Layered bismuth oxyhalide nanomaterials have received much more interest as promising photocatalysts because of their unique layered structures and high photocatalytic performance, which can be used as potential inorganic photosensitizers in tumor photodynamic therapy (PDT). In recent years, photocatalytic materials have been widely used in PDT and photothermal therapy (PTT) as inorganic photosensitizers. This investigation focuses on applying layered bismuth oxyhalide nanomaterials toward cancer PDT, an application that has never been reported so far. The results of our study indicate that the efficiency of UV-triggered PDT was highest when using BiOCl nanoplates followed by BiOCl nanosheets, and then TiO2. Of particular interest is the fact that layered BiOCl nanomaterials showed excellent PDT effects under low nanomaterial dose (20 μg mL−1) and low UV dose (2.2 mW cm−2 for 10 min) conditions, while TiO2 showed almost no therapeutic effect under the same parameters. BiOCl nanoplates and nanosheets have shown excellent performance and an extensive range of applications in PDT.

Objective This study developed a multifunctional MnFe2O4/TiVNbMoC3-cRGD (MXenes) nanocomposite for targeted photothermal- photodynamic therapy against bladder cancer. Methods A multifunctional MXene-based nanocomposite was constructed by integrating MnFe2O4 nanoparticles onto TiVNbMoC3 MXene nanosheets to form a layer-on-layer Schottky junction, followed by conjugation of cRGD peptides for tumor targeting. Successful synthesis, colloidal stability, broad optical absorption, efficient photothermal conversion and ROS generation under 808 nm laser were confirmed. In vitro assays were conducted using 5637 bladder cancer cells, and a murine bladder tumor model was established to evaluate the anticancer efficacy. Biocompatibility assessments were also carried out. Results The composite was constructed by integrating MnFe2O4 nanoparticles onto TiVNbMoC3 MXene nanosheets, which formed a layer - on - layer Schottky junction. Then, cRGD peptides were conjugated to the surface for tumor targeting. Comprehensive characterization confirmed successful synthesis, favorable colloidal stability, broad optical absorption, and efficient photothermal conversion and reactive oxygen species (ROS) generation under 808 nm laser irradiation. In vitro, the cRGD-targeted nanocomposite was efficiently internalized by 5637 bladder cancer cells and showed significantly higher anticancer efficacy than doxorubicin (DOX), including inhibition of proliferation, migration, colony formation, and induction of apoptosis and immunogenic cell death. In a murine bladder tumor model, the nanocomposite achieved the strongest tumor growth suppression and reduced final tumor volume more effectively than DOX. Biocompatibility assessments showed minimal hemolysis and no major organ damage at therapeutic doses. Conclusion This MXene-based Schottky junction nanocomposite represents a promising targeted platform for dual-modal photothermal/photodynamic therapy combined with immune activation in bladder cancer, warranting further translational investigation.

Graphical Ag 2 S quantum dots are shown to be capable of killing tumor cells by photodynamic therapy with near-infrared light irradiation, and after modification with polydopamine, they can induce more reactive oxygen species as a result of the regulation of polydopamine, which is demonstrated by experiments. Given their superior penetration depths, photosensitizers with longer absorption wavelengths present broader application prospects in photodynamic therapy (PDT). Herein, Ag 2 S quantum dots were discovered, for the first time, to be capable of killing tumor cells through the photodynamic route by near-infrared light irradiation, which means relatively less excitation of the probe compared with traditional photosensitizers absorbing short wavelengths. On modification with polydopamine (PDA), PDA-Ag 2 S was obtained, which showed outstanding capacity for inducing reactive oxygen species (increased by 1.69 times). With the addition of PDA, Ag 2 S had more opportunities to react with surrounding O 2 , which was demonstrated by typical triplet electron spin resonance (ESR) analysis. Furthermore, the PDT effects of Ag 2 S and PDA-Ag 2 S achieved at longer wavelengths were almost identical to the effects produced at 660 nm, which was proved by studies in vitro. PDA-Ag 2 S showed distinctly better therapeutic effects than Ag 2 S in experiments in vivo, which further validated the enhanced regulatory effect of PDA. Altogether, a new photosensitizer with longer absorption wavelength was developed by using the hitherto-unexplored photodynamic function of Ag 2 S quantum dots, which extended and enhanced the regulatory effect originating from PDA.

Effectively prolonging the residence time of nanoprobes in the tumor region and reducing the accumulation of nanoprobes in the vital organs (for example, lung, liver and spleen) is crucial for high-efficiency photodynamic therapy (PDT) of cancer. Herein, we systematically report an ultra-small and highly stable nanoplatform with diameters of 4, 8 and 13 nm that exhibited excellent photodynamic therapeutic efficacy using Fe 3 O 4 @polymer-NPO/PEG-Glc@Ce6 nanoprobes. Based on the small size effect, the nanoprobes displayed lower cytotoxicity and excellent biocompatibility. Owing to the synergistic virtues of markedly active targeting and intrinsic small size effect, the Fe 3 O 4 @P-NPO/PEG-Glc@Ce6 nanoprobes can effectively prolong their residence time in the tumor region and reduce accumulation in the normal organs. Benefitting from the small size effect, the synthesized Fe 3 O 4 @P-NPO/PEG-Glc@Ce6 nanoprobes exhibited excellent tumor-targeting capability and photodynamic therapeutic efficacy by inhibiting the growth of tumors in mice under visible red light irradiation with a relatively lower power. The successful application of the small size effect in Fe 3 O 4 @P-NPO/PEG-Glc@Ce6 nanoprobes to significantly improve the PDT efficiency in our strategy suggests new building blocks for PDT of tumors and paves a new way for clinical therapies and translation in the near future.

Traditional photodynamic therapy (PDT) suffers from poor tissue penetration due to using external light as excitation source. Cerenkov radiation (CR) from radionuclides can serve as an internal light source to activate photosensitizers inside lesion tissues. Using CR instead of external light in PDT is thought to be an effective way to overcome the shortcoming of traditional PDT. However, the weakness of CR emission limits the outcomes of CR-induced PDT (CR-PDT). We anticipated that CR-induced combined therapy could effectively enhance the therapeutic outcome of CR-PDT. Herein, a ROS-responsive Chemo-PDT agent ( TTC ) was synthesized by conjugation of tetraphenylporphyrin (photosensitizer) with camptothecin (chemotherapeutic drug) through a ROS-cleavable thioketal linker. Under activation by CR emission from gallium-68 ( 68 Ga), the porphyrin moiety could generate ROS , which then cleaves the thioketal linker to induce the subsequent release of CPT. Using [ 68 Ga]Ga-NOTA-Nb109 as an intracellular CR emitter, in comparison with its non-cleavable analogue ( TUC ) only displaying phototoxicity, TTC exhibited much higher cytotoxicity toward A375-hPD-L1 cells due to its combined effect of PDT and chemotherapy. Our work presents a new strategy to use ROS-responsive Chemo-PDT agent to improve the outcomes of CR-PDT.

The emergence of drug-resistant bacteria and their easy recurrence have been challenging in the clinical treatment of skin abscesses. Therefore, there is an urgent need to develop treatments against bacterial infections. This study developed an antibacterial PDT/CDT dual-mode treatment platform (Ce6/TA-Fe3+ NPs) to treat subcutaneous abscesses and promote wound healing. The Ce6/TA-Fe3+ NPs can quickly generate 1O2 under illumination and ·OH through the Fenton reaction in the microenvironment, leading to the eradication of biofilms. In particular, the Ce6/TA-Fe3+ NPs demonstrated over 90% effectiveness in fighting Staphylococcus aureus (S. aureus) bacteria and successfully eliminated biofilms after being exposed to a 660 nm laser for 10 min. Antibacterial mechanism research showed that the Ce6/TA-Fe3+ NPs induced a speedy rupture of the bacterial cell membranes, membrane lipid peroxidation, protein leakage, inhibition of intracellular ATP, and DNA damage based on the synergistic effect of PDT and CDT. Meanwhile, the Ce6/TA-Fe3+ NPs demonstrated good biosafety to normal cells and can clear the bacteria from the infected wound to promote the healing process of the infected area. The experimental results in vivo demonstrated that Ce6/TA-Fe3+ NPs effectively treated skin abscesses. Therefore, this study suggested that the Ce6/TA-Fe3+ NPs possessed the ability to kill bacteria, eliminate biofilms, and effectively treat subcutaneous abscesses based on the synergistic mechanism of PDT and CDT, which provided the promising strategy for the antibacterial medication and treatment of subcutaneous abscesses.

Photodynamic/photothermal therapy (PDT/PTT) has become a research focus of cancer treatment due to the non-invasiveness, spatio-temporal controllability, and effectiveness of repeated treatment. Here, Au@MOF core–shell hybrids were designed and constructed by the layer-by-layer method, and the thickness of the MOF shell can be adjusted by controlling the coordination reaction between the layers. Au nanorod cores mainly produce the PTT effect due to their strong absorbance at 650 nm. The porphyrin ligand in the MOF shell can convert O2 into 1O2 under light conditions, resulting in a high PDT effect. Moreover, the metal node Fe3O(OAc)6(H2O)3+ cluster of the MOF can catalyze the decomposition of H2O2 into O2 to overcome the hypoxic environment of tumors, which further improves the effect of PDT. The combination of the porphyrin ligand in the MOF structure and Au nanorods has promoted the synergistic effects of PDT/PTT. As expected, the results confirmed that Au@MOF hybrids showed no obvious biotoxicity in both cells and animal experiments, and exhibited good biocompatibility. With the synergistic effects of PDT/PTT, cancer cells could be effectively killed and tumor growth could be inhibited. In addition, the modification of folic acid on the surface of Au@MOF can further enrich the hybrids at the tumor site and enhance the inhibitory effect on tumors. These studies have proved that PDT and PTT can be effectively combined and have greater advantages in enhancing the treatment of tumors.

The integration of hypoxia-activated chemotherapy with photodynamic therapy (PDT) has newly become a potent strategy for tumor treatment. Herein, a reactive oxygen species (ROS)-responsive drug carriers (PS@AQ4N/mPEG- b -PSe NPs) are fabricated based on the amphiphilic selenium-containing methoxy poly(ethylene glycol)-polycarbonate (mPEG- b -PSe), the hydrophobic photosensitizer (PS), and hypoxia-activated prodrug Banoxantrone (AQ4N). The obtained nanoparticles are spherical with an average diameter of 100 nm as characterized by transmission electron microscope (TEM) and dynamic laser scattering (DLS) respectively. The encapsulation efficiency of the PS and AQ4N reaches 92.83% and 51.04% at different conditions, respectively, by UV–vis spectrophotometer. It is found that the drug release is accelerated due to the good ROS responsiveness of mPEG- b -PSe and the cumulative release of AQ4N is up to 89% within 30 h. The cell test demonstrates that the nanoparticles dissociate when triggered by the ROS stimuli in the cancer cells, thus the PS is exposed to more oxygen and the ROS generation efficiency is enhanced accordingly. The consumption of oxygen during PDT leads to the increased tumor hypoxia, and subsequently activates AQ4N into cytotoxic counterpart to inhibit tumor growth. Therefore, the synergistic therapeutic efficacy demonstrates this drug delivery has great potential for antitumor therapy.

Currently, the low survival rate and poor prognosis of patients with nasopharyngeal carcinoma are ascribed to the lack of early and accurate diagnosis and resistance to radiotherapy. In parallel, the integration of imaging-guided diagnosis and precise treatment has gained much attention in the field of theranostic nanotechnology. However, constructing dual-modal imaging-guided nanotheranostics with desired imaging performance as well as great biocompatibility remains challenging. Therefore, we developed a simple but multifunctional nanotheranostic GdCPP for the early and accurate diagnosis and efficient treatment of nasopharyngeal carcinoma (NPC), which combined fluorescence imaging and magnetic resonance imaging (MRI) onto a single nanoplatform for imaging-guided subsequent photodynamic therapy (PDT). GdCPP had an appropriate particle size (81.93 ± 0.69 nm) and was highly stable, resulting in sufficient tumor accumulation, which along with massive reactive oxygen species (ROS) generation upon irradiation further significantly killed tumor cells. Moreover, GdCPP owned much stronger r1 relaxivity (9.396 mM–1 s–1) compared to clinically used Gd-DTPA (5.034 mM–1 s–1) and exhibited better T1WI MRI performance. Under dual-modal imaging-guided PDT, GdCPP achieved efficient therapeutic outcomes without causing any noticeable tissue damage. The results of in vitro and in vivo studies indicated that GdCPP may be a suitable candidate for dual-modal imaging-guided precision tumor therapy.

Triple-negative breast cancer (TNBC) presents a formidable challenge due to its poorest prognosis and limited array of treatment options available. Photodynamic therapy (PDT) has emerged as a potent therapeutic modality to generate intratumoral toxic reactive oxygen species (ROS) in combating refractory triple-negative breast cancer (TNBC). However, its therapeutic efficacy is compromised due to insufficient tumor accumulation and therapeutic resistance. Herein, an “all-in-one” tumor-therapeutic nanomedicine named HA@IR780@KU55933@BSA (HIKB) which integrated photosensitizer IR780 with ATM kinase inhibitor KU55933 was designed to facilitate drug delivery and target specific pathways involved in tumor PDT treatment resistance. Co-delivery of IR780 and KU55933 exacerbated intracellular ROS production, mitochondrial dysfunction and DNA damage to form a potent anti-TNBC therapeutic cyclical feedback loop and then induced pyroptosis and apoptosis of TNBC cells by activating the Caspase3/GSDME signaling pathway and regulating apoptosis-related protein expression, respectively. In vivo evaluations in the TNBC orthotopic xenograft mouse model demonstrated that the designed HIKB NPs could accumulate in tumor tissues and exert synergistic therapeutic effects. Altogether, this study described a self-assembling strategy for constructing an all-in-one nanomedicine that effectively integrates multiple therapeutic modalities to provide a comprehensive and systemic approach to tumor suppression.

In photodynamic therapy (PDT), the concentration of reactive oxygen species (ROS) generated in cells directly determines the therapeutic effect. However, excessive glutathione (GSH) concentration at the tumor site and hypoxic environment strongly reduce the production of ROS. Nitric oxide (NO) has a variety of anti-tumor activities, including the induction of cell apoptosis and sensitization to PDT, so it is in the spotlight in the area of cancer intervention. Therefore, it is very innovative to combine NO therapy with PDT and simultaneously reduce GSH content and relieve hypoxia. Metal-organic framework (MOF) is an emerging drug delivery vehicle that can exert photodynamic effects when a photosensitizer is used as its organic ligand. In this study, we constructed a GSH-responsive NO producing nanosystem to enhance ROS production through GSH depletion and hypoxia alleviation. Briefly, nicorandil (Nic) is encapsulated into porous porphyrinic MOF nanoparticles, and hyaluronic acid (HA) is electrostatically adsorbed on the surface of the MOF nanoparticles for CD44 receptor-targeting and prevention of Nic leakage. Notably, the Nic reacts with GSH to produce NO gas with vasodilation and GSH reduction, thus improving oxygen supply for efficient ROS generation. Moreover, the generated NO can further react with superoxide radical anion to produce highly reactive peroxynitrite (ONOO − ) molecules with higher cytotoxicity. Overall, the [email protected] nanosystem combines NO gas therapy with PDT, resulting in a significant increase in ROS production and greatly enhancing the efficacy of PDT.

Reactive oxygen species (ROS) generated during photodynamic therapy (PDT) can trigger autophagy. However, little research is focused on whether there is a synergistic anticancer effect with PDT if extra autophagy promoter or inhibitor is added. Here, it is found that autophagy promotion significantly enhances the PDT activity to cancer cells. Based on this preliminary result, a ROS-sensitive self-assembled dendrimer nanoparticle is exploited as a carrier to codeliver an autophagy promoter (rapamycin, Rapa) and photosensitizer (phthalocyanine, Pc) to the tumor. After entrapped by cancer cells and irradiated by light, the ROS generated in PDT process of Pc can trigger nanoparticle destruction to release Rapa, thus initiating the autophagy process and remarkably enhancing the efficacy of PDT, leading to efficient tumor suppression.
