Nanozyme-catalyzed reactive oxygen species (ROS) generation
has
emerged as a promising strategy for advanced ROS-based therapeutics.
However, achieving efficient ROS production with favorable catalytic
kinetics in a confined microenvironment remains challenging. Herein,
a cancer-targeting heterogeneous Janus nanozyme (JN, Cu 2– x Se–Au-TLS11a) was rationally fabricated via
a facile modular synthesis and assembly. Upon near-infrared II (NIR-II)
light irradiation, the well-designed JN realized multimodal synergistic
anticancer effects via three coordinated mechanisms: (i) efficient
photothermal conversion with a high efficiency of 61.9% for photothermal
therapy (PTT); (ii) NIR-II-triggered hot-electron transfer from the
Au domain to Cu 2– x Se promoted the
generation of Cu + , which accelerated Fenton-like reactions
to produce hydroxyl radicals (•OH); and (iii) NIR-II photoexcitation
induced the formation of electron–hole (e – -h + ) pairs on the Janus nanostructure, enabling additional
photocatalytic •OH generation. Under NIR-II light activation,
the JNs, integrating photothermal, photoenhanced chemodynamic, and
photocatalytic effects, demonstrated highly specific and potent in
vitro anticancer efficacy, achieving an IC 50 value of approximately
12.5 μg mL –1 against HepG2 cells and reducing
cell viability to 13.8%, thereby providing an innovative paradigm
for designing spatiotemporally controlled, ROS-amplifying nanoplatforms
toward precision cancer therapy.

Occlusive thrombi within blood vessels are a primary cause of life-threatening cardiovascular diseases, which continue to pose significant global health challenges. Here, we report a polymer nanoplatform that integrates near-infrared II (NIR-II) light-triggered photothermal thrombolysis with intrinsic antithrombotic functionality. A novel semiconducting polymer, featuring intense NIR-II absorption and exceptionally high photothermal conversion, is rationally engineered to enable rapid and localized light-to-heat thrombolysis. A thermosensitive hydrogen sulfide (H 2 S) donor is also incorporated, generating multifunctional “nanosweepers” that combine synergistic photothermal and gas-mediated thrombolytic activity. The formulation is further functionalized with bis-serotonin groups, which confer myeloperoxidase- and hydrogen peroxide-responsive thrombus-targeting capability, ensuring selective accumulation and site-specific therapy. This agent possesses robust NIR-II absorption, high photothermal conversion efficiency (77.2%), and pronounced thrombus-specific enrichment. Importantly, the combination of localized photothermal heating and on-demand H 2 S release enable rapid, near-complete thrombolysis in both carotid and lower-limb arterial thrombosis models, achieving almost full restoration of blood flow. This work establishes a versatile and highly efficient strategy that integrates precise NIR-II photothermal conversion with H 2 S-mediated redox regulation, providing a promising platform for site-specific multimodal synergistic treatment of life-threatening thrombotic disorders. The findings offer new insights into the design of multifunctional, precision therapeutics for vascular disease management.

Rare-earth doped oxysulfides have gained considerable attention as a luminescence host material owing to its good chemical stability and high efficiency. In this study, Er3+-sensitized La2O2S nanoparticles are prepared by the high-temperature co-precipitation method. La2O2S:2%Er3+ is selected to construct La2O2S:Er3+@La2O2S core/shell structures through epitaxial growth, and the luminescence properties are studied. The surface defects of La2O2S:Er3+ are repaired by the inert protective shell, and the luminescence intensity of the core/shell nanocrystals is increased by 9.6 times. Finally, MAPbI3 is spin-coated over the La2O2S:Er3+@La2O2S film to construct narrow band near-infrared photodetectors. Under 1550 nm excitation, the response rate is 23 mA W−1, detection rate is 2.57 × 108 Jones, and response time is 220 ms.

Healing wounds presents a significant challenge due to bacterial biofilm infections and the inherent drug resistance of these biofilms. This report introduces a multifunctional nanoplatform (NPs) designed to combat wound biofilm infections using NIR-II photothermal therapy. The NPs are self-assembled from amphiphilic polymers (AP) to encapsulate photothermal polymers (PT) through classic electrostatic interactions. Importantly, these NPs are electrically neutral, which enhances their ability to penetrate biofilms effectively. Once inside the biofilm, the NPs achieve complete thermal ablation of the biofilm under NIR-II laser irradiation. Additionally, when exposed to laser and the GSH microenvironment, the NPs exhibit strong photothermal effects and self-degradation capabilities. In vitro tests confirm that the NPs have excellent antibacterial and anti-biofilm properties against methicillin-resistant Staphylococcus aureus (MRSA). In vivo studies demonstrate that the NPs can efficiently clear wound biofilm infections and promote wound healing. Notably, the NPs show superior photothermal effects under NIR-II laser irradiation compared to NIR-I lasers. In summary, the developed NPs serve as an integrated diagnostic and therapeutic nano-antimicrobial agent, offering promising applications for biofilm wound infections and wound healing.

The current strategy of co-delivering copper ions and disulfiram (DSF) to generate cytotoxic CuET faces limitations in achieving rapid and substantial CuET production, specifically in tumor lesions. To overcome this challenge, we introduce a novel burst-release cascade reactor composed of phase change materials (PCMs) encapsulating ultrasmall Cu 2-x Se nanoparticles (NPs) and DSF (DSF/Cu 2-x Se@PCM). Once triggered by second near-infrared (NIR-II) light irradiation, the reactor swiftly releases Cu 2-x Se NPs and DSF, enabling catalytic reactions that lead to the rapid and massive production of Cu 2-x Se-ET complexes, thereby achieving in situ chemotherapy. The mechanism of the burst reaction is due to the unique properties of ultrasmall Cu 2-x Se NPs, including their small size, multiple defects, and high surface activity. These characteristics allow DSF to be directly reduced and chelated on the surface defect sites of Cu 2-x Se, forming Cu 2-x Se-ET complexes without the need for copper ion release. Additionally, Cu 2-x Se-ET has demonstrated a similar (to CuET) anti-tumor activity through increased autophagy, but with even greater potency due to its unique two-dimensional-like structure. The light-triggered cascade of interlocking reactions, coupled with in situ explosive generation of tumor-suppressive substances mediated by the size and valence of Cu 2-x Se, presents a promising approach for the development of innovative nanoplatforms in the field of precise tumor chemotherapy.

Photothermal therapy has a remarkable effect on the destruction of tumors. It kills tumor cells by photothermal ablation and induces immunogenic cell death by activating the immune response in tumor tissues. However, inhibition of the tumor immune microenvironment suppresses PTT-induced body-specific anti-tumor immunity. In this study, we designed the GdOF@PDA-HA-R837-hydrogel complex to achieve NIR-II imaging-guided photothermal ablation and enhanced immune response. Due to the doping of Yb and Er elements and the presence of a polydopamine coating, the synthesized nanoparticles enable NIR-II and photoacoustic imaging of tumor tissues, which will help in the integration of multimodal tumor imaging for diagnosis and treatment. Polydopamine is used as a photothermal agent and drug carrier because of its excellent photothermal ability and high drug loading capacity under 808 nm near infrared light. Hyaluronic acid can bind to specific receptors on the surface of cancer cells, allowing nanoparticles to aggregate around the tumor, thus enhancing the targeting ability of nanoparticles. In addition, imiquimod (R837) has been used as an immune response modulator to enhance the immunotherapeutic effect. The presence of a hydrogel enhanced the retention effect of nanoparticles in the tumor. We demonstrate that the combination of photothermal therapy with immune adjuvants effectively induces ICD, which in turn stimulates the activation of specific anti-tumor immunity and enhances the effect of photothermal therapy in vivo.

Accurate detection and localization of micrometastases are extremely important for tumor diagnosis and comprehensive treatment. However, it is clinically difficult to finely localize micrometastases, especially focal lesions smaller than 1 mm. To that purpose, this work designed a tumor microenvironment factor (glutathione, GSH)-activated second near-infrared transparency window (NIR-II) fluorescent probe for visualization of micrometastases. It is constructed by complexing copper ions with 3-mercaptopropionic acid-modified silver sulfide (Ag 2 S-MPA) quantum dots . The experimental results showed that the probe has good and stable GSH activation properties and can achieve tumor-specific NIR-II fluorescence imaging. Further, in vivo imaging experiments demonstrated that the probe can precisely localize melanomas with a diameter of < 0.1 mm in lung micrometastases. This work provides a potential strategy for the development of novel tools for localization and visualization of micrometastases.

Fallopian tube blockage-related diseases are the main cause of female infertility. Diagnosis of tubal patency and accurate determination of tubal obstruction lesions is of great value for infertility treatment. Conventionally, the common used hysterosalpingogram (HSG) has the disadvantages of radioactive hazard and high false positive rate. Thus, the development of a safe and accurate method which can replace traditional HSG for tubal patency diagnosis is urgently needed. Recently, fluorescence imaging in the second near-infrared region (NIR-II) was widely used in in-vivo imaging research due to its excellent tissue penetration and signal-to noise ratio. With the aid of fluorescence agents, NIR-II imaging has the potential to diagnose tubal patency, which, unfortunately, has not been explored. Herein, a kind of rare-earth Erbium-based nanoprobe (Er-RENPs) with outstanding NIR-II fluorescence property was synthesized. Based on the Er-RENPs, the potential of NIR-II fluorescence imaging was explored. Results show that the outline of fallopian tubes can be clearly visualized with the help of Er-RENPs. And the stenotic, occlusive fallopian tubes and hydrosalpinx can be diagnosed. Additionally, the Er-RENPs exhibit good biocompatibility. In this study, we reported that NIR-II rare-earth fluorescent nanoprobe can serve as a promising fluorescent agent, and exhibits considerable potential application for tubal diagnosis.

Lanthanide-doped near-infrared-II (NIR-II) luminescent nanoparticles have shown great potential in many fields. However, most previous studies were focused on the sensitizer-activator coupled systems in which the NIR-II luminescence intensity is seriously restricted by the low dopants concentration . Improving the NIR-II emission intensity in lanthanide-doped nanoparticles has remained a daunting challenge. Herein, we report a systematic research on the enhancement of the NIR-II luminescence of Er 3+ through lanthanide-mediated energy modulation. It is found that introducing proper content of Yb 3+ , Ce 3+ , Tm 3+ , and Ho 3+ in the erbium sublattice can indeed enhance the NIR-II emission under 980 and 808 nm excitations by mediating the energy transfer processes. Doping of other lanthanide ions (e.g., Pr 3+ , Tb 3+ , Dy 3+ , and Sm 3+ ) only led to a quick decline in luminescence intensity. The NIR-II emission was further enhanced by designing a core-shell-shell nanostructure which consists of an NaYbF 4 interlayer to maximize the absorption of 980 nm excitation. The NIR-II luminescence exhibits superior penetration depth in biological tissue, showing great promise in biological applications .

Due to the deep biological penetrability and therapeutic depth, the photothermal therapy over second near-infrared region (NIR-II) is booming against deep-seated tumors. Intensive endeavors are committed to looking for suitable photothermal agents (PTAs), but the progress seems not so satisfied toward the choice of PTA dosage. Herein, a comprehensive parameter, incident photon-to-thermal conversion coefficient (IPTCE), is used to evaluate the overall conversion of PTAs at different dosage, which will benefit for determining the optimized dosage of PTAs in pursuit of complete healing together with reduced long-term damages of nanodrugs. To prove the possibility, a series of anionic solid solution MXenes are chosen as hosts due to their versatile chemical compositions and correspondingly tunable light response. By deconvoluting fundamental structure–composition–property relationships, anionic regulation with extra electron injection leads to tunable free carrier densities and enhanced NIR-II harvesting. Ti 3 C 1.23 N 0.77 with high-level nitrogen exhibits extraordinary extinction coefficient (43.5 L g −1 cm −1 ) than other MXenes. The parameter of IPTCE can guide the choice of PTA concentration for complete photothermal healing in vitro and vivo. This proof-of-principle demonstration highlights synthetically tailoring of the light harvesting over NIR-II biowindow for a given host material by anionic regulation and further optimizes tumor photothermal therapy at low dose.

Photoacoustic (PA) imaging with functional nanoprobes in the second near-infrared region (NIR-II, 1000–1700 nm) has aroused much interest due to its deep tissue penetration and high maximum laser permissible exposure. However, most NIR-II PA imaging is performed using the two-dimensional (2D) imaging modality, which impedes the comprehension of the in vivo biodistribution, angiography and molecular-targeted performance of NIR-II nanoprobes (NPs). Herein, we report the systematic monitoring of biomineralized copper sulfide (CuS) NPs, typical NIR-II NPs, in mouse models by employing NIR-II three-dimensional (3D) PA imaging. The advanced imaging modality provides dynamic information about the 3D biodistribution and metabolic pathway of CuS NPs. We also achieved contrast-enhanced 3D PA imaging of abdominal and cerebral vessels at a high signal-to-background ratio. Moreover, the tumor-targeted CuS NPs conjugated with the bombesin peptide endowed NIR-II 3D PA with superior performance in imaging orthotopic tumors both deep in the prostate and in the brain beneath the intact scalp and skull. Our results highlight the potential of NIR-II 3D PA imaging for the evaluation of the in vivo behavior of NPs, thus providing a promising strategy for screening NPs in clinical translational studies.

Graphical Quinoid conjugated polymer PIS with broad absorption in the NIR-II region is developed based on multiple strategies. Benefiting from its remarkable photothermal conversion behavior under NIR-II laser (1064 nm), PIS nanoparticles show excellent antitumor performances in both in vitro and in vivo studies. This work presents a rational design strategy to explore NIR-II photothermal agents for hyperthermia therapy. Recently, extensive efforts have been devoted to the development of the second near-infrared bio-window (NIR-II, 1000–1700 nm) theranostic agents owing to the excellent tissue-penetration capability of NIR-II light. The exploration of organic NIR-II photothermal therapy materials, especially those with absorption peak over 1000 nm, is an appealing yet significantly challenging task. Herein, we have designed conjugated polymer nanoparticles (PIS NPs) with NIR-II absorption peak at 1026 nm through a combined strategy of introducing quinoid donor-acceptor (D–A) structures, constructing intramolecular “conformational locks” and extending the conjugation area to narrow the band gap. Irradiated at 1064 nm, PIS NPs showed remarkable photothermal conversion performance for efficient photothermal ablation of tumor cells in vitro and in vivo. This study provides useful insights into the rational design of organic NIR-II photothermal agents based on multiple strategies.
