Synergistic protective effects of β-glucan and Lactobacillus johnsonii NSP009 in ulcerative colitis

Shanshan Zhang,Yonggan Sun,Linlin Gao,Ziyi Zhang,Yongkang Yu,Dechang Xu,Shaoping Nie

The combination of probiotics and prebiotics has emerged as a promising strategy for managing ulcerative colitis (UC). Among these, β-glucan (BG) and Lactobacillus johnsonii have individually shown potential in alleviating colitis, with BG notably enriching L. johnsonii populations, suggesting a synergistic therapeutic potential when combined. This study investigates the protective effects of a novel synbiotic formulation, BG and L. johnsonii NSP009 (LJ), termed BGLJ, in a dextran sulfate sodium (DSS)-induced colitis mouse model. Our results reveal that BGLJ significantly mitigated DSS-induced weight loss, reduced the disease activity index, and alleviated intestinal inflammation and barrier dysfunction. Notably, BGLJ markedly increased L. johnsonii abundance in mouse feces, with LJ producing substantial levels of indole-3-lactic acid (ILA), an aryl hydrocarbon receptor (AhR) agonist. BGLJ treatment robustly elevated ILA levels in the gut, activating AhR and upregulating IL-22 production. Collectively, our findings highlight the therapeutic potential of the BGLJ-based synbiotic in maintaining intestinal homeostasis and underscore its promise as a novel intervention for UC management.

Effects of fermented Arctium lappa L. root by Lactobacillus casei on hyperlipidemic mice

MingJu Chen,Yuxiao Wu,Hongxuan Yang,Tianfeng Liu,Tongkun Han,Wangqiang Dai,Junyue Cen,Fan Ouyang,Jingjing Chen,Jianxin Liu,Lin Zhou,Xuguang Hu

Introduction: This study aimed to establish a fermentation system based on Lactobacillus casei (LC) and Arctium lappa L. root (AR) to investigate its effects. The objectives included comparing metabolite profiles pre- and post-fermentation using untargeted metabolomics and evaluating the impact of LC-AR in high-fat diet-induced hyperlipidemic mice. Methods: Untargeted metabolomics was used to analyze differences in metabolites before and after fermentation. In vitro antioxidant activity, liver injury, lipid levels, pro-inflammatory cytokine levels, and cholesterol-related mRNA expression were assessed. 16S rRNA sequencing was conducted to evaluate changes in gut microbiota composition. Results: LC-AR exhibited stronger antioxidant activity and higher metabolite levels than AR. It also improved liver injury as well as better regulation of lipid levels, pro-inflammatory cytokine levels, and cholesterol-related mRNA. 16S rRNA analysis revealed that LC-AR decreased the Firmicutes/Bacteroidetes ratio, which correlated negatively with triglycerides, total cholesterol, and low-density lipoprotein cholesterol levels.Discussion: These findings suggest that LC-AR may serve as a promising functional food and drug raw material for improving hyperlipidemia, particularly through its beneficial effects on gut microbiota and lipid regulation.

Bioinspired gelated cell sheet–supported lactobacillus biofilm for aerobic vaginitis diagnosis and treatment

Yueyue Gui,Qingfei Sun,Kexin Li,Longjia Lin,Han Zhou,Jiehua Ma,Chao Li

Aerobic vaginitis (AV) is a long-standing inflammatory disease that affects female patients. The use of antibiotics is a common means for AV treatment, but it will indiscriminately kill both pathogenic bacteria and beneficial strains, which easily causes vaginal dysbacteriosis and infection recurrence. Herein, we describe a bioinspired strategy for fabricating gelated cell sheet–supported lactobacillus biofilms (GCS-LBs) for AV treatment. Compared with common planktonic probiotic formulations, probiotic biofilms forming on a robust GCS exhibit enhanced stress tolerance and better colonization capacity in the mouse vagina. Moreover, DNA nanodevices are decorated on the GCS and dynamically report the microenvironment change of biofilms for timely evaluating bacterium activity, both in vitro and in vivo. Consequently, GCS-LBs are used for treating AV in an Escherichia coli–infected mouse model, which shows enhanced therapeutic efficacy compared with conventional antibiotic or lactobacillus monotherapy. Overall, the GCS-LB shows promise as a potent multifunctional tool to combat bacterial infection.

Glycerin-Betaine-Based Natural Eutectic Solvent Enhanced the Spray Drying Resistance of Lactobacillus

Jiang Jingya,Song Xiaoning,Zhao Liming,Wang Song,Hou Baochao,Li Baolei,Dudu Olayemi Eyituoyo,Yi Huaxi,Zhang Lanwei,Gong Pimin

Spray drying is a promising technique for embedding and stabilizing lactic acid bacteria. However, bacteria are unable to withstand high-temperature environment. Naturally small molecules fall short in meeting the evolving demands of production. Therefore, it is imperative to explore non-chemical pretreatment methods urgently, with the aim of enhancing their protective efficacy. In this study, natural deep eutectic solvents (NADES) were synthesized by glycerin-betaine to improve the survival rates of spray-dried Lactobacillus bulgaricus sp1.1. Results revealed that the spray drying survival rate of L. bulgaricus sp1.1 after adding NADES was 30.54%, which was 4.72 times higher than adding equal amounts of untreated glycerin and betaine, and the number of viable bacteria decreased less than 1 log after 6 months of storage. The investigation revealed that the impact of NADES was localized external to the cell rather than within. Through crosslinking with the peptidoglycan on the cell wall, NADES bolstered the cell wall structure thereby creating a protective barrier. This resulted in a reduction of damage to the membrane and ribosome induced by high temperatures during the lactic acid bacteria’s exposure. This study synthesized NADES using current protective agents as an effective method to enhance heat resistance of lactic acid bacteria for spray drying for the first time, and the lactic acid bacteria powder with high survival rate and high storage stability was successfully prepared, which provides a new approach for its industrial application. Lactobacillus plantarum ameliorates intrahepatic cholestasis by modulating the gut microbiota and tryptophan metabolism

Shijing Dong,Liping Guo,Chenyang Xuan,Chenhui Zhu,Zongze Han,Nian Chen,Xinyu Wang,Meng Shen,Xiaoxuan Wei,Hui Yang,Xue Zhang,Man Liu,Simin Zhou,Lu Zhou

Lactobacillus plantarum (L. plantarum ) has shown protective effects in various liver diseases, yet its therapeutic potential and mechanisms in intrahepatic cholestasis remain unelucidated. This study aimed to investigate the therapeutic efficacy and mechanism of L. plantarum in intrahepatic cholestasis using two well-established mouse models induced by α-naphthylisothiocyanate (ANIT) and 3,5-diethoxycarbonyl-1,4-dihydrocollidine (DDC). The results indicated that L. plantarum significantly alleviated cholestasis-related hepatic inflammation, ductular proliferation, fibrosis, and improved gut barrier integrity. The 16S rRNA sequencing and untargeted metabolomics analysis revealed that the intestinal abundance of L. plantarum was significantly increased after intervention and showed a positive correlation with the level of indole-3-acetic acid (IAA), a key functional metabolite of tryptophan ( r  = 0.712, p  = 0.003). The elevated IAA levels were closely associated with the improvement of key liver injury indicators. Exogenous IAA supplementation replicated the hepatoprotective effects of L. plantarum in intrahepatic cholestasis. Transcriptome analysis indicated that IAA mediated these benefits primarily through aryl hydrocarbon receptor (AHR) activation, and AHR inhibition markedly attenuated its protective effects. This study reveals that L. plantarum ameliorates intrahepatic cholestasis by enhancing tryptophan metabolism to promote IAA production, which in turn activates the IAA-AHR axis, providing new insights into probiotic and metabolite-based therapies for intrahepatic cholestasis.

Mechanism study on enhanced foaming properties of individual albumen proteins by Lactobacillus fermentation

Jie Jia,Bing Ji,Liangjie Tian,Mei Li,Mei Lu,Long Ding,Xuebo Liu,Xiang Duan

Our recent research showed that moderate Lactobacillus fermentation endowed egg white with stronger foaming properties. But the individual contribution of albumen proteins and the relative mechanisms during the process were still not clear. In this study, three major egg white proteins (ovalbumin, ovomucin and ovomucoid) were singly fermented by multiple Lactobacillus ( Lactobacillus delbruekii ssp. bulgaricus , Lactobacillus acidophilus , Streptococcus thermophilus and Bifidobacterium animalis ) for 0, 3, 6 and 9 h, and the surface activity, degradation, apparent viscosity and foaming properties were investigated. For ovalbumin, the surface hydrophobicity largely increased at 3 h and then began to decrease; combined with the gradual degradation by protease, the foaming ability of ovalbumin significantly increased at 3 h and then decreased at 6 and 9 h of fermentation. The fermented ovomucin and ovomucoid possessed an improved surface characteristics and appropriate apparent viscosity; correspondingly, the both proteins exhibited the enhanced foaming ability and stability after fermentation. This work revealed the molecular mechanism behind Lactobacillus fermentation treatment increased foaming properties of egg white, and provided basic information on the application of fermentation approach in improving functionalities of egg white in industry.

Selective uptake determines the variation in degradation of organophosphorus pesticides by Lactobacillus plantarum

Shaofeng Yuan,Changjian Li,Hang Yu,Yunfei Xie,Yahui Guo,Weirong Yao

Organophosphorus pesticides (OPPs) are widely used worldwide, leading to varying degrees of residues in food. Lactic acid bacteria (LAB) can degrade OPPs by producing phosphatase. This study explored the reasons for the variation in the degradation of different OPPs by Lactobacillus plantarum . The results showed that the degradation effects of OPPs by L. plantarum (intact cells) varied greatly, the degradation rate constant of phoxim was 1.65-fold higher than that of dichlorvos. However, the phosphatase extracted from L. plantarum had no degradation selectivity for OPPs in vitro . It was speculated that the selective uptake of cells determines this degradation selectivity. The results of molecular docking supported this hypothesis because there was no difference in the binding energies between phosphatase and OPPs, while the binding energies between phosphate-binding protein and pesticides were different, and they were negatively correlated with the degradation rate constants of the eight OPPs by L. plantarum .

Regulatory effects of Lactobacillus fermented black barley on intestinal microbiota of NAFLD rats

Chuang Zhu,Qi Guan,Chenwei Song,Lingyue Zhong,Xinwen Ding,Hui Zeng,Pan Nie,Lihua Song

Gut microbiota dysbiosis and oxidative stress may play important roles in the progression of nonalcoholic fatty liver disease (NAFLD). Fermented foods contain probiotics and other bioactive components that may improve gastrointestinal health and provide other health benefits. Here, we investigated the effect of Lactobacillus -fermented black barley on NAFLD rats. Adult Sprague Dawley rats were randomized into four groups: normal chow diet (NC), high-fat diet (HF), HF + fermented black barley treatment (HB) and HF +  Lactobacillus treatment (HL). The rats in the HB and HL groups were continuously administered fermented black barley or Lactobacillus , respectively, for 12 weeks (1 mL/100 g·BW, containing 1 × 10 8 CFU/mL Lactobacillus ). Compared with the HF treatment, HB treatment effectively inhibited the increase in body weight, liver and abdominal fat indexes and hepatic lipids ( p  < 0.01), increased hepatic SOD activity ( p  < 0.05), decreased thiobarbituric acid reactive substances (TBARSs) ( p  < 0.01) and improved liver function. Moreover, Lactobacillus -fermented black barley exhibited regulatory effect on high-fat diet-induced intestinal microbiota dysbiosis by increasing the relative microbiota abundance and diversity, increasing the relative abundance of Bacteroidetes , decreasing the Firmicutes/Bacteroidetes ratio, increasing the abundances of some intestinal probiotics (such as Akkermansia and Lactococcus ), and influencing some of the fecal metabolites related to hormones and lipid metabolism. The supplementation of fermented cereal food might be a new effective and safe preventive dietary strategy against NAFLD.

Physiological function analysis of Lactobacillus plantarum Y44 based on genotypic and phenotypic characteristics

Yuan Gao,Yujun Liu,Mengying Sun,Heping Zhang,Guangqing Mu,Yanfeng Tuo

ABSTRACT In our previous studies, Lactobacillus plantarum Y44 showed antioxidant activity and favorable gastric and intestinal transit tolerance. In the current study, we investigated the physiological function of L. plantarum Y44 based on an analysis of its genotype and phenotype. The complete genome of L. plantarum Y44 contained a single circular chromosome of 3,255,555 bp, with a GC content of 44.6%, and a single circular plasmid of 51,167 bp, with a GC content of 38.8%. The L. plantarum Y44 genome contained 3,293 genes including 3,112 protein coding sequences, 16 rRNAs, 66 tRNAs, 4 small (s)RNAs, and 95 pseudo genes. Lactobacillus plantarum Y44 could metabolize 24 different carbohydrate sources. Nineteen complete phosphoenolpyruvate-dependent sugar phosphotransferase system complex genes and intact Embden-Meyerhof-Parnas pathway and hexose monophosphate pathway enzyme genes, as well as abundant carbohydrate active enzyme genes, were identified in the L. plantarum Y44 genome. We also identified genes related to the biosynthesis of exopolysaccharide and surface proteins. Surface proteins played an important role in the L. plantarum Y44 adhesion to HT-29 cell monolayers, as evidenced by the removal of cell surface proteins leading to decreased adhesion capacity. The L. plantarum Y44 genome contained genes encoding chaperones, intracellular proteases, and 2-component systems, which were associated with the general stress response. Genes encoding bile salt hydrolase, F 0 F 1 -ATPase, Na + /H + -antiporter, H + /Cl − exchange transporter, cyclopropane-fatty acyl-phospholipid synthase, and alkaline shock protein were identified in the L. plantarum Y44 genome, which might explain the strain's favorable gastric and intestinal transit tolerance. Some genes associated with encoding the NADH system, glutathione system, and thioredoxin system were predicted via in silico analysis and might account for the strain's ability to scavenge reactive oxygen species. Lactobacillus plantarum Y44 was susceptive to 7 antibiotics and did not produce biogenic amines, likely due to the absence of acquired antibiotic resistance genes and amino acid decarboxylase genes. The phenotype profile of L. plantarum Y44 was associated with its genetic characteristics, indicating that strains with certain physiological functions can be screened by analyzing their phenotypic and genotypic characteristics. Lactobacillus plantarum Y44 has the potential to be used as a starter culture in fermented dairy products.

Composition and Characteristics of Yam Juice Fermented by Lactobacillus plantarum and Streptococcus thermophilus

Wenxue Chen,Jianzhong Zhu,Hui Niu,Yizhe Song,Weimin Zhang,Haiming Chen,Weijun Chen

In this study, Lactobacillus plantarum (LP), alone or in combination with Streptococcus thermophilus , was used to ferment yam juice. Changes in the composition (phenols, organic acids, reducing sugars and volatile substances) and functional characteristics (antioxidative activity and ability to regulate the intestinal flora) of yam juice during fermentation were investigated. The results showed that the total phenolic (TP) content increased from 201.27 to 281.27 and 285.77 μg/mL for LP- and L. plantarum and S. thermophilus (LPST)-fermented yam juice, respectively. The antioxidative activity of yam juice improved significantly after fermentation, highly correlating with its TP content. In addition, LP- or LPST-fermented yam juice had positive effects on members of the human intestinal flora, improving the activity of Bifidobacterium and inhibiting the growth of Escherichia coli . Sensory analysis showed that LPST-fermented yam juice had a highest score. The results of this study showed that fermented yam juice can serve as a healthy beverage for consumers with low immunity or an imbalance of the intestinal flora.

Study of soybean gel induced by Lactobacillus plantarum: Protein structure and intermolecular interaction

Xiaoyu Yang,Yue Su,Liang Li

The change of protein structure and intermolecular interaction during the formation of soy protein isolate (SPI) gel induced by Lactobacillus plantarum ( L . plantarum ) were investigated. In the present study, the peak value of UV–Vis spectra decreased gradually with the prolongation of fermentation time. Fermentation treatment resulted in α-helix content decreasing by 60.16%, while β-sheet content increased by 68.15% (P < 0.05). The surface hydrophobicity was decreased by 62.31% (P < 0.05) due to the molecular aggregation of SPI. Correspondingly, the free sulfydryl group content decreased by 58.48% (P < 0.05). These results highlighted that the protein structure and intermolecular interaction of SPI could be significantly changed by L . plantarum fermentation. The main intermolecular interactions maintaining the fermentation-induced gel were hydrophobic interactions and disulfide bonds. This work provided theoretical support for the establishment of the mechanism of fermentation-induced gel and the rational application of L . plantarum in soybean products.

Hypocholesterolaemic action of Lactobacillus casei F0822 in rats fed a cholesterol-enriched diet

C.F. Guo,J.Y. Li

Elevated serum cholesterol is a major risk factor for coronary artery disease. Probiotics may manage elevated cholesterol. The hypocholesterolaemic effects of Lactobacillus casei F0822 and its functional mechanisms were investigated in rats . The serum total cholesterol, low-density lipoprotein cholesterol and liver total cholesterol levels significantly decreased in rats fed a high-cholesterol diet plus drinking water supplemented with viable cells of strain F0822; however, there was no significant difference in the serum high-density lipoprotein cholesterol levels among the treatment groups. The hypocholesterolaemic mechanisms of strain F0822 were attributed to its ability to suppress the reabsorption of bile acids into the enterohepatic circulation though hydrolysis of conjugated bile acids in the small intestine, binding of deoxycholic acid and hyodeoxycholic acid in the large intestine, and increase in propionate:acetate ratio in the large intestine in rats. Strain F0822 may be a promising probiotic culture with potential hypocholesterolaemic action in human.
