Aptamer-based fluorometric assay for direct identification of methicillin-resistant Staphylococcus aureus from clinical samples

Jinjuan Qiao,Xiangying Meng,Yufang Sun,Qian Li,Ronglan Zhao,Yun Zhang,Jiaqi Wang,Zhengjun Yi

Accurate and rapid identification of methicillin-resistant Staphylococcus aureus (MRSA) is of important clinical significance. In this study, a novel aptamer-based fluorometric assay was developed for detection of MRSA in clinical samples by coupling with immunomagnetic separation . The S. aureus cells in clinical specimens were enriched by magnetic separation. Following lysis by staphylococcal lysin , the PBP2a proteins were released from S. aureus cells and detected by the aptamer-based fluorometric assay. Without lengthy period of bacteria cultivation in the traditional susceptibility testing, this test has an overall testing time of only 2 h with the detection limit of 2.63 × 10 3 and 1.38 × 10 3  CFU/mL in PBS and spiked nasal swab, respectively. Since it is simple, rapid and sensitive, this method could be used for the detection of MRSA in various clinical samples.

Nano-microcapsules loaded with nano-antibacterial agents for long-term inhibition of drug-resistant strains

He Ke,Pengjiazi Fu,Wu Yanqi,He Yanting,Hu Qing,Wu Jia,Cui Guangxun,Zhang Jun,Jin Jun

There are growing concerns and worries about the safety of public facilities and equipment. Therefore, coating products with antibacterial properties have become the key to solve this problem, and there is an urgent need to develop new antibacterial architectural coatings. In this study, polystyrene nano-microspheres (PS NMS) were prepared from styrene as raw material. PS nano-microcapsules (PS NMC) were prepared by solute co-diffusion method (SCM) using PS NMS as a template, which were modified to obtain PS-OH NMC. CuS nanoparticles (CuS NPs) with antibacterial activity were further loaded to prepare CuS@PS-OH NMC with long-term antibacterial activity. The SEM results showed that the particle size of CuS@PS-OH NMC was about 410 nm, which was larger than that of PS-OH NMC. The characterization of UV and IR spectra confirmed that CuS NPs were not only encapsulated into PS NMC in large quantities, but also adhered to its surface in a small amount. CuS@PS-OH NMC has good water solubility and can slowly release Cu2+, showing good long-term antibacterial properties against Methicillin-resistant Staphylococcus aureus (MRSA). The CuS@PS-OH NMC developed in this study has excellent performance, good antibacterial effect, environmental friendliness, low price, etc. More importantly, it can efficiently and long-term inhibit MRSA, and can be used as a potential antibacterial architectural coating for special scenarios such as hospitals, schools and densely populated places, which has excellent social and economic value.

Exercise-driven gut microbiota alterations enhance colonization resistance against methicillin-resistant Staphylococcus aureus

Tingting Xu,Dan Fang,Tianqi Xu,Xiuying Tao,Zhiqiang Wang,Yuan Liu

Gut microbiota plays a crucial role in resisting the invasion of pathogens, particularly multidrug-resistant (MDR) bacteria, which pose a significant threat to public health. While exercise offers numerous health benefits, its impact on host colonization resistance remains largely unclear. In this study, we demonstrate that moderate exercise significantly reduces gut colonization by methicillin-resistant Staphylococcus aureus (MRSA), a clinically important MDR pathogen. Moreover, we identify an understudied strain of the intestinal probiotic Dubosiella newyorkensis (L8) as a critical factor in mediating exercise-induced colonization resistance against MRSA. Mechanistically, L8 enhances the deprivation of fucose, a crucial carbon source essential for MRSA growth and pathogenicity. This process relies on the high binding affinity of pyruvate to the ILE257 site of the lactate dehydrogenase in L8. Overall, our work highlights the importance of moderate exercise in maintaining host colonization resistance and demonstrates L8 as a probiotic in protecting against MRSA colonization.

Graphene oxide-enhanced colorimetric detection of Mec A gene based on toehold-mediated strand displacement

Zongkang Guo,Xueting Fan,Xiaoli Wang,Zhemin Zhou,Yuting Zhang,Nandi Zhou

Mec A, as a representative gene mediating resistance to β‐lactam antibiotics in methicillin-resistant Staphylococcus aureus (MRSA), allows a new genetic analysis for the detection of MRSA. Here, a sensitive, prompt, and visual colorimetry is reported to detect the Mec A gene based on toehold‐mediated strand displacement (TMSD) and the enrichment effect of graphene oxide (GO). The Mec A triggers to generate the profuse amount of signal units of single-stranded DNA (SG) composed of a long single‐stranded base tail and a base head: the tail can be adsorbed and enriched on the surface of GO; the head can form a G quadruplex structure to exert catalytic function towards 2,2′‐azino‐bis (3‐ethylbenzthiazoline‐6‐sulphonic acid). Therefore, through the enrichment effect of GO, the signal units SG reflects different degrees of signal amplification on different substrates (such as aqueous solution or filter membrane). This strategy demonstrates a broad linear working range from 100 pM to 1.5 nM (solution) and 1 pM to 1 nM (filter membrane), with a low detection limit of 39.53 pM (solution) and 333 fM (filter membrane). Analytical performance in real samples suggests that this developed colorimetry is endowed with immense potential for clinical detection applications.

Multifunctional self-pumping microneedle dressing with pH monitoring for MRSA-infected diabetic wounds

Lubin Zhou,XinXing Sun,Shishi Xu,Qian Chen,Yang Chen,Hongchun Gu,Biaotong Huang

MSAMD is composed of a composite of nanofibers and microneedles. • MSAMD integrates multiple functions into a single platform. • MSAMD exhibits antibacterial activity against E. coli , S. aureus , and MRSA. • MSAMD facilitates the unidirectional drainage of inflammatory exudates. • MSAMD monitors wound pH to assess the healing status of infected wounds. Infected diabetic wounds face multiple challenges, including bacterial infection, uncontrolled inflammation, and impaired angiogenesis, leading to prolonged wound healing. However, traditional dressings for infected diabetic wounds lack effective antibacterial properties, cannot manage inflammatory exudate, or cannot monitor healing in real time. Hence, we propose a pH-monitoring self-pumping antibacterial microneedle dressing that simultaneously provides antibacterial activity, inflammatory exudate management, and wound status monitoring. The dressing consists of a pH-monitoring absorbent layer, an adhesive drainage layer, and an antimicrobial microneedle layer. The first two layers form a pH-monitoring self-pumping backing layer. The microneedle layer of this dressing punctures the MRSA biofilm in diabetic wounds and eradicates MRSA. The dissolved microneedle tips drain inflammatory exudate into the pH-monitoring self-pumping backing layer, where unidirectional drainage enables exudate removal and pH colorimetric monitoring to assess wound healing status. In MRSA-infected diabetic wound healing experiments and immunofluorescence analyses, this dressing demonstrates excellent antibacterial activity, inflammatory regulation, and vascular regeneration. This multifunctional integrated dressing offers a novel treatment paradigm for infected diabetic wounds.

An antioxidant and injectable hydrogel dressing for repairing MRSA biofilm–infected diabetic wounds

Bei Zhongwu,Tong Qi,Yang Yun,Ye Lin,Liang Shiyu,Li Jianan,Liu Jie,Liang Xuyue,Chen Wen,Liu Wenwen,Yu Xiaorui,Chu Bingyang,Shi Bi,Qian Zhiyong

Chronic non-healing wounds represent a severe complication of diabetes mellitus, which frequently progress to infection, limb amputation, and even mortality. The dysregulated wound microenvironment, marked by persistent inflammation and oxidative stress, severely impedes tissue repair, and the presence of MRSA biofilm infection further worsens these impairments and poses major clinical challenges. To address these challenges, we constructed a multifunctional injectable hydrogel (SOT) that integrates antibiofilm, antioxidant, and immunomodulatory properties. This hydrogel is formed through dynamic covalent crosslinking between thiolated hyaluronic acid (HA-SH) and dopamine-modified oxidized dextran (ODex-DA), which enables favorable injectability, self-healing, and in situ gelation. Tannic acid–silver nanoparticles (TA-Ag NPs) incorporated into the system impart antibiofilm and reactive oxygen species (ROS)-scavenging properties. In a diabetic MRSA biofilm infection model, the SOT hydrogel eradicated biofilms, reduced excessive ROS, and promoted wound closure. These findings suggest that this immuno-instructive hydrogel platform may offer a promising therapeutic approach for the treatment of MRSA biofilm-infected chronic diabetic wounds. Graphical

Versatile Chlorin e6-based magnetic polydopamine nanoparticles for effectively capturing and killing MRSA

Chunbo Lu,Feifei Sun,Yinyin Liu,Yao Xiao,Yuanhao Qiu,Haibo Mu,Jinyou Duan

Bacterial infections are a growing global challenge for public health as antibiotic resistance could cause the failure of anti-infective treatment eventually. So, it is urgent to develop new potential antibacterial materials. Herein, a multifunctional chitosan (CS) functionalized magnetic Chlorin e6 (CS-MP-Ce6) was constructed to combat methicillin-resistant Staphylococcus aureus (MRSA) infection by integrating bacterial conjugation and enrichment, and near-infrared (NIR)-triggered photodynamic sterilization. CS-MP-Ce6 could efficiently capture bacteria due to positively charged property of CS, and Ce6 acted as an effective photodynamic killer to convert NIR light into local energy to enhance antibacterial activity. Specifically, after being trapped together with MRSA, CS-MP-Ce6 showed an excellent in vitro photodynamic sterilization ability. In vivo MRSA-induced abscess treatment studies showed faster healing when CS-MP-Ce6 was used as subcutaneous nano-localized energy sources with the assistance of external magnet to concentrate CS-MP-Ce6–bacteria conjugate. This work provides a promising framework for constructing a new system for efficiently combating MRSA.

Modulation of MRSA virulence gene expression by the wall teichoic acid enzyme TarO

Lu Yunfu,Chen Feifei,Zhao Qingmin,Cao Qiao,Chen Rongrong,Pan Huiwen,Wang Yanhui,Huang Haixin,Huang Ruimin,Liu Qian,Li Min,Bae Taeok,Liang Haihua,Lan Lefu

Phenol-soluble modulins (PSMs) and Staphylococcal protein A (SpA) are key virulence determinants for community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA), an important human pathogen that causes a wide range of diseases. Here, using chemical and genetic approaches, we show that inhibition of TarO, the first enzyme in the wall teichoic acid (WTA) biosynthetic pathway, decreases the expression of genes encoding PSMs and SpA in the prototypical CA-MRSA strain USA300 LAC. Mechanistically, these effects are linked to the activation of VraRS two-component system that directly represses the expression of accessory gene regulator ( agr ) locus and spa . The activation of VraRS was due in part to the loss of the functional integrity of penicillin-binding protein 2 (PBP2) in a PBP2a-dependent manner. TarO inhibition can also activate VraRS in a manner independent of PBP2a. We provide multiple lines of evidence that accumulation of lipid-linked peptidoglycan precursors is a trigger for the activation of VraRS. In sum, our results reveal that WTA biosynthesis plays an important role in the regulation of virulence gene expression in CA-MRSA, underlining TarO as an attractive target for anti-virulence therapy. Our data also suggest that acquisition of PBP2a-encoding mecA gene can impart an additional regulatory layer for the modulation of key signaling pathways in S. aureus .

Synthesis and Biological Evaluation of Novel Thioether Pleuromutilin Derivatives

Shuhua Mu,Huixian Liu,Lifang Zhang,Xiaoyang Wang,Feiqun Xue,Yue Zhang

To develop new pleuromutilin derivatives as veterinary antibiotic medicines, we designed and synthesized a series of new thioether pleuromutilin derivatives possessing acylthiazolyl moiety based on previously designed derivatives. The antibacterial properties of the prepared pleuromutilin derivatives were assessed in vitro by the broth dilution method against five kinds of bacteria and the mycoplasma Mycoplasma gallisepticum (MG). All of the tested compounds displayed moderate to good antibacterial activity to methicillin-sensitive Staphylococcus aureus (MSSA), methicillin-sensitive Staphylococcus epidermidis (MSSE), methicillin-resistant S. aureus (MRSA), Streptococcus agalactiae (S. aga) and MG. However, the activity to Pyogeniccoccus (Pyogens) was generally poor. Compounds 13i and l showed potent antibacterial activity against MSSE and MRSA which are better than that of valnemulin. The structural modification for pleuromutilin affected the antibacterial activity. Amino substituents in the benzene ring can effectively improve activity. Compared with the analogue 13a that possesses unsubstitution benzoyl group, the nitro, methoxy, hydroxy and dichloro substituent contributed little to antibacterial activity. Increasing a methylene between benzene moiety and carbonyl group decreased the bioactivity of derivative. The analogues that obtained by the reaction of amino acids and intermediate 9 showed moderate activity.

Ratiometric fluorescent nanoprobe for imaging and screening of hydrogen sulfide related bacterial resistance

Deyan Gong,Xiang Huang,Zineng Yi,Qianpeng Zhou,Xiaona Xu,Xiaohan Meng,Congyu Liu,Zhaohua Miao,Zhengbao Zha

As a metabolite or reductive cytoprotective gas of bacteria to mediate and mitigate the oxidative stress imposed by antibiotics, hydrogen sulfide (H 2 S) related drug-resistant mechanisms represent a growing point in bacteria drug-resistant research. Thereby, novel diagnostic strategies exploration for H 2 S related drug-resistant pathways recognition are important for drug-resistant bacteria monitoring. Herein, a new organic fluorescent dye ( E )− 3-(3,3-dimethyl-2-styryl-3 H -benzo[ f ]indol-1-ium-1-yl) propane-1-sulfonate self-assembled nanoprobe (DPS NP, ~200 nm) was designed for ratiometric fluorescent monitoring the change of H 2 S concentration. DPS NP exhibited ratiometric fluorescence alteration from yellow (λ em = 553 nm) to blue (λ em = 464 nm) due to nucleophilic addition reaction with H 2 S within five minutes. DPS NP exhibits remarkable capabilities with selectively sensitive colorimetric and ratiometric fluorescent alteration toward H 2 S over other analytes. Fluorescence microscopy imaging or ratiometric fluorescent visualization for monitoring the concentration of bacterial released H 2 S show DPS NP can act as particular diagnostic technique for screening of β -lactam antibiotic resistance in Methicillin-resistant Staphylococcus aureus (MRSA) and nonresistant bacteria Staphylococcus aureus ( S. aureus ) .

Callistemon citrinus bioactive metabolites as new inhibitors of methicillin-resistant Staphylococcus aureus biofilm formation

Amr M. Shehabeldine,Rehab M. Ashour,Mona M. Okba,Fatema R. Saber

Ethnopharmacological relevance The development of new inhibitors of bacterial virulence factors from natural origin has recently received significant attention. Callistemon citrinus Skeels is an important plant of great medicinal value. Its antimicrobial activity is well documented. Although several compounds were isolated from this plant, the actual bioactive compounds responsible for its antimicrobial activity are still unrevealed. Aim of the study To evaluate the effect of C. citrinus crude extract and isolated compounds on methicillin-resistant and sensitive Staphylococcus aureus. Materials and methods The methylene chloride-methanol extract (MME) of C. citrinus leaves was prepared by Soxhlet apparatus. Biologically guided fractionation of MME was accomplished using several normal and reversed phase silica gel columns. The potency of MME and its isolated compounds against methicillin-resistant Staphylococcus aureus (MRSA) and methicillin-sensitive S. aureus (MSSA) was evaluated. In addition, the mechanism of resistance was studied using three virulence factors; antibiofilm activity, inhibition of staphyloxanthin biosynthesis and effect on acid tolerance. Ultrastructural changes in MRSA and MSSA were observed by TEM to understand mode of action of these compounds. Results Pulverulentone A (C1), 8- desmethyl eucalyptin (C2) and eucalyptin (C3) were isolated from the most bioactive fraction of MME. Confocal scanning laser microscopy images revealed that C. citrinus isolated compounds destroyed the intact architecture of biofilm, thickness and reduced its biomass. Pulverulentone A (C1) showed the most potent anti-biofilm activity up to 71% and 62.3% against MRSA and MSSA, respectively. It also exhibited the highest inhibition of staphyloxanthin biosynthesis of MRSA and MSSA by 55.6% and 54.5%, respectively. The bacterial cell membrane was compromised, losing its integrity and releasing important cellular constituents when exposed to C1-C3 Conclusions C. citrinus phenolics and acylphloroglucinols may serve as potential source of plant-based antibacterials and thus could be implicated to control MRSA biofilm formation.

Antimicrobial and antitumor activity of peptidomimetics synthesized from amino acids

Huan Li,Shuang Fu,Yudan Wang,Xuan Yuan,Lijia Liu,Hongxing Dong,Qiang Wang,Zhijia Zhang

Thirteen cationic peptidomimetics derived from amino acids bearing an alkyl or ethynylphenyl moiety that mimic the structure of cationic antibacterial peptides were designed and synthesized using a simple coupling reaction of an amino acid with a substituted amine. Antibacterial activities of the resulting peptidomimetics against drug-sensitive bacteria, such as Gram-positive Staphylococcus aureus ( S. aureus ) and Bacillus subtilis , Gram-negative Escherichia coli ( E. coli ) and Salmonella enterica , and a drug-resistant bacterium, methicillin-resistant S. aureus (MRSA), were systematically evaluated. Most peptidomimetics show significant broad-spectrum antibacterial activity. A-L-Iso-C 12 (isoleucine derivative bearing a dodecyl moiety) show MICs of 2.5 μg/mL against S. aureus and 4 μg/mL against MRSA and A-L-Val-C 12 (valine derivative bearing a dodecyl moiety) show MICs of 1.67 μg/mL against E. coli and 8.3 μg/mL against MRSA. A-L-Val-C 12 showed low cytotoxicity toward L929 cells in comparison with SGC 7901 cells, indicating tumor-directed killing by peptidomimetics while avoiding toxicity to normal cells. The influences of type of amino acid and substituent, length of substituent, and stereochemistry of amino acids on antibacterial activity and cytotoxicity of peptidomimetics were systematically investigated. The results indicate that this series of cationic peptidomimetics derived from amino acids display antitumor activity and may be useful for treatment of bacterial infections.
