A cyclable detection of histamine by using the DNA microfluidic chip with high SERS performance

Yansheng Liu,Yunjie Zhan,Haonan Hu,Guofu Wang,Xiaobo Jia,Jin Zhou,Hongqi Li,Haixin Chang,Zhaoxu Li

This study reports a SERS-based microfluidic chip for the cyclic detection of histamine with high specificity. Easily fabricated Au nanoparticles (AuNPs) were synthesized on Si substrates by using a thermally treating approach. The designed probe DNA (pDNA) functionalized AuNPs is used as a sensing surface. The Cyanine 3 (Cy3) functionalized complementary DNA (cDNA) is modified on the chemically synthesized AuNPs and the complex cDNA/AuNPs are used as nanotags. Due to the hybridization between the pDNA and the cDNA, nanotags-pDNA/AuNPs structure is formed and illustrates a high SERS intensity of Cy3. With the presence of the histamine, due to the stronger affinity between cDNA and histamine, nanotags-pDNA/AuNPs structure is destroyed and the nanotags are separated from the sensing surface resulting in a decrease in the SERS intensity of Cy3. By adding fresh nanotags, nanotags-pDNA/AuNPs structure is regenerated, and a new detection cycle can be performed. By utilizing the proposed SERS-based microfluidic chip, the limit of detection (LOD) of histamine in DI water is 1 × 10 −9  g/mL. The concentration is well below the U.S. Food and Drug Administration (FDA) histamine toxicity threshold of 5 × 10 −5  g/mL. Moreover, the proposed SERS-based microfluidic chip can be reused 7 times. Furthermore, this chip has accuracy and applicability were validated through its application in beer samples. Indicating that this method has great potential for histamine detection.

Monitoring Spatiotemporal Dynamics of Extracellular pH in Breast Tumor Spheroid and Tissue by SERS Microsensors

Zedong Zhang,Chunyan An,Jianguo Dong,Xuyu Zhang,Tianyu Shen,Dekun Wang,Xue Mi,Shijing Yue,Xiaoyue Tan,Yuying Zhang

Extracellular pH (pHe) acidification is a hallmark of solid tumors, linked to cancer initiation, progression, and drug resistance. Non-invasive monitoring of pHe spatiotemporal dynamics during cancer growth and in response to drug is crucial for understanding cancer mechanisms and developing novel therapeutic strategies. Here, a micro-sized pH sensor is developed by encapsulating Au@Ag@MBA nanosensors within biocompatible agarose hydrogel microspheres. These Aga microsensors in a size range of 3–10 µm, may avoid cellular endocytosis and stably reside in the extracellular environment. pHe dynamics are monitored via surface-enhanced Raman scattering (SERS) mapping in tumor spheroids and solid tumor tissues over 7 and 25-day periods, respectively. Distinct pH gradients are observed between the outer and core regions of the tumor spheroid, which evolved over time and are disrupted by doxorubicin treatment. In vivo, a decrease of average tumor pHe from ≈6.2 to ≈5.5 is observed with tumor progression, and doxorubicin administration resulted in a pHe elevation of ≈0.6 units. This microsensor-based SERS sensing approach provides a facile method for long-term monitoring of spatiotemporal dynamics within the disease microenvironment.

Dual-mode SERS and colorimetric sensor for lung cancer VOC-biomarker detection using hydrogel patches

Jialin Li,Shaowei Liu,Amil Aligayev,Hao Jiang,Muhammad,Shenhao Wang,Xin Yu,Jin Tao,Jiaqi Wang,Agnieszka Jastrzębska,Qing Huang

Hexanal, a volatile organic compound (VOC), is a potential biomarker for the early detection of lung cancer. In this study, we developed a dual-mode flexible biosensor that integrates surface-enhanced Raman scattering (SERS) and colorimetric detection for the quantitative analysis of hexanal in human exhaled breath. The biosensor employs Ag nanocubes wrapped with Co-Ni layered double hydroxide (AgNCs@Co-Ni LDH) as a functional matrix, offering both superior SERS enhancement and efficient VOC adsorption properties. To achieve selective detection, AgNCs@Co-Ni LDH was incorporated into agarose hydrogels along with 3-methyl-2-benzothiazolinone hydrazone (MBTH). The resulting MBTH-AgNCs@Co-Ni LDH/hydrogel patches facilitate the oxidation of hexanal, enabling simultaneous colorimetric and SERS signal generation, while producing acrizine, a blue-colored reaction product. This hydrogel-based dual-mode sensing platform exhibits high selectivity, excellent stability, and precision in SERS-based hexanal detection. The detection limit for the SERS method was determined to be as low as 3.34 × 10 −13  M. Furthermore, developed and optimized compact CNN-based multi-terminal intelligent recognition system for enhanced hydrogel-patch detection through AI-driven, portable, and real-time colorimetric analysis. Therefore, this work not only enables the effective detection of hexanal in the exhaled breath of suspected lung cancer patients, underscoring its potential for early lung cancer screening, but also establishes a foundation for the development of multimodal hydrogel biosensors for broader applications in disease diagnosis.

Dongjie Zhang,Zhaoyang Cheng,Yali Song,Huandi Li,Lin Shi,Nan Wang,Yingwen Peng,Renan Chen,Nianzheng Sun,Min Han,Fengjiao Hu,Chuntao Zong,Rui Zhang,Si Chen,Conghui Zhu,Xiaoli Zhang,Xiaobo Li,Xiaopeng Ma,Changbei Shi,Xiaofei Zhang,Rui Liu,Ziqi Ren,Lin Wang,Qi Zeng,Tingting Zeng,Xueli Chen

Rapid identification and accurate diagnosis are critical for individuals with acute leukemia (AL). Here, we propose a combined deep learning and surface-enhanced Raman scattering (DL-SERS) classification strategy to achieve rapid and sensitive identification of AL with various subtypes and genetic abnormalities. More than 390 of cerebrospinal fluid (CSF) samples are collected as targets, encompassing healthy control, AL patients, and individuals with other diseases. Sensitive SERS detection could be achieved within 5 min, using only 0.5 μL volume of CSF. Through integrated feature fusion (1D spectra and 2D image) with a transformer model, the classification method is developed to screen and diagnose AL patients, demonstrating exceptional classification performances of accuracy, sensitivity, specificity, or reliability. Also, this approach demonstrates remarkable versatility and could be extended to the classifications of meningitis diseases. The sensitive DL-SERS classification platform has the potential to be a powerful auxiliary in vitro diagnostic tool.

Continuous and Online Detection of Melamine, 6-Thioguanine and Histamine Using Silver Colloid-Based SERS

Hao Zhang,Wanyi Li,Ying Zhong,Meng Zhang,Kaile He,Yihan Yuan,Jinyi Wang

Surface-enhanced Raman scattering (SERS) based on noble metal nanoparticles has garnered significant attention in analytical chemistry due to its simplicity and rapidity for low-concentration detection. Although widely applied in fields like environmental monitoring, food safety, and biomedical diagnostics, the impact of silver nanoparticle shape on SERS performance remains insufficiently studied. Furthermore, traditional stationary solid and liquid SERS approaches are limited by inconsistent reproducibility and temporal variations in uniformity, respectively. To overcome these limitations, we introduce a simple continuous and online Ag SERS platform that employs spherical silver colloids within a capillary-Teflon tube system for dynamic, real-time analyte detection. Continuous fluid flow reduces variations in colloids, ensuring uniformity, while the enclosed system limits dynamic behaviors of colloids, enhancing stability. This platform demonstrates outstanding stability, with relative standard deviations of 2.30%. Its versatility is showcased through the quantitative detection of melamine in milk, 6-thioguanine in serum, and histamine in beer, demonstrating its capability for accurate, real-time detection in complex food matrices. This continuous and online Ag SERS platform represents a significant advancement in reliable, high-performance analyte detection, offering promising prospects for real-time analytical applications and providing a cost-effective, scalable solution for widespread use.

Selective SERS detection of dopamine in human urine using catechol oxidase-mimicking AgNPs@MOF-818

Shuojiang Liu,Gongke Li,Yuling Hu

The integration of surface-enhanced Raman scattering (SERS) and nanozymes has proven to be a highly effective strategy in bioanalytical applications, owing to its superior capabilities in signal transduction and amplification. Nevertheless, clinical applications still face obstacles, particularly in achieving sufficient selectivity. In this work, we have presented a SERS strategy for the selective detection of dopamine in human urine with MOF-818 which exhibits selective catechol oxidase-like activities. AgNPs@MOF-818 catalyze the oxidation of o -phenylenediamine (OPD) to diaminobenzazine (DAP) molecules, when dopamine (DA) is present, the oxidation of OPD is inhibited while DA is catalyzed into o -quinones, thereby suppressing the SERS signal intensity. An indirect SERS detection strategy of dopamine with high sensitivity and selectivity was established. The catechol oxidase-like activity of MOF-818 provides ideal selectivity to dopamine, while the OPD-dopamine catalytic system induces significant changes in SERS intensity. The detection protocol was subsequently optimized to achieve high sensitivity for dopamine, yielding a linear response range of 0.1–30 μmol/L and a low limit of detection (LOD) of 0.04 μmol/L. The method was successfully applied to the quantitative analysis of dopamine in human urine samples, demonstrating excellent recovery rates and minimal relative error in comparison with results obtained by commercial dopamine ELISA kit. This strategy presents a cost-effective, efficient, and accurate approach for dopamine detection while delivering insights into the integration of selective multifunctional nanozymes with sensitive SERS technique.

Lingli Zhang,Yilin Yu,Xinyi Zhu,Liping Song,Yu Zhao,Lei Jiang,Xiaoyu Zhao,Bihu Lv,Yixuan Sun,Youju Huang

Electromagnetic hotspot engineering plays a crucial role in surface-enhanced Raman scattering (SERS) biosensors . However, the construction of optical hotspots with high density and the concentration of analytes into hotspot centers remain challenging tasks. Additionally, achieving high sensitivity and signal reproducibility in SERS biosensors is an ongoing challenge. In this study, we utilise Janus micro/nano arrays to develop a superhydrophobic SERS platform. The high roughness of these arrays facilitates the distribution of rich hotspots ranging from nano to microscale and encourages the concentration of analytes within hotspot centers. The Janus plasmonic micro/nano arrays were meticulously crafted through the synergy of site-specific growth and instant interfacial assembly of nanoparticles (NPs). In the realm of SERS measurements targeting organic dyes in river water , our sensing platform demonstrates high sensitivity (limit of detection (LOD): 10 −9 M) and signal reproducibility (relative standard deviation (RSD)=4.25   %). Furthermore, the SERS platform exhibited remarkable sensitivity to multiple dyes in river water . Thus, our superhydrophobic SERS platform lays the groundwork for sensitive analysis in diverse fields such as environmental analysis and healthcare monitoring.

Stabilizing Enzymes in Plasmonic Silk Film for Synergistic Therapy of In Situ SERS Identified Bacteria

Zhangkun Liu,Shengkai Li,Zhiwei Yin,Zhaotian Zhu,Long Chen,Weihong Tan,Zhuo Chen

Increasing antibiotic resistance becomes a serious threat to public health. Photothermal therapy (PTT) and antibacterial enzyme-based therapy are promising nonresistant strategies for efficiently killing drug-resistant bacteria. However, the poor thermostability of enzymes in PTT hinders their synergistic therapy. Herein, antibacterial glucose oxidase (GOx) is embedded in a Ag graphitic nanocapsule (Ag@G) arrayed silk film to fabricate a GOx-synergistic PTT system (named silk-GOx-Ag@G, SGA). The SGA system can stabilize GOx by a vitrification process through the restriction of hydrogen bond and rigid β -sheet, and keep the antibacterial activity in the hyperthermal PTT environment. Moreover, the arrayed Ag@G possesses excellent chemical stability due to the protection of graphitic shell, providing stable plasmonic effect for integrating PTT and surface enhanced Raman scattering (SERS) analysis even in the GOx-produced H 2 O 2 environment. With in situ SERS identification of bacterial intrinsic signals in the mouse wound model, such SGA realizes superior synergistic antibacterial effect on the infected Escherichia coli , Staphylococcus aureus , and methicillin-resistant Staphylococcus aureus in vivo, while without causing significant biotoxicity. This system provides a therapeutic method with low resistance and in situ diagnosis capability for efficiently eliminating bacteria.

Dongjian Li,Mulinde Timothy Ezra,Haoran Li,Yifei Chen,Chengyun Si,Xuefang Luo

Neonicotinoids (NEOs) are widely used in agricultural cultivation, and the presence of residual NEOs poses a serious threat to consumer health. Therefore, trace detection of NEOs is particularly important. In this study, a response surface experimental design was employed to develop AgNP with high enhancement factor and surface height anisotropy, effectively enhancing the Raman signals of acetamiprid (ACE) and thiacloprid (THI) through densely populated hotspots on the surface. Gaussian simulation calculations were used to assign peaks in the ACE and THI spectra. Subsequently, convolutional neural network, backpropagation neural network, and AlexNet neural network were employed to identify the ACE, THI, and mixed component spectra, successfully distinguishing the spectra of the three compounds and achieving qualitative analysis of unknown spectra. Furthermore, genetic algorithm-partial least squares was used to model the spectral data, achieving a correlation coefficient of 0.97. The method was applied for the detection of spiked tea samples, achieving a precision RSD of 4.84 %. Therefore, this sensor combined with intelligent algorithms can be used for spectral identification and trace detection of mixed pesticide samples with structurally similar compounds.

Kai Liu,Qikui Fan,Zhun Qiao,Zhixue Zhang,Yongzhao Zhang,Zhiqun Chen,Hao Ma,Zemin He,Zongcheng Miao,Jiaofei Huo,Duqiang Xin,Jianxin Guo,Chuanbo Gao

Plate-like metal nanomaterials have attracted much interest due to their anisotropy, high aspect ratio and large surface area. The synthesis of non-noble metal nanocrystals with high purity remains challenging due to the poor regulation mechanism of their seeds, which limits their property and application studies. This paper presents a selective etching mechanism that effectively facilitates the identification and dissolution of non-target seeds during the synthesis of Cu nanoplates. This innovative approach not only enables the production of highly pure nanoplates but also allows for the shape transformation of nanoparticles into nanoplates. The resulting Cu nanoplates exhibit tunable localized surface plasmon resonance (LSPR) characteristics and high enhanced performance in surface-enhanced Raman scattering (SERS). This method offers a novel approach to the synthesis and purification of metal nanocrystals.

Fabrication of Ag/ZnO composite nanoflowers for ultrasensitive and recyclable SERS detection toward Rhodamine 6G

Yanfen Wang,Pengfei Jia,Jie Ai,Gangzheng Liu,Shuangshuang Liu,Juan Gao,Zhaoqi Sun

Novel Ag/ZnO composite nanoflowers (CNFs) had been designed as a surface-enhanced Raman scattering (SERS) substrate for organic molecule detection. Flower-like ZnO architecture was firstly synthesized by a low-temperature precipitation method, and subsequently Ag nanoparticles were decorated on ZnO nanoflowers through a photochemical reaction. The optimal Ag component was investigated by changing the photo-reduction time of Ag + in the ZnO precursor solution. It is found that the Ag nanoparticles with suitable size and crystallinity are significantly related to the light absorption and Raman signals for Ag/ZnO substrate. For Rhodamine 6G (R6G) as probe molecules, the optimal Ag/ZnO-40 min CNFs achieve a low detection limit of 1.0 × 10 −12  M and a high enhanced factor (EF) of 1.74 × 10 6 . Due to its excellent photocatalytic activity, the Ag/ZnO active substrate effectively degrades R6G molecules adsorbed on the surface, displaying an excellent self-cleaning property and stable recyclability. Such a superior SERS performance can be attributed to the synergy effect of two components (Ag nanoparticles and ZnO nanoflowers) such as its unique architecture, charge transfer and high-density hot spots.
