BACKGROUND Insect glutathione S-transferases (GSTs) play a crucial role in insecticide detoxification. However, there remains a distinct lack of information regarding the role of GSTs in the detoxification of Tolfenpyrad (TFP) in insects. RESULTS Real-time quantitative PCR showed significant upregulation of PxGSTs after exposure to TFP for 6 h. An in vitro inhibition assay showed that TFP could inhibit PxGSTδ, PxGSTε and PxGSTσ, and the most pronounced inhibitory effect was on PxGSTσ. Metabolism assays displayed that PxGSTσ was superior to other test PxGSTs in metabolizing TFP. The molecular docking of TFP and PxGSTσ revealed that the H-bond provided by the sidechains of Tyr107 and Tyr162 were key to the detoxification of TFP by PxGSTσ. Further tests using mutant PxGSTσ proteins at the sites of Tyr107 (PxGSTσY107A) and Tyr162 (PxGSTσY162A) corroborated that the individual replacement of Tyr107 and Tyr162 could greatly weaken the binding and metabolic abilities to TFP. CONCLUSION Metabolic interactions between the Plutella xylostella (L.) GSTs (PxGSTs) and TFP were deciphered. This study illustrates the molecular metabolism mechanism of PxGSTσ towards TFP and provides theoretical underpinnings for the design and optimization of novel TFP-like insecticides. © 2020 Society of Chemical Industry

A novel smartphone-assisted fluorescent sensor based on europium/zirconium metal-organic framework (Eu 0.5 /Zr 0.5 -MOF) was developed for the fast and sensitive determination of doxycycline (DOX) and L-arginine (Arg). After the addition of DOX, the fluorescence of Eu 0.5 /Zr 0.5 -MOF was quenched owing to the inner filter effect (IFE). When Arg was introduced into the Eu 0.5 /Zr 0.5 -MOF@DOX complex system, the fluorescence was recovered because the interaction between Arg and Eu 0.5 /Zr 0.5 -MOF@DOX weakened the IFE. Moreover, the Eu 0.5 /Zr 0.5 -MOF produced continuous fluorescence color changes for the visual measurement of DOX and Arg. The fluorescent probe for DOX and Arg offered broad linear ranges of 0.05–80 and 0.1–60 μg/mL, respectively, with detection limits as low as 2.07 and 67.5 ng/mL. The proposed method was successfully applied to monitor DOX in eggs and Arg in human serum. This work provides a powerful platform for the real-time and visual analysis of DOX and Arg in food and biological samples.

The study aimed to prepare the chelate of L-theanine (LT) with zinc (Zn) and evaluate its stability, antioxidant and antidiabetic activity. The chelation mechanism between Zn 2+ and LT was investigated by ultraviolet-visible, Fourier transform infrared, scanning electron microscopy/energy dispersive spectrometer and 1 H nuclear magnetic resonance spectroscopy. L-theanine-Zn chelate (LT-Zn) was obtained using LT to Zn citrate in a mass ratio of 3:1 at pH 6 and 70 °C for 180 min with the chelation efficiency of 61.86 ± 1.26%. The results revealed the amino nitrogen atoms and carboxyl oxygen atoms in LT chelated with Zn ions, and the mass of the chelate was confirmed by liquid chromatography with tandem mass spectrometry. Scanning electron microscopy with the element analyzed by energy dispersive spectroscopy showed that the morphology of LT-Zn consisted of irregular granular particles with rough surface. The LT-Zn exhibited higher solubility and improved stability under 100 °C, various pH condition and different concentrations of glucose and NaCl ( p < 0.05) than that of sulfate Zn and Zn gluconate. Compared to sulfate Zn and Zn gluconate, the LT-Zn showed better stability during simulated gastrointestinal digestion. The chelate also exhibited antioxidant activity, as well as an inhibitory effect on α-glucosidase and α-amylase. Overall, the study provided a novel approach to prepare Zn nutraceuticals with antioxidant and antidiabetic properties.

β-Alanine is an important non-essential amino acid and used in the manufacture of a wide range of drugs, in particular it plays a key role in the biosynthesis of carnosine and pantothenic acid. L-aspartate-α-decarboxylase (PanD) is the key enzyme in the enzymatic preparation of β-alanine. In this study, BsPanD* was modified by protein structure prediction and directed evolution, and a mutant BsPanD* PQ4 with good catalytic properties was obtained, which showed a 40 % increase in activity and remained stable in the pH 6.0–9.0 range. The strain E.coli pRSF Duet-1 - smaiA - ecaspA (pMA) was constructed for efficient co-expression of maleate cis - trans isomerase and L-aspartase, with an apparent activity of 253.4 U/g. The multi-enzyme cascade catalytic process for the preparation of β-alanine with maleate as the substrate was developed. The results showed that when the concentration of ammonium maleate was 1 mol/L, the substrate conversion rate was greater than 99.9 %, and the β-alanine concentration was 87.5 g/L with a yield of 98.3 %. This study not only solved the problem of low activity and poor stability of PanD, but also realized the preparation of β-alanine from maleate, a cheap chemical raw material, and laid the foundation for its industrial application.

Mentha arvensis L. (M. arvensis) is an aromatic plant of the Mentha genus, renowned for its medicinal and economic importance. The primary components of its essential oils (EOs) are monoterpenoids, synthesized and stored in peltate glandular trichomes (PGTs). In general, the EO content in M. arvensis is relatively low. Methyl jasmonate (MJ) has been reported as an effective elicitor of terpenoid biosynthesis in medicinal plants, but the specific mechanisms underlying MJ’s influence on M. arvensis remain unclear. In this study, exogenous application of MJ significantly increased the EO content, yield, and PGT density in a dose-dependent manner. At a 5 mM dose, the EO content and PGT density peaked, with increases of 71.20% and 53.69%, respectively. Gas chromatography-mass spectrometry (GC-MS) analysis indicated that, in general, MJ treatment did not significantly alter the types or relative proportions of EO components of M. arvensis. However, L-menthol content decreased slightly by 7.90% under 5 mM MJ treatment. Transcriptome analysis identified 4,659 differentially expressed genes (DEGs) in MJ-treated leaves. KEGG enrichment analysis revealed that “Monoterpenoid biosynthesis” was among the most significantly enriched metabolic pathways. Key genes involved in jasmonic acid (JA) signaling (JAZs and MYCs) and monoterpenoid biosynthesis (GPPSs, LSs, L3Hs, and IPRs) were significantly up-regulated. Co-expression analysis, promoter binding element analysis and weighted gene co-expression network analysis (WGCNA) indicated that transcription factors (TFs) such as AP2/ERF, WRKY, MYB, and bHLH play crucial roles in regulating MJ-mediated monoterpenoid biosynthesis. Several key candidate TFs potentially involved in regulating monoterpenoid biosynthesis in M. arvensis were identified. These findings provide valuable insights into the molecular mechanisms regulating monoterpenoid accumulation in the Mentha genus.

l-Tryptophan decarboxylase (TDC) and l-3,4-dihydroxyphenylalanine decarboxylase (DDC) catalyze the decarboxylation of l-tryptophan, 5-hydroxytryptophan, and l-3,4-dihydroxyphenylalanine. In this study, we analyzed the amino acid compositions of the substrate-binding pockets of TDC from Catharanthus roseus (CrTDC) and DDC from Sus scrofa (SsDDC), explored the specificity of key amino acids within these pockets, and elucidated mechanisms influencing substrate selectivity and catalytic activity in both enzymes, using whole-cell catalysis to screen mutants and determine enzymatic kinetic parameters. The results demonstrated that residues Ala-103 and Val-122 in CrTDC, along with their corresponding sites Thr-82 and Ile-101 in SsDDC, significantly influence substrate selectivity and catalytic efficiency. Molecular dynamics simulations revealed that substrate selectivity and catalytic efficiency depends on the nucleophilic attack distance between the substrate’s amino group and the C4′ of pyridoxal 5′-phosphate. This study elucidates the catalytic mechanisms and structural bases of TDC and DDC, guiding enhancements in the related aromatic monoamine biosynthesis.

Chirality is a ubiquitous phenomenon in nature. The advent of nanomaterials has led to a gradual evolution of chiral studies from the molecular scale to the nanoscale. The emergence of carbon dots (CDs) has inaugurated a novel domain in the scientific and technological realms of carbon nanomaterials. In recent years, CDs have attracted increasing attention due to their luminescent properties, excellent biocompatibility and remarkable bioactivity. However, little attention has been paid to the chirality of the CDs and, in particular, the differences between CDs synthesized from racemic compounds and other chiral CDs, as well as the differences in the biological effects of chiral CDs, remain to be explored. Here, chiral CDs were synthesized from L-/D-/DL-arginine, and the differences in various aspects of chiral CDs were evaluated. We found that L-CDs without extreme differences in structure had brighter fluorescence and a more significant ability of NO generation and lipid droplet inhibition than the other two chiral CDs. Combined with its better drug loading and release ability, we validated its superior efficacy in tumor treatment. Therefore, this study provides a basis for further research on chiral carbon dots and their differences.

The design of synthetic driving forces for biosynthetic pathway is crucial for directing carbon flux toward the target product. Optimizing cellular redox status is one of the key strategies for constructing microbial cell factories. In this study, we attempt to create a novel redox imbalance force-driven (RIFD) strategy to direct carbon flow toward the target synthetic pathway. Initially, we increased the NADPH pool through a strategy of “open source and reduce expenditure” employing four approaches to achieve excessive NADPH levels and growth inhibition: (I) the expression of cofactor-converting enzymes, (II) the expression of heterologous cofactor-dependent enzymes, (III) the expression of enzymes involved in the NADPH synthesis pathway, and (IV) reduced NADPH wastage by knocking down non-essential genes that consume NADPH in vivo. Next, multiple automated genome engineering (MAGE) techniques were employed to evolve redox-imbalanced engineered strains and drive the metabolic flux to L-threonine production. Finally, we developed a NADPH and L-threonine dual-sensing biosensor, combined it with Fluorescence-Activated Cell Sorting (FACS), and a high-yield (0.65 g/g) L-threonine-producing strain with a titer of 117.65 g L −1 was obtained. This research presents a general approach to increasing the production of cofactor-related products. Utilizing redox imbalance forces to drive metabolic flow toward the target product, it is possible to increase production while simultaneously restoring cell growth.

Syzygium jambos (L.) Alston fruits, a traditional medicinal species in the Myrtaceae family, are rich in polyphenolic compounds. This study optimized polyphenol extraction from S. jambos fruits using ultrasonic-assisted extraction combined with response surface methodology, achieving a total polyphenol content (TPC) of 31.24 mg/g. To further enrich polyphenols, crude extracts (CEs) were fractionated using solvents of increasing polarity (petroleum ether, dichloromethane, and ethyl acetate). Results showed that the ethyl acetate phase (EAP) had the highest TPC (96.25 mg/g, threefold higher TPC than CE) and strongest antioxidant activity. In addition, polyphenol extracts demonstrated excellent biocompatibility, significant tyrosinase inhibition, and antibacterial activity against Escherichia coli and Staphylococcus aureus , with inhibition zones of 16.2 ± 0.5 and 14.3 ± 0.6 mm at 100 mg/mL. Finally, 41 polyphenolic compounds were identified by liquid chromatography–mass spectrometry (LC–MS) in EAP, including flavonoids (quercetin, isorhamnetin, and rutin), phenolic acids (caffeic acid and ferulic acid), and tannins (corilagin). These results demonstrate the potential of S. jambos polyphenols as natural additives for antioxidant, skin-whitening, and antimicrobial applications. Graphical The study, for the first time, systematically explored the impact of different polarity solvents on the distribution of polyphenols and their antioxidant and skin-whitening activities in the fruits of Syzygium jambos . Through the integration of cellular experiments and zebrafish embryo studies, the biocompatibility was comprehensively assessed.

Phyllanthus emblica L . ( P. emblica L.) is a medicine and food homologous plant, whose fruit is acknowledged by the International Health Organization for its high nutritional value. However, eco-friendly and highly efficient methods for extracting the active components from P. emblica L. fruits are currently lacking. In this study, we explored the potential of an eco-friendly enzyme-assisted extraction method for enhancing the extraction of phenolic compounds from P. emblica L., then optimized the extraction process using response surface methodology. After optimization, the contents of gallic acid and total polyphenols reached 33.98 ± 0.15 mg/g and 147.92 ± 1.21 mg GAE/g, respectively. The optimized P. emblica L. extract showed promising anti-inflammatory, antioxidant, and hypoglycemic properties. These findings suggest a viable alternative for obtaining multifunctional extracts from P. emblica L., which have important applications in the functional food and pharmaceutical sectors.

The quality of yogurt deteriorates during storage due to exposure to oxygen and UV light, and high-barrier packaging effectively mitigates this issue. Concurrently, there is increasing interest in replacing petroleum-based packaging with eco-friendly alternatives. In this study, poly(lactic acid-butylene itaconate) (PLBI), developed as a modified version of poly (L-lactic acid) (PLLA) to enhance toughness, was combined with poly (vinyl alcohol) grafted with acryloyl chloride (PVA-g-AC) through UV curing. The resulting PLBI/PVA-g-AC films exhibited significantly improved barrier properties. They were used to package stirred yogurt alongside polyethylene/nylon (PE/NY) films for comparison. Quality assessments, including acidity, water-holding capacity, and viable bacterial counts, demonstrated that PLBI/PVA-g-AC films provided superior preservation. Metabolomics analysis revealed increased flavor compounds, such as acids and alcohols, and decreased sugar levels in these packaged yogurt. This work expands potential applications of PLLA-based materials in food packaging and offers insights into the influence of oxygen on yogurt storage quality.

Polymeric polyphenols in different parts (fruits, leaves and calyces) of persimmon ( Diospyros kaki L. cv. Mopan) were characterized via four different degradation methods, followed by HPLC/UPLC-UV/vis, HPLC/UPLC-Q-Exactive-Orbitrap/MS and UPLC-QqQ/MS to obtain comprehensive structural constituent unit information of polymeric polyphenols. The results indicated that persimmon polymeric polyphenols not only consisted of proanthocyanidins as terminal and extension units, but also contained anthocyanidins, flavonols and phenolic acids as terminal units. Among these different fragments detected, (−)-gallocatechin, (−)-gallocatechin-3- O -gallate, (+)-catechin and (+)-catechin-3 -O- gallate as extension units and (−)-epicatechin, (−)-gallocatechin, (−)-epigallocatechin, eight phenolic acids and four flavonols as terminal units were found for the first time in persimmon polymeric polyphenols. The molar percentages of constitutive units of persimmon polymeric polyphenols varied considerably. Compared to polymeric polyphenols in persimmon fruits and calyces, anthocyanidins, flavonols and phenolic acid units with the highest molar percentages were found in polymeric polyphenols in persimmon leaves. In contrast, the mean polymerization degrees of polymeric polyphenols in persimmon fruits, leaves and calyces were 26.45 ± 0.43, 6.23 ± 0.08 and 11.60 ± 0.07, respectively. These findings provide, for the first time, comprehensive structural information on persimmon polymeric polyphenols, and offer valuable insights into their composition and characteristics.
