MicroRNAs (miRNAs) are small molecules with significant regulatory functions that have served as biomarkers for the early diagnosis of disease. Accordingly, it is purposeful to develop a rapid, targeted assay for the sensitive, selective, and determination of target miRNAs. Single-band red emission of upconversion nanoparticles (UCNPs) offer high photostability, large anti-Stokes shifts, and low background. Additionally, improving the accuracy via dual-signal fluorescence is also effective for reducing false positive signals induced by background. Herein, taking advantage of the single-band red emission of UCNPs and DNA complementary nanotechnologies, a target-triggered self-assembled dual-signal response biosensor via a strand displacement cascade is reported. The reported approach was used for the determination of miRNA-222 (a model analyte) in serum and the results were similar to those obtained by the polymerase chain reaction (PCR). The spiked recoveries were from 92.5 to 108.4%, indicating that this biosensor has potential for accurate and sensitive determination of miRNAs in biological samples.

Acute detection and high-resolution imaging of microRNAs (miRNAs) in living cancer cells have attracted great attention in clinical diagnosis and therapy. However, current methods suffer from low detection sensitivity or heavy dependence on expensive and sophisticated spectrometers. Herein, a novel algorithm-assisted system of detecting and imaging miRNAs in living cancer cells was developed via the disassembly of plasmonic core-satellite probes coupled with strand displacement amplification (SDA). The target miRNAs in the system could trigger the disassembly of plasmonic core-satellite probes, leading to the color change in the scattering light of the probes, which could be captured by dark-field microscopy (DFM). The concentration of the target miRNAs was obtained by analyzing the dark-field image based on the proposed algorithm with a detection limit of 2 pM for miRNA-21. Thus, the performance in terms of simplicity and sensitivity of the system compared with one of the conventional spectrophotometers was well presented, which could inspire more clinical applications of inexpensive, intelligent, and rapid screening of cancer cells. The application software based on the proposed algorithm running on the Android platform was also developed, demonstrating the potential of remote diagnosis.

Herein, a novel photoelectrochemical (PEC) biosensor was developed for the ultrasensitive detection of dual microRNAs (miRNAs), with the detection being based on energy transfer (ET) between carbon quantum dots (CQDs) and gold nanoparticles (AuNPs). The PEC platform consisted of a [email protected] 2 C nanotube modified ITO electrode. Two hairpin probes (H1 and H2) carrying the Au NPs were used “switch off” and “switch on” the PEC signal of the CQDs, with a close approach of the tagged AuNPs to the CQDs quenching the PEC signal. The introduction of different miRNAs (miRNA-159b and miRNA-166a) altered the interparticle distance between the AuNPs and CQDs, thereby affecting the intensity of the PEC response. This approach allowed the highly sensitive detection of both miRNA-159b and miRNA-166a. The linear range of the biosensor for miRNA-159b and miRNA-166a detection were 0.5–5000 fM, with low detection limits of 0.15 fM and 0.21 fM, respectively. To our knowledge, this is the first reported CQDs-based ET biosensor for the PEC detection of dual miRNAs. Results suggest that this approach offers a promising platform for the ultrasensitive detection of multiple miRNAs.

A novel biosensing platform is proposed that integrates catalytic hairpin assembly (CHA) and hybridization chain reaction (HCR) cascade isothermal amplification with the CRISPR/Cas12a system, enabling ultrasensitive detection of microRNAs (miRNAs) targets. Within this platform, two modules are integrated: a target recognition and signal amplification module constructed by the cascade of CHA and HCR, and a signal transduction module in which the CRISPR/Cas12a system acts in concert with DNA probes loaded onto gold nanoparticles (AuNPs). This design achieves cascaded amplification from target recognition to signal output, thereby conferring high signal gain. Experimental results demonstrate that the proposed biosensor had high sensitivity toward the target miRNA, with a detection limit as low as 37 fM. Moreover, it shows remarkable single-base discrimination capability, effectively distinguishing sequences with single-nucleotide mismatches. Notably, the sensor maintains stable and reliable performance in complex biological matrices, including serum samples and lysates from various tumor cells. This strategy effectively couples signal amplification with the CRISPR system, achieving both high sensitivity and specificity, making it a useful tool for miRNA detection and early cancer screening. A high-throughput and sensitive fluorimetric analysis method has been developed with wettable microwells array for probing short-chain microRNAs (miRNAs) in blood using silver nanoclusters (AgNCs) with red fluorescence amplified and stabilized by metal organic frameworks (MOFs). Glass slides were first spotted with polyacrylic acid to form hydrophilic microdots and then patterned with hydrophobic hexadecyltrimethoxysilane, followed by etching the microdots to yield the microwells array. Furthermore, the DNA capture probes with silver-binding G sequences were covalently bound onto the functionalized microwells to hybridize with targeting miRNAs. Exonuclease I catalytic digestion was then conducted to remove any single-strand DNA probes unhybridized. Eventually, AgNCs were applied to specifically recognize the G sequences of DNA probes survived to be further coated with MOFs of zeolitic imidazolate framework (ZIF-8). Unexpectedly, the red fluorescence of AgNCs probes could be dramatically enhanced due to the “electron-donor effect” of nitrogen-containing ligands of ZIF-8 coatings, together with improved sensing stability. High detection sensitivity and reproducibility could thereby be expected for detecting miRNAs in the blood with the concentrations linearly ranging from 0.175 to 500 pM. Markedly different from the common sandwiched DNA detection methods, the developed fluorimetric strategy using AgNCs coated with MOFs and DNA probes designed with silver-recognizing sequences would be tailored for quantifying various kinds of short-chain miRNAs with low levels in the complicated blood media.

Detection of microRNAs (miRNAs) reveals tumor information at the transcriptional level, which is crucial for early diagnosis and prognosis of tumors. However, the rapid detection of miRNAs in biological samples without relying on large equipment remains a significant challenge. In this work, a double-layer NiCo heterostructure (DL-H) for enhancing the reaction efficiency of DNA labels, and dual-target cycling reaction system were developed to improve detection sensitivity and shorten the detection time within 30 min. The DL-H and catechol provide favorable condition for methylene blue (MB) oxidation and single-electron transfer of ferrocene (Fc). The target cycling reaction allows one target to release multiple Fc/MB labels from the electrode surface, enhancing the signal differentiation triggered by the target. The obtained sensor possesses a linear detection range of 10 fM-900 pM for miRNA-574-5p and miRNA-1254. The detection limit is 302 aM for miRNA-574-5p, while 85 aM for miRNA-1254. And the detection results of clinical samples present a strong correlation with the PCR. More importantly, a sensing automatic preparation-detection system was developed based on microfluidic chip, to resolve the reliance on highly trained technicians for the electrochemical sensing preparation. This development provides a valuable foundation for the advancement of portable detection tool of tumor biomarker.

Previous studies have shown that maternal resveratrol improved growth performance and altered the microbial composition of suckling piglets under hot summer conditions. However, it remains unclear how maternal resveratrol improves growth performance of suckling piglets under high summer temperatures. A total of 20 sows (landrace × large white; three parity) were randomly assigned to 2 groups (with or without 300 mg/kg resveratrol) from d 75 of gestation to d 21 of lactation in a high temperature environment (from 27 to 30 °C). The results showed that maternal resveratrol supplementation increased total daily weight gain of piglets under hot summer conditions, which is consistent with previous studies. Furthermore, we found that maternal resveratrol improved the intestinal morphology and intestinal epithelial proliferation in suckling piglets. Dietary resveratrol supplementation affected the characteristics of exosome-derived microRNAs (miRNAs) in sow colostrum, as well as the genes targeted by differentially produced miRNAs. MiRNAs are concentrated in the tight junction pathway. As a result, the expression of intestinal tight junction proteins was increased in suckling piglets ( P < 0.05). Notably, maternal resveratrol increased the intestinal secretory immunoglobulin A (sIgA) levels of suckling piglets via colostrum immunoglobins ( P < 0.05), which could increase the abundance of beneficial microbiota to further increase the concentration of short chain fatty acids (SCFA) in suckling piglets' intestine ( P < 0.05). Finally, our correlation analysis further demonstrated the positive associations between significantly differential intestinal microbiota, intestinal sIgA production and SCFA concentrations, as well as the positive relation between total daily weight gain and intestinal health of suckling piglets. Taken together, our findings suggested that maternal resveratrol could promote intestinal health to improve piglet growth under high summer temperatures, which might be associated with the immunoglobin and exosome-derived miRNAs in sows’ colostrum.

An Exo-III-triggered target-reconstructed DNAzyme walker enables ultrasensitive multiplex miRNA detection. • Cascade amplification integrates Exo-III recycling, polymerase/endonuclease cycling, and DNAzyme walking. • Orthogonal Pt, Ir, Rh tags allow simultaneous ICP-MS quantification of three miRNAs without spectral cross-talk. • Detection limits reach 0.15 fM (miR-21), 0.32 fM (miR-1246), and 0.26 fM (miR-20a) with excellent linearity. • The method discriminates single-base mismatches and performs well in diluted human serum (92–108% recovery). Cervical cancer remains a leading cause of cancer-related mortality among women, and the prognosis of patients after radiotherapy varies considerably due to differences in tumor radiosensitivity and treatment response. Reliable biomarkers for post-radiotherapy prognosis are therefore critically needed. MicroRNAs (miRNAs) have emerged as promising non-invasive indicators, yet accurate detection is hindered by their low abundance and the lack of multiplexing capability in conventional methods. Herein, we report a highly sensitive and specific platform for simultaneous detection of three cervical cancer radiotherapy-associated miRNAs (miR-21, miR-1246, and miR-20a) by integrating an Exo-III-triggered target-reconstructed DNAzyme walker with inductively coupled plasma mass spectrometry (ICP-MS). The strategy employs a structurally locked DNA walker that is sequentially activated through Exo-III-mediated target recycling and DNA polymerase/endonuclease-assisted cyclic amplification, ultimately unleashing a DNAzyme walker to cleave element-labeled substrates. Using orthogonal platinum group element tags (Pt, Ir, Rh), the released tags are quantified by ICP-MS without spectral cross-talk. The method achieves detection limits as low as 0.15 fM, 0.32 fM, and 0.26 fM for miR-21, miR-1246, and miR-20a, respectively, with excellent single-nucleotide discrimination and negligible cross-reactivity. Successful spike-recovery experiments in diluted human serum validate its practical feasibility. This cascade amplification strategy offers a robust, multiplex, and ultrasensitive tool for non-invasive liquid biopsy-based prognostic assessment of post-radiotherapy cervical cancer, and can be readily adapted for other disease-related miRNA panels.
