Electrospun membranes are attracting interest as a drug delivery system because of their material composition flexibility and versatile drug loading. In this study, the electrospun membrane was loaded with doxorubicin (DOX) via electrostatic adsorption for long-term drug delivery. DOX loading process was optimized by varying temperature, time, drug concentration, pH and ionic strength of solutions. The loading process did not impair the structural properties of the membrane. Next, we investigated the drug release kinetics using spectroscopic techniques. The composite membranes released 22% of the adsorbed DOX over the first 48 h, followed by a slower and sustained release over 4 weeks. The DOX release was sensitive to acidic solutions that the release rate at pH 6.0 was 1.27 times as that at pH 7.4. The DOX-loaded membranes were found to be cytotoxic to U-87 MG cells in vitro that decreased the cell viability from 82.92% to 25.49% from 24 to 72 h of co-incubation. These membranes showed strong efficacy in suppressing tumour growth in vivo in glioblastoma-bearing mice that decreased the tumour volume by 77.33% compared with blank membrane-treated group on Day 20. In conclusion, we have developed an effective approach to load DOX within a clinically approved poly (L-lactic acid)/gelatine membrane for local and long-term delivery of DOX for the treatment of glioblastoma.

To address the uneven nutrient distribution within three-dimensional (3D) tissue models and organoids currently used in medical research, this study introduces a microvascular network based on the Hilbert curve. Our aim was to develop innovative solutions for enhancing nutrient supply in thick tissue models in vitro. By using 3D bioprinting, we engineered microvascular networks of varying Hilbert orders and validated their efficacy in enhancing nutrient uniformity through numerical simulations and experiments. These networks facilitated broader and more uniform nutrient distribution throughout the thick tissue models, particularly the 2° Hilbert microvascular structure, which occupies less space and significantly reduces regions of cellular death. Furthermore, we explored the potential of assembling larger tissue constructs using the 2° Hilbert microvascular network, showcasing its applicability in constructing large-scale biological models. The findings suggest that the 2° Hilbert microvascular structure is particularly effective in ensuring adequate nutrient delivery, thus enhancing the viability and functionality of large-volume tissue models. These innovations hold significant promise for advancing the fields of tissue engineering and regenerative medicine by improving nutrient delivery to in vitro thick tissue block models. This provides a robust foundation for future in vitro research and clinical applications, potentially leading to more effective treatments and interventions in the medical field. The development of these microvascular networks represents a crucial step forward in overcoming the limitations of current 3D tissue models and organoids, paving the way for more sophisticated and reliable biomedical research tools.

Sequential administration and controlled release of different drugs are of vital importance for regulating cellular behaviors and tissue regeneration, which usually demands appropriate carriers like microspheres (MS) to control drugs releases. Electrospray has been proven an effective technique to prepare MS with uniform particle size and high drug-loading rate. In this study, we applied electrospray to simply and hierarchically fabricate sphere-in-sphere composite microspheres, with smaller poly(lactic-co-glycolic acid) MS (∼8–10 μm in diameter) embedded in a larger chitosan MS (∼250–300 μm in diameter). The scanning electron microscopy images revealed highly uniform MS that can be accurately controlled by adjusting the nozzle diameter or voltage. Two kinds of model drugs, bovine serum albumin and chlorhexidine acetate, were encapsulated in the microspheres. The fluorescence-labeled rhodamine-fluoresceine isothiocyanate (Rho-FITC) and ultraviolet (UV) spectrophotometry results suggested that loaded drugs got excellent distribution in microspheres, as well as sustained, slow release in vitro. In addition, far-UV circular dichroism and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) results indicated original secondary structure and molecular weight of drugs after electrospraying. Generally speaking, our research proposed a modified hierarchically electrospraying technique to prepare sphere-in-sphere composite MS with two different drugs loaded, which could be applied in sequential, multi-modality therapy.

Wound healing dressing is increasingly needed in clinical owing to the large quantity of skin damage annually. Excessive reactive oxygen species (ROS) produced through internal or external environmental influences can lead to lipid peroxidation, protein denaturation, and even DNA damage, and ultimately have harmful effects on cells. Aiming to sufficiently contact with the wound microenvironment and scavenge ROS, superabsorbent poly (acrylic acid) and antioxidant poly (ester amide) (PAA/PEA) hybrid hydrogel has been developed to enhance wound healing. The physical and chemical properties of hybrid hydrogels were studied by Fourier-transform infrared (FTIR) absorption spectrum, compression, swelling, degradation, etc. Besides, the antioxidant properties of hybrid hydrogels can be investigated through the free radical scavenging experiment, and corresponding antioxidant indicators have been tested at the cellular level. Hybrid hydrogel scaffolds supported the proliferation of human umbilical vein endothelial cells and fibroblasts, as well as accelerated angiogenesis and skin regeneration in wounds. The healing properties of wounds in vivo were further assessed on mouse skin wounds. Results showed that PAA/PEA hybrid hydrogel scaffolds significantly accelerated the wound healing process through enhancing granulation formation and re-epithelialization. In summary, these superabsorbent and antioxidative hybrid hydrogels could be served as an excellent wound dressing for full-thickness wound healing.

Infected bone defects present a significant clinical challenge, as persistent bacterial burden and exacerbated inflammatory response induce microenvironmental acidification, which severely compromises tissue regeneration. Therefore, this study aims to construct a smart platform to regulate the local microenvironment and achieve the therapeutic effect of bone regeneration. Here, pitaya‑inspired compartmentalized microspheres (IBPR‑CuTA) were developed to provide spatiotemporally controlled pH‑responsive release of Cu 2+ , antibiotic rifampicin (RF) and natural extract icariin (ICA), coupling infection control with tissue regeneration. Specifically, under infected inflammation, the microspheres rapidly release Cu 2+ and RF, which facilitates early anti-bacterial and anti-inflammatory effects and promotes pH normalization. This is succeeded by a delayed release of remaining Cu 2+ to foster angiogenesis, and a subsequent sustained release of encapsulated RF and ICA, collectively providing long-term inhibition of infection recurrence and promotion of bone regeneration. Mechanistically, IBPR‑CuTA was associated with suppressed glucose metabolism-related signaling and TNF/NF‑κB activation, and induced a shift from pro‑inflammatory M1 to reparative M2 polarization. In vivo , IBPR‑CuTA concurrently eradicated infection and accelerated vascularized bone regeneration in an infected defect model. Collectively, these results demonstrate an immunometabolically guided, spatiotemporally programmed microsphere platform that integrates early bactericidal/antioxidant activity with sustained pro‑regenerative cues, highlighting translational potential for managing complex infected bone defects.

Oxidative stress is a central pathological driver in diabetic nephropathy (DN), leading to excessive reactive oxygen species (ROS) generation, chronic inflammation and progressive renal fibrosis. Effective modulation of oxidative imbalance thus represents a vital therapeutic strategy for mitigating DN progression. Here, we developed a bioinspired polyphenolic nanoplatform by first constructing polymerized grape seed polyphenol nanoparticles via enzymatic polymerization, followed by supramolecular assembly with the bioactive polyphenol resveratrol. This design combined the intrinsic bioactivity of polyphenols with active resveratrol integration, yielding a stable and biocompatible strategy that alleviated DN and enabled superior restoration of renal structure and function. In diabetic models, it promoted coordinated recovery of renal morphology, key structural proteins and physiological function, suggesting potential advantages for long-term renal protection beyond short-term biochemical improvement. These findings demonstrated that enzymatically engineered polyphenolic nanotherapeutics provided a potent and biocompatible approach for oxidative regulation and functional renal restoration in DN therapy.

The escalating global prevalence of obesity and its associated metabolic disorders now represent one of the most urgent public health crises. Despite the pressing need for safe and effective anti-obesity interventions, current therapeutic options are often limited by suboptimal efficacy and various side effects. Herein, we present a microneedle (MN)-based drug delivery platform, termed RES@hmPDA-Dpep/MN, for photothermal browning therapy (PBT) to combat obesity. This system integrates hemispherical mesoporous polydopamine (hmPDA) nanoparticles with resveratrol (RES) and an adipose-homing peptide (Dpep) for localized PBT. Under identical conditions, hmPDA maintains photothermal performance comparable to spherical mPDA, while exhibiting advantageous drug-loading behavior particularly under higher drug-feeding conditions. In addition, the platform supports localized delivery with NIR-triggered, on-demand RES release and thermogenic activation. In vivo studies demonstrate that the RES@hmPDA-Dpep/MN system effectively suppresses WAT expansion, reduces body weight by 16.1% ± 2.5%, and significantly improves obesity-associated metabolic dysfunctions, including hyperlipidemia and hepatic steatosis. Mechanistic analyses reveal marked upregulation of UCP1 and other key browning genes. Furthermore, the nanoplatform shows favorable in vitro biocompatibility and hemocompatibility, supporting its potential for further translational evaluation. Collectively, this PBT strategy may serve as a potential localized intervention for fat reduction and systemic metabolic improvement in the management of obesity and related metabolic disorders.

Soft tissue defects represent a major challenge in plastic surgery, for which autologous fat grafting remains the standard treatment. However, in the early post-transplant period, the grafted fat tissue undergoes ischemia and hypoxia, leading to low and markedly unpredictable survival rates that consequently compromise the aesthetic outcomes. Nitric oxide (NO), an endogenous gasotransmitter, play a vital role in mediating angiogenesis and vascular remodeling. Therefore, we developed a near-infrared (NIR)-responsive hydrogel that releases NO on demand to promote adipocytes vascularization and thereby improve graft retention and enable precise volumetric filling. To ensure high-filling, the platform incorporates a photosensitive Gelatin Methacryloyl (GelMA) hydrogel inspired by the rapid solidification of “Frost Formation”. The hydrogel can be precisely delivered to the target area and photocrosslinked in situ to form a three-dimensional network that supports cell growth. Notably, this photocrosslinking process does not require ultraviolet (UV) irradiation, thereby eliminating the associated risks and significantly greatly enhancing the safety of adipose remodeling and filling technologies. Experimental studies conducted both in vitro and in vivo have validated that this hydrogel platform ensures precise grafting, promotes graft integration, and significantly improves the long-term survival of transplanted adipose tissue.

Cardiovascular diseases have become one of the major threats to human health. There is an urgent clinical need for small-diameter vascular grafts to achieve long-term implantation in the body. In previous studies, some progress has been made, but the prognosis is still poor. Heparin (HEP) is an anticoagulant that is widely applied in the clinical field and commonly used for anticoagulation modification of biomaterials. However, there is an important problem: HEP has almost no direct inhibitory effect on platelet adhesion, which may become one of the potential dangers after graft implantation. In recent years, it has been found that calcification is also a key factor affecting the long-term patency of small-diameter vascular grafts, which is often ignored. Therefore, in this study, based on HEP, procyanidin (PC) was introduced to modify the thermoplastic polyurethane (TPU) grafts fabricated by electrospinning to enhance the anticoagulation and anticalcification properties simultaneously. To verify the effects of drug modification on the material, the physical and chemical characterization, biocompatibility experiments and effectiveness tests of this vascular graft were conducted. The results indicated that the HEP-PC grafts could not only inhibit the excessive proliferation of human umbilical artery smooth muscle cells to some extent but also have excellent hemocompatibility. Particularly, in vivo experiments showed that this graft also achieved dual anticoagulation and delayed the calcification after implantation. These results suggested that HEP-PC-modified TPU might be a promising candidate for inhibiting intimal hyperplasia and anticalcification of small-diameter grafts.

Myocardial Infarction (MI) is a leading cause of high global cardiovascular mortality and disability. Platelet-rich plasma (PRP), as a widely available autologous biological product rich in various growth factors, holds significant potential for promoting tissue healing and regeneration. However, its clinical application is limited by short in vivo retention time and transient therapeutic effects, necessitating the development of novel drug delivery systems to enhance its clinical value. In this study, a novel injectable PRP-gel composite was developed. Based on the Schiff base reaction, the composite gel exhibits excellent biocompatibility, biodegradability, appropriate mechanical properties, and enables controlled release of multiple growth factors from PRP. Results at 28 days post-intramyocardial injection demonstrated that, compared with the control group, the PRP-gel composite effectively promoted angiogenesis, inhibited cardiac cell apoptosis and injury, significantly reduced infarct size, decreased myocardial fibrosis, and preserved ventricular wall thickness, ultimately leading to a marked improvement in cardiac function. In summary, the composite gel developed in this study provides an optimized strategy for the clinical application of PRP and represents a promising therapeutic approach for myocardial regeneration following MI.

The healing of infected wounds is a formidable clinical challenge that demands advanced biomaterials capable of simultaneously eradicating pathogens and orchestrating the complex regenerative process. Herein, we engineered an immunomodulatory and angiogenic multifunctional hydrogel dressing by integrating choline phosphorylated chitosan (CS-MCP) with tetracycline-loaded polydopamine nanoparticles (PDA@TH NPs) for the programmed healing of infected wounds. This composite hydrogel demonstrates on-demand, spatiotemporally controlled therapeutic responses. Under near-infrared (NIR) irradiation, it enables synergistic bactericidal activity through photothermal effects and triggered antibiotic release. Moreover, the system exhibits pH-responsive drug release behavior, specifically targeting the acidic microenvironment of infected tissues. Beyond its potent antibacterial function, the hydrogel actively promotes regenerative processes. In vitro, CS-MCP markedly enhanced the adhesion, proliferation, and tube formation of human umbilical vein endothelial cells, demonstrating potent pro-angiogenic effects. Furthermore, the polydopamine nanoparticles effectively scavenged reactive oxygen species (ROS), attenuating oxidative stress and inducing M2 macrophage polarization to foster an immunoregulatory microenvironment conducive to tissue repair. In a rat model of Staphylococcus aureus-infected full-thickness skin defects, the hydrogel significantly accelerated wound healing by comprehensively modulating the entire regeneration cascade: eliminating infection, mitigating inflammation, promoting angiogenesis, and enhancing collagen deposition. This study presents a novel immune-engaging and pro-regenerative strategy, representing a highly promising platform for the treatment of refractory infected wounds.

Bioadhesives are being increasingly used in clinical practice, but pose significant health risks due to their uncontrollable intraoperative leakage. Nevertheless, there is currently a critical lack of effective solutions. Herein, we propose a second near-infrared window (NIR-II) fluorescence-visible indocyanine green (ICG)-BioGlue adhesive for precision surgical adhesion with intraoperative leakage prevention. The fabricated ICG-BioGlue adhesive based on the non-covalent interaction exhibits nearly 100% fluorescence labeling efficiency and maintains long-term stable NIR-II fluorescence for up to 3 months. During surgery, ICG-BioGlue adhesive enables real-time visualization of wound adhesion in various fields such as aortic dissection adhesion and liver or kidney wound hemostasis, preventing leakage into surrounding tissues. Moreover, the stable fluorescence of ICG-BioGlue adhesive allows effective imaging-guided removal of long-term adhesive fragments in the body when they cause compression symptoms after surgery. Considering it consists solely of Food and Drug Administration-approved drugs ICG and BioGlue, with a straightforward synthesis method and versatile applicability, ICG-BioGlue adhesive holds excellent potential for clinical translation. Our study provides a leakage-preventing strategy to enhance biosafety and promote the widespread application of bioadhesives in clinical settings.
