Traumatic brain injury (TBI) disrupts oxygen homeostasis in the brain, leading to excessive reactive oxygen species (ROS) production and dysregulated antioxidant mechanisms, which fail to clear excess ROS. This ROS overload promotes the expression of pro-inflammatory genes, releasing cytokines and chemokines and creating a vicious “ROS-neuroinflammation” cycle, making it essential to break this cycle for effective TBI treatment. In this study, we developed cysteine-alanine-glutamine-lysine (CAQK) peptide-modified antioxidant nanoparticles (C-PPS/C) for co-delivery of curcumin (Cur) to modulate oxidative and neuroinflammatory disturbances after TBI. In TBI mice, C-PPS/C nanoparticles accumulated in injured brain regions, where poly (propylene sulfide) 120 scavenged ROS, reducing oxidative stress, while Cur release further suppressed ROS and inflammation. C-PPS/C nanoparticles broke the “ROS-neuroinflammation” cycle, protecting the blood-brain barrier (BBB), reducing acute brain edema, and promoting long-term neurological recovery. Further investigation showed that C-PPS/C nanoparticles inhibited the NF-κB pathway, reducing pro-inflammatory gene expression and mitigating inflammation, suggesting a promising approach for TBI treatment.

Thiol-modified hyaluronic acid (HASH) improves zein nanoparticles (ZNPs) stability by disulfide bonding with zein. However, the effect of the degree of thiol substitution in HASH on the stability of the ZNPs remains unclear. In this study, HASH with 1%, 3%, and 5% degrees of thiol substitution was synthesized and used as stabilizers to prepare three composite NPs. The pH and salt ion stability of the composite NPs increased with increasing degree of thiol substitution, which improved the antioxidant and anti-inflammatory capacities of the composite NPs. Interestingly, when the degree of thiol substitution exceeded 3%, the physical stability of composite NPs did not improve further. Our results revealed the relationship between the degree of thiol substitution in HASH and the stability of ZNPs, and provided the optimal degree of thiol substitution. These findings provided important information for improving the physical stability of ZNPs and contributed to the development of a delivery system for natural actives.

The harms caused by ultraviolet (UV) and blue light to eyes are attracting momentous concern due to growing exposure to artificial illumination and modern IT devices. Herein, a simple and eco-friendly adsorption approach was employed to integrate curcumin, a natural bioactive compound, into the cellulose substrate for the development of flexible and biodegradable filters capable of blocking harmful light. The curcumin/cellulose films demonstrate excellent UV-screening competence and photostability, with UV-A and UV-B screening ratios ranging from 92.8 % to 100 % and 89.2 % to 100 %, respectively. The films could block >96 % of blue light in the wavelength range of 400–500 nm. Meanwhile, the films maintain high transmittance (85.2–89.4 %) and low haze (2.0–2.7 %). The films can efficiently block blue light emanated from sunlight, light-emitting diodes, lighting systems, computer and mobile phone screens. Encouragingly, the incorporation of curcumin led to a substantial increase in the water contact angle, elevating it from 41.6 to 81.3°. Furthermore, the films exhibit excellent antimicrobial properties, biodegradability, and tensile strength in excess of 72 MPa. Therefore, these films fabricated entirely from natural resources have the potential to achieve practical applications such as food packaging and spectacle lens, especially suitable for electronic screen protectors.

Inspired by the milk fat globule membrane composition, we investigated the interaction between whey protein isolate (WPI) and soybean lecithin (SL) at various mass ratios (10:0, 10:1, 10:2, 10:5, 10:10, and 10:15) and their effects on curcumin emulsions. Our results demonstrated that SL interacted with WPI through hydrophobic interactions and hydrogen bond restructuring. The WPI–SL complex effectively enveloped oil droplets, resembling natural milk fat globules. Notably, the 10:10 sample exhibited the smallest droplet size (217.33 nm), lowest polydispersity index (0.202), and lowest zeta potential (−43.56 mV). After heat treatment at 90 °C for 1 h, the 10:10 sample exhibited the highest thermal stability , and its polydispersity index (0.236) was significantly lower than at other mass ratio (0.332–0.268). During in vitro digestion, emulsions stabilized by the WPI–SL complex exhibited less oil droplet aggregation, resulting in enhanced curcumin bioaccessibility (58.73–66.32 %) compared to pure WPI-stabilized emulsions (57.45 %). These findings signify the potential of WPI–SL complexes in constructing effective delivery systems mimicking natural fat globule structures, thereby improving curcumin bioavailability. The systematic investigation of different WPI–SL mass ratios in this study provides valuable insights for optimizing emulsifier compositions in the food industry.

The potential of dual protein–polyphenol supramolecular complexes for constructing delivery systems based on high internal phase Pickering emulsions (HIPPEs) is significant, yet the underlying mechanism has not been fully revealed. This study systematically investigated hybrid supramolecular complexes fabricated through physical processing techniques such as ultrasound (US) and high pressure (HP), integrating sunflower-walnut protein hydrolysates (SW) with naringenin (Nar) as novel stabilizers for oil-in-water HIPPEs. While physical treatments effectively enhanced SW emulsification capacity through particle size reduction, interfacial tension decrease, and wettability increase, particularly under US treatment, the physical field (US and HP) has limited effectiveness in improving the emulsion’s cream stability. The synergistic application of physical processing and naringenin complexation induced SW structural unfolding, facilitating the adsorption and anchoring of SW at the oil–water interface. Compared to the SW-Nar (US), SW-Nar (HP) generated emulsions with smaller droplets, reinforced viscoelastic networks, and improved curcumin bioaccessibility. In vitro digestion induced SW-Nar complexes interfacial structural unfolding, hydrophobic exposure, and α-helix-to-β-sheet transition, enhancing emulsification and micelle formation via strengthened hydrogen bonds and hydrophobic interactions, particularly in SW-Nar (HP). These findings collectively establish SW-Nar-stabilized HIPPEs as advanced delivery platforms for lipophilic bioactives, with system performance critically dependent on supramolecular interfacial microstructure modulated by physical processing methods.

Alzheimer’s disease (AD) is characterized by the accumulation of amyloid beta (Aβ) plaques, leading to neuronal death. Notably, there are no therapeutically efficacious treatments for this disorder. This study investigates the neuroprotective mechanisms of curcumin. In vitro experiments demonstrated that curcumin significantly promoted the proliferation of neural stem cells (NSCs) derived from E16 mouse embryos. Additionally, in the APP/PS1 transgenic mouse model, curcumin improved cognitive function, as evidenced by a shorter escape latency and increased platform crossing frequency in the Morris water maze test and enhanced novel object recognition. Histological analysis showed that curcumin reduced Aβ accumulation in the hippocampal and cortical regions. Molecular studies revealed that curcumin upregulated the expression of BDNF and enhanced phosphorylation of CREB. Collectively, these findings indicate that curcumin exerts neuroprotective effects through multiple mechanisms, including promoting NSC proliferation, reducing Aβ accumulation, and the potential modulation of the BDNF-CREB signaling pathway.

Freshness deterioration of salmon during cold storage poses significant safety concerns, highlighting the need for real-time monitoring strategies. Herein, a novel starch-pectin film incorporating Nisin-A and curcumin nano-emulsion (SP-NC film) was developed for real-time monitoring of salmon freshness. FTIR, XRD, and molecular dynamics simulations confirmed strong molecular compatibility and stable hydrogen bonding among film components. The SP-NC film exhibited high mechanical strength, with a tensile strength of 4.51 MPa and elongation at break of 43.58 %, as well as outstanding barrier properties reflected by low water vapor permeability (6.82 × 10 −10 g·mm −1 ·s −1 ·Pa −1 ) and oxygen permeability (6.10 × 10 −9 g·mm·mm −2 ·s −1 ). Under blue light, the SP-NC film reduced S. aureus and Salmonella survival rates to 1.85 % and 3.79 %, respectively. When applied to 4 °C refrigerated salmon, the SP-NC film extended shelf life from 4 to 8 days and provided visual spoilage indication. Overall, this study offers a valuable reference for the design of intelligent food packaging.

Zein-fucoidan nanoparticles (ZFNPs) as an excellent colloidal delivery system for bioactive compounds, such as curcumin (CUR), show a great application potential in the food industry. Herein, various amount of ZFNPs, ZFNPs loaded with CUR (CZFNPs) and free CUR were added into golden threadfin bream ( Nemipterus virgatus ) surimi to explore their impacts on the gel properties. Dynamic rheological tests showed that storage modulus (G') and loss modulus (G") of surimi gels with the additive amount of ZFNPs and CZFNPs under 0.8 % significantly increased and their loss tangent (tan δ) slightly reduced during thermal gelation, indicating stronger networks and better structural configuration. Puncture test and textural profile analysis confirmed the improvements in breaking force, hardness, chewiness and gumminess of surimi gels. Furthermore, 0.8 % of ZFNPs and CZFNPs induced conformational changes of surimi proteins from α-helix to β-sheet and random coil, and then facilitating the cross-link between proteins. Low-field nuclear magnetic resonance revealed that ZFNPs and CZFNPs promoted the redistribution of water in surimi gel networks, converting more free water to tightly bounded water, and thus increasing the water holding capacity. However, the addition of equivalent amount of CUR caused detrimental to surimi gel properties. In conclusion, ZFNPs and CZFNPs conferred better gel properties and nutritional characteristics to surimi gels, and this study provided a theoretical basis for the development of high-quality surimi products.

Curcumin (Cur) as a natural pigment and biological component, can be widely used in food and beverages. However, the water insolubility of Cur significantly limits its applications. In this study, we prepared a series of nanocrystals via ultrasound-assisted method to improve the solubility and availability of Cur. The results showed artemisia sphaerocephala krasch polysaccharide (ASKP), gum arabic (GA) and wheat protein (WP) were outstanding stabilizers for nanocryatals except traditional agent, poloxamer 188 (F68). The obtained curcumin nanocrystals (Cur-NC) displayed a rod-shaped, crystal- and nanosized structure, and extremely high loading capacity (more over 80 %, w/w). Compared with raw powder, Cur-NC greatly improved the water solubility and dispersibility, and the slow and complete release of Cur of Cur-NC also endowed them excellent antioxidant capacities even at 10 μg/mL. Importantly, as functional factor additive in beverages (e.g. water and emulsion), Cur-NC could increase the content of Cur to at least 600 μg/mL and retain a good stability. Overall, we provided an effective improvement method for the liposoluble active molecules (e.g. Cur) based on the nanocrystals, which not only tremendously enhanced its water solubility, but also strengthened its bioactivity. Notably, our findings broadened the application of water-insoluble compounds.

Due to the complex physiological and pathological barriers in tumor tissues, the penetration depth of light is limited and the accumulation and distribution of photosensitizers in the tumor are insufficient, which seriously affects the therapeutic effect of photothermal therapy (PTT) on solid tumors. The preparation process of traditional photothermal agents is complicated, and their biosafety needs further improvement. Moreover, photothermal agents are difficult to rapidly excrete from the body after realizing the therapeutic function, which may cause long-term potential harm to the body. In this study, multifunctional nanoparticles (FeCNPs) consisting of iron(III), curcumin, and poly(ethylene glycol) grafted polyglutamate (PLG-g-mPEG) were obtained through a simple one-pot method. The synthesized FeCNPs exhibited an average hydrodynamic size of 125 nm and a polydispersion index (PDI) of 0.462. Furthermore, the nanoparticle has excellent biocompatibility and a photothermal effect. After the PPT, the FeCNPs break down into harmless elements via a dynamic disassembly mechanism triggered by deferoxamine mesylate (DFO), facilitating swift elimination and preventing possible long-term toxicity to major organs. The therapeutic synergies enable efficient tumor removal. In summary, this study demonstrates an approach for the dynamic disassembly of metal-polyphenol nanoparticles, effectively addressing the challenge of balancing the EPR effect and renal clearance.

Radiation-induced oral mucositis (RIOM) substantially compromises the efficacy of cancer treatment and patient survival, while current therapeutic approaches exhibit limited efficacy in addressing its multifactorial pathogenesis. Curcumin possesses multi-targeted pharmacological properties relevant to RIOM, including antioxidative, anti-inflammatory, and mucosal repair effects, rendering it an ideal therapeutic agent. This study developed a curcumin-based intelligent-responsive integrated therapeutic platform (Cur-VE-Zn@gel) via a hierarchical assembly strategy. Specifically, curcumin was covalently bound to vitamin E to form the active center, while its coordination with Zn 2+ facilitates self-assembly into stable polymeric structures. This design not only significantly enhanced the biosecurity of curcumin but also amplified its antioxidant, anti-inflammatory, and tissue repair capabilities, while exhibited stable DNA repair characteristics, thus establishing a comprehensive radioprotective barrier. This bioactive complex was further incorporated into an adhesive thermosensitive hydrogel consisting of chitosan and poloxamer, which enabled targeted delivery to oral mucosa and bypassed the low systemic bioavailability of curcumin. Comprehensive in vitro and in vivo evaluations demonstrated that the Cur-VE-Zn@gel platform effectively modulated DNA damage repair, oxidative stress, inflammatory cascades, and epithelial regeneration, thereby achieving synergistic multi-targeted therapy for the suppression of RIOM. Ultimately, this biomaterial-based strategy successfully addresses the key pharmacological limitations of curcumin through structural modification and targeted delivery, providing a highly promising, safe, and effective approach for the clinically challenging management of RIOM.

In infected full-thickness skin injuries, deep wound exposure accompanying bacterial invasion and persistent inflammatory lead to prolonged wound healing, remaining a major challenge in clinical wound healing. In this work, an emulsion gel (SCEG) with pH/enzyme-responsive release for promoting infected wound repair was successfully constructed based on the Spirulina protein isolate/carboxymethyl chitosan (SPI/CMC) system. The SCEG was prepared by cross-linking the SPI/CMC emulsion with tetrakis (hydroxymethyl) phosphonium sulfate (THPS) through the Mitchell reaction. It effectively encapsulated the curcumin (Cur) with an encapsulation efficiency of 87.26 % and exhibited pH/enzyme-responsive release behavior. The SCEG showed excellent antibacterial activity against both S. aureus and E. coli . Additionally, it can evidently alleviate oxidative stress and maintain cell activity. Meanwhile, it could significantly reduce the secretion of pro-inflammatory cytokines TNF-α and IL-1β. Finally, a model of full-thickness skin damage infected with S. aureus was built to assess the wound healing effect of SCEG. The result showed that it can efficaciously inhibit bacterial infection of the wound, reduce inflammatory response, promote angiogenesis, accelerate tissue regeneration, and thus boost the healing process. Altogether, the emulsion gel dressing is expected to be a safe, efficient and promising candidate material for the field of infectious wound care.
