Efficient synthesis of 5-hydroxymethyl-2-furancarboxylic acid by Escherichia coli overexpressing aldehyde dehydrogenases

Xue-Ying Zhang,Xiao-Yang Ou,Yi-Jing Fu,Min-Hua Zong,Ning Li

Catalytic transformation of biomass-derived furans into value-added chemicals and biofuels has received considerable interest recently. In this work, aldehyde dehydrogenases (ALDHs) were identified from Comamonas testosteroni SC1588 for the oxidation of bio-based furans into furan carboxylic acids. Of the whole-cell biocatalysts constructed, Escherichia coli expressing a vanillin dehydrogenase ( E. coli _CtVDH1) proved to be the best for the oxidation of 5-hydroxymethylfurfural (HMF). 5-Hydroxymethyl-2-furancarboxylic acid (HMFCA) was obtained in the yield of approximately 92 % within 12 h using this recombinant strain when the HMF concentration was up to 200 mM. In a fed-batch process, 292 mM of HMFCA was produced within 20.5 h, thereby providing a productivity as high as 2.0 g/L h. Other furan carboxylic acids were synthesized in the yields of 83–95%. Besides, the partially purified HMF was smoothly converted into HMFCA by this recombinant strain, with a 90% yield.

Thiolation of methanol with H2S using core-shell structured ZSM-5@t-ZrO2 catalyst

Piao Liu,Jianxin Cao,Zheng Xu,Chunliang Yang,Xiaodan Wang,Fei Liu

Core-shell structured ZSM-5@t-ZrO 2 catalysts were prepared by a hydrothermal coating method, and their physicochemical properties, catalytic performance, and deactivation behavior in thiolation reactions were studied using a ZSM-5/t-ZrO 2 physically-blended catalyst as a comparison. Reactions using the core-shell ZSM-5@t-ZrO 2 catalyst were carried out at a pressure of 0.7 MPa, 370 °C, a H 2 S:CH 3 OH molar ratio of 2:1, and a N 2 flow rate of 90 ml·min −1 . The conversion of CH 3 OH, and the selectivity and yield of CH 3 SH under these conditions were 92.07%, 90.89%, and 83.68%, respectively, and the catalytic lifetime was longer than 30 h. Compared with the ZSM-5/t-ZrO 2 physically-blended catalyst, the CH 3 SH yield and the catalytic lifetime of the core-shell structured ZSM-5@t-ZrO 2 catalyst were higher by 5.37% and 150%, respectively. The unique reaction pathway of the shell (t-ZrO 2 , Lewis (L) base active site)-core (ZSM-5, Brǿnsted (B) acid active site) and the Lewis-Brønsted acid active center based on a composite solid-solution interface were mainly responsible for the high activity of the thiolation reaction. Meanwhile, the formation of a micro-mesoporous system increased the diffusion of molecules and reduced the deactivation rate of carbon- and sulfur-containing deposits. Generally, carbon- and sulfur-containing substances deposited on catalysts were mainly composed of polymethylbenzene, polymethyl naphthalene, cyclic elemental sulfur, and other compounds containing C S and S S bonds. Finally, the core-shell structured catalysts showed excellent resistance to deactivation by carbon- and sulfur-containing deposits.

Catalytic synthesis of 2,5-bis(hydroxymethyl)furan from 5-hydroxymethylfurfual by recombinant Saccharomyces cerevisiae

Zhi-Hao Xia,Min-Hua Zong,Ning Li

2,5-Bis(hydroxymethyl)furan (BHMF) is a versatile building block in the synthesis of polymers, fuels, and macrocycle polyethers. In this work, alcohol dehydrogenases (ADHs) were identified from Meyerozyma guilliermondii SC1103 and were heterologously expressed in Saccharomyces cerevisiae for the synthesis of BHMF from 5-hydroxymethylfurfural (HMF). Of recombinant strains constructed, S. cerevisiae expressing an aryl ADH ( Mg AAD1669) was observed to be the best catalyst. Upon process optimization, BHMF was afforded with a 99% selectivity and a 94% yield within 24 h at the substrate concentration of 250 mM. The space-time yield up to 3.4 g/L h was achieved in the fed-batch synthesis of BHMF. Inexpensive corncob hydrolysate proved to be a promising alternative to glucose as co-substrate for biocatalytic synthesis of BHMF, thus resulting in the significantly reduced production cost.

Comparative Study on the Dehydration of Biomass-Derived Disaccharides and Polysaccharides to 5-Hydroxymethylfurfural

Yuan Zhao,Kaifeng Lu,Hao Xu,Yang Qu,Lingjun Zhu,Shurong Wang

5-Hydroxymethylfurfural (HMF), a well-known value-added platform chemical, can be produced by biomass dehydration for the synthesis of bio-based liquid fuels. In this study, several kinds of saccharides that are widely available from biomass resources, namely, sucrose, maltose, cellobiose, lactose, inulin, and cellulose, were selected as feedstocks. A sulfonated ionic liquid catalyst was employed for the production of HMF from the selected saccharides in a renewable butanone–water solvent mixture. The yields of primary dehydration products versus time on stream from the saccharides were comparatively studied, and the distribution of the liquid-phase product was also analyzed. Results showed that the proposed reaction system could achieve fast conversion of disaccharides and polysaccharides with high selectivity toward HMF. Complete conversion of disaccharide and up to 65.1% yield of HMF were achieved within 1 min. Sucrose and inulin, which are rich in the fructose unit, showed the highest HMF selectivity. The next were maltose, cellobiose, and cellulose, which mainly consist of the glucose unit. They produced more oxygenated aliphatics than sucrose and inulin via side reactions. Lactose had the lowest HMF selectivity and tended to be converted into furan byproducts during the dehydration process.

Protein from Hylocereus polyrhizus protects MRC-5 cells against hydrogen peroxide (H2O2)-induced damage

Ding Haomiao,Zhang Yuzhe,Zha Yue,Zhou Sijia,Cao Chaoqing,Zhu Huajian,Xu Feng,Zhang Xiuqiang,Wang Caisheng

The cytoprotective and potential molecular mechanisms of Hylocereus polyrhizus protein (RFPP) were investigated on the hydrogen peroxide (H 2 O 2 )-triggered damage in normal human embryonic lung (MRC-5) cells. An MTT assay was conducted to assess the MRC-5 cell viability after exposure to H 2 O 2 or RFPP. Cell cycle distribution and apoptosis were explored via flow cytometry. The contents of related proteins were assessed via western blot. MRC-5 cells exhibited markedly decreased cellular viability after treatment with H 2 O 2 ; however, treatment with RFPP suppressed this decrease. Additionally, RFPP interference dampened H 2 O 2 -triggered intracellular apoptosis levels and increased H 2 O 2 -triggered intracellular S phase. In these processes, the contents of phosphorylated ( p )-AKT along with p -mTOR proteins were downregulated in 120 µM H 2 O 2 -treated cells compared with vehicle-treated cells. Nevertheless, in MRC-5 cells inoculated with RFPP, the levels expression of these proteins were reversed. To conclude, RFPP protected MRC-5 cells from H 2 O 2 -triggered damage via activation of the PI3K/AKT/mTOR cascade.

Insights into the active Fe species of Fe/ZSM-5 catalysts for ethane dehydrogenation

Zhengxi Zou,Zhenke Xu,Weiming Hua,Yinghong Yue,Zi Gao

Catalytic ethane dehydrogenation to ethylene is an attractive route for ethylene production following the shale gas revolution. However, its industrial implementation remains challenging due to high costs and environmental concerns. Iron-based catalysts have been identified as promising alternatives, while their catalytic performance is significantly dependent on the coordination environment and oxidation state of the iron species. Herein, a series of Fe/ZSM-5 catalysts were synthesized via urea precipitation (FeZ-P) and incipient wetness impregnation (FeZ-I) methods to investigate the relationship between Fe species evolution and catalytic behavior during ethane dehydrogenation. The FeZ-P catalysts exhibited significantly enhanced activity compared with FeZ-I, which was attributed to the improved dispersion of iron species and their favorable transformation behavior under reaction conditions. Detailed characterizations of fresh and spent catalysts revealed that the initial Fe 2 O 3 species on FeZ-P surface possessed limited dehydrogenation activity but underwent reaction-induced reduction and carbonization, generating Fe 3 C with enhanced ethane dehydrogenation performance. In contrast, the Fe 2 O 3 bulks on FeZ-I exhibited slower transformation rates, resulting in inferior catalytic activity. Air oxidation treatment enabled partial regeneration of the catalyst by removing carbon deposits and modifying Fe species evolution. This study highlights the importance of regulating Fe species dispersion and dynamic reduction-carbonization behavior, through which catalytically active Fe 3 C can be generated for enhanced ethane dehydrogenation performance. These findings provide valuable insights into the rational design of efficient Fe-based catalysts for non-oxidative alkane dehydrogenation.

Wen-Lin Tang,Yu Zhang,De-Xuan Hu,Hui Yang,Qian Yu,Jian-Wen Chen,Keli Agama,Yves Pommier,Lin-Kun An

DNA topoisomerase IB (TOP1) regulates DNA topological structure in many cellular metabolic processes and is a validated target for development of antitumor agents. Our previous study revealed that the benzophenanthridone scaffold is a novel chemotype for the discovery of TOP1 inhibitors. In this work, a series of novel 5-aminoethyl substituted benzophenanthridone derivatives have been synthesized and evaluated for TOP1 inhibition and cytotoxicity. Compound 12 exhibits the most potent TOP1 inhibition (+++) and cytotoxicity in human cancer cell lines with GI 50 values at nanomolar concentration range. 12 induces the cellular TOP1cc formation and DNA damage, resulting in HCT116 cell apoptosis . The pharmacokinetics , acute toxicity and antitumor efficiency in vivo of 12 were also studied.

Amphiphilic HZSM-5 for Cyclopentene Hydration at the Liquid–Liquid Interface in Pickering Emulsion

Changhao Bing,Xubin Zhang,Fumin Wang,Yi Zhai,Yongwang Li,Kaiwei Wang,Xiaolu Fan,Jinjin Zhang,Qi Shen,Xinyuan He

Zeolite is considered an ideal catalyst for olefin hydration due to its high specific surface area and abundant acid sites. However, the immiscibility of the water–oil two phases in olefin hydration limits mass transfer, and the side reaction of etherification occurs acutely, resulting in a low yield of alcohol. Thus, water–oil amphiphilic HZSM-5 was prepared by sulfonating silanized zeolite. The successful introduction of organic and sulfonic acid groups is demonstrated by FT-IR, TG, and water contact angles. Amphiphilic HZSM-5 can stabilize the Pickering emulsion and catalyze cyclopentene hydration at the phase interface. In addition, NH3-TPD and Py-IR show that the amount of strong Bro̷nsted acid sites of zeolites increases significantly after sulfonation. This facilitates the rate-determining step of cyclopentene activation by H+ to form carbocation. Moreover, the nucleophilic side reactions are inhibited by a high concentration of H+. Finally, under the optimized reaction condition, the conversion of cyclopentene can achieve 5.066% with a selectivity of 85.37% to cyclopentanol, which almost reaches the reaction equilibrium.

Promising valorisation method of chitin biomass by producing 5-hydroxymethylfurfural using microwave hydrothermal treatment

Yuyang Long,Liqun Xiao,Dan Zhou,Yanjun Meng,Lulu Wang,Dongsheng Shen

Chitin biomass is the second largest biomass resource on Earth but under-utilized. In this study, pretreated shrimp shells were converted into value-added platform chemical 5-hydroxymethylfurfural (HMF) using microwave hydrothermal treatment. Under the combined pretreatment of acid decalcification at room temperature and microwave-assisted alkali deacetylation, the HMF yield could reach 1.8 wt%. The key process parameters, including the holding temperature, holding time, and pH value, were evaluated and optimised. The highest HMF yield of 6.5 wt% was obtained at 202.6°C at a holding time of 5.8 min and a pH value of 1.5. This result demonstrates the potential of synchronously treating waste and recycling it, thereby offering a highly promising valorisation strategy for chitin-biomass utilisation.

Photothermal effect act as controllable switch for tunable photocatalytic selective oxidation of 5-Hydroxymethylfurfural

Zhenfei Yang,Xinnian Xia,Ming Fang,Longlu Wang,Yutang Liu

The selective oxidation of 5-hydroxymethylfurfural (HMF) to 2,5-dicarbofuran (DFF), and 2,5-furanedicarboxylic acid (FDCA) is essential to its practical application. Herein, a novel Pt/ZIS/MnO 2 (PMZIS) photocatalysts with photothermal effect were well designed for selective and controlled oxidation of HMF toward DFF or FDCA. The product distribution of HMF photo-oxidation on PMZIS can be regulated by the photothermal effect. At a low temperature (10 °C) with PMZIS, DFF is the primary oxidation product with the yield of 54.6% and selectivity of 85.4%. At a high temperature (60–70 °C) induced by photothermal effect, the FDCA can be obtained with the yield of 61.5 % and selectivity of 82.9%. Moreover, the Z-scheme transfer pathway and Pt cocatalysts realize the spatial charge separation structure of PMZIS heterojunctions. This unique structure greatly inhibits charge recombination and maximizes the activation ability of molecular oxygen to efficiently generate of the reactive oxygen species (e.g., 1O 2 and O 2– ), thus significantly improving the photocatalytic performance. This work would provide valuable insight in the product regulation through photothermal effect for the photocatalytic specific selective oxidation of organic molecules.

Catalytic Conversion of Chitin Biomass to 5-Hydroxymethylfurfural in Lithium Bromide Molten Salt Hydrates

Lu Lu,Xinlei Ji,Xuewei Wang,Fangming Jin,Xi Chen

5-Hydroxymethylfurfural (5-HMF) is one of the top valuable platform chemicals that can be derived from biomass. Herein, a facile, mild, and efficient catalytic conversion method was developed to convert chitin biomass into 5-HMF in lithium bromide molten salt hydrates (LiBr MSHs) with boric acid (BA) as the catalyst. About 49.6% 5-HMF yield was obtained from glucosamine (GlcN) in 65 wt % LiBr MSHs at 130 °C for 2 h with 800% BA addition. The system is applicable to other sugars, and the C2-substituted group of sugars played a significant role in the 5-HMF yield. The 5-HMF yields were 81.3, 40.3, and 3.9% from fructose, glucose, and N-acetyl glucosamine (GlcNAc), respectively, under identical conditions. Combined with acid addition and Box–Behnken experiment designs, the direct conversion of chitosan and chitin were achieved at 140 °C for 5 h to produce 5-HMF with 21.6 and 15.3% yield. Ammonia quantitative analysis and NMR tests have been conducted, based on which a plausible reaction pathway was proposed. The LiBr MSHs and the BA catalyst facilitated the hydrolysis, deacetylation, and ring-opening of chitin biomass. Especially, the catalyst BA played a key role in the formation of 5-HMF, as it was found to promote dehydration at the last step. The study put forward new avenues for chitin refinery to produce high-value furanic platform chemicals in cheap and green solvent systems.
