PLLA/(MWCNTs-ZnO)@PDA composite with NIR-light induced shape memory effect, antibacterial properties and 3D printability

Yulong Zou,Beibei Zhou,Wenqiang Guo,Zhijie Sun,Xili Lu,Li Li

PLLA based shape-memory polymer composites with multifunctions have attracted growing attention in biomedical applications. Generally, a single nanofiller has the problem of insufficient functionalization and easy aggregation. Therefore, we design and fabricate polydopamine (PDA) surface modified MWCNTs-ZnO hybrid nanostructure ((MWCNTs-ZnO)@PDA). The dispersion of (MWCNTs-ZnO)@PDA in PLLA is improved in comparison with the pristine MWCNTs or ZnO. The results indicate that the (MWCNTs-ZnO)@PDA nanosystem shows a synergistic effect on the mechanical properties and photothermal properties of the composites. Comparative analysis with neat PLLA demonstrates a remarkable 17% increase in tensile strength and a 19% increase in elongation at break for PLLA/(MWCNTs-ZnO)@PDA composite. Additionally, the composites exhibit excellent NIR light-induced shape memory effects with a shape recovery ratio reaching 100%. Moreover, PLLA/(MWCNTs-ZnO)@PDA composites show favorable antibacterial activity against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ). Furthermore, the polymer composites display good 3D printability, and the 3D-printed porous scaffold possesses high compressive strength and modulus. Also, the PLLA/(MCNTs-ZnO)@PDA scaffold can be compressed into a compact size and recover its original shape under NIR light irradiation. This work provides an approach to prepare novel shape-memory polymer composites with multifunctions including high strength, light-induced shape-recovery, antibacterial properties and printability.

NIR-activatable proenzyme DSF/CuS@M for synergistic in situ chemotherapy/CDT/PTT/cuproptosis-based antitumor therapy

Lushuang Xu,Qinyue Chen,Yangwei Qian,Jin Wang,Chenxi Wang,Rui Fan,Jia-Wei Shen,Qiying Shen

Nanozymes provide therapeutic benefits against a diverse range of malignant tumors. However, their antitumor potencies are seriously impaired by their low catalytic efficiencies and the off-target toxicities caused by their “always-on” catalytic activity. Herein, we developed multifunctional nanoparticles co-loaded with ultra-small copper sulfide nanoparticles (CuS NPs) and disulfiram (DSF) (DSF/CuS@M). These nanoparticles had peroxidase-like activity and served as (near-infrared) NIR-activatable proenzymes to achieve synergistic in situ chemotherapy, chemodynamic therapy (CDT), photothermal therapy (PTT), and cuproptosis-based antitumor therapy. DSF/CuS@M not only provided PTT as NIR-activatable proenzymes but also enabled complete DSF and Cu 2+ release. Non-toxic DSF was converted into diethyldithiocarbamic acid (DDC) through a glutathione (GSH)-mediated metabolic reaction, leading to the generation of cytotoxic Cu(DDC) 2 in situ , which enhanced cell apoptosis. Cu 2+ was converted into Cu + by GSH, and this Cu + catalyzed the conversion of H 2 O 2 into hydroxyl radicals (·OH) to induce CDT. Moreover, Cu + could also bind to dihydrolipoyl transacetylase (DLAT) in the tricarboxylic acid (TCA) cycle, thereby inducing cuproptosis. These results demonstrate that the NIR-activatable proenzyme developed in this study is effective and safe for synergistic therapy against malignant tumors.

NIR-Propelled Thermosensitive Bowl-Shaped Nanomotors with High Penetration and Targeting for Photoacoustic Imaging Guided Thrombolysis Therapy

Yina Su,Linjie Huang,Guizhen Xu,Simin Chen,Jiaqiong Wu,Siyu Wang,Yichao Zhang,Xiahui Lin

Traditional antithrombotic therapeutic strategies encounter challenges including heightened bleeding risks, short circulation times, low targeting ability, and inferior thrombus penetration. Therefore, a novel thrombolysis nanodrug (APBUL) is designed that incorporates urokinase (UK) loaded onto the surface of bowl-shaped nanomotors (APBs) encapsulated within fibrin peptide (CREKA)-modified thermosensitive liposomes, presenting an innovative therapeutic platform for thrombolysis. APBUL leverages CREKA's targeting ability for thrombus accumulation. Subsequently, under the irradiation of near-infrared light, the thermosensitive liposomal shell undergoes controlled disruption, releasing internal APBs and UK. Then, the APBs move directionally though thermophoresis effect, facilitating photothermal therapy and deep thrombus penetration, and synergistically enhancing UK release and diffusion to optimize thrombolysis. Moreover, the APBUL possesses a catalase-like activity, catalyzing hydrogen peroxide into oxygen to alleviate oxidative stress and inflammatory factors at the thrombus site, thereby lowering the recurrence risk. Combined with the ability of APBUL's photoacoustic imaging, this new strategy is expected to provide an inspiring idea for the integrated use of clinical thrombolytic therapy in diagnosis, imaging, and treatment.

NIR-II light-activated and Cu nanocatalyst-enabled bioorthogonal reaction in living systems for efficient tumor therapy

Hui Huang,Wendi Xuan,Jiakang Hai,Xue Wang,Meng Chen,Guobing Hong,Xinyue Dai,Lili Xia,Wei Feng,Yu Chen

Bioorthogonal reaction refers to chemical reactions that occur within a biological system without interfering the normal biochemical process, offering the unprecedented versatility in engineering chemical reactions within cells. However, the precise regulation of bioorthogonal reaction in living systems is mired by the complexity of the physiological environment and the toxicity of catalysts. Herein, considering the deeper tissue penetration and reduced phototoxicity compared to visible light and ultraviolet, a second near infrared (NIR-II) light-activated Cu-based bioorthogonal reaction is developed to achieve precise spatiotemporal control and effective switching for Cu(I)-catalyzed azide-alkyne cycloaddition (CuAAC) mediated chemical transformations in tumor, reducing the off-target effects. The catalytic activity of Cu catalyst through valence state interconversion between Cu(II) and Cu(I) can be precisely regulated in a reversible manner under NIR-II light irradiation-induced photoelectron transfer, which controls the extent of desired drug synthesis in bioorthogonal reaction. Meanwhile, the adverse effects of Cu(I) can be substantially mitigated within normal tissues due to their oxygen-rich condition. By utilizing NIR-II light and oxygen level, the Cu bioorthogonal catalyst achieves a balance between catalytic activity and biocompatibility. The ability to achieve precise spatiotemporal control and reversible catalysis makes this NIR-II light-mediated CuAAC platform an efficient and adaptable tool for bioorthogonal chemistry in living systems.

NIR-II photo-accelerated polymer nanoparticles boost tumor immunotherapy via PD-L1 silencing and immunogenic cell death

Tian Zhang,Dongsheng Tang,Pengfei Wu,Shaoping Jiang,Yuquan Zhang,Abid Naeem,Yong Li,Chunhui Li,Bo Hu,Shuai Guo,Caixia Sun,Haihua Xiao,Ran Yan,Yuhua Weng,Yuanyu Huang

Immune checkpoint blockade (ICB) therapy is a widely favored anti-tumor treatment, but it shows limited response to non-immunogenic “cold” tumors and suffers from drug resistance. Photodynamic therapy (PDT), as a powerful localized treatment approach, can convert a “cold tumor” into a “hot tumor” by inducing immunogenic cell death (ICD) in tumor cells, thereby enhancing tumor immunogenicity and promoting tumor immunotherapy. However, the effectiveness of PDT is largely hindered by the limited penetration depth into tumor tissues. To address these issues, we proposed an all-in-one drug system with NIR-II photo-accelerated PDT effects, efficient immune checkpoint gene silencing, and a facile manufacturing process. The so-called all-in-one drug system comprises a multi-modal designed polymer PPNP and siRNA. PPNP is an amphipathic polymer that includes the near infrared-II (NIR-II) photosensitizer Aza-boron-dipyrromethene (Aza-BODIPY), a glutathione (GSH)-cleavable linker, and a cationic monomer derived from cholesterol. PPNP can self-assemble and efficiently load siRNA. Under laser irradiation, PPNP triggers a potent ICD cascade, causing the on-demand release of siPD-L1, reshaping the tumor's immunosuppressive microenvironment, effectively inhibiting the growth of various tumors, and stimulating the immune memory. This study represents a generalized platform for PDT and gene silencing, designed to modulate immune-related signaling pathways for improved anticancer therapy.

Tumor-specific enhanced NIR-II photoacoustic imaging via photothermal and low-pH coactivated AuNR@PNIPAM-VAA nanogel

Sun Xiaodong,Li Yujie,Liu Xiaowan,Cui Dandan,Shi Yujiao,Huang Guojia

Background Properly designed second near-infrared (NIR-II) nanoplatform that is responsive tumor microenvironment can intelligently distinguish between normal and cancerous tissues to achieve better targeting efficiency. Conventional photoacoustic nanoprobes are always “on”, and tumor microenvironment-responsive nanoprobe can minimize the influence of endogenous chromophore background signals. Therefore, the development of nanoprobe that can respond to internal tumor microenvironment and external stimulus shows great application potential for the photoacoustic diagnosis of tumor. Results In this work, a low-pH-triggered thermal-responsive volume phase transition nanogel gold nanorod@poly(n-isopropylacrylamide)-vinyl acetic acid (AuNR@PNIPAM-VAA) was constructed for photoacoustic detection of tumor. Via an external near-infrared photothermal switch, the absorption of AuNR@PNIPAM-VAA nanogel in the tumor microenvironment can be dynamically regulated, so that AuNR@PNIPAM-VAA nanogel produces switchable photoacoustic signals in the NIR-II window for tumor-specific enhanced photoacoustic imaging. In vitro results show that at pH 5.8, the absorption and photoacoustic signal amplitude of AuNR@PNIPAM-VAA nanogel in NIR-II increases up obviously after photothermal modulating, while they remain slightly change at pH 7.4. Quantitative calculation presents that photoacoustic signal amplitude of AuNR@PNIPAM-VAA nanogel at 1064 nm has ~ 1.6 folds enhancement as temperature increases from 37.5 °C to 45 °C in simulative tumor microenvironment. In vivo results show that the prepared AuNR@PNIPAM-VAA nanogel can achieve enhanced NIR-II photoacoustic imaging for selective tumor detection through dynamically responding to thermal field, which can be precisely controlled by external light. Conclusions This work will offer a viable strategy for the tumor-specific photoacoustic imaging using NIR light to regulate the thermal field and target the low pH tumor microenvironment, which is expected to realize accurate and dynamic monitoring of tumor diagnosis and treatment.

Synergistic Cancer Therapy: An NIR-Activated Methylene Blue-Nitrogen Mustard Prodrug for Combined Chemotherapy and Photodynamic Therapy

Tianyu Zhu,Jipeng Ding,Fan Zheng,Yanpeng Fang,Wenzhi Huang,Ying Yin,Wenbin Zeng

Nitrogen mustard, a widely used chemotherapeutic agent for more than 70 years, exhibits significant efficacy. However, its clinical applications are severely limited by poor tumor selectivity and severe side effects on normal tissues. To address these limitations, we developed a near-infrared (NIR)-activatable nitrogen mustard prodrug, MBNM. Upon NIR irradiation, the controlled cleavage of the urea bond within MBNM facilitates the simultaneous release of nitrogen mustard and methylene blue (MB), enabling a synergistic approach combining chemotherapy and photodynamic therapy (PDT) for effective tumor suppression. Moreover, the release of MB upon activation enables real-time monitoring of prodrug activation. Notably, MBNM demonstrated significantly improved biosafety compared to free nitrogen mustard. These findings suggest that the photocleavable prodrug MBNM offers a promising strategy for safe and effective combination cancer therapy.

Cross relaxation-enhanced NIR-Ⅱ fluorescence in negative thermal expansion phosphor Sc2(MoO4)3 for thermal sensing

Xiuna Tian,Hongjian Dou,Youfeng Wen,Lingyuan Wu,Changjun Wang,Zongwei Zhang,Youzi Zhang

In the field of disease diagnosis and treatment, achieving highly sensitive temperature sensing via non-contact methods based on fluorescence continues to present a significant challenge. This study investigated the influence of the cross-relaxation (CR) process of Tm 3+ ions at varying concentrations and the negative thermal expansion (NTE) property of the matrix on the luminescent properties of Sc 2 (MoO 4 ) 3 :Yb 3+ /Tm 3+ materials under diverse temperature conditions, and further evaluated their potential application value in the field of temperature sensing. At room temperature, the analysis of the concentration dependence of the emission spectra reveals that the intensities of the emission peaks originating from higher energy levels ( 1 G 4 , 3 H 4 , and 3 H 5 ) and the lowest excited state ( 3 F 4 ) exhibit significant variation with the doping concentration. As the doping concentration increases, the temperature enhancement factor for the fluorescence intensity of high-energy level transition ( 1 G 4 → 3 H 6 ) progressively decreases from 21.6 to 1.2. In contrast, the temperature enhancement factor for the lowest excited state transition ( 3 F 4 → 3 H 6 ) markedly increases from 471 to 2964. At a concentration of 8%, the temperature extension factor of the fluorescence lifetime at 1627 nm ( 3 F 4 → 3 H 6 ) can reach 7.2, whereas at a concentration of 0.5%, this factor is 2.7. These suggests that the CR process between Tm 3+ ions exerts markedly distinct influences on the luminescence intensities and lifetime in Sc 2 (MoO 4 ) 3 :Yb 3+ /Tm 3+ materials. In addition, this study employs the fluorescence intensity ratio of I 1629 nm / I 1197 nm for temperature sensing, thereby achieving high sensitivity in temperature measurement.

Multiple amplifications of NIR-I ECL signals of Au6PtAg nanoclusters and applications in fast detections of noradrenaline

Fanfan Guo,Yuanyuan Hu,Huiwen Zhou,Yuanjin Li,Jiadi Li,Guichuan Wei,Jing Long Han,Wenyue Gao,Sisi He,Wenxin Niu,Ling Zhang

NIR-I electrochemiluminescence (ECL) is emerging as highly sensitive detection strategy due to its low background at the ambient conditions. Nonetheless, the applications of current NIR-I ECL nanoprobes are limited to the harsh synthetic/testing condition, high toxicity, low signal, and poor biocompatibility. In this study, glutathione/lipoic acid-protected trimetallic Au 6 PtAg nanoclusters (GSH/LA/Au 6 PtAg NCs) are demonstrated as an efficient nanoprobe with strong ECL signals around 850 nm in the presences of triethylamine. Critically, the alloying of Ag  +  into the bimetallic Au 2.5  Pt NCs altered the electronic structures of Pt(II), valences of Au(0), and the zata potentials of NCs, promoting the ECL intensities at 850 nm by 1.0 time. Additionally, the partly ligand-exchange of GSH by LA further increased the ECL intensities at 850 nm by 2.2 times through accelerating electron transfer processes. Meanwhile, the NIR-I fluorescence ( λ emit  = 910 nm) was enhanced. More importantly, the utilization of GSH/LA/Au 6 PtAg NCs μg −1 is enhanced by 3000 times after embedding into SiO 2 microspheres ( D , 318 nm). Because noradrenalin has substantial inhibition effects to NIR-I ECL of GSH/LA/Au 6 PtAg/SiO 2 , 1.0 nM−0.1 mM noradrenalin was detected with the low limit of detection as 0.04 nM (S/N = 3) selectively. Biomolecules, glucose, ascorbic acid, or uric acid didn't interrupt the detection of noradrenalin. The sensitivity of this ECL method to detect noradrenalin is much higher than those in previous reports, enabling the early diagnosis of neurogenic diseases.

Morphology-Preserved Ag Alloying in Au Nanosheets Enables Tunable NIR-I/II Plasmonics for Enhanced Photothermal Conversion

Qi He,Qiuping Yang,Xiangyu Tong,Xiaotong Zhang,Yi Wang,Xiaohu Wu,Yanyun Ma,Yiqun Zheng

Precise spectral modulation of plasmonic absorptions in the near-infrared (NIR-I and NIR-II) regions is crucial for matching laser wavelengths in practical applications, driving significant interest in sensing, photothermal therapy, and bioimaging. In this work, we show that silver (Ag) alloying in gold (Au) nanosheets provides tunable plasmonic absorption across both NIR-I (700–950 nm) and NIR-II (1000–1350 nm) spectral windows while preserving their intrinsic two-dimensional morphology. Through controlled variation of the Ag/Au ratio in a seed-mediated synthesis, we observe a continuous blue-shift of the in-plane dipole plasmon resonance from 1000 nm (NIR-II) to 700 nm (NIR-I). Theoretical simulations confirm that Ag incorporation tunes plasmonic responses by modulating both the free electron density and local dielectric environment. This morphology-preserved alloying approach establishes a versatile platform for developing optically switchable nanomaterials with promising applications in multiplexed biosensing and wavelength-selective photothermal therapy.

Long-term dynamic tracking of acute kidney injury by Au nanoclusters-based reversible NIR-II fluorescent probe

Qing Hu,Fanyi Qin,Huijiao Qu,Huayu Xiong,Shengfu Wang,Xiuhua Zhang,Yan Xiao

Acute kidney injury (AKI) is a common kidney disease characterized by a sudden decline in renal function. It usually has no clear clinical symptoms in its early stages, but can easily progress to renal failure or even result in death. Real-time monitoring of AKI progression significantly enhances the prospects for early diagnosis and prompt intervention. However, there is no available to monitor AKI as developing a renal-clearable and reversibly responsive probe capable of long-term in vivo track biomarkers of AKI, such as reactive oxygen species (ROS), remains an extremely challenging task. Herein, we have constructed a redox-reversible second near-infrared (NIR-II) fluorescent probe based on Au nanoclusters (Au NCs) for long-term dynamic monitoring of AKI. The probe features NIR-II excitation and emission capabilities, along with effective kidney targeting and prolonged retention, facilitating renal enrichment and enabling long-term high-resolution renal imaging. Notably, it demonstrates rapid and reversible responsiveness to ROS with high sensitivity and selectivity, permitting real-time monitoring of redox fluctuations in vivo. Given its exceptional performance, the probe achieves unprecedented real-time dynamic tracking of AKI progression, providing crucial redox status information during the disease process. Additionally, the probe can serve as an effective new tool for screening of potential therapeutic drugs for AKI. This work may offer some substantial promotion for the diagnosis and treatment of kidney diseases.
