Inhibition of the double-edged effect of curcumin on Maillard reaction in a milk model system by a nanocapsule strategy

Yingnan Liu,Shaoxuan Yu,Lizhi Liu,Xiaoyue Yue,Wentao Zhang,Qingfeng Yang,Li Wang,Yanru Wang,Daohong Zhang,Jianlong Wang

In this work, the double-edged effect of curcumin (Cur) on Maillard reaction (MR) in model milk pasteurization process was demonstrated for the first time, where Cur inhibits MR at low concentrations (<5 μg/mL), while promotes it at high concentrations (>5 μg/mL). A nanocapsule strategy was introduced to resolve the disadvantages of Cur on MR at high temperature. In vitro experiments confirmed that curcumin @ polyvinylpyrrolidone nanocapsules (Cur@PVP NCs) synthesized by a one-pot method can control the continuous and slow release of Cur and exert inhibitory effect on MR by competing with lactose for casein, trapping the reactive dicarbonyl compounds and scavenging ROS, which might provide a new strategy for the application of functional food ingredients in the control of MR during food heat processing , including milk pasteurization.

pH-Responsive reversibly cross-linked micelles by phenol–yne click via curcumin as a drug delivery system in cancer chemotherapy

Yuancheng Liu,Fan Chen,Kui Zhang,Quan Wang,Yuanwei Chen,Xianglin Luo

Conventional cross-linked micelles may suffer from low drug loading efficiency, side effects or toxicity of residual cross-linkers and complicated preparation processes of drug-loaded micelles. Herein, pH-sensitive reversibly cross-linked micelles by phenol–yne click via curcumin (Cur) were developed by combining drug loading and cross-linking using poly(ethylene glycol)-b-poly(2-methacrylate ethyl 5-hexynoicate), (mPEG-b-PHEMA-5HA), as a drug delivery system. mPEG-b-PHEMA-5HA was synthesized by ATRP of 2-hydroxyethyl methacrylate (HEMA) using mPEG-Br as a macroinitiator and then by esterification with 5-hexynoic acid. mPEG-b-PHEMA-5HA could form micelles and the micelles could load Cur. Meanwhile, the Cur loaded micelles were readily cross-linked by a phenol–yne click reaction. The cross-linked micelles showed excellent stability after high dilution with water, and turned into smaller micelles under an acid environment (pH 5.0). The cross-linked micelles achieved a higher drug loading of Cur (17.81%, wt%) than the noncross-linked micelles, due to physical entrapment and chemical click reaction. Cur release from the Cur-cross-linked micelles was inhibited under physiological conditions (pH 7.4) but was faster at pH 5.0 in comparison with the noncross-linked micelles. The cross-linked micelles exhibited a superior antitumor activity with a lower IC50 of 4.86 and 9.6 μg mL−1 toward HeLa and 4T1 cells, respectively, resulting from effective internalization by tumor cells. In pharmacokinetics, the cross-linked micelles showed a prolonged circulation time, and thus exhibited more effective inhibition of tumor growth compared to free Cur and Cur loaded noncross-linked micelles in in vivo experiments. Therefore, pH-sensitive reversibly cross-linked micelles via Cur are promising as an antitumor drug delivery system.

A Co-Delivery System of Curcumin and p53 for Enhancing the Sensitivity of Drug-Resistant Ovarian Cancer Cells to Cisplatin

Xinli Guo,Zhou Fang,Min Zhang,Deyu Yang,Shuyue Wang,Kehai Liu

In order to enhance the sensitivity of drug-resistant ovarian cancer cells to cisplatin (DDP), a co-delivery system was designed for simultaneous delivery of curcumin (CUR) and p53 DNA. Firstly, the bifunctional peptide K14 composed of tumor targeting peptide (tLyP-1) and nuclear localization signal (NLS) was synthesized. A nonviral carrier (PEI-K14) was synthesized by cross-linking low molecular weight polyethyleneimine (PEI) with K14. Then, CUR was coupled to PEI-K14 by matrix metalloproteinase 9 (MMP9)-cleavable peptide to prepare CUR-PEI-K14. A co-delivery system, named CUR-PEI-K14/p53, was obtained by CUR-PEI-K14 and p53 self-assembly. Furthermore, the physicochemical properties and gene transfection efficiency were evaluated. Finally, ovarian cancer cisplatin-resistant (SKOV3-DDP) cells were selected to evaluate the effect of CUR-PEI-K14/p53 on enhancing the sensitivity of drug-resistant cells to DDP. The CUR-PEI-K14/DNA complexes appeared uniformly dispersed and spherical. The particle size was around 20–150 nm and the zeta potential was around 18–37 mV. It had good stability, high transfection efficiency, and low cytotoxicity. CUR-PEI-K14/p53 could significantly increase the sensitivity of SKOV3-DDP cells to DDP, and this effect was better as combined with DDP. The sensitizing effect might be related to the upregulation of p53 messenger RNA (mRNA), the downregulation of P-glycoprotein (P-gp) mRNA, and the upregulation of BCL2-Associated X (bax) mRNA. CUR-PEI-K14/p53 can be used as an effective strategy to enhance the sensitivity of drug-resistant ovarian cancer cells to DDP.Keywords:ovarian cancer;drug resistance;polyethyleneimine;curcumin;p53;co-delivery system

Novel T7-Modified pH-Responsive Targeted Nanosystem for Co-Delivery of Docetaxel and Curcumin in the Treatment of Esophageal Cancer

Lian Deng,Xiongjie Zhu,Zhongjian Yu,Ying Li,Lingyu Qin,Zhile Liu,Longbao Feng,Rui Guo,Yanfang Zheng

Background Although single-drug chemotherapy is still an effective treatment for esophageal cancer, its long-term application is limited by severe side-effects, poor bioavailability, and drug-resistance. Increasing attention has been paid to nanomedicines because of their good biological safety, targeting capabilities, and high-efficiency loading of multiple drugs. Herein, we have developed a novel T7 peptide-modified pH-responsive targeting nanosystem co-loaded with docetaxel and curcumin for the treatment of esophageal cancer. Methods Firstly, CM-β-CD-PEI-PEG-T7/DTX/CUR (T7-NP-DC) was synthesized by the double emulsion (W/O/W) method. The targeting capacity of the nanocarrier was then investigated by in vitro and in vivo assays using targeted (T7-NP) and non-targeted nanoparticles (NP). Furthermore, the anti-tumor efficacy of T7-NP-DC was studied using esophageal cancer cells (KYSE150 and KYSE510) and a KYSE150 xenograft tumor model. Results T7-NP-DC was synthesized successfully and its diameter was determined to be about 100 nm by transmission electron microscopy and dynamic light scattering. T7-NP-DC with docetaxel and curcumin loading of 10% and 6.1%, respectively, had good colloidal stability and exhibited pH-responsive drug release. Good biosafety was observed, even when the concentration was as high as 800 μg/mL. Significant enhancement of T7-NP uptake was observed 6 hours after intravenous injection compared with NP. In addition, the therapeutic efficacy of T7-NP-DC was better than NP-DC and docetaxel in terms of growth suppression in the KYSE150 esophageal cancer model. Conclusion The findings demonstrated that T7-NP-DC is a promising, non-toxic, and controllable nanoparticle that is capable of simultaneous delivery of the chemotherapy drug, docetaxel, and the Chinese Medicine, curcumin, for treatment of esophageal cancer. This novel T7-modified targeting nanosystem releases loaded drugs when exposed to the acidic microenvironment of the tumor and exerts a synergistic anti-tumor effect. The data indicate that the nanomaterials can safely exert synergistic anti-tumor effects and provide an excellent therapeutic platform for combination therapy of esophageal cancer.

Enhanced In Vitro and In Vivo Anticancer Properties by Using a Nanocarrier for Co-Delivery of Antitumor Polypeptide and Curcumin

Qi Mengjiao,Zou Shaohua,Guo Chunjing,Wang Kaili,Yu Yueming,Zhao Feng,Fan Huaying,Wu Zimei,Liu Wanhui,Chen Daquan

In this paper, a novel pH and redox dual-sensitive nanocarrier loaded with curcumin (Cur) and anticancer polypeptide (AP) was developed for dual targeting mitochondrial and CD44 receptor. The amphiphilic block copolymer was prepared by triphenylphosphonium (TPP)/oligomeric hyaluronic acid (oHA)/disulfide-menthone 1,2-glycerol ketal (SM), hereinafter referred to as TPP-oHSM. The TPP targeted the mitochondria, pH/redox dual-sensitive SM served as a hydrophobic part, and the CD44 receptor targeting oHA worked as a hydrophilic part. The chemical structure of the TPP-oHSM was identified using 1H NMR and FTIR technologies. Cur and AP were loaded into the TPP-oHSM micelles by self-assembly and denoted as C/A@TM. The C/A@TM prepared in this study exhibited an approximately spherical structure, with a mean diameter of 191.3 ± 3.1 nm and a negative zeta potential of –26.10 ± 0.45 mV. The in vitro release study and cellular uptake test revealed that the C/A@TM targeted the mitochondria and CD44 receptor, as well as it showed sensitivity towards pH and redox. In addition, the C/A@TM demonstrated satisfactory cytotoxic effects against MDA-MB-231 cells and MCF-7 cells. Finally, the in vivo application of the C/A@TM showed excellent therapeutic effects. The C/A@TM developed in this study exhibited promising multifunctional properties as a co-delivery carrier of polypeptide and chemical drug for an effective clinical therapy for cancer.

Maillard-Reacted Whey Protein Isolates and Epigallocatechin Gallate Complex Enhance the Thermal Stability of the Pickering Emulsion Delivery of Curcumin

Gang Liu,Qi Wang,Zhongze Hu,Jie Cai,Xinguang Qin

In this study, whey protein isolates (WPI), lactose (Lac) Maillard-reacted products, and epigallocatechin gallate (EGCG) complex were used to enhance the thermal stability of the Pickering emulsion delivery of curcumin. Atomic force microscopy combined with Fourier transform infrared spectroscopy were employed to study the morphological characteristics and structural changes of WPI-Lac/EGCG nanoparticles. Results proved that WPI-Lac and EGCG were combined by hydrogen bonding and hydrophobic interaction. The mechanism underlying WPI-Lac/EGCG-stabilized Pickering emulsion was further characterized by confocal laser scanning microscopy. The optimal binding ratio of WPI-Lac to EGCG was 1:1 (w/w) at pH 3.0. The particle size and zeta potential of the WPI-Lac/EGCG nanoparticles were about 110 nm and 27 mV, respectively. Analysis of microstructure and droplet size distribution revealed that the glycated WPI-Lac/EGCG-stabilized emulsions exhibited more uniform droplet distribution, stronger thermal stability, and higher curcumin percentage retention than WPI. These results indicate that the WPI-Lac/EGCG nanoparticles are potential stabilizers for Pickering emulsion requirements. This study provides a basis for the construction of Pickering emulsion systems while carrying pro-/hydrophobic bioactive components.

Development of pH-responsive antioxidant soy protein isolate films incorporated with cellulose nanocrystals and curcumin nanocapsules to monitor shrimp freshness

Yaqing Xiao,Yingnan Liu,Shufang Kang,Mengdi Cui,Huaide Xu

Spherical curcumin/polyvinylpyrrolidone nanocapsules (CurNC) with the improved water solubility and thermal stability than native curcumin (Cur) were synthesized. Subsequently, soy protein isolate-based pH-sensitive multifunctional packaging films reinforced with cellulose nanocrystals (CNC), CurNC, CNC/CurNC, and CNC/Cur were developed for shrimp freshness detection. It was found that adding the CNC-containing agents improved the tensile strength, hydrogen bond interactions, thermal stability, and crystalline structure of the film and decreased its water solubility and water vapor and oxygen permeability. The Cur-containing agents decreased the L* (lightness/darkness) and a* (redness/greenness) values and increased the b* (yellowness/blueness) and ΔE (total color difference) values of the film. A more dense and uniform matrix of the nanocomposite films was likely responsible for delaying the release of Cur from the film matrix. Interestingly, CNC/CurNC film displayed higher antiradical scavenging activity than CNC/Cur film, as well as remarkable color responsiveness to pH changes and NH 3 presence. Moreover, the CNC/CurNC film was able to decrease the total volatile basic nitrogen of stored shrimp and visually monitor shrimp freshness in real-time.

Oral delivery of curcumin via multi-bioresponsive polyvinyl alcohol and guar gum based double-membrane microgels for ulcerative colitis therapy

Yan Hu,Shangwen Zhang,Zhijie Wen,Hudie Fu,Jie Hu,Xuexin Ye,Li Kang,Xiaojun Li,Xinzhou Yang

Anti-inflammatory drugs for ulcerative colitis (UC) treatment should specifically penetrate and accumulate in the colon tissue. Herein, a multi-bioresponsive anti-inflammatory drug (curcumin, CUR)-loaded heterogeneous double-membrane microgels ( [email protected] ) for oral administration was fabricated in this study, in which the inner core was derived from polyvinyl alcohol (PVA) and guar gum (GG) and the outer gel was decoration with alginate and chitosan by polyelectrolyte interactions. The structure and morphology of microgels were characterized. In vitro, the formulation exhibited good bio-responses at different pH conditions and sustained-release properties in simulated colon fluid with a drug-release rate of 84.6 % over 34 h. With the assistance of the outlayer gels, the microgels effectively delayed the premature drug release of CUR in the upper gastrointestinal tract. In vivo studies revealed that [email protected] specifically accumulated in the colon tissue for 24 h, which suggest that the interlayer gels were apt to reach colon lesion. As expected, the oral administration of microgels remarkably alleviated the symptoms of UC and protected the colon tissue in DSS-induced UC mice. The above results indicated that these facilely fabricated microgels which exhibited excellent biocompatibility and multi-bioresponsive drug release, had an apparent effect on the treatment of UC, which represents a promising drug delivery strategy for CUR in a clinical application.

Curcumin Alleviates Oxygen-Glucose-Deprivation/Reperfusion-Induced Oxidative Damage by Regulating miR-1287-5p/LONP2 Axis in SH-SY5Y Cells

Teng Zhang,Xiaomin Chen,Yueqing Qu,Yanbing Ding

Oxidative stress-induced neuronal damage is a main cause of ischemia/reperfusion injury. Curcumin (Cur), the principal constituent extracted from dried rhizomes of Curcuma longa L. (turmeric), exhibits excellent antioxidant effects. Previous studies have indicated that miR-1287-5p was downregulated in patients with ischemic stroke. Additionally, we predicted that Lon Peptidase 2, Peroxisomal (LONP2), which is involved in oxidative stress regulation, is targeted by miR-1287-5p. The aim of the current study is to investigate the effect of Cur on ischemia/reperfusion damage and its underlying mechanism. To mimic ischemia/reperfusion damage environment, SH-SY5Y cells were subjected to oxygen-glucose-deprivation/reperfusion (OGD/R). OGD/R treatment downregulated miR-1287-5p and upregulated LONP2 in SH-SY5Y cells, but Cur alleviated OGD/R-induced oxidative damage and reversed the effect of OGD/R on the expression of miR-1287-5p and LONP2. Furthermore, we confirmed the interactive relationship between miR-1287-5p and LONP2 (negative regulation). We revealed that miR-1287-5p overexpression alleviated OGD/R-induced oxidative damage alleviation, similar to the effect of Cur. MiR-1287-5p inhibition accentuated OGD/R-induced oxidative damage in SH-SY5Y cells, which was reversed by Cur. The expression of LONP2 in OGD/R-treated SH-SY5Y cells was decreased by miR-1287-5p overexpression and increased by miR-1287-5p inhibition, and Cur counteracted the increase in LONP2 expression induced by miR-1287-5p inhibition. In conclusion, we suggest that Cur alleviates OGD/R-induced oxidative damage in SH-SY5Y cells by regulating the miR-1287-5p/LONP2 axis. The findings provide a theoretical basis for the clinical application of curcumin.

Anti-inflammatory activity of curcumin-loaded solid lipid nanoparticles in IL-1β transgenic mice subjected to the lipopolysaccharide-induced sepsis

Jiao Wang,Haoyue Wang,Rongrong Zhu,Qiang Liu,Jian Fei,Shilong Wang

Sepsis is a significant public healthcare problem, affecting millions of people worldwide each year, killing one in four, and increasing in incidence. Thus, advanced therapeutic strategies are required to treat sepsis patients. Curcumin (Cur) is a promising anti-inflammatory agent for various inflammatory disorders. However, the therapeutic efficacy of Cur is limited due to poor aqueous solubility, rapid degradation, and low bioavailability. The aims of this study were to evaluate the therapeutic potential of Cur-loaded solid lipid nanoparticles (Cur-SLNs) for sepsis treatment. A firefly luciferase transgenic mouse was used to monitor real time interleukin 1β (IL-1β) expression in lipopolysaccharide (LPS)-induced sepsis model to examine the protective effect of Cur-SLNs, and to elucidate its underlying molecular mechanisms. Mice (female or male) were intraperitoneally administered with free Cur or Cur-SLNs (30 mg/kg) before the intraperitoneal delivery of LPS (3 mg/kg). Our results indicated that Cur-SLNs can effectively reduced levels of IL-1β expression compared to free Cur, especially at 3 h after LPS injection. Also, Cur-SLNs significantly decreased the expression of serum pro-inflammatory cytokines, including IL-6, TNF-α, and IL-1β as compared with free Cur, but augmented anti-inflammatory cytokine IL-10 by ELISA assay. Further, marked alleviation of the sepsis-induced damage to organs, including kidney, liver, and heart was observed with Cur-SLNs treatment as determined by hematoxylin/eosin-staining. Western blot analyses revealed that Cur-SLNs can significantly lower the expression levels of TLR4 , TLR2 , and TNF-α in lymph node tissues. Meanwhile, it showed suppressions of NF-κB activation and IκBα degradation levels. In conclusion, we suggested that Cur-SLNs may be used as an effective and safe therapeutic agent in treating sepsis in high-risk patient groups.

Redox-Based Strategy for Selectively Inducing Energy Crisis Inside Cancer Cells: An Example of Modifying Dietary Curcumin to Target Mitochondria

Ya-Long Zheng,Zhi-Shan Tu,Hong-Mei Cui,Shuai Yan,De-Chen Duan,Wei Tang,Fang Dai,Bo Zhou

Reprograming of energy metabolism is a major hallmark of cancer, but its effective intervention is still a challenging task due to metabolic heterogeneity and plasticity of cancer cells. Herein, we report a general redox-based strategy for meeting the challenge. The strategy was exemplified by a dietary curcumin analogue (MitoCur-1) that was designed to target mitochondria (MitoCur-1). By virtue of its electrophilic and mitochondrial-targeting properties, MitoCur-1 generated reactive oxygen species (ROS) more effectively and selectively in HepG2 cells than in L02 cells via the inhibition of mitochondrial antioxidative thioredoxin reductase 2 (TrxR2). The ROS generation preferentially mediated the energy crisis of HepG2 cells in a dual-inhibition fashion against both mitochondrial and glycolytic metabolisms, which could hit the metabolic plasticity of HepG2 cells. The ROS-dependent energy crisis also allowed its preferential killing of HepG2 cells (IC50 = 1.4 μM) over L02 cells (IC50 = 9.1 μM), via induction of cell-cycle arrest, apoptosis and autophagic death, and its high antitumor efficacy in vivo, in nude mice bearing HepG2 tumors (15 mg/kg). These results highlight that inhibiting mitochondrial TrxR2 to produce ROS by electrophiles is a promising redox-based strategy for the effective intervention of cancer cell energy metabolic reprograming.

Thermodynamic compatibility and conformational adaptability enable the matrix-adaptive protection of glycated pea protein isolate-curcumin nanoparticles in frying oils

Zhi-feng Tan,Cong Sun,Shi-hui Zhao,Zhen-wen Shao,Da-yong Zhou,Liang Song

g PPI-CUR NPs exhibit matrix-adaptive protection during simulated frying. • Antioxidant efficacy positively correlates with oil unsaturation levels. • Nanoparticles inhibit acrolein and cis-trans fatty acid isomerization. • Dual-adaptive behavior is driven by thermodynamic affinity with lipids. • Structural expansion in highly unsaturated oils maximizes the scavenging. The solvent effects exerted by continuous lipid matrices on nanocarrier antioxidant efficacy remain insufficiently defined. Herein, glycated pea protein isolate-curcumin nanoparticles ( g PPI-CUR NPs) were engineered to elucidate their matrix-adaptive kinetic responses across frying oils with distinct saturation profiles. Protective efficacy correlated positively with lipid unsaturation, demonstrating 55.1% relative peroxide inhibition in highly unsaturated soybean oil versus 31.7% in saturated palm oil. Concurrently, the system suppressed cytotoxic aldehydes accumulation, mitigated viscosity increments, and inhibited thermal cis -to- trans isomerization. Molecular dynamics simulations elucidated this structural adaptability. The protein shell underwent conformational expansion driven by thermodynamic compatibility with linoleic acid, while the internal core maintained hydrophobic retention for ligand stability. This structural equilibrium maximized solvent-accessible surface area, increasing interfacial collision probability for in situ lipid radical scavenging. These findings generalize the dynamic balance between interfacial flexibility and core retention as a fundamental design principle for targeted lipid protection in high-stress processing.
