Ruddlesden-Popper type Sr(La,Gd)2Al2O7:Cr3+ layered perovskite new phosphors for NIR pc-LED application

Zhiyuan Pan,Sihan Feng,Fan Li,Xuejiao Wang,Qi Zhu,Ji-Guang Li

Near-infrared phosphor converted light-emitting diodes (NIR pc-LEDs), as a state-of-the-art type of NIR sources, are finding multifaceted applications, where phosphor is a crucial component. In hope to provide new choices for NIR lighting, we synthesized in this work the two series of Ruddlesden -Popper type layered perovskite phosphors of SrLa 2 Al 2- x O 7 : x Cr 3+ ( x  = 0–0.02) and SrLa 2- y Gd y Al 1.984 O 7 :0.016Cr 3+ ( y  = 0–2), followed by detailed structure and luminescence investigation. SrLa 2 Al 2- x O 7 : x Cr 3+ was found to emit ∼700–800 nm NIR light under UV excitation and be able to maintain ∼54.3 % of its room temperature intensity at 423 K, which is remarkably higher than those (∼25–30 %@423 K) of the Mn 4+ activated perovskite NIR phosphors. Luminescence regulation was achieved by substituting La 3+ with Gd 3+ , and it was revealed that Gd 3+ may act as a sensitizer to significantly enhance Cr 3+ luminescence (∼346 % at 77 K) through Gd 3+ → Cr 3+ energy transfer, which was confirmed by spectral and fluorescence lifetime analysis. The observed spectral features and the influence of Gd 3+ were rationalized by considering crystal structure, local coordination, and crystal field. Finally, an NIR pc-LED device showing strong ∼700–900 nm luminescence was fabricated to show potential application of the phosphor in night vision and plant lighting.

Quintuple Free-Radical Therapy: An Ultralong-Retention FAND for NIR-Involved Multiple Site-Acting Hypoxic Tumor Therapy

Chuang Zhang,Dongxu Zhao,Fang Fang,Lin Zhu,Weiyu Li,Sa Wang,Yueyun Fan,Jiani Yang,Yanhong Liu,Jinfeng Zhang

The heavy dependence on intratumoral O 2 and H 2 O 2 availability has greatly restricted ROS-based therapy. Although O 2 /H 2 O 2 -irrelevant free-radical nanogenerator has garnered tremendous attention as a promising anticancer candidate to overcome the above intrinsic limitations of ROS-based treatment, the practical therapeutic efficacy of free-radical therapy is still hindered by limited free-radical type and inferior tumor retention performance. Herein, inspired by the new concept of full-API nanodrug (FAND) with 100% active pharmaceutical ingredient (API) content and repurposing of clinical anti-malaria drug artesunate (ARTE) as an anticancer free-radical generator, the AFeI FANDs composed of ARTE, human essential Fe 3+ , and FDA-approved fluorescent agent ICG for hypoxic tumor therapy are rationally designed and constructed. Attractively, the completely pharmaceutically active components ARTE, Fe 3+ , and ICG can be responsively liberated in the acidic tumor microenvironment and synergistically produce five types of free radicals including •O 2 − , •C, •OH, LOO•, and 1 O 2 , leading to robust mitochondrial injury, nuclear DNA damage, and lipid peroxides. More importantly, the AFeI FANDs displayed ultralong tumor retention longer than 108 h and favorable tumor suppression outcomes under mild NIR irradiation. Collectively, the presented first paradigm of FAND-based quintuple free-radical therapy expands new horizons for the development of clinically transferable nanomedicine for tumor therapy.

Multiple amplifications of NIR-I ECL signals of Au6PtAg nanoclusters and applications in fast detections of noradrenaline

Fanfan Guo,Yuanyuan Hu,Huiwen Zhou,Yuanjin Li,Jiadi Li,Guichuan Wei,Jing Long Han,Wenyue Gao,Sisi He,Wenxin Niu,Ling Zhang

NIR-I electrochemiluminescence (ECL) is emerging as highly sensitive detection strategy due to its low background at the ambient conditions. Nonetheless, the applications of current NIR-I ECL nanoprobes are limited to the harsh synthetic/testing condition, high toxicity, low signal, and poor biocompatibility. In this study, glutathione/lipoic acid-protected trimetallic Au 6 PtAg nanoclusters (GSH/LA/Au 6 PtAg NCs) are demonstrated as an efficient nanoprobe with strong ECL signals around 850 nm in the presences of triethylamine. Critically, the alloying of Ag  +  into the bimetallic Au 2.5  Pt NCs altered the electronic structures of Pt(II), valences of Au(0), and the zata potentials of NCs, promoting the ECL intensities at 850 nm by 1.0 time. Additionally, the partly ligand-exchange of GSH by LA further increased the ECL intensities at 850 nm by 2.2 times through accelerating electron transfer processes. Meanwhile, the NIR-I fluorescence ( λ emit  = 910 nm) was enhanced. More importantly, the utilization of GSH/LA/Au 6 PtAg NCs μg −1 is enhanced by 3000 times after embedding into SiO 2 microspheres ( D , 318 nm). Because noradrenalin has substantial inhibition effects to NIR-I ECL of GSH/LA/Au 6 PtAg/SiO 2 , 1.0 nM−0.1 mM noradrenalin was detected with the low limit of detection as 0.04 nM (S/N = 3) selectively. Biomolecules, glucose, ascorbic acid, or uric acid didn't interrupt the detection of noradrenalin. The sensitivity of this ECL method to detect noradrenalin is much higher than those in previous reports, enabling the early diagnosis of neurogenic diseases.

PLLA/(MWCNTs-ZnO)@PDA composite with NIR-light induced shape memory effect, antibacterial properties and 3D printability

Yulong Zou,Beibei Zhou,Wenqiang Guo,Zhijie Sun,Xili Lu,Li Li

PLLA based shape-memory polymer composites with multifunctions have attracted growing attention in biomedical applications. Generally, a single nanofiller has the problem of insufficient functionalization and easy aggregation. Therefore, we design and fabricate polydopamine (PDA) surface modified MWCNTs-ZnO hybrid nanostructure ((MWCNTs-ZnO)@PDA). The dispersion of (MWCNTs-ZnO)@PDA in PLLA is improved in comparison with the pristine MWCNTs or ZnO. The results indicate that the (MWCNTs-ZnO)@PDA nanosystem shows a synergistic effect on the mechanical properties and photothermal properties of the composites. Comparative analysis with neat PLLA demonstrates a remarkable 17% increase in tensile strength and a 19% increase in elongation at break for PLLA/(MWCNTs-ZnO)@PDA composite. Additionally, the composites exhibit excellent NIR light-induced shape memory effects with a shape recovery ratio reaching 100%. Moreover, PLLA/(MWCNTs-ZnO)@PDA composites show favorable antibacterial activity against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ). Furthermore, the polymer composites display good 3D printability, and the 3D-printed porous scaffold possesses high compressive strength and modulus. Also, the PLLA/(MCNTs-ZnO)@PDA scaffold can be compressed into a compact size and recover its original shape under NIR light irradiation. This work provides an approach to prepare novel shape-memory polymer composites with multifunctions including high strength, light-induced shape-recovery, antibacterial properties and printability.

Long-term dynamic tracking of acute kidney injury by Au nanoclusters-based reversible NIR-II fluorescent probe

Qing Hu,Fanyi Qin,Huijiao Qu,Huayu Xiong,Shengfu Wang,Xiuhua Zhang,Yan Xiao

Acute kidney injury (AKI) is a common kidney disease characterized by a sudden decline in renal function. It usually has no clear clinical symptoms in its early stages, but can easily progress to renal failure or even result in death. Real-time monitoring of AKI progression significantly enhances the prospects for early diagnosis and prompt intervention. However, there is no available to monitor AKI as developing a renal-clearable and reversibly responsive probe capable of long-term in vivo track biomarkers of AKI, such as reactive oxygen species (ROS), remains an extremely challenging task. Herein, we have constructed a redox-reversible second near-infrared (NIR-II) fluorescent probe based on Au nanoclusters (Au NCs) for long-term dynamic monitoring of AKI. The probe features NIR-II excitation and emission capabilities, along with effective kidney targeting and prolonged retention, facilitating renal enrichment and enabling long-term high-resolution renal imaging. Notably, it demonstrates rapid and reversible responsiveness to ROS with high sensitivity and selectivity, permitting real-time monitoring of redox fluctuations in vivo. Given its exceptional performance, the probe achieves unprecedented real-time dynamic tracking of AKI progression, providing crucial redox status information during the disease process. Additionally, the probe can serve as an effective new tool for screening of potential therapeutic drugs for AKI. This work may offer some substantial promotion for the diagnosis and treatment of kidney diseases.

Upconverted/downshifted NaLnF4 and metal-organic framework heterostructures boosting NIR-II imaging-guided photodynamic immunotherapy toward tumors

Qi-Xian Wang,Yuan-Fan Yang,Xiang-Fei Yang,Yue Pan,Ling-Dong Sun,Wen-Yuan Zhang,Yulei Shao,Jie Shen,Jian Lin,Lele Li,Chun-Hua Yan

Heterostructures of NaLnF 4 and porphyrinic metal-organic frameworks (MOF) have been reported as a promising platform for near-infrared (NIR)-driven photodynamic therapy (PDT), while the energy transfer from NaLnF 4 to MOF greatly decreased the upconversion emissions. To develop NaLnF 4 and MOF heterostructures with significantly improved PDT and imaging simultaneously is important to track their location to implement therapy accurately. Herein, we constructed Er 3+ -doped NaLnF 4 @MOF [email protected] nanoparticles to integrate NIR-driven PDT and NIR-II imaging. NaLnF 4 nanoparticles are designed with upconversion and NIR emission simultaneously under a single 980 nm excitation, where the upconversion luminescence (UCL) overlaps well with the absorption of Zr-based porphyrin MOF, enabling efficient energy transfer from NaLnF 4 to the MOF shell to generate 1 O 2 . And the NIR luminescence, which gave out under a lower excitation power than that of UCL, could be used to track the location and distribution of the heterostructures via NIR-II imaging. Moreover, antiprogrammed death-ligand 1 (α-PD-L1) immunotherapy was introduced synergistically to improve antitumor immunity efficacy. The combination of NaLnF 4 @MOF and α-PD-L1 not only inhibited primary tumors but also suppressed distant one, leading to an effective tumor inhibition rate of 95%. This work provides an integrated nanoplatform for boosting NIR-II imaging-guided PDT/immunotherapy efficacy toward tumor treatment.

IC-MAC: A novel NIR fluorescent probe for monitoring the glutathione fluctuations in drug-induced liver injury

Xiao Li,Chengqing Zhao,Zhiyuan Wang,Suntao Shi,Huiling Gao,Jiang Wu,Baoxin Zhang

Drug-induced liver injury (DILI) is widely recognized as a major threat to global public health, posing significant obstacles to clinical treatment and drug innovation. The core mechanism underlying DILI progression is the depletion of intracellular glutathione (GSH), which is widely regarded as a key marker of oxidative liver injury and an important biomarker for early-stage DILI. In this study, we developed IC-MAC , a novel near-infrared (NIR) fluorescent probe for the highly sensitive and selective detection of GSH, which exhibited excellent photostability, a large Stokes Shift, and a significant fluorescence enhancement upon reaction with GSH. The probe demonstrated high biocompatibility and enabled real-time imaging of intracellular GSH fluctuations under physiological and oxidative stress conditions. In a mice model of acetaminophen-induced DILI, IC-MAC enabled dynamic visualization of GSH depletion in the liver and its recovery following therapeutic intervention. Fluorescence intensity was significantly correlated with the degree of GSH depletion and treatment response, providing robust evidence for the quantitative assessment of liver injury severity and drug efficacy. Collectively, these findings highlighted the potential of IC-MAC as a powerful platform for early DILI diagnosis and in vivo redox monitoring.

Controlled co-release of doxorubicin and reactive oxygen species for synergistic therapy by NIR remote-triggered nanoimpellers

Beibei Hou,Weitao Yang,Chunhong Dong,Bin Zheng,Ying Zhang,Jing Wu,Hanjie Wang,Jin Chang

How to encapsulate and transport the payload of multiple therapeutic compounds avoiding premature leakage, and simultaneously co-release them rapidly at specific lesions still remains the major concern in clinic. Herein, we designed the UCN@mSiO 2 -(Azo   +   RB) (azobenzene groups and Rose Bengal) nanoimpellers, which used the multicolor-emission capability of the core-shell upconverting nanoparticles (UCNs) at a single excitation wavelength to co-release anticarcinogen doxorubicin (Dox) and reactive oxygen species (ROS) for combined chemotherapy and photodynamic therapy (PDT). The nanoimpeller was formed from UCN inner core, mesoporous silica shell, and light triggers Azo and RB molecules. The UCNs emitting UV/blue and green/red multiband light were used to activate the photoresponsive Azo and photosensitizer RB molecules; The mesoporous silica shell offered the possibilities to load anticancer drug and conjugate the light triggers; As there are strong charge interaction and hydrogen bonds between Dox and surface silanols of mesoporous silica, the azobenzene molecules worked as “gatekeeper” and “molecular stirrer” to precisely trap and propel the release of Dox under the external stimuli. The time-dependent drug release analysis, ROS production test and PDT test suggested that the nanoparticles may serve as a useful multifunctional nanoplatform for synergistic therapy and cancer diagnostic.

Polypeptide nanoparticles obstruct glucose supply for NIR-II fluorescence-guided tumor starvation and enhanced mild photothermal therapy

Yating Wang,Yixuan Xu,Dejia Chen,Xin Li,Dalong Yin,Lifeng Yan

Photothermal therapy (PTT) utilizes localized hyperthermia above 50 °C generated by nanomaterials upon exposure to near-infrared (NIR) laser light for effective cancer cell eradication. Yet, in high-temperature PTT, tumor cells develop heat stress tolerance due to elevated heat shock protein (HSP) levels, diminishing therapeutic efficacy. Moreover, excessive heat can trigger inflammatory conditions and promote cancer metastasis. Targeting the glycolytic pathway in highly metabolically active tumor cells offers a promising strategy for inducing starvation therapy, capitalizing on their vigorous energy demands amidst rapid proliferation. Here, we synthesized a highly luminous NIR-II dye, FNF, followed by its encapsulation alongside myricetin (My) within amphiphilic polypeptide carriers through a self-assembly method. The resulting nanoparticles exhibited great NIR-II imaging capabilities and boasted a notable photothermal conversion efficiency of 55.58 %. Furthermore, My effectively impeded glucose transport facilitated by glucose transporter protein 1 (GLUT1), curtailing glucose supply to tumor cells. This interference disrupted mitochondrial energy production, resulting in decreased adenosine triphosphate (ATP) synthesis and subsequent downregulation of HSP70 expression. By leveraging this approach, which targeted HSP expression via GLUT1 inhibition, we enhance the efficacy of PTT while achieving a synergistic effect for mild photothermal therapy through starvation. Statement of significance High expression of heat shock proteins (HSPs) in cancer cells impairs the efficacy of photothermal therapy (PTT) and triggers inflammation or metastasis, among other effects. Rapid malignant proliferation of tumor cells results in high energy metabolism, so interfering with their glucose metabolism to inhibit the glycolytic process is a feasible route for tumor starvation therapy. Here, we employed an amphiphilic polypeptide encapsulated photosensitizer (FNF) and myricetin (My) to construct nanoparticles with both NIR-II imaging capability and high photothermal conversion efficiency (55.58 %). Among them, My blocked glucose transport mediated by glucose transporter protein 1 (GLUT1), reduced the glucose supply and ATP synthesis in cancer cells, and then down regulated HSP70 expression. Thus, this strategy achieves starvation synergistic mild photothermal therapy through metabolic disruption.

Sn-Ag2S quantum dots with enhanced photoluminescence quantum yield in the NIR-II window for bone imaging

Qing Li,Ning Liu,Dayu Wang,Lie Ma,Mingzhou Wu,Enna Ha,Junqing Hu,Zhe Zhang,Shuqing He

Silver sulfide (Ag 2 S) quantum dots (QDs), which emit light in the near-infrared II window (NIR-II, 1000–1700 nm), exhibit unique and remarkable potential in bioluminescence. However, their widespread application in experimental research is currently limited by a relatively low quantum efficiency. Cation exchange reactions have emerged as a remarkably versatile approach for synthesizing nanomaterials with adjustable structures and customizable compositions. In this study, we developed a NIR-II fluorescent probe based on Sn-doped silver sulfide (Sn-Ag 2 S) QDs via a straightforward cation exchange method. The stoichiometric ratios of precursors play a crucial role in tailoring the optical properties of the resultant materials during structural modifications involving cation exchange processes. By optimizing the ratio of Ag and Sn, we obtained ca. 16-fold quantum yields enhancement of Sn-Ag 2 S QDs in the NIR-II range, specifically around strong emission peak at ca. 1200 nm. Following hydrophilic modification, the Sn-Ag 2 S-GSH/MPA QDs exhibited high NIR-II fluorescence signal signal-to-noise ratio (S/N) with a ca. 1.7-fold increase compared to Ag 2 S-GSH/MPA QDs in the femur of mice, showing significant potential for bone imaging. To further investigate the QDs biodistribution mechanisms within osseous microenvironments, we devised a comprehensive multimodal analytical approach incorporating: (1) optical spectral analysis, (2) ultrastructural validation through transmission electron microscopy (TEM), and (3) spatial resolution mapping via laser scanning confocal microscopy (LSCM). These integrated methodologies consistently demonstrated preferential QDs accumulation within trabecular bone marrow compartments, revealing distinct biodistribution patterns in mineralized tissues.

CD44 and αV-Integrins Dual-targeting Bimetallic Nanozymes for Lung Adenocarcinoma Therapy via NIR-Enhanced Ferroptosis/Apoptosis

Jingchuan He,Tingting Li,Xiaoqin Pan,Zhihua Deng,Jifu Huang,Xiaocheng Mo,Xiaoju Shen,Xiumei Qin,Xingye Yang,Ming Gao,Jie Yang

Combination therapy is a promising strategy for lung adenocarcinoma (LUAD), due to the advantages of overcoming drug resistance, side effects, and tumor heterogeneity. Herein, we report a novel dual-targeting bimetallic nanozyme (MH-iRGD) consisting of nanosized manganese ferrite (MF) after encapsulating with dopamine and methacrylic anhydride to modify hyaluronic acid, followed by integrin receptor targeting peptide (HS-PEG3400-iRGD) modification for LUAD targeted therapy. Our study confirmed that MH-iRGD combined with near-infrared irradiation (NIR) possessed dramatic photothermal effects and reactive oxygen species (ROS) production and GSH depletion abilities. Importantly, MH-iRGD possessed dual-targeting capacities for LUAD cells overexpressed CD44 and αV-integrin receptors owing to hyaluronic acid coating and iRGD modification. Inhibitors of CD44 and integrins could impair the uptake of MH-iRGD in LUAD cells. Moreover, MH-iRGD+NIR displayed excellent anti-LUAD effects as a result of the production of intracellular ROS, consumption of glutathione (GSH) and mitochondrial dysfunction. Mechanistically, NIR robustly strengthened MH-iRGD-induced ferroptosis and apoptosis by down-regulating SLC7A11, GPX4, Bcl-2 levels while up-regulating Bax level. Specifically, ferroptosis and apoptosis were increased while the LUAD progression was inhibited after intravenous injection of MH-iRGD+NIR in xenograft mouse models. Taken together, our results indicate that MH-iRGD+NIR serves as a promising targeted therapy for LUAD, which broadens the applications of highly active dual-targeting bimetallic nanozymes.

Crystal Chemistry and NIR Spectral Responses of Fe/Mg-Rich Smectites: Implications for Clay Detection on Mars

Qi Tao,Yonglei Zhang,Tianqi Zhang,Xiaorong Qin,Binglong Ye,Hongping He,Sridhar Komarneni

Over the past decade, remote sensing exploration has revealed widespread detection of clay minerals in Noachian terrains (>3.7 Ga) on Mars, suggesting a period of active near-surface water–rock interaction. The in-depth characterization of the crystal structure and chemistry of laboratory-synthesized smectite minerals with varying compositions offers a unique perspective to further constrain their formation mechanism, environmental conditions, and the climate of early Mars. This study synthesized smectites with varying Fe/Mg ratios to compare their near-infrared (NIR) spectral characteristics with those of representative areas on Mars. The study revealed that the characteristic metal–OH (M–OH) band in the range of 2270–2320 nm shifted toward longer wavelengths as the octahedral Fe3+ content decreased. When the octahedral Fe/Mg ratio was close to 1, the position of M–OH was similar to that of Mg-smectite (saponite), indicating that although the M–OH band of smectite is located at ca. 2308–2312 nm, it may still contain a significant amount of octahedral Fe3+. This suggests that there are likely still many Fe-smectites in Mg-rich smectite outcrops (ca. 20% as in the literature) on the Martian surface. Furthermore, correlation analysis between the smectite layer stacking order index (V/P) and the second-order derivative band intensity ratio (Deriv2 ratio) showed that when the Deriv2 ratio is small (generally <8), smectite has a relatively high degree of layer stacking order. When the Deriv2 ratio is >10 and tends toward infinity (+∞), the layer stacking order of smectite is very low. The above results suggest that the crystal chemical characteristics of smectites should be carefully studied when analyzing the mineral composition of the Mars surface using remote sensing or in situ NIR spectra. Smectites on Mars generally exhibit a high layer stacking order, indicating the presence of a relatively long-term warm and humid climate in Martian history. However, later alteration processes can significantly reduce their crystallinity.
