Heavy metal–organic complexes widely distributed in wastewater are hazardous and urgently need to be removed and recycled. Herein, copper glycinate (CuGly) was employed as a model complex and removed with the environmentally friendly oxidant ferrate(VI) (Fe(VI)). We found that Fe(VI) can be used to effectively remove CuGly and recycle Cu via a decomplexation–degradation–immobilization coupled pathway, i.e., decomplexing CuGly, degrading organic ligands, and immobilizing the released Cu ions. Theoretical calculation results revealed that CuGly was favored to be oxidized by Fe(VI), and the CuGly decomplexation and organic ligand degradation could spontaneously thermodynamically proceed at room temperature. Although different active oxidation species, including Fe(V), Fe(IV), H2O2, and •OH, are present during the Fe(VI) decomposition process, control experiments suggested that Fe(VI), Fe(V), and Fe(IV), rather than H2O2 and •OH, played critical roles in CuGly removal. The resultant Fe and Cu precipitate could be further converted to a magnetic composite material, which could be used as a catalyst for antibiotic degradation. This study presents an effective strategy and the comprehensive underlying mechanisms of synergistic heavy metal–organic complex removal and metal resource recycling using ferrate, which could be extended to green chemical treatment of heavy metal–organic complexes in various wastewater bodies.

The conventional environmental remediation materials, which are applied for the long-term treatment of mine tailings are easy to fail when affected by acid rain. Therefore, dynamic column simulation experiments were conducted to compare the immobilization persistence of As, Mn, Pb and Zn by the addition of triethylenetetramine modified montmorillonite (TTA-Mt) and calcium-based montmorillonite (Ca-Mt) in sulfide tailings during a simulated 5-year study of acid rain leaching. The structural and morphological characteristics of TTA-Mt were analyzed with FTIR, XRD, SEM, TEM and EDS. Results showed that the additions of TTA-Mt could persistently immobilize the heavy metal ions while Ca-Mt additions had little effect. After long-term simulated leaching for 5 years, TTA-Mt still had superior environmental stability for Zn and As with the immobilization rates of 96% and 86%, and the leaching amounts of Mn and Pb were less than 4.23 and 0.66 mg/kg, respectively. Morphology and chemical fraction analyses confirmed that the immobilization was mainly due to the formation of relatively stable phases by complexation and precipitation. Results presented here demonstrated the significant potential of TTA-Mt for long-term immobilization of heavy metals in tailings against acid rain and thus could provide guidance for remediation of tailings.

Senescence is a major inductive factor of aging-related diseases in connection with an accumulation of reactive oxygen species (ROS). Therefore, it is important to maintain ROS at an appropriate level to keep homeostasis in organisms. Superoxide dismutase (SOD) is a vital enzyme in defending against oxidative damage in vivo. Because of the defects in the direct application of SOD and SOD mimics, mounting delivery systems have been developed for the efficient applications of SOD to realize antioxidant treatment. Among these systems, mesoporous silica nanoparticles (MSNs) have been widely studied because of various advantages such as desirable stability, low toxicity, and adjustable particle sizes. Herein, SOD was immobilized on MSNs using a physical absorption strategy to construct the nanosystem SOD@MSN. The nematode Caenorhabditis elegans (C. elegans) was selected as the model organism for the subsequent antioxidant and anti-aging studies. The research results suggested the nanosystem could not only be effectively internalized by C. elegans but could also protect the nematode against external stress, thus extending the lifespan and healthspan of C. elegans. Therefore, SOD@MSN could be applied as a promising medicine in anti-aging therapeutics.

The contamination of soils by mercury (Hg) seriously threatens the local ecological environment and public health. S-functionalized amendments are common remediation technology , yet, Hg re-activation often occurs in the commonly used immobilization remediation by S-functionalized amendments, resulting in an unsatisfactory remediation effect. In this study, a novel FeS–Se functionalized biochar composite (FeS-Se-BC) amendment was prepared and applied for the efficient remediation of Hg-polluted soil. An immobilization efficiency of 99.62% and 99.22% for H 2 O-extractable Hg and TCLP solution-extractable Hg was achieved with the application of FeS-Se-BC (0.05) after 180 d. The analyses of XPS, Hg-TPD, SEM-EDS demonstrated that excellent remediation performance by FeS-Se-BC resulted from the synergistic effect of FeS and Se to form HgS and HgSe concurrently. In comparison to the treatments of biochar and FeS-functionalized biochar (FeS-BC), FeS-Se-BC promoted the transformation of exchangeable, carbonate-bound, and Fe–Mn oxides-bound Hg fractions into organic material-bound, and residual fractions, effectively reducing the risk of Hg-contaminated soil from a highly dangerous level to a low risk. Furthermore, the introduction of Se clearly inhibited the re-activation of Hg and reduced the release of Hg by 81.12% compared to FeS-BC when the ratio of S 2− to Hg was 5: 1 due to the formation of extremely stable HgSe. These results suggest that FeS-Se-BC has good potential for remediation of Hg-polluted soils which provides a new inhibitory idea for Hg re-activation after immobilization.

In recent years, some studies have found that oriented immobilization of antibodies to microspheres can fully expose the antigen binding sites of antibodies, which can improve the sensitivity of sandwich immunoassays for the detection of proteins. Can this antibody immobilization strategy also improve the sensitivity of competitive immunoassays for the detection of small molecules ? To answer this question, the conjugate MS-SPG-Ab (oriented immobilization of aflatoxin B1 antibody to time-resolved fluorescent microspheres via streptococcal protein G) and the conjugate MS-Ab (nonoriented immobilization of aflatoxin B1 antibody to time-resolved fluorescent microspheres) were prepared, and a lateral flow immunoassay (LFIA) for the detection of aflatoxin B1 (AFB1) was established. The detection performance of the two methods was compared. The results showed that under the condition that the number of “effective” antibodies immobilized on TRF-MS was similar, compared with the nonoriented immobilization strategy (IC50 = 0.21 ng mL −1 ), the LFIA method established by the oriented immobilization strategy reduced the sensitivity of AFB1 detection (IC50 = 0.37 ng mL −1 ). However, this method can obtain higher detection precision for AFB1, the CV values were all below 8%. And it has stronger tolerance to the matrix of maize and peanut samples. The bias of LFIAs based on oriented immobilization technology (−14.93%–7.92%) was lower than nonoriented immobilization technology (28.16%–34.19%) for AFB1 detection in the two sample extracts. This study suggests that the LFIA method based on the oriented immobilization of antibodies can improve the accuracy of the detection results when performing rapid screening of small molecules.

Here, an aptamer-based SERS method for the detection of Staphylococcus aureus ( S. aureus ) without immobilization using [email protected] NPs/slide as enhanced substrate was constructed. ROX-aptamer of S. aureus was modified on the surface of [email protected] NPs/slide through electrostatic interaction. Based on the specific binding effect of the aptamer to S. aureus , the ROX-aptamer fell off from the surface of the substrate, resulting in a decrease of the SERS signal intensity of the substrate. Under the optimal experimental conditions, a good linear relationship was found between SERS intensity at 1500 cm −1 and the logarithm of concentration of S. aureus in the range of 10 2 cfu/mL-10 7 cfu/mL (y = 6623-796lgx, R 2 = 0.994) with a detection limit of 6 cfu/mL. The selectivity analysis revealed that the method had higher selectivity toward the corresponding target. The results for milk sample using the developed SERS method for the detection of S. aureus were similar to those of the plate counting method. The recovery ratio was from 90.60% to 107.26%, indicating the accuracy and reliability of the developed method. This method eliminates the need for bacterial immobilization and improves the convenience and efficiency of detection.

Metal-organic frameworks (MOFs) are excellent platforms for enzyme immobilization. However, strong or weak interactions between enzymes and MOFs often lead to inactivation and enzyme leaching. Herein, single-stranded DNA (ssDNA) with Janus properties was used as a linker between enzymes and the dual carriers of MOFs and graphene oxide (GO), which provided a simple and effective platform to tightly co-immobilize glucose oxidase (GO X ) and horseradish peroxidase (HRP). Mechanistic investigation revealed that the enzyme was immobilized mainly through π-π stacking between nucleobases and GO and coordination of the sugar-phosphate backbone with zirconium clusters in MOFs. This strategy ameliorated traditional surface adsorption issues for enzyme immobilization on MOFs. By introducing GO and ssDNA, the GO X &HRP cascade reaction on MOFs was enhanced significantly, showing better kinetics than the corresponding free GO X &HRP. Furthermore, the immobilized GO X &HRP exhibited substrate selectivity and a wide linear range for glucose detection. This enzyme immobilization strategy should have broad application prospects and practical value in biocatalysis and diagnostics.

Developing a functional affinity monolithic column towards in-tube solid-phase microextraction (IT-SPME) for selective sample pretreatment is critical. Herein, a high-performance capillary affinity monolithic column with an ultra-high aptamer coverage density was rapidly fabricated via a simple adsorption strategy, in which aptamers with natural sequences were directly immobilized on an ammonium-based strongly cationic matrix. Limitations of the traditional biological or covalent methods such as time-consuming modification reactions, special requirement of active groups (e.g. –NH2 and –SH) on the aptamer, and low aptamer coverage density levels were avoided. An ultra-high coverage density of 8616 pmol μL−1 was achieved with excellent stability, and the highest aptamer-modification level among all the current methods was reached. Selective recognition and high recovery yields of the model mycotoxin ochratoxin A (OTA) were achieved in 95.9 ± 0.98%–97.9 ± 0.28% (n = 3). In particular, there was little cross-reactivity towards the OTB analogue of only 0.5% even in the case of 250 fold of the analogue OTB, which was not reported in previous affinity monoliths. Upon sample analysis, satisfactory discriminations of trace OTA were obtained at 93.7 ± 1.4%–95.5 ± 2.5% (n = 3) in beer and wheat. A facile and direct method for efficiently fabricating an aptamer-based affinity monolith towards online selective IT-SPME was proposed.

Functional carrier capable of covalently binding proteins from crude lysate under mild condition has become increasingly important in bioprocess. In this paper, a magnetic microbead that can covalently immobilize specific proteins directly from crude protein solution was developed, and its application conditions and stability were evaluated. The SpyCatcher was firstly modified with N-terminal cysteine by genetic methods and then the cys-SpyCatcher was covalently immobilized on epoxy-modified magnetic beads by epoxy-thiol reaction. The reaction between the epoxy and the only terminal thiol group in cys-SpyCatcher ensured the same orientation of the cys-SpyCatcher on the particle surface, while the maximum binding capacity of cys-SpyCatcher reached 23.6 μg/mg under the condition of 250 mM Na 2 SO 4 , pH 8.5 at initial cys-SpyCatcher concentration of 3 mg/mL. The functional magnetic microsphere ( [email protected] ) showed covalent site-specific capture property for enhanced green fluorescent protein fused with SpyTag (SpyTag-EGFP). A maximum binding capacity of (25 μg SpyTag-EGFP)/(mg beads) was far more than that of the previous carrier with SpyCatcher. In addition, the [email protected] could specifically capture the SpyTagged protein directly from the cell lysate, and this carrier showed good stability at pH 4–9 and temperature of 30–70ºC. Therefore, as a universal covalent capture carrier, it is believed that the [email protected] is useful in one-step purification and site-specific immobilization of the SpyTagged protein in bioprocess or biomedical application.

In this work, TiO 2 nano particle was used for two purposes: it played as a role of a solid carrier for immobilization of HRP, and meanwhile acted as a photo-catalyst itself. In this way, the loss of enzymatic activity during enzyme immobilization can be compensated by the coupling of TiO 2 photo-catalysis. Then, the surface of TiO 2 nanoparticles were modified by phase-transitioned lysozyme (PTL) and connected with horseradish peroxidase (HRP) via a DNA helix ( [email protected] 2 ). The DNA double helix chain was utilized as a flexible spacer arm to keep the flexibility of the immobilized enzyme. The [email protected] 2 showed a high recovered activity (90.52%) of the original activity of free HRP. This data is better than most reported activity recovery of immobilized enzyme. Moreover, [email protected] 2 can maintain high catalytic activity, stability and reusability when degrading phenol in wastewater. The phenol of 0.5 mmol∙L −1 could be totally degraded within 10 min. Moreover, a visual colorimetric method for phenol concentration by [email protected] 2 was established, and the phenol concentration of 0–0.2 mmol∙L −1 can be visually detected with naked eyes. Furthermore, an enzymatic-photocatalytic synergetic effect of [email protected] 2 is found and investigated. The efficiency of [email protected] 2 ensures it a potential application in the wastewater treatment.

Aiming to a durably antibacterial function, stable immobilization of silver nanoparticles (Ag NPs) on cotton textiles still represents challenging in textile industry. In the present work, by grafting oligomeric polyacrylic acid (PAA) onto cotton fiber surfaces, we effectively prepared the Ag NPs with controllable size on the fiber surface, which are of average size of 16.43 nm, and therefore achieved a desirable antibacterial durability. The oligomer PAA binder can give the modified fabrics with a remarkable antibacterial durability against washing, as the bacterial reduction rates against both Staphylococcus aureus and Escherichia coli remained over 90 % even after 50 laundering cycles. Moreover, this graft to approach using oligomeric PAA shows merits in maintaining the desired cotton properties such as flexibility, water absorptivity and vapor permeability. Finally, this finishing process ensures a reliable safety for human skin.

Uniform poly(ester-amino) microspheres (PEAs) were prepared by catalyst-free aza-Michael addition precipitation polymerization of trihydroxymethylpropyl triacrylate (TMPTA) and piperazine (PA) in acetonitrile. The effects of monomer feed ratio and concentration, polymerization temperature, and time on the size and morphology of PEAs were investigated. Monodisperse PEAs with size about 4 μm were successfully obtained at 2/3 molar ratio of TMPTA/PA and 4°C. The chemical structure of PEAs was characterized. There were amount of unreacted acrylate groups remained on the surface of PEAs prepared at the imino/acrylate molar ratio of 1/1.1, which provide lots of active sites for further chemical modification, especially for bioactivator immobilization. Laccase was used as an example to determine bio-immobilization activity of PEAs. Immobilized laccase exhibited higher tolerance to pH and temperature. The immobilized laccase was used as biocatalyst for degradation of Malachite Green. About 53% of its initial catalytic activity was retained after 4 cycles of reuse.
