In this work, core-satellite assemblies and exonuclease assisted double amplification strategy is developed to produce surface-enhanced Raman scattering (SERS) biosensor towards ultrasensitive detection of biotoxin. In the presence of target molecules, the exonuclease III (Exo III) assisted efficient recycling amplification provides an excellent pathway for the fabrication of core-satellite SERS sensor. Briefly, the proposed strategy includes the following double amplifications: (i) Exo III induced target-related signal amplification; (ii) core-satellite assemblies assisted formation of SERS “hot-spots” induced signal amplification. To show the applicability of the suggested strategy, the detection of ochratoxin A (OTA), one of the most toxic and widely distributed biotoxin, is demonstrated as an example. The results show that the limit of detection (LOD) of OTA is 0.83 fg mL −1 (S/N = 3). On the basis of the DNA aptamer induced specific target recognition, hence our sensing strategy is easy to be expended to the ultrasensitive detection of other targets, e.g., DNAs, RNAs, and other molecules that have corresponding DNA aptamers.

With the use of gold nanostructures, photothermal therapy (PTT) of cancer has great advantages compared to conventional methods, such as noninvasive targeted destruction and easily operation. Generally speaking, respective diagnosis and therapy of tumor require at least two instruments, leading to incongruence of tumor borders between diagnosis and therapy. To tackle this problem, tunable SERS-tags-hidden gold nanorattles (STHGNRs) have been designed and developed here for theranosis of cancer with single laser beam. The surface plasma resonance peak of STHGNRs can be tuned from visible region to near-infrared region by controlling the cavity size and shell thickness. The outer shells not only improve the stability of the SERS reporters but also enhance the brightness by more than two order magnitude compared to gold nanoparticles. In vitro study, immuno STHGNRs can serve as theranosis agents simultaneously for sensitive and efficient theranosis of cancer cells.

In response to the shortcomings of traditional surface-enhanced Raman spectroscopy (SERS) substrates, such as short shelf life, poor uniformity, and low selectivity, this study innovatively proposed a three-dimensional composite substrate of CNTs/Ag/AgNWs/SiO 2 . This substrate demonstrates excellent SERS enhancement effects, with a detection limit of 10 −12 mol/L for the probe molecule Rhodamine 6G (R6G) and an enhancement factor (EF) of 8.947 × 10 8 . Further experiments confirmed the substrate’s superior uniformity and stability. The enhancement mechanism was investigated using both experimental methods and the Finite Difference Time Domain (FDTD) approach. When commonly used pesticide thiram was used as the target analyte, the detection limit of the substrate reached 0.1 mg/L, which is significantly lower than the pesticide residue standards of China and the European Union. Additionally, the genetic algorithm (GA)-optimized Back Propagation (BP) neural network was introduced for the quantitative analysis of thiram concentrations. The experimental results indicated that the GA-BP algorithm achieved the training prediction accuracy of 92.5% for thiram, demonstrating good network performance. This method shows good selectivity and has broad application prospects in the detection of toxic chemicals, environmental pollutants, and food additives.

Herein, a high-performance Ta2O5/AgNPs composite Surface-Enhanced Raman Scattering (SERS) substrate is engineered for highly sensitive detection of microplastics. Through morphology modulation and band-gap engineering, the semiconductor Ta2O5is structured into spheres and composited with silver nanoparticles (AgNPs), facilitating efficient charge transfer and localized surface plasmon resonance (LSPR). This architecture integrates electromagnetic (EM) and chemical (CM) enhancement mechanisms, achieving an ultra-low detection limit of 10−13M for rhodamine 6G (R6G) with excellent linearity. Furthermore, the three-dimensional “pseudo-Neuston” network structure exhibits superior capture capability for microplastics (PS, PET, PMMA). To address spectral interference in simulated complex environments, a multi-scale deep-learning model combining wavelet transform, Convolutional Neural Networks (CNN), and Transformers is proposed. This model achieves a classification accuracy of 98.7% under high-noise conditions, significantly outperforming traditional machine learning methods. This work presents a robust strategy for environmental monitoring, offering a novel solution for precise risk assessment of microplastic pollution.

In recent years, biomaterials have increasingly attracted attention on surface-enhanced Raman spectroscopy (SERS) due to their well Raman performance while metal particles are combined with biological substrates. Therefore, we propose an environmentally friendly substrate based on silver-plated cicada wings with seamless graphene layer (Gr-AgNPs-C.w.), which can be prepared with a simple and inexpensive method. Compared with AgNPs-C.w., Gr-AgNPs-C.w. hybrids show better SERS performance with high sensitivity, good uniformity and good stability with R6G detection. The minimum detected concentration can reach 10−15 M, and the value of R2 can reach 0.996, respectively. Theoretical simulation demonstrates the situation of electromagnetic field through COMSOL software. In addition, due to the affinity of graphene for biomolecules, we can successfully detect the DNA biomolecules through a simple process. Therefore, this cheap and efficient natural SERS substrate has great potential for a considerable number of biochemical SERS applications and can broaden the way in which multiple SERS platforms derived from other natural materials are prepared.

Melanin plays an indispensable role in the human body. It serves as a biological reducer for the green synthesis of precious metal nanoparticles. Melanin–Ag nanocomposites were successfully produced which exhibited very strong surface-enhanced Raman scattering (SERS) effect because of the reducibility property of melanin. A melanin–Ag composite structure was synthesized in situ in melanin cells, and SERS technique was performed for the rapid imaging and quantitative assay of intracellular melanin. This imaging technique was also used to successfully trace the formation and secretion of intracellular melanin after stimulation with melanin-stimulating hormones. Based on the self-reducing property of melanin, the proposed SERS imaging method can provide potentially powerful analytical detection tools to study the biological functions of melanin and to prevent and cure melanin-related diseases.Keywords:surface-enhanced Raman scattering;melanin–Ag nanocomposites;bioimaging;melanogenesis

γ-Glutamyl transpeptidase (GGT) has attracted considerable attention for its regulatory effect on glutathione metabolism in living organisms; further, its close relationship with physiological dysfunctions such as hepatitis and liver cancers has enhanced its applicability. Therefore, the accurate detection of GGT levels is particularly important for the early diagnosis of diseases. Thus, we herein report the development of a surface-enhanced Raman spectroscopic (SERS) probe, namely bis-s,sʹ-(( s )-4,4ʹ-thiolphenylamide-Glu) (b-( s )-TPA-Glu), that comprises of a γ-glutamyl moiety for detection of the GGT activity. In this system, detection was achieved by observing differences in the SERS spectral profiles of the b-( s )-TPA-Glu probe and its corresponding hydrolysis product that resulted from the catalytic action of GGT. This SERS probe system exhibited a high selectivity toward GGT due to a combination of its specific catalytic action and the distinctive spectroscopic fingerprint of the SERS technique. The developed SERS approach was also found to be approximately linear in the range of 0.2–200 U/L, and a limit of detection of 0.09 U/L was determined. Furthermore, the proposed SERS method was suitable for detection of the GGT activity of clinical serum samples and also for evaluation of the inhibitors of GGT. Consequently, this approach is considered to be a promising diagnostic and drug screening tool for GGT-associated diseases.

AgNPs–CNP enabled NaOH-assisted SERS detection of salbutamol in pork. • AgNPs–CNP-10 gave the strongest response among the tested formulations. • Bands at 1603 and 1360 cm −1 supported preliminary dual-band calibration. • Response-based LOD and LOQ were 0.119 and 0.396 μg/g, respectively. • SAL/RAC mixtures showed composition-dependent spectral overlap. Salbutamol (SAL), a representative β-adrenergic agonist, poses potential risks to food safety when illegally present in animal-derived foods. In this study, silver nanoparticles prepared in the presence of carbon nanopowder (AgNPs–CNP) were examined for surface-enhanced Raman spectroscopy (SERS) measurement of SAL-containing extracts prepared from fortified pork. Among the tested preparations, AgNPs–CNP-10 produced the strongest SAL response, and a 0.9 mol/L NaOH stock solution produced the strongest response among the tested aggregation conditions. Across the recorded fortification series, the bands at 1603 and 1360 cm −1 showed level-dependent spectral changes. All droplets at a given recorded level originated from the same fortified and extracted pork portion, and the held-out droplets therefore described only droplet-level variation within the same extract series. Moreover, because the aliquot volume used in the original fortification step could not be reconstructed from the surviving records, the recorded level labels and derived regression statistics are not presented as traceable sample concentrations or method-validation parameters. An exploratory SAL/RAC coexistence experiment showed composition-dependent changes in adjacent spectral regions and progressively greater band overlap as the SAL:RAC ratio decreased. This work therefore provides a matrix-oriented exploratory description of the AgNPs–CNP/NaOH response. Verified fortification, independently prepared samples, recovery and trueness assessment, reference-method confirmation, broader selectivity testing, spatial and batch reproducibility, day-to-day evaluation, and storage-stability studies are required before quantitative or practical screening performance can be assessed. A noble-metal-free, aggregation-independent surface-enhanced Raman scattering (SERS) platform is reported based on a bifunctional α-MoO 3−x -CuCl 2 nanocomposite for the sensitive and selective detection of phosphate species. The defect-rich α-MoO 3−x provides strong SERS enhancement, and the introduction of CuCl 2 not only enhances its peroxidase like activity, but also endows it selective response to phosphate species. The composite α-MoO 3−x -CuCl 2 can efficient catalytic oxidation of colorless tetramethylbenzidine (TMB) to blue oxidized TMB (ox-TMB), which yields a strong SERS signal. At the assay pH of 4.0, the protonated phosphate species (mainly H 2 PO 4 ⁻) selectively coordinate with Cu 2+ , suppressing the nanozyme activity and causing a dose-dependent decrease in both colorimetric and SERS responses. Under optimized conditions, the method exhibits a linear detection range from 8.0 × 10 − 9 M to 8.0 × 10 − 5 M and a limit of detection as low as 2.0 × 10 − 9 M. The platform also demonstrates good anti-interference capability against common coexisting ions and satisfactory recoveries (103.75%–107.5%) in spiked lake water and tap water samples. This work provides a low-cost, reliable, and reproducible strategy for indirect SERS detection of phosphate species without reliance on noble metals or nanoparticle aggregation. Graphical abstract

Residual pesticides in agricultural production are one of the major food safety concerns around the world. In this study, we investigated the amount of isofenphos-methyl in corns by using surface-enhanced Raman spectroscopy (SERS) coupled with chemometric methods. The highest isofenphos-methyl peak at 1043 cm −1 was used to detect and quantify the amount of isofenphos-methyl spiked in methanol-water (1:1) solvent and corns. The limits of detection (LODs) of isofenphos-methyl in methanol-water solvent and corns were 0.01 μg/mL and 0.01 μg/g, respectively. Quantitative detection of isofenphos-methyl residues (0–5 μg/mL) was conducted using partial least squares (PLS) model. Based upon the results of leave-one-out cross-validation, isofenphos-methyl could be predicted by PLS (the correlation coefficient was 0.9964) with a low root mean square error of calibration (RMSEC=0.146). The developed SERS method provides a much simpler and more sensitive way to monitor isofenphos-methyl for safe production of corns, which can be extended to the other agricultural products.

Hollow urchin-like Au nanocrystals have been widely studied due to their excellent surface plasmon resonance properties and large specific surface area, but the controllable preparation of hollow urchin-like Au nanocrystals is still a challenge. In this article, we successfully prepared hollow urchin-like Au nanocrystals using HAuCl4·3H2O and AgNO3as precursors and ascorbic acid as the reducing agent. No surface ligands or polymer stabilizers are required in the preparation process. HAuCl4·3H2O and AgNO3will first form AgCl cubes, then the reducing agent, ascorbic acid, will reduce the Au3+in the solution to Au0, and Au0will be deposited on the pre-formed AgCl cubes to form AgCl@Au nanocrystals. We characterized the morphology of the prepared Au nanocrystals by scanning electron microscopy and found that by increasing the amount of HAuCl4·3H2O in the reaction, the surface morphology of the Au nanocrystals would change from a rough spherical shape to an urchin-like shape. By further increasing the amount of the precursor HAuCl4·3H2O, urchin-like Au will convert into flake-like morphology. The AgCl in the interior was removed with ammonia water, and finally, hollow urchin-like Au crystals were formed. In addition, we used R6G molecule to explore the surface-enhanced Raman spectroscopy (SERS) enhancement effect of prepared Au crystals. The results show that the minimum detectable concentration of R6G reaches 10−8M. Moreover, we applied hollow urchin-like Au nanocrystals as catalysts and SERS enhancing materials to detect the photocatalytic reaction of 4-NTP. We used a 785 nm laser as both the SERS light source and the catalytic light source to monitor the photocatalytic effect of the laser on 4-NTP in situ by adjusting the laser power. Keywords:Au nanocrystals;sea urchin-like structure;SERS;photocatalytic reaction

Silica-coated SERS tags have been attracting greater attention in recent years. However, the reported methods to synthesize these tags are tedious and often subjected to the limited signal intensity. Here, we report a facile and general method to prepare the silica-coated Ag SERS tags with the enhanced signal intensity by no introducing the primers. This approach mainly depends on the colloidal stability of the Ag NPs in alcohol solution. By decreasing the concentration of salt in Ag NP solution, the citrate-stabilized Ag NPs can be well dispersed in alcohol solution. Based on this, the Ag SERS tags can be directly coated with thickness-controlled and homogeneous silica shells. This approach is highly reproducible for silica shell growth and signal intensity, not depending on the properties of Raman molecules, proved by 7 kinds of the Raman molecules. Moreover, this kind of SERS tags coated with silica hold the stronger SERS signals than the traditional method due to no interference from the priming molecules.
