en Isobolographic analysis was used to assess the antioxidant interactions (synergism, addition, and antagonism) of 4 common vegetables (tomato [T], carrot [C], eggplant [E], and purple potato [P]). The lipophilic (L) extracts of T and C (main carotenoids), the hydrophilic (H) extracts of E and P (main phenolics) were mixed by the certain ratios (1:9, 3:7, 1:1, 7:3, 9:1, w/w) and their antioxidant activities were investigated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2’-Azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS) radical scavenging assays, respectively. Most of the binary mixtures (LC-HE, LC-HP, HE-HP, LT-HE, and LT-HP combinations) showed the synergistic antioxidant effects. In DPPH assay, the greatest antioxidant activity of vegetable combinations was 1:9 LT-HP (EC 50 : 2.45 ± 0.13 mg/mL), followed by 9:1 HE-HP (EC 50 : 3.62 ± 0.12 mg/mL) and 1:9 LC-HE (EC 50 : 3.74 ± 0.47 mg/mL). In ABTS assay, the greatest antioxidant activity of vegetable combinations was 9:1 HE-HP (EC 50 : 4.20 ± 0.10 mg/mL), followed by 7:3 HE-HP (EC 50 : 4.41 ± 0.63 mg/mL) and 1:1 HE-HP (EC 50 : 5.35 ± 0.85 mg/mL). Among these combinations, 1:1 LC-HE combination showed the highest synergistic antioxidant effects in DPPH assay (synergistic rate: 87.4%), and 7:3 LC-HE combination showed the highest synergistic antioxidant effects in ABTS assay (synergistic rate: 87.0%). The mixtures of phenolics and carotenoids with suitable ratios in vegetables effectively enhanced the synergistic antioxidant effects. Practical Application pt In this study, the results indicate that the combination of different vegetables with suitable ratios could improve the antioxidant activities through synergistic effects of phytochemicals. Also, isobolographic analysis could be well used in evaluating the antioxidant interactions between antioxidants.

To analyze the components of the fatty oil extracted from Antrodia camphorata fungus powder and to evaluate the antioxidant activity of A. camphorata crude oil. A derivatized gas chromatography mass spectrometry (GC–MS) was developed for the quantification of 19 fatty acids in extracts of A. camphorata obtained by supercritical carbon dioxide (SC-CO 2 ). Good separation was obtained under the optimized chromatographic conditions and 19 target compounds after methyl-esterification were identified by GC–MS on a HP-VOC capillary column (60 m × 320 μm × 1.8 μm) with an initial temperature set at 80 °C. The validity of the established method was examined experimentally with good linearity, intra-assay precisions, repeatability, stability and recovery. The antioxidant activity of A. camphorata crude oil was evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging assay. Nineteen fatty acids showed good linearity over the tested ranges (r > 0.9956) and the recovery ranged from 93.47 to 104.89 %. The crude oil extracted from A. camphorata fungus powder also revealed its antioxidant activity. It was the report about simultaneous determination of 19 fatty acids in A. camphorata and antioxidant activity of its crude oil for the first time. The established method might also be utilized for the investigation of edible plant materials and agricultural products containing fatty acids.

Ferns are famous traditional medicinal plants and 300 fern species have been used as phytomedicines by Southwestern of China. However, there is little information on the phytochemicals and pharmacological characteristics of most fern species. The total flavonoid concentrations, antioxidant activity, anticancer activity, and acetylcholinesterase inhibition of extracts from 19 species of ferns were investigated. The total flavonoid concentrations ranged from 8.6 to 306.4 mg/g (w/w). The antioxidant activity including DPPH free radical scavenging, ABTS radical scavenging, and superoxide anion scavenging, reducing power and ferric reducing antioxidant potential (FRAP) of flavonoid extracts from these ferns showed a weak linear relationship with their total flavonoid concentrations. Selaginella frondosa with very low total flavonoid concentration (13.4 mg/g) showed highest inhibition against A549 cells and it is necessary to further investigate the anti-cancer compounds in S. frondosa . The extracts from Davallia cylindrica Ching and Stenoloma chusanum Ching significantly inhibited AChE activity, which illustrated that it is worthy of attention to investigate flavonoids from D. cylindrica and S. chusanum as AChE inhibitors.

Novel galloyl phytosterols were rationally designed by incorporation of gallic acid into phytosterols through straightforward esterification . The esterification was successfully achieved by coupling of gallic acid and phytosterols through a mild chemical Steglich esterification reaction that is more straightforward than the enzymatic method . The identity of the newly synthesized galloyl phytosterols was confirmed by NMR, HPLC–MS and IR spectroscopies . Further evaluation of the novel galloyl phytosterols with radical scavenging, ferrous ion chelating, and Rancimat methods revealed its excellent antioxidant activities that are comparable to the most potent fat-soluble antioxidants . This novel antioxidant offers an intriguing solution for naturally derived antioxidants and will have great potential application as antioxidant in food industry. The methods developed in this study will be valuable for development of other phenolic phytosterols.

Myricetin shows low oral bioavailability (<10%) in rats due to poor aqueous solubility, though it has various pharmacological activities. Complexation with cyclodextrins (CDs) is a potent pharmaceutical method to enhance the bioavailability of poorly soluble compounds. The myricetin/HP-β-CD inclusion complex was prepared and confirmed by DSC, PXRD, and SEM. Here, the inclusion mode is described in detail with regard to structural and energetic aspects using a phase solubility diagram and 1 H NMR, NOESY, and FT-IR spectra. The water solubility and dissolution rate of myricetin were greatly enhanced by forming the myricetin/HP-β-CD inclusion complex. Consequently, the oral bioavailability of the myricetin/HP-β-CD inclusion complex in rats was effectively increased 9.4-fold over free myricetin, and its antioxidant activity was also improved. The present study provides useful information for the potential application of complexation with myricetin, a naturally occurring hydrophobic phenolic compound in herbal medicine.

Four polysaccharides (TPS-F, TPS-V, TPS-S and TPS-M) were obtained from tea ( Camellia sinensis ) leaves by freeze-drying, vacuum-drying, spray-drying and microwave-vacuum drying, respectively. Their chemical composition and biological properties were comparatively studied. The results showed these TPS were similar in IR, UV absorption and distribution of molecular weight. However, they showed significant differences ( P < 0.05) in yields of crude polysaccharides and contents of protein and total polyphenols. Furthermore, morphological analysis showed their surface differed from each other in size and shape when viewed by SEM, from which surface of TPS-F was rough and porous, surface of TPS-S was evenly particles, and surfaces of the other two were similar to anomalistic bricks. In addition, the bioactivity of them was also evaluated. It was found TPS-F exhibited relatively better ability on metal chelating and superoxide radicals scavenging assays than others, and TPS-V exhibited higher ability on α-glycosidase and α-amylase inhibition assays than others with inhibitory percentages of 82.75% and 92.8%.

Three derivatives of polymannuronic acid (PM) were prepared by chemically polyanionic modification. PM was substituted with phosphate ( OPO 3 2− ), H-phosphonate ( OPO 2 H − ) and sulfate ( OSO 3 − ) groups with the degree of substitution (DS) of 0.41, 1.42 and 1.04, respectively. The structures of all PM derivatives were characterized by FT-IR, 31 P NMR and 13 C NMR spectroscopy . The weight average molecular weight (Mw) was determined by high performance gel permeation chromatography (HPGPC). The antioxidant activities of PM and its derivatives were evaluated in vitro. The results indicated that both phosphate and H-phosphonate groups improved the hydroxyl radical scavenging activity, while sulfate group enhanced the superoxide radical scavenging activity. However, all of the derivatives scavenged DPPH less effectively than PM. The mechanism for how the different anionic substituent groups influenced the antioxidant activities was discussed.

The ultrasound-assisted extraction of phenolic compounds from Euryale ferox seed shells was modeled using response surface methodology. A three-level-three-factor Box–Behnken design was employed to optimize three extraction variables, including extraction time ( X 1 ), ethanol concentration ( X 2 ), and ratio of aqueous ethanol to raw material ( X 3 ), for the achievement of high extraction yield of the phenolic compounds. The statistical analyses show that the independent variables ( X 1 , X 2 ), the quadratic terms ( X 1 2 , X 2 2 and X 3 2 ), and the interactions of X 1 with X 2 and X 3 have significant effect on the yield ( p < 0.01). The optimized conditions are X 1 of 21 min, X 2 of 52%, and X 3 of 31 mL/g. Under these conditions, the experimental yield is 15.69 ± 0.082% ( n = 3), which is well matched with the predicted yield of 15.70%. The evaluation of antioxidant activity by DPPH assay indicates that the phenolic compounds from E. ferox seed shells possess significant antioxidant activity. HPLC analysis reveals that pyrogallol, gallic acid, chlorogenic acid and rutin are the major composition in the extracts.

Summary In this study, microwave-assisted extraction with ethanol (MAEE) of star anise oil from Illicium verum Hook.f . has been optimised by response surface methodology (RSM). A maximum yield of star anise oil was obtained at an optimum condition: the ratio of solvent to sample 17 mL g −1 , extraction time 16 min and microwave power 505 W. Accordingly, the highest yield of star anise oil was about 24.98%, which was much higher than that of steam distillation (SD), 7.17%. Oxygenated organic compounds in representative of trans-anethole are a major component in star anise oil, nearly 94.21% for SD and 86.66% for MAEE, identified and determined by GC-MS. The oils extracted by SD and MAEE both have strong antioxidant activities that were demonstrated by the DPPH and ABTS assays.

The antidiabetic and antioxidant activities of the ethyl acetate-soluble extract (MFE) of mulberry fruit (Morus alba L.) were investigated. In vitro , MFE showed potent α-glucosidase inhibitory activity and radical-scavenging activities against DPPH and superoxide anion radicals. In vivo , MFE could significantly decrease fasting blood glucose (FBG) and glycosylated serum protein (GSP), and increase antioxidant enzymatic activities (SOD, CAT, GSH-Px) in streptozotocin (STZ)-induced diabetic mice. Bioactivity-guided fractionation of the MFE led to the isolation of 25 phenolic compounds, and their structures were identified on the basis of MS and NMR data. All the 25 compounds were isolated from mulberry fruit for the first time. Also, the α-glucosidase inhibitory activity and antioxidant activity of the phenolics were evaluated. Potent α-glucosidase inhibitory and radical-scavenging activities of these phenolics suggested that they may be partially responsible for the antidiabetic and antioxidant activities of mulberry fruit.

Ethnopharmacological relevance Smilax riparia A. DC., known as “Niu-Wei-Cai” in China, is distributed through the south and middle of China. The roots and rhizomes of Smilax riparia have been used not only as traditional Chinese medicines (TCMs) for the treatment of bronchitis , lumbago of renal asthenia , traumatic injury, asthenia edema, and cancer but also as edible wild herbs in some areas of China. Aim of the study To identify the phytochemicals in the roots and rhizomes of Smilax riparia and to investigate their antioxidant activities and cytotoxicities toward several tumor cell lines. Materials and methods Four fractions and five phenylpropanoid glycosides were obtained from roots and rhizomes of Smilax riparia under bioassay-guided screenings. The structures of five compounds were elucidated by spectroscopic methods and compared with published data. We evaluated their antioxidant activities and their cytotoxicities on five cancer cell lines: human promyelocytic leukemia (HL-60), human hepatocellular carcinoma (SMMC-7721), human lung cancer (A-549), human breast cancer (MCF-7), and human colon cancer (SW480). Results Of the five glycosides, one new compound (3, smilaside P) was isolated from an EtOAc fraction. Compound 1 was cytotoxic toward HL-60, SMMC-7721, A-549, MCF-7, and SW480 (IC 50 2.70, 3.80, 11.91, 3.79, and 3.93 μM, respectively). Moreover, compounds 1 – 3 showed moderate scavenging activities against the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical (IC 50 339.58, 330.66, 314.49 μM, respectively). Conclusions Five phenylpropanoid glycosides were reported for the first time from this TCM. Each was studied, as observed here for the first time, in the cytotoxic experiments toward HL-60, SMMC-7721, and SW480 cell lines. Compound 1 , bearing three feruloyl groups and three acetyl groups, had the greatest cytotoxicity toward the five tumor cell lines. Compounds 1 – 3 showed moderate antioxidant activities. All results reflect that compounds 1 – 3 are cytotoxic for a wide variety of cancer cell lines of differing tissue origins and that the cytotoxicities of these compounds may be related to their antioxidant activities.

Caulis Bambusae in Taenia is a medicinal preparation from Bambusa tuldoides Munro consisting of skinless slices of the stem (bamboo shavings) and used as a traditional health food in tea, wine, and soup in Asia. Three novel lignans, (−)-7′-epi-lyoniresinol 4,9′-di-O-β-d-glucopyranoside (7), (−)-lyoniresinol 4,9′-di-O-β-d-glucopyranoside (8), bambulignan A (10), and seven known lignan compounds (1–6 and 9) were isolated from Caulis Bambusae in Taenia. The structures of the lignans were determined by detailed spectroscopic analysis (HRESIMS, HSQC, HMBC, NOE). All the isolated lignans were tested for antioxidant activities by DPPH and FARP assays. The results showed that the compounds (+)-lyoniresinol 9′-O-β-d-glucopyranoside (1) and (−)-7′-epi-lyoniresinol 9′-O-β-d- glucopyranoside (9) have strong free radical scavenging activity and reducing power.
