NIR laser-responsive liquid metal-loaded polymeric hydrogels for controlled release of doxorubicin

Linlin Fan,Xuyang Sun,Xuelin Wang,Hongzhang Wang,Jing Liu

Liquid metals (LMs) have recently emerged as a new class of promising multifunctional materials with attractive properties. They have excellent photothermal conversion efficiency, generating heat under near-infrared (NIR) laser irradiation. This work reports encapsulating LM droplets into poly(NIPAm-co-MBA) hydrogels (PNM) to achieve nanodispersed liquid metals in bulk polymeric hydrogels for NIR laser-responsive materials. LM droplets (∼530 nm) are produced by dispersing an alloy of gallium and indium (EGaIn) into glycerol. The LM-loaded PNM hydrogels (PNM/LM) exhibited excellent thermal-/NIR laser-responsive ability. In a water bath, the weight of the PNM/LM can decrease 92% at 50 °C. And the volume of PNM/LM can decrease 62% under NIR laser irradiation for 12 min. Because of its thermal-/NIR laser-responsive ability and porous three-dimensional (3D) networks, PNM/LM is very suitable for use as a drug carrier. We also prepared doxorubicin (DOX)-loaded PNM/LM hydrogels (PNM/LM/DOX) and demonstrated that the PNM/LM/DOX hydrogel can generate heat and raise its temperature under NIR laser irradiation. When the temperature becomes higher than the lower critical solution temperature (LCST), such a hydrogel would shrink immediately and extrude the DOX encapsulated in its networks simultaneously, then complete the controlled release of the pre-loaded drug. Further, an in vitro cytotoxicity test indicated the biocompatibility and feasibility as a chemophotothermal synergistic therapeutic of the present hydrogel. This NIR laser-responsive hydrogel fully exhibits its superiority as a drug carrier which promises great potential in future targeted controlled drug release.

An Enzyme-Responsive Controlled Release System of Mesoporous Silica Coated with Konjac Oligosaccharide

Wei Guo,Chunyu Yang,Liru Cui,Huiming Lin,Fengyu Qu

A simple and green method to fabricate an ingenious enzyme-responsive drug controlled release system was presented. Mesoporous silica material (mSiO2) 100 nm in size was used as the host, and Konjac oligosaccharide (KOGC) was employed to seal the nanopores of mSiO2 to inhibit the drug release. Rhodamine B was used as the model cargo to reveal the release behavior of the system. The KOGC-modified mSiO2 (mSiO2@KOGC) retains the drug until it reaches the colonic environment where bacteria secrete enzymes (β-mannanase) can degrade KOGC and make drug release. The amount of KOGC and enzyme can be used to adjust the release performance. And all the release behaviors fit the two-step Higuchi model, which predominate by KOGC degradation and mesoporous structure, respectively. With well bioactivity and selectivity, the system has potential application as an oral medicine carrier for treating intestinal disease.

Narrow pH response multilayer films with controlled release of ibuprofen on magnesium alloy

Li Lei,Ran Yan,Shougang Chen,Xiangping Hao,Wenwen Dou,Hu Liu,Zhanhu Guo,Dan Kilula,Ilwoo Seok

An intelligent narrow pH-triggered multilayer film was prepared on magnesium alloys, aiming to solve the implant infections during the implantation period and improve the corrosion resistance of magnesium alloys. The encapsulation of ibuprofen by chitosan (IBU@CS) makes the release of IBU sensitive to narrow pH (pH 6.8–7.4). Positive charged IBU@CS was assembled with heparin (Hep) to fabricate (Hep/IBU@CS) 10 film on AZ31 alloys using layer-by-layer method. The microstructure, composition and anticorrosion properties of the film were characterized by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FT-IR) and electrochemical experiments. Cellular activity was studied by MTT cell viability assay. The results showed that the Hep/IBU@CS multilayer films improved the corrosion resistance of magnesium alloys. The in vitro test demonstrated that the release of IBU in the film presented narrow pH sensitivity. The films showed no obvious signs of cytotoxicity conformed by the MTT assay and presented antibacterial properties. These preliminary results demonstrate the potential use of the Hep/IBU@CS multilayer films on magnesium-based implants.

Dynamic Tannic Acid Hydrogel with Self-Healing and pH Sensitivity for Controlled Release

Peihong Li,Yanlong Sui,Xueyan Dai,Qiangsheng Fang,Haofei Sima,Chunling Zhang

Dynamic hydrogels constructed with dynamic chemical bonds often have mechanical strength and self-healing properties. In this paper, tannic acid is combined with lysine-containing F127 through Schiff base. A series of FLaT hydrogels cross-linked by hydrogen bonds and dynamic chemical bonds is prepared, and the influence of Schiff base amount on the performance is discussed. The FLaT hydrogel exhibits reversible sol–gel transition, self-healing, injectability, and pH sensitivity. Increasing the amount of Schiff base can improve the strength, stability, and self-healing ability of the hydrogel. Owing to their low cytotoxicity, linear release pattern, and pH-controlled release rate, the FLaT hydrogels show potential use in drug delivery systems for cancer treatment.

Photo-induced synthesis glucose-responsive carriers for controlled release of insulin in vitro

Du Xiangxiang,Jiang Guohua,Li Lei,Liu Yongkun,Chen Hua,Huang Qin

The glucose-responsive amphiphilic poly(PEGMA- co -AAPBA) copolymer micelles as drug carriers have been developed using 3-phenylpyruvic acid (3-PPA) as photoinitiator. Under the light irradiation, 3-PPA could be self-polymerized into the trimer structure to execute the emulsifier and initiator functions that confirmed by gas chromatography-mass spectrum (GC-MS) analysis. The structure and morphology of resultant poly(PEGMA- co -AAPBA) micelles were characterized by 1 H NMR and TEM. Insulin, as the model drug of diabetes, was loaded into micelles during the photopolymerization process. The as-prepared polymeric micelles exhibited the excellent glucose sensitivity. The loaded insulin could be released from micelles triggered by regulation of temperature and glucose concentration in the environment. The new drug carriers provided a potential application for the therapy of diabetes based on glucose-sensitive controlled drug delivery systems.

Silk Fibroin Microneedles with Controlled Release Kinetics for Extended Protection Against Soman Intoxication

Chong Li,Yi Zhang,Guixiang Yang,Qian Jin,Wang Chen,Ruihua Zhang,Tong Shi,Jingchen Wei,Xuejun Chen,Liqin Li

The rapid aging kinetics of soman (< 2 min) necessitate the development of effective prophylactic countermeasures, as conventional post-exposure therapies are often inadequate. To address this challenge, we engineered a novel separable microneedle (MN) system based on silk fibroin (SF) for sustained transdermal delivery of huperzine A (Hup A)—a reversible acetylcholinesterase inhibitor with demonstrated blood - brain barrier penetration capabilities. The optimized SFMNs design featured a distinctive tip-separation mechanism that facilitated reliable detachment of drug-loaded needle tips within 2 min after dermal application. The characterization results showed that the MN patch exhibited robust mechanical strength (155.49±11.18 N/patch), uniform morphology, effective skin penetration and homogeneous distribution of Hup A within the needle tips. Notably, the SFMNs platform provided controlled release kinetics, achieving sustained in vitro release of Hup A over 12 h, with an initial burst phase conducive to rapid drug onset. Pharmacokinetic evaluations in mice revealed that transdermal delivery via SFMNs (4 mg/kg Hup A) maintained therapeutic plasma concentrations for up to 12 h, significantly prolonging the mean residence time (11.87 ± 0.88 h) compared to oral administration (2.39 ± 0.31 h). This sustained-release profile markedly reduced systemic toxicity; whereas oral Hup A elicited severe cholinergic adverse effects (e.g., sweating, tremors), SFMNs-mediated delivery circumvented these toxic responses. In a prophylactic efficacy study, Hup A-loaded SFMNs (4 mg/kg) conferred complete protection (100% survival) for 12 h against a 95% lethal dose soman challenge, substantially extending the protective window compared to both oral Hup A (2 h) and intramuscular pyridostigmine bromide (45 min). These findings demonstrate that the SFMNs platform represents a promising, convenient, and well-tolerated strategy for long-acting prophylaxis against soman intoxication.

Capillary-Wave-Induced ‘Endocytosis’ Enables Programmable Core-Shell Capsules Formation for Controlled Release

Yi Huang,Sihang Liu,Shuai Yin,Lian Xiao,Yugang Zhao,Zhi Tao,Haiwang Li

Understanding droplet-liquid pool interactions in immiscible systems is critical for advancing fluidic encapsulation strategies. Here, we uncover a previously unreported ‘endocytosis’ phenomenon in which an aqueous droplet impacts a deep immiscible pool and spontaneously engulfs the pool liquid, forming a stable double-layered liquid structure. This process origins from the focusing of capillary waves at the droplet-pool interface and followed by a pinch-off of a central oil column via Rayleigh-Plateau instability. We propose a dimensionless criterion for central column formation, determined by whether viscous dissipation of the droplet's kinetic energy. By introducing a UV-crosslinkable precursor, gelatin methacryloyl (GelMA) into the droplet, we succeed in encapsulating pool liquid into hydrogel capsules with tunable core-shell ratios by simply adjusting the impact velocity. This enables precise control of release mechanism: thick-shell capsules exhibit non-Fickian sustained release, while thin-shell capsules display delayed burst Case II release upon swelling. The proposed ‘endocytosis’ phenomena offer a surfactant-free, single-parameter controlled strategy compared to conventional liquid encapsulation methods, which offers a new opportunity for on-demand release in drug delivery and responsive materials.

Controlled-release hydrogel loaded with engineered extracellular vesicles for treatment of chronic endometritis

Haixia Li,Ruixue Hu,Yue Zhang,Junli Zhang

Chronic endometritis (CE) is an important factor contributing to infertility and adverse pregnancy outcomes. Current clinical management mainly relies on antibiotics such as ceftizoxime (CTX), whose efficacy is limited by poor oral bioavailability and short half-life. To address these limitations, this study developed an intelligent, responsive hydrogel incorporating Leonurus japonicus Houtt derived extracellular vesicles (EVs) loaded with CTX. The hydrogel forms sustained-release drug reservoir in situ within the uterine cavity, enabling controlled release in response to the local inflammatory microenvironment. EVs can effectively regulate the inflammatory microenvironment by inhibiting activation of NF-κB pathway and NLRP3 inflammasome, downregulating the pro-inflammatory factor TGF-α, and upregulating the anti-inflammatory factors IL-10 and TGF-β1, thereby significantly promoting endometrial repair and reducing fibrotic areas. Meanwhile, CTX provides targeted antibacterial activity to eliminate pathogens and control infection. Through exerting antibacterial, anti-inflammatory, and tissue repairing, the developed intelligent, responsive hydrogel system offers a promising strategy for treatment of CE.

Controlled Release of Thermosensitive Hydrogel Incorporating Betaxolol-Loaded Resin Microspheres for Glaucoma Therapy

Liu Hongfei,Zhang Zhen,Tang Xinyi,Feng Yingshu,Firempong Caleb Kesse,He Haibing,Zhang Guoqing

Glaucoma therapy is often limited by poor ocular bioavailability and the short residence time of conventional eye drops, highlighting the need for sustained-release delivery systems. This study presents the development of a thermosensitive in situ hydrogel incorporating cation-exchange resin (CER) for sustained delivery of betaxolol hydrochloride (BH) in glaucoma therapy. CER microspheres were prepared using a seed swelling method, and drug loading was achieved through an ion-exchange interaction to obtain BH@CER, which was characterized by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and scanning electron microscopy (SEM). A thermosensitive hydrogel-microspheres composite (BH@CER@P407) was formulated using optimized concentrations of poloxamer 407 (P407), exhibiting a sol-to-gel transition at approximately 34°C for easy administration and prolonged ocular retention. In vitro release studies indicated that BH@CER@P407 achieved sustained release (85% over 12 h), in contrast to the pronounced burst release of commercial BH eye drops (Betoptic® S, 90% within 2 h). In vivo evaluations in New Zealand rabbits revealed strong ocular adhesion and retention for up to 8 h, correlating with sustained intraocular pressure (IOP) reduction. Ocular irritation studies confirmed excellent biocompatibility. Compared to Betoptic® S, the BH@CER@P407 offers enhanced ocular residence and prolonged therapeutic efficacy, presenting a promising strategy for improving glaucoma treatment.  Graphical

Nisin Z-Loaded Chitosan-Pectin Coated Liposomes for Controlled Release and Antibiofilm Activity

Minh Quan Ho,Nam-Son Dinh-Ngoc,Quynh-Anh Vu-Yen,Dang Nguyen Nha Khanh,Tan Anh Nguyen,Ngoc Thuy Trang Le

Abstract Pathogenic biofilms and complex biological environments significantly limit the therapeutic efficacy of antimicrobial peptides such as Nisin Z due to rapid degradation and poor penetration. In this study, we developed a Nisin Z-loaded chitosan-pectin coated liposome (LCPN) as a lipid-polymer hybrid nanosystem to overcome these limitations. The optimized LCP formulation exhibited an average particle size of 278.4 ± 3.1 nm, a polydispersity index of 0.229 ± 0.326, and a surface charge of −28.7 ± 0.8 mV. Furthermore, the nanosystem achieved a high encapsulation efficiency of 63.84% and demonstrated a sustained release profile, delivering 63.64% of Nisin Z over 72 h. In addition, LCPN exhibited a sustained release profile, contributing to prolonged antimicrobial activity. Functionally, the LCPN system demonstrated strong antibiofilm efficacy (88.13% at 350 μg/mL), effectively enhancing disruption of pre-formed biofilms and significantly reducing bacterial biomass compared to free Nisin Z. Notably, in vivo evaluation in a shrimp infection model showed enhanced antibacterial performance in hemolymph, resulting in improved survival rates (83.3% at 14 days post-infection) and growth performance following Vibrio challenge. These findings suggest that the chitosan-pectin coated liposomal platform is a highly stable and controlled-release delivery system for antimicrobial peptides, offering a promising and targeted strategy for disease management in aquaculture.

Mechanistic insights of the controlled release properties of amide adhesive and hydroxyl adhesive

Zheng Luo,Xiaocao Wan,Chao Liu,Liang Fang

Although interactions between drugs and acrylate pressure sensitive adhesives (PSAs) containing amide groups were reported in the previous studies, detailed studies elucidating their mechanism of action are still lacking. In the present study, an amide PSA (AACONH 2 ) and a hydroxyl PSA (AAOH, as the control) were synthesized, and their molecular mechanism of controlled drug release was described. Using zolmitriptan (ZOL) and etodolac (ETO) as model drugs, in vitro drug release and skin permeation experiments were performed. Intermolecular interactions between drugs and PSAs were determined by Flory-Huggins model, FT-IR spectroscopic analysis and molecular modeling. In addition, PSA mobility was evaluated using differential scanning calorimetry and rheology study. Release percent of ZOL and ETO from AACONH 2 were 43.9 ± 0.3% and 50.0 ± 2.0% respectively, while from AAOH, the release percent of ZOL and ETO were 61.4 ± 1.2% and 81.0 ± 1.2% separately. As a consequence of controlled drug release , skin permeation of both drugs was significantly controlled by AACONH 2 . It was demonstrated that AACONH 2 markedly interacted with drugs, especially with ETO, through hydrogen bonding and weak intermolecular forces ( e . g . dipole-dipole and van der waals). PSA mobility of AACONH 2 was significantly increased due to drug-PSA interactions. In conclusion, AACONH 2 had stronger controlled release properties compared with AAOH, which was mainly caused by the stronger interactions between amide groups and drugs. The amide PSA synthesized in the present study was a potential sustained-release excipient for transdermal drug delivery system .

Characterization of Menthol-Loaded Electrospun Nanofibrous Membranes and the Temperature-Controlled Release Properties

Zhiqiang Li,Shanzhai Shang,Xinyi Fang,Jingmei Han,Tinghua Li,Xia Zhang,Dong Xiao,Qiaomei Sun,Hui Li,Ping Lei

The controlled release of volatile compounds is one of the important research topics in the field of food. In order to improve the flavor release ability of menthol and to achieve a temperature-controlled release of menthol, chitosan (CS) and polycaprolactone (PCL) are used as raw materials to prepare nanofibrous membranes by electrostatic spinning and menthol-loaded nanofibrous membranes are subsequently prepared by incorporating a menthol flavor. Scanning electron microscopy (SEM), Fourier transform infrared (FTIR), XRD, and solid-state NMR are applied to characterize the morphology and chemical structure of the nanofibrous membranes. Thermogravimetric analysis (TGA) is used to test the thermal stability and release characteristics of the menthol-loaded nanofibrous membranes. The gas chromatography (GC) assay shows that the CS/PCL nanofibrous membrane has a menthol loading of 57.64% and can gradually release menthol from 85 to 260°C, thus, achieving a temperature-controlled release of menthol. A 14-day stability test shows that the nanofibrous membrane retains menthol up to 40.23% and cytotoxicity experiments confirm their good biocompatibility and biosafety. The method of embedding menthol proposed in this paper can offer the possibility of controlling the temperature to release menthol and providing potential ideas for the application of menthol in food and pharmaceuticals.
