To date, extensive studies have focused on developing proteins as stabilizers to fabricate food-grade emulsions for encapsulating bioactive compounds aimed at targeted delivery. This paper aimed to develop a novel stabilizer using foxtail millet prolamin (FMP) to fabricate medium internal-phase Pickering emulsions (MIPEs) and investigate the stability andin vitrorelease behavior of curcumin (Cur) encapsulated within the MIPEs. Ultrasound treatment modified the secondary and tertiary structures of FMP, along with its particle size, zeta potential, and wettability, enhancing its functionality as a stabilizer for MIPEs. The MIPEs stabilized by 65% ultrasound-treated FMP (FMP-U) exhibited better rheological properties and stability, significantly improving the storage stability and antioxidant activity of Cur.In vitrodigestion results demonstrated that the MIPEs delayed the release of Cur, achieving a final release rate of 84.0 ± 1.47% after 4 h of gastrointestinal digestion and the DPPH radical scavenging activity (RSA) of 39.9 ± 1.31%, which was notably higher than the RSA of free Cur in oil at only 5.8 ± 1.37%. Moreover, MIPEs with Cur increased the bioaccessibility of Cur. This study provides new insights into a novel delivery system designed with FMP-U for encapsulating hydrophobic compounds, thereby enhancing their stability, sustained release, and bioaccessibility.

Visual pH-responsive packaging material is particularly important in food supply chain safety monitoring due to their non-destructive monitoring method and intuitive result. However, it has always been limited by the instability performance of pH-response components and carriers, which further hinders its wide food safety application. To address these challenges, we selected cellulose with remarkable biocompatibility and mechanical properties as the carrier, and high pH-responsive curcumin to develop a smart packaging material (RC/GC composite film) with real-time food safety monitoring. Compared with pure cellulose film, the RC/GC composite film exhibited excellent mechanical properties (4-fold enhancement) and thermal stability (100 °C increasing). Meanwhile, based on the first reported strategy of curcumin in-situ growth during cellulose film formation, the RC/GC composite film exhibited exceptional antioxidant activity (89.2 %), antimicrobial property (91.6 %), and significant pH-responsive sensitivity (within 15 s). This innovative approach offers a new strategy for easy-to-use and effective monitoring of food spoilage in packaging materials.

Dopamine detection and concentration identification using electrochemical sensing technique is critically important for a wide range of disease diagnosis and monitoring. However, the sensitivity of electrochemical sensing can be significantly affected from the non-specific protein adsorption in biological fluid samples. In this study, we aim to creating a modified electrode with excellent resistance to protein fouling. Herein, we have developed an antifouling electrochemical dopamine sensing interface integrated hydrophilicity hyaluronic acid (HA) with curcumin/multi-walled carbon nanotubes (CM/MWCNTs) via a facile method. The HA integrated with CM/MWCNTs composites-based electrochemical sensor exhibited synergistic effects: (i) the abundant hydrophilic groups in the HA structure (water contact angle, 30.88°) facilitated the formation of a hydrated layer on the electrode surface to prevent fouling; (ii) CM/MWCNTs catalyzed the electrooxidation of dopamine; (iii) electrostatic interactions between the negatively charged HA/CM/MWCNTs and positively charged dopamine in neutral condition. The morphology and structure of the nanocomposite were characterized. The HA/CM/MWCNTs-modified electrode exhibited the improved hydrophilicity with a water contact angle of 30.92° and enhanced electrochemical response of the modified electrode for dopamine sensing in a variety of conditions with protein in the electrolyte. Moreover, the HA/CM/MWCNTs-modified sensor also exhibited superior dopamine analytical performance in human serum samples, with the dopamine detection accuracy approaching 97 % and be interference-free from proteins. The constructed electrochemical sensor presented great potential in dopamine detection in clinic setups.

Oral squamous cell carcinoma (OSCC) is one of the most prevalent malignant tumors in the oral and maxillofacial region. Traditional treatments for OSCC, including surgery, radiotherapy, and chemotherapy, often lead to severe adverse effects. Therefore, the development of safe and effective novel cancer therapies is crucial for achieving synergistic cancer treatment, significantly enhancing patient survival and quality of life. In this study, triaxial electrospinning combined with freeze-shortening technology was employed to prepare injectable short fibers that provided a sustained release of curcumin (CUR). Subsequently, polydopamine (PDA) was modified on the surface of the short fibers to create PDA@CUR@PCL/PLA. The developed injectable short fibers represented a significant advancement in reducing the risks associated with the surgical implantation of traditional two-dimensional fibrous membranes. These fibers were coated with dopamine, which imparted notable significant photothermal effects, facilitating the rapid destruction of cancer cells. Furthermore, the core-loaded CUR was released in a pH-responsive manner and demonstrated a sustained anti-tumor effect. Cellular and animal experiments indicated that these composite short fibers could effectively eliminate oral cancer cells through a synergistic combination of photothermal and drug therapies without apparent toxic side effects. The developed injectable coaxial PDA@CUR@PCL/PLA short fibers are expected to provide a novel treatment approach for OSCC.

This study investigated the effect of different-polarity aqueous ethanol solutions on the formation of V -type starch originating from corn starch. Scanning electron microscopy revealed that the morphology of starch transformed from a random lamellar structure to a granular structure with decreasing solution polarity. When the ethanol concentration increased from 40 % to 60 %, the crystallinity and single-helix ratio of V -type starch increased from 9.66 % and 6.90 % to 29.77 % and 12.84 %, respectively. When the ethanol concentration was 100 %, the aforementioned parameters decreased to 21.31 % and 11.07 %, respectively. The crystallinity and curcumin encapsulation efficiency of the V -type starch reached their maximum values when this starch was prepared using 60 % aqueous ethanol. Furthermore, curcumin release experiments indicated that the produced starch–curcumin complex could resist gastric acid, and V -type starch with higher crystallinity exhibited a better sustained release effect in intestinal fluid. These results indicate that the formation of V -type starch can be finely regulated using aqueous ethanol solutions with different polarities, and such regulation is beneficial for the application of V -type starch in the oral delivery of guest molecules.

Herein, a novel oral nano-delivery system (CR-NPs) was designed for co-delivering curcumin and resveratrol to treat ulcerative colitis (UC) using Mesona chinensis polysaccharides and zein via antisolvent precipitation and continuous water evaporation crosslinking. It showed high encapsulation rates for both bioactives due to hydrogen bonding and hydrophobic forces. CR-NPs have favorable in vitro gastrointestinal stability and low pre-colonic release rates, resulting in increased accumulation in the colon. In vivo therapeutic effects of CR-NPs were confirmed through alleviation of UC symptoms and inflammation of UC mice. CR-NPs maintained the balance of intestinal flora and promoted the production of short-chain fatty acids in UC mice. Furthermore, CR-NPs intervention can alleviate the development of UC by modulating arachidonic acid, linoleic acid, and tryptophan metabolism. This study provides some theoretical basis using a novel dietary protocol option that co-delivering natural bioactives to the colon for alleviation of UC.

Diabetic wounds often exhibit poor angiogenesis, infection, and prolonged inflammation, posing significant challenges for effective treatment. To address these issues, the thermoresponsive self-healing hydrogel dressings based on hydroxybutyl chitosan and 4-carboxyphenylboronic acid was developed for the delivery of curcumin. The hydrogel exhibited unique properties, including thermoresponsive sol–gel irreversibility, efficient self-healing, and pH/glucose responsive drug release. The irreversible gelation behavior was attributed to the formation of a strong dual crosslinking network involving hydrogen bonds and dynamic boronate ester interactions. Furthermore, the hydrogel dressings exhibited excellent self-healing properties and an injectable nature, making it suitable for in situ wound dressing applications. The pH/glucose dual-responsive release of curcumin was further verified, with accelerated release under acidic conditions and hyperglycemic environments, potentially benefiting diabetic wound treatment. The in vitro studies confirmed the hydrogel's antibacterial, antioxidant and anti-inflammatory activity and ability to promote cell migration and blood coagulation. The in vivo experiments further validate the hemostatic efficacy. This work presents a novel strategy for constructing self-healing, thermoresponsive hydrogel dressings with pH/glucose dual-responsive drug delivery capabilities for the effective management of chronic diabetic wounds.

Traumatic brain injury (TBI) disrupts oxygen homeostasis in the brain, leading to excessive reactive oxygen species (ROS) production and dysregulated antioxidant mechanisms, which fail to clear excess ROS. This ROS overload promotes the expression of pro-inflammatory genes, releasing cytokines and chemokines and creating a vicious “ROS-neuroinflammation” cycle, making it essential to break this cycle for effective TBI treatment. In this study, we developed cysteine-alanine-glutamine-lysine (CAQK) peptide-modified antioxidant nanoparticles (C-PPS/C) for co-delivery of curcumin (Cur) to modulate oxidative and neuroinflammatory disturbances after TBI. In TBI mice, C-PPS/C nanoparticles accumulated in injured brain regions, where poly (propylene sulfide) 120 scavenged ROS, reducing oxidative stress, while Cur release further suppressed ROS and inflammation. C-PPS/C nanoparticles broke the “ROS-neuroinflammation” cycle, protecting the blood-brain barrier (BBB), reducing acute brain edema, and promoting long-term neurological recovery. Further investigation showed that C-PPS/C nanoparticles inhibited the NF-κB pathway, reducing pro-inflammatory gene expression and mitigating inflammation, suggesting a promising approach for TBI treatment.

Overcoming gastrointestinal tract barriers to enhance bioavailability is a major challenge for oral delivery. The innovative design of multi-compartment functional Pickering emulsions would provide an ideal option for targeted co-delivery of bioactive components. In this study, pH-responsive water-in-oil-in-water (W/O/W) Pickering double emulsions co-loaded with curcumin and fluorouracil were fabricated by zein/anthocyanin (ACN)/Zn 2+ nanoparticles (ZAZn NPs) via a one-step emulsification process. The unique emulsifier was designed by depositing ACN-Zn 2+ complexes on zein nanoparticles (zein NPs). The binding of ACN to the nanoparticle surface through hydrogen bonding interaction enhanced the hydrophilization and stimuli-response of zein NPs. Furthermore, the co-ordination of Zn 2+ and ACN facilitated the formation of nanoparticles with near-neutral wettability, which served as the dominant driving force for the one-step generation of W/O/W Pickering emulsions. The droplet size, distribution and internal structure of Pickering double emulsions were influenced by nanoparticle concentration and oil fraction. Due to Zn 2+ complexation with curcumin at the oil/water interface, the W/O/W Pickering emulsions obtained exhibited high curcumin encapsulation efficiency (91.2%) and free fatty acid release degree (65.9%), resulting in increased bio-accessibility (71.0%). The simulated gastrointestinal release demonstrated that curcumin and fluorouracil achieved controlled intestinal release, which was driven by a pH-mediated competitive adsorption-desorption mechanism. Moreover, in vitro cell experiments showed that co-encapsulated Pickering double emulsions exhibited 2.5 times greater bio-activity than those containing fluorouracil only. This facile strategy for preparing Pickering double emulsions is promising for the co-delivery of different physico-chemical bio-active compounds, which would improve oral bio-accessibility and bio-activity.

The harms caused by ultraviolet (UV) and blue light to eyes are attracting momentous concern due to growing exposure to artificial illumination and modern IT devices. Herein, a simple and eco-friendly adsorption approach was employed to integrate curcumin, a natural bioactive compound, into the cellulose substrate for the development of flexible and biodegradable filters capable of blocking harmful light. The curcumin/cellulose films demonstrate excellent UV-screening competence and photostability, with UV-A and UV-B screening ratios ranging from 92.8 % to 100 % and 89.2 % to 100 %, respectively. The films could block >96 % of blue light in the wavelength range of 400–500 nm. Meanwhile, the films maintain high transmittance (85.2–89.4 %) and low haze (2.0–2.7 %). The films can efficiently block blue light emanated from sunlight, light-emitting diodes, lighting systems, computer and mobile phone screens. Encouragingly, the incorporation of curcumin led to a substantial increase in the water contact angle, elevating it from 41.6 to 81.3°. Furthermore, the films exhibit excellent antimicrobial properties, biodegradability, and tensile strength in excess of 72 MPa. Therefore, these films fabricated entirely from natural resources have the potential to achieve practical applications such as food packaging and spectacle lens, especially suitable for electronic screen protectors.

This research examines the formulation and characterization of a pH-sensitive nanocarrier for the targeted and controlled release of curcumin (Cur) in the treatment of brain cancer. The nanocarrier consists of gelatin (G), agarose (Aga), and magnesium-doped carbon quantum dots (Mg-CQD), produced by a water-in-oil-in-water (w/o/w) double emulsification method. These materials demonstrate pH responsiveness, biocompatibility, and biodegradability, crucial for addressing Cur's limitations, including inadequate solubility and fast metabolism. In vitro cytotoxicity assays on U-87 MG glioma cells showed a 49 % reduction in cell viability, whereas minimal toxicity was observed in L929 fibroblast cells, indicating selective anticancer potential. The nanocarrier exhibited a regulated and extended release of Cur under acidic conditions, achieving encapsulation and loading efficiencies of 45.75 % and 85.5 %, respectively, hence enhancing its bioavailability and therapeutic efficacy. Characterization confirmed uniform nanoparticle distribution (average size: 193.5 nm; zeta potential: −42 mV) and successful drug encapsulation. FTIR, XRD and FE-SEM tests validated the effective drug encapsulation, uniform distribution of nanoparticles, and stability. The findings underscore the nanocarrier's potential for targeted cancer therapy, providing both therapeutic efficacy and safety, with intriguing applications in nanomedicine.

Thiol-modified hyaluronic acid (HASH) improves zein nanoparticles (ZNPs) stability by disulfide bonding with zein. However, the effect of the degree of thiol substitution in HASH on the stability of the ZNPs remains unclear. In this study, HASH with 1%, 3%, and 5% degrees of thiol substitution was synthesized and used as stabilizers to prepare three composite NPs. The pH and salt ion stability of the composite NPs increased with increasing degree of thiol substitution, which improved the antioxidant and anti-inflammatory capacities of the composite NPs. Interestingly, when the degree of thiol substitution exceeded 3%, the physical stability of composite NPs did not improve further. Our results revealed the relationship between the degree of thiol substitution in HASH and the stability of ZNPs, and provided the optimal degree of thiol substitution. These findings provided important information for improving the physical stability of ZNPs and contributed to the development of a delivery system for natural actives.
