Ratiometric Fluorescence Imaging of Intracellular MicroRNA with NIR-Assisted Signal Amplification by a Ru-SiO2@Polydopamine Nanoplatform

Xunxun Deng,Xiaobo Liu,Shuo Wu,Shiyu Zang,Xiaotong Lin,Yanqiu Zhao,Chunying Duan

Accurate and sensitive fluorescence imaging of intracellular miRNA is essential for understanding the mechanism underlying some physiological and pathological events, as well as the prevention and diagnosis of diseases. Herein, a highly sensitive ratiometric fluorescent nanoprobe for intracellular miRNA imaging was fabricated by integrating a Ru-SiO2@polydopamine (Ru-SiO2@PDA) nanoplatform with a near-infrared light (NIR)-assisted DNA strand displacement signal amplification strategy. The Ru-SiO2@PDA spheres have excellent biosafety, high photothermal effect, and unique photophysical properties that can both emit a stable red fluorescence and well quench the fluorophores getting closer to them. So, when the fuel DNA and carboxyfluorescein (FAM)-labeled signal DNA are co-assembled on their outer surfaces, the FAM’s green fluorescence is quenched, and a low ratiometric signal is obtained. However, in the presence of miRNA, the target displaces the signal DNA from the capture DNA, releasing the signal DNA far away from the Ru-SiO2@PDA. Then, the green fluorescence recovers and leads to an enhanced Igreen/Ired value. Under NIR light irradiation, the Ru-SiO2@PDA increases the local temperature around the probe and triggers the release of fuel DNA, which thus recycles the target miRNA and effectively amplifies the ratiometric signal. Using A549 cells as a model, the nanoprobe realizes the highly sensitive ratiometric fluorescence imaging of miRNA let-7a, as well as its in vivo up- and down-regulation expressions. It provides a facile tool for highly sensitive and accurate intracellular miRNA detection through one-step incubation and may pave a new avenue for single-cell analysis.

Ratiometric emission NIR-fluorescent probe for the detection of lysosomal pH in living cells and in vivo

Weifen Niu,Juan Jia,Junkai Li,Chao Zhang,Keming Yun

The lysosome is known to be an acidic organelle that is ubiquitous in cells. Acidic pH is a characteristic feature of lysosomes, and aberrant lysosomal pH values have been manifested to be associated with diverse diseases. In order to monitor the lysosomal pH changes, we developed a ratiometric emission NIR-fluorescent probe (CzQl) for the quantitative analysis of the lysosomal pH in living cells and in vivo. CzQl was constructed via the ethylene bridging of carbazole derivatives and quinoline. The protonation of the probe resulted in a marked green-to-red emission color change. The probe exhibited an ideal pKa value of 4.60 with a linear relationship in the pH range of 3.9–5.3, which covered most of the pH ranges of lysosomes, and was useful for the quantitative detection and imaging of lysosomal pH changes. Moreover, CzQl exhibited easy loading capability, high selectivity, a large Stokes shift, specific lysosome-targeting ability and low cytotoxicity; thereby, it was successfully used for monitoring the lysosomal pH changes at a cellular level as well as the LPS-mediated inflammation in vivo.

A novel NIR-controlled NO release of sodium nitroprusside-doped Prussian blue nanoparticle for synergistic tumor treatment

Tao Feng,Jingyuan Wan,Pan Li,Haitao Ran,Huali Chen,Zhigang Wang,Liangke Zhang

Nitric oxide (NO) has shown positive effects in tumor treatment. However, controlling NO release in specific targets is still a crucial challenge for antitumor therapy. Considering that sodium nitroprusside (SNP) and potassium ferricyanide have similar chemical structures , a near infrared (NIR) laser-controlled NO release nanoplatform has been fabricated by allowing SNP to participate in mesoporous Prussian blue (m-PB) nanoparticle formation. The resulting SNP-doped m-PB (m-PB-NO) exhibited a good NIR-controlled NO release behavior, and the amount of NO released can be controlled by adjusting the laser intensity and irradiation time. Given that m-PB-NO still has strong absorption in NIR region , it exhibited an excellent photothermal effect in vitro and in vivo. After carrying antitumor drug, docetaxel (DTX)-loaded m-PB-NO ( [email protected] ) can simultaneously achieve NIR-controlled NO release, good photothermotherapy, and chemotherapy. The combination therapy of [email protected] showed a significant synergistic effect compared with each monotherapy and can significantly improve the therapeutic effect. Combination therapy also significantly inhibited the lung metastasis of 4T1 breast cancer cells in tumor-bearing mice by ablating primary tumors.

A NIR ratiometric fluorescent probe for the ‘naked-eye’ detection of endogenous hypochlorous acid in practical samples

Kangming Xiong,Fangjun Huo,Yongbin Zhang,Ying Wen,Caixia Yin

Hypochlorous acid (HClO), as one of the reactive oxygen species (ROS), is very important to the human immune system. Although many hypochlorous acid fluorescent probes have been developed, most of them are not suitable for detecting endogenous HClO due to their own limitations such as a slow response time and short emission wavelength. In this work, we designed a near-infrared ratiometric (2.1 to 15.3, I608/I735) fluorescent probe with a quick response (20 s) based on cyanine polyene derivatives. The system could detect endogenous hypochlorous acid in HepG-2 cells. It is significant that the color change in the measurement process makes the probe applicable to the determination of hypochlorous acid content in an 84 disinfectant and other actual samples.

A crystal-growth boundary-fusion strategy to prepare high-quality MAPbI3 films for excellent Vis-NIR photodetectors

Tao Wang,Gang Lian,Liping Huang,Fei Zhu,Deliang Cui,Qilong Wang,Qingbo Meng,Haihui Jiang,GuangJun Zhou,Ching-Ping Wong

Rendering high-quality perovskite films is integral for excellent performance of optoelectronic devices and highly challenging as well. Here, we establish a two-round pressure-assisted solvent-engineering (TPS) strategy to achieve remarkable enhancement in grain size (~400 μm 2 ), crystallinity , orientation and, especially, boundary fusion. Crystal growth and boundary welding of MAPbI 3 grains are mainly attributed to pressure-enhanced ion diffusion and defect elimination, which facilitates lateral enlargement of grains and formation of ultra-smooth film. Dramatic diminution of trap density determines long carrier lifetime and improved lateral charge transport, as demonstrated by the superior performance of MAPbI 3 -based photodetectors , e.g. ultrahigh on/off ratio (~10 6 ), good stability, fast response, high responsivity and high detectivity (1.3 × 10 14 Jones). When upconversion nanoparticles are uniformly assembled on the high-quality MAPbI 3 films, interestingly, the hybrid device exhibits dominantly elevated photoelectric capability in near infrared range (NIR, 980 nm) with long-term stability in ambient air. The results and method pave a novel way for ongoing fabrication of efficient and stable optoelectronic devices based on high-quality perovskite films.

When a Semiconductor Utilized as an NIR Laser-Responsive Photodynamic/Photothermal Theranostic Agent Integrates with Upconversion Nanoparticles

Ruichan Lv,Yanxing Wang,Jun Liu,Miao Feng,Fan Yang,Xue Jiang,Jie Tian

Photoactive theranostics including photodynamic therapy (PDT) hace aroused an intensive interest that combines medical diagnostics with treatment under a single irradiation laser. However, the conventional organic PDT agents usually suffer from aggregation caused by the quenching effect. In this research, we designed a core–shell–shell (CSS) UCNPs with NaGdF4 as the host, and the UCNPs could emit higher blue/green light by the energy transfer processes among Nd/Yb/Tm/Gd/Tb under 808 nm laser. When the CSS UCNPs were combined with the MnO2 semiconductor as the energy acceptor, CSS@Mn could be utilized as an effective PDT agent under a NIR laser. The temperature also increased more than 12.2 °C under the 808 nm laser, indicating the potential of synergistic photothermal therapy (PTT) and PDT. Interestingly, cells with small amounts of H2O2 could be responsive to the CSS@Mn platform. The antitumor effect was proved by the in vitro and in vivo experiments. Also, the biocompatibility, biodistribution, and excretion pathway were detected and revealed by the H&E stained pathological section and photoacoustic (PA) images. That means this platform could be potentially used as antitumor PDT/PTT agents.

NIR-Activated Polymeric Nanoplatform with Upper Critical Solution Temperature for Image-Guided Synergistic Photothermal Therapy and Chemotherapy

Jia Tian,Baoxuan Huang,Haiquan Li,Hongliang Cao,Weian Zhang

Premature and incomplete drug release is the typical bottleneck of drug release in traditional chemotherapy. Synergistic therapies are highly desirable in medicine and biology because they can compensate for the drawbacks of single therapy and significantly enhance the therapeutic efficacy. Herein, a novel near infrared (NIR)-activated polymeric nanoplatform with upper critical solution temperature (UCST) was constructed for image-guided synergistic photothermal therapy (PTT) and chemotherapy. UCST-responsive amphiphilic block copolymers were synthesized by reversible addition–fragmentation chain-transfer (RAFT) polymerization and then co-assembled with IR780 and cabazitaxel (Cab) to form spherical nanoparticles (NPs). IR780/Cab dual-loaded UCST polymeric NPs can produce local heating upon NIR laser irradiation and further lead to the dissociation of cargo-loaded NPs and controlled release of Cab. IR780 plays the role of both a heating generator and an activator for “on-demand” drug release. The investigation of in vivo fluorescence and photothermal imaging clearly demonstrated tumor targeting. Notably, both in vitro and in vivo studies illustrated that the synergistic PTT and chemotherapy presented better anticancer efficacy than that of PTT and chemotherapy simplely combined. Thus, the well-defined polymeric nanoplatform opens a versatile and effective path to develop image-guided synergistic therapies for tumor treatment.

Shape memory, self-healing property, and NIR photothermal effect of epoxy resin coating with polydopamine@polypyrrole nanoparticles

Yuhua Dong,Ming Gong,Dong Huang,Jie Gao,Qiong Zhou

Raspberry-like polydopamine@polypyrrole nanoparticles were synthesized by in situ chemical oxidative polymerization. Morphology was observed by scanning electron microscopy, and photothermal conversion performance was investigated. A shape memory coating with self-healing ability was prepared based on a thermo-responsive shape memory epoxy resin that utilized polydopamine@polypyrrole nanoparticles as the photothermal agent. Damage to the coating was healed via 808 nm near infrared light. The healing performance was evaluated by comparing the macroscopic morphologies of the scratched and healed coatings via optical microscopy. Linear sweep voltammetry was used to assess the recovery of the coating’s barrier properties. The results confirmed that the coating had self-healing ability based on the photothermal conversion effect of the polydopamine@polypyrrole nanoparticles.

Multifunctional NIR-responsive poly(vinylpyrrolidone)-Cu-Sb-S nanotheranostic agent for photoacoustic imaging and photothermal/photodynamic therapy

Meirong Hou,Chenggong Yan,Zelong Chen,Qingliang Zhao,Miaomiao Yuan,Yikai Xu,Bingxia Zhao

Ternary copper-based chalcogenide nanomaterials have become rather attractive due to the near-infrared (NIR) response in cancer theranostic fields. However, it is still challenging to further improve the theranostic efficiency of these nanomaterials. Herein, Cu-Sb-S nanoparticles (NPs) around 24 nm are synthesized facilely and functionalized with poly(vinylpyrrolidone) (PVP). Under the NIR irradiation, the resultant PVP-Cu-Sb-S NPs exhibit a relatively high photothermal conversion efficiency of 53.16% and a simultaneous reactive oxygen species (ROS) generation effect. Due to these outstanding photothermal/photodynamic effects, excellent tumor ablation results can be achieved by the combination of PVP-Cu-Sb-S NPs and 808 nm NIR laser treatments without obvious side effect. In addition, they show remarkable contrast enhancement according to in vitro and in vivo photoacoustic (PA) imaging. These PVP-Cu-Sb-S NPs could be served as a multifunctional nanotheranostic agent for PA imaging, photothermal/photodynamic cancer therapy. Statement of Significance Highly theranostic efficiency ternary copper-based chalcogenide nanomaterials has not been fully developed yet. Herein we report the PVP-Cu-Sb-S nanoparticles (NPs) with relatively high photothermal efficiency, simultaneous reactive oxygen species generation effect and photoacoustic imaging capability. The photothermal conversion efficiency of PVP-Cu-Sb-S NPs is higher than most of copper-based chalcogenide nanomaterials reported before. These findings provide a new kind of ternary copper-based chalcogenide with an enhanced theranostic effect, which could be served as a promising multifunctional nanotheranostic agent in the field of biomedical application.

Chengwan Xia,Diya Xie,Lang Xiong,Qian Zhang,Yang Wang,Zezheng Wang,Yuxin Wang,Bin Li,Chao Zhang

Herein, we reported nitroxide radical-modified CuS nanoparticles (CuS–NO˙ NPs), and they exhibited a typical absorption peak at 1182 nm. Due to such a long wavelength absorbance, CuS–NO˙ NPs exhibited excellent therapeutic outcome and low damage to normal tissues. Besides, we simultaneously achieved CuS–NO˙ NPs for MRI and CT dual-modal imaging, which successfully provided a new strategy for imaging-guided tumor treatment, thus increasing potential clinical applications for cancer treatment.

Fe3+-Induced Synchronous Formation of Composite Hydrogels for Effective Synergistic Tumor Therapy in NIR-I/II Biowindows

Jiulong Zhao,Chunhua Zhou,Chenyao Wu,Huan Wu,Chunping Zhu,Changqing Ye,Shige Wang,Duowu Zou

Alginate-Ca2+ hydrogel has been used to immobilize photothermal materials as well as chemotherapy drugs at lesion sites to prevent their entry into the bloodstream. However, the alginate-Ca2+ gelation mechanism may result in hardening of the blood vessels because of Ca2+ migration to the lesion site. In this study, a unique and facile one-pot formation of chemotherapeutic (doxorubicin, DOX) and polypyrrole-containing alginate hydrogel was designed by introducing Fe3+, which can synchronously induce the polymerization of pyrrole and gelatinization of alginate, into the DOX/pyrrole/alginate solution. The formed composite hydrogel was endowed with superior photothermal conversion properties in both the NIR-I (650–950 nm) and NIR-II (1000–1700 nm) biowindows and light-to-heat conversion efficiency higher than 50%, which enabled effective tumor hyperthermia treatment. Besides, near-infrared (NIR) irradiation could be used as a remote controller to trigger the DOX-release because of the heat generation, thus achieving continuous and on-demand tumor chemotherapy. The composite polymer hydrogels exhibited favorable hemo-, cyto-, and histocompatibility, as well as simple and cost-effective preparation and good clinical prospects.

Colloidal β-KYF4:Yb3+,Er3+/Tm3+ nanocrystals: tunable multicolor up-conversion luminescence from UV to NIR regions

Bin Zhou,Youfa Wang,Donglin Xia

Pure β-KYF4 nanocrystals codoped with Er3+/Yb3+ and Tm3+/Yb3+ were successfully synthesized via the thermal decomposition of trifluoroacetate precursors using oleic acid and octadecylene as coordinating solvents. The up-conversion optical properties of the β-KYF4 nanocrystals with different lanthanide (Ln)-doped ions (Yb3+/Er3+, Yb3+/Tm3+) were investigated. It is found that the colloidal dispersion of β-KYF4:Yb,Er/Tm nanocrystals display strong multiple up-conversion emission spanning from the deep UV-to-NIR regions under 980 nm excitation. Pure β-KYF4 nanocrystals codoped with Er3+/Yb3+ and Tm3+/Yb3+ can emit bright blue, green, yellow, and red emissions visible to the naked eye by adjusting the concentrations of Yb3+ sensitizer ion and Er3+ or Tm3+ activator ions. Moreover, the UC luminescent colors can be tuned for the four colors from blue to green to yellow and finally to red emission in the β-KYF4:X%Yb3+/(0.2%–2%)Er3+ and β-KYF4:25%Yb3+/0.2%Tm3+ colloidal samples. This work substantially expands our understanding of this category of KYF4 upconversion nanocrystals.
