Osteomyelitis is a devastating bone disease caused by bacterial infection, the treatment of which remains challenging due to bacterial biofilm formation and impaired bone regeneration. The development of an integrated therapeutic platform capable of locally filling bone defects, enabling controlled drug release and modulating the infectious microenvironment is of crucial importance. This study constructed an injectable self-healing hydrogel based on Schiff base bonds for pH-triggered delivery of ZIF-8-loaded curcumin, aiming to achieve synergistic therapy for osteomyelitis. Curcumin-encapsulated zeolitic imidazolate framework-8 nanoparticles were synthesized via a one-pot method and embedded into an oxidized starch–gelatin hydrogel network formed by dynamic Schiff base crosslinking, yielding the composite hydrogel. Systematic characterization revealed that the ZIF-8@Cur nanoparticles exhibited a well-defined rhombic dodecahedral structure, high crystallinity and a curcumin encapsulation efficiency of 85%. The composite hydrogel demonstrated excellent injectability, rapid self-healing behavior and remarkable pH-responsive drug release: under the acidic condition mimicking an infectious microenvironment (pH 5.5), the cumulative release over 72 h reached 85%, significantly higher than the 25% observed at physiological pH 7.4. In vitro antibacterial assays confirmed its potent broad-spectrum antibacterial activity against both Gram-positive and Gram-negative bacteria ( > 98% inhibition). More importantly, the system effectively promoted macrophage polarization from pro-inflammatory M1 to anti-inflammatory and reparative M2 phenotypes while exhibiting good biocompatibility. This study successfully developed an intelligent responsive injectable hydrogel delivery system integrating efficient antibacterial, anti-inflammatory and immunomodulatory functions, offering a promising strategy for the comprehensive local treatment of osteomyelitis.

Fabrication of indicator array based on varying proportions of curcumin and alizarin. • Composite films exhibited a lower water solubility compared with pure PVA films. • The detection limit of indicator array for volatile amine gases was down to 1 ppm. • Color alterations of the array strongly correlated with pork spoilage indexes. In this work, a synergistic polyvinyl alcohol (PVA)/zein electrospun nanofiber array was fabricated using curcumin and alizarin as dual responsive pigments for visual pork freshness monitoring. The water solubility of the PVA/zein composite reduced from 100.00% to 31.74%–35.42% ( P < 0.05), exhibiting remarkably enhanced water resistance. The dual-pigment array displayed continuous color transitions from pH 4.0 to 11.0 and detected ammonia and dimethylamine at 1 ppm, whereas alizarin-only films needed 5 ppm. The total color difference ( ΔE ) of the array showed strong linear correlations with thiobarbituric acid reactive substances (TBARS, r = 0.98), pH ( r = 0.95), and total volatile basic nitrogen (TVB-N, r = 0.98) in pork during refrigerated storage. Notably, the array enabled visual grading of pork freshness (fresh/sub-fresh/spoiled) and exhibited excellent color stability under ambient and refrigerated conditions. This study presents an eco-friendly and scalable intelligent packaging platform with great potential for real-time, on-site food freshness assessment. In this study, a colorimetric array based on polyvinyl alcohol-zein composite substrates and varying ratios of curcumin and alizarin as responsive materials was fabricated via electrospinning for monitoring pork freshness during refrigeration. Rheumatoid arthritis (RA) therapy demands agents that are both effective and safe, yet many natural products with promising bioactivity, such as curcumin, face major translational challenges due to poor solubility and bioavailability. To overcome these limitations, we designed and synthesized a series of dual-functional curcumin-borneol prodrugs via a succinate ester linkage, including the parent ester (CBS), its water-soluble sodium salt (CBS-Na), and a disubstituted analog (DCBS), to enhance delivery and synergize therapeutic action. These derivatives markedly improved cellular uptake and nuclear localization, potently scavenged reactive oxygen species, and effectively suppressed the MAPK/AP-1 signaling axis by inhibiting ERK and p38 phosphorylation and c-Fos expression, leading to downregulation of matrix metalloproteinase MMP9 and pro-inflammatory cytokines (TNF-α, IL-6, IL-1β). In a collagen-induced arthritis rat model, the derivatives demonstrated superior efficacy over native curcumin in alleviating joint inflammation and destruction, while exhibiting an excellent safety profile, thus representing a novel and promising therapeutic strategy for RA.

An asymmetric adhesive hydrogel is developed. • The sprayable and biodegradable hydrogel possesses antibacterial and antioxidant properties. • The hydrogel possesses excellent hemocompatibility and cytocompatibility, and can promote cell migration. Injectable hydrogel adhesives with asymmetric adhesion properties are of great significance in reducing the risk of postoperative tissue adhesion. In this study, a sprayable, in situ–forming hydrogel was synthesized by grafting 2-aminoethyl methacrylate (AEMA) and dopamine (DA) onto a sodium alginate (SA) backbone, followed by loading curcumin-PF127 micelles (Cur-PF127) and ultraviolet (UV)- initiated free-radical crosslinking. The prepared hydrogel exhibited porous microstructure, sprayability, degradability, and suitable swelling properties (>1000%). Rheological analysis revealed that the hydrogel exhibited stable storage and loss moduli. In addition, the hydrogel exhibited good adhesion to diverse substrates, including glass, rubber, plastic, porcine skin, and various mouse organs (liver, lung, stomach, spleen, kidney, and heart). Notably, the adhesive performance of the hydrogel could be reduced after UV exposure, enabling controllable adhesion behavior. Furthermore, the hydrogel exhibited antibacterial efficacy against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ), potent antioxidant activity (scavenging over 80% of free radicals), and good in vitro cytocompatibility, as validated by the CCK-8 assay, cell staining, the cell scratch test, and hemocompatibility analysis. Owing to its combination of controllable adhesion, sprayability, antioxidant and antibacterial properties, and cytocompatibility, this hydrogel shows potential as a multifunctional biomedical adhesive. Infection and excessive oxidative stress are major challenges in wound healing. Herein, quaternary electrospun membranes were fabricated using polylactic acid (PLA) and microcrystalline cellulose (MCC) as the matrix, with zinc oxide (ZnO) and curcumin (Cur) as functional additives to achieve dual antibacterial and antioxidant functions. MCC acted as a structural reinforcer and hydrophilic modifier to improve the mechanical properties and hydrophilicity of the PLA matrix, while ZnO and Cur served as synergistic bioactive components to endow the membranes with outstanding antibacterial and antioxidant capacities. ZnO-only groups (Z1 and Z2) exhibited moderate antibacterial and antioxidant activities. In contrast, the incorporation of Cur remarkably boosted these effects. The Z1C3 sample displayed a DPPH radical scavenging rate of 64.5%, alongside antibacterial efficiencies of 71.27% against Escherichia coli and 92.04% against S. aureus . Further elevation of ZnO and Cur loading yielded superior bioactivity; Z2C5 achieved antibacterial rates of 77.38% against E. coli , 95.68% against S. aureus , and a DPPH scavenging ratio of 89.98%. These results indicate that ZnO and Cur loading can be adjusted to balance bioactivity and safety. From the biosafety perspective, Cur greatly improved biocompatibility and reduced the hemolysis rate below 5%. Overall, the PLA/MCC/ZnO/Cur nanofibrous membranes hold promise as functional wound dressing materials, offering a potential strategy for the design of sustainable and multifunctional wound care materials. Osteoporosis is one of the most common chronic metabolic diseases that mainly caused by an imbalance in bone homeostasis. Considered its severe concurrent disorders, osteoporosis has become a global health issue, however, the anti-osteoporosis medications available in clinical practice are confronted with challenges such as high costs, adverse side effects, and limited efficacy. Herein, we reported construction of pH-responsive and bone-targeting nanomedicine (termed HPGCA) via click conjugation of alkyne functionalized hyperbranched polyglycerol (HPG) with curcumin (Cur) and alendronate (ALN) for the first time. We demonstrated that HPGCA exhibit superior water dispersibility, small particle size, and pH-dependent drug release behavior. Results from biological assays indicated that HPGCA displayed desirable bone-targeting capability, strong anti-osteoporosis efficacy, good hemocompatibility, and negligible adverse effects. More importantly, HPGCA showed better anti-osteoporosis efficacy than both free Cur and ALN according results from micro-CT and TRAP staining. The synergistic osteoporosis therapeutic efficacy would be explained by targeting delivering HPGCA to bone tissues and in-situ releasing cargos under acidic environment. Therefore, considered the above aspects, we believe that the innovative nanomedicine is promising for osteoporosis therapy in clinic and will provide new insights for full realization anti-osteoporotic properties of natural compounds

This study elucidates the mechanism of curcumin encapsulation in soy protein isolate (SPI) modified via enzymatic deamidation and pH-shifting. A novel Chryseobacterium sediminis -derived glutaminase (CS-PG) demonstrated superior thermal stability, catalytic efficiency, and specificity over commercial CP-PG. Controlled deamidation by both enzymes enhanced SPI functionality (solubility/emulsification/foaming) with minimal hydrolysis, where CS-PG induced hydrophobic self-assembly and CP-PG promoted hydrophilic domain formation. These distinct conformations dictated binding modes: CS-PG-SPI achieved 92.97 % higher encapsulation efficiency versus SPI through hydrophobic encapsulation, while attained an 85.00 % increase via surface-layer stabilization. pH-shifting further altered SPI conformation by inducing reversible unfolding and refolding, enabling formation of amorphous, stable protein–curcumin complexes. Dual modification significantly enhanced bioaccessibility versus free curcumin—1034 % for CS-SPI–Cur–pH and 1058 % for CP-SPI–Cur–pH. The work establishes enzymatic deamidation as a precision tool for tailoring SPI self-assembly and bioactive delivery in functional foods.

In infected full-thickness skin injuries, deep wound exposure accompanying bacterial invasion and persistent inflammatory lead to prolonged wound healing, remaining a major challenge in clinical wound healing. In this work, an emulsion gel (SCEG) with pH/enzyme-responsive release for promoting infected wound repair was successfully constructed based on the Spirulina protein isolate/carboxymethyl chitosan (SPI/CMC) system. The SCEG was prepared by cross-linking the SPI/CMC emulsion with tetrakis (hydroxymethyl) phosphonium sulfate (THPS) through the Mitchell reaction. It effectively encapsulated the curcumin (Cur) with an encapsulation efficiency of 87.26 % and exhibited pH/enzyme-responsive release behavior. The SCEG showed excellent antibacterial activity against both S. aureus and E. coli . Additionally, it can evidently alleviate oxidative stress and maintain cell activity. Meanwhile, it could significantly reduce the secretion of pro-inflammatory cytokines TNF-α and IL-1β. Finally, a model of full-thickness skin damage infected with S. aureus was built to assess the wound healing effect of SCEG. The result showed that it can efficaciously inhibit bacterial infection of the wound, reduce inflammatory response, promote angiogenesis, accelerate tissue regeneration, and thus boost the healing process. Altogether, the emulsion gel dressing is expected to be a safe, efficient and promising candidate material for the field of infectious wound care.

This study aims to develop a novel chitosan-based pH-responsive hydrogel for sustained curcumin (Cur) delivery. Chitosan was modified with norbornene dicarboxylic anhydride to introduce reactive double bonds and enhance solubility, while cysteine-modified Pluronic F127 was crosslinked via UV-induced thiol–ene chemistry to form a stable three-dimensional network. The hydrogel structure was tuned by adjusting the CSNB/CPC ratio and characterized using 1 H NMR, solid-state 13 C NMR, FTIR, TGA, XRD, SEM, and rheology. The optimized hydrogel achieved a drug loading capacity of 13.3 mg/g and exhibited pH-responsive release in simulated gastrointestinal fluids, with kinetics well-fitted to the Peppas–Sahlin model (R 2 > 0.98), dominated by Fickian diffusion. Functional evaluation showed that NC4 displayed nearly 90% DPPH radical scavenging efficiency, and its antioxidant potential was further supported by the Folin–Ciocalteu (GAE) assay and the phosphomolybdenum (AAE) assay, indicating enhanced overall reducing capacity and total antioxidant capacity. NC4 also exhibited strong antibacterial activity against Staphylococcus aureus and Escherichia coli . Biocompatibility was confirmed by CCK-8 and hemolysis assays, with cell viability exceeding 90% and hemolysis rates below 0.35%. Collectively, this multifunctional hydrogel integrates sustained release, antioxidant, antibacterial, and excellent biocompatibility, providing a promising material platform for oral controlled drug delivery and wound healing applications.

Food-grade co-delivery systems have garnered significant attention for their ability to deliver two or more bioactive components simultaneously. Co-delivery systems possessing programmed sequential release properties allow sequential delivery of bioactive components to different sites in the gastrointestinal tract (GIT) to enhance their bioavailability. This study constructed solid-in-oil-in-water multilayer emulsions (S/O/W-E) using carboxymethyl konjac glucomannan-coated gliadin nanoparticles as the solid phase, coconut oil as the oil phase, and carboxymethyl starch/propylene glycol alginate complexes as the aqueous phase, which realized the co-encapsulation of curcumin (Cur) and fucoxanthin (FUC) and their programmed sequential release in the GIT. The programmed sequential release behavior of S/O/W-E was further evaluated by in vitro digestion models. It was demonstrated that both Cur and FUC released less than 16.5% in simulated gastric fluid, following a Fickian diffusion. The Cur located in the oil phase was released in large quantities (67.3%) in simulated intestinal fluid, predominantly through erosion. Owing to the action of β-mannanase, 60.3% of FUC located in the solid phase was released into simulated colonic fluid and dominated by the erosive mechanism. In addition, in vivo bioavailability evaluation and fluorescence imaging experiments confirmed that S/O/W-E enhanced Cur bioavailability by 6.4-fold through delivering it to the small intestine and inhibited FUC release in the upper GIT by delivering substantial amounts of FUC to the colon. This study is beneficial for effectively expanding the application of S/O/W-E in co-delivery systems.

The aim of this study was to construct a series of BA-esterified Lanzhou lily polysaccharides (BLLP) capable of self-assembling into nanoparticles (BLLP NPs). And BLLP NPs achieved synergistic immune-enhancing effects with curcumin (Cur). The successful synthesis of BLLP was confirmed by FT-IR and 1 H NMR. The BLLP NPs exhibit a regular spherical morphology, with a particle size range of 260–410 nm and good dispersion (PDI < 0.32) as determined by DLS. These nanoparticles showed good biosafety (cytotoxicity >100 %, hemolysis <5 %) and can be effectively taken up by cells. The loading capacity of BLLP NPs for Cur can reach 17.60 %, and the encapsulation rate was 48.69 %. After encapsulation, the water solubility, photostability and thermal stability of Cur in BLLP-Cur NPs were enhanced by 83.1-fold, 2.6-fold and 1.4-fold, respectively, compared with free Cur. In vitro simulated release and in vivo absorption showed that BLLP-Cur NPs had a better release from the intestines (in accordance with the Korsmeyer-Peppas model) and a longer intestinal residence time. In in vitro immunoreactivity assays (neutrophil phagocytosis, NO release, cytokine and transcription factor levels) BLLP-Cur NPs exhibited the best immunoreactivity due to the synergistic effect of Cur and BLLP NPs. This study could support the construction of curcumin synergistic bioactive nano-delivery system and diversified applications of Lanzhou lily polysaccharides in functional foods.

To address the challenges posed by excessive inflammatory responses and reactive oxygen species (ROS) in the complex microenvironment during pathological bone defect repair, this study developed a composite scaffold. First, zeolitic imidazolate framework-8 (ZIF-8) loaded with curcumin (CCM) was grown in situ on the surface of strontium carbonate (SrCO3) to form SrCO3@CZIF-8 composite particles with a core−shell structure. Subsequently, these particles were integrated into a poly-l-lactic acid (PLLA) matrix using selective laser sintering technology. Experimental results indicate that the CCM and Zn2+ ions released during scaffold degradation effectively scavenge hydroxyl radicals (with a scavenging efficiency of 61.98% after 24 h) and significantly alleviate inflammatory responses. Furthermore, the released Sr2+ ions enhance osteogenic activity and promote osteogenic differentiation, suggesting that this scaffold holds potential for bone defect repair in complex pathological environments.
