Oat beta-glucan has been reported to be beneficial to glucose and lipid metabolism in human and animal studies. However, weight-loss was shown only in animal studies, whereas human studies reported an improvement in hyperlipidemia. This study investigated the effects of oat beta-glucan on obesity and related metabolic disturbances at the doses based on the Chinese recommended daily intake of 100 g of whole grains which contain approximately 10 % β-glucan. Our data showed that 200 mg/kg efficiently blocked the process of obesity as determined by significantly decreased body weight (7.57 g) and adipose tissue mass (5.24 g) after the 4-week intervention. Lower serum low-density lipoprotein cholesterol (0.32 mmol/L) and reduced atherosclerosis were also observed. Oat beta-glucan improved hepatic lipid metabolism at the low dose (100 mg/kg) and restored it to normal levels in lean mice at the high dose. To our knowledge, our study is the first to analyze the effects of oat beta-glucan on adipose stem cells and reveal the anti-obesity mechanism of oat β-glucan is modulating the adipose stem cell microenvironment. This study determined an efficient supplementary dose for blocking high-fat diet-induced obesity, which not only improved lipid metabolism but also altered stem cell composition in adipose tissue.

The purpose of this study was to permit bone marrow mesenchymal stem cells (BMSCs) to reach their full potential in the treatment of chronic wounds. A biocompatible multifunctional crosslinker based temperature sensitive hydrogel was developed to deliver BMSCs, which improve the chronic inflammation microenvironments of wounds. A detailed in vitro investigation found that the hydrogel is suitable for BMSC encapsulation and can promote BMSC secretion of TGF-β1 and bFGF. In vivo, full-thickness skin defects were made on the backs of db/db mice to mimic diabetic ulcers. It was revealed that the hydrogel can inhibit pro-inflammatory M1 macrophage expression. After hydrogel association with BMSCs treated the wound, significantly greater wound contraction was observed in the hydrogel + BMSCs group. Histology and immunohistochemistry results confirmed that this treatment contributed to the rapid healing of diabetic skin wounds by promoting granulation tissue formation, angiogenesis, extracellular matrix secretion, wound contraction, and re-epithelialization. These results show that a hydrogel laden with BMSCs may be a promising therapeutic strategy for the management of diabetic ulcers.

The age-associated decline in intestinal stem cell (ISC) function is a key factor in intestinal aging in organisms, resulting in impaired intestinal function and increased susceptibility to age-related diseases. Consequently, it is imperative to develop effective therapeutic strategies to prevent ISC aging and functional decline. In this study, we utilized an aging Drosophila model screening of amino acids and found that asparagine (Asn), a nonessential amino acid in vivo, exhibits its profound anti-aging properties on ISCs. Asn inhibits the hyperproliferation of aging ISCs in Drosophila , maintains intestinal homeostasis, and extends the lifespan of aging flies. Complementarily, Asn promotes the growth and branching of elderly murine intestinal organoids, indicating its anti-aging capacity to enhance ISC function. Mechanistic analyses have revealed that Asn exerts its effects via the activation of the autophagic signaling pathway. In summary, this study has preliminarily explored the potential supportive role of Asn in ameliorating intestinal aging, providing a foundation for further research into therapeutic interventions targeting age-related intestinal dysfunction.

The liver is known to play a pivotal role in modulating blood glucose homeostasis through intrahepatic glucose metabolism. Here, we reveal a unique mechanism by which fatty liver exacerbates hyperglycemia through remote communication from hepatocytes to intestinal stem cells (ISCs), independent of enhanced intrahepatic gluconeogenesis. Mechanistically, hepatocyte-derived alkaline phosphatase (ALP) targets α2δ-1 in ISCs to promote the membrane translocation of Ca v 1.2. This process triggers increased intracellular calcium levels, which subsequently activates the calcineurin/NFATC2 signaling axis, thereby inhibiting SOX21 expression. Then, decreased expression of SOX21 downregulated bone morphogenetic protein 7 (BMP7), ultimately hindering ISCs differentiation into intestinal L-cells. Consequently, the levels of hypoglycemic enteroendocrine hormones secreted by L-cells are decreased, thereby promoting hyperglycemia. Therapeutically, inhibiting ALP synthesis in fatty liver independently reduces blood glucose and synergistically enhances the hypoglycemic effect of metformin. Our study highlights the role of liver-gut communication in regulating the fate of ISC differentiation and blood glucose homeostasis.

Introduction Glioblastoma, the most aggressive form of brain tumor, continues to present significant therapeutic challenges, including the limited delivery of drugs posed by the blood-brain barrier (BBB) and the blood-brain tumor barrier (BBTB), severe systemic toxicity associated with conventional chemotherapy, and the complexity arising from tumor heterogeneity. Methods To overcome these challenges, this study developed a novel biomimetic drug delivery system. Specifically, we prepared poly(lactic-co-glycolic acid) (PLGA) nanoparticles co-loaded with the chemotherapeutic agent doxorubicin (DOX) and the natural polyphenol curcumin (CUR), and subsequently functionalized them with the membrane of human umbilical cord mesenchymal stem cells (hUC-MSCs), which possess inherent tumor-homing capability. Results In vitro studies demonstrated that the hUC-MSCs membrane coating significantly enhanced targeted recognition and cellular uptake by glioblastoma cells, and the biomimetic nanoplatform exhibited superior synergistic cytotoxicity and induced greater cellular apoptosis compared to free drug combinations and uncoated nanoparticles. Antitumor mechanism analysis indicated that biomimetic nanoplatform inhibited glioblastoma migration, invasion, and angiogenesis. In vivo anti-tumor efficacy studies showed that the biomimetic nanoparticles effectively suppressed the growth of tumor. Notably, CUR contributed to the system by amplifying the anticancer activity of DOX and alleviating its associated toxicity.Discussion This work demonstrates that hUC-MSC membrane-camouflaged PLGA nanoparticles enable successful co-delivery of DOX and CUR, offering a promising strategy to address the critical barriers of delivery and toxicity in GBM chemotherapy, supported by their excellent in vitro targeting, in vivo anti-tumor efficacy, and reduced toxicity profile.

Mesenchymal stem cells (MSCs) are one of the most promising cell populations for tissue engineering and regenerative medicine. Of utmost importance to MSC research is identification of MSC sources that are easily obtainable and stable. Several studies have shown that MSCs can be isolated from amniotic fluid. The sheep is one of the main types of farm animal, and it has many biophysical and biochemical similarities to humans. Here, we obtained MSCs from ovine amniotic fluid and determined the expansion capacity, surface and intracellular marker expression, karyotype, and multilineage differentiation ability of these ovine amniotic fluid mesenchymal stem cells (oAF-MSCs). Moreover, expression levels of differentiation markers were measured using reverse transcription-qPCR (RT-qPCR). Our phenotypic analysis shows that the isolated oAF-MSCs are indeed MSCs.

Embryonic stem cells (ESC) are pluripotent, with the potential to differentiate into multiple cell types, making them a valuable tool for regenerative medicine and disease therapy. However, common culture methods face challenges, including strict operating procedures and high costs. Currently, Leukemia inhibitory factor (LIF), an indispensable bioactive protein for ESC culture, is typically applied to maintain self-renewal and pluripotency, but its instability and high cost limit its effectiveness in stable culture conditions. Hence, we have developed an innovative strategy using a soluble nanomaterial, metal-organic polyhedra (MOPs), to effectively maintain the self-renewal and pluripotency of ESC. The selected amino-modified vanadium-based MOP not only exhibits excellent biocompatibility and high stability but also possesses similar or even superior biological functions compared to commercial LIF. Due to the precise structure of MOPs, the active site responsible for maintaining ESC pluripotency has been identified and regulated at the molecular level. The new ESC culture method significantly reduces costs, simplifies preparation, and enhances the practicality of biopharmaceutical preparation and storage. This represents the first case of using MOPs to maintain self-renewal of ECS, opening an avenue for introducing advanced materials into the development of innovative ESC culture methods.

Due to the ability to combine multiple osteogenic induction “cues” at the same time, hydrogels are widely used in the three-dimensional (3D) culture of human mesenchymal stem cells (hMSCs) and osteoinduction. However, the survival and proliferation of stem cells in a 3D culture system are limited, which reduces their osteogenic differentiation efficiency. In addition, the cells inside the hydrogel are prone to apoptosis due to hypoxia, which is a serious challenge for tissue engineering based on stem cells. In this study, a tripeptide-based macroporous alginate hydrogel was prepared to improve the osteogenic microenvironment of stem cells. The arginine–glycine–aspartate (RGD) peptide promoted the adhesion and proliferation of stem cells, and the degradation of gelatin microspheres (GMs) produced a macroporous structure to enhance further the migration and aggregation of stem cells. Mesoporous silica nanoparticles (MSNs) sustained-release bone-forming peptide-1 (BFP-1) induced osteogenic differentiation, and the sustained release of the QK peptide from the GMs promoted angiogenesis. In vitro experiments have shown that this functionalized hydrogel stimulates the proliferation of hMSCs, encourages larger cell cluster formation, and enhances the osteogenic differentiation efficiency. The released QK facilitates the proliferation and migration of endothelial cells. In vivo experiments have also verified that this system has a better osteogenic effect, and more blood vessels were observed inside the hydrogel, than in other systems. In general, this research has led to the development of a tripeptide macroporous hydrogel that can simultaneously promote osteogenesis and angiogenesis, showing great promise for applications of 3D cultures and stem cell-based tissue engineering.

Background: Mesenchymal stem cells (MSCs) have strong self-renewal ability and multiple differentiation potential. Some studies confirmed that spreading shape and area of single MSCs influence cell differentiation, but few studies focused on the effect of the circularity of cell shape on the osteogenic differentiation of MSCs with a confined area during osteogenic process. Methods: In the present study, MSCs were seeded on a micropatterned island with a spreading area lower than that of a freely spreading area. The patterns had circularities of 1.0 or 0.4, respectively, and areas of 314, 628, or 1256 μm 2 . After the cells were grown on a micropatterned surface for 1 or 3 days, cell apoptosis and F-actin were stained and analyzed. In addition, the expression of β-catenin and three osteogenic differentiation markers were immunofluorescently stained and analyzed, respectively. Results: Of these MSCs, the ones with star-like shapes and large areas promoted the expression of osteogenic differentiation markers and the survival of cells. The expression of F-actin and its cytosolic distribution or orientation also correlated with the spreading shape and area. When actin polymerization was inhibited by cytochalasin D, the shape-regulated differentiation and apoptosis of MSCs with the confined spreading area were abolished. Conclusion: This study demonstrated that a spreading shape of low circularity and a larger spreading area are beneficial to the survival and osteogenic differentiation of individual MSCs, which may be regulated through the cytosolic expression and distribution of F-actin.

Imitating the physiological microenvironment of living cell and tissues opens new avenues of research into the application of electricity to medical therapies. In this study, dynamic piezoelectric stimulation is generated in a dynamic culture because of the piezoelectric effect of the poly(vinylidene fluoride)–polypyrrole (PVDF–PPy) electroactive composite. Combined with PPy nanocones, dynamic piezoelectric signals are effectively and continuously provided to cells. In the presence of dynamic piezoelectric stimulation and PPy nanocones, PPy-PVDF NS samples show promoted bone mesenchymal stem cell (BMSCs) adhesion, spreadin, and osteogenic differentiation. On the basis of the results of this study, PPy nanocones and dynamic piezoelectric stimulation can be administered to modulate cell behavior, paving the way for the exploration of cellular responses to dynamic electrical stimulation.

The mechanical environment of extracellular matrix (ECM) plays an important role in adjusting the behaviors of cells. Natural ECM are highly viscoelastic materials with stress-relaxion behavior. Hydrogel is considered as a promising and attractive material for cell carrier, but they are typically elastic serving as synthetic ECM. Double-network (DN) hydrogel has an interpenetrating network of special structure combining the advantages of both rigid and ductile components, due to which the mechanical properties of the system can be very different from that of the single-network ones, and some special biological properties can be obtained. In this study, GG/PEGDA DN hydrogel was prepared by combining gellan gum (GG) with polyethylene glycol diacrylate (PEGDA), and then the influence of the two individual networks on the viscoelasticity of the system were investigated. Furthermore, the effects of viscoelasticity of GG/PEGDA DN hydrogel on the biological behavior of bone mesenchymal stem cells (BMSCs) were explored in vitro and in vivo . The results indicate that the spreading of BMSCs was closely related to the relaxation behavior of the hydrogels. GG/PEGDA DN hydrogel shows excellent mechanical and relaxation properties which provide a favorable physical environment for cell proliferation and spreading, and induce chondrogenic differentiation. Our study demonstrates that this DN hydrogel has bright prospects in the fields of cell carrier and cartilage tissue engineering.

Understanding the shape effect of hydroxyapatite (HAp) microparticles on cellular behavior is important for enabling new kinds of biological and biomedical applications. However, it is still a challenge to prepare HAp microparticles with different shapes but similar physicochemical properties, and then to investigate their relationships with cellular behavior. Herein, we developed a novel, facile route to regulate the morphology of HAp microparticles, and investigated the interaction between the particles and bone marrow mesenchymal stem cells (BMSCs). Our results revealed that the shape of HAp has a strong influence on cellular behavior, and that the sphere-like particles performed better than the rod-like particles. These findings highlight the importance of the shape characteristics of HAp microparticles, and may provide new insights for the utility of HAp-based materials.
