We report a plasmon-mediated route to drive the azo coupling of 4-aminothiophenol to 4,4′-dimercaptoazobenzene on a gold nanostructure on a nanoelectrode (NSoNE) platform. Gold nanostructures were grown on electrochemically etched gold nanoelectrodes to form NSoNE substrates. The curved tip showed a larger enhancement factor than the planar rear surface, with values of 1.31× 104 and 3.08 × 103. Surface-enhanced Raman spectra tracked the azo coupling reaction in real time. At 88 μW μm–2, the curved tip reached a steady state in 37.5 s, while the planar region required 108.4 s. Reducing the power to 44 μW μm–2 slowed the process. Phenyl isocyanide (PIC) served as a molecular thermometer. The C≡N peak of PIC showed a linear red shift with temperature, with a slope of −0.246 cm–1 °C–1. Calibrated maps revealed a nonuniform thermal field that peaked at the tip. Potassium bromate was used to consume hot electrons. The reaction still proceeded in 0.8 mmol L–1 of KBrO3 but with longer steady-state times of 56.4 s at the curved tip and 109.3 s at the planar region. Dark-field heating experiments indicate that thermal effects can effectively activate the 4-ATP coupling reaction, yet they are insufficient to achieve the maximum reaction state. Overall, the results demonstrate that thermal effects can activate and accelerate the reaction process, with the most pronounced influence near the tip; moreover, electron-related reaction pathways also play a certain role.

Ankylosing spondylitis (AS) and osteoarthritis (OA) are two common rheumatic diseases. Although their underlying mechanisms differ significantly—the former being an autoimmune inflammatory condition and the latter a degenerative disorder—they exhibit substantial overlap in early clinical symptoms. Conventional serological tests lack sufficient specificity, leading to a high risk of misdiagnosis. This study developed a highly sensitive surface-enhanced Raman scattering (SERS) biosensing platform based on Ag NPs loaded onto Ti 3 C 2 T x MXene for the early diagnosis of AS and OA. High-density, uniform growth of Ag NPs was achieved on MXene surfaces via in situ self-reduction of AgNO 3 . This composite system fully leverages MXene's unique metalloid electronic structure to enable efficient interfacial charge-transfer pathways. This facilitates strong synergistic coupling between MXene-induced chemical enhancement (CM) and electromagnetic enhancement (EM) arising from localized plasmon resonance in Ag NPs, significantly boosting the substrate's detection sensitivity. Experiments demonstrated that the composite substrate exhibited optimal SERS activity at an AgNO 3 concentration of 0.1 M. This substrate enabled ultrasensitive detection of the probe molecule (Rhodamine 6G, R6G) with a detection limit as low as 10 −8 M. By comparing SERS spectra from healthy controls (HS), AS, and OA subjects, this study revealed significant spectral differences reflecting specific alterations in serum antioxidant, lipid, and amino acid levels. To achieve intelligent classification and diagnosis of HS, AS, and OA, this study integrated SERS spectroscopy with deep learning algorithms. Three deep learning architectures—Artificial Neural Network (ANN), One-Dimensional Convolutional Neural Network (CNN), and Residual Network (ResNet)—were constructed, and their diagnostic performance was compared. Quantitative evaluation demonstrated that the ANN model established a strong classification benchmark, whereas the ResNet model performed poorly due to overfitting. The CNN model demonstrated clear superiority across all evaluation metrics, achieving 91.36% accuracy and an AUC of 0.9896, effectively capturing subtle spectral fingerprint differences. This platform integrates SERS technology with deep learning, offering a novel tool with significant clinical potential for the rapid, non-invasive, and precise differential diagnosis of AS and OA.

Dual-mode detection integrating SERS and UCL for sensitive and visualized detection of CA125. • A type-II band alignment for efficient charge transfer was featured via MXene/MoS 2 heterostructure. • The dual-mode immunoassay kit was developed as a practical tool for clinical ovarian cancer diagnostics. Ovarian cancer (OC) is one of the most common cancers with a high mortality rate, posing a severe threat to female health. Cancer antigen 125 (CA125) has been broadly acknowledged as a significant oncomarker for monitoring OC. Herein, we present a dual-mode immunosensor that integrates surface-enhanced Raman scattering (SERS) and upconversion luminescence (UCL) to facilitate the sensitive and visualized detection of CA125. The MXene/MoS 2 composites were employed as the SERS substrate, whose abundant surface functional groups enable robust antibody immobilization while efficient interfacial charge transfer markedly boost the chemical enhancement. Specifically, a typical type-II band alignment was observed in MXene/MoS 2 heterostructure, which was favorable for efficient charge transfer and resulted in a significant SERS effect with an enhancement factor of 5.24 × 10 7 . Furthermore, the co-doped Yb 3+ /Er 3+ ions UCL probes were employed to improve the detection capability for CA125 by visible green fluorescence. The developed dual-mode immunosensor demonstrated a wide linear detection response, covering the range from 10 to 10 −4 IU/mL. Moreover, the immunosensor displayed a highly sensitivity in detecting of CA125, with a limit of detection as 4.46 × 10 −5 IU/mL for SERS mode and 5.01 × 10 −5 IU/mL for UCL mode. Based on the favorable dual-mode immunosensor, it is proposed as a reliable tool that shows great potential for the detection of CA125. AgNP/MXene arrays enabled a compact biomimetic SERS e-nose. • Chemically distinct MXenes generated cross-reactive VOC responses. • MXene adsorption and AgNP enhancement produced VOC fingerprints. • Four similar VOCs were classified with 100% accuracy by PCA/HCA. • Three sensing channels achieved efficient VOC discrimination. The discrimination of structurally similar volatile organic compounds (VOCs) remains a significant challenge due to the limited selectivity of existing sensing materials. To address this, we present a methodologically novel, novel bio-inspired electronic nose platform based on a surface-enhanced Raman scattering (SERS) sensor array of silver nanoparticles/MXene (AgNP/MXene) composites for intelligent VOC identification. The proposed approach combines a rationally designed, chemically orthogonal MXene library (Ti 3 C 2 , V 2 C, and a high-entropy MXene, TiVCrMoC 3 ) with an optimized multivariate analysis workflow, thereby establishing a generalizable methodology for constructing cross-reactive SERS sensing systems. The MXene components serve as superior adsorbents to preconcentrate VOC molecules, while the in-situ grown AgNPs provide strong plasmonic enhancement, translating adsorption events into distinct molecular “fingerprint” spectra. The cross-reactive responses of the array to four similar VOCs (dipentene, ethyl lactate, isopropyl acetate, and propylene glycol monomethyl ether). The generated multidimensional SERS data is effectively decoded using pattern recognition algorithms, namely hierarchical cluster analysis (HCA) and principal component analysis (PCA), which provided a reliable proof-of-concept for the identification of VOC gases. This work thus introduces a new materials–data co-design strategy for biomimetic SERS e-nose development, enabling reliable discrimination of structurally similar VOCs in complex environments. The substrate serves as the functional “chip” in surface-enhanced Raman spectroscopy (SERS) analysis. In this work, a rationally designed core–shell composite sensing material (AgNW@COF) was synthesized by encapsulating silver nanowires (AgNWs) within a covalent organic framework (COF) shell (TAPB-BTCA-COF) via an in-situ growth strategy. The AgNWs core provides strong electromagnetic enhancement through localized surface plasmon resonance, while the porous, chemically robust COF shell not only stabilizes the AgNWs against oxidation and aggregation but also acts as a selective molecular concentrator. Optimization of the core–shell architecture revealed that precise control over COF shell thickness is critical: a balanced shell ensures efficient analyte enrichment near the “hot spots” without compromising plasmonic coupling. Leveraging this engineered substrate with a streamlined QuEChERS-based sample pretreatment, we developed a rapid, sensitive, and reliable SERS method for the quantification of triazophos, a representative organophosphate pesticide, in Shanghai green vegetable samples. The method exhibits linearity over the range of 30–800 ng/mL in spiked matrix (R 2 = 0.9785), with a limit of detection of 1.19 ng/g (S/N = 3), and mean recovery rates of 89.5–126.5% (n = 3) at three spiking levels. Crucially, total analysis time was reduced from ∼1 h/sample using the national standard LC–MS/MS method to less than 30 min, without sacrificing accuracy or reproducibility. This study thus advances both the rational design of multifunctional SERS substrates and the practical deployment of SERS for on-site food safety monitoring.

Considering the irreversible nature of Alzheimer's disease (AD), early diagnosis is of great importance for AD treatment. Both β-amyloid (Aβ) and monoamine oxidase B (MAO-B) are potential biomarkers for AD. This study developed a surface-enhanced Raman scattering (SERS) sensor using gold-nanocube (OX-AuNCs) to detect Aβ42 and MAO-B. OX-AuNCs, a type of gold-nanocube with open gaps, achieved a Raman enhancement factor of 8.99 × 10 8 due to numerous hot spots. The specific affinity between Thioflavin T (ThT) and Aβ42 inhibits ThT's intramolecular rotation, reducing its SERS signal. This reduced signal showed a linear correlation with Aβ42 concentrations from 66.70 pM to 0.20 μM, achieving a limit of detection (LOD) at 44.30 pM, while also distinguishing different aggregation levels of Aβ42. Interestingly, SERS intensity of phenethylamine (PEA) also decreased significantly when interacting with MAO-B. A linear relationship was found between the change in PEA intensity and MAO-B concentration (0.01–20.00 μg mL −1 ), yielding an LOD of 5.00 ng mL −1 . The proposed SERS sensor effectively detected two potential AD biomarkers, Aβ42 and MAO-B, in artificial cerebrospinal fluid (ACSF) and human serum with satisfactory recovery rates, respectively. These results show promise for clinical diagnosis and drug screening for AD.

Therapeutic drug monitoring (TDM) of mitoxantrone (MIT) is essential for balancing efficacy and toxicity, yet current methods are limited by complex pretreatment, long turnaround times, and matrix interference. Herein, we developed a SERS sensor based on a pectin/AgNPs@CMC/CS/PAAM composite hydrogel for separation-free detection of cationic anticancer drugs in plasma. Fabricated via one-step UV photopolymerization, the hydrogel's dense network provides size-sieving to exclude plasma proteins, while abundant carboxyl groups confer a strong negative surface charge for selective electrostatic enrichment of positively charged drug molecules. This dual mechanism enhances specificity toward cationic analytes in complex matrices. Using MIT as a model, the sensor achieved 82.6%–101.7% recoveries in 10% plasma, a detection limit of 4.7 × 10 −10 M, and excellent reproducibility. Requiring only simple dilution, this platform offers a practical and ultrasensitive tool for TDM of cationic drugs in combination chemotherapy.

A highly stable Au@Au SERS nanolabels-based LFIA was developed for rapid FT analysis. • The AuNPs DTNB @Au exhibited high stability during a 90-day storage period with a RSD value of 7.71%. • The LOD of 2.27 fg mL −1 and the LDR of 10 fg mL −1 to 10 μg mL −1 were achieved. • The entire analysis process was completed within 15 min. Free testosterone (FT) serves as a crucial biomarker for endocrine assessment. Nevertheless, the rapid and accurate analysis of FT presents a significant challenge due to its low abundance and the interference from the matrix. In this study, we developed a lateral flow immunoassay (LFIA) integrated with gold-core gold-shell nanoparticles (Au@Au NPs)-based surface-enhanced Raman scattering (SERS) nanolabels for the rapid analysis of FT. The Raman signal molecules (RSMs) were embedded within the interstitial gap between the Au core and the Au shell, significantly contributing to SERS enhancement while also improving the stability of the SERS nanolabels. The proposed biosensor demonstrated a linear dynamic detection range (LDR) of 10 fg mL −1 to 10 μg mL −1 (R 2 = 0.9914) and a limit of detection (LOD) of 2.27 fg mL −1 , with the entire analysis process completed within 15 min. Additionally, the results indicated that the platform exhibited remarkable reproducibility, stability, and selectivity, along with reliable performance in human serum samples. This generates a simple, rapid, and highly sensitive strategy for FT detection, demonstrating considerable potential for point-of-care testing of low-abundance biomarkers. Conventional synthetic routes to Ag nanocubes (AgNCs) often leave reagent residues on the particle surface, thereby compromising analyte accessibility and surface-enhanced Raman scattering (SERS) sensitivity. Here, AgNCs were grown in-situ on two-dimensional (2D) nanocellulose films via a photo-induced reduction process, yielding surfactant-free particles with tunable morphology. No additional reducing agents or surfactants were introduced during synthesis, reducing surfactant-derived surface coverage and improving the accessibility of the resulting AgNCs. Notably, the dissolved oxygen in the reaction solution played an important etching role in eliminating the twins during the nucleation process and generating AgNCs with a single crystal structure. The nanocellulose film served both as a photo-reactive substrate, where surface hydroxyl groups acted as reductive sites for the photo-induced conversion of Ag(I) to Ag(0), and as a 2D support that stabilized interfacial nucleation and growth. In addition, the in-situ synthesized AgNCs were uniformly distributed across the nanocellulose film surface, enabling one-step fabrication of flexible SERS substrates under mild conditions. The optimized substrate enabled highly sensitive detection of Rhodamine 6G (R6G), with a limit of detection of 6.24 × 10 −12 g L −1 , approximately fivefold lower than that of the same substrate after surfactant adsorption. This strategy provides a simple platform for constructing nanocellulose-based SERS substrates for trace detection.

The excessive activation of sympathetic nerve is closely related to cardiovascular diseases (CVDs). In this study, we developed a novel MIL-100(Fe)@Ag surface-enhanced Raman scattering (SERS) substrate for the simultaneous and effective detection of plasma norepinephrine (NE) and adenosine—two pivotal sympathetic hormones. The microscopic morphology and crystal structure of the substrate were characterized using techniques such as transmission electron microscope (TEM) and X-ray diffraction (XRD). Mechanistically, the HOMO-LUMO energy levels of the detection molecules were calculated using Gaussian. The substrate realized a dual SERS enhancement mechanism through the synergistic effect of localized surface plasmon resonance and interfacial charge transfer. Consequently, the MIL-100(Fe)@Ag platform exhibited high-sensitivity quantitative detection capabilities for NE and adenosine: the detection limit for NE is 0.596 nmol/L; the detection limit for adenosine is 0.24 nmol/L. Crucially, when applied to patients with pheochromocytoma—a neuroendocrine tumor characterized by plasma catecholamine surges, this method achieved robust and stable detection of plasma NE. It offered comparable diagnostic accuracy to conventional mass spectrometry while significantly reducing analysis complexity. Therefore, this innovative strategy may hold great promise for the long-term, decentralized monitoring of sympathetic nerve activity in populations at risk for CVDs or related clinical disorders.

Qualitative and semi-quantitative detection and identification of multiple targets is of great importance in complex sample analysis, while surface-enhanced Raman scattering (SERS) could be an efficient strategy. Here, to efficiently promote the enhancement effect, Ni doping and Au nanoparticles (AuNPs) surface modification were adopted to develop a MoS 2 SERS substrate composed of intercrossed nanoflowers, named AuNPs@Ni-MoS 2 , which exhibits high SERS enhancement towards the Raman reporter (4-mercaptobenzoic acid, 4-MBA) with an enhancement factor (EF) of 10 8 . In complex matrices of different mushrooms, this substrate can also sensitively detect α -amanitin and β -amanitin with a limit of detection (LOD) of 10 −8 M for both, which laid a solid foundation for the accurate discrimination via the following machine learning-based approach. Meanwhile, based on the AuNPs@Ni-MoS 2 substrate, sensitive and specific detection of Hg 2+ was attributed to the formation of an Au-Hg alloy, which could greatly lower the catalytic potential required for the oxidation of the colorless chromogenic agent (3,3’,5,5’-tetramethylbenzidine, TMB) to a yellow oxidized TMB (TMB 2+ ). This study not only provides a reference model for improving charge transfer efficiency in SERS system but also offers technical support for rapid screening multiple analytes in complex samples.

Squamous cell carcinoma antigen (SCCA) a magnetic enrichment–assisted core–satellite surface-enhanced Raman scattering (SERS) biosensor for ultrasensitive squamous cell carcinoma antigen (SCCA) detection is reported. Magnetic nanoparticles functionalized with SCCA capture antibodies act as enrichment cores, while 4-mercaptobenzoic acid (MBA)–encoded Au@Ag core–shell nanoparticles conjugated with SCCA detection antibodies serve as SERS satellites. Target-mediated sandwich immunorecognition induces a dynamically assembled core–satellite structure, bringing plasmonic nanogaps into close proximity and generating reproducible electromagnetic hot spots for signal amplification. Magnetic pre-enrichment and controlled assembly help reduce nonspecific protein interference in serum and contribute to stable signal generation. The biosensor exhibited a monotonic concentration-dependent Raman response over the range of 1 fg/mL to 10¹¹ fg/mL and achieved a limit of detection (LOD) of 60.0 fg/mL. Quantitative determination of serum samples was performed using an independently established matrix-matched calibration curve. The platform was further validated using real human serum samples from early-stage lung SqCC patients ( n = 8) and healthy controls ( n = 8), showing higher SCCA concentrations in patient samples than in healthy controls and clear discrimination between the two groups. Overall, the proposed magnetic enrichment–assisted core–satellite SERS immunoassay provides a robust analytical strategy for sensitive SCCA quantification in complex serum matrices.
