Bioreductive co-metabolic degradation of azo dyes represents a promising green technology for addressing the environmental pollution caused by azo dyes. This study investigated the impact of co-metabolized substances on NADH production in microbial systems, focusing on the production of reducing power, electron transfer, and the synergistic effects of microbial communities and associated mechanisms during azo dye degradation. A culture system was developed to maximize NADH production at 3 g/L yeast extract, and it was observed that the system exhibited a significant increase in reducing power, with NADH concentration reaching 909.48 pg/mL (60 h). The electron transfer process in this system primarily depended on factors such as redox mediators, azoreductase, and formic acid. Azo dye reductive degradation and decolorization occurred through an indirect electron transfer pathway. Flavin-based redox mediators (riboflavin and flavin mononucleotide) played a key role in the system, with the application of riboflavin and flavin mononucleotide increasing the system's dye reduction ratio by 14.45% and 14.40%, respectively. They were endogenously expressed by the system and facilitated efficient electron transfer by synergizing with specific reductases, particularly when the electron transport chain was inhibited, and alternative pathways ensured the continuation of the reduction reaction. NADH production primarily occurred through glycolysis, the TCA cycle, and fatty acid β-oxidation, with glycolysis contributing the most. Microorganisms such as Enterococcus , Burkholderia , and Escherichia within the microbial community played a crucial role in NADH production while regulating community behavior through a quorum sensing system, thereby enhancing the stability and efficiency of dye degradation. This study investigated the bioreduction of azo dyes in terms of reducing power, offering a theoretical foundation and practical guidance for optimizing the microbial system and enhancing the biodegradation efficiency of azo dyes.

Spatiotemporal monitoring of reduced nicotinamide adenine dinucleotide (NADH) is critical for understanding cellular bioenergetics and redox homeostasis. However, conventional electrochemical methods are hindered by electrode fouling, interference, and a lack of spatial resolution required for subcellular analysis. Here, we present a “needle-type”, nanosized field-effect transistor (nano-FET) probe constructed on dual-barrel carbon nanoelectrodes for ultrasensitive NADH detection. The device features a nanoscopic polypyrrole (PPy) channel, modified with 1-methoxy-5-methylphenazinium methyl sulfate (mPMS) as a specific redox mediator. We establish a sensing mechanism based on coupled electron and ion transport, where mPMS-mediated NADH oxidation triggers ion dedoping in PPy channel to achieve intrinsic signal amplification. The nano-FET probe exhibits an exceptionally low NADH detection limit of 0.1 pM and a broad dynamic range (1 pM to 100 μM), accompanied by robust operational stability. This work significantly expands the toolkit of nanopipette-based electrochemical sensors, offering a powerful platform for spatially resolved metabolic profiling at the single-cell level.

As a state-of-the-art conjugated polymer photocatalyst, graphitic carbon nitride(abbreviated as g-C 3 N 4 ) has shown great potential in photocatalytic cofactor(reduced form of nicotinamide adenine dinucleotide, NADH) regeneration. Herein, Fe-doped g-C 3 N 4 was engineered for photocatalytic NADH oxidation. The π-π interaction between the NADH molecule and the conjugated heptazine building block facilitates the adsorption of NADH onto the framework, as revealed by density functional theory(DFT) calculations. Furthermore, iron doping promoted the oxidation kinetics of NADH under blue LED illumination. The conversion ratio of NADH to its oxidized form could be up to 85.7% in 20 min, comparing with 59.4% for metal-free counterpart. Enzyme assay employing formate dehydrogenase(FDH) further verified the selectivity of the products, with 67.5%±2.6% of enzymatically active 1,4-NADH being regenerated following the oxidation process. Scavenger experiments suggest the dominant role of photo-induced electrons in the oxidation of NADH. This work could shed light on developing a novel cofactor regeneration route through the synergistic effect between the metal doping and noncovalent interaction based on the conjugated polymer.

Background Conventional biochemical methods for detecting nicotinamide adenine dinucleotide (NADH) struggle with issues such as low sensitivity, limited cellular permeability, and challenges in longitudinal studies within intact organs or deep tissue signal detection. Upconversion nanoparticle-based fluorescence sensing technologies emerge as a promising alternative, offering benefits like low cytotoxicity and high sensitivity. This study introduces a novel ratiometric approach to overcome these limitations and improve NADH detection in cellular studies. Results We have developed highly sensitive upconversion nanoparticle-based probes that enhance NADH detection. These probes consist of core-shell upconversion nanoparticles (UCNPs) surface-modified with mesoporous silica, which immobilizes a NADH-specific recognition molecule, Probe A. This setup facilitates a spironolactone ring-opening reaction, enhancing fluorescence resonance energy transfer (FRET) between Probe A and NADH. Our approach demonstrates a linear response over a 0–70 nM concentration range with a low detection limit of 0.053 nM. Application in PC12 and ρ0 cells confirms the method's efficacy and potential for broader biomedical research applications. Significance This innovative ratiometric sensing approach significantly enhances the accuracy and sensitivity of NADH detection. By providing a robust and precise measurement tool, it offers a substantial improvement over traditional biochemical methods, facilitating deeper insights into cellular metabolism and pathophysiological processes. Its adaptability and effectiveness herald potential transformative applications in biomedical research, particularly in studies involving metabolic and degenerative diseases.

The synthesis of chiral intermediates for the traditional antidepressant duloxetine has gained significant attention as the number of depressed patients continues to grow. S - N , N -Dimethyl-3-hydroxy-3-(2-thienyl)-1-propanamide ( S -DHTP) is a critical intermediate in the synthesis of duloxetine, and the chemical synthesis process is complex and environmentally unfriendly. Reduced nicotinamide adenine dinucleotide phosphate (NADPH) is a major cost driver in the biocatalytic production of S -DHTP from N , N -Dimethyl-3-keto-3-(2-thienyl)-1-propanamide (DKTP). Here, we successfully modified the coenzyme preference of an aldo-keto reductase (AKR7-2-1) to use the cheaper reduced nicotinamide adenine dinucleotide (NADH) through a coenzyme preference modification approach. We utilized protein engineering to create a superior mutant, Y53F, which increased the coenzyme specificity of AKR7-2-1 by 875-fold and improved its thermal stability , enhancing its potential for industrial applications. Molecular dynamics simulations were performed to demonstrate the effect of mutations at key sites on the protein, revealing the altered coenzyme preference and increased thermal stability from structural and energetic changes. This study validates the viability of the coenzyme preference modification strategy for aldo-keto reductase, offering valuable insights for fellow researchers and guiding future investigations.

Three-dimensional TiO 2 hierarchical nanowire arrays were synthesized featuring nanocavities via a hydrogen peroxide–assisted wet chemical reaction, followed by a simple liquid-phase deposition process. We subsequently decorated the TiO 2 arrays with dopamine, leveraging its enediol ligand for chelation. Reduced nicotinamide adenine dinucleotide (NADH), a cofactor for over 300 dehydrogenases, serves as the targeted analyte to assess the PEC sensing performance of the sensor. The developed sensor exhibited a linear concentration range for NADH detection of 0.05 to 50 µM, achieving a high sensitivity of 0.29 μA∙μM −1 ∙cm −2 and a low detection limit of 0.03 μM (signal-to-noise ratio of 3). Notably, PEC performance of the resulting sensor surpassed that of commercial TiO 2 (P25)-based sensors. The enhanced sensing capabilities can be attributed to several factors: the TiO 2 hierarchical nanowire arrays with nanocavities provide a larger surface area, abundant active sites, and shorter molecular diffusion lengths for surface adsorption and reaction, thereby reducing response times and improving sensitivity. Additionally, the coupling of TiO 2 with dopamine enhances visible light absorption and optimizes charge transfer dynamics. These findings open a new avenue for designing high-performance three-dimensional nanoarchitectures for PEC sensing applications. Graphical

The commonly believed mechanism of colistin against Gram-negative bacteria is to cause cell membrane lysis, whereas the mechanism of colistin against Gram-positive bacteria is extremely fragmented. In this study, we found that colistin treatment onBacillus subtilisWB800,Paenibacillus polymyxaC12 andPaenibacillus polymyxaATCC842 enhances not only the activities of α-ketoglutaric dehydrogenase and malate dehydrogenase in tricarboxylic acid (TCA) cycle, but also the relative expression levels of their encoding genes. Additionally, the oxaloacetate concentration also increases. Interestingly, the analysis of the relative expression of genes specific for respiratory chain showed that colistin treatment stimulates the respiratory chain in Gram-positive bacteria. Accordingly, the NAD+/NADH ratio increases and the oxidative level is then boosted up. As a result, the intensive oxidative damages are induced in Gram-positive bacteria and cells are killed. Notably, both rotenone and oligomycin, respectively, inhibiting NADH dehydrogenase and phosphorylation on respiratory chain can downgrade oxidative stress formation, thus alleviating the colistin-induced killing of Gram-positive cells. Besides, thiourea-based scavenging for reactive oxygen species also rescues the colistin-subjected cells. These data collectively demonstrate that colistin stimulates both TCA cycle and respiratory chain in Gram-positive bacteria, leading to the enhancement of NADH metabolism and resulting in the generation of oxidative damages in Gram-positive cells. Our studies provide a better understanding of antibacterial mechanism of colistin against Gram-positive bacteria, which is important for knowledge on bacterial resistance to colistin happening via the inhibition of respiratory chain and manipulation of its production.Keywords:colistin;Gram-positive bacteria;oxidative stress;TCA cycle;respiratory chain

CO 2 activation and conversion by enzymes to produce value-added multi-carbon compounds under mild conditions show significant impact on carbon neutrality. However, the enzyme co-factor NADH is expensive and the regeneration is still challenging for the enzyme reaction process. Inspired by natural photosynthesis, this study successfully developed a biomimetic photocatalyst Ce/TCPP-UiO-66-NH 2 (TCPP: meso-tetrakis (4-carboxyphenyl) porphine) for NADH regeneration and CO 2 activation. Structure analysis and spectra showed that the co-doping of Ce and TCPP reduced the band gap width of UiO-66-NH 2 , improved the separation efficiency of photo-generated carriers, and endowed it with excellent photocatalytic performance. The synthesized biomimetic photocatalyst Ce/TCPP-UiO-66-NH 2 was then applied for NADH regeneration. The regeneration efficiency of NADH can reach up to 95 % after 3 h of light reaction, which was about 1.6 times higher than that of the control group. Ultra-fast femtosecond time-resolved transient absorption (fs-TA) showed that Ce/TCPP-UiO-66-NH 2 possessed the shortest average decay time ( τ ¯ = 1.12 ps), indicating shorter average life-time of carriers after Ce doping, thus suppressing hole-electron recombination. Finally, the key enzyme formate dehydrogenase (FDHPa) for CO 2 activation was immobilized on the membrane, combining with light regeneration of NADH for CO 2 production to formate. The formate yield (3.1 mM) was 16 times higher than that of the control group (0.19 mM). This work provided an effective strategy and efficient photocatalyst for the biomimetic photocatalytic activation of CO 2 to formate, which is an indispensable bridge for the further high-valuable C n compounds production by biocatalysis.

Glycolysis, a pivotal step in yeast metabolism, plays an indispensable role as a carbohydrate utilization process crucial for cellular survival. Developing advanced technologies to elucidate this fundamental physiological process holds significant scientific implications. Electrochemiluminescence (ECL) imaging exhibits the advantage of negligible background interference and facilitates straightforward visualization, thereby conferring significant value in biomolecular observation. In this study, we present an ECL imaging method for investigating yeast metabolism by utilizing the endogenetic NADH as an efficient coreactant for ECL generation. The yeast glycolysis process drives the conversion of NAD+ to NADH, resulting in enhanced ECL response as well as the increased brightness of ECL images that can be used for quantification of yeast activity. There was a linear correlation between the reciprocal of both the gray value of ECL image and yeast concentration within the range of 6.25 × 106 – 6.25 × 108 CFU/mL. Due to the highly efficient coreactant behavior of NADH, our method demonstrated excellent selectivity with minimal interference. Furthermore, we employed this approach to investigate some toxic inhibitors on yeast metabolism, yielding reliable results. This ECL imaging method not only avoids the use of additional coreactants but also provides a sensitive and intuitive approach for monitoring yeast metabolism, demonstrating great potential in revealing various complex biological processes.

Artificial photosynthesis shows a promising potential for sustainable supply of nutritional ingredients. While most studies focus on the assembly of the light-sensitive chromophores to 1-D architectures in an artificial photosynthesis system, other supramolecular morphologies, especially bioinspired ones, which may have more efficient light-harvesting properties, have been far less studied. Here, MCpP-FF, a bioinspired building block fabricated by conjugating porphyrin and diphenylalanine, was designed to self-assemble into nanofibers-based multiporous microspheres. The highly organized aromatic moieties result in extensive excitation red-shifts and notable electron transfer, thus leading to a remarkable attenuated fluorescence decay and broad-spectrum light sensitivity of the microspheres. Moreover, the enhanced photoelectron production and transfer capability of the microspheres are demonstrated, making them ideal candidates for sunlight-sensitive antennas in artificial photosynthesis. These properties induce a high turnover frequency of NADH, which can be used to produce bioproducts in biocatalytic reactions. In addition, the direct electron transfer makes external mediators unnecessary, and the insolubility of the microspheres in water allows their easy retrieval for sustainable applications. Our findings demonstrate an alternative to design new platforms for artificial photosynthesis, as well as a new type of bioinspired, supramolecular multiporous materials.

NADH, composed of metal binding, charged and redox groups, acts as a “three-in-one” template to construct peroxidase mimicking Pd nanoclusters (NCs). The physicochemical properties of Pd NCs are highly dependent upon the molar ratio of [NADH]/[Na 2 PdCl 4 ], thereby contributing to distinct peroxidase mimicking activities. The ultrasmall Pd nanozyme that contains 30% metallic Pd 0 species, exhibits the K m of 0.063 mM toward TMB and 80.8 mM toward H 2 O 2 . Notably, addition of heparin enables the enhancement of its peroxidase-like activity in neutral media. A colorimetric assay was well-established at pH 6 for quantitatively monitoring heparin in aqueous solution and biological fluid. The linear response lies in the range of 0.5–25 µg mL −1 , with the limit of detection of 1.1 ng mL −1 . This work paves a promising pathway to manufacture highly active enzyme mimetics with desirable physicochemical properties. Graphic Abstract

Coenzyme regeneration is important for the industrial utilization of oxidoreductase. In this work, reduced nicotinamide adenine dinucleotide (NADH) was regenerated under visible light via a photosensitizer and an electron donor. First, several photosensitizers including 5,10,15,20-tetrakis(4-carboxyphenyl)porphyrin (TCPP), zinc porphyrin (ZnTCPP) and eosin Y (EY) with varied concentrations were compared. Next, using TCPP as the photosensitizer, the type and concentration of the electron donor, pH value and concentration of the buffer system for the reaction were optimized. Following that, TCPP was immobilized onto the surface of various supports for screening. Then, the immobilization conditions were optimized and the photocatalytic performance of silica-attached TCPP was studied. Lastly, the effect of a rhodium mediator on NADH regeneration was evaluated. The results show that the optimum concentration of TCPP, ZnTCPP and EY was 2, 1 and 4 mM, respectively, and the highest NADH yield of the TCPP system was 2.37 and 1.47 times those of the ZnTCPP and EY systems. With TCPP as the photosensitizer, triethanolamine was the most efficient electron donor and its appropriate concentration was 5 mM. The optimal pH value and concentration of the phosphate buffer was 7.0 and 0.05 M. Under these conditions, a high NADH yield of 81.5% was achieved after 60 min. For the supports examined, the thiol-containing silica microspheres coated with polydopamine and polyethyleneimine had the highest loading capacity of TCPP (9.15 mg per g-support). Via sensitization, the NADH yield was 9.64% after 90 min and was upgraded to 22.5% with the addition of the rhodium mediator.
