(E)-2-Hexenal is one of the most prevalent and abundant volatile substances in fruits. Many researchers have attempted fumigation with (E)-2-hexenal to control fungal diseases with poor or no results; the key is the inability to maintain an effective concentration for a long time. Therefore, a new starch-based antifungal coating with (E)-2-hexenal microcapsules was prepared. The obtained results showed that the microcapsule embedding rate reached 78.95% by orthogonal optimization experiments and that the (E)-2-hexenal-based coating was effective at reducing the lesion diameter of apple fruit during storage at 25 °C after Penicillium expansum infection, and in other fruits; it also achieved a good antifungal effect. Moreover, the activities of the main defense-related enzymes and the expression of key genes involved in the salicylic acid (SA) signaling pathway, such as CHI, GLU, PAL , NPR1 , and PR1 , were enhanced by (E)-2-hexenal-based coating treatment. These findings suggest that the (E)-2-hexenal-based coating has the dual effect of direct antifungal and induction of apple system resistance. E2H-based coating is a promising approach to control postharvest Penicillium expansum in fruit.

A major citrus postharvest pathogen,Penicilliumitalicum(P. italicum), causes substantial economic losses in citrus. In this study, a citral nanoemulsion containing polymethoxylated flavonoids (PMFs), the antimicrobial compounds from citrus, was prepared. The antifungal activity and potential antifungal mechanisms of the nanoemulsion againstP. italicumwere evaluated. The results showed that the growth ofP. italicumwas effectively inhibited by the nanoemulsion, with a minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) of 62.5 and 250 mg L−1, respectively. The nanoemulsion significantly inhibited spore germination and mycelial growth, and it altered the morphology ofP. italicum. In addition, the permeability of the cell membrane increased with increasing nanoemulsion concentrations, as evidenced by a rapid rise in extracellular electric conductivity and stronger red fluorescence from mycelia (propidium iodide staining). Compared with the control, the nanoemulsion treatment induced a decrease in total lipid and ergosterol contents inP. italicumcells by 64.61% and 60.58%, respectively, demonstrating that membrane integrity had been disrupted. The results indicated that the PMFs-loaded nanoemulsion exerted antifungal activity againstP. italicumby disrupting cell membrane integrity and permeability; such a nanoemulsion may be used as a potential fungicide substitute for preservation in citrus fruits. Keywords:antifungal mechanism;citrus blue mold;membrane integrity;membrane permeability;P. italicum

Penicillium italicum, the cause of citrus blue mold, is a pathogenic fungus that seriously affects the postharvest quality of citrus fruit and causes serious economic loss. In this study, a eugenol nanoemulsion containing limonin, an antimicrobial component from citrus seeds, was prepared using a high-pressure microfluidizer and the antifungal activity of the nanoemulsions againstP. italicumwas evaluated based on the conidial germination rate, mycelial growth, and scanning electron microscopy analysis. The results showed that the minimum inhibitory concentration and the inhibition rate of limonin-loaded eugenol nanoemulsion was 160 μg/mL and 59.21%, respectively, which was more potent than that of the limonin-free eugenol emulsion. After treatment with the nanoemulsions, the integrity of theP. italicumcell membrane was disrupted, the cell morphology was abnormal, and the leakage of nucleic acid and protein was observed. In addition, the challenge test on citrus fruits revealed that the limonin-loaded eugenol emulsion inhibited citrus infection for longer periods, with an infection rate of 29.2% after 5 days. The current research shows that nanoemulsions containing limonin and eugenol have effective antifungal activity againstP. italicum, and may be used as a substitute for inhibiting blue mold in citrus fruits. Keywords:eugenol nanoemulsion;citrus blue mold;limonin;Penicillium Italicum;antifungal activity

Mycelial pellets prepared using a marine-derived Penicillium janthinellum sp. strain (P1) were applied as biosorbents to remove dyes from aqueous solutions. Nine different dyes were studied and the effects of pH and salt concentration on the decolorization processes were investigated. The results showed that the mycelial pellet was effective in removing dyes from their water solutions. Moreover, it was found that pH was an important factor of the decolorization process and the best dye removal performance was achieved when pH = 5.0. The adsorption process of the live mycelial pellets was not affected by high salt concentrations. In addition, the mycelial pellets retained 91.3% of the original decolorization activity after five reusing cycles without any desorption treatments. The adsorption isotherms indicated that the biosorption process agreed well with the Langmuir isotherm model with a maximum biosorption capacity of 344.83 mg g(dry) − 1 and the kinetic adsorption results showed that the biosorption process followed a pseudo-second-order kinetic model.

Herein, we demonstrated a environmental-friendly biocatalytic route for the synthesis of dimethyl carbonate (DMC), which is the first example to use enzyme in the process. Moreover, immobilization of Penicillium expansum lipase (PEL) using environmentally benign and biodegradable CMC–PVA polymer has significantly enhanced the catalytic activity thus making them an eligible biocatalyst for synthesis of DMC. The biocatalyst revealed high catalytic performance even under ambient pressure and low temperature (conversion of EC to 94%, yield of DMC to 93% and selectivity of DMC to 99%). The immobilized lipase was effectively recycled for four consecutive cycles providing good yields of the desired product. Accompanying with the intense requirement for the green chemicals and process, our work can provide a useful idea for “green and clean” of harmful chemical reaction.

Penicillium expansum is a globally significant postharvest pathogen causing substantial economic losses in fruit and vegetable production. Despite the importance of this pathogen to global food security, the regulatory roles of transcription factors - particularly APSES family members - remain poorly characterized in P. expansum . Here, we identified PeStuA, a homolog of the key APSES member StuA, in P. expansum . Targeted deletion of PeStuA resulted in three independent deletion mutants. These mutants exhibited decreased biomass and the lack of the ability to form conidiophores and conidia, accompanied by significantly reduced expression of genes encoding fungal development regulators (BrlA, AabA, WetA, and VosA), confirming the role of PeStuA in fungal growth and development. PeStuA also influences the metabolism of P. expansum in terms of carbon and nitrogen sources. Notably, although Δ PeStuA mutants completely lost patulin production capacity in vitro , only a few genes within the patulin cluster were significantly down-regulated in expression, implying potential new regulatory mechanisms governing patulin production in P. expansum . Furthermore, the Δ PeStuA mutants presented significantly decreased pathogenicity on apple fruits. RNA sequencing analysis demonstrated the global regulatory role of PeStuA in spore development, primary metabolism and secondary metabolism during P. expansum infecting apple hosts. Overall, this study reveals the role of PeStuA and offers new perspectives on the molecular basis of P. expansum , providing a theoretical foundation for developing strategies against fruit diseases and patulin pollution it poses.

Penicillium expansum is a major pathogen causing blue mold rot in fruits and vegetables. Given the increasing concerns over the use of chemical fungicides in postharvest treatments, this research explores the effects of pulsed light (PL) on the growth and patulin production of P. expansum . Our results exhibited that PL significantly suppressed spore germination and mycelial growth while impairing the cell membrane integrity. Mechanistic studies revealed that PL treatment caused disruption of antioxidant defense systems and the excessive accumulation of reactive oxygen species (ROS), leading to the oxidative damage and mitochondrial dysfunction. Moreover, PL treatment significantly downregulated the expression of key genes associated with P. expansum growth and patulin biosynthesis. Specifically, the expression levels of global regulators CreA and LaeA were decreased by 39 % and 62 %, and 47 % and 82 % after PL30 and PL90 treatments, respectively. Additionally, genes within the patulin biosynthesis gene cluster, including patA , patK , patN , and patL , were significantly downregulated. In apples with PL90-treated P. expansum , the rot onset was delayed by 4 d. On day 7, PL90 reduced lesion diameter by 87.9 %, decay incidence by 67 %, and patulin content by 95 %. The findings indicate that PL is an effective technology for controlling postharvest diseases in fruits, making it a promising alternative to chemical treatments.

Green mold formed byPenicillium digitatumis a major disease that limits the yield and overall value of postharvest citrus fruits. The antifungal activity of sodium cuminate (SC) againstP. digitatumand the corresponding mechanism were explored in this research. The minimal inhibitory concentration (MIC) and minimal fungicidal concentration (MFC) of SC againstP. digitatumwere 0.4 and 0.8 g L−1, respectively. SC (8× MFC) reduced the incidence of disease in Ponkan fruits without compromising their quality. The results of CFW staining and extracellular alkaline phosphatase assays revealed that 1/2MIC SC for 30 min had no impact on the cell wall integrity ofP. digitatum. In contrast, 1/2MIC SC apparently destroyed cell membrane integrity, as shown by the increase in the content of reactive oxygen species (ROS), malondialdehyde, and H2O2. The addition of exogenous cysteine (Cys) or diphenyleneiodonium chloride (DPI) significantly mitigated the cytotoxic effects of SC. At the same time, mitochondrial membrane potential was significantly decreased by 1/2MIC SC, and the addition of exogenous Cys or DPI restored it to normal levels. In summary, the antifungal capacity of SC might be attributable to membrane damage inP. digitatumcaused by oxidative stress.

Perillaldehyde (PAE) is a green and safe active ingredient of natural plants. The inhibitory activity of PAE against the citrus postharvest pathogen Penicillium digitatum and its ability to control green mold were evaluated by in vitro and in vivo experiments. The results revealed that the minimum inhibitory concentration (MIC) and the minimum fungicidal concentration (MFC) of PAE against P. digitatum were 0.5 and 1 μL/mL, respectively. The results of conductivity, cell activity staining, and calcium fluoride white staining revealed that PAE damaged the membrane and cell wall integrity of P. digitatum . PAE promoted ROS accumulation in P. digitatum , as shown by 2',7'-Dichlorofluorescein diacetate (DCHF-DA) staining, which may induce cell death. To further investigate the molecular regulatory network of PAE in P. digitatum , we performed an RNA-sequencing analysis. The results displayed that differentially expressed genes were significantly enriched in the biogenetic pathway of ribosomes, and all of these genes were downregulated, suggesting that PAE treatment interfered with ribosome biogenesis in P. digitatum and inhibited the normal life processes of cells. Furthermore, PAE treatment reduced the incidence and severity of citrus green mold, and the overall control effect of the PAE treatment group with 5 × MFC was greater than that with 10 × MFC. Therefore, we determined the activity of enzymes related to fruit disease resistance. The results revealed that PAE treatment significantly increased the activities of CAT, POD, and PAL in citrus. These findings demonstrate that PAE prevents green mold disease by increasing the activity of citrus disease-resistant enzymes.

Blue mold caused by Penicillium italicum is one of the most devastating postharvest diseases in Citrus fruit. This work aims to determine the antifungal potential of ( E )-2-octenal, a component of plant essential oil, on the mycelial growth and disease incidence of a prochloraz-resistant strain of P. italicum isolated from an orchard in Luxi, Hunan Province, China. An in vitro assay revealed that the minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) of ( E )-2-octenal were 0.25 and 0.50 mL L −1 , respectively. An in vivo assay showed that wax + 4 × MFC ( E )-2-octenal treatment could reduce the blue mold incidence in Ponkan fruit inoculated with P. italicum without impairing fruit quality. Scanning electron microscopy analysis illustrated that the mycelia of P. italicum were anomalously twisted and shrunken upon treatment with ( E )-2-octenal. The primary antifungal targets were attributed to the cell membrane instead of the cell wall based on propidium iodide and calcofluor white staining results. In addition, ( E )-2-octenal induced massive accumulation of reactive oxygen species in hyphae in a dose-dependent manner, leading to oxidative stress and dysfunction of energy and respiration metabolism. At the same time, the activities of phosphofructokinase (PFK) and pyruvate kinase (PK) were decreased in ( E )-2-octenal-treated samples. Taken together, our present study suggested that ( E )-2-octenal could exert its antifungal activity against P. italicum by damaging the function of mitochondria and may be a potential antifungal substance for an alternative to chemical fungicides.

This study investigated the antifungal effect of curcumin-mediated photodynamic inactivation (PDI) with ascorbic acid (Vc) as a synergist on Penicillium italicum and its biofilms, and evaluated the efficacy in inactivating P. italicum on fresh-cut orange. Results showed that the number of P. italicum spores decreased by 4.61 Log CFU/mL when the concentration of curcumin, Vc and illumination time was 75 μmol/L, 250 mg/L and 20 min, respectively. After PDI treatment, the cell structure of P. italicum was destroyed and leakage of cell contents was observed. Mature P. italicum biofilms were formed after 72 h of incubation, and PDI could reduce the spores in biofilms by 3.23 Log CFU/mL. In addition, under the optimal photodynamic conditions, the number of P. italicum spores in fresh-cut orange decreased by 2.48 Log CFU/g, and the sensory quality was well maintained. In conclusion, curcumin-mediated photodynamic technology could better maintain the freshness of fresh-cut orange. Therefore, this study deepened the understanding of the antimicrobial efficacy of curcumin-mediated PDI on food borne pathogens and its impact on food quality.

Nuclease P1 (NP1) can hydrolyze nucleic acids into four 5′-mononucleotides, which are widely used in the pharmaceutical and food industries. In this paper, an aqueous two-phase system (ATPS) was developed to purify NP1 from Penicillium citrinum . Polyethylene glycol (PEG) and nucleotides salts were studied to form ATPSs, among which PEG3000/disodium guanosine monophosphate (GMPNa 2 ) was researched, including the phase composition and pH. Using 14% (w/w) PEG3000 and 20% (w/w) GMPNa 2 ATPS at pH 5.0, the best recovery and purification factor, 82.4% and 3.59, were obtained. The recovery of NP1 was 98.3% by the separation of ultrafiltration from the PEG-rich phase. The recycling use of GMPNa 2 was also studied, and 95.1% of GMPNa 2 in the salt-rich phase was obtained with the addition of ethanol as the solvent. These results showed that the ATPS was effective for purification of NP1.
