In this study, the soluble, but non-digestible, longan (Dimocarpus longan Lour.) polysaccharides (LP) were extracted from dried longan fruits and then chemically selenylated to produce two selenylated products, namely SeLP1 and SeLP2, with different selenylation extents. The aim was to investigate their protective effects on rat intestinal epithelial (IEC-6) cells exposed to the food toxin fumonisin B1 (FB1). LP only contained total Se content of less than 0.01 g/kg, while SeLP1 and SeLP2 were measured with respective total Se content of up to 1.46 and 4.79 g/kg. The cell viability results showed that these two selenylated products were more efficient than LP in the IEC-6 cells in alleviating FB1-induced cell toxicity, suppressing lactate dehydrogenase (LDH) release, and decreasing the generation of intracellular reactive oxygen species (ROS). These two selenylated products were also more effective than LP in combating FB1-induced barrier disruption via increasing the transepithelial electric resistance (TEER), reducing the paracellular permeability, decreasing the mitochondrial membrane potential (MMP) loss, and maintaining cell barrier integrity by upregulating the tight-junction-related genes and proteins. FB1 caused cell oxidative stress and barrier dysfunction by activating the MAPK and mitochondrial apoptosis signaling pathways, while SeLP1 and SeLP2 could regulate the tMAPK- and apoptosis-related proteins to suppress the FB1-mediated activation of the two pathways. Overall, SeLP2 was observed to be more active than SeLP1 in the IEC-6 cells. In conclusion, the chemical selenylation of LP caused an activity enhancement to ameliorate the FB1-induced cell cytotoxicity and intestinal barrier disruption. Meanwhile, the increased selenylation of LP would endow the selenylated product SeLP2 with more activity. Keywords:longan polysaccharides;selenylation;fumonisin B1;IEC-6 cells;cytotoxicity;barrier function;oxidative stress;apoptosis

Glutathione transferase (GST P1-1) is a potential target for anticancer drugs. In this work, a series of 6-(7-nitro-2,1,3-benzoxadiazol-4-ylthio)hexanol (NBDHEX) derivatives as GST P1-1 inhibitors were designed, synthesized, and evaluated for their biological activity. Among the target compounds, 4n showed more selective inhibition toward GST P1-1 and GST M2-2, better water solubility, and more potent anticancer activities toward all the tested cancer cells (except for HOS) than its parent molecule. Detailed biological studies on the effect of 4n toward 143b cells revealed that 4n could arrest the cell cycle at the G2 phase and induced cell apoptosis in a dose-dependent manner. Like NBDHEX, 4n displayed good pharmacokinetic characteristics. An in vivo study on 143b xenograft models demonstrated that 4n could significantly reduce tumor growth in a dose-dependent manner, showing stronger antitumor activity than NBDHEX. Thus, 4n deserves to be further investigated as a potential antitumor agent for cancer therapy.

In polymer blends , the macroscopic properties are directly affected by the phase separation morphology. For polyamide/hydrogenated nitrile blends, the factors that influence the relationship between the reinforcement effect and the phase separation morphology are unclear. In this study, the reinforcing effects of polyamide 6 (short-chain polyamide) and polyamide 1212 (long-chain polyamide) in the hydrogenated nitrile (HNBR) matrix were comparatively analyzed. Deep analysis was performed on the mechanical properties, oil resistance, dynamic mechanical properties, and tribology properties of the blends. Results show that the interaction of polyamide 6 (PA6) with the HNBR chains was stronger than that of polyamide 1212 (PA1212). However, PA1212 exhibited a better enhancement effect than PA6 on the matrix. In the 20 wt% PA1212/HNBR blend, no friction marks were observed on the surface of the specimen. This phenomenon can be attributed to the synergistic effect of the uniformly dispersed particle and fibrous phase of PA1212 induced by the HNBR matrix. The formation of this special structure was closely related to the weaker physical interaction between two different phases (i.e., lower T g , smaller wavenumber shift, and weaker heterogeneous nucleation effect). This result was creative and could provide theoretical and experimental support for the regulation of the phase separation morphology of PA/HNBR blends with high performance.

Two novel low- ε r gallates MGa 2 O 4 (M = Ca, Sr) have been synthesized via a standard solid-state reaction method. According to the X-ray diffraction results, CaGa 2 O 4 crystallizes in space group Pna2 1 with an orthorhombic symmetry, while SrGa 2 O 4 belongs to the monoclinic P2 1 / c system. Both ceramics show ever-improving microstructures with the increasing sintering temperature . The optimal microwave dielectric properties ( ε r = 9.2, Qf = 66,000 GHz, τ f =–85 ppm/°C for SrGa 2 O 4 and ε r = 10.6, Qf = 15,400 GHz, τ f =–58 ppm/°C for CaGa 2 O 4 ) are obtained when sintered at 1275 °C. A patch antenna is fabricated using SrGa 2 O 4 ceramics as the substrate, which realizes a return loss of –19.94 dB and total efficiency of –1.38 dB (72.8 % in power ratio) at 4.84 GHz. The exceptional performances indicate that SrGa 2 O 4 ceramics are promising candidates for antenna applications at the Sub-6 GHz band.

Calcium aluminate cement (CAC) is widely used as a binder for refractory materials, and thus the improvement in compressive strength is of vital importance for CAC applied at high temperature. For this purpose, nano-Ca 10 (PO 4 ) 6 (OH) 2 additive with a ratio of 0.5–1.5 mass% was added with the water-cement ratio to be 0.4. X-ray diffraction and isothermal calorimetry analysis demonstrate that nano-Ca 10 (PO 4 ) 6 (OH) 2 additive can shorten the hydration process and promote the formation of main hydrates of CaAl 2 O 4 ·10H 2 O (CAH 10 ) and Ca 2 Al 2 O 5 ·8H 2 O (C 2 AH 8 ). In addition, scanning electron microscopy results suggest that nano-Ca 10 (PO 4 ) 6 (OH) 2 can protect CAH 10 and C 2 AH 8 from being destroyed during the calcination, guaranteeing that these thin lamellar crystals are intertwined to form the denser microstructure. Benefited from above effects, nano-Ca 10 (PO 4 ) 6 (OH) 2 can obviously improve the compressive strength of the CAC mortar samples cured for 7 d after calcination at 1100 °C, while the improving effect is dependent upon its contents. Especially, compared with the one without the additive, the compressive strength of the sample with 1.0% nano-Ca 10 (PO 4 ) 6 (OH) 2 is increased by 14%.

The effective detection of CA15-3 is of great significance for the postoperative monitoring and early diagnosis of breast cancer. Here, a fluorescent biosensor based on the fluorescence resonance energy transfer (FRET) between the prismatic hollow Metal-polydopamine frameworks (PHMPF) and 6-carboxyfluorescein (FAM)-labeled protein aptamer (FAM-Apt) was established to detect CA15-3. Characterization of SEM, TEM, EDS, FT-IR, XRD and XPS confirmed that the synthesized PHMPF was a polyhedral prismatic hollow organometallic frame containing the elements of C, N, O, and Co. PHMPF can quench the fluorescent group modified on the aptamer efficiently. Under optimized conditions, the proposed fluorescent biosensor can detect CA15-3 low to 2.56 × 10 −5 U mL −1 . In addition, the excellent selectivity and spiked experiments indicate the proposed biosensor owns potential applications for the clinical diagnosis of tumor markers in body fluids.

Phenol is highly toxic and carcinogenic even at low concentrations. This study focused on preparing high-performance mixed matrix membranes (MMMs) for the pervaporation separation of phenol from its aqueous solution. A highly-efficient zeolitic metal azolate framework, RHO-[Zn(Heim) 2 ] (MAF-6, Heim = 2-ethylimidazole) was incorporated into the polyether block amide (PEBA-2533) supported on the polyvinylidene fluoride (PVDF) ultrafiltration membrane. The MAF-6 was chosen as the filler due to its inherent hydrophobic surfaces and aperture size (7.6 Å), which is larger than the molecular kinetic diameter of phenol (6.6 Å). Effects of the casting solution concentration, active layer thickness, MAF-6 loading, operating temperature, and phenol concentration were systematically investigated. With the increase of the MAF-6 loading ratio X from 0 to 7, the phenol flux of the membrane was gradually enhanced, but the water flux was reduced. For 1000 ppm phenol aqueous solution at 80 °C, MAF-6–7@MMM displayed the optimal separation factor of 25.9 with a phenol flux of 89.2 g∙m −2 ∙h −1 . The pervaporation separation index (PSI) of the MAF-6–7@MMM was 1317.7 kg∙µm∙m −2 ∙h −1 at 80 °C, which was much higher than the PEBA/PVDF membrane (787 kg∙µm∙m −2 ∙h −1 ). Impressively, an increasing feed temperature enhanced the phenol flux more prominently than water, owing to the hydrophobicity and porosity of MAF-6. Consequently, the resultant MAF-6-X@MMMs broke the trade-off limitation between phenol permeability and selectivity. The as-prepared membrane also exhibited exceptional long-term stability, suggesting its prospect of industrial application.

This study aimed to determine the anti-obesity effects and mechanisms of Cerasus humilis polyphenol (CHP) in C57BL/6 obese mice and 3T3-L1 cells. High-performance liquid chromatography–electrospray ionization-tandem mass spectrometry was used for the qualitative and quantitative identification of CHP components. The obese mice, induced by feeding high-fat diet (HFD), were treated with CHP (250 mg/kg/day) by gavage for 12 weeks. Orlistat was gavaged at 15.6 mg/kg bw/day, as a positive control group. The analysis revealed that the main components of CHP were procyanidin B2, cyanidin-3-glucoside, and pelargonidin-3-glucoside. CHP dietary supplementation significantly reduced body weight and improved blood lipid measurements in HFD-fed mice (p < 0.01). Moreover, it inhibited mRNA expression of miR-122, Srebp-1c, and Cpt1a (p < 0.01) and reduced hepatic lipid deposition, as seen by hematoxylin and eosin staining. CHP downregulated the protein expression of PPARγ and C/EBPα in HFD-induced obese mice and inhibited adipocyte differentiation (p < 0.01). Compared with the HFD group, CHP supplementation had an obvious anti-inflammatory effect (decreased protein expression, such as TNF-α, IL-6, and MCP1), reducing leptin levels and TNF-α secretion in serum and cells (p < 0.01). CHP significantly inhibited the expression of miR-27a/b (53.3 and 29.9%, p < 0.01) in mice retroperitoneal white adipocytes, enhancing the expression of the target gene Prdm16 and significantly upregulating Sirt1 (105.5%, p < 0.01) compared with the HFD group. Moreover, CHP supplementation effectively improved oxidative stress (ROS, T-AOC, SOD, CAT, and GSH-Px) induced by HFD in obese mice (p < 0.01). Thus, CHP mitigates adipocyte differentiation, browning of white adipocytes, and reduction of inflammation and antioxidant activity to reduce obesity. Consequently, these results provide novel insights into the anti-obesity roles of CHP in HFD-induced obesity.

An inherent flame-retardant PA6 (FR-PA6) containing polydiphenylsiloxane (PDPS) was synthesized under the action of ethylene glycol (EG) via a facile “2-step” bulk polymerization. Inspiringly, EG acted not only as a ‘chain-linker’ to connect PA6 oligomer with PDPS moiety but also as a charring agent to increase the charring capacity of FR-PA6 after combustion. The prepared FR-PA6 reached 28.3% of limiting oxygen index (LOI), and passed the V-0 level of UL94 test with suppressed melt-dripping. After investigating the char residues and pyrolysis volatiles of FR-PA6 after combustion, the formation of silicon-rich protective layer was proved to be the essential factor to increase the flame retardancy of FR-PA6 via reducing the diffusion of pyrolysis volatiles, restricting the heat transfer and mass loss as well as preventing polymer melts from dripping. This work thus makes a worthwhile contribution to the development of FR-PA6 by offering a novel route that is easily accessible to industrial fabrication.

Although Fraxinellone (Frax) isolated from Dictamnus albus L. possessed excellent anti-hepatic fibrosis activity, oral administration of Frax suffered from the inefficient therapeutic outcome in vivo due to negligible oral absorption. At present, the oral formulation of Frax is rarely exploited. For rational formulation design, we evaluated preabsorption risks of Frax and found that Frax was rather stable while poorly dissolved in the gastrointestinal tract (78.88 μg/mL), which predominantly limited its oral absorption. Further solubility test revealed the outstanding capacity of cyclodextrin derivatives (CDs) to solubilize Frax (6.8–12.8 mg/mL). This led us to study the inclusion complexes of Frax with a series of CDs and holistically explore their drug delivery performance. Characterization techniques involving 1H-NMR, FT-IR, DSC, PXRD, and molecular docking confirmed the most stable binding interactions when Frax complexed with 6-O-α-D-maltosyl-β-cyclodextrin (G2-β-CD-Frax). Notably, G2-β-CD-Frax exhibited the highest solubilizing capacity, fast dissolution rate, and superior Caco-2 cell internalization with no obvious toxicity. Pharmacokinetic studies demonstrated markedly higher oral bioavailability of G2-β-CD-Frax (5.8-fold that of free drug) than other Frax-CDs. Further, long-term administration of G2-β-CD-Frax (5 mg/kg) efficiently inhibited CCl4-induced hepatic fibrosis in the mouse without inducing any toxicity. Our results will inspire the continued advancement of optimal oral Frax formulations for anti-fibrotic therapy.

The fabrications of hydrogel with both good mechanical properties and self-healing ability are still challenging issues. Herein, we report a high strength and pH responsive hydrogel based on polyvinyl alcohol (PVA) and poly 6-acrylamidohexanoic acid (PAACA) using dual physically cross-linking strategy. Owing to the H-bonds and the crystallites domains, the prepared hydrogel present considerable tensile strength . Meanwhile, the PAACA could facilitate the crystallization behavior of PVA, resulting in the fabrication of hydrogel through one simple freezing step instead of repeated freeze/thaw cycles. Furthermore, the hydrogel exhibit reversible pH responsive self-healing and swelling property stemming from protonated and deprotonated of the carboxyl groups in PAACA side chains, allowing external control over the healing and swelling process.

The Investigator 24plex QS Kit (QIAGEN, Hilden, Germany) is a 6-dye fluorescent chemistry short tandem repeat (STR) polymerase chain reaction (PCR) amplification system that simultaneously amplifies 20 of the expanded Combined DNA Index System (CODIS) core STR loci, SE33, DYS391, and the standard sex-determining locus, amelogenin, as well as two special internal performance quality sensor controls (QS1 and QS2), which are included in the primer mix to check the PCR performance. This study was designed to be a pilot evaluation of this STR-PCR kit in a Chinese Han population regarding the PCR conditions, sensitivity, precision, accuracy, repeatability, reproducibility, and concordance; tolerance to PCR inhibitors; applicability to real “forensic-type” samples; species specificity; mixture, balance and stutter analyses, and utility in a population investigation. The exhaustive validation studies demonstrated that the Investigator 24plex QS system is accurate, sensitive and robust for STR genotyping. In addition, these genetic markers in the population data in our study indicated that they can also be useful for forensic identification and paternity testing in the Chinese Han population.
