The potential of chitosan nanoparticles (CHNPs) for improving the quality stability of pork patties under freeze-thaw (F-T) cycles was studied. Chitosan nanoparticles were successfully produced by the ionotropic gelation method. Pork patties were incorporated with 0%, 0.5% chitosan, 1% chitosan, 0.5% CHNPs, and 1% CHNPs and then subjected to repeated F-T cycles. Results showed that CHNPs were able to inhibit lipid and protein oxidation with a significant decrease in the lipid malondialdehyde and protein carbonyl formation in the patties after seven F-T cycles. Low-field nuclear magnetic resonance (LF-NMR) results indicated that CHNPs effectively restrained water mobility of samples. In addition, CHNPs exhibited a distinguishable improvement in the textural properties and water holding capacity in the stored patties during F-T cycles. Thus, CHNPs could be used as a natural ingredient to protect frozen patties from quality degradation caused by temperature fluctuations.

Pseudomonas aeruginosa ( P. aeruginosa ) infections present a grave threat to immunocompromised individuals, particularly those with cystic fibrosis due to the development of bacterial biofilms. In this study, we engineered self-assembling chitosan-ceftazidime nanoparticles (CSCE) capable of effectively penetrating biofilms and eradicating P. aeruginos a. The CSCE nanoparticles were synthesized through ionic cross-linking, combining negatively charged ceftazidime with positively charged chitosan, resulting in uniform nanoparticles measuring approximately 40 nm in diameter, exhibiting high dispersity and excellent biocompatibility. Remarkably, these nanoparticles exhibited significant inhibition of P. aeruginosa growth, reduced pyocyanin production, and diminished biofilm formation, achieving a maximum inhibition rate of 22.44%. Furthermore, in vivo investigations demonstrated enhanced survival in mice with abdominal P. aeruginosa infection following treatment with CSCE nanoparticles, accompanied by reduced levels of inflammatory cytokines Interleukin-6 (125.79 ± 18.63 pg/mL), Interleukin-17 (125.67 ± 5.94 pg/mL), and Tumor Necrosis Factor-α (135.4 ± 11.77 pg/mL). Critically, mice treated with CSCE nanoparticles showed no presence of bacteria in the bloodstream following intraperitoneal P. aeruginosa infection. Collectively, our findings highlight the potential of these synthesized nanoparticles as effective agents against P. aeruginosa infections.

Spinal cord injury (SCI) is a central system disease with a high rate of disability. Pathological changes such as ischemia and hypoxia of local tissues, oxidative stress and apoptosis could lead to limb pain, paralysis and even life-threatening. It was reported that catalase (CAT) was the main antioxidant in organisms, which could remove reactive oxygen species (ROS) and release oxygen (O 2 ). However, the efficacy of the drug is largely limited due to its poor stability, low bioavailability and inability to cross the blood spinal cord barrier (BSCB). Therefore, in this study, we prepared folic acid-functionalized chitosan nanoparticles to deliver CAT (FA-CSN CAT) for solving this problem. In vivo small animal imaging results showed that FA-CSN could carry CAT across the BSCB and target to the inflammatory site. In addition, Immunofluorescence, ROS assay and JC-1 probe were used to detect the therapeutic effect of FA-CSN CAT in vitro and in vivo. The results showed that FA-CSN CAT could alleviate the hypoxic environment at the injured site and remove ROS, thereby inhibiting oxidative stress and protecting neurons, which may provide a new idea for clinical medication of SCI.

Porcine epidemic diarrhea (PED), a highly contagious intestinal disease caused by PEDV, threatens newborn piglets and causes enormous losses in the swine industry. Intestinal mucosal immunity is vital for defense against PEDV. Chitosan (CS), a mucin-adhesive biodegradable polysaccharide, has emerged as a promising mucosal vaccine delivery vector and antigen adjuvant. To develop a PEDV mucosal vaccine, we prepared S1-CS nanoparticles (NPs) loaded with the S1 domain of the PEDV spike (S) protein. We evaluated their ability to induce mucosal immune responses in mice via different inoculation routes and to confer immune protection in piglets after oral inoculation. The results showed that S1-CS NPs exhibited no cytotoxicity and remained stable in simulated gastric fluid in vitro. Compared with oral administration, intramuscular inoculation of S1-CS NPs induced higher levels of PEDV-specific serum IgG in mice. However, PEDV-specific IgA was detected only in the serum and intestinal lavage fluid of orally vaccinated mice. Oral administration also elicited higher IFN-γ levels than intramuscular injection, suggesting stronger activation of cellular immunity. In piglets, oral S1-CS NPs induced serum neutralizing antibodies and IgA responses in both serum and intestinal mucosa, increased the number of IgA-secreting cells in the intestine, reduced viremia and virus shedding, and improved intestinal villus morphology. These findings indicate that the S1-CS NPs developed in this study represent promising candidates for an oral mucosal PEDV vaccine.

The aim of this work was to evaluate the potential of polyelectrolyte complex nanoparticles (PENPs) based on hyaluronic acid/chitosan (HA/CS) as carriers for water-insoluble curcuminoid (CUR) and explore in vitro performance against brain glioma cells. PENPs were observed to be affected by the order of addition, mass ratios and initial concentrations of the HA/CS, pH and ionic strength . PENPs remained stable over a temperature range of 5–-55 (C. CUR was successfully encapsulated into the PENPs. CUR-PENPs showed spherical shape with a mean diameter of 207 nm and positive charge of 25.37 mV. High encapsulation efficiency (89.9%) and drug loading (6.5%) was achieved. Drug release studies revealed initial burst release of drug from the PENPs up to 4 h followed by sustained release pattern. DSC thermograms and XRD patterns showed that CUR was encapsulated inside the PENPs in a molecular or amorphous state. Compared with CUR-solution, CUR-PENPs showed stronger dose dependent cytotoxicity against C6 glioma cells and higher performance in uptake efficiency in C6 cells. Cellular uptake of CUR-PENPs was found to be governed by multi-mechanism in C6 cells, involving active endocytosis , macropinocytosis , clathrin-, caveolae-, and CD44-mediated endocytosis . In conclusion, CUR-PENPs might be a promising carrier for therapy of brain gliomas.

Efficient recovery and reuse of lipase remain major challenges in lipid biocatalysis. In this study, aldehyde-functionalized gum arabic-modified magnetic chitosan nanoparticles (GA-CHO/MCNPs) were developed as a biopolymer-based magnetic carrier for lipase immobilization. The carrier was prepared by in situ coprecipitation followed by surface modification with periodate-oxidized gum arabic. Under optimized conditions, the protein loading and protein-based immobilization yield reached 112.5 ± 4.8 mg g −1 and 84.6 ± 2.9%, respectively, while the recovered activity was 62.7 ± 3.1%. Compared with free lipase, Lipase@GA-CHO/MCNPs showed a higher optimum temperature (45 versus 35 °C), improved tolerance to pH and thermal treatment, and convenient magnetic recovery. Immobilization increased the apparent K m from 2.12 to 3.85 mM and decreased V max and k cat , indicating diffusional and steric limitations associated with the carrier. Nevertheless, the immobilized lipase achieved a soybean oil hydrolysis yield of 91.2%, retained 82.3% of its initial activity after 10 reuse cycles, and retained 84.8% activity after 30 d of storage at 4 °C. The results support successful lipase immobilization through an aldehyde-amine-mediated coupling strategy and demonstrate the potential of GA-CHO/MCNPs as a recoverable biopolymer-based carrier for soybean oil hydrolysis.

This study aimed to investigate the impact of substituting pork back fat with Pickering emulsion (PE) stabilized by soybean protein isolate-chitosan (SPI-CH) nanoparticles on the quality of pork batters. Cooking loss, rheological properties, water distribution, microstructures, texture, color, and oxidative stability of pork batters were evaluated. The results revealed that replacing fat with PE led to a reduction in fat content and an increase in moisture content, as well as higher L * and b * values ( P < 0.05). Furthermore, the substitution resulted in improved gel performance by enhancing rheological properties and texture, reducing water mobility and cooking loss, as well as effectively inhibiting lipid and protein oxidation in pork batters. However, no significant differences were observed in sensory acceptability. The findings suggest that using SPI-CH nanoparticles as a stabilizer for PE presents significant potential as a nutritionally improved fat alternative in meat products, offering enhanced functional characteristics.

Limitations of the blood-brain barrier (BBB) are responsible for the low efficacy and high toxicity of many antidepressants. However, strategies to enhance BBB targeting to improve the efficacy and safety of antidepressants remain challenging. In this study, we prepared transferrin (Tf)-functionalized chitosan (CS)- N , O -carboxymethy chitosan (NOCMS) nanosystems (NPs) (NOCMS-CS-Tf NPs), which can enhance BBB targeting in the depressive environment and allow more NPs to enter the brain. Experiments demonstrated that NOCMS-CS-Tf NPs had enhanced BBB targeting in the depressed environment compared to the normal environment and were safe and non-toxic. In vitro experiments demonstrated that NOCMS-CS-Tf NPs could target multiple cell lines in the brain and had stronger BBB targeting in neuroinflammatory and neuronal injury environments than in normal environments; in vivo experiments demonstrated that NOCMS-CS-Tf NPs had stronger BBB targeting in the brains of depressed mice than in normal mice, and were safe and non-toxic. The nanosystem enhanced BBB targeting in the setting of neuroinflammation and neuronal damage in depression, providing a promising approach to drug delivery systems for depression and bringing new hope for the return to the market of many first-line antidepressants that have been withdrawn.

Severe acute pancreatitis (SAP) is a prevalent gastrointestinal disease with no effective treatment to control inflammation currently. Macrophages, particularly peritoneal macrophages (PMOs), play a pivotal role in SAP inflammation by polarizing into M1 or M2 phenotypes, which exhibit distinct functional properties and cytokine expression profiles. Galectin-9 (Gal-9) modulates macrophage polarization, but its specific effect on PMOs in SAP remains unclear. In this study, hyaluronic acid-chitosan nanoparticles encapsulating Gal-9 (HA-CS-Gal-9 NPs) were developed for delivery. In vitro, HA-CS-Gal-9 NPs enhanced M2 marker expression and suppressed M1 markers in both naive (M0) and LPS-induced M1 macrophages. In vivo, HA-CS-Gal-9 NPs effectively delivered Gal-9, showing effective uptake by PMOs without notable toxicity, resulting in reduced IL-6, and increased IL-10 expression in PMOs. Treatment with these nanoparticles (NPs) decreased systemic pro-inflammatory cytokines, thereby alleviating pancreatitis severity. These findings demonstrate that Gal-9-loaded NPs robustly promoted M2 macrophage polarization, highlighting a promising therapeutic strategy for SAP.

This study aimed to develop self-assembled gelatin-chitosan (Gel-CS) nanocapsules as a water-soluble delivery system for alcohol-soluble compounds. Eugenol (Eug) selected as the model molecule for its high solubility in alcohol and insolubility in water. The study also examined different reaction conditions and reagent loading ratios to optimize eugenol-embedded gelatin-chitosan nanocapsules (Eug-Gel-CS NPs) formation. The optimal loading ratio of Gel, CS, and Eug was 3:1:2, which yielded an average particle size of 229.09 nm and an encapsulation efficiency of 50.69%. The morphological characteristics of Eug-Gel-CS NPs were further demonstrated by transmission electron microscopy. The prepared Eug-Gel-CS NPs also exhibited potential controlled-release properties in aqueous phase with the in vitro release percentage of 69.47 ± 1.24% over 36 h. This nanocapsule system may provide a platform to expand the potential applications of Eug in food and nutrition.

The outcome of application of epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs) in patients with non-small-cell lung cancer (NSCLC) always suffers from acquired drug resistance. Its combination with other therapies has been explored with promising synergistic results. Photodynamic therapy (PDT) is an established treatment modality for NSCLC, but the low tumor selectivity of currently available photosensitizers (PSs) hampered the wide clinical application of PDT. Here, we synthesized erlotinib modified chitosan (ECs) by click coupling and developed ECs/indocyanine green (ICG) self-assembled nanoparticles (GECs) for combined targeted chemo-photodynamic therapy. GECs showed oval-shaped particles with a diameter of 267.5 nm, a zeta potential of −15.2 mV, and considerable photostability. In vitro release experiment showed that erlotinib and ICG could be slowly released from GECs in acidic condition with lysozyme. GECs were capable of generating reactive oxygen species (ROS) for PDT under near-infrared laser irradiation . GECs showed synergistic molecular targeted and photodynamic therapeutic effects in inhibition of cellular growth and induction of apoptosis in NSCLC cells. This study demonstrated that the GECs could be a promising platform by combination of targeted chemo-photodynamic therapy for NSCLC treatment.

Lumbrokinase (LK) has strong fibrinolytic and thrombolytic activities, but it has a short half-life, can be easily inactivated, and may cause hemorrhage as a side effect. This study develops a potential thrombolytic therapy by fabricating N,N,N-Trimethyl Chitosan (TMC) nanoparticles modified with the cyclic Arg-Gly-Asp-Phe-Lys peptide (c-RGD) and loaded with LK (i.e. c-RGD-LK-NPs). The binding of c-RGD to platelet membrane GPIIb/IIIa receptors is expected to enable targeted delivery of the c-RGD-conjugated TMC to the thrombus. The synthesized c-RGD-LK-NPs had a mean particle size of 232.0 nm, zeta potential of 19.8 mV, entrapment efficiency of 52.7% ± 2.5%, and loading efficiency of 17.4% ± 0.65%. Transmission electron microscopy showed that they were generally spherical. The c-RGD-LK-NPs gave a cumulative in vitro LK release of 80.6% over 8 h, and the activity of LK was close to 80%, indicating that the nanoparticles protected the activity of LK. In vitro blood clot lysis assays were carried out and in vivo thrombolysis effect was tested in Sprague-Dawley rats carotid artery thrombus model. In all cases, the c-RGD-LK-NPs showed superior performance compared with the free LK and the unmodified TMC nanoparticles loaded with LK. The c-RGD-LK-NPs reagent is expected to be potentially useful in treating thromboembolic diseases.
