Post-craniotomy scalp infection with antibiotic-resistant bacteria remains a great challenge in neurosurgery clinical practice, which hinders the healing process and increases the risk of intracranial infection, thereby endangering life. In this study, a versatile injectable hydrogel dressing with antibacterial and angiogenic activity composed of Pluronic F-127, silk fibroin (SF), and graphene oxide quantum dots (GOQDs) is fabricated (F-127/SF/GOQDs). The hydrogel is rapidly formed through thermosensitive gelation at body temperature and has controllable degradation and good biocompatibility. In vitro studies show that the F-127/SF/GOQDs hydrogel demonstrates excellent antibacterial performance (including drug-resistant strains) through photothermal treatment (PTT) via disruption of cellular energy metabolism and membrane function. In the meantime, the F-127/SF/GOQDs hydrogel promotes endothelial cell migration and differentiation toward CD31 hi Emcn hi type H vessels. In vivo results using post-craniotomy scalp infection models show that the hydrogel significantly enhances bacterial clearance to accelerate wound closure, and promotes angiogenesis and the immune modulation, which are related to enhanced collagen deposition, re-epithelialization and, ultimately accelerates wound healing of post-craniotomy scalp infections. Collectively, this multifunctional F-127/SF/GOQDs hydrogel, with dual action of photothermal antibacterial efficacy and type H vessels-related angiogenesis promotion, emerges as an innovative injectable dressing with potential for advancing post-craniotomy scalp infection management. Background Metabolic reprogramming is a hallmark of cutaneous squamous cell carcinoma (cSCC), yet the underlying mechanisms remain incompletely understood. SLC25A1, a mitochondrial citrate transporter, has been implicated in tumor progression, but its role in cSCC is unclear. Objective This study aimed to investigate the role of SLC25A1 in cSCC progression and elucidate the underlying molecular mechanisms. Methods SLC25A1 expression was examined in cSCC tissues and cell lines using immunohistochemistry, RT-qPCR, and Western blotting. Functional assays, including proliferation, apoptosis, migration, and invasion analyses, were performed following SLC25A1 knockdown or overexpression. RNA sequencing and metabolic assays were conducted to explore underlying mechanisms. Chromatin immunoprecipitation (ChIP)-qPCR was used to assess pan-histone H3 acetylation. In vivo effects were evaluated using DMBA/TPA-induced cSCC mouse models, with pharmacological inhibition by CTPI-2. Results SLC25A1 was significantly upregulated in cSCC and promoted tumor cell proliferation, migration, and invasion while inhibiting apoptosis. Mechanistically, SLC25A1 enhanced glycolysis by increasing pan-histone H3 acetylation at the ALDOC promoter, thereby upregulating ALDOC expression. Knockdown of SLC25A1 reduced glycolytic activity and tumor growth, whereas ALDOC overexpression partially rescued these effects. Pharmacological inhibition of SLC25A1 with CTPI-2 suppressed tumor progression and altered metabolic profiles in vivo. Conclusions SLC25A1 promotes cSCC progression by regulating glycolysis through pan-acetylated H3-mediated transcriptional activation of ALDOC. Targeting SLC25A1 may represent a promising therapeutic strategy for cSCC. Graphical

Background Acute lung injury (ALI) is a life‑threatening respiratory disease that lacks precise diagnostic tools and specific therapeutic interventions, highlighting an urgent need for theranostic strategies. Here, we developed a biomimetic multienzyme‑mimetic Prussian blue nanozyme (MM@HMPB/Sim) that couples catalytic ROS neutralization with macrophage reprogramming for photoacoustic (PA) imaging‑guided immunotherapy of ALI. Results The nanoplatform was constructed by cloaking simvastatin (Sim)‑loaded hollow mesoporous Prussian blue (HMPB) metal–organic framework (MOF) nanozyme with a native macrophage membrane (MM). The HMPB core exhibited catalase (CAT)‑, peroxidase (POD)‑, and superoxide dismutase (SOD)‑like activities, enabling cascade catalytic scavenging of reactive oxygen species (ROS). MM@HMPB/Sim efficiently scavenged ROS, alleviated oxidative cellular damage, promoted M1-to-M2 macrophage repolarization, and inhibited monocyte adhesion to inflamed endothelium via competitive receptor engagement. Mechanistically, the HMPB core appeared to upregulate NRF2-dependent antioxidant responses, while simvastatin seemed to enhance AMPKα1-mediated metabolic reprogramming. Their combination was associated with synergistic restoration of the PPARα/AMPKα1/NRF2 axis and suppression of NF-κB p65, correlating with reduced Th17 differentiation and IL-17A production, which may have facilitated a shift toward an M2-like phenotype. Together with MM-mediated inhibition of immune cell infiltration, these effects were accompanied by improved pulmonary immune homeostasis and attenuated lung injury. The nanozyme also displayed favorable biocompatibility and gradual in vivo clearance. Conclusion This biomimetic multienzyme‑mimetic nanozyme offers a safe, PA imaging‑guided catalytic immunotherapy that integrates ROS neutralization with synergistic metabolic and antioxidant pathway modulation to resolve ALI, holding translational potential for ROS-driven inflammatory lung diseases. To improve the performance in electrochemical immunoassays, paper-based devices typically necessitate the introduction of transducing materials through additional preparation steps, and the assay procedures often require labeling steps. Here we show that laser fabrication can generate both transducing and microfluidic components of paper-based devices to support label-free immunoassays. Laser parameters were adjusted to improve the electrochemical properties of paper-based graphenic transducing structures. Surface treatment for label-free biosensing was performed, and the device configuration and operation for electrochemical impedance spectroscopy sensing were explored. Interleukin-6 protein was used as a representative target to evaluate the analytical performance of the developed immunosensors. In phosphate buffered saline, the immunosensors exhibited a sensitivity of 67.95 Ω/decade and a limit of detection of 4.66 pg/mL. In artificial serum, the immunosensors exhibited a sensitivity of 57.08 Ω/decade and a limit of detection of 5.35 pg/mL. The clinical application potential was further demonstrated with human serum samples. The presented approach for developing paper-based microfluidic immunosensors can enhance their performance while maintaining ease of fabrication and usage to meet different requirements in clinical practice. With the increasingly severe environmental pollution, developing multifunctional fabrics integrating photocatalysis, oil–water separation, and photothermal anti-icing properties has become a significant challenge. In this study, a photothermal superhydrophobic fabric was successfully fabricated via a simple impregnation method, with polyester/cotton woven fabric (PTCO) as the substrate and polydimethylsiloxane (PDMS), dopamine hydrochloride (DA), and zinc oxide (ZnO) nanoparticles as raw materials. This work aimed to overcome the limitations of weak photothermal response and slow heating rate in traditional superhydrophobic fabrics while enhancing their functional versatility and application potential. The prepared PDA-ZnO@PDMS@PTCO exhibited a water contact angle of 160.3 ± 1.6° and demonstrated excellent stability. Owing to the photocatalytic property of ZnO, the modified PTCO achieved degradation rates of 97.1 ± 1.8% for methylene blue and 95.8 ± 1.2% for rhodamine B solutions. Furthermore, modifying ZnO with PDA endowed it with efficient photothermal performance. Under xenon lamp irradiation for 300 s, the surface temperature increased from 22.6 to 99.8 ℃, melting 1 cm 3 of ice within 146 s. Moreover, it exhibited high separation efficiency (96.3 ± 0.3–98.5 ± 0.2%) for six kinds of oil–water mixtures. The PDA-ZnO@PDMS@PTCO with photothermal and photocatalytic properties provides a promising approach for marine oil pollution treatment.

Estimating the time since deposition (TSD) of bloodstains at crime scenes is critical in forensic science. Although various approaches have demonstrated feasibility and accuracy for TSD estimation, analysis of volatile organic compounds (VOCs) offers a promising alternative because it is noninvasive, rapid, and potentially cost-effective. In this study, human blood was chemically profiled using solid-phase microextraction coupled with gas chromatography-mass spectrometry (SPME-GC-MS) to investigate changes in VOC profiles over time and under different storage conditions. A total of 39 metabolites were associated with TSD. This study compared the types, abundances, and temporal patterns of VOCs among three substrates: gauze, wood, and glass. Based on 13 characteristic VOCs commonly detected in dried bloodstains across these three substrates within a 7-day period, a machine learning regression model was developed and evaluated. The optimal model was a random forest (RF) model, achieving root mean square error (RMSE), coefficient of determination ( R 2 ), and mean absolute error (MAE) values of 0.8217, 0.9401, and 0.5103, respectively, for the validation set, and 0.7742, 0.8533, and 0.5437, respectively, for the external validation set. These findings demonstrated that volatolomics analysis has strong potential for estimating bloodstain TSD, offering reliable biomarkers, high model accuracy, and robustness to substrate effects. This study provides a new methodological framework for TSD estimation and lays the foundation for further research in this field.

Steroidal estrogens are a type of compound that disrupts endocrine function and could potentially threaten food safety and human health. So, there is a real need for a sensitive and convenient method to detect estrogens in food samples. Magnetic silica (Fe 3 O 4 @SiO 2 ) has been recognized as a versatile adsorbent material with a porous structure that provides numerous imprinting sites while reducing mass transfer resistance, making it a promising candidate for adsorption applications. Fe 3 O 4 @SiO 2 denotes core-shell nanoparticles with magnetite as core and silica as outer shell. However, to further enhance the adsorption capacity and selectivity, the incorporation of metal-organic frameworks (MOFs) can be highly beneficial. In this work, estrogens were detected using molecularly imprinted polymers (MIPs) based on composites of Fe 3 O 4 @SiO 2 and aluminum metal-organic framework (Al-MOF). The Al-MOF, specifically MIL-68(Al), was selected for its high surface area, tunable pore size, and strong stability, which contribute to its excellent adsorption properties. The incorporation of MIL-68(Al) into the Fe 3 O 4 @SiO 2 matrix not only increases the available surface area but also provides additional specific binding sites for estrogens, resulting in enhanced selectivity and sensitivity. The resulting Fe 3 O 4 @SiO 2 @MIL-68(Al)@MIP material proved to be a powerful adsorbent for estrogens with a maximum adsorption capacity of 34.52 mg g −1 . Notably, Fe 3 O 4 @SiO 2 @MIL-68(Al)@MIP exhibited high adsorption capacity and selectivity and achieved a limit of detection (LOD) of 1.1–4.6 ng mL −1 (0.04–0.18 µg kg −1 ) and a linear response range of 40.96–8000 ng mL −1 , with recoveries between 87.34% and 99.86%, and relative standard deviations (RSDs) ≤ 5.92%. These impressive results are attributed to the synergistic effect of the magnetic core, porous silica shell, and high-affinity MOF, making the material a highly efficient sorbent for the pretreatment of estrogenic veterinary drugs in traditional Chinese medicine. Background Insoluble dietary fiber (IDF) from Actinidia arguta is a valuable functional food ingredient, but its inherent dense structure severely limits its functional properties and industrial application potential. In this study, IDF obtained via enzymatic extraction was used as the raw material to investigate the improvement effects of physical modification technologies on its comprehensive performance. Results Four physical modification methods, including microwave, ultrasound, high-pressure moist heat, and high hydrostatic pressure treatments, were applied. All treatments effectively disrupted the tight compact structure of unmodified IDF and promoted the exposure of internal active groups, thus significantly enhancing the structural and functional properties of the fiber ( p < 0.05). Among the modified samples, microwave-modified IDF (WIDF) exhibited a well-developed honeycomb network structure and the best overall performance. Its water-holding, oil-holding, and swelling capacities increased approximately 1.5– to 2.1-fold compared with the unmodified IDF. The antioxidant activities were also considerably improved, with the hydroxyl radical scavenging rate showing the highest increase of 2.4-fold. In terms of adsorption capacity, WIDF showed a 4.7-fold increase in sodium cholate adsorption, and also exhibited enhanced binding capacities for cholesterol, glucose, nitrite, and heavy metals in a pH-dependent manner. Conclusions Microwave-modified IDF holds promising potential as a multifunctional food ingredient. This study provides a feasible approach for the high-value valorization of fruit processing by-products, which is conducive to promoting the sustainable development of the food processing industry. The mechanism of unconventional protein secretion remains an unresolved issue. Here, we describe an unconventional protein secretion pathway for galectin-3 that is mediated by phase separation and condensation. Using four lysosomal damage models, we observed a rapid, pronounced release of galectin-3 in large, non-exosomal particles. This secretion is driven by glycoprotein-induced galectin-3 phase separation and is independent of pyroptosis and secretory autophagy. During phase separation, the S-face of galectin-3 carbohydrate recognition domain binds glycoproteins that triggers galectin-3 N-terminal tail release and condensation. These condensates then recruit ALG-2 via the exposed N-terminal tail. ALG-2 directs the condensates to the endoplasmic reticulum–late endosome interface. After translocation into late endosomes, galectin-3 condensates are secreted into the extracellular milieu by SNARE-dependent vesicular transport. This mechanism of exporting phase-separated protein condensates may serve as a clean-up response to membrane damage.

The catalytic upcycling of plastic mixtures into H 2 and carbon nanotubes (CNTs) represents a transformative strategy for advancing plastic management and the circular economy. Herein, an in-situ constructed high-performance FeNi/Ni/C catalyst combined with prompt microwave catalysis enables efficient conversion of LDPE-PVC, LDPE-HDPE-PP-PS-PVC-PTFE and landfilled waste mixture into valuable H 2 and CNTs, resulting in near-quantitative atom economy (i.e., H 2 and carbon efficiencies are nearly 100%) together with H 2 and CNTs yields reaching >922 mmol g -1 catalyst and >10600 mg g -1 catalyst , respectively. Furthermore, this microwave catalysis strategy presents an excellent stability (>35 cycles) via Cl/F-based regeneration and energy efficiency (80% reduction vs. traditional thermal catalysis), alongside a high turnover number of 107.6 mol carbon mol -1 NiFe . Fundamentally, the Ni species contribute to C-H breaking, and Fe is beneficial for CNT formation during plastic decomposition. Here, we show a cost-effective platform for upcycling of chlorinated/fluorinated plastic mixtures, aligning with resource circularity objectives.

Fe-based polyanionic cathodes are promising for large-scale Na-ion batteries owing to their stability, safety and elemental abundance, however their capacity remains limited by electrochemically inactive Na sites and irreversible Na loss. Here we identify that the Na + coordination environment critically influences the Na-site accessibility and redox activity in Na 4 Fe 3 (PO 4 ) 2 P 2 O 7 -type cathodes. Combined experimental and theoretical analyses reveal that precise V 3+ substitution at the Fe2 site harmonizes Na + coordination geometry and softens the polyanionic framework, thereby activating previously inert Na sites and stabilizing high-voltage redox reactions above 4 V. The optimized Na 3.4 Fe 2.4 V 0.6 (PO 4 ) 2 P 2 O 7 achieves full Na + utilization (3.4 Na + , 150.7 mAh g −1 ) and a 52% increase in energy density (487 Wh kg −1 ), approaching the practical limit of Li-ion phosphate cathodes. It also demonstrates exceptional durability over 10,000 cycles in the 2.1-4.5 V range and stable pouch-cell performance. These findings provide a coordination-based strategy to overcome intrinsic capacity limitations in phosphate cathodes, enabling high-energy, durable Na-ion batteries.
