N-halamines functionalized PCL nanofibrous yarns with antimicrobial properties for sutures applications

Yingfeng Wang,Ziyi Liao,Ming Huang,Kaizhun Li,Nengyu Pan,Hao Hou,Wei Ma,Maoli Yin

Post-surgical site infections (PSSI) pose significant challenges in clinic treatment, and the antibacterial modification of medical materials is an effective strategy to combat these issues. In this work, a monomeric N-halamines (DCDMH) was doped in PCL matrix via electrospinning process, and further processed using a conventional twisting method to yield nanofiber-based yarns. The micro/nano-structure of the PCL-based electrospun nanofibrous yarns (ENYs) enables the effective release of N-halamines and exhibits suitable storage stability after 30 days. The N-halamine modified ENYs effectively inhibited 100 % of E. coli O157:H7, S. aureus , and C. albicans within 1 min, resulting impressive antimicrobial capacities. The effect of N-halamines dosage on the morphology and mechanical properties of fabricated ENYs was assessed. Furthermore, the in vitro degradation, hemocompatibility, and protein adsorption properties of the synthesized nanofibrous yarns were also evaluated. The prepared ENYs with good hemocompatibility and antimicrobial properties, suggesting a novel approach for the development of medical materials, specifically for surgical sutures materials.

Functionalized 3D-Printed PCL/β-TCP Scaffolds with ZnO for Enhanced Bone Healing

Ayijiamila Mohetaer,Shan Rong,Zhang Jing,Wang Yuxin,Wan Xiaofei,He Qian,Ma Chunquan,Liu Bin,Song Fuxiang

Alveolar ridge deficiency remains a significant clinical challenge in dental implantology, often necessitating bone augmentation procedures. While autogenous bone grafting is considered the gold standard, its limitations, including donor site complications and limited availability, have prompted the development of alternative bone graft materials. In this study, we fabricated a novel polycaprolactone (PCL)/β-tricalcium phosphate (β-TCP)@zinc oxide (ZnO) composite scaffold via direct ink writing (DIW) three-dimensional (3D) printing. The scaffold integrates a biomimetic porous architecture with a controlled ion-release strategy to enhance osteogenic performance. The characterization of the material confirmed excellent chemical stability, hydrophilicity, and tunable degradation behavior. The incorporation of ZnO facilitated the sustained release of bioactive ions (Ca 2+ , PO 4 3− , and Zn 2+ ), significantly improving the scaffold's bioactivity. In vitro assessments revealed that the scaffold with 2% ZnO (PTZ-2) exhibited optimal cytocompatibility and osteogenic differentiation capacity, promoting alkaline phosphatase activity and mineralized nodule formation in MC3T3-E1 cells. In a rat cranial defect model, the PTZ-2 scaffold achieved approximately 95% new bone area after 12 weeks. Histological analysis further confirmed the presence of continuous, highly mineralized bone matrix with excellent osseointegration capability. These findings collectively indicate that the 3D-printed PCL/β-TCP@ZnO composite scaffold has great potential for clinical translation in bone defect regeneration.

Inhibitory effect of Vibrio parahaemolyticus phage-functionalized PCL films on the host bacterium

Han Deng,Tian Shi,Liang Ke,Wei Xu,Mengni Zhou,Huajuan Wang,Min Zhou

An environmentally stable Vibrio parahaemolyticus phage 475s1 was isolated. • Phage 475s1 was stably immobilized onto biodegradable PCL films. • The films displayed strong antibacterial efficacy in vitro and in salmon. Vibrio parahaemolyticus ( V. parahaemolyticus ) is recognized as a major foodborne pathogen with global prevalence. The overuse of traditional antibiotics has not only induced the emergence of drug-resistant strains, but also posed a potential threat to human health through the food chain. Therefore, the development of green and precisely targeted antibacterial agents is urgent. In this study, a V. parahaemolyticus phage 475s1 was isolated and exhibited remarkable stability over a broad temperature range (0-70 °C), pH range (4–11) and ultraviolet (UV) irradiation durations (0-60 min). It was capable of lysing 12 out of 23 tested V. parahaemolyticus strains, with a burst size of 110 plaque-forming units (PFU) per infected cell. The phage 475s1 was immobilized onto the surface of biodegradable polycaprolactone (PCL) films. The results showed that the binding density of phage on the PCL films reached 0.04 phages/μm 2 , while the host-capturing density was approximately 0.05 cells/μm 2 . After ultrasonic treatment for 20 min, 35% of the phages remained stably adsorbed on the PCL films. The phage-functionalized PCL films were further applied to artificially contaminated salmon samples. At an incubation temperature of 37 °C, the phage-functionalized PCL films exhibited significant antibacterial efficacy. Compared with the control group, the bacterial concentration decreased by 1.72 log CFU/mL at 2 h and 1.60 log CFU/mL at 3 h, respectively. Therefore, these findings indicate that the V. parahaemolyticus phage-functionalized PCL films hold great promise as a novel and effective antibacterial packaging material. This work provides a feasible green antibacterial strategy against seafood-borne V. parahaemolyticus , offering valuable theoretical support for the development of biodegradable active packaging for industrial aquatic preservation.

Construction and Properties of SPI/PLA-PCL Composite Coating on Pure Titanium Surface

Chunmei Wang,Congyi Zhu,Qi Zhang,Shuangsheng Zhang,Ling Zhang,Jiang Wu,Guoliang Zhang

Titanium and titanium alloys have been widely used in clinical implants such as dental implants due to their high strength and corrosion resistance. However, their inherent biological inertness and mismatch with the elastic modulus of human bone tissue restrict bone healing and reconstruction. In this study, pure titanium was first modified by micro-arc oxidation (MAO), and then a biodegradable soybean protein isolate (SPI)/polylactic acid (PLA)–polycaprolactone (PCL) composite coating was prepared by using the spin-coating method. The coating was systematically characterized by scanning electron microscopy (SEM), an energy spectrometer (EDS), Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), and atomic force microscopy (AFM), confirming that the composite coating was successfully prepared. An evaluation of the physical and chemical properties showed that the introduction of SPI significantly improved the hydrophilicity, surface roughness and adhesion to the substrate of the coating, regulated the degradation rate, and reduced the elastic modulus to the range of 10–30 GPa, which matches human bone tissue. At the same time, it enhanced corrosion resistance. In vitro MC3T3-E1 osteoblast experiments showed that the coating containing 50% SPI showed better cell compatibility, adhesion ability and osteogenic inducibility, and all groups had good blood compatibility. The SPI/PLA–PCL composite coating effectively improves the biological activity of the pure titanium surface and provides a feasible strategy for the surface modification of titanium implants and bone defect repair.

Enhancing wound healing through sonodynamic silver/barium titanate heterostructures-loading gelatin/PCL nanodressings

Yusen Zhang,Shuai Ke,Xiao Hu,Shuangying Wang,Wanqi Peng,Xinhang Qian,Linghui Tian,Huijun Wu,Binghui Li,Xiantao Zeng,Lingling Zhang

Bacterial infections present a formidable challenge in surgical procedures, and are a major threat to wound healing. Sonodynamic therapy (SDT) is a non-invasive approach for fighting pathogens; however, it is hindered by the efficiency of sonosensitizers and effective antibacterial time. In this study, we developed a biocompatible nanodressing to improve the antibacterial efficacy and accelerate wound healing via SDT. Silver nanoparticles (NPs) were synthesized on tetragonal barium titanate (BTO) NPs to create an Ag@BTO heterostructure of sonosensitizers to improve their piezocatalytic activities, which were then incorporated into gelatin/polycaprolactone (PCL) to form Ag@BTO-gelatin/PCL nanofiber (ABT-gP NFs) dressings. The loading of Ag@BTO NPs resulted in ABT-gP NFs with better mechanical properties and excellent piezocatalysis, which produced reactive oxygen species and Ag + to kill bacteria during ultrasound (US) irradiation. Additionally, nanodressing released moderate amounts of silver ions without US, prolonging the antibacterial time, while promoting fibroblast migration. This approach was effective in killing Gram-positive and Gram-negative bacteria (100 % and 90.8 %, respectively), promoting cell migration in vitro and accelerating wound healing without adverse effects in vivo . This study extends the potential applications of ultrasound-triggered nanodressing to the field of wound healing.

Humidity-responsive PCL/ICs/PVA pad with directional water transport for fruit preservation

Xuling Song,Zhengfeng Wang,Minxin Zhou,Yuwei Li,Yujie Wei,Yi Luo,Jianfeng Li,Bei Xu,Baijun Shang,Qingshan Duan

A smart humidity-responsive antimicrobial pad with directional water transport (DWT) was developed to address the rapid spoilage of fruits after harvest due to high humidity and water accumulation. This pad was fabricated from polycaprolactone/cinnamaldehyde β-cyclodextrin inclusion complexes/polyvinyl alcohol (PCL/ICs/PVA) using electrospinning and co-precipitation methods. The inclusion complexes formed by β-cyclodextrin and the guest molecule cinnamaldehyde (CIN) undergo crystalline and conformational changes upon moisture absorption, generating gaps or channels enabling humidity-responsive release of CIN. Meanwhile, capillary action, water uptake by the hydrophilic layer, and reverse water penetration resistance from the hydrophobic layer jointly promote DWT. In the PCL/ICs/PVA pad, the hydrophobic PCL coalesces free water molecules in the air to form tiny droplets to promote DWT, ICs release CIN in response to humidity, and the hydrophilic PVA absorbs droplets, together enhancing the antibacterial performance of the pad under high humidity conditions. The 14 PCL layer (electrospun from 14 wt% PCL solution) with honeycomb-like morphology was beneficial for loading ICs and humidity response. At 98 % RH, the pad with 33.3 % ICs sustained CIN release for 168 h and extended strawberry shelf life. This study holds significant implications for fruit preservation in high-humidity environments.

Oral delivery of WR-1065 by ROS-responsive PEG-PCL nanoparticles for radioprotection

Xiaona Lin,Xinxin Wang,Hongqi Tian

When a burst of reactive oxygen species (ROS) is induced after radiation exposure, a sufficient radioprotector drug must be quickly released to neutralize the ROS . An oral polymeric carrier of poly(ε-caprolactone)-b-poly(ethylene glycol)-b-poly(ε-caprolactone) was synthesized via the ROS-response linker thioketal (TK-linked PCEC) and used for delivery of the drug WR-1065. The drug-loaded nanoparticles (PCEC/WR-1065 NPs) had high WR-1065 content (19.7 % w/w) in the core and a particle size of 183 nm. The PCEC/WR-1065 NPs could protect the drug from degradation at gastric pH. More than 70 % of the TK-linked PCEC copolymer could be cleaved in 1 h. Therefore, a sufficient amount of WR-1065 could be quickly released from the PCEC/WR-1065 NPs to neutralize the burst of free radicals generated under irradiation. The in vitro cell uptake results of TK-linked PCEC NPs indicated that the NPs could be efficiently endocytosized by Caco-2 cells. Oral administration of the PCEC/WR-1065 led to survival benefit as well as protective effects on hematopoiesis and main organs with respect to the irradiation and oral free WR-1065 groups after 7.2 and 8.0 Gy total body irradiation, respectively. In vivo biodistribution studies indicated that the PCEC/WR-1065 NPs accumulated mainly in intestinal tissue. These results suggest that the design presented here provides a promising oral ROS-sensitive platform for acute radiation syndrome.

A facile strategy for fabricating PCL/PEG block copolymer with excellent enzymatic degradation

Haipeng Wang,Dejin Tong,Lei Wang,Lei Chen,Na Yu,Zhanxiong Li

Poly(ω-caprolactone) (PCL) has been the focus in many research initiatives, especially regarding various ways though which to introduce reactive sites to the polyester chains. In this work, hydroxyl groups were covalently introduced into the chain of PCL by reacting ester groups with 6-amino-1-hexanol. Poly(ethylene glycol) (PEG) was crosslinked to the PCL chain through esterification , by using N,N’-carbonyldiimidazole (CDI) as catalyst. The prepared PCL-PEG copolymers were characterized by various analytical techniques such as Fourier transform infrared spectroscopy (FT-IR), 1 H NMR and X-ray photoelectron spectroscopy (XPS). Static water contact angles measurement indicated that hydrophilicity of the copolymer films modified by PEG has improved considerably. The copolymer films were prepared by casting the polymer solution onto a stainless plate. The degradation of the copolymer samples, in a phosphate buffer solution containing a type of Lipase (Aspergillus Oryzae), was observed at different time intervals. The results of weight loss ratio, during the degradation process, indicated that blocking PEG chains onto the PCL chains can effectively accelerate the process of enzymatic degradation of the resulting copolymer. The changes in molecular weight of the copolymer films were investigated by Gel Permeation Chromatography (GPC) and the copolymers were characterized by 1 H NMR during degradation. Surface morphology of PCL and PCL-PEG polymers, before and after degradation, was observed by Scanning electron microscopy (SEM), and emphasized that the enzymatic degradation of PCL-PEG copolymers occurred faster by comparison with PCL, starting from the surface layer of the films.

Preparation and characterization of PCL@β-TCP composite antibacterial scaffolds by additive manufacturing

Chen Zhang,Yi Qi,Binying Dai,Runyi Lin,Jinchao You,Yaomei Wang,Shunxue Xing,Yuansong Ye,De-Jing Li

Poly(ε-caprolactone) (PCL) and β-tricalcium phosphate (β-TCP) are promising materials for bone tissue engineering due to their complementary biodegradability and osteoconductivity. However, their individual limitations hinder clinical application. In this study, PCL@β-TCP composite scaffolds containing 5%, 10%, and 20% β-TCP were fabricated via fused deposition modeling (FDM). The effects of β-TCP content on the physicochemical, mechanical, and antibacterial properties of the scaffolds were systematically investigated. Results showed that increasing β-TCP content enhanced hydrophilicity (contact angle reduced from 89.1° to 66.9°), accelerated biodegradation (higher weight loss after 103 days in PBS), and improved antibacterial activity, as evidenced by larger inhibition zones against E. coli and S. aureus. These findings demonstrate that β-TCP incorporation effectively enhances the bioactivity and antibacterial performance of PCL scaffolds, highlighting their potential for bone tissue engineering applications, particularly in non-load-bearing contexts.

Fabrication of PCL/CMARX/GO Composite Nanofibrous Mats for Dye Adsorption: Wastewater Treatment

Mona Saad Binkadem

The effluents of industrial wastewater contain several toxic organic and inorganic pollutants that may contaminate clean and freshwater sources if untreated or poorly treated. These toxic pollutants include colors; hazardous compounds; surfactants; cosmetics; agrochemicals; pharmaceutical by-products; and agricultural, pharmaceutical, and medical contaminants. Treating wastewater has become a global problem. Many projects have been started in the last two decades to treat wastewater, resultant water pollution, and associated waste management problems. Adsorbants based on graphene oxide (GO) are viable wastewater treatment materials due to their adaptability, photocatalytic action, and capacity for self-assembly. Here, we report the fabrication of nanofibrous mats from polycaprolactone (PCL), carboxymethyl arabinoxylan (CMARX), and carboxyl-functionalized-graphene oxide using an electrospinning technique. The silver nanoparticles were loaded onto the mat to enhance their photocatalytic activity. These mats were characterized using different techniques, including Fourier transform infrared (FTIR), scanning electron microscope (SEM), and transmission electron microscope (TEM). The water contact angles were used to study their hydrophilic and hydrophobic behavior. The Langmuir isotherm model and adsorption kinetics were studied to evaluate their adsorption capabilities against methylene blue (MB). Sample 2 followed the Langmuir isotherm model (R2= 0.9939). Adsorption kinetics exhibited pseudo-second order behavior (R2= 0.9978) due to their maximum correlation coefficient values. MB has excellent adsorption at room temperature and the formation of the monolayer at the surface of the adsorption mat. An enhanced PO43−and MB adsorption was observed, providing recyclability up to 4–5 times. Hence, the fabricated nanofibrous mat would be a potential candidate for more effective wastewater treatment applications. Keywords:adsorption;bacterial cellulose;electrospinning;thin nanofibrous mat;wastewater treatment;environmental pollution treatment

Electrohydrodynamic Printing of PCL@CsPbBr3 Composite Fibers with High Luminescence for Flexible Displays

Maolin Liu,Changqing Lin,Weicheng Ou,Han Wang,Chunyang Pan,Yuchen Ji,Hehui Zheng

Metal halide perovskite quantum dots (PQDs) are widely used in the display field due to their excellent photoelectric properties, such as ultra-narrow half-peak widths and ultra-pure luminescence color purity. Inkjet printing, laser direct writing and electrospinning are all common methods for PQDs printing to prepare micropattern displays. In order to produce large-scale and high-resolution PQDs micropatterns, electrohydrodynamic (EHD) printing technology is capable of large-scale deposition of highly oriented nanofibers on rigid or pliable, flat or bent substrates with the advantages of real-time regulation and single control. Therefore, it has a lot of potential in the fabrication of pliable electronic devices for one-dimensional ordered light-emitting fibers. Polycaprolactone (PCL) as an EHD printing technology polymer material has the advantages of superior biocompatibility, a low melting point, saving energy and easy degradation. By synthesizing CsPbBr3quantum dots (QDs) and PCL composite spinning stock solution, we used the self-built EHD printing platform to prepare the PCL@CsPbBr3composite light-emitting optical fiber and realized the flexible display of high-resolution micropatterns in polydimethylsiloxane (PDMS) packaging. An x-ray diffractometer (XRD), scanning electron microscope (SEM) and photoluminescence (PL) were used to characterize and analyze the fiber’s morphology, phase and spectral characteristics. EHD printing technology may open up interesting possibilities for flexible display applications based on metal halide PQDs.Keywords:perovskite;flexible display;photoluminescent fibers;EHD;CsPbBr3

Fe3O4/ poly(caprolactone) (PCL) electrospun membranes as methylene blue catalyst with high recyclability

Wen Xue,Yang Hu,Fujun Wang,Xudong Yang,Lu Wang

High specific-surface-area porous poly(caprolactone) (PCL) membranes combined with Fe 3 O 4 nanoparticles were fabricated by electrospinning technology. Their applications as catalysts in Fenton reactions to degrade methylene blue (MB) were evaluated in the presence of H 2 O 2 and UV light . The results indicated that the fibrous catalyst system illustrated efficient MB removal performance (99% of MB degradation rate) and was viable after 6 cycles (96% of MB degradation rate). Besides, the morphology, chemical composition , magnetic and mechanical properties were also accessed by scanning electron microscopy (SEM), X-ray diffraction (XRD), Fourier transform infrared (FT-IR) spectroscopy, magnetometry , and strain-stress curves. The outstanding catalytic behaviors of the composites are attributed to synergy of porous surface and embedded Fe 3 O 4 nanoparticles. It also shows advantages of being environment-friendly and separated easily without an external magnetic field.
