Hierarchically porous zeolitic imidazole framework-8/cellulose membrane installed in filter as sorbent for microextraction in packed syringe towards trace tetracyclines in water samples

Lin Li,Sijia Zhang,Yingying Li,Ting Wang,Xiaohong Hou

In this study, zeolitic imidazole framework-8/cellulose membrane (ZIF-8/CM) was fabricated through an in situ synthesis method. The composite was installed in a dismountable filter and used as a sorbent for lab-made microextraction in packed syringe (MEPS). The entire MEPS procedure can be performed through simple operation of filtration, since the macroporous cellulose membrane was introduced as the substrate for ZIF-8 powder. Coupled to a high performance liquid chromatography tandem ultraviolet detector (HPLC-UV), the proposed method was applied for the analysis of tetracyclines in water samples. The extraction parameters, including the adsorbent type, sample solution pH, sample volume, type and volume of the elution solvent, were systematically studied. Under optimized conditions, desirable linearity (r > 0.99 for three analytes), high sensitivity (limit of detection: 0.1373–0.8917 μg L−1), good precision (RSD < 6.3%) and ideal recovery (72.9–97.6%) were obtained. The proposed method was demonstrated to be efficient, easily operative and environmentally friendly.

Degradation of sulfadiazine in aqueous media by peroxymonosulfate activated with biochar-supported ZnFe2O4 in combination with visible light in an internal loop-lift reactor

Yan Wang,Tao Gan,Jingyu Xiu,Ganghua Liu,Haiming Zou

Solid waste resource utilization and the treatment of wastewater are two important aspects in environmental protection. Here, biochar (BC) derived from municipal sewage sludge has been combined with ZnFe2O4 to form the photocatalyst ZnFe2O4/biochar (ZnFe/BC), and it was used to degrade sulfadiazine (SDZ) in the presence of peroxymonosulfate (PMS) under visible (Vis) light irradiation in an internal loop-airlift reactor (ALR). The surface morphology and structure of ZnFe/BC have been characterized by X-ray diffraction (XRD), scanning electron microscopy equipped with an attachment for energy-dispersive spectroscopy (SEM-EDS), X-ray photoelectron spectroscopy (XPS), and UV-Vis diffuse reflectance spectroscopy (UV-Vis DRS). ZnFe/BC displays outstanding photocatalytic performance and reusability. After four reuse cycles of ZnFe/BC in the Vis/ZnFe/BC/PMS system, the SDZ degradation rate and efficiency still reached 0.082 min−1 and 99.05%, respectively. Reactive species in this system included free radicals SO4˙−, ˙OH, and ˙O2−, as well as non-radicals 1O2, e−, and h+, as established from the results of chemical quenching experiments and electron paramagnetic resonance (EPR) analyses. Moreover, a mechanism of action of the Vis/ZnFe/BC/PMS system for SDZ degradation was proposed. The acute toxicity of the reaction solution towards Photobacterium phosphoreum T3 spp. in the Vis/ZnFe/BC/PMS process increased during the first 40 min and then decreased, illustrating that Vis/ZnFe/BC/PMS provided an effective and safe method for the removal of SDZ.

Transparent and highly efficient full-band UV-shielding bioplastic designed by in-situ embedded metal-organic frameworks (MIL-68(In)-NH2) in cellulose matrix

Sun Lijian,Qian Xueren,An Xianhui

Cellulose-based bioplastics with degradability and remarkable mechanical performance have recently attracted increasing attention. As a high-performance material, cellulose-based bioplastics have a strong potential to replace synthetic plastic substrates in many applications. However, its poor shielding performance to UV light limit its application. In this work, transparent and UV-blocking MIL-68(In)-NH 2 @cellulose bioplastic (M(In)CBP) sheets are fabricated via a featured processing method, i.e., cellulose liquefaction, rapid solidification, in-situ synthesis and hot-press drying. The as-obtained functional bioplastic sheet (M(In)CBP1) exhibits high visible-light transparence (74.7% a 600 nm) and the existence of MIL-68(In)-NH 2 allows superior UV-blocking ability (below 400 nm, transmittance was under 4%). The M(In)CBP sheets show high photostability even after continuous UV irradiation (365 nm) for over 12 h. The superior UV-shielding performance is attributed to the uniform distribution and the high UV absorbance of MIL-68(In)-NH 2 in M(In)CBP sheets. High resistance to penetration by aqueous/nonaqueous liquids was proved. Moreover, the M(In)CBP sheets also show excellent mechanical properties and thermal stability. The present work designed a UV-blocking bioplastic constructed from cellulose and MIL-68(In)-NH 2 , which is potential candidates to replace synthetic plastics in many advanced applications such as a UV protection layer for electronic products, or useful as packaging materials to prevent food spoilage. Characterization of chemical constituents and metabolites in vivo and in vitro after oral administration of Wuteng tablets in rats by UHPLC–Q/TOF–MS

Pengyi Chen,Chengguo Pang,Tianen Xu,Peiliang Dong,Hua Han

Waste medicinal plants are widely used in drug production. With the increasing demand for botanical drugs, there is an urgent need to identify new and effective drugs and improve the utilization of medicinal plant resources. Wuteng tablets (WTP) are extracted from the stem of Schisandra chinensis and have a good therapeutic effect on Alzheimer’s disease. In this study, a holistic identification strategy based on UHPLC–Q/TOF–MS was developed for the first time to investigate the metabolites and metabolic pathways involved in the in vitro metabolism and liver microsomal incubation and in the in vivo metabolic system of rats after WTP administration. After the oral administration of WTP, 21 metabolites were identified in the serum and 25 metabolites were identified in the urine, of which six were new metabolites; 33 metabolites were inferred from the microsomal metabolites in vitro . The metabolic pathways related to WTP mainly involve demethylation, hydroxylation, dehydroxylation and dehydrogenation. In this study, the metabolites and metabolic pathways of WTP were elucidated via UHPLC–Q/TOF–MS, which provided a basis for an in-depth study of the pharmacodynamic and pharmacotoxicological effects of WTP.

Insights into spirotetramat-induced thyroid disruption during zebrafish (Danio rerio) larval development: An integrated approach with in vivo, in vitro, and in silico analyses

Fang Jiao,Hua Rong,Yang Zhao,Panfeng Wu,Yaohui Long,Jie Xu,Tao Zhao,Lingxi Han,Jun Wang,Huirong Yang

Spirotetramat (SPT), a tetronic acid-derived insecticide, is implicated in reproductive and lipid metabolism disorders, as well as developmental toxicity in fish. While these effects are documented, the precise mechanisms underlying its developmental toxicity are not fully elucidated. In this study, zebrafish embryos (2 h post-fertilization, hpf) were exposed to four concentrations of SPT (0, 60, 120, and 240 μg/L) until 21 dpf (days post-fertilization). We delved into the mechanisms by examining its potential disruption of the thyroid endocrine system, employing in vivo , in vitro , and in silico assays. The findings showed notable developmental disturbances, including reduced hatching rates, shortened body lengths, and decelerated heart rates. Additionally, there was an increase in malformations and a decline in locomotor activity. Detailed analyses revealed that SPT exposure led to elevated thyroid hormone levels, perturbed the hypothalamic-pituitary-thyroid (HPT) axis transcript levels, amplified deiodinase type I (Dio1) and deiodinase type II (Dio2) activities, and both transcriptionally and proteomically upregulated thyroid receptor beta (TRβ) in larvae. Techniques like molecular docking and surface plasmon resonance (SPR) confirmed SPT's affinity for TRβ, consistent with in vitro findings suggesting its antagonistic effect on the T3-TR complex. These insights emphasize the need for caution in using tetronic acid-derived insecticides.

Identification of novel antioxidant collagen-peptides for preventing and treating H2O2-induced oxidative stress in HepG2 cells through in vitro and in silico approaches

Bowei Du,Chao Zhang,Guiya Deng,Shuai Zhang,Shiqi Wang,Yuepeng Guan,Yaqin Huang

Background Nowadays, the prevalence of oxidative stress-related chronic diseases is increasing. The identification of novel antioxidant collagen-peptides to counteract oxidative stress for individuals’ health have gained significant intention. Results In this study, the collagen-peptides with antioxidant activities were separated and identified by ion chromatography (IEC), reversed-phase high-performance liquid chromatography (RP-HPLC), and liquid chromatography-tandem mass spectrometry (LC-MS/MS). The identified antioxidant collagen-peptides were further screened by molecular docking for Keap1-targeted peptide inhibitors and their theoretical interaction mechanisms were investigated. Four novel antioxidant collagen-peptides, GPAGPIGPVG, GPAGPpGPIG, ISGPpGPpGPA and IDGRPGPIGPA with high binding affinity to Keap1 were selected. Molecular docking results demonstrated that the putative antioxidant mechanism of four antioxidant collagen-peptides contributed to their blockage of Keap1-Nrf2 interactions. The results of antioxidant activity of four antioxidant collagen-peptides proved that IDGRPGPIGPA exerted a high scavenging capacity to DPPH and ABTS free radicals, while GPAGPpGPIG improved the resistance of cells to hydrogen peroxide (H 2 O 2 ) induced oxidative damage by promoting the activation of intracellular antioxidant enzymes and the production of reduced glutathione in human hepatoma (HepG2) cells. Conclusion The antioxidant collagen-peptides (GPAGPIGPVG, GPAGPpGPIG, ISGPpGPpGPA and IDGRPGPIGPA) will be developed as the novel functional food for human health in the near future. In situ stabilization of the adsorbed Co2+ and Ni2+ in rice straw biochar based on LDH and its reutilization in the activation of peroxymonosulfate

Qiang Yang,Peixin Cui,Cun Liu,Guodong Fang,Meiying Huang,Qiuyue Wang,Yiyi Zhou,Hongbo Hou,Yujun Wang

The stabilization of heavy metals adsorbed in the spent biochar remains a challenge, and the risk of secondary pollution under environmental changes will rise under inadequate post-treatment. Moreover, the safe and sufficient re-utilization of the spent biochar with heavy metals loaded have attracted extensive attention. In this research, the performance and mechanism of rice straw biochar (RSBC) pyrolyzed at different temperature for nickel and cobalt adsorption were investigated, and the stabilization of the adsorbed heavy metals was achieved via a simple two-step strategy: the adsorption of metal ions and the hydrothermal process to form the stable layered double hydroxides (LDH) on biochar, with a leaching rate below 0.005% evaluated by EPA toxicity characteristic leaching procedure (TCLP). Meanwhile, the stabilized RSBC-LDH can be reused as an excellent catalyst in the activation of peroxymonosulfate (PMS) to degrade organic pollutants efficiently. This work eliminated the risk of heavy metal desorption from the spent biochar, and enabled a new strategy for the optimized utilization of biochar in environmental remediation.

In Situ Synthesis of FeOCl in Hollow Dendritic Mesoporous Organosilicon for Ascorbic Acid-Enhanced and MR Imaging-Guided Chemodynamic Therapy in Neutral pH Conditions

Tianyao Li,Fei He,Bin Liu,Tao Jia,Boyang Shao,Ruoxi Zhao,Hui Zhu,Dan Yang,Shili Gai,Piaoping Yang

Chemodynamic therapy (CDT) based on the Fenton reaction is a promising strategy for nonlight cancer treatment. However, the traditional Fenton reaction is only efficient in strongly acidic conditions (pH = 2–4), resulting in the limited curative effect in a weakly acidic tumor microenvironment (TME). Herein, we first developed a simple in situ growth method to confine FeOCl nanosheets into hollow dendritic mesoporous organosilicon (H-DMOS) nanoparticles to obtain FeOCl@H-DMOS nanospheres. Ascorbic acid (AA) was then absorbed on the nanosystem as a H2O2 prodrug and, meanwhile, was used for the regeneration of Fentons reagent for Fe2+. Finally, poly(ethylene glycol) (PEG) was coated on FeOCl@H-DMOS-AA to enhance the permeability and retention (EPR) effect in tumor tissue. The as-fabricated FeOCl@H-DMOS-AA/PEG can generate a large amount of highly toxic hydroxyl radicals (•OH) by catalyzing H2O2 even in neutral pH conditions with the help of AA. As a result, the effect of CDT has been markedly enhanced by the increased amount of H2O2 and the efficient Fenton reaction in mild acidic TME, which can remove almost all of the tumors in mice. In addition, FeOCl also endows the nanosystem with T2-weighted MR imaging capability (r2 = 34.08 mM–1 s–1), thus realizing the imaging-guided cancer therapy. All in all, our study may contribute a new direction and may have a bright future for enhanced CDT with a neutral pH range.

Preparation and characterization of paclitaxel palmitate albumin nanoparticles with high loading efficacy: an in vitro and in vivo anti-tumor study in mouse models

Hang Chen,Sifan Huang,Heyi Wang,Xinmei Chen,Haiyan Zhang,Youfa Xu,Wei Fan,Yun Pan,Qiuyan Wen,Zhizhe Lin,Xuena Wang,Yongwei Gu,Baoyue Ding,Jianming Chen,Xin Wu

Background Combination of the prodrug technique with an albumin nano drug-loaded system is a novel promising approach for cancer treatment. However, the long-lasting and far-reaching challenge for the treatment of cancers lies in how to construct the albumin nanometer drug delivery system with lead compounds and their derivatives. Methods In this study, we reported the preparation of injectable albumin nanoparticles (NPs) with a high and quantitative drug loading system based on the NabTM technology of paclitaxel palmitate (PTX-PA).Results Our experimental study on drug tissue distribution in vivo demonstrated that the paclitaxel palmitate albumin nanoparticles (Nab-PTX-PA) remained in the tumor for a longer time post-injection. Compared with saline and paclitaxel albumin nanoparticles (Abraxane®), intravenous injection of Nab-PTX-PA not only reduced the toxicity of the drug in normal organs, and increased the body weight of the animals but maintained sustained release of paclitaxel (PTX) in the tumor, thereby displaying an excellent antitumor activity. Blood routine analysis showed that Nab-PTX-PA had fewer adverse effects or less toxicity to the normal organs, and it inhibited tumor cell proliferation more effectively as compared with commercial paclitaxel albumin nanoparticles. Conclusions This carrier strategy for small molecule drugs is based on naturally evolved interactions between long-chain fatty acids (LCFAs) and Human Serum Albumin (HSA), demonstrated here for PTX. Nab-PTX-PA shows higher antitumor efficacy in vivo in breast cancer models. On the whole, this novel injectable Nab-PTX-PA has great potential as an effective drug delivery system in the treatment of breast cancer.

Comparison of lipid radical scavenging capacity of spice extract in situ in roast beef with DPPH and peroxy radical scavenging capacities in vitro models

Yingjie Bao,Xiaopu Ren,Yuxia Zhu,Yawei Zhang,Zengqi Peng,Guanghong Zhou

The effects of 7 spice extracts on lipid radical formation were first collectively studied in situ in roasted beef by electron spin resonance (ESR). And the correlations among the ESR and conventional indexes including 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging capacity, reducing power, oxygen radical absorption capacity (ORAC), conjugated diene value (CDV) and thiobarbituric acid-reactive substances (TBARS) were also analysed. Clove was observed the highest scavenging lipid radical and DPPH radical capacity, and rosemary showed the highest peroxyl radical scavenging capacity, while tert-butylhydroquinone (TBHQ) significantly increased lipid radicals in roasted beef ( p  < 0.05). ESR showed significant correlations with the DPPH and CDV assays ( p  < 0.05) but no correlation with the ORAC and TBARS assays ( p  > 0.05), indicating that with the addition of spice extracts, the lipid radical formation in roasted beef could be predicted by the DPPH assay but not be appropriate to evaluate the oxidation stability in roasted beef.

Effects of Tin in La–Sn-Codoped Zn(O,S) Photocatalyst to Strongly Cleave the Azo Bond in Azobenzene with in Situ Generated Hydrogen

Hairus Abdullah,Yu-Rou Ko,Dong-Hau Kuo,Noto Susanto Gultom

La–Sn-codoped Zn(O,S) catalysts were synthesized with different amounts (0%, 2.5%, 5%, and 10%) of Sn and a constant amount (10%) of La to improve the photocatalytic hydrogenation reaction (PHR) of azobenzene to aniline. The as-prepared catalysts were carefully characterized and tested for a hydrogenation reaction. The incorporation of Sn significantly enhanced the PHR activities since the incorporated Sn in the Zn(O,S) lattices could increase the conductivity of the catalysts to improve the charge transfer during the catalytic reaction as indicated with EIS measurement. Further measurement with a photoresponse of La–Sn-codoped Zn(O,S) catalysts also exhibited relatively higher intensities as compared to those of La-doped Zn(O,S) and Sn-doped Zn(O,S) catalysts. On the basis of the measurement results of EIS and transient photo current, the La–Sn-codoped Zn(O,S) with the best properties was further utilized for PHR to convert azobenzene to aniline. GC–MS measurement confirmed that 15 ppm azobenzene could be totally converted to aniline in only 60 min which was achieved with a catalyst that was prepared with 5%-Sn and 10%-La doping. The relatively short reaction time indicated that the as-prepared catalyst had a strong reduction capability able to cleave the strong bonding of N═N in azobenzene. The reaction kinetic of the N═N bond cleavage was elucidated based on solvation, adsorption, pinning, and photocatalytic hydrogenation processes on the catalyst surfaces.

Combination of Polypropylene Mesh and in Situ Injectable Mussel-Inspired Hydrogel in Laparoscopic Hernia Repair for Preventing Post-Surgical Adhesions in the Piglet Model

Wanjun Hu,Zhigang Zhang,Long Zhu,Yazhou Wen,Tianzhu Zhang,Pengfei Ren,Faming Wang,Zhenling Ji

Polypropylene (PP) mesh has been used successfully for a long time in clinical practice as an impressive prosthesis for ventral hernia repair. To utilize a physical barrier for separating mesh from viscera is a general approach for preventing adhesions in clinical practice. However, a serious abdominal adhesion between the mesh and viscera can possibly occur post-hernia, especially with the small intestine; this can lead to a series of complications, such as chronic pain, intestinal obstruction, and fistula. Thus, determining how to prevent abdominal adhesions between the mesh and viscera is still an urgent clinical problem. In this study, a dopamine-functionalized polysaccharide derivative (oxidized-carboxymethylcellulose-g-dopamine, OCMC-DA) was synthesized; this was blended with carboxymethylchitosan (CMCS) to form a hydrogel (OCMC-DA/CMCS) in situ at the appropriate time. The physical and chemical properties of the hydrogel were characterized successfully, and its excellent biocompatibility was presented by the in vitro cell test. The combination of this hydrogel and PP mesh was used in laparoscopic surgery for repairing the abdominal wall defect, where the hydrogel could become fixed in situ on the PP mesh to form an anti-adhesion gel-mesh. The results showed that the gel-mesh could prevent abdominal adhesions effectively in the piglet model. Moreover, the histology and immunohistochemical staining proved that the gel-mesh could effectively alleviate the inflammation reaction and deposition of collagen around the mesh, and it did not disturb the integration between mesh and abdominal wall. Thus, the gel-mesh has superior tissue compatibility.
