In this research, a fluorescent probe of 7-(diethylamine) coumarin derivatives with multiple binding sites to detect biothiols in tumor cell with strong NIR II luminescence in vivo was synthesized. The biothiols include cysteine (Cys) and glutathione (GSH) in tumor cells, and the tumor-response luminescence was proved by the cell experiment. Importantly, the monolayer functional phospholipid (DSPE-PEG) coating and aggregation induced emission (AIE) dye of TPE modification made the probe have good stability and biocompatibility with little luminescence quenching in aqueous phase, which was proved by in vitro and in vivo experiments. The final aqueous NIR II probe combined with bevacizumab (for VEGF recognition in the cancer cells) and Capmatinib (for Met protein recognition in the cancer cells) has stronger targeted imaging on head and neck squamous cell carcinoma (HNSCC) cancer with intravenous injection. This GSH/Cys detection in the tumor cell and strong dual-molecular NIR II bioimaging in vivo may provide new strategy to tumor detection.

Early diagnosis is critical and challenging for tongue squamous cell carcinoma (TSCC), which is a kind of tumor with high malignancy, poor prognosis, and a high incidence of invasion and metastasis. In this research, dual-modal optical imaging rare earth nanoparticle (RENP) probes with peptide functionalization are designed for targeted TSCC imaging and therapy. RENP@C@Au (UCA) with enhanced red upconversion luminescence (UCL) and near infrared II (NIR II) imaging intensity is designed by metal and codopant modulation, and the two peaks at about 650 nm and 1064 nm of Nd ions are relatively stable with less quenching and suitable for luminescence bioimaging. Then, the cMBP peptide targeted to Cal 27 TSCC cells with highly expressed c-MET proteins is combined with UCA. Compared with human's normal cells of MCF-10A, other tumor cell lines such as A549 and HeLa, together with mice tumor cells of 4T1, the designed probe has targeted imaging and therapy to Cal 27. The final in vivo experiment shows that the probe with high NIR II luminescence signals is not easy to retain when injected into the tail vein, indicating its potential precise clinical application for the diagnosis and therapy of TSCC.

Regulating the level of reactive oxygen species (ROS) in a tumor is an efficient and innovative anticancer strategy. However, the therapeutic efficacy of ROS-based therapies, such as chemodynamic therapy (CDT) and photodynamic therapy (PDT), offers finite outcomes due to the oxygen dependence and limited concentration of hydrogen peroxide (H2O2) and overexpression of glutathione (GSH) within the tumor microenvironment (TME), so a single therapeutic strategy is insufficient to completely eliminate tumors. Therefore, we demonstrated an omnipotent nanoplatform MnO2/Ag3SbS3 (abbreviated as MA) with strong optical absorbance in the NIR-II biowindow and oxygen self-sufficient ROS-mediated ability, which not only relieves tumor hypoxia significantly but also enhances the photothermal therapy (PTT)/PDT/CDT efficacy. By 1064 nm laser irradiation, MnO2/Ag3SbS3 nanoparticles (NPs) reveal a favorable photothermal conversion efficiency of 23.15% and achieve a single-laser-triggered NIR-II PTT/PDT effect, resulting in effective tumor elimination. Once internalized into the tumor, MnO2/Ag3SbS3 NPs will be degraded to Mn2+ and Ag3SbS3. The released Ag3SbS3 NPs as a NIR-II phototherapy agent could be utilized for photoacoustic imaging-guided NIR-II PDT/PTT. Mn2+ could be used as a Fenton-like catalyst to continuously catalyze endogenous H2O2 for generating highly virulent hydroxyl radicals (•OH) for CDT and O2 for PDT, enhancing the efficiency of PDT and CDT, respectively. Meanwhile, Mn2+ realizes magnetic resonance imaging-guided accurate tumor therapy. Moreover, the MnO2/Ag3SbS3 NPs could deplete intracellular GSH in TME to promote oxidative stress of the tumor, further strengthening ROS-mediated antitumor treatment efficacy. Overall, this work presents a distinctive paradigm of TME-responsive PDT/CDT/PTT in the second near-infrared biowindow by depleting GSH and decomposing H2O2 for efficient and precise cancer treatment.

A NIR-II fluorescent probe Py-RP is constructed via spirolactamization of rhodamine–polymethine and 2-hydrazinobenzothiazole. • Intermolecular hydrogen bond-triggered metal-free spiroring opening enables highly selective and sensitive NIR-II imaging of SA. • The probe achieves real-time monitoring of plant abiotic stress and visual identification of SA residues on fruit surfaces. Fluorescence imaging of salicylic acid (SA) is key to understanding plant protective mechanisms. However, existing SA probes are limited by their visible or NIR-I emission, where tissue scattering and autofluorescence compromise imaging quality. To address this limitation, we developed a rhodamine-based NIR-II fluorescent probe, named Py-RP, by functionalizing a rhodamine–polymethine scaffold with 2-hydrazinobenzothiazole via spirolactamization. Its fluorescence turn-on mechanism originates from a metal-free spirocyclic ring-opening process, initiated by a cooperative hydrogen-bonding network formed between the spirolactam carbonyl group of Py-RP and the ortho-hydroxyl/carboxyl moieties of SA. This structural opening extends the π-conjugated system, enabling distinct NIR-II fluorescence activation upon SA recognition. Py-RP exhibited high selectivity and sensitivity toward SA, with a detection limit of 0.15 μM, allowing for dynamic monitoring of plant responses to abiotic stress and visual analysis of SA residues on fruit surfaces. This rhodamine-based NIR-II probe opens an avenue for advanced research in plant stress physiology. Download: Download high-res image (168KB) Download: Download full-size image

The combination of multidrug chemotherapy and photothermal therapy (PTT) enhances cancer therapeutic efficacy. Herein, we develop a simple and smart pH/NIR dual-stimulus-responsive degradable mesoporous CoFe2O4@PDA@ZIF-8 sandwich nanocomposite. The mesoporous CoFe2O4 core acts as T2-weighted magnetic resonance (MR) imaging probe, PTT agent, and loading platform of hydrophilic doxorubicin (DOX). A polydopamine (PDA) layer is used to avoid the premature leakage of DOX before arriving at tumor site, enhance PTT efficiency, and facilitate the integration of ZIF-8 (a kind of metal–organic framework). The ZIF-8 shell serves to encapsulate hydrophobic camptothecin (CPT) and as the switch for the pH and NIR stimulation-responsive release of the two drugs. Therefore, T2-weighted MR imaging-guided multidrug chemotherapy and PTT synergistic treatment is achieved. Two kinds of anticancer drugs, hydrophilic DOX and hydrophobic CPT, are successfully loaded in CoFe2O4 and ZIF-8, respectively, so no mutual interference between the two drugs exists. A unique two-stage stepwise release process is exhibited for CPT and DOX with an interval of 12 h to improve the anticancer efficacy under the acidic microenvironment of tumor tissue. NIR irradiation achieves the burst drug-release and PTT after laser stimulation, simultaneously. With this smart design, high drug concentration is achieved at the tumor site by quick release, especially for the therapeutic drugs that show nonlinear pharmacokinetics, and PTT is integrated efficiently. Furthermore, negligible biotoxicity and a remarkable synergic antitumor effect of the hybrid nanocomposites are validated by HepG2 cells and tumor-bearing mice as models. Our multidrug delivery-releasing composite improves tumor therapeutic efficiency significantly compared with a single-drug chemotherapy system. The simple multifunctional composite system can be applied as an effective platform for personal nanomedicine with diagnosis, smart drug delivery, and cancer treatment through its remarkable photothermal property and controllable multidrug release.

Anisotropic plasmonic metals have attracted significant attention in enhancing the catalytic performance of catalysts due to their broad light-harnessing capabilities and active hot electrons; however, limited investigations have been dedicated towards improving their electrochemical reaction performance in the visible and near infrared (NIR) regions. Herein, anisotropic Pt-edged Au nanodisks (NDs) were synthesized by controlling the preferential loading of Pt and used as catalysts for plasmon-enhanced electrochemical methanol oxidation reactions (MORs) under visible-NIR light irradiation by, and the light-enhanced electric current over the Pt-edged Au NDs was found to be 3-fold higher than that under dark conditions. Wavelength-dependent electric current over the Pt-edged Au NDs for the MOR in the visible-NIR light region demonstrates that the light-induced enhancement of the electric current is due to surface plasmon resonance (SPR) of the Au NDs. Furthermore, plasmonic hot electron transfer was studied by the single-particle photoluminescence images and spectra of Au NDs and Pt–Au NDs, and the dipole surface plasmon resonance (DSPR) mode was proved to be the main channel for hot electron transfer. During the electrochemical reaction under visible-NIR light irradiation, a plasmonic hot electron is transferred to the electrode, and a “hot hole” is left on the surface, boosting the MOR.

Mechanoluminescence (ML) is a phenomenon upon external mechanical stimuli and it has found diverse applications such as stress sensing, structure health diagnosis, 3-D signature, energy harvesting etc because of its unique properties of in situ and real-time response to the stimuli. However, ML of most of the state-of-art phosphors primarily appears within the spectral range from ultraviolet to visible, which does not lie in the biological transparent windows, and, therefore, limits its applications in biological field. Here, we report a strong near infrared (NIR) ML from orthorhombic Cmcm perovskite Sr 3 Sn 2 O 7 : Nd 3+ , which is peaked at ~ 900 nm and located exactly within the first optical transparent window of tissues. The ML apparates after gradual discharge of the traps deeper than 0.73 eV triggered by the external mechanical stimuli and subsequently excitation of Nd 3+ to the state of 4 F 3/2 . The rechargable ML presents well repeatable linearity to loaded force at least up to 5000 N, and it can penetrate tissues easily that are thick up to 30 mm such as pigskin and the ceramic disk of hydroxy apatite which is the main constituent of bone. These results demonstrate the potential application for in situ biomechanical sensor. Meanwhile, by recording and processing these ML signals during signing on a pellet sample, for the first time, we provide a novel signature system based on NIR ML. This could raise the security level of existed signature anti-countering to a higher level.

Cuproptosis has gained significant attention in cancer therapy. Use of copper-overloaded combination therapy that employs a variety of cell death mechanisms has emerged as a promising avenue in the treatment of cancer. However, integrating different modes in a system with optimal synergy and minimal side effects remains a challenge. In this study, a multifunctional copper ion-overloaded nanoparticle was fabricated via one-pot self-assembly for enhanced cuproptosis, chemo- and phototherapy. Molecular dynamics simulation was performed to study the self-assembly mechanism, demonstrating they assembled into stable nanoparticles (NPs) in 50 ns. The NPs possess dual-targeting capabilities towards tumor cells and mitochondria, and exhibit excellent stability in neutral PBS, while showing responsive payload release under acidic conditions or NIR laser irradiation. Upon reaching the mitochondria, the NPs release their payloads, where accumulated copper ions trigger aggregation of lipoylated proteins, etc. Released disulfiram chelates excessive copper ion to form toxic Cu(DTC) 2 , which induces apoptosis combining with hydroxycamptothecin. Meanwhile, indocyanine green-generated ROS and photothermal effects further enhance cuproptosis for synergistic therapy. In vitro results revealed potent cytotoxicity of the NPs against CT26 cells while negligible effects on normal cells. Furthermore, significant reactive oxygen species generation, mitochondrial membrane potential alteration, increased apoptosis, and modulated protein mRNA expression levels, confirmed the synergistic combination of chemotherapy, cuproptosis, and photothermal/photodynamic therapies. Notably, in vivo experiments demonstrated selective tumor accumulation and outstanding antitumor efficacy (89.5% inhibition rate) with excellent biosafety profile. The NPs provide a novel strategy for cuproptosis-based combination cancer therapy and shows significant clinical translation potential.

Blue-light-excitable near-infrared (NIR) emitting materials exhibit significant potential for compact light sources in bioimaging, nondestructive detection, and night vision. However, achieving efficient NIR-II/III (1000–1800 nm) emission under blue excitation remains challenging due to the large excitation–emission Stokes shift and the forbidden nature of f – f or d – d transitions. In this study, we report on the Ni 2 + -doped monoclinic double perovskite Ca 2 ScTaO 6 , which exhibits broadband NIR-II/III with a full width at half maximum of 240 nm and a photoluminescence quantum yield of approximately 21% when excited at 425 nm. The incorporation of Fe 3+ ions significantly improves the photoluminescence quantum yield to approximately 37% through energy transfer pathways from Fe 3+ to Ni 2+ and crystal field modulation. Structural and spectroscopic investigations reveal the presence of Fe 3+ ions at various lattice sites, resulting in localized distortions that facilitate the relaxation of d – d transitions in Ni 2+ . A prototype NIR light-emitting diode utilizing Ca 2 ScTaO 6 :Fe 3+ , Ni 2+ exhibits strong NIR-II/III emission, facilitating the visualization of subsurface damage in various materials, including fruits, capsules, meat, and silicon wafers. This work demonstrates an effective strategy for designing blue-light-excitable NIR materials through synergistic energy transfer and crystal field engineering.

Lanthanide NIR-II luminescence free from thermal quenching is attractive for various applications at elevated temperatures. Here, we present a thermal enhancement of Er 3+ NIR-II luminescence based on the synergy of phonon assisted Er 3+ -Er 3+ cross relaxations and controllable Er 3+ -Ho 3+ energy transfer. The lattice expansion of NaYGeO 4 olivine phosphor upon heating (298–823 K) suppresses the Er 3+ -Ho 3+ energy transfer and leads up to 46.1 times enhancement of Er 3+ NIR-II luminescence under 379 nm excitation. The NIR-II luminescence evolution as increasing temperature allows dual-mode ratiometric thermometry, i.e., luminescence intensity ratios (LIRs) based on Er 3+ 4 I 13/2 Stark sublevels and on Er 3+ 4 I 13/2 and Ho 3+ 5 I 6 levels. Obtained relative sensitivities above 0.46% K ‒1 over a broad temperature range of 298–673 K with a maximum of 2.32% K -1 suggest NaYGeO 4 :Er 3+ /Ho 3+ phosphor as a promising NIR-II luminescence thermometer. Moreover, this research offers an available perspective for designing thermal-enhanced NIR luminescence in ordinary thermal expansion materials.

We report the synthesis of an organic/inorganic architecture, a rare-earth upconversion nanoparticle (UCNP)-based core with organic porous silicon shell nanocapsules, that enables visible and ultraviolet light emission under 980 nm near-infrared (NIR) irradiation for photodegradation. A mesoporous organosilica shell containing disulfide bonds was uniformly coated onto UCNP cores via a combined sol–gel and hard-template strategy. Glutathione (GSH) – triggered degradation of the nanocarriers was quantified using the molybdenum blue method. A photoresponsive azobenzene derivative, activatable by both UV and visible light, was synthesized and grafted onto the nanocarrier surface. Doxorubicin (DOX) was loaded into the nanocapsules, achieving a drug-loading capacity of 12.2 wt%. At neutral pH, minimal DOX leakage occurred; maximal release was observed under a composite stimulus environment. The Baker-Lonsdale model was employed to calculate diffusion coefficients under varying release conditions, providing quantitative guidance for designing multi-responsive drug-release systems. In vitro cellular assays showed that synergistic stimuli—NIR irradiation combined with an acidic, GSH-rich microenvironment—trigger controllable DOX release, enabling efficient tumor cell ablation. This work demonstrates a versatile, multi-responsive delivery platform combining UCNP-based photoreactivity, GSH-triggered degradation, and photoactive surface gating to achieve targeted, low-toxicity, on-demand chemotherapy.

Tumor hypoxia limits the efficacy of photodynamic therapy (PDT), necessitating photosensitizers with hypoxia-adaptive therapy and imaging. Here, we present a fluorinated N -oxide aza-BODIPY ( OFBD ) nanoemulsion ( OFBD-NP ) for hypoxia-responsive, multimodal imaging-guided phototherapy. OFBD generates robust singlet oxygen under normoxia for PDT, but is reduced by CYP450 enzymes in hypoxic cells to photothermal-potent FBD , enabling switchable PDT/PTT. Co-assembly with fluorinated oil enhances oxygen delivery, boosts 19 F MRI sensitivity, and promotes J -aggregation, shifting fluorescence into the NIR-II window for deep-tissue imaging. The redox conversion also activates photoacoustic signals, enabling responsive tri-modal imaging (NIR-II FLI/ 19 F MRI/PAI). OFBD-NP shows potent cytotoxicity in vitro under both normoxic and hypoxic conditions via apoptosis. In vivo, it selectively accumulates in tumors, offers high-contrast imaging, and leads to complete tumor regression in subcutaneous A549 models after laser irradiation, without systemic toxicity. This work demonstrates a smart nanoplatform that integrates deep-tissue imaging and hypoxia-triggered therapeutic switching, addressing major limitations of conventional photosensitizers in cancer imaging and phototherapy.
