Stabilizing Enzymes in Plasmonic Silk Film for Synergistic Therapy of In Situ SERS Identified Bacteria

Zhangkun Liu,Shengkai Li,Zhiwei Yin,Zhaotian Zhu,Long Chen,Weihong Tan,Zhuo Chen

Increasing antibiotic resistance becomes a serious threat to public health. Photothermal therapy (PTT) and antibacterial enzyme-based therapy are promising nonresistant strategies for efficiently killing drug-resistant bacteria. However, the poor thermostability of enzymes in PTT hinders their synergistic therapy. Herein, antibacterial glucose oxidase (GOx) is embedded in a Ag graphitic nanocapsule (Ag@G) arrayed silk film to fabricate a GOx-synergistic PTT system (named silk-GOx-Ag@G, SGA). The SGA system can stabilize GOx by a vitrification process through the restriction of hydrogen bond and rigid β -sheet, and keep the antibacterial activity in the hyperthermal PTT environment. Moreover, the arrayed Ag@G possesses excellent chemical stability due to the protection of graphitic shell, providing stable plasmonic effect for integrating PTT and surface enhanced Raman scattering (SERS) analysis even in the GOx-produced H 2 O 2 environment. With in situ SERS identification of bacterial intrinsic signals in the mouse wound model, such SGA realizes superior synergistic antibacterial effect on the infected Escherichia coli , Staphylococcus aureus , and methicillin-resistant Staphylococcus aureus in vivo, while without causing significant biotoxicity. This system provides a therapeutic method with low resistance and in situ diagnosis capability for efficiently eliminating bacteria.

3D composite SERS substrates with COF spaced Au nanoparticles for enrichment and selective sensitive detection

Xiaomu Shan,Jie Pan,Xuejian Du,Jian Shi,Zhuofan Wei,Yuanze Xu,Shouzhen Jiang

The excellent performance of surface-enhanced Raman spectroscopy (SERS) is closely related to the intensity, density and uniformity of hotspots. In this study, we introduce a PET/Au NPs@COF Raman substrate featuring three-dimensional hotspots. The spacing between each layer of gold nanoparticles (NPs) is precisely controlled by adjusting the thickness of the COF via the interfacial synthesis technique, causing the strong coupling between gold particles across the layers. The porosity of the COF enables the substrate to adsorb numerous analyte molecules into the large-area uniform hotspots. Through simulations and adsorption detection on the substrate, Raman testing further demonstrated that the substrate with double-layer Au NPs@COF exhibited the most favorable SERS performance. Additionally, the substrate allows for selective detection based on the particle size of the target molecule. The flexibility of the substrate enables it to conform to irregular surfaces, effectively capturing analytes from a variety of real samples.

Shuang-Feng Pan,Xiang-Dong Tian,Ying Lu,Yu-Qin Liao,Mei-Qing Liu,Ling-Yi Meng,Lei Zhou,Yun Zhang

Small chiral metabolic molecules are increasingly recognized as pivotal biomarkers for disease monitoring and treatment. Here, a wearable microfluidic patch is presented that integrates chiral 3D plasmonic nanostructures with surface-enhanced Raman spectroscopy (SERS) sensing activity for the in situ and real-time metabolic profiling of chiral molecules in sweat. The microfluidic patch is designed for the direct, in situ capture and storage of microliter volumes of sweat. By exploiting the 3D chiral plasmonic coupling interactions within the nanostructures, the integrated SERS sensor on the patch allows for highly sensitive and quantitative enantiomer detection through their unique fingerprint SERS spectra. In a proof-of-concept demonstration, the first in situ, real-time quantitative detection of chiral drug metabolite and pH in human sweat using this device is successfully conducted. This capability enables the capturing of an individual's dynamic metabolic profile. Leveraging this solution, pharmacokinetic correlations are established, showcasing the potential application of the device in assessing human health.

Superhydrophobic SERS substrates based on silver dendrite-decorated filter paper for trace detection of nitenpyram

Qinzhi Wang,Yingnan Liu,Yaowen Bai,Siyu Yao,Zijie Wei,Min Zhang,Liming Wang,Li Wang

In the present work, highly sensitive Raman detection of nitenpyram using superhydrophobic filter paper as substrates is introduced. The process is simple, and efficient. By sequentially coating silver dendrites and Octyltrimethoxysilane (OTMOS) on filter paper, we produced highly active surface-enhanced Raman scattering (SERS) substrates which show advancing and receding water contact angles of θ A / θ R  = 159°/156°. Nitenpyram, a type of pesticides popularly used in agriculture, can be easily detected with the detection limit as low as 1 nM using the superhydrophobic filter paper as SERS substrates, which validates their use in Raman applications.

Production of stable and sensitive SERS substrate based on commercialized porous material of silanized support

Han Zhang,Lin Sun,Yuan Zhang,Yan Kang,Huilian Hu,Huirong Tang,Yiping Du

This paper reports accurate synthesis of a new type of surface-enhanced Raman spectroscopy (SERS) substrate based on gold nanoparticles decorated 201 red silanized diatomaceous supports. The developed SERS substrate is easy to fabricate, cost effective and offers sensitive rapid detection. The performance and stability of the SERS substrate was investigated and the results showed good SERS activity of substrate that can last for more than 6 months. Tiazophos and phosmet pesticides in aqueous solutions can be detected clearly at a low concentration of 0.01   mg/L. The new substrate was applied to detect triazophos in apple pulp at 0.2   mg/kg. The results indicate that this substrate have good potential in rapid monitoring of pesticide residues in fruits and could be suitable for field-based applications, and routine laboratory analysis of chemicals.

Gap-enhanced V-shaped SERS substrate for multiplex trace dye pollutants detection in river water

Qianqian Ding,Jisong Qian,Shijie Mei,Zebin Zhu,Wenge Yang,Kexin Huang,Yanqiang Cao,Liyong Jiang

Surface-enhanced Raman scattering (SERS) is a powerful analytical tool for detecting trace pollutants due to its molecular fingerprint specificity and single-molecule sensitivity. However, conventional two-dimensional gap-enhanced substrates suffer from limited analyte diffusion into nanogap caused by plasmonic material hydrophobicity. Herein, we present a V-shaped SERS substrate designed to overcome these challenges through three synergistic mechanisms: (i) three-dimensional (3D) plasmonic nanogaps, (ii) gravity-driven analyte enrichment enabling efficient aqueous-phase molecule transport, and (iii) light-trapping nanopores amplifying electromagnetic fields. By utilizing atomic layer deposition (ALD)-defined ultrathin alumina sacrificial layers, we achieved precisely tunable nanogaps (2–10 nm) between gold nanoparticles (AuNPs) and the underlying gold film (Au) within V-shaped architectures. The optimized 6 nm AuNPs-gap-Au nanostructure balances near-field intensity and aqueous analyte accessibility, achieving sub-nanomolar sensitivity for rhodamine 6G with outstanding reproducibility (RSD < 9 % across 10 −9 –10 −5  M). Significantly, the substrate enables multiplex detection of dye pollutants (basic blue 3, malachite green, methylene blue) at sub-nanomolar level in river water, showing great potential for real-time, on-site monitoring of emerging pollutants in natural water systems.

AI-Enhanced Rapid SERS Screening of Trace Quinolone Antibiotics across the Source-Pathway-Sink Ecosystem

Jing Xu,Si-Heng Luo,Chen-ru Xiong,Jia-hao Liu,Zeyu Zhao,Wei-qi Lin,Wei Zhang,Ping Guo,Cheng Pan,Quanlong Li,Zhong-Qun Tian,Bin Ren,Guo-Kun Liu

The overused quinolone antibiotics in animal husbandry and clinical medicine pose a growing threat for global health as they enter ecosystem via agricultural discharge and medical wastewater. Consequently, risk assessment for environmental and human exposure is highly demanded, which calls for an accurate, reliable and rapid qualitative and quantitative analysis of trace antibiotics in different matrices. Surface-enhanced Raman spectroscopy (SERS), providing fingerprint chemical information with near-single-molecule sensitivity, has received considerable attention. Herein, considering the common physicochemical properties of various quinolone antibiotics, we first developed sample pretreatment strategy consisting of solid-phase extraction (SPE), liquid–liquid extraction (LLE), salting-out assisted liquid–liquid extraction (SALLE), and NPs purification (NPs (p)). The modular SPE–LLE–SALLE–NPs (p) coupling with spectral separate network (SSNet) (a deep-learning based spectral unmixing algorithm) enabled a rapid (<8 min per sample), sensitive (μg L–1), on-site, and intelligent SERS analysis of 19 quinolone antibiotics in food, biological, and environmental matrices. Besides, the identification of each QN in a five-target mixture sample enabled SPE–LLE–SALLE–NPs (p)–SSNet–SERS to be a highly promising rapid detection strategy in blind sample screening qualitatively and quantitatively. The integrated SPE–LLE–SALLE–NPs (p)–SSNet–SERS offers a cross-media, full-process, and big data-enhanced rapid monitoring solution for antibiotic pollution within the “source-pathway-sink” framework.

Plasmonic microsphere lens arrays-integrated microfluidic SERS chip for mixed pesticides identification with machine learning

Zhenyong Dong,Feixiang Zheng,Hao Wang,Yukai Liu,Xiao Ren,Feng Lin,Song Zhou,Li Chen,Yan-qing Lu,Guanghui Wang

The use of multiple pesticides in agriculture has caused mixed residues, complicating food safety testing. Surface-enhanced Raman scattering (SERS) offers high sensitivity and fingerprinting capabilities for pesticide detection; however, its application is limited by spot size, uneven sample distribution, spectral analysis challenges in mixed samples, and the lack of integrated automated systems for sample preparation, detection, and analysis. Hence, we developed a SERS microfluidic chip integrated with plasmonic microsphere lens arrays, which integrates optical field-modulated microspheres and plasmonic nanostructures to realise the transition of SERS detection from point-hotspots to uniform area-hotspots, achieving high-sensitivity, high-stability Raman detection under wide-field laser excitation. Moreover, a comprehensive a sample-to-answer total analysis system was established with this chip as the core, incorporating the micro-QuEChERS pretreatment module for efficient extraction of pesticides in complex matrices and the random forest-dual annealing spectral parsing algorithm for mixed spectral analysis. The results indicate that the limits of detection for 2,4-dichlorophenoxyacetic acid, acetamiprid, and thiabendazole were 1.15 nM, 0.63 nM, and 0.69 nM, respectively. The linear ranges were 10⁻⁴–10⁻⁹ M, with R² values of 0.9982, 0.9989, and 0.9996, respectively. Average recoveries ranged from 90.8 % -104.66 %, 89.1 %-107.89 %, and 89.73 %-108.4 %, respectively, with relative standard deviations below 8 %. The algorithm quantified mixed pesticides with mean absolute percentage errors of 4.96 %, 5.35 %, and 7.72 %, respectively. The system showed 3 s detection response and 15-minute analysis time. Thus, the method provides a rapid, on-site solution for detecting mixed pesticides in complex matrices by exceptional optical performance and automated processing functionality.

Rapid qualitative identification and quantitative detection of three common spice extracts based on SERS technology

Manhong Zhang,Shijie Liu,Zhengyan Gai,Miaoyun Li,Lingxia Sun,Huihui Zhi,Lijun Zhao,Dong Liang,Yaodi Zhu,Yanxia Liu,Yangyang Ma

In the quality control of braised meat products, the content of spice extract in braised soup is often used as one of the important indexes to evaluate its flavor and quality. However, the existing research methods mostly rely on chromatography-mass spectrometry for quantitative analysis, which has some problems such as complex operation, long period, high requirements for equipment and professional technology. To enable rapid detection of spice extracts while reducing analytical costs and improving efficiency, this study took the main extracts of star anise, Sichuan pepper and chili peppers, namely shikimic acid xanthoxylin and capsaicin, as the research objects. By preparing AuNPs as the substrate material, a rapid classification and recognition method for SERS spectra of the three different spice extracts was constructed. The results showed that the Raman signals of three spice extracts could be obviously enhanced by using AuNPs as substrate, with good reproducibility ( RSD  < 5.5 %). SERE spectrum could clearly distinguish the differences of SERS peaks of shikimic acid, xanthoxylin and capsaicin. Principal component analysis (PCA) and linear discriminant analysis (LDA) further confirmed clear visual classification of the spice extracts. Finally, based on the SERS spectra of different concentrations, the digital quantitative equations of SERS spectra of three spice extracts were constructed. This work provides a theoretical foundation and technical framework for rapid detection of spice extracts based on SERS technology.

Band-Engineered MoS2/F4TCNQ Heterojunction for Ultrasensitive SERS Detection of Hg2+ in Water Pollution Monitoring

Yuxin You,Abdur Rahim,Mingyue Li,Xiaoyu Zhang,Min Li,Chengju Guo,Muhammad Saleem,Muhammad Zahid,Xueyou Xu,Mei Liu

As human society continues to develop and progress, the hazards posed to human health by emerging pollutants are becoming increasingly serious due to their unique characteristics such as specific toxicity, persistence, and bioaccumulation. It is critical to develop highly sensitive, renewable nonmetallic surface-enhanced Raman scattering (SERS) platforms for environmental monitoring, particularly for the trace detection of mercury ions in water. Here, we utilized a band-tunable F4TCNQ/MoS2 heterojunction achieving an ultrasensitive limit of detection (LOD) of 2.77 × 10–11 M for Hg2+ in pure water. The enhancement factor (EF) for the 989 cm–1 peak was found to be approximately 4.2 × 105, while that for the 722 cm–1 peak was 2.8 × 105. Such sensitivity arises due to the interfacial charge transfer (CT) and the weak electromagnetic interaction between the heterojunction and Hg2+ with the Raman reporter molecule 4-methylpyridine (4-MPy). The substrate exhibits remarkable selectivity against interfering ions (Ca2+, Mg2+, Pb2+, etc.,), along with high reproducibility (RSD < 9.33%), and stability up to one month under environmental conditions. The results indicate that the detection sensitivity is comparable to or even better than that achieved with the conventional SERS substrate to enable specific detection of Hg2+. Moreover, the substrate can be reused through photocatalytic degradation showing a 30 min duration for complete removal of Hg2+, enabled by the F4TCNQ/MoS2 heterostructure. This offers significant advantages for detecting Hg2+ in complex environmental samples, such as seawater and soil leachate, while also proving valuable for environmental monitoring and food safety applications.

SERS-active vertically aligned silver/tungsten oxide nanoflakes for ultrasensitive and reliable detection of thiram

Fengli Gao,Weiqian Kong,Guangli He,Yanzhen Guo,Huili Liu,Shouren Zhang,Baocheng Yang

As an emerging technique for rapid detection, Surface-enhanced Raman scattering (SERS) has received much practical attention. Here, we report a uniformly distributed silver nanoparticles on well-defined tungsten oxide nanoflakes hierarchical nanostructures (Ag/WO 3-x ) as flexible solid SERS substrate. The vertically aligned Ag/WO 3-x nanoflakes were prepared by in situ redox reaction without reductant additives, ensuring the clean surface and free of extra interferences with good signal-to-noise ratio. Using rhodamine 6G (R6G) as probe molecule, the enhancement factor of developed Ag/WO 3-x nanoflakes SERS substrate was found to be high as 2.41 × 10 7 with an ultrasensitive detection concentration of 10 -12 M. Additionally, Ag/WO 3-x nanoflakes SERS substrate exhibited good reproducibility and long-time stability. Meanwhile, the pesticide thiram with a 1.3 pM limit of detection (LOD) can be readily determined via developed SERS substrate. To illustrate the potential practical value of the prepared substrates, 2.4 ppb spiked thiram on apple peel surface were both successfully detected. Notably, the developed SERS substrates could be cut into small pieces for detecting trace chemical or biological molecules and the flexible utilization of substrates would be improved dramatically. As a result, vertically aligned Ag/WO 3-x nanoflakes with well-defined shape and hierarchical structure have great potential as solid SERS substrate in the field of fast detection and trace analysis.

Versatile self-assembled MXene-Au nanocomposites for SERS detection of bacteria, antibacterial and photothermal sterilization

Zizhen Yu,Li Jiang,Rongyang Liu,Weidan Zhao,Ziheng Yang,Jinyao Zhang,Shangzhong Jin

The emergence of antibiotic-resistant bacterial strains has caused bacterial infections to become a more serious worldwide health problem. The development of multi-functional platform that can quickly and sensitively detect bacteria and effectively inhibit or kill bacterial is important and urgent. Herein, a novel versatile MXene-Au nanocomposite was successfully synthesized by self-assembly method for rapid detection of bacteria and photothermal sterilization. The typical gram-negative Escherichia coli (E. coli) and the gram-positive Bacillus subtilis (B. subtilis) were used as models to perform label-free, rapid and sensitive detection of bacteria based on SERS method. The antibacterial performance of the material was proved by the colony counting method, and the survival rates of E. coli and B. subtilis were as low as 8.05% and 0.06%, respectively. The Ti 3 C 2 T x -Au nanoparticles (NPs) irradiated with 808 nm light showed significant antibacterial effect within only 6 min, and the germicidal rates for E. coli and B. subtilis were 99.25% and 100%, respectively. The photothermal conversion efficiency reached as high as 43.40%. Accordingly, this multi-functional nanocomposite material can not only detect bacteria sensitively, but also has antibacterial and photothermal sterilization effects, which provides a greatly promising countermeasure for clinical treatment of diseases caused by multi-drug resistant bacteria.
