d-tryptophan potentiates dielectric barrier discharge cold plasma inactivation of Shewanella baltica through selective singlet oxygen generation

Jian Chen,Shiya Guo,Huan Li,Shigenobu Koseki,Jianhao Zhang,Xiaowei Sheng,Yanbo Wang

Dielectric barrier discharge (DBD) cold plasma combined with d -tryptophan ( d -Trp) exhibited a pronounced synergistic inactivation effect against Shewanella baltica , a dominant spoilage bacterium in aquatic products, and the underlying reactive oxygen species (ROS)-mediated mechanism was elucidated. 40 s combined treatment (DBD + d -Trp) achieved a 7.31 log CFU/mL reduction, markedly exceeding the 1.3 log reduction obtained with DBD treatment alone. Physicochemical analysis showed elevated hydrogen peroxide and nitrite levels accompanied by decreased pH in the synergistic system. Electron paramagnetic resonance and radical scavenging assays revealed that d -Trp selectively enhanced singlet oxygen ( 1 O 2 ) generation while suppressing hydroxyl radicals and superoxide anions, thereby reshaping the ROS profile compared to DBD treatment alone. Severe cellular damage was observed by scanning electron microscopy, and increased alkaline phosphatase release, potassium leakage, and membrane depolarization confirmed disruption of the cell envelope integrity. Elevated intracellular ROS accumulation and dysregulation of antioxidant enzyme activity indicated aggravated oxidative stress leading to bacterial death. The synergistic inactivation effect was further validated in fish juice, demonstrating superior efficacy compared to individual treatments. These findings demonstrate that d -Trp potentiates plasma inactivation through selective modulation of ROS composition and provide a mechanistic foundation for developing targeted control strategies against specific spoilage bacteria in aquatic products.

Biomineralized Shewanella Exploits Tumor Lactate Metabolism via Biological Nanowires for Radiotherapy-Synergized Bioelectric Ablation and Systemic Immunity

Mengen Guo,Wenjie Xu,Pengcheng Sun,Jiayi Liu,Xinyan Hao,Muyan Li,Hai Huang,Dehua Liao,Jiangshan Zhao,Tiantian Tang,Jianing Lin,Ruiqi Chen,Daxiong Xiang,Junyong Wu

Bioelectronic therapy is an emerging strategy that ablates tumor cells via electrical signals. However, achieving precisely controllable bioelectronic ablation remains challenging. Here, we report a living bioelectronic platform, termed PSDT, for radiotherapy (RT)-activated tumor intervention and immune activation. PSDT consists of a Pd/Pt-modified Shewanella core (SDT) encapsulated within a ROS-responsive polymeric shell (PEI-Se-Se-PEG). The ROS-responsive shell keeps the platform inert systemically and confines its activity to irradiated tumors, achieving spatiotemporal control. Upon RT activation, the released SDT core utilizes the anaerobic characteristics of the tumor microenvironment to specifically construct nanowire connections with tumor cells, thereby establishing the electronic synapse for current delivery. Simultaneously, the Pd/Pt bio-alloy enhances Shewanella 's lactate metabolism and electron transfer. This enhancement enables the platform to generate a continuous bioelectric current fueled by tumor lactate. RT also induces the Pd/Pt bio-alloy to release high-energy electrons, which are then delivered to the tumor cells alongside the amplified bioelectric current through the nanowire. This dual-pathway electron delivery enhances tumor cell ablation and yields prominent inhibition of primary tumor growth. The treatment also induces systemic antitumor effects, as the release of tumor antigens from electro-ablation and bacterial antigens from RT-mediated bacterial disruption triggeres local immune activation and an abscopal effect. Moreover, the combination with anti-PD-1 treatment further enhances these effects, resulting in nearly complete tumor eradication and durable tumor control. This work demonstrates a bioelectronic therapy that integrates tumor metabolic intervention with systemic immune activation for the treatment of immunologically cold solid tumors.

Antibacterial mechanism of ultrasound combined with thymol against Shewanella putrefaciens: implications for cell integrity and biofilm control

Yongji Gu,Weiqing Lan,Jing Xie

Summary The study aimed to assess the in vitro antibacterial effect of thymol (Thy) in conjunction with ultrasound (US) on Shewanella putrefaciens ( S. putrefaciens ). The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of Thy against S. putrefaciens were 0.50 and 1.00 mg mL −1 , respectively. US was found to augment the antibacterial efficacy of Thy, resulting in a significant reduction of 2.36 log CFU mL −1 in the total colony count of S. putrefaciens , ensuring that the bacterial population maintained a consistently low growth rate. Quantitative detection of crystal violet revealed that the combined treatment exhibited a pronounced inhibitory effect on biofilm formation, with a clearance rate of mature biofilms reaching 91.60%. Alkaline phosphatase (AKPase), lactate dehydrogenase (LDHase), UV absorbing substance permeability, and other indicators were employed to measure changes in the bacterial cell membrane and internal environment. Scanning electron microscopy (SEM) was further used to observe the bacteria morphology before and after treated by US and Thy, a notable collapse of bacterial cells was observed, accompanied by ruptured outer membranes, damaged cell walls, and extensive leakage of cellular contents. This study presents a novel approach for controlling bacterial growth in food preservation methods.

Shewanella oneidensis Metabolically Engineered with Prussian Blue for Synergistic Tumor Microenvironment Remodeling and Photodynamic Breast Tumor Elimination

Qilong Yan,Xian Yu,Jinming Wei,Chao Fang,Junwei Zhao,Xiang Li,Yike Fu,Xiang Li

Microorganism-based tumor therapy has attracted increasing interest due to the intrinsic tumor tropism and metabolic plasticity of certain bacterial species. However, their therapeutic efficacy is often hindered by insufficient metabolic activity within the tumor microenvironment and safety concerns associated with systemic administration. Here, we develop a biohybrid system, termed Ce6@PB@MR-1, to enhance the intrinsic tumor-targeting behavior of chlorin e6 (Ce6) and lactate-metabolizing capability of Shewanella oneidensis MR-1 (MR-1) through engineered material–microbe coupling. In this system, Prussian blue (PB) is metabolically precipitated onto the MR-1 surface during anaerobic respiration, forming an electron-mediating interface in which PB functions as an efficient exogenous electron acceptor to strengthen respiratory electron flux and accelerate the lactate metabolism of MR-1. The PB coating simultaneously attenuates bacterial immunogenicity, enabling the improved in vivo persistence and safety of MR-1. Furthermore, bioconjugated Ce6 enables 660 nm-triggered photodynamic therapy to induce immunogenic cell death and activate antitumor immune responses. In vivo, the biohybrid system achieves durable tumor eradication accompanied by a marked remodeling of the tumor microenvironment. This work establishes a metabolically assisted material–microbe hybridization framework that expands microbial functionality and offers a versatile strategy for developing safe and effective microbe-based cancer therapies.

Screening and preservation application of quorum sensing inhibitors of Pseudomonas fluorescens and Shewanella baltica in seafood products

Yanbo Wang,Yongzheng Wang,Jian Chen,Shigenobu Koseki,Qing Yang,Hejun Yu,Linglin Fu

Pseudomonas fluorescens and Shewanella baltica are specific spoilage organisms (SSOs) of most seafood products . Both of them can cell-to-cell communicate through quorum sensing (QS), facilitating the spoilage activities such as protease secretion, flagellar movement, and biofilm formation. Thus, it is important to screen the quorum sensing inhibitors (QSI) of P. fluorescens and S. baltica. In this study, we tried to apply a high-throughput virtual screening technology to select QSIs candidates. Three amino acid sequences from the key QS regulator–LuxR/I family (the LuxR and LuxI proteins of P. fluorescens, and the LuxR of S. baltica ) were targeted and analyzed . Their 3D structures were subsequently constructed for the QSIs candidates' screen of both P. fluorescens and S. baltica . The candidates of cytidine-5′-monophosphate (5′-CMP) and 5′-adenylic acid (5′-AMP) with high scores exerted strong antagonistic effects against both P. fluorescens and S. baltica QS. The addition of 5′-CMP and 5′-AMP also inhibit QS-controlled phenotypes (swimming motility, protease activity, EPS, and biofilm formation) in P. fluorescens and S. baltica . Furthermore, the exogenous addition of 5′-CMP and 5′-AMP could retard the spoilage process of salmon slices during low-temperature storage by inhibiting the production of extracellular protease, siderophores, and Total Volatile Basic Nitrogen (TVB-N).

An excellent anaerobic respiration mode for chitin degradation by Shewanella oneidensis MR-1 in microbial fuel cells

Shan-Wei Li,Raymond J. Zeng,Guo-Ping Sheng

Shewanella oneidensis MR-1, a model electroactive bacterium, can oxidize a variety of fermentative organics and utilize an electrode as an electron acceptor for anaerobic respiration. Chitin as the main component of food wastes (shrimp and crab shells) is hardly degraded for bioenergy generation. This study demonstrated that S. oneidensis MR-1 could degrade chitin in microbial fuel cell (MFC). The metabolites during chitin degradation in MFC were succinate, lactate, acetate, formate, and ethanol. Their concentrations produced in MFC were higher than those in the fermentation system, as well as additional electricity could be recovered in MFC. Furthermore, the degradation of GlcNAc (the intermediate of chitin hydrolysis) by S. oneidensis MR-1 was faster than chitin in MFC and fermentation systems. Moreover, the mechanism of enhanced chitin degradation in MFC was speculated. This work might provide a new insight for biomass treatment and energy recovery by S. oneidensis MR-1 in MFC.

Electron shuttles enhance the degradation of sulfamethoxazole coupled with Fe(III) reduction by Shewanella oneidensis MR-1

Chen Zhou,Huiqing Wang,Youbin Si,Kang Wu,Amina Yousaf

The ability of anthraquinone-2,6-disulfonate (AQDS) and riboflavin to enhance the sulfamethoxazole (SMX) degradation coupled with the Fe(III) reduction by Shewanella oneidensis MR-1 was investigated. The results indicated that the SMX degradation rate was 38.5% with an initial SMX concentration at 0.04 mM. For the overall performance of AQDS and riboflavin mediated SMX degradation and iron reduction, the SMX degradation rate was gradually increased with the enhancement of iron reduction. Riboflavin had a stronger enhancement on SMX degradation and iron reduction than AQDS, but the enhancement was not positively correlated with electron shuttles concentration. A quantitative characterization of the electron transfer capacity (ETC) of the electron shuttles showed that the ETC was higher for riboflavin than AQDS. The S. oneidensis MR-1 16S rRNA gene copies results indicated that electron shuttles had a positive effect on the microbial activity of S. oneidensis MR-1. The LC MS result indicated that the products of the SMX biodegradation were 3-amino-5-methylisoxazole and 4-aminobenzenesulfonic acid, which suggested that the SMX biodegradation was caused by S N bond cleavage. This study indicates that the biochemical mechanisms play a vital role in SMX transformation and Fe(II) generation in this system.

Changes of membrane fatty acids and proteins of Shewanella putrefaciens treated with cinnamon oil and gamma irradiation

Lyu Fei,Gao Fei,Wei Qianqian,Liu Lin

Background In order to detect the antimicrobial mechanism of combined treatment of cinnamon oil and gamma irradiation (GI), the membrane fatty acids and proteins characteristics of Shewanella putrefaciens ( S. putrefaciens ) treated with cinnamon oil and GI, and the distribution of cinnamon oil in S. putrefaciens were observed in this study. Results The membrane lipid profile of S. putrefaciens was notably damaged by treatments of cinnamon oil and the combination of cinnamon oil and GI, with significantly fatty acids decrease in C14:0, C16:0, C16:1, C17:1, C18:1 ( p  < 0.05). The SDS-PAGE result showed that GI did not have obvious effect on membrane proteins (MP), but GI combined with cinnamon oil changed the MP subunits. Cinnamaldehyde, the main component of cinnamon oil, can not transport into S. putrefaciens obviously. It was transformed into cinnamyl alcohol in the nutrient broth with the action of S. putrefaciens . This indicated that the antimicrobial action of cinnamon oil mainly happened on the membrane of S. putrefaciens. Conclusion Cinnamon oil could act on the membrane of S. putrefaciens with the damage of fatty acids and proteins, and GI would increase the destructive capability of cinnamon oil on the membrane fatty acids and proteins of S. putrefaciens .

Insight into the antibacterial activity and mechanism of allicin and antioxidant of bamboo leaves against Shewanella putrefaciens

Weiqing Lan,Zhe Shao,Jintao Du,Xuening Chen,Jing Xie

Summary The antibacterial activity and mechanism of allicin (All) and antioxidant of bamboo leaves (AOB) against Shewanella putrefaciens was studied. The minimum inhibitory concentration (MIC) and growth curve were used to evaluate the antibacterial activity of combining All and AOB against S. putrefaciens . The inactivation mechanism was reflected by the changes in Na + /K + -ATPase activity, confocal laser scanning microscopy (CLSM), and the leakage of nucleic acid and protein. Scanning electron microscopy (SEM) was used to observe the morphology of bacterial cells and crystal violet biofilm reflected the ability to inhibit the formation of biofilm. Results showed that after being treated by All and AOB, S. putrefaciens was effectively inactivated, and the combined treatment had the strongest inactivation effect. The cell wall and cell membrane were destroyed by All and AOB, resulting in the release of cell contents. The results of Na + /K + -ATPase activity demonstrated that the membrane permeability could be changed, and the cell structural proteins could be destroyed by All and AOB. SEM observation further proved the effects of All and AOB on bacterial morphology destructing. Therefore, the combination of All and AOB can be used as an efficient method for the preservation of aquatic products.

Fully Stretchable Microbial Fuel Cell with 75% Stretchability

Shizhe Peng,Jia Li,Yihan Hu,Jinwei Cao,Ming Zhou,Lu Lu,Yude Su

A decent stretchability is of paramount significance to operate microbial fuel cell (MFC) under mechanically dynamic conditions. However, it remains a grand challenge to fabricate fully stretchable MFC without compromising its power output. Here, using Shewanella oneidensis MR-1 ( S. oneidensis ) as the model electrogenic bacteria, the study demonstrates a fully stretchable MFC device that can operate with a stretchability of 75%. The design takes advantage of a stretchable and ion-conductive polyurethane membrane, which encapsulates the biohybrids composed of S. oneidensis and reduced graphene oxide (rGO) on the polydimethylsiloxane (PDMS) current collector for synchronous stretching. It is discovered that the “stretchable” living biohybrids can sustain an adaptive bio-current output under stretching/releasing stimulation. The design also employs a stretchable air cathode. The stabilized peak power density of the stretchable MFC follows an increasing trend with the applied strain, and reaches 5.0 ± 0.7, 5.9 ± 0.9, 6.2 ± 1.1, 6.6 ± 1.4 µW cm −2 at strains of 0%, 25%, 50%, and 75%, respectively (n = 3). At 75% strain, the stretchable MFC yields a maximum current output of 104 ± 27 µA cm −2 and an open-circuit voltage of 283 ± 30 mV (n = 3). The results provide insights to design stretchable MFCs to power the next-generation on-skin devices, soft robotics, and sustainable electronics.

Synergistic U(VI) immobilization by a ternary Shewanella putrefaciens–chlorite–humic acid system: coupled adsorption and bio-reduction

Wang Xinyu,Qin Yilin,Gong Junyuan,Zhang Guohao,Peng Yinyin,Sun Gen,Ji Yanhao,Huang Yaling,Li Yahui,Nie Xiaoqin

This study aimed to enhance U(VI) removal from water by constructing a ternary system of Shewanella putrefaciens ( S. putrefaciens ), chlorite, and humic acid (HA). The ternary system achieved significantly higher U(VI) immobilization than single or binary systems, owing to synergistic effects of adsorption enrichment, electron shuttling, and bioreduction. Under optimal conditions, the U(VI) removal efficiency reached 91.6%. Importantly, pre-forming the ternary composite before U(VI) addition avoided false removal caused by HA-U(VI) complexation, ensuring accurate assessment of reductive immobilization. This work provides a reliable strategy for uranium-contaminated water remediation. Graphical abstract

Common Divalent Cations Elicit Sublethal Effects of Negatively Charged Nanoplastics on Shewanella oneidensis by Compromising Membrane Protein Functionality

Siyu Zhang,Boyu Jia,Weilin Huang,Changdong Ke,Yuekang Chen,Yanping Deng,Guining Lu,Zhi Dang,Chuling Guo

The toxicity of negatively charged nanoplastics (NNP) to bacteria is generally subtler than that of positively charged counterparts, owing to limited NNP-cell interaction. This study hypothesized that common environmental cations (Na+, Mg2+, Ca2+) could enhance interaction between NNP and Shewanella oneidensis, thereby inducing biological effects. Settling experiments and dynamic light scattering analyses showed that NNP-cell interaction increased in the order of Ca2+ ≈ Mg2+ > Na+, which can be attributed to the decreased electrostatic repulsion, as confirmed by extended Derjaguin–Landau–Verwey–Overbeek theory calculations. Although coexposure to NNP and cations did not result in significant lethality, extracellular electron transfer (EET) to insoluble electron acceptors was significantly inhibited by coexposing to NNP and Ca2+ (NNP+Ca2+, by 37%) or Mg2+ (NNP+Mg2+, by 20%), but not by NNP alone or NNP and Na+ treatments. Two-dimensional correlation spectroscopy indicated that membrane proteins predominantly mediate bacterial interactions with NNP. Physical membrane damage and structural alterations of membrane proteins were observed following coexposure to NNP+Ca2+ and NNP+Mg2+, impairing the direct EET pathways. Transcriptomic and physiological analyses further revealed that NNP+Ca2+ upregulated persister marker genes (spoT, ppx, relA) and induced ATP depletion, triggering cellular dormancy and suppressing membrane protein-mediated processes. By contrast, NNP+Mg2+ exposure activated protective responses, including two-component systems and flagellar assembly, consistent with the milder impairment of EET. Notably, these effects were absent in treatments with either cations alone or NNP alone. These findings reveal an overlooked ecological impact of NNP and underscore the potential for distinct bacterial responses to NNP in varying aquatic environments.
