In vitro antioxidant and anti-inflammatory activities of Radix Isatidis extract and bioaccessibility of six bioactive compounds after simulated gastro-intestinal digestion

Ping Xiao,Hongzhi Huang,Jianwei Chen,Xiang Li

Ethnopharmacological relevance Radix Isatidis called “Ban-Lan-Gen” is one of the most commonly-used traditional Chinese medicines for antiviral, anti-inflammatory, antioxidant and antipyretic purposes. Investigate the bioaccessibility of uridine , epigoitrin, adenosine, clemastanin B, indigoticoside A and isolariciresinol as well as the antioxidant and anti-inflammatory activities during an in vitro gastro-intestinal digestion of the Radix Isatidis extract (RIE). Materials and methods High performance liquid chromatography (HPLC) technique was adopted to determine the bioaccessibility of six bioactive compounds in RIE. Antioxidant activities of RIE in different digestive stages were determined by 1,1-Diphenyl-2-picrylhydrazyl (DPPH), superoxide anion and hydroxyl radical scavenging abilities. Anti-inflammatory activity was assayed by the inhibitions of inflammatory cytokines such as nitrous oxide (NO), prostaglandin E 2 (PGE 2 ) and tumor necrosis factor α(TNF-α) producted by lipopolysaccharide (LPS) stimulated RAW264.7 cells. Results The bioaccessibility of uridine, epigoitrin, adenosine, clemastanin B, indigoticoside A and isolariciresinol were 15.38%, 18.28%, 24.01%, 6.50%, 8.65% and 17.78%, respectively. Also, the digestion products still possessed certain antioxidant activities. The antioxidant activity was highly correlated with lignans (clemastanin B, indigoticoside A and isolariciresino). The anti-inflammation activity of the three samples decreased in the order: IN sample (the solution that had diffused into the dialysis tubing)>Nondigested sample (RIE solution)>Gastric sample (post-gastric digestion)>OUT sample (material that remained in the gastro-intestinal tract). Conclusions Results obtained in this research reveal the amount of bioactive compounds from RIE that could be available for absorption in vivo . The antioxidant activity decreased significantly but the anti-inflammatory activity was enhanced in serum-available fraction after gastro-intestinal digestion in vitro . This study could provide a scientific basis for a deeper pharmacological activity study of Radix Isatidis and a simple method for pharmacodynamic material basis research.

Characterization and antioxidant activity of β-carotene loaded chitosan-graft-poly(lactide) nanomicelles

Wenjiao Ge,Dong Li,Meiwan Chen,Xiaohui Wang,Shijie Liu,Runcang Sun

β-Carotene (β-C) is a well-established natural antioxidant agent, but its poor water-solubility, low chemical stability and low bioavailability limit its application in food, pharmaceuticals and cosmetics industries. Thus it is critical to develop an efficient method to improve the water solubility and stability of β-C. In this research, amphiphilic chitosan-graft-poly (lactide) (CS-g-PLA) copolymer was synthesized via a homogeneous ring-opening polymerization (ROP) in ionic liquid. The obtained CS-g-PLA copolymer was able to self-assemble into about 14   nm micelles in water at low concentration, and β-C loaded micelles (β-C/M) had low β-C degradation after 15 days. The antioxidant properties of the β-C/M were investigated by the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) method and ferric reducing antioxidant power (FRAP) method, respectively. Significantly improved antioxidant activity was observed for β-C/M in compare with free β-C. The aqueous dispersion of β-C/M has potential to be applied in the field of functional food and cosmetics.

Echinacoside inhibits amyloid fibrillization of HEWL and protects against Aβ-induced neurotoxicity

Di Zhang,Hua Li,Jian-Bo Wang

Amyloid protein depositions play crucial roles in a variety of degenerative disorders composing amyloidosis . There is a great interest in developing small molecule inhibitors of amyloidogenic processes. We examined the inhibitory effects of echinacoside (ECH) with different concentrations and at different fiber-forming stages in vitro utilizing the hen egg-white lysozyme (HEWL) model system. We also evaluated the antioxidant capacity of ECH by using elimination tests for the 2,2-diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl ( HO) free radicals. We investigated the protection provided by ECH against neurotoxicity induced by β-amyloid protein (Aβ). Through spectroscopic analyses, electron microscopy , cell viability assay, and hemolysis assay, we found that ECH dose dependently inhibited HEWL aggregation, and this inhibition occurred in different fiber-forming stages. ECH could also scavenge the DPPH and OH free radicals in a concentration-dependent manner. 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and 2′,7′-dichlorodihydrofluoresceindiacetate (DCFH-DA) fluorescent measurement results indicated that ECH could increase viability of rat pheochromocytoma PC12 cells injured by Aβ and suppress the increase in intracellular reactive oxygen species (ROS) triggered by Aβ. The present study findings facilitate a better understanding of the interaction between ECH and amyloid-forming proteins and also shed light on the protection of ECH against amyloid fibril-induced neuronal cell death.

Folic acid–polydopamine nanofibers show enhanced ordered-stacking via π–π interactions

Hailong Fan,Xiang Yu,Yang Liu,Zujin Shi,Huihui Liu,Zongxiu Nie,Decheng Wu,Zhaoxia Jin

Recent research has indicated that polydopamine and synthetic eumelanins are optoelectronic biomaterials in which one-dimensional aggregates composed of ordered-stacking oligomers have been proposed as unique organic semiconductors. However, improving the ordered-stacking of oligomers in polydopamine nanostructures is a big challenge. Herein, we first demonstrate how folic acid molecules influence the morphology and nanostructure of polydopamine via tuning the π–π interactions of oligomers. MALDI-TOF mass spectrometry reveals that porphyrin-like tetramers are characteristic of folic acid–polydopamine (FA–PDA) nanofibers. X-ray diffraction combined with simulation studies indicate that these oligomers favour aggregation into graphite-like ordered nanostructures via strong π–π interactions. High-resolution TEM characterization of carbonized FA–PDA hybrids show that in FA–PDA nanofibers the size of the graphite-like domains is over 100 nm. The addition of folic acid in polydopamine enhances the ordered stacking of oligomers in its nanostructure. Our study steps forward to discover the mystery of the structure–property relationship of FA–PDA hybrids. It paves a way to optimize the properties of PDA through the design and selection of oligomer structures.

Nutraceutical potential and antioxidant benefits of selected fruit seeds subjected to an in vitro digestion

Guan-Lin Chen,Song-Gen Chen,Fu Chen,Ying-Qing Xie,Men-Di Han,Chun-Xia Luo,Ying-Ying Zhao,Yong-Qing Gao

In order to find new sources of natural antioxidants, total phenolic contents and antioxidant capacities of 11 fruit seeds before and after in vitro digestion were evaluated by Folin–Ciocalteu method, 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity assay (DPPH), ferric reducing antioxidant power assay (FRAP), and ABTS •+ scavenging activity (ABTS) assay, respectively. The correlations between DPPH, FRAP, ABTS values and total phenolic contents were also evaluated. The results showed that 11 fruit seeds had diverse antioxidant capacities and the variation was very large. Furthermore, the main compounds of 11 fruit seeds were identified and quantified. Four fruit seeds, Dimocarpus longgana Lour, Vitis vinifera Linn (red), Vitis vinifera Linn (black), and Litchi chinensis Sonn showed the strongest antioxidant activities whether before or after digestion, which implied that these fruit seeds are important natural sources for preventing oxidative stress diseases.

Hydrolysis of rapeseed meal protein under simulated duodenum digestion: Kinetic modeling and antioxidant activity

Cunshan Zhou,Xiaojie Yu,Xiaopei Qin,Haile Ma,Abu ElGasim A. Yagoub,Jiali Hu

Kinetics modelling, DPPH radical scavenging activity and reducing power of rapeseed meal protein were investigated under simulated duodenum digestion. Kinetics of the reaction was considered in relation to initial substrate concentration, initial enzyme concentration, and hydrolysis time. Optimum hydrolysis conditions were trypsin concentration of 10 g/L, substrate concentration of 4 g/L, time of 40 min and temperature of 37 °C. From the results a general kinetic enzymolysis equation was suggested, providing a rational theoretical basis for determining the parameters of the reaction. The highest antioxidant activity was in accordance with the optimum reaction conditions. Antioxidant free amino acids increased after hydrolysis. Antioxidant oligopeptides of molecular weights of 2608, 1695 and 211 Da were identified. At a concentration of 2 mg/mL, the peptide fraction of CS-F 2 -CC 2 ; with molecular weight of 1000–1500 Da; showed the highest DPPH radical scavenging activity and reducing power, accounting for 70.32% and 0.538, respectively. The trypsin-hydrolysate displayed a potential capacity to scavenge free radicals and binding irons after duodenal digestion, appearing as promising ingredient to formulate functional foods with antioxidant activity.

The aim of this study was to compare the flavonoids compositions, content and antioxidant activities of Hawk tea from six botanical origins for screening the most suitable botanical origins. The main flavonoids of Hawk teas from Litsea coreana Levl. var. lanuginose were identified, and six high contents of individual flavonoids (hyperin, isoquercitrin, quercitrin, astragalin, quercetin and kaempferol) were quantitative and comparative analysis. Moreover the potential antioxidant capacity of Hawk tea was measured by using in vitro antioxidant assays. The above experimental data were all used for the principal component analysis. The statistical results showed that the extracts from Machilus chuanchienensis S. Lee and L. coreana Levl. var. lanuginose appeared closely positioned in the positive regions, indicated the higher flavonoids and antioxidant activity compared with other species. Thus, they were temporarily considered as good sources of Hawk tea.

Curcumin encapsulated in the complex of lysozyme/carboxymethylcellulose and implications for the antioxidant activity of curcumin

Zhenshun Li,Yuntao Wang,Yaqiong Pei,Wenfei Xiong,Chunlan Zhang,Wei Xu,Shilin Liu,Bin Li

A facile approach was investigated to encapsulate and protect curcumin (Cur) by self-assembly of lysozyme (Ly) and carboxymethylcellulose (CMC) of different degrees of substitution (DSs). This work studied the influence of Ly–CMC coacervates on the binding, solubility and stability of Cur. The interactions of Cur with Ly–CMC coacervates were researched by UV–vis, steady-state fluorescence, synchronous fluorescence and circular dichroism spectra. These results were explained in terms of the formation of Ly–CMC coacervates with electrostatic interaction, which led to unfold the structure of Ly for providing Cur with more hydrophobic microenvironment than free Ly. Meanwhile, the CMC of higher DS provide more negative charges, and produce more attraction to Ly than that of lower DS values. Moreover, the coacervates of Ly–CMC of higher DS are found to be superior for enhancing the stability of Cur. Our work provided a new insight for understanding the biomolecule protective system based on protein/polysaccharide complexes.

Enzymatic Hydrolysis of Oleuropein from Olea europea (Olive) Leaf Extract and Antioxidant Activities

Jiao-Jiao Yuan,Cheng-Zhang Wang,Jian-Zhong Ye,Ran Tao,Yu-Si Zhang

Oleuropein (OE), the main polyphenol in olive leaf extract, is likely to decompose into hydroxytyrosol (HT) and elenolic acid under the action of light, acid, base, high temperature. In the enzymatic process, the content of OE in olive leaf extract and enzyme are key factors that affect the yield of HT. A selective enzyme was screened from among 10 enzymes with a high OE degradation rate. A single factor (pH, temperature, time, enzyme quantity) optimization process and a Box-Behnken design were studied for the enzymatic hydrolysis of 81.04% OE olive leaf extract. Additionally, enzymatic hydrolysis results with different substrates (38.6% and 81.04% OE) were compared and the DPPH antioxidant properties were also evaluated. The result showed that the performance of hydrolysis treatments was best using hemicellulase as a bio-catalyst, and the high purity of OE in olive extract was beneficial to biotransform OE into HT. The optimal enzymatic conditions for achieving a maximal yield of HT content obtained by the regression were as follows: pH 5, temperature 55 °C and enzyme quantity 55 mg. The experimental result was 11.31% ± 0.15%, and the degradation rate of OE was 98.54%. From the present investigation of the antioxidant activity determined by the DPPH method, the phenol content and radical scavenging effect were both decreased after enzymatic hydrolysis by hemicellulase. However, a high antioxidant activity of the ethyl acetate extract enzymatic hydrolysate (IC50= 41.82 μg/mL) was demonstated. The results presented in this work suggested that hemicellulase has promising and attractive properties for industrial production of HT, and indicated that HT might be a valuable biological component for use in pharmaceutical products and functional foods. Keywords:Olea europeaL.;hydroxytyrosol;oleuropein;enzymatic hydrolysis;hemicellulase

Preparation of the high purity gingerols from ginger by dummy molecularly imprinted polymers

Wenhua Ji,Xiuli Ma,Jinghua Zhang,Hongkai Xie,Feng Liu,Xiao Wang

In this work, a dummy molecularly imprinted polymers (MIPs) were developed as the selective sorbents for preparation of the high purity gingerols from ginger for the first time. The dummy template molecule with similar structural skeleton to gingerols, N-vanillylnonanamide, has been designed and synthesized. The performance of the MIPs and non-imprinted polymers (NIPs) were evaluated including selective recognition capacity, adsorption isotherm, and adsorption kinetics. Optimization of various parameters affecting dummy molecular imprinted solid phase extraction (MISPE), such as the type and flow rate of the loading solvent, the composition and volume of the eluting solvent, and the composition and volume of the washing solvent were investigated. Gingerols with the percent recovery of 80 and the percent purity of 99.1 were obtained from the extracts of ginger by MISPE. Besides, gingerols obtained by MISPE had more powerful activity of eliminating free radical compared with extracts before extraction with the MISPE column. Application of MIPs with a high affinity toward three gingerols provides a novel method for obtaining a group of compounds which have likely active groups from natural products.

Enhanced in Vitro Antioxidant Activity of Polysaccharides From Enteromorpha Prolifera by Enzymatic Degradation

Jie Xu,Li-Li Xu,Qin-Wei Zhou,Shu-Xian Hao,Tao Zhou,Hu-Jun Xie

In order to improve the antioxidant activity of polysaccharide derived from E nteromorpha prolifera (PEP), PEP was hydrolyzed to relatively low molecular weight polysaccharides in the presence of either pectinase or gluco-amylase. Response surface methodology was employed to optimize the hydrolysis conditions based on the polysaccharide degradation rate. Optimum hydrolysis for pectinase was found to be: enzyme concentration 9.36 U/mL, reaction temperature 56C and pH 4.45; for gluco-amylase, the optimum conditions were: enzyme concentration 14.20 U/mL, reaction temperature 49C and pH 4.48. The enzymatic hydrolysates of polysaccharide obtained under optimal conditions were evaluated for their antioxidant activities by determining their ability to scavenge 1,1-diphenyl-2-picrylhydrazyl, hydroxyl and superoxide anion radicals. The results indicate that the enzymatic hydrolysates of polysaccharide exhibited stronger free radical scavenging ability than nondegraded polysaccharide, while pectinase hydrolyzed polysaccharide was found to possess superior radical scavenging ability to that of gluco-amylase hydrolyzed polysaccharide. Practical Applications In order to fully utilize polysaccharide resources, an effective method to improve the antioxidant activity of E nteromorpha prolifera was established by enzymatic degradation. The hydrolysates obtained by this method may be used as ingredients of functional foods due to their antioxidant activity.

Enrichment and purification of total flavonoids from Cortex Juglandis Mandshuricae extracts and their suppressive effect on carbon tetrachloride-induced hepatic injury in Mice

Pan Zhao,Chao Qi,Gang Wang,Xinpeng Dai,Xiaohong Hou

In the present work, a simple and efficient chromatographic separation method was developed for preparative separation and enrichment of total flavonoids (TFs) from Cortex Juglandis Mandshuricae (CJM) extracts and then the protective effect of TFs against CCl 4 -induced acute liver injury in mice was investigated. Enrichment and purification of TFs from CJM extracts were studied using six macroporous resins and HPD-750 resin was selected as the best resin according to its adsorption and desorption properties. The operating parameters of resin column chromatography were optimized. Under the optimal conditions, TFs from CJM with purity larger than 50% were produced and their antioxidant activity was further evaluated in vitro. The mice were orally administrated with the purified TFs for seven days and then given CCl 4 (0.3%, 10   mL/kg i.p.). The results showed that TFs of CJM significantly attenuated the activities of serum aspartate transaminase (AST) and alanine transaminase (ALT) compared with model group, as well as the relative liver weight. Histopathological observation also revealed that TFs reduced the incidence of liver lesions and improved hepatocyte abnormality. Moreover, oral administration of TFs significantly enhanced antioxidant enzyme activities (superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px)) and decreased the content of malondialdehyde (MDA). Histopathological and biochemical results elicited that TFs of CJM had significant hepatoprotective activity comparable to the standard silymarin. This is the first time to reveal the protective actions of the TFs from CJM against CCl 4 -induced liver damage in mice and this natural product should be developed as a new drug for treatment of live injury in future.
