activity, which not only depletes glucose to induce starvation therapy, but also generates hydrogen peroxide (H2O2) for cascade reactions. Moreover, Pt@Au nanozymes demonstrate CAT-like activity, catalyzing the conversion of H2O2 to O2. This conversion effectively alleviates hypoxia, stabilizes ROS, increases DNA damage, significantly enhancing RT efficacy and sustaining the effects of starvation therapy. As high-Z materials, Pt@Au nanozymes can deposit more X-ray energy. Furthermore, the POD-like activity catalyzes the conversion of H2O2 into highly reactive hydroxyl radicals (·OH), which increases ROS levels and enhances RT. Pt@Au nanozymes serve as X-ray computed tomography (CT) imaging agents, allowing for clear differentiation between tumor and normal tissue boundaries and enhancing the precision of RT. In summary, Pt@Au nanozymes serve as effective radiosensitizers by depleting glucose to induce starvation therapy, enhancing cascade reactions, and inhibiting tumor proliferation. Through their self-cascade reactions, these nanozymes dramatically increase oxygen levels within tumors, reduce hypoxia, and enhance ROS levels. This advancement addresses the radioresistance associated with hypoxic tumors, paving the way for innovative strategies in RT.

Photothermal therapy (PTT)-based combination therapy is an effective approach to cancer treatment. However, there is a possibility of tumor development due to the inflammation caused by PTT. In addition, hyperthermia can cause damage to normal tissues. This work reported a drug delivery/cascade enzyme system based on Ti 3 C 2 T x MXene nanosheets. Ultra-small, degradable mesoporous silica nanoparticles (DS-MSN) were prepared to load phloretin (Phl), which has anti-inflammatory, antioxidant, and anticancer properties. Subsequently, glucose oxidase (GOx) was adsorbed on the surface of DS-MSN. The nanoparticles were then modified on Ti 3 C 2 T x MXene nanosheets and finally treated with polyethylene glycol (PEG) to obtain the multifunctional system named Ti 3 C 2 T x -Phl@DS-MSN-GOx-PEG (TPDMGP). The high near-infrared photothermal conversion efficiency of Ti 3 C 2 T x made it suitable for PTT. GOx and Ti 3 C 2 T x formed a cascade enzyme system inside cancer cells: Ti 3 C 2 T x exhibited catalase (CAT)-like activity that decomposed intracellular hydrogen peroxide (H 2 O 2 ) into O 2 , which was provided to GOx for enhanced starvation therapy. GOx in the presence of O 2 produces H 2 O 2 , for the oxygen production of Ti 3 C 2 T x . Meanwhile, DS-MSN could be degraded in the reducing environment inside cancer cells as it contains disulfide bonds in its backbone, thus releasing Phl for chemotherapy. Such a combination therapy strategy could achieve a satisfactory therapeutic outcome by low-temperature PTT while evading normal tissue damage by high temperature. The anti-inflammatory effects of both Ti 3 C 2 T x and Phl enabled TPDMGP to eliminate the inflammation caused by PTT via the scavenging of reactive oxygen species (ROS). The multifunctional system is expected to generate new paradigms for cancer PTT.

In this study, we developed an indirect competitive plasmonic immunoassay using glucose oxidase (GOx)-induced redox reaction to etch Au nanorods (AuNRs) for qualitative and quantitative detection of aflatoxin M1 (AFM1) in milk. In this system, streptavidin (SA) was selected as a linker between biotinylated anti-AFM1-monoantibody (bio-mAb) and biotinylated GOx (bio-GOx) to form the immunocomplexes bio-mAb-SA-bio-GOx. After the oxidation of the glucose and I - , the resultant I 2 could etch cetytrimethylammonium bromide (CTAB)-stabilized AuNRs . This resulted in the blue shift of the longitudinal localized surface plasmon resonance (LSPR) extinction peak, with a color change from blue to pink. The linear range and limit of detection (LOD) of the plasmonic immunoassay were 0.25–10 ng mL −1 and 0.11 ng mL −1 , respectively. It displayed quantitative detection with high sensitivity, specificity, recovery, and accuracy, which is promising for qualitative and quantitative detection of AFM1 in food safety.

Surgical resection is the primary treatment for melanoma; however, preventing tumor recurrence after resection remains a significant clinical challenge. To address this, we developed a multifunctional nanocomposite hydrogel (H-CPG) composed of glucose oxidase (GOx)-coated CuS@PDA@GOx (CPG) nanoparticles, aminated hyaluronic acid (HA-ADH), and oxidized rhizomatous polysaccharides (OBSP), which are interconnected through hydrogen bonds and dynamic Schiff base linkages. In the acidic tumor micro-environment, the hydrogel releases GOx, catalyzing the production of hydrogen peroxide (H2O2), which enhances chemokinetic activity through a Cu2+-mediated Fenton-like reaction. This process generates hydroxyl radicals that intensify oxidative stress and promote macrophage polarization from the M2 to M1 phenotype. This polarization triggers the release of pro-inflammatory cytokines, thereby inhibiting tumor recurrence. Additionally, the hydrogel induces photothermal effects that help eradicate residual bacteria at the wound site. Overall, the H-CPG hydrogel offers a dual mechanism to prevent melanoma recurrence and reduce resistance to monotherapy, presenting a promising strategy for postoperative tumor management.

Tumor cells can survive when detached from the extracellular matrix (ECM) or lose cell–cell connections, a phenomenon known as anoikis-resistance (AR). AR is closely associated with tumor cell metastasis and recurrence, enabling tumor cells to disseminate, migrate, and invade after detachment. To address this issue, a novel intervention method combining intraoperative hemostasis with multifunctional nanozyme driven-enhanced chemodynamic therapy (ECDT) has been proposed, which holds the potential to weaken the AR capability of tumor cells and suppress tumor recurrence. Here, a nanocomposite containing a dendritic mesoporous nanoframework with Cu 2+ was developed using an anion-assisted approach after surface PEG grafting and glucose oxidase (GOx) anchoring (DMSN-Cu@GOx/PEG). DMSN-Cu@GOx/PEG was further encapsulated in a thermal-sensitive hydrogel (H@DMSN-Cu@GOx/PEG). DMSN-Cu@GOx/PEG utilizes its high peroxidase (POD) activity to elevate intracellular ROS levels, thereby weakening the AR capability of bladder cancer cells. Additionally, through its excellent catalase (CAT) activity, DMSN-Cu@GOx/PEG converts the high level of hydrogen peroxide (H 2 O 2 ) catalyzed by intracellular GOx into oxygen (O 2 ), effectively alleviating tumor hypoxia, downregulating hypoxia-inducible factor-1α (HIF-1α) expression, inhibiting epithelial-mesenchymal transition (EMT) processes, and ultimately suppressing the migration and invasion of bladder cancer cells. Interestingly, in vivo results showed that the thermosensitive hydrogel H@DMSN-Cu@GOx/PEG could rapidly gel at body temperature, forming a gel film on wounds to eliminate residual tumor tissue after tumor resection surgery. Importantly, H@DMSN-Cu@GOx/PEG exhibited excellent hemostatic capabilities, effectively enhancing tissue coagulation during post-tumor resection surgery and mitigating the risk of cancer cell dissemination and recurrence due to surgical bleeding. Such hydrogels undoubtedly possess strong surgical application. Our developed novel nanosystem and hydrogel can inhibit the AR capability of tumor cells and prevent recurrence post-surgery. This study represents the first report of using dendritic mesoporous silica-based nanoreactors for inhibiting the AR capability of bladder cancer cells and suppressing tumor recurrence post-surgery, providing a new avenue for developing strategies to impede tumor recurrence after surgery. Herein, a Au-Au/IrO2 nanocomposite with tandem enzyme-mimicking activity was innovatively synthesized, which can show outstanding glucose oxidase (GOx)-like activity and peroxidase-like activity simultaneously under neutral conditions. Moreover, a Au-Au/IrO2@Cu(PABA) reactor was prepared via encapsulation of the Au-Au/IrO2 nanocomposite in a Cu(PABA) metal organic framework. The reactor not only exhibits excellent organic solvent stability, acid resistance, and reusability but also displays better cascade reaction catalytic efficiency (kcat/Km = 148.86 min–1 mM–1) than the natural free enzyme system (GOx/HRP) (kcat/Km = 98.20 min–1 mM–1) and Au-Au/IrO2 nanocomposite (kcat/Km = 135.24 min–1 mM–1). In addition, it is found that the reactor can catalyze glucose or dissolved oxygen to produce active oxygen species (ROS) including HO, 1O2, and O2–· through its enzyme-mimicking activity. Finally, the novel reactor was successfully used in organic dye degradation and antibacterial application. The results show that it can effectively degrade methyl orange, methylene blue, and rhodamine B, which all can reach a degradation rate of nearly 100% after interacting with Au-Au/IrO2@Cu (PABA) for 3.5 h. Furthermore, the reactor also exhibits excellent antibacterial activity, so as to achieve a complete bactericidal effect to Staphylococcus aureus and Escherichia coli at a concentration of 12.5 μg mL–1.
