Nanoengineered mesenchymal stem cell therapy for pulmonary fibrosis in young and aged mice

Meng-Meng Han,Xing-Yue He,Ling Tang,Liang Qi,Ming-Yuan Yang,Yi Wang,Lei Xing,Jee-Heon Jeong,Hu-Lin Jiang

Pulmonary fibrosis (PF) is an age-related interstitial lung disease that results in notable morbidity and mortality. The Food and Drug Administration–approved drugs can decelerate the progression of PF; however, curing aged patients with severe fibrosis is ineffective because of insufficient accumulation of these drugs and wide necrocytosis of type II alveolar epithelial cells (AEC IIs). Here, we constructed a mesenchymal stem cell (MSC)–based nanoengineered platform via the bioconjugation of MSCs and type I collagenase–modified liposomes loaded with nintedanib (MSCs-Lip@NCAF) for treating severe fibrosis. Specifically, MSCs-Lip@NCAF migrated to fibrotic lungs because of the homing characteristic of MSCs and then Lip@NCAF was sensitively released. Subsequently, Lip@NCAF ablated collagen fibers, delivered nintedanib into fibroblasts, and inhibited fibroblast overactivation. MSCs differentiated into AEC IIs to repair alveolar structure and ultimately promote the regeneration of damaged lungs in aged mice. Our findings indicated that MSCs-Lip@NCAF could be used as a promising therapeutic candidate for PF therapy, especially in aged patients.

Win-win integration: A mitochondria targeted AIE photosensitizer for hypochlorite detection and type I & type II photodynamic therapy

Jiahao Ni,Lichao Yu,Yixuan Wang,Tong Yang,Yueqi Bai,Bowen Zheng,Manshan Liang,Xiaoxia Ye,Yun-Yun Quan,Feifei Lin,Zu-Sheng Huang

Background Photodynamic therapy (PDT) is a pioneering and effective anticancer modality with low adverse effects and high selectivity. Hypochlorous acid or hypochlorite (HClO/ClO − ) is a type of inflammatory cytokine. The abnormal increase of ClO − in tumor cells is related to tumor pathogenesis and may be a “friend” for the design and synthesis of responsive phototherapy agents. However, preparing responsive phototherapy agents for all-in-one noninvasive diagnosis and simultaneous in situ therapy in a complex tumor environment is highly desirable but still remains an enormously demanding task. Results An acceptor-π bridge-donor-π bridge-acceptor (A-π-D-π-A) type photosensitizer TPTPy was designed and synthesized based on the phenothiazine structure which was used as the donor moiety as well as a ClO − responsive group. TPTPy was a multifunctional mitochondria targeted aggregation-induced emission (AIE) photosensitizer which could quickly and sensitively respond to ClO − with fluorescence “turn on” performance (19-fold fluorescence enhancement) and enhanced type I reactive oxygen species (ROS) generation to effectively ablate hypoxic tumor cells. The detection limit of TPTPy to ClO − was calculated to be 185.38 nM. The well-tailored TPTPy anchoring to mitochondria and producing ROS in situ could disrupt mitochondria and promote cell apoptosis. TPTPy was able to image inflammatory cells and tumor cells through ClO − response. In vivo results revealed that TPTPy was successfully utilized for PDT in tumor bearing nude mice and exhibited excellent biological safety for major organs. Significance and novelty A win-win integration strategy was proposed to design a tumor intracellular ClO − responsive photosensitizer TPTPy capable of both type I and type II ROS production to achieve photodynamic therapy of tumor. This work sheds light on the win-win integration design by taking full advantage of the characteristics of tumor microenvironment to build up responsive photosensitizer for in situ PDT of tumor.

Preparation of Ti-based homologous heterojunction and its photocatalytic degradation properties of tetracycline in seawater

Yubo Zhou,Xinbo Zhao,Zan Li,Junchao Yin,Liangmin Yu

To address ion interference in photocatalytic antibiotic degradation within high-salt seawater, this study innovatively designed and synthesized a titanium-based metal–organic framework (MOF) heterojunction composite. Specifically, a solvothermal technique was used to create MIL-125(HAC)/TiO 2 (TAC) photocatalysts using commercial TiO 2 (P25) and MIL-125(Ti) that had been treated with acetic acid. Remarkably, under 60 min of visible-light irradiation, the optimized TAC-2 sample achieved 95.7 % tetracycline (TC) degradation in seawater (compared to 98.1 % in deionized water) and retained >85 % efficiency after four cycles. Critically, its degradation rate constant (k=0.0355 min −1 ) in seawater was 44.4 times higher than that of P25, demonstrating exceptional resistance to salt-ion interference and photocatalytic activity. Mechanistic studies revealed that the performance enhancement arises from three synergistic mechanisms: (1) A mesoporous structure mitigating ion interference and strengthening TC-selective adsorption; (2) A Type-II heterojunction enabling efficient charge separation; (3) •O 2 − and h + identified as dominant active species (confirmed by EPR and quenching tests). Collectively, this work establishes an effective MOF heterojunction engineering strategy for efficient antibiotic removal in saline environments, ultimately contributing to marine pollution remediation.

FeNC with atomically dispersed iron atoms as a photosensitizer for combined photodynamic and photothermal therapy

Dan Li,Mengyao Zhang,Yudai Huang,Xin Hu,Junqing Hu

This research explores the efficacy of photodynamic therapy (PDT) and photothermal therapy (PTT) in combating chemotherapy-resistant diseases. This study focuses on enhancing tumor treatment effectiveness by leveraging the synergetic effects of combining PDT and PTT through the development of Fe-nitrogen-carbon (FeNC) nanoparticles with superior photostability. These nanoparticles, functioning as photosensitizers for the combined PDT/PTT treatment, can generate both type I and type II ROS and heat upon 808 nm irradiation. Notably, the FeNC nanoparticles demonstrate an exceptional photothermal conversion efficiency (34 %), surpassing commonly used PTT photosensitizers. In vitro and in vivo experiments corroborate the efficiency of FeNC as a photosensitizer in achieving significant tumor inhibition. In conclusion, the FeNC nanoparticles present promising applicability in the synergistic PTT/PDT treatment of tumors.

Therapeutic effects of polysaccharides from Anoectochilus roxburghii on type II collagen-induced arthritis in rats

Yaoli Guo,Qi Ye,Shuling Yang,Jinzhong Wu,Bingzhu Ye,Yanbin Wu,Zehao Huang,Chengjian Zheng

Anoectochilus roxburghii , a famous Chinese herbal medicine, has been commonly used for the treatment of liver disease, diabetes, and rheumatoid arthritis. Our study aimed to investigate the anti-rheumatoid arthritis effects of A . roxburghii polysaccharides (ARP), using the rat's model of type II collagen-induced arthritis (CIA). ARP was prepared by alcohol sedimentation and structurally characterized based on combined chemical, chromatographic and spectroscopic methods. High Performance Size Exclusion Chromatography-Multiangle Laser Light Scattering-Refrative Index (HPSEC–MALLS–RI) analysis revealed that ARP includes two peaks, and the weight-average molecular weight ( Mw ) of the principal one was estimated as 5.90 kDa with a relative content of 98.2%. Pharmacological results exhibited that ARP significantly decreased the arthritis index and ameliorated the inflammatory cell infiltration and the synovial tissue destruction in CIA rats. Additionally, ARP possessed significant NO production inhibitory effects and antioxidant activity. Further anti-inflammatory mechanism investigations indicated that ARP significantly inhibited the activation of nuclear factor κB (NF-κB) pathway by suppressing the phosphorylation of IκB and p65, which subsequently down-regulated the mRNA expressions of IL-1β and IL-6 in LPS-stimulated RAW 264.7 cells. These findings suggested that ARP has great potential in the development of functional foods and dietary supplements for the treatment of rheumatoid arthritis.

Organic semiconductor poly-2,6-diaminoanthraquinone composite and Bi loading synergistically enhance the photoelectrochemical cathodic protection performance of BiVO4

Juan Liu,Yingnan Qin,Meiqi Wang,Hengjun Liu,Tao Zheng,Jing Tian,Jing Wang

BiVO 4 -based materials have attracted extensive research as advanced photoanode for photogenerated cathodic protection. However, the performance of BiVO 4 -based photoanodes is greatly hindered by its weak charge migration and positive conduction band potential. Herein, poly-2,6-diaminoanthraquinone (PDAAQ)/Bi was grafted on BiVO 4 to improve the separation efficiency of photogenerated carriers and enhance the reducing ability of photogenerated electrons. Studies show that a type II heterojunction can be formed between PDAAQ and BiVO 4 , which accelerates the separation of photogenerated carriers at the interface. In addition, the Bi monomers anchored on BiVO 4 /PDAAQ then expand the light absorption range through the surface plasmon resonance effect. Consequently, the obtained BiVO 4 /PDAAQ/Bi photoanode exhibits a promising photocurrent density of 40 μA·cm -2 , while the photoinduced potential drop is 540 mV, which is approximately 1.4 times of BiVO 4 materials. These outcomes provide general ideas for the design of organic-inorganic heterozygous systems for achieving eco-friendly corrosion protection.

Two-pronged strategy: A mitochondria targeting AIE photosensitizer for hydrogen sulfide detection and type I and type II photodynamic therapy

Jizhong Shi,Manshan Liang,Yiting Qiu,Jin Zhang,Shihua Wang,Heng Fang,Yongsheng Jiang,Xiaoxia Ye,Yanshu Luo,Zu-Sheng Huang,Yun-Yun Quan

Multifunctional type-I photosensitizers (PSs) for hydrogen sulfide (H 2 S) detection and photodynamic therapy (PDT) of hypoxia tumors exhibits attractive curative effect but remains a challenging task. Herein, a mitochondria targeted aggregation-induced emission (AIE) photosensitizer TSPy-SS-P was designed and synthesized, which could be used for H 2 S detection and simultaneously type I and type II PDT. TSPy-SS-P had excellent selectivity and anti-interference abilities for endogenous and exogenous H 2 S detection in tumor cells. TSPy-SS-P was able to distinguish tumor cells with high level of H 2 S from normal cells by fluorescence “turn off” response to H 2 S. In addition, TSPy-SS-P showed type Ⅰ and type Ⅱ reactive oxygen species (ROS) generation ability to effectively ablate hypoxic tumor cells. TSPy-SS-P showed mitochondria targeting capacity which could produce ROS in situ to disrupt mitochondria and promote cell apoptosis. In vivo PDT experiments showcased that TSPy-SS-P had excellent tumor retention capability, effective tumor ablation ability and good biocompatibility. This work provided a two-pronged strategy to design organelles targeted photosensitizers for H 2 S detection and effective PDT of tumors.

Loss of PTPN21 disrupted mitochondrial metabolic homeostasis and aggravated experimental pulmonary fibrosis

Lian Hui,Xu Kai,Chang Airu,Wang Yaxuan,Ma Shuaichen,Cheng Lianhui,Zhao Wenyu,Xia Cong,Wang Lan,Yu Guoying

Idiopathic pulmonary fibrosis (IPF) is a high-mortality lung disease with unclear pathogenesis. Convincing evidence suggests that an imbalance in mitochondrial homeostasis resulting from repeated injury to alveolar epithelial type 2 cells (AEC2) underlies IPF. Non-receptor protein tyrosine phosphatase 21 (PTPN21) performs various functions in cancer; however, its role in IPF has not been studied. This study aimed to investigate the role of PTPN21 in lung fibrosis. The experimental results showed that loss of PTPN21 exacerbated lung fibrosis by increasing cell numbers in bronchoalveolar lavage fluid, lung hydroxyproline content, and extracellular matrix protein expression of fibronectin and α-smooth muscle actin (α-SMA) in bleomycin-challenged mouse lungs. In A549 cells (AEC2), knockdown of PTPN21 suppressed focal adhesion and migration, reduced mitochondrial fission and increased fusion, increased the level of mitochondrial superoxide, decreased mitochondrial membrane potential and ATP levels. Simultaneously, knockdown of PTPN21 impaired autophagy, and increased intracellular reactive oxygen species levels. Treatment of fibroblasts (MRC-5) and primary human lung fibroblasts (PHLF)) with the supernatant from PTPN21-knockdown A549 cells increased the expression of fibronectin, collagen 1 and α-SMA. Conversely, overexpression of PTPN21 in A549 cells produced opposite effects. However, treatment of MRC-5 and PHLF with the supernatant from PTPN21-overexpressing A549 cells only slightly reduced the expression of fibronectin, collagen 1 in MRC-5 cells, but did not change the expression of α-SMA. In summary, this study revealed that the loss of PTPN21 in epithelial cells disrupted mitochondrial metabolic homeostasis, leading to epithelial cell inactivation and increased the deposition of extracellular matrix proteins in fibroblasts, thereby exacerbating experimental pulmonary fibrosis.

Oxymatrine Liposomes for Intervertebral Disc Treatment: Formulation, in vitro and vivo Assessments

Huan Wang,Yifan Ding,Wei Zhang,Kang Wei,Yaping Pei,Chenming Zou,Chong Zhang,Jiahui Ding,Huang Fang,Songwei Tan

Purpose Intervertebral disc degeneration (IVDD) is the main cause of modern low back pain, leading to high societal economic costs. To find an effective medical treatment for this disease, oxymatrine liposomes (OMT-LIP) were prepared with the pH-gradient method.Materials and Methods Nucleus pulposus (NP) cells from Sprague–Dawley rats were used for the cell experiments. Kunming mice were used for in vivo imaging. LIP were employed to deliver OMT, and the particle size, ζ-potential, morphology, in vitro stability and in vitro release characteristics were evaluated. The OMT-LIP targeting effect was measured by in vivo imaging. Cell Counting Kit-8 assays were used to detect the cytotoxicity of OMT and OMT-LIP on NP cells. Therapeutic efficacy was measured by Western blot, real-time quantitative polymerase chain reaction, and apoptosis assays. Radiologic analysis was performed to evaluate the therapeutic effects in vivo. Results Orthogonal test results revealed that the mass ratio of egg yolk phosphatidylcholine to cholesterol was the key factor to effectively trap OMT in LIP. Optimal OMT-LIP showed multivesicular structure with entrapment efficiency of 73.4 ± 4.1%, particle size of 178.1 ± 2.9 nm, and ζ-potential of –13.30 ± 2.34 mV. OMT-LIP manifested excellent stability in vitro and presented significantly longer sustained release compared to OMT solution in phosphate-buffered saline (pH 7.4). OMT-LIP conspicuously increased OMT accumulation in the degenerative disc, attenuated NP cell apoptosis, reduced the expression of matrix metalloproteinases 3/9 and interleukin-6, and decreased degradation of type II collagen. In in vivo study, X-ray demonstrated that OMT-LIP inhibited IVDD.Conclusion OMT-LIP may be a useful treatment to alleviate disc inflammation and IVDD.

Phosphorus Quantum Dots-Facilitated Enrichment of Electrons on g-C3N4 Hollow Tubes for Visible-Light-Driven Nicotinamide Adenine Dinucleotide Regeneration

Dong Yang,Yishan Zhang,Hongjian Zou,Shaohua Zhang,Yizhou Wu,Ziyi Cai,Jiafu Shi,Zhongyi Jiang

Nicotinamide cofactors (e.g., NADH) are essential hydrogen sources for the majority of enzyme reduction reactions. Currently, highly efficient and in situ cofactor regeneration is urgently required but remains challenging. In this study, red phosphorus quantum dots were decorated onto graphitic carbon nitride hollow tubes to prepare high-performance heterojunction photocatalyst (g-C3N4-HTs@rP-QDs) for visible-light-driven NADH regeneration. Energy band analysis combined with photoelectrochemical measurements demonstrates rP-QDs not only act as additional photosensitizers for charge generation but also constitute a type II heterojunction with g-C3N4-HTs for charge transfer, facilitating enrichment of electrons on the g-C3N4-HTs surface. Such enrichment of electrons for g-C3N4-HTs@rP-QDs offers ∼5 times higher reducing active sites than bulk g-C3N4 to trigger the NADH regeneration reaction under visible-light (LED, 405 nm) illumination, thus acquiring superior NADH regeneration efficiency with an initial reaction rate (2 min) of 0.247 ± 0.012 mmol g–1 min–1. In the concerned experimental range, higher g-C3N4-HTs/rP-QDs mass ratios or light intensities that are beneficial for the electron enrichment could result in enhanced regeneration efficiency. The bioactivity of NADH toward enzyme catalysis is further demonstrated by alcohol dehydrogenase-catalyzed hydrogenation of formaldehyde, which enables the sustainable production of methanol.

In situ formation of red/black phosphorus-modified SiO2@g-C3N4 multi-heterojunction for the enhanced photocatalytic degradation of organic contaminants

Jiancheng Li,Chi Wang,Yixing Ma,Kai Li,Yi Mei

A new heterojunction material BP/RP-g-C3N4/SiO2 was obtained by a one-step ball milling method, and its photocatalytic capacity was researched by the degradation of Rhodamine B (RhB) and ofloxacin (OFL) in simulated sunlight. The construction of an in situ BP/RP heterojunction can achieve perfect interface contact between different semiconductors and effectively promote the separation of photogenerated carriers. The composite material was well characterized, which proved that the multi-heterogeneous structure was prepared. Furthermore, the type II heterojunction was formed between the g-C3N4 and BP/RP interface, playing an important role in the degradation and promoting electron transfer. The degradation effect of BP/RP-g-C3N4/SiO2 on RhB reached 90% after 26 min of simulated solar irradiation, which was 1.8 times that of g-C3N4/SiO2. The degradation of OFL by BP/RP-g-C3N4/SiO2 reached 85.3% after illumination for 50 min, while the degradation of g-C3N4/SiO2 was only 35.4%. The mechanisms were further discussed, and ˙O2− and h+ were found to be the main active substances to degrade RhB. The catalyst also revealed distinguished stability of catalyst and recyclability, and the degradation effect of RhB can still realize 85% after 4 runs of experiment. Thus, this study provided a novel method for the design and preparation of multi-heterojunction catalysts in the removal of organic pollutants from wastewater.

A Novel Curcumin-Loaded Nanoplatform Alleviates Osteoarthritis by Inhibiting Chondrocyte Ferroptosis

Weibei Sheng,Aikang Li,Yaohang Yue,Qichang Wang,Fei Yu,Jian Weng,Jianjing Lin,Yingqi Chen,Hui Zeng,Deli Wang,Jun Yang,Peng Liu

Osteoarthritis (OA) is a chronic degenerative joint disease characterized by the degradation of articular cartilage. Recent studies have demonstrated that chondrocyte ferroptosis plays a crucial role in the progression of OA. Consequently, developing nanomedicines that suppress chondrocyte ferroptosis is a promising strategy for OA treatment. However, there are few reports on nanomedicines specifically targeting chondrocyte ferroptosis for OA therapy. In this study, Curcumin-loaded nanoparticles (Cur-NPs) are fabricated to suppress chondrocyte ferroptosis by regulating reactive oxygen species (ROS), ferrous ion (Fe 2 ⁺), and Acyl-CoA Synthetase Long-Chain Family Member 4 (ACSL4) levels of chondrocyte. This is achieved by combining the functions of curcumin and an amphiphilic block copolymer with ROS scavenging and iron-chelating properties. The in vitro anti-ferroptotic effects of Cur-NPs are thoroughly investigated. The findings indicate that Cur-NPs decrease the expression of ferroptosis markers such as ROS, Fe 2 ⁺, and ACSL4, while protecting the mitochondrial membrane potential of chondrocytes. Additionally, Cur-NPs attenuated lipid peroxidation in chondrocytes. Furthermore, Cur-NPs significantly reduced the expression of the catabolic factor Matrix Metallopeptidase 13 (MMP13) and increased the expression of the anabolic factor Collagen type II (Col II) in vitro. This study demonstrates that Cur-NPs exhibit enhanced chondroprotective effects through anti-ferroptotic actions, presenting a promising approach for inhibiting chondrocyte ferroptosis using bioactive nanomaterials in OA treatment.
