PVC microplastics facilitate uropathogenic Escherichia coli pathogenicity by enhancing host cell invasion and mitochondrial-dependent pyroptosis

Zhiyun Wang,Ziwei Chen,Sheng Wang,Luna Cui,Minghao Tang,Xianhua Liu,Tao Wang,Xueping Li

Microplastics (MPs), ubiquitous environmental pollutants, can enter the human body and exacerbate health risks. Polyvinyl chloride MPs (PVC-MPs) are emerging contaminants of particular concern due to direct clinical exposure pathway. However, the role of PVC-MPs in pathogenic bacterial infections remains poorly understood. Here, we investigate the role of PVC-MPs for the pathogenicity of uropathogenic Escherichia coli (UPEC), the primary causative agent of urinary tract infections (UTIs). Results demonstrate that PVC-MPs significantly enhance both acute and chronic UTIs. Mechanically, PVC-MPs act as carriers, enriching UPEC on their surface and enhancing UPEC invasion of bladder epithelial cells (BECs) via receptor-independent pathway, thereby promoting UPEC colonization in mouse bladders and kidneys. After entering BECs, intracellular PVC-MPs induce mitochondrial dysfunction, causing mitochondrial reactive oxygen species accumulation and mitochondrial DNA leakage, which activate Caspase-1-dependent pyroptosis, promoting urothelial cell shedding and UPEC chronic infection. These findings establish PVC-MPs as mediators of UTI pathogenesis, providing critical mechanistic insights into how environmental pollutant exposure may enhance susceptibility to infectious diseases. Polyvinyl chloride microplastics have been detected within clinical settings and may influence bacterial infections. Here, the authors demonstrate that these microplastics facilitate acute infection by uropathogenic E. coli, helping the bacteria evade immune cells and increase shedding.

Enhancing adipogenic differentiation efficiency and gene expression in yak stromal vascular cells through 3D spheroid culture

Ma Yannan,Zheng Lei,He Pengjia,Pan Chanchan,Wang Bo,Song Shuzhen,Hou Mingjie,Chen Wen,Ma Xiaoyan,Lei Zhaomin

Intramuscular fat deposition is a key determinant of meat quality in livestock species; however, yak ( Bos grunniens ) stromal vascular cells (SVCs) exhibit relatively low adipogenic differentiation efficiency under conventional in vitro conditions. In this study, we established a three-dimensional (3D) spheroid culture system to improve adipogenic differentiation of yak SVCs and compared it with traditional two-dimensional (2D) monolayer culture. Yak SVCs were isolated from longissimus dorsi muscle and characterized prior to differentiation. Cells were subjected to adipogenic induction under both 2D and 3D conditions. Adipogenic differentiation was evaluated by morphological observation, Oil Red O staining and quantification, and quantitative real-time PCR (RT-qPCR) analysis of adipogenic marker genes. Additionally, Western blot analysis of key adipogenic proteins was performed specifically on yak SVCs. To validate the induction system, murine C3H10T1/2 cells were included as a parallel control group and evaluated using the aforementioned non-protein assays. The results showed that 3D culture significantly enhanced lipid accumulation and increased the expression of adipogenic marker genes compared with 2D culture in yak SVCs. Consistently, protein levels of PPAR γ were significantly elevated in the 3D group of yak SVCs, while FABP4 showed an upward trend. Furthermore, the C3H10T1/2 control group exhibited successful adipogenic differentiation with similarly enhanced lipid accumulation and gene expression under 3D conditions, confirming the reliability of our system. In conclusion, 3D spheroid culture improves adipogenic differentiation of yak SVCs and provides a useful in vitro model for studying adipose development in yak. This system may facilitate future studies on the regulation of intramuscular fat deposition in livestock species.

A self-sufficient nanoreactor orchestrating calcium overload and sonodynamic/chemodynamic-amplified ferroptosis for oral squamous cell carcinoma therapy

Wu Lei,Wang Chuiyin,Chen Qian,Wang Aiqi,Peng Meiying,Zhang Gengming,Cao Mingxin,Guo Zhaoyang,Yang Xiaoying,Wang Yinsong

Bacteria-derived components hold great promise for cancer therapy owing to their versatile biological properties. We previously reported that Porphyromonas gingivalis cell walls (PgCW), rich in intrinsic µ-oxo bisheme ([Fe(III)PPIX] 2 O), can effectively mediate both sonodynamic therapy (SDT) and chemodynamic therapy (CDT). More recently, we identified PgCW’s inherent potential to trigger ferroptosis. However, its therapeutic efficacy is limited by the hypoxic tumor microenvironment (TME) and insufficient endogenous hydrogen peroxide (H 2 O 2 ) required for catalytic reactions. Herein, a self-sufficient nanoreactor (DCP) was developed by depositing calcium peroxide (CaO 2 ) within dendritic large-pore mesoporous silica nanoparticles (DLMSNs), followed by the electrostatic adsorption of PgCW. Under ultrasound irradiation, DCP combines SDT and CDT with continuous glutathione (GSH) depletion to trigger ferroptosis. In the acidic TME, the decomposition of CaO 2 generates O 2 to enhance PgCW-mediated SDT, supplies H 2 O 2 for Fenton-like CDT, and releases Ca 2+ to induce calcium overload. The abundant reactive oxygen species (ROS) from CaO 2 -supplemented SDT and CDT, together with GSH exhaustion, amplify ferroptosis, driving severe lipid peroxidation (LPO) and cell membrane rupture to release damage-associated molecular patterns (DAMPs) for primary tumor regression. Furthermore, intracellular calcium overload synergistically promotes this ferroptosis-mediated immunogenic cell death (ICD), which, combined with the innate adjuvanticity of PgCW, drives robust systemic immune activation to suppress distant tumors. In summary, this nanoreactor overcomes TME limitations through self-sufficient substrate supply and efficiently combines calcium overload with amplified ferroptosis to suppress the growth and metastasis of oral squamous cell carcinoma (OSCC). These findings suggest a potential strategy for precision cancer immunotherapy. Graphical

Macrophage FOLR2 drives liver fibrosis via intercellular engagement of TGFβRII

Yuan Man-man,Zheng Bing-feng,Wang Jing-lin,Xu Shuai-qi,Wang Sheng-lan,Tan Yang,Feng Yong,Dong Xian-chi,Zhu Qi-hua,Yan Jie,Xu Qiang,Wu Xing-xin

Hepatic stellate cells (HSCs) and macrophages are key regulators of liver fibrosis, yet their direct communication during fibrogenesis remains incompletely characterized. We showed that genetic deletion of folate receptor beta ( Folr2 ) significantly attenuated experimental liver fibrosis in mice. In both human and murine fibrotic livers, FOLR2 + macrophages are frequently juxtaposed with activated HSCs. Functionally, FOLR2 expressed by reparative, but not M1 or scar-associated, macrophages promotes HSC activation in a contact-dependent manner. Mechanistically, FOLR2 binds to transforming growth factor beta receptor II (TGFβRII) on HSCs, sustaining transforming growth factor beta 1 (TGF-β1) signaling and driving fibrogenesis. Finally, the natural compound fraxinellone targets FOLR2, disrupts its interaction with TGFβRII, and attenuates HSC activation and liver fibrosis. These findings identify the FOLR2-TGFβRII intercellular interaction as a critical mediator of macrophage-HSC crosstalk and highlight its disruption as a promising therapeutic strategy against liver fibrosis.

Dual-targeting oral nanoparticle mucosal vaccine against brucella infection: co-targeting M cells and dendritic cells

Tian Tingting,Qi Xinxin,Shi Juan,Shang Kaiyu,Shi Huidong,Luo Jiarui,Chai ZhengLong,Li Chuang,Ding Jianbing,Zhu Yuejie,Zhang Fengbo

This study designed and evaluated a chitosan nanoparticle-based mucosal vaccine (CS-CKS9-MEV-DCpep-NPs) targeting M cells and dendritic cells against Brucella . From four immunogenic outer membrane proteins (OMP10, Omp25, Omp31, BtpB), 41 specific epitope peptides were screened and optimized via bioinformatics and ELISPOT for high antigenicity and safety. The dual-targeting strategy uses CKS9 to enhance M cell-mediated antigen uptake and DCpep, together with CS-NPs, to activate DCs and promote their maturation. This coordinates cDC1, cDC2, and pDC subsets, inducing CD8⁺ and CD4⁺ T cell responses and strengthening Th1-biased immunity against intracellular bacteria. The vaccine activates mucosal (sIgA), humoral (IL-21, memory B cells), and cellular (Th1/CTL, central memory T cells) immunity. It significantly reduces bacterial load and tissue damage, offering comprehensive protection. Compared with conventional vaccines, this oral nanovaccine provides high safety, precise targeting, and multi-dimensional immune activation, representing a novel strategy for brucellosis control. Cooked meat-derived extracellular vesicles ssc-miR-1 induces metabolic disorders in the mice liver via PI3k/AKT pathway

Shen Linyuan,Ma Jianfeng,Liang Shuang,Yang Yiting,Liao Tianci,Lei Yuhang,Chen Lei,Wang Yan,Zhao Ye,Niu Lili,Gan Mailin,Zhu Li

Diet derived extracellular vesicles (EV) can be absorbed and influence physiological processes. Herein, this study investigates the potential health impacts of extracellular vesicles in meat products. Experimental results showed the extracellular vesicles derived from cooked red meat (RM-EV) and white meat (WM-EV) were successfully isolated via ultra-high speed centrifugation. The median particle size of RM-EV is 137.5 nm and that of WM-EV is 116.1 nm. RM-EV and WM-EV are added to the drinking water of mice for ten weeks. The mice developed insulin resistance and abnormal lipid metabolism in the liver. The influences of RM-EV are more pronounced for mice than WM-EV. High-throughput sequencing indicated that ssc-miR-1 (52.05%) and scc-miR-133a-3p (14.21%) are the most abundant microRNA in RM-EV. However, the highest content of microRNA is gga-miR-133a-3p (44.16%) in WM-EV, followed by gga-miR-1 (21.58%). Therefore, the most abundant ssc-miR-1 in red meat EV (RM-EV-miR-1) was selected for in vitro studies. In vitro experiments revealed that RM-EV-miR-1 inhibited cell proliferation by targeting IGF1 in NCTC1469 liver cell. Besides, RM-EV-miR-1 exacerbates insulin resistance in NCTC1469 insulin resistance cell model by targeting PI3K/AKT signaling pathway. Our findings show that the differential health impacts of red and white meat may be partially attributed to the presence of EV. These results provide novel insights into dietary health from the perspective of EV.

Mild and efficient catalytic nitration of o-xylene with NO2 over silicon sulfonic acid to 4-nitro-o-xylene under solvent-free conditions

Yan Jiaqi,Fu Weiwen,You Kuiyi,Zhao Fangfang,Luo He’an

A mild and efficient method for the solvent-free catalytic nitration of o-xylene (OX) employing silicon sulfonic acid (SSA) as a catalyst and NO 2 as a clean nitrating agent, leads to high selectivity for 4-nitro-o-xylene (4-NOX). Under optimal conditions, a 64.6% selectivity for 4-NOX and a 46.7% conversion of OX were achieved. The results showed that the enhanced OX conversion and target product selectivity were attributed to the synergistic catalysis of the combination of the SSA catalyst and the NO 2 –O 2 system. The characterization results showed that the immobilized SSA, which is not only simple and economical to prepare but also exhibits excellent nitration activity, was effectively synthesized via the chemical bonding method. The highly dispersed sulfonic acid groups were covalently anchored to the surface of the silicon acid (SA), thereby enriching acid sites and stabilizing the catalytic performance. Additionally, a possible reaction mechanism for nitrating OX with NO 2 –O 2 over the SSA catalyst was proposed. Compared with the traditional method, this study promotes the selectivity of the target product while reducing acidic wastewater streams, indicating potential application prospects.

Waterborne polyurethane nanocomposites based on reduced graphene oxide/Ag for light-induced antibacterial properties

Li Hui,Qu Jianbo

Graphene oxide was functionalized by dopamine to synthesize polydopamine-reduced graphene oxide (PDRGO). PDRGO was further applied as a template and reductant to reduce silver ion to form silver nanoparticles (PDRGO-Ag). Then, the PDRGO-Ag was incorporated into waterborne polyurethane (WPU) by in situ emulsification to fabricate the PDRGO-Ag embedded WPU nanocomposite films (WPU/PDRGO-Ag). Herein, the PDRGO-Ag was regarded as a photothermal agent that offers effective and controllable photothermal converters for bactericidal applications. Meanwhile, silver nanoparticles acted as an antibacterial agent that endow synergistically enhanced antibacterial properties to the PDRGO-Ag. The resulting film exhibited exceptional antibacterial performance against both E. coli and S. aureus . Moreover, the WPU/PDRGO-Ag films showed improved mechanical properties. The development of this light-induced functional WPU-based film opens a new pathway to antibacterial composite coatings. Graphical

Synthesis of Mg/Fe bimetallic oxide loaded biochar by hydrothermal co-pyrolysis method for efficient removal of uranium(VI)

Feng Zihao,Wang Yang,Ren Qi,Yi Cui,Li Yang,Du Yanjun,Wang Changfu,Wang Yun

Effective U(VI) separation from uranium wastewater is crucial for nuclear energy's sustainable growth. A magnetic, cauliflower-shaped magnesium-iron bimetallic oxide loaded biochar (MFBC) was fabricated via a green hydrothermal co-pyrolysis. Its U(VI) adsorption process was pH-dependent and fitting Langmuir–Freundlich and pseudo-second-order models, the theoretical maximum adsorption capacity of U(VI) by MFBC peaked at 1354.8 mg·g −1 (pH 5.0, 30 min). After five regenerations, 90.3% removal remained. The adsorption mechanism of MFBC for U(VI) involved electrostatic interaction, surface complexation, ion exchange and reduction reaction, making it a highly efficient and excellent adsorbent for treating uranium-containing wastewater.

Unraveling the modulation of covalently bonded carbon matrix on Fe-N4 active sites for enhanced laccase-like activity: toward advanced detection and degradation of phenolic pollutants

Zhang Guo-Qi,Long Xia,Shi Yu-Han,Wu Wei,Zhou Xin-Yue,Jiang Wen-Cai,Li Tian-Qi,Xiang Wen-jing,Liu Wei-Ping,Zhao Yan

Herein, we present a proof-of-concept investigation into the assessment of the laccase-like activity of covalently bonded carbon environments connected to Fe-N 4 sites, using the closed π conjugated phthalocyanine-based intrinsic covalent organic polymers (COP) with well-designed structures. Based on the theoretical prediction and experimental implementation, the impact of the covalent-bonded carbon matrix on the laccase-like activity of COP-X (X represents the degree of conjugation) was systematically investigated. Further calculation results by density functional theory showed that the strongest laccase-like catalytic activity of COP-2 may be due to the facile desorption of catalytic intermediates, and the laccase-like catalytic mechanism of COP-2 nanozymes was also deeply understood. As anticipated, the utilization of COP-2 nanozymes with laccase-like activity enabled the achievement of excellent sensitivity for monitoring epinephrine in human serum. In addition, the COP-2 nanozymes also facilitate the detection and degradation of phenolic pollutants present in the environment. This finding presents a novel perspective for the rational design of high-performance laccase-like iron based nanozymes, and also lays the foundation for clarifying the origin of the laccase-like activity of nanozymes. Effect of annealing on self-assembly kinetics and structural colors of supramolecular polymer patterns

Deng Shuai,Mao Xi,Wang Mian,Deng Renhua,Zhu Jintao

Block copolymer (BCP) photonic crystals (PCs) have great potential in color decoration, optical encryption, and smart sensing because of their excellent flexibility and stimuli-responses. Supramolecular BCPs (SBCPs) provide a versatile platform for engineering structural colors through reversible non-covalent interactions, yet their precise dynamic modulation under confined environments remains underexplored. Here, we develop an integrated real-time monitoring system to elucidate the temperature-solvent synergistic annealing strategy to regulate the disorder-to-order transition of hydrogen-bonded comb-like SBCPs and homopolymers binary co-assembly system, which can yield a lamellar structure and exhibit structural color. Systematic investigation revealed that increasing solvent content accelerates ordering kinetics and induces a red-shift of structural color in their swollen states, while the equilibrium structural color remains solvent-independent upon solvent removal. Unexpectedly, elevating the temperature decelerates ordering kinetics and induces a blue-shift in both swollen and dry states. This counterintuitive phenomenon can be attributed to thermally reduced solvent uptake (by SBCPs) and partial hydrogen-bond dissociation, diminishing the molecular weight and rigidity of SBCPs. These findings elucidate the critical role of swelling and dynamic hydrogen-bond interactions in governing assembly pathways, structure and optical properties of SBCPs, providing a versatile platform for designing stimuli-responsive PC materials with on-demand colors and color switching.

Modification of zero-valent iron with xanthan gum and oxalic acid enhances Cr(VI) removal, reduces bacterial toxicity, and improves transport

Guo Jia,Xu Jiaqi,Zhang Xiangwei,Li Zhengmao,Liu Fuyang,He Lei,Long Mingce,Tong Meiping

The rapid aggregation and sedimentation, the high toxicity towards bacteria, and the passivation of surfaces hinder the application of zero-valent iron (ZVI) particles in groundwater remediation. To overcome the shortages, ZVI particles were modified using oxalic acid (an environmental-friendly low-molecular-weight organic acid) together with xanthan gum (a safe and cost-effective high-molecular-weight polymer). We found that ZVI modified with both oxalic acid and xanthan gum exhibited superior Cr(VI) removal performance than those modified solely with oxalic acid and unmodified ZVI. Due to the reduced Fe 2+ release, the toxicity towards Escherichia coli by oxalic acid-xanthan gum modified ZVI was greatly inhibited. Via enhancing electrostatic repulsion through increasing the negative zeta potentials, modification of ZVI by oxalic acid and xanthan gum improved the dispersion of ZVI and reduced their sedimentation in suspension. Moreover, the mobile performance of ZVI in porous media was greatly improved by the modification with oxalic acid and xanthan gum due to the enhanced electrostatic repulsive interaction between ZVI and quartz sand. The observations held true in actual groundwater samples without and with heavy metal contamination. Clearly, the modification of ZVI by using oxalic acid and xanthan gum could be a feasible approach to enhance the practical application potential for the in-situ decontamination of groundwater.
