Facile synthesis, characterization, and in vitro biocompatibility of free-standing titania hollow microtubes

Yixuan Wang,Fenghao Zhang,Song Chen,Akiyoshi Osaka,Weiyi Chen

Titania nanotube (NT) arrays have been widely used as cell-supporting matrices. However, cells are always seeded on the porous surface of the NT array and have very limited interactions with each individual NT in the array. In this study, titania hollow microtubes (HMTs) were synthesized via a gelatin-template sol-gel route and then utilized as free-standing cell-supporting matrices for the first time. The resultant titania HMTs were studied by field emission scanning electron microscopy, energy-dispersed spectroscopy, X-ray diffraction, and Fourier-transform infrared spectroscopy. Each HMT was composed of rutile-type titania nanoparticles with diameters of 50–100 nm and a diameter of 50–100 µm. The results from a leaching liquor assay demonstrated good biocompatibility of titania HMTs. Each HMT has been demonstrated to independently support the adhesion and proliferation of osteoblast MC3T3-E1 cells. For comparison, titania NT arrays, not independent titania NT, only supported the adhesion of cells on their porous surface. Thus, the resultant titania HMTs are applicable to free-standing and biocompatible cell-supporting matrices.

Endowing polyetheretherketone with anti-inflammatory ability and improved osteogenic ability

Yue Yu,Kenan Xie,Lu Xie,Yi Deng

Both osteogenesis and anti-inflammatory bioactive materials play a vital role in the regeneration of skeletal defects. Bone inflammation is hard to cure and can lead to malformation or amputation. The purpose of this study is to use anti-inflammatory drugs to endow polyetheretherketone (PEEK) with the dual ability to achieve anti-inflammatory effects while maintaining favorable biocompatibility. In this experiment, the porous PEEK was immersed in an aspirin (ASP) solution after sulfonation, and the obtained porous PEEK had significantly improved the anti-inflammatory abilities. Additionally, grafting the bone forming peptide (BFP) onto the porous PEEK can distinctly enchance the osteogenesis capability. The effects of the BFP polypeptide on the proliferation of MC3T3-E1 cells and ALP activity, and the effects of aspirin on inflammation were systematically investigated. The modified material showed favorable biocompatibility and osteogenic ability. The results suggest that the combination of the BFP polypeptide with aspirin may lead to a synergetic effect on the stimulation of osteogenesis and on the reduction of inflammation.

Facile Preparation and Properties of Porous Poly(vinyl alcohol)/Trehalose/Nano-Clay Hydrogels with High Mechanical Strength for Potential Application in Bone Tissue Engineering

Changxin Xiang,Qing Zhang,Zijian Guo,Zehua Wang,Weiyi Chen,Xiaona Li,Xiaochun Wei,Pengcui Li

Among various biomaterials, hydrogels are considered as the most attractive tissue engineering material due to their resemblances to extracellular matrix. However, due to the poor mechanical properties, poly(vinyl alcohol) hydrogels are restricted to be applied in load-bearing tissue repair and regeneration. In this work, we designed a series of porous biocompatible composite hydrogels with tough mechanical properties for bone tissue engineering, consisting of poly(vinyl alcohol), trehalose and nano-clay. These novelty hydrogels were prepared by a facile one-pot method. Compositions, microscopic structures, and mechanical properties of hydrogels were investigated in detail. The maximum compressive strength, compressive modulus, tensile strength, elastic modulus, and toughness of hydrogels attained about 2.71 MPa, 0.163 MPa, 0.741 MPa, 0.084 MPa, and 0.94 MJ/m 3 , respectively. These as-prepared hydrogels exhibited the excellent mechanical properties, which were ascribed to the hydrogen bond interactions between molecules and nano-enhancement effect of the nano-clay. Meanwhile, as-prepared hydrogels also exhibited a three-dimensional and interconnected porous microstructure, which facilitated the transport of nutrients to promote cells growth. In addition, the mechanical properties and microscopic structures of hydrogels could be controlled by adjusting the concentrations of trehalose and nano-clay. In vitro cell experiments demonstrated that the hydrogels had good biocompatibility. And hydrogels possessed the excellent ability to induce MC3T3-E1 cells osteogenesis to accelerate bone defects repair. Above illustrated results demonstrated that the potential applications of the hydrogels used as bone tissue engineering materials. Therefore, we anticipated this kind of poly(vinyl alcohol)-based composite hydrogel could develop a new strategy for bone tissue repair

Controlling Preosteoblast Behavior through Manganese Vacancy-Rich Birnessite with Enhanced Divalent Cation Modulation of Fibronectin–Integrin Interactions

Shiwei Liu,Kai Li,Tao Hu,Xiaodong Wu,Haiyong Ao,Youtao Xie,Xuebin Zheng

The osteoblast-function associated integrin–ligand interactions are known to be dependent on divalent cations, but there is scarce information about the effect of Ca 2+ and Mg 2+ adsorption properties of an implant surface on integrin α 5 β 1 –fibronectin interactions and the subsequent osteoblast behavior. Cation adsorption properties are closely associated with the extent of structural defects in materials. Here, the birnessite-type MnO 2 coatings with different amounts of manganese vacancy ( V Mn ) are prepared on Ti substrates via hydrothermal treatment (denoted as 1M100-Ti, 5M100-Ti, and 5M120-Ti). The 5M120-Ti surface with hierarchical flower-like structure and the most stacked [MnO 6 ] layers possess the highest V Mn content, which is determined by the ratios of Mn 3+ /Mn 4+ and K/Mn. The increased V Mn content enhances the Ca 2+ and Mg 2+ adsorption properties of the birnessite coatings. When immersed in Ca 2+ /Mg 2+ aqueous solution, the birnessite coatings significantly promote fibronectin adsorption and the exposure of cell-binding domains on adsorbed fibronectin with 5M120-Ti showing the greatest enhancement. Accordingly, the 5M120-Ti surface provides more α 5 β 1 -integrin-specific instructions to MC3T3-E1 preosteoblasts capable of enhancing spreading and osteoblastic differentiation compared to two other samples. This study may provide a prospective approach of modulation of integrin–ligand interactions for controlling cell behavior by adjusting metal vacancies in metal oxides coatings.

A novel icariin-encapsulated PLGA/nBM composite biomimetic microspheres for bone repair

Zhenxu Wu,Guoqing Niu,Zongtai Xiong,Xinxiang Ding,Yongbo Li,Peibiao Zhang,Feng Wu

Deer bone in traditional Chinese medicine is valued for its unique biomimetic bone matrix components. Icariin (Ica), which is claimed by Chinese medicine as a bone-repairing agent, is also widely accepted. In this report, a novel Ica-encapsulated PLGA/nano deer bone meal (Ica@PLGA/nBM) biomimetic microsphere was fabricated. The Ica could be released from microspheres in a controlled manner. In vitro results showed that the microsphere had good biocompatibility due to the addition of nBM and the MC3T3-E1 cells could adhere to and grow well on the microspheres. Moreover, Ica and nBM in microspheres could synergically promote the ALP activity of MC3T3-E1 cells significantly ( p < 0.05), which indicated the acceleration of osteogenic differentiation. The RT-PCR results showed that the expression levels of bone matrix proteins (Col1 & OPN) were enhanced by the composite ( p < 0.05). In addition, Ica and nBM in microspheres also obviously promoted the expression of RUNX2 and BMP-2, respectively ( p < 0.05). The in vivo results demonstrated the effective promotion of Ica@PLGA/nBM on complete bony connection to cover the defect site and the bone defect repair. And the Ica(M)@PLGA/nBM microspheres provided the best repair results. All the results confirmed that the Ica@PLGA/nBM composite biomimetic microsphere has the potential for clinical application.

Hydroxyapatite Gradient Poly (Vinyl Alcohol)/Bacteria Cellulose Bone Scaffold via Buoyancy-Driven Gradient Method

Zhang Quanchao,Wang Huiwen,Shi Jian,Luo Honglin,Yin Chen,Wan Yizao

The repair of bone defects necessitates the establishment of sophisticated gradients. However, most strategies for gradient casting require specialized apparatus and have high equipment requirements. Therefore, there is a strong desire to develop a simple method for constructing gradient scaffolds that mimic the hierarchical structure of native bone. In this study, we prepared poly (vinyl alcohol)/bacterial cellulose (PVA/BC) scaffolds with gradient hydroxyapatite (HAp) content using the buoyancy-driven gradient (BG) method. Scanning electron microscopy characterization revealed that HAp gradient scaffold exhibited the gradient microstructure and HAp content. Furthermore, HAp gradient scaffold demonstrated enhanced adhesion, spreading, and proliferation of MC3T3-E1 cells and displayed excellent osteogenic ability. Additionally, when applied in subcutaneous implantation models in mice, HAp gradient scaffold showed superior biocompatibility compared to other scaffolds tested in vitro. These results present a straightforward approach for fabricating gradient HAp bone scaffolds that hold great potential as candidate materials for bone regeneration.

Tough physically crosslinked poly(vinyl alcohol)-based hydrogels loaded with collagen type I to promote bone regeneration in vitro and in vivo

Changxin Xiang,Zehua Wang,Qing Zhang,Zijian Guo,Xiaona Li,Weiyi Chen,Xiaochun Wei,Pengcui Li

Poly(vinyl alcohol) (PVA) hydrogels exhibit great potential as ideal biomaterials for tissue engineering, owing to their non-toxicity, high water content, and strong biocompatibility. However, limited mechanical strength and low bioactivity have constrained their application in bone tissue engineering. In this study, we have developed a tough PVA-based hydrogel using a facile physical crosslinking method, comprising of PVA, tannic acid (TA), and hydroxyapatite (HA). Systematic experiments were conducted to examine the physicochemical properties of PVA/HA/TA hydrogels, including their compositions, microstructures, and mechanical and rheological properties. The results demonstrated that the PVA/HA/TA hydrogels possessed the porous microstructures and excellent mechanical properties. Furthermore, collagen type I (Col I) was used to further improve the biocompatibility and bioactivity of PVA/HA/TA hydrogels. In vitro experiments revealed that PVA/HA/TA/COL hydrogel could offer a suitable microenvironment for the growth of MC3T3-E1 cells and promote their osteogenic differentiation. Meanwhile, the PVA/HA/TA/COL hydrogel demonstrated the ability to promote bone regeneration and osteointegration in a rat femoral defect model. This study provides a potential strategy for the use of PVA-based hydrogels in bone tissue engineering.

Interleukin-4-Loaded Heparin Hydrogel Regulates Macrophage Polarization to Promote Osteogenic Differentiation

Yuhao Zhao,Xiaofei Feng,Zhenrui Zhao,Zhengdong Song,Wenji Wang,Haiyan Zhao

In bone tissue engineering, biological scaffolds are designed with structural and functional properties that closely resemble the extracellular environment, aiming to establish a microenvironment conducive to osteogenesis. Macrophages hold significant potential for promoting osteogenesis and modulating the biological behavior of tumor cells. Multiple coculture experiments of macrophages and osteoblasts have demonstrated that macrophage polarization significantly impacts osteogenesis. Therefore, exploring bone biomaterials that can modulate macrophage polarization holds great clinical significance. In this study, heparin was modified with maleimide and was used as a raw material to form a hydrogel with 4-am-PEG-SH. The compound was used to polarize macrophages and promote osteogenesis after combining with interleukin 4 (IL-4) by taking advantage of the electronegativity of heparin. The results revealed overexpressed M2 macrophage-related phenotypic genes and cocultivation with MC3T3-E1 cells demonstrated the osteogenesis-promoting effect of the loaded IL-4 heparin hydrogel. Previous research reported that hydrogel loaded with IL-4 can be used as a biomaterial for osteogenesis promotion. Heparin materials used in this paper are derived from clinically anticoagulant drugs and feature a simple operation. The synthesized hydrogel effectively binds cytokines, regulates macrophages to induce osteogenesis and has many potential clinical applications.

Apelin-13 treats postmenopausal osteoporosis by reducing mitochondrial oxidative phosphorylation and modulating the OPG/RANK/RANKL/IGF-1 pathway

XiaoRong Meng,Dandan Meng,Chenhui Xiang,Lingling Pu,Xiaofei Yin,Jing Yang

Objective Postmenopausal osteoporosis (PMOP) is a prevalent metabolic bone disorder characterized by decreased bone mineral density and an increased risk of fractures. This study aims to investigate the molecular mechanisms underlying the therapeutic effects of Apelin-13 in PMOP. Methods A PMOP model was established using mouse embryonic pre-osteoblast MC3T3-E1 cells treated with dexamethasone (DEX). A variety of experimental techniques—including MTT assays, Western blotting, flow cytometry, immunofluorescence staining, mitochondrial membrane potential assays, and ELISA—were employed to elucidate the potential mechanisms through which Apelin-13 exerts its therapeutic effects. Results Apelin-13 significantly enhanced osteoblast activity and effectively reversed the inhibitory effects of DEX on cell proliferation. It suppressed apoptosis by modulating the expression of apoptosis-related proteins. Furthermore, Apelin-13 promoted osteogenic differentiation, upregulated osteogenic protein expression, increased alkaline phosphatase (ALP) activity, and facilitated calcium nodule formation. It also improved mitochondrial function by reducing levels of reactive oxygen species (ROS) and malondialdehyde (MDA), boosting antioxidant enzyme activity, and activating the Nuclear factor erythroid 2-related factor 2 (NRF2) signaling pathway. Critically, Apelin-13 was found to regulate the OPG/RANK/RANKL/IGF-1 signaling axis, and its pro-osteogenic effects were significantly enhanced when used in combination with SPA0355. Conclusion Apelin-13 exhibits promising therapeutic potential for PMOP in in vitro models by modulating key processes related to bone metabolism. It achieves this by attenuating mitochondrial oxidative phosphorylation and regulating the OPG/RANK/RANKL/IGF-1 pathway, thereby laying a foundation for future in vivo studies and clinical applications.

In situ polymerization of poly(γ-benzyl-L-glutamate) on mesoporous hydroxyapatite with high graft amounts for the direct fabrication of biodegradable cell microcarriers and their osteogenic induction

Linlong Li,Xincui Shi,Zongliang Wang,Yu Wang,Zixue Jiao,Peibiao Zhang

Large-scale cell culture for cell expansion in tissue engineering is currently a major focus of research. One method to achieve better cell amplification is to utilize microcarriers. In this study, different amounts of poly(γ-benzyl-L-glutamate) (PBLG) (from 11 wt% to 50 wt%) were grafted on mesoporous hydroxyapatite (MHA) by the in situ ring opening polymerization of γ-benzyl-L-glutamate N-carboxyanhydride (BLG-NCA), and biodegradable and biocompatible PBLG-g-MHA microcarriers were directly fabricated using the oil-in-water (O/W) solvent-evaporation technique for bone tissue engineering. The amount of grafted PBLG could be controlled by adjusting the feed ratio of MHA and BLG-NCA. The relationships between sphere morphology and graft amount or solution concentration were explored. Furthermore, cytological assays were performed to evaluate the biological properties of the PBLG-g-MHA microcarriers. For a solution concentration of 3% (w/v) and PBLG graft amounts of 33 wt% and 50 wt%, the microspheres could be harvested with optimal spherical shapes. In vitro cell culture revealed that the PBLG-g-MHA microspheres had favorable properties for cell proliferation and significantly enhanced the osteogenic differentiation of MC3T3-E1 cells and bone matrix formation.

CircZfp644-205 inhibits osteoblast differentiation and induces apoptosis of pre-osteoblasts via sponging miR-455-3p and promoting SMAD2 expression

Zhang Peng,Liu Jie,Chai Zijia,Fu Jinjin,Li Shuwen,Yang Zhe

Background Circular RNAs (circRNAs) are involved in the progression of osteoporosis; however, their impact on osteogenic differentiation has yet to be fully elucidated. In this study, we identified a novel circRNA known as circZfp644-205 and investigated its effect on osteogenic differentiation and apoptosis in osteoporosis. Methods CircZfp644-205, miR-445-3p, and SMAD2 levels were measured using quantitative real-time polymerase chain reaction (qRT-PCR). MC3T3-E1 cells were subjected to microgravity (MG) to establish a cell model. Osteogenic differentiation was assessed using qRT-PCR, Alizarin Red S staining, alkaline phosphatase staining, and western blot. The apoptosis was evaluated using flow cytometry. The relationship between miR-445-3p and circZfp644-205 or SMAD2 was determined using bioinformatics, RNA pull-down, and luciferase reporter assay. Moreover, a hindlimb unloading mouse model was generated to investigate the role of circZfp644-205 in vivo using Micro-CT. Results CircZfp644-205 expression was up-regulated significantly in HG-treated MC3T3-E1 cells. Further in vitro studies confirmed that circZfp644-205 knockdown inhibited the osteogenic differentiation and induced apoptosis of pre-osteoblasts. CircZfp644-205 acted as a sponge for miR-455-3p, which reversed the effects of circZfp644-205 on pre-osteoblasts. Moreover, miR-455-3p directly targeted SMAD2, thus inhibiting the expression of SMAD2 to regulate cellular behaviors. Moreover, circZfp644-205 alleviated the progression of osteoporosis in mice. Conclusions This study provides a novel circRNA that may serve as a potential therapeutic target for osteoporosis and expands our understanding of the molecular mechanism underlying the progression of osteoporosis.

Protective effects of low-magnitude high-frequency vibration on high glucose-induced osteoblast dysfunction and bone loss in diabetic rats

Fu Zhaoyu,Huang Xu,Zhou Pengcheng,Wu Bo,Cheng Long,Wang Xinyu,Zhu Dong

Objective Low-magnitude high-frequency vibration (LMHFV) has been reported to be capable of promoting osteoblast proliferation and differentiation. Reduced osteoblast activity and impaired bone formation were related to diabetic bone loss. We investigated the potential protective effects of LMHFV on high-glucose (HG)-induced osteoblasts in this study. In addition, the assessment of LMHFV treatment for bone loss attributed to diabetes was also performed in vivo. Method MC3T3-E1 cells induced by HG only or treated with LMHFV were treated in vitro. The experiments performed in this study included the detection of cell proliferation, migration and differentiation, as well as protein expression. Diabetic bone loss induced by streptozotocin (STZ) in rats was established. Combined with bone morphometric, microstructure, biomechanical properties and matrix composition tests, the potential of LMHFV in treating diabetes bone loss was explored. Results After the application of LMHFV, the inhibiting effects of HG on the proliferation, migration and differentiation of osteoblasts were alleviated. The GSK3β/β-catenin pathway was involved in the protective effect of LMHFV. Impaired microstructure and biomechanical properties attributed to diabetes were ameliorated by LMHFV treatment. The improvement of femur biomechanical properties might be associated with the alteration of the matrix composition by the LMHFV. Conclusion LMHFV exhibited a protective effect on osteoblasts against HG by regulating the proliferation, migration and differentiation of osteoblasts. The function of promoting bone formation and reinforcing bone strength made it possible for LMHFV to alleviate diabetic bone loss.
