Enhanced Lethal Effects of Combined P-tert-Butylcatechol and L-Lysine on Microcystis aeruginosa

Heyun Jiao,Gangwei Jiao,Ruitong Jiang,Yifei Shen,Peimin He,Liu Shao

Simple SummaryHarmful algal blooms caused byMicrocystis aeruginosathreaten aquatic ecosystems and human health. This study explores the combined effects of two natural allelochemicals, P-tert-butylcatechol (TBC) and L-lysine, in controllingM. aeruginosa. When combined at low concentrations (0.04 mg/L TBC + 1 mg/L L-lysine), they achieved over 90% growth inhibition within 96 h. The treatment damaged algal cell membranes, reduced photosynthetic efficiency, and triggered oxidative stress. Furthermore, intracellular and extracellular microcystin levels decreased significantly compared to untreated algae, minimizing ecological risks. These findings highlight the potential of the combination of TBC and L-lysine as a safe and cost-effective strategy for mitigating harmful algal blooms.Allelochemicals are recognized as promising algaecides due to their environmental safety. Para-tert-butylcatechol (TBC) and L-lysine exhibit significant potential in suppressing harmful algal blooms (HABs); however, their combined effects and algae inhibition mechanisms remain unelucidated. Therefore, this study systematically investigated the growth inhibition ofMicrocystis aeruginosaby TBC and L-lysine individually and in combination, while simultaneously examining their combined effects on algal growth, cell membrane integrity, photosynthetic activity, antioxidant responses, and microcystin production. The results revealed a significant interactive effect between TBC (0.04 mg/L) and L-lysine (1 mg/L), achieving over 90% growth inhibition within 96 h. The combined treatment significantly inhibitedM. aeruginosagrowth through impaired photosynthetic efficiency and elevated oxidative stress. Compared to the control group, the treatment group exhibited a continuous decline in chlorophyll-a content, phycobiliprotein levels,Fv/Fm, YII, α, and rETRmax, while phosphoenolpyruvate carboxylase (PEPC) activity decreased by 96.48% by day 8. And antioxidant enzymes, including superoxide dismutase (SOD) and reduced glutathione (GSH), showed a progressive increase in activity. In addition, the structure and integrity of the cell membrane ofM. aeruginosawere damaged after treatment, and the conductivity of the treatment groups increased continuously from 2.32 to 4.63 μs/cm. In addition, under combined treatment, intra- and extracellular microcystin levels initially increased (peaking at day 2) but sharply declined thereafter, becoming significantly lower than controls by day 8. These findings highlight the potential of combining TBC and L-lysine as an eco-friendly and cost-effective strategy for mitigatingM. aeruginosa-dominated harmful algal blooms.

Genkwanin impairs triple-negative breast cancer aggressiveness and metastasis by targeting Lysine Demethylase 4C

Hanfeng Xu,Jie Gao,Shiyong Fu,Zihao Liang,Ying Zhang,Huijuan Chen,Qin Zheng

Background Triple-negative breast cancer, the most aggressive breast cancer with highest metastatic capacity and the worst prognosis, accounts for 15% of breast cancer. According to prior studies, the natural active component genkwanin suppressed cell growth in estrogen-receptor-positive breast cancer. Nonetheless, its impact on triple-negative breast cancer and its target remains poorly understood. Purpose To explore the effects of genkwanin and identify the direct targets and cellular mechanism of genkwanin in triple-negative breast cancer. Study design The effects of genkwanin were investigated in SUM-159PT and MDA-MB-231 cells. Direct target protein of genkwanin were identified by Quantitative proteomics. The in-vivo effect of genkwanin on triple-negative breast cancer were investigated using mouse models. Methods The anti-metastasis effects of genkwanin were investigated by individual cell-tracking assay and 3D-on top assay. Quantitative proteomics was used to identify the target of genkwanin and microscale thermophoresis ligand-binding assay was performed to further confirmed it. In-vitro and in-vivo extreme limiting diluting assays were performed to investigate the inhibitory effect of stemness. Lung metastasis mouse model was used to evaluate the anti-metastasis effects of genkwanin. Results This study demonstrated that genkwanin could significantly attenuate migration and invasion in triple-negative breast cancer cells. Notably, genkwanin could efficiently modulate the stemness of breast cancer cells and improve its sensitivity to paclitaxel (PTX). Lysine demethylase 4C (KDM4C) was identified as the target of genkwanin and the in-vivo therapeutic effect of genkwanin was also confirmed in 4T1-luc lung metastasis model. Mechanically, genkwanin reduced KDM4C expression, which regulated the lysine demethylation of histone H3, thereby suppressing KLF14 transcription. KLF14 acts as a switch of lipid synthesis and regulates intracellular cholesterol levels, inhibiting cancer metastasis. Conclusion These results demonstrate genkwanin's therapeutic potential in triple-negative breast cancer and its effect on targeting metastasis.

Hyaluronic Acid-Graft-Poly(L-Lysine): Synthesis and Evaluation as a Gene Delivery System

Viktor Korzhikov-Vlakh,Polina Teterina,Nina Gubina,Apollinariia Dzhuzha,Tatiana Tennikova,Evgenia Korzhikova-Vlakh

The synthesis of novel biodegradable polymers as non-viral vectors remains one of the challenging tasks in the field of gene delivery. In this study, the synthesis of the polysaccharide-g-polypeptide copolymers, namely, hyaluronic acid-g-polylysine (HA-g-PLys), using a copper-free strain-promoted azide-alkyne cycloaddition reaction was proposed. For this purpose, hyaluronic acid was modified with dibenzocyclooctyne moieties, and poly-L-lysine with a terminal azido group was obtained using ring-opening polymerization ofN-carboxyanhydride of the corresponding protected amino acid, initiated with the amino group azido-PEG3-amine. Two HA-g-PLys samples with different degrees of grafting were synthesized, and the structures of all modified and synthesized polymers were confirmed using1H NMR and FTIR spectroscopy. The HA-g-PLys samples obtained were able to form nanoparticles in aqueous media due to self-assembly driven by electrostatic interactions. The binding of DNA and model siRNA by copolymers to form polyplexes was analyzed using ethidium bromide, agarose gel electrophoresis, and SybrGreen I assays. The hydrodynamic diameter of polyplexes was ˂300 nm (polydispersity index, PDI ˂ 0.3). The release of a model fluorescently-labeled oligonucleotide in the complex biological medium was significantly higher in the case of HA-g-PLys as compared to that in the case of PLys-based polyplexes. In addition, the cytotoxicity in normal and cancer cells, as well as the ability of HA-g-PLys to facilitate intracellular delivery of anti-GFP siRNA to NIH-3T3/GFP+ cells, were evaluated.

Alkyl-substituted cucurbit[6]uril–based fluorescent probes for glutamic acid and lysine sensing

Xin Lin,Fang-Fang Shen,Ren-Jie Song

In this study, two novel fluorescent probes, H 1 /G and H 2 /G, were constructed using two alkyl-substituted cucurbit[6]urils (CB[6]s), H 1 and H 2 , and a cationic dye guest, G. The G molecules were encapsulated within the cavities of CB[6] hosts, forming 1:1 host–guest complexes. While free G molecules exhibited negligible fluorescence in solution, the inclusion complexes demonstrated a strong fluorescence signal (618 nm). The detection limits for glutamic acid (Glu) recognition by the two probes were as low as 2.98 × 10 −7  M and 4.20 × 10 −7  M, respectively. Notably, although the two modified CB[6]s differed by only a single substituent, their resulting probes demonstrated different degrees of amino acid recognition ability. The H 2 /G probe was capable of recognizing both Glu and lysine (Lys), while the H 1 /G probe detected only Glu. By employing structurally modified CB[6]s as hosts and constructing CB[6]-based fluorescent probes capable of selectively recognizing non-aromatic amino acids, this study paves the way for the biological applications of CB[6]-based supramolecular host–guest systems.

Lysine-polydopamine nanoparticles for ameliorating acidic microenvironment and neuroprotection in spinal cord injury repair

Yi Li,Bing Ran,Qihang Ye,Huisheng Zhong,Nayin Zhong,Yi Zhong,Xinyu Fu,Xinying Liu,Guanghua Wu,Xiaohui Hu,Junming Ye,Qinwen Zhong

Spinal cord injury (SCI) is exacerbated by the formation of an acidic microenvironment and extensive neuronal loss, both of which contribute to poor functional recovery. To address this, we developed lysine-polydopamine nanoparticles (Lys-PDA) as a multifunctional therapeutic platform for SCI. Lysine, a naturally occurring amino acid, possesses weak alkalinity and neuroprotective properties, but suffers from poor in vivo stability and non-specific distribution. Polydopamine (PDA), a biocompatible polymer with inherent antioxidant and anti-inflammatory capabilities, was employed as a nanocarrier to enhance lysine delivery and local retention. In vitro and in vivo assessments confirmed that Lys-PDA exhibit excellent biocompatibility and enable sustained release of lysine for up to three days. Upon administration into SCI mice, Lys-PDA significantly neutralized the acidic lesion milieu, with pH values increasing by approximately 0.33 units—approaching levels observed in uninjured spinal tissue. After 28 days of treatment, Lys-PDA markedly reduced neuronal apoptosis, suppressed reactive oxygen species (ROS) and lactate accumulation, and significantly improved hindlimb motor function. Behavioral analyses demonstrated a substantial increase in Basso Mouse Scale (BMS) scores from 0.0 to 5.8, alongside a corresponding increase in hindlimb stride length from 2.0 cm to 4.3 cm. Collectively, these findings suggest that Lys-PDA not only modulate the hostile post-injury microenvironment but also promote neuroprotection and functional recovery, positioning them as a promising candidate for SCI repair.

A Polymer-Based Polar Stationary Phase Grafted With Modified Lysine for Hydrophilic Interaction Chromatography

Yufeng Shen,Rui Jin,Feifang Zhang,Bingcheng Yang

The high hydrophobicity and chemical inertness of poly(styrene-divinylbenzene) (PS-DVB) microspheres make their surface hydrophilic modification difficult. Here we describe a facile way to convert PS-DVB microspheres to hydrophilic, then can be used as polar stationary phase for hydrophilic interaction chromatography. This approach utilizes the grafting of an acrylamide-terminated lysine zwitterionic monomer onto PS-DVB microspheres via free radical polymerization. The obtained stationary phase shows good hydrophilicity and a typical retention mechanism of hydrophilic interaction chromatography toward several model polar analytes. It also exhibits obvious zwitterionic properties and is capable of separating cationic and anionic analytes simultaneously. The column shows negligible bleeding level, much superior to silica-based ones.

Qian Sun,Xielong Dong,Juan Xu,Ting Wang

The study aimed to develop a multifunctional wound dressing with enhanced antibacterial properties and wound healing promotion. The synthesis process involved preparing oxidized regenerated cellulose (ORC) following a modified procedure, synthesizing chitosan/silver nanoparticles (CS/Ag NPs) via an in-situ reduction method, and subsequently preparing ORC/CS/Lys@Ag NPs hydrogels. Characterization techniques including FTIR, XRD, SEM, and EDS were employed to analyze functional groups, lattice structure, morphology, and elemental composition. Gelation time, swelling behavior, water retention, mechanical properties, viscosity, self-healing capacity, rheological behavior, oxygen permeability, in vitro degradation, release of Ag + , and antibacterial properties were evaluated using various experimental methods. Results indicated that the novel wound dressing has the capability to evenly distribute Ag NPs to effectively counteract bacteria. It can maintain moist conditions for 86 h, resist a sturdy mechanical pressure of 11.3 KPa, and degrade by 11.045 % ± 0.429 within 8 h. Combining its efficient gas exchange abilities, self-repairing function, and biocompatibility, almost full recovery was observed in injured mouse skin within 13 days, highlighting its promising clinical utility.

Poly(L-lysine) modified glassy carbon electrode for sensitive electrochemical measurement of 2,3,5,6-tetramethylpyrazine

Mingjing Yin,Jicheng Yuan,Lina Yu,Xinying Ma,Kun Long,Mingyong Chao

2,3,5,6-Tetramethylpyrazine (TMP), an organic compound with numerous potential health benefits, is extensively used in medical and food products. This research introduces a novel approach for the precise quantification of TMP by employing a poly(L-lysine) modified glassy carbon electrode (pLL/GCE). The pLL/GCE is fabricated through the electrochemical polymerization of L-lysine onto a glassy carbon electrode (GCE) surface. This modification enhances the electrode's electrocatalytic efficiency, resulting in a significant increase in the reduction peak current of TMP. Exploiting this distinctive property, we establish an electrochemical method for quantifying TMP using cyclic voltammetry (CV). Optimized conditions reveal a robust linear correlation between the reduction peak current of TMP and its concentration within two adjacent linearity ranges: 40.0 nmol/L to 1.0 μmol/L and 1.0 μmol/L to 100.0 μmol/L. Notably, this method achieves an impressive detection limit of 20.0 nmol/L. To validate the practicality of this approach, we analyzed various real-world samples, obtaining recovery rates ranging from 98.2% to 100.8%. The developed method holds significant promise for the determination of TMP in medical and food products, underscoring its potential for widespread application.

Study on polyarginyl lysine grafted modified nano biochar as an environmentally friendly shale inhibitor

Qiannan Wang,Guancheng Jiang,Xingxing Wang,Yinbo He,Lili Yang,Yijie Wang

In the study, JL-BC, an environmentally friendly shale inhibitor with a temperature tolerance of 220 °C was synthesized by grafting modified nano biochar (BC) onto polyarginyl lysine (JL) for the first time. Fourier transform infrared spectroscopy (FTIR) and thermogravimetric (TGA) tests indicated that JL-BC had been successfully synthesized and had good thermal stability. The inhibition performance of JL-BC was experimentally evaluated by linear swelling test, hot-rolling recovery test and sodium bentonite (Na-BT) block immersion test, and the inhibition mechanism of JL-BC was studied by various experimental characterization methods. The experimental results showed that compared with potassium chloride (KCl), polyether amine (PEA), bionic inhibitor dopamine (DA), 2, 3-epoxypropyltrimethylammonium chloride (EPTAC), and poly dimethyl diallyl ammonium chloride (PDMDAAC), Na-BT had the lowest linear swelling height of 4.49 mm in 4 % JL-BC solution at 150 °C. The recovery rates of shale cuttings were highest in 4 % JL-BC solution at 200 °C and 220 °C, which were 96 % and 92 %, respectively. The Na-BT block was immersed in 4 % JL-BC solution for 16 h basically retained its original morphology. The excellent inhibition performance of JL-BC was mainly attributed to the positive charge of JL-BC in aqueous solution at pH 9, which was strongly adsorbed on Na-BT, lowering the zeta potential of Na-BT, destabilizing Na-BT and causing it to aggregate. With the increase of JL-BC concentration, Na-BT gradually aggregated into larger flocs, increasing the particle size of Na-BT. Nano BC reduced water intrusion to a certain extent by physically blocking micropores. In addition, the EC 50 value of JL-BC was 2.93 × 10 5 mg/L, indicating that it was non-toxic. The addition of JL-BC to the soil effectively increased the content of organic matter, ammonium nitrogen and available potassium, and promoted the growth of wheat seedlings. This work may open a new avenue for the development and use of environmentally friendly treatment agents.

Multi-response kinetic study of Maillard reaction hazards in the glucose-lysine model system

Kaihua Liu,Zhijie Liu,Junjian Miao,Yiqun Huang,Keqiang Lai

BACKGROUND N ε-carboxymethyllysine (CML), N ε-carboxyethyllysine (CEL) and α -aminoadipic acid (AAA) are important foodborne hazards and their intake can cause a variety of diseases in humans. It is extremely important to investigate the formation mechanism of CML, CEL and AAA, as well as their association with each other when aiming to control their production. RESULTS A multi-response kinetic model was developed within the glucose-lysine Maillard reaction model system. The concentrations of glucose, lysine, glyoxal (GO), methylglyoxal (MGO), CML, CEL and AAA were quantified at different temperature (100–160 °C) and at different intervals (0–60 min). The experimental data were fitted to the proposed model to calculate kinetic parameters for the corresponding steps. The results indicated that the production of CML was primarily relied on the direct oxidative cleavage of the Amadori product, rather than the reaction between GO and Lys, whereas CEL and AAA were generated through the reaction of MGO with Lys. Significantly, the reaction between α -dicarbonyl compounds and Lys preferentially generated CML and CEL, resulting in the lower concentrations of AAA compared to CML and CEL. CONCLUSION The multi-response kinetic model developed in the present study can be applied well to the Maillard reaction. The relationship between the formation mechanisms of CML, CEL and AAA is also explained. © 2024 Society of Chemical Industry.

Lysine-Polydopamine Nanocrystals Loaded with the Codrug Abemaciclib-Flurbiprofen for Oral Treatment of Cancer

Ting Sun,Faxing Zhang,Yuyi Xu,Xiaowei Wang,Jiajia Jia,Lihong Sang,Ji Li,Dongkai Wang,Zhiguo Yu

Nonsteroidal anti-inflammatory drugs (NSAIDs) combined with chemotherapeutic agents for the treatment of colorectal cancer (CRC) are a promising therapeutic strategy. NSAIDs can effectively boost the antitumor efficacy of chemotherapeutic agents by inhibiting the synthesis of COX-2. However, hazardous side effects and barriers to oral drug absorption are the main challenges for combination therapy with chemotherapeutics and NSAIDs. To address these issues, a safe and effective lysine–polydopamine@abemaciclib-flurbiprofen (Flu) codrug nanocrystal (Lys-PDA@AF NCs) was designed. Abemaciclib (Abe), a novel and effective inhibitor of the CDK4/6 enzyme, and Flu were joined to prepare Abemaciclib-Flu codrug (AF) by amide bonds, and then the AF was made into nanocrystals. Lysine-modified polydopamine was selected as a shell to encapsulate nanocrystals to enhance intestinal adhesion and penetration and lengthen the duration time of drugs in vivo. Nuclear magnetic resonance, Fourier transform infrared, Massspectrometry, X-ray photoelectron spectroscopy, Transmission electron microscopy, and drug loading were used to evaluate the physicochemical characteristics of the nanocrystals. In our study, Abe and Flu were released to exert their synergistic effect when the amide bond of AF was broken and the amide bond was sensitive to cathepsin B which is overexpressed in most tumor tissues, thus increasing the selectivity of the drug to the tumor. The results showed that Lys-PDA@AF NCs had higher cytotoxicity for CRC cell with an IC50 of 4.86 μg/mL. Additionally, pharmacokinetics showed that Abe and Flu had similar absorption rates in the Lys-PDA@AF NCs group, improving the safety of combination therapy. Meanwhile, in vivo experiments showed that Lys-PDA@AF NCs had excellent antitumor effects and safety. Overall, it was anticipated that the created Lys-PDA@AF NCs would be a potential method for treating cancer.

Regulation of fibrinolytic protein adsorption on polyurethane surfaces by modification with lysine-containing copolymers

Zengchao Tang,Xiaoli Liu,Yafei Luan,Wei Liu,Zhaoqiang Wu,Dan Li,Hong Chen

A new strategy to regulate the attachment of plasminogen and its activator, t-PA, to a polyurethane surface is reported. The method is based on the one-step graft copolymerization of a lysine-containing methacrylic monomer and hydroxyethyl methacrylate (HEMA). The copolymer-modified PU surfaces showed high capacity for plasminogen and t-PA binding. The release of pre-adsorbed t-PA from the lysine-containing PU surfaces in contact with plasma was also investigated. It was shown that the interactions of plasminogen and t-PA on this type of surface can be controlled simply by changing the monomer feed ratio, and thus the relative quantities of HEMA and lysine in the grafts. This approach may be useful for the development of fibrinolytic, blood-contacting materials and, more generally, for controlling protein-surface interactions for biocompatibility.
